We employ a rigorous analysis using the photometric classifier SuperNNova on state-of-the-art simulations of SN samples to determine cosmological biases due to such ‘non-Ia’ contamination in the Dark Energy Survey (DES) 5-yr SN sample. Depending on the non-Ia SN models used in the SuperNNova training and testing samples, contamination ranges from 0.8 to 3.5 per cent, with a classification efficiency of 97.7–99.5 per cent. Using the Bayesian Estimation Applied to Multiple Species (BEAMS) framework and its extension BBC (‘BEAMS with Bias Correction’), we produce a redshift-binned Hubble diagram marginalized over contamination and corrected for selection effects, and use it to constrain the dark energy equation-of-state, w. Just as a single polypeptide strand can self-fold into a complex 3D structure, a single strand of DNA can self-fold into DNA origami. Most DNA origami structures (i.e., the scaffold-staple and DNA tiling systems) utilize hundreds of short single-stranded DNA. As such, these structures come with challenges inherent to intermolecular construction. Many assembly challenges involving intermolecular interactions can be resolved if the origami structure is constructed from one DNA strand, where folding is not concentration dependent, the folded structure is more resistant to nuclease degradation, and the synthesis can be achieved at an industrial scale at a thousandth of the cost. This review discusses the design principles and considerations employed in single-stranded DNA origami and its potential benefits and drawbacks.

DNA-PAINT enabled super-resolution imaging through the transient binding of fluorescently-labelled single-stranded DNA (ssDNA) imagers to target ssDNA. However, its performance is constrained by imager background fluorescence, resulting in relatively long image acquisition and potential artifacts. We designed a molecular beacon (MB) as the PAINT imager. Unbound MB in solution reduces the background fluorescence due to its natively quenched state. They are fluorogenic upon binding to target DNA to create individual fluorescence events. We demonstrate that MB-PAINT provides localization precision similar to traditional linear imager DNA-PAINT. We also show that MB-PAINT is ideally suited for fast super-resolution imaging of molecular tension probes in living cells, eliminating the potential of artifacts from free-diffusing imagers in traditional DNA-PAINT at the cell-substrate interface.

Objective Type 2 diabetes mellitus (T2D) is the result of a dysregulation of insulin production and signalling, leading to an increase in both glucose concentration and pro-inflammatory cytokines such as interleukin (IL)-6 and tumour necrosis factor (TNF)-α. Previous work showed that T2D patients exhibited immune dysfunction associated with increased adhesion molecule expression on endothelial cell surfaces, accompanied by decreased neutrophil rolling velocity on the endothelial cell surface. Changes in cell rolling adhesion have direct vascular and immune complications such as atherosclerosis and reduced healing time in T2D patients. While previous studies focused primarily on how endothelial cells affect neutrophil rolling under T2D conditions, little is known about changes to neutrophils that affect their rolling. In this study, we aim to show how the rolling behaviour of neutrophils is affected by T2D conditions on a controlled substrate. Results We found that neutrophils cultured in T2D-serum mimicking media increased cell rolling velocity compared to neutrophils under normal conditions. Specifically, glucose alone is responsible for higher rolling velocity. While cytokines further increase the rolling velocity, they also reduce the cell size. Both glucose and cytokines likely reduce the function of P-selectin Glycoprotein Ligand-1 (PSGL-1) on neutrophils.

Tavangar et al., We use 6 yr of data from the Dark Energy Survey to perform a detailed photometric characterization of the Phoenix stellar stream, a 15° long, thin, dynamically cold, low-metallicity stellar system in the Southern Hemisphere. We use natural splines, a nonparametric modeling technique, to simultaneously fit the stream track, width, and linear density. This updated stream model allows us to improve measurements of the heliocentric distance (17.4 ± 0.1 (stat.) ±0.8 (sys.) kpc) and distance gradient (−0.009 ± 0.006 kpc deg−1) of Phoenix, which corresponds to a small change of 0.13 ± 0.09 kpc in heliocentric distance along the length of the stream. We measure linear intensity variations on degree scales, as well as deviations in the stream track on ∼2° scales, suggesting that the stream may have been disturbed during its formation and/or evolution. We recover three peaks and one gap in linear intensity along with fluctuations in the stream track. Compared to other thin streams, the Phoenix stream shows more fluctuations and, consequently, the study of Phoenix offers a unique perspective on gravitational perturbations of stellar streams. Leukocyte rolling adhesion, facilitated by selectin-mediated interactions, is a highly dynamic process in which cells roll along the endothelial surface of blood vessel walls to reach the site of infection. The most common approach to investigate cell-substrate adhesion is to analyze the cell rolling velocity in response to shear stress changes. It is assumed that changes in rolling velocity indicate changes in adhesion strength. In general, cell rolling velocity is studied at the population level as an average velocity corresponding to given shear stress. However, no statistical investigation has been performed on the instantaneous velocity distribution. In this study, we first developed a method to remove systematic noise and revealed the true velocity distribution to exhibit a log-normal profile. We then demonstrated that the log-normal distribution describes the instantaneous velocity at both the population and single-cell levels across the physiological flow rates. The log-normal parameters capture the cell motion more accurately than the mean and median velocities, which are prone to systematic error. Lastly, we connected the velocity distribution to the molecular adhesion force distribution and showed that the slip-bond regime of the catch-slip behavior of the P-selectin/PSGL-1 interaction is responsible for the variation of cell velocity.

Introduction Smoking is a modifiable risk factor for many diseases. The public should recognize the impact of smoking on their health and their wealth. The current study aimed to evaluate the cost burden of smoking to target Asia-Pacific countries. Material and Methods The current study estimated the annual spending and lifetime spending of smokers in the target Asia-Pacific countries (Hong Kong, Malaysia, Thailand, South Korea, Singapore, and Australia) on purchasing cigarettes, as well as predicted the revenue that could be generated if smokers spent the money on investment instead of buying cigarettes. Smokers’ spending on cigarettes and the potential revenue generated from investment were estimated based on the selling prices of cigarettes, Standards & Poor’s 500 Index, and life expectancies of smokers. Data were extracted from reports released by the World Health Organization or government authorities. Results The annual expenses (in US$) on purchasing one pack of cigarettes, in decreasing order, were: Australia ($5628.30), Singapore ($3777.75), Hong Kong ($2799.55), Malaysia ($1529.35), South Korea ($1467.30), and Thailand ($657.00). The lifetime spending on purchasing one pack of cigarettes each day were: Australia ($308993.67), Singapore ($207398.48), Hong Kong ($151735.61 for male and $166853.18 for female), South Korea ($80261.31), Malaysia ($72338.26), and Thailand ($31207.50). Conclusions The cost burden of smoking is high from a smoker’s perspective. Smokers should recognize the high economic burden and quit smoking to enjoy better health and wealth.

Studies have provided evidence that Interprofessional Education (IPE) can improve learners’ attitudes, knowledge, skills, behaviors, and competency. Traditionally, IPE is commonly seen in the healthcare professional training in tertiary education. Aging is a global issue that requires more than just a single healthcare sector. It requires interdisciplinary collaboration and understanding to tackle the issues. Therefore, IPE is essential for nurturing university students to tackle the ever-changing global challenges. In addition, different hurdles can hinder IPE development. To have a better understanding of the feasibility, acceptance, and educational value of IPE in Hong Kong, we conducted a cross-sectional quantitative study. We invited teachers and students from a Hong Kong university to fill in an online survey that evaluated their understanding and participation in IPE, their attitude toward IPE, and the barriers to developing IPE from March to June 2020. Among the 37 academic staff and 572 students who completed the survey, 20 (54.1%) teachers and 422 (73.8%) students had never heard of IPE before, and 26 (70.3%) teachers and 510 (89.2%) students had never participated in any IPE activities. Major barriers reported by teachers included an increase in teaching load (72.9%), lack of administrative support (72.9%), lack of financial support and limited budget (67.5%), difficulty to make logistic arrangements (64.8%), and problems with academic schedules and calendars (62.1%). The survey findings revealed that despite the positive attitude of university teachers and students toward IPE, barriers that could hinder the development of IPE included heavy teaching and administrative load and logistic arrangement for classroom arrangement and academic scheduling involving multiple faculties.

Monkeypox is a rare infectious disease caused by the monkeypox virus, which used to occur endemically in central and western Africa. As of 25th May, a total of 219 recently confirmed cases of monkeypox have been reported from 19 non-endemic countries. This outbreak unusually takes place in non-endemic areas for monkeypox virus and has exhibited features of high risk of human-to-human transmission. Onset of multiple human monkeypox cases may be related to the decreased level of herd cross-immunity after the cessation of smallpox vaccination. Moreover, behavioral patterns in specific populations may account for the human-to-human transmission in this outbreak. Currently, possibility of global epidemic of monkeypox is extremely low, but China should be cautious about risks of importation of monkeypox cases. The key to prevention and control is to establish a surveillance system to identify suspicious cases and close contacts as soon as possible.猴痘是由猴痘病毒感染引起的罕见疾病，主要发生在中非和西非国家。截至2022年5月25日，19个非流行国家及地区报道了219例确诊猴痘病例。本次猴痘疫情不同寻常地发生于非流行地区，且出现较高人际传播性。其暴发原因可能与天花疫苗接种停止后群体猴痘免疫水平降低相关，加之特定人群中行为模式相关的传播可能是本次疫情出现人际传播的原因。目前猴痘全球流行可能性极低，但我国仍有输入风险；建立监测体系，及时识别可疑病例及密切接触者是防控的关键。.

Melatonin is a human neurotransmitter and plant signalling metabolite that perceives and directs plant metabolism. The mechanisms of melatonin action in plants remain undefined. We hypothesized that roots have a melatonin-specific receptor and/or transporter that can respond to melatonin-mediating pharmaceuticals. To test this hypothesis Arabidopsis seedlings were grown with melatonin pharmaceutical receptor agonists: ramelteon and tasimelteon, and/or antagonists: luzindole and 4-P-PDOT. Ramelteon was found both to mimic and competitively inhibit melatonin metabolism in plants. Due to the higher selectivity of ramelteon for the MT1 receptor type in humans, a sequence homology search for MT1 in Arabidopsis identified the rhomboid-like protein 7 (RBL7). In physiological studies, Arabidopsis rbl7 mutants were less responsive to ramelteon and melatonin. Quantum dot visualizations of the effects of ramelteon on melatonin binding to root cell membranes revealed a potential mechanism. We propose that RBL7 is a melatonin-interacting protein that directs root architecture and growth in a mechanism that is responsive to environmental factors.

DELVE DR2 combines new DECam observations with archival DECam data from the Dark Energy Survey, the DECam Legacy Survey, and other DECam community programs. DELVE DR2 consists of ∼160,000 exposures that cover >21,000 deg2 of the high-Galactic-latitude (∣b∣ > 10°) sky in four broadband optical/near-infrared filters (g, r, i, z). DELVE DR2 provides point-source and automatic aperture photometry for ∼2.5 billion astronomical sources with a median 5σ point-source depth of g = 24.3, r = 23.9, i = 23.5, and z = 22.8 mag. A region of ∼17,000 deg2 has been imaged in all four filters, providing four-band photometric measurements for ∼618 million astronomical sources. DELVE DR2 covers more than 4 times the area of the previous DELVE data release and contains roughly 5 times as many astronomical objects. Objectives With the prolonged survival time of AIDS patients, complications of various systems and organs of HIV infection are increasingly prominent. These diseases have become the major factors influencing the quality of life and prognosis of HIV-infected persons, and multidisciplinary cooperation treatment is urgently needed. Methods The Chinese HIV/AIDS Clinical Trial Network has conducted a series of multicentre clinical cohort studies over the past 16 years, in which studies related to people living with HIV systemic complications. Based on the results of previous studies, this review establishes the complications of Chinese people living with HIV after long-term cART. Results HIV's direct damage to human cells, chronic abnormal inflammatory activation after HIV infection, long-term drug side effects caused by cART and persistent reservoirs cause systemic complications in people living with HIV. We summarised the clinical characteristics of the complications of HIV infection in China from the aspects of the liver, cardiovascular, the nervous system, the kidney, bone metabolism, blood glucose, and lipid metabolism. Conclusions The management of the complications of HIV infection is a major link in improving the survival treatment and prognosis of patients in the future. The joint participation of doctors from different departments of general hospitals in the management of comorbidities is the main theme for future improvement of quality of life and prognosis for people living with HIV.

Introduction: Atopic dermatitis (AD) is a type of allergic/inflammatory dermatitis characterized by itch and an impairment in quality of life.Areas covered: Herein, the authors review drug discovery efforts for AD, highlighting the clinical efficacy of novel drugs, with a particular focus on the relief of pruritus. Topical agents include emollients, topical antihistamines, corticosteroids, calcineurin inhibitors and herbs. Recently, topical phosphodiesterase E4 (PDE4) inhibitors like crisaborole have become available and are efficacious for mild to moderate AD with few side effects. For more severe AD, monoclonal antibodies like dupilumab are considered as efficacious subcutaneous treatment options. In severe and recalcitrant AD, systemic treatment can ameliorate AD symptoms.Expert opinion: Many topical and systemic medications have demonstrated therapeutic benefits for AD. Indeed, randomized trials have shown that topical PDE4 inhibitors and subcutaneous dupilumab are safe and efficacious. Objective tools to evaluate itch and gauge treatment efficacy is important, but current methodology relies primarily on clinical scores. AD is a systemic atopic disease with a lot of complicated psychosocial issues. Suboptimal efficacy is often due to poor compliance and unrealistic expectation of curative treatment, rendering treatment difficult despite the existence of effective medications.

The advancement in microfluidics has provided an excellent opportunity for shifting from conventional sub-micron-sized isolation and purification methods to more robust and cost-effective lab-on-chip platforms. The acoustic-driven separation approach applies differential forces acting on target particles, guiding them towards different paths in a label-free and biocompatible manner. The main challenges in designing the acoustofluidic-based isolation platforms are minimizing the reflected radio frequency signal power to achieve the highest acoustic radiation force acting on micro/nano-sized particles and tuning the bandwidth of the acoustic resonator in an acceptable range for efficient size-based binning of particles. Due to the complexity of the physics involved in acoustic-based separations, the current existing lack in performance predictive understanding makes designing these miniature systems iterative and resource-intensive. This study introduces a unique approach for design automation of acoustofluidic devices by integrating the machine learning and multi-objective heuristic optimization approaches. First, a neural network-based prediction platform was developed to predict the resonator’s frequency response according to different geometrical configurations of interdigitated transducers In the next step, the multi-objective optimization approach was executed for extracting the optimum design features for maximum possible device performance according to decision-maker criteria. The results show that the proposed methodology can significantly improve the fine-tuned IDT designs with minimum power loss and maximum working frequency range. The examination of the power loss and bandwidth on the alternation and distribution of the acoustic pressure inside the microfluidic channel was carried out by conducting a 3D finite element-based simulation. The proposed methodology improves the performance of the acoustic transducer by overcoming the constraints related to bandwidth operation, the magnitude of acoustic radiation force on particles, and the distribution of pressure acoustic inside the microchannel.
