The regulation of the Glucocorticoid Receptor (GR) transcript was investigated in the gills of Atlantic salmon (Salmo salar) during the parr-smolt transformation. Sampling of parr and smolt fish was performed between December and July and in particular during the smoltification period occurring in spring. Quantification of GR transcripts revealed differences between the two groups in March and at the beginning of April. During these dates, the amounts of GR mRNA in parr gills were respectively three and two fold lower than those measured in smolts. In order to determine which factors are responsible for these differences, we studied the long-term effects of prolactin and Cortisol treatments on GR transcript in the gills of presmolt fish. The plasma levels of these two hormones respectively drop and rise during smoltification. Contrary to Cortisol long-term treatment which did not modify the amount of gill GR transcript, short-term treatment induced a significant decrease within 12 hours. Prolactin long-term treatment caused a significant increase of GR transcript abundance after 13 days of implant treatment. This result is unexpected with regard to those obtained in the smoltification analysis but is in agreement with previous studies performed in mammary gland revealing a positive control of PRL on GR in epithelial cells. Our data suggest that the regulation of the GR transcript during the parr-smolt transformation probably involves several hormonal factors.

The distribution of the glucocorticoid receptor (GR)within the forebrain of rainbow trout has been studied by means of in situ hybridization (ISH) and immunohistochemistry (ICC) using probes and antibodies directed against the AB-domain of the rainbow trout GR (rtGR). Both rtGR mRNA and protein exhibited the same pattern of distribution. A strong labelling was observed in all subdivisions of the preoptic nucleus, in the thalamic region and in the mediobasal hypothalamus (nucleus recessus lateralis, nucleus lateralis tuberis).It was notably found that all magnocellular neurons of the nucleus preopticus pars magnocellularis (NPOmc) were positive with both ISH and ICC. Furthermore, numerous cells of the anterior lobe of the pituitary were positively stained. Double staining studies allowed to identify part of the rtGR expressing cells. At the telencephalic level, it was found that GnRH neurons frequently express rtGR in vitellogenic females. Virtually all tyrosine hydroxylase (TH) cells in the anteroventral part of the preoptic nucleus (NPOav) were rtGR-positive. In this same area, a colocalization between rainbow trout estradiol receptor (rtER) and TH has already been documented, indirectly demonstrating that rtER and rtGR are colocalized. The same holds true for the GTH-II cells of the anterior pituitary which express both types of receptors. These results provide information on the potential sites of cortisol action on the reproductive axis directly at the pituitary level or indirectly on neuroendocrine Systems controllingGTH-II secretion, possibly via GR-ER interactions.

During the past decade there has been a substantial advance in our understanding of estrogen signaling both from a clinical as well as a preclinical perspective. Estrogen signaling is a balance between two opposing forces in the form of two distinct receptors (ER alpha and ER beta) and their splice variants. The prospect that these two pathways can be selectively stimulated or inhibited with subtype-selective drugs constitutes new and promising therapeutic opportunities in clinical areas as diverse as hormone replacement, autoimmune diseases, prostate and breast cancer, and depression. Molecular biological, biochemical, and structural studies have generated information which is invaluable for the development of more selective and effective ER ligands. We have also become aware that ERs do not function by themselves but require a number of coregulatory proteins whose cell-specific expression explains some of the distinct cellular actions of estrogen. Estrogen is an important morphogen, and many of its proliferative effects on the epithelial compartment of glands are mediated by growth factors secreted from the stromal compartment. Thus understanding the cross-talk between growth factor and estrogen signaling is essential for understanding both normal and malignant growth. In this review we focus on several of the interesting recent discoveries concerning estrogen receptors, on estrogen as a morphogen, and on the molecular mechanisms of anti-estrogen signaling.

Estrogens, by binding to and activating two estrogen receptors (ERalpha and ERbeta), are critically involved in the development of the mammary gland and breast cancer. An isoform of ERbeta, ERbeta2 (also called ERbetacx), with an altered COOH-terminal region, is coexpressed with ERalpha in many human breast cancers. In this study, we generated a stable cell line from MCF7 breast cancer cells expressing an inducible version of ERbeta2, along with endogenous ERalpha, and examined the effects of ERbeta2 on the ERalpha protein levels and function. We showed that ERbeta2 inhibited ERalpha-mediated transactivation via estrogen response element and activator protein-1 sites of reporter constructs as well as the endogenous genes pS2 and MMP-1. Chromatin immunoprecipitation assays revealed that ERbeta2 expression caused a significant reduction in the recruitment of ERalpha to both the pS2 and MMP-1 promoters. Furthermore, ERbeta2 expression induced proteasome-dependent degradation of ERalpha. The inhibitory effects of ERbeta2 on ERalpha activity were further confirmed in HEK293 cells that lack functional endogenous ERs. We also showed that ERbeta2 can interact with ERalpha both in vitro and in mammalian cells, which is compatible with a model where ERbeta2/ERalpha heterodimers are targeted to the proteasome. Finally, in human breast cancer samples, we observed that expression of ERbeta2 significantly correlated with ERalpha-negative phenotype. Our data suggest that ERbeta2 could influence ERalpha-mediated effects relevant for breast cancer development, including hormone responsiveness.

In this study, an estrogen receptor (ER) alpha-expressing T47D cell line containing an inducible tet-off FLAG-ERbeta was used to examine the influence of ERbeta on ERalpha activity. Real-time PCR analysis of mRNA levels of two well-studied estrogen-responsive genes, pS2 and progesterone receptor (PR), showed that the expression levels of both genes were reduced in the presence of ERbeta. Chromatin immunoprecipitation assays showed that the 17beta-estradiol (E2)-induced recruitment patterns to the pS2 and PR promoters were similar for both ERalpha and ERbeta. ERbeta expression did not significantly influence the kinetic recruitment profile of ERalpha to the pS2 promoter, but it was evident that ERalpha occupancy at the PR promoter was reduced. The E2-induced recruitment of c-Fos to a 12-O-tetradecanoylphorbol-13-acetate response element site in the PR promoter was significantly reduced in the presence of ERbeta, whereas only a slight reduction in the recruitment of c-Fos to the pS2 promoter was observed. ERbeta expression resulted in a significant reduction in the E2-induced expression of c-Fos mRNA. The recruitment pattern of c-Jun was also altered by ERbeta, although the expression levels of c-Jun were not. Expression of ERbeta caused a further 30-50% decrease of the E2-induced reduction in ERalpha protein after 3 h of E2 treatment, showing that ERbeta influences ERalpha protein levels. The altered recruitment of the activating protein-1 complex, combined with the reduction in ERalpha protein levels, may partly explain the antagonistic effect of ERbeta on ERalpha-mediated transcription.

Steroid receptors are transcription factors that regulate hormone-responsive genes and whose activity is controlled by their interaction with numerous other proteins. Observations reported here reveal that estrogen receptors alpha and beta (ERalpha and ERbeta), androgen receptor, and glucocorticoid receptor bind in vitro to vinexin alpha, a multiple SH3 motif-containing protein associated with the cytoskeleton. The SH3 domains are not involved in this interaction. Furthermore, we demonstrate that vinexin alpha stimulates the ligand-induced transactivation function of these receptors, although it is devoid of intrinsic transcriptional activity when tethered to DNA. In addition, the ectopic coexpression of vinexin alpha and ERalpha results in a loss of ERalpha phosphorylation on serines and the partial redistribution of vinexin alpha into the nucleus, where it colocalizes with ERalpha. These results establish a new model of transcriptional regulation where components of the cell-cell and cell-substrate adhesion complexes can regulate the phosphorylation and activity of steroid receptors.

Our appreciation of the physiological functions of estrogens and the mechanisms through which estrogens bring about these functions has changed during the past decade. Just as transgenic mice were produced in which estrogen receptors had been inactivated and we thought that we were about to understand the role of estrogen receptors in physiology and pathology, it was found that there was not one but two distinct and functional estrogen receptors, now called ER alpha and ER beta. Transgenic mice in which each of the receptors or both the receptors are inactive have revealed a much broader role for estrogens in the body than was previously thought. This decade also saw the description of a male patient who had no functional ER alpha and whose continued bone growth clearly revealed an important function of estrogen in men. The importance of estrogen in both males and females was also demonstrated in the laboratory in transgenic mice in which the aromatase gene was inactivated. Finally, crystal structures of the estrogen receptors with agonists and antagonists have revealed much about how ligand binding influences receptor conformation and how this conformation influences interaction of the receptor with coactivators or corepressors and hence determines cellular response to ligands.

Human estrogen receptors alpha (ERalpha) and beta (ERbeta) are ligand-inducible transcription factors that are highly homologous in their central DNA-binding and carboxyl-terminal ligand-binding domains. In contrast, there is very little conservation between ERalpha and ERbeta in the amino-terminal domain. Using different human cell lines, we show that wild-type ERbeta transcriptional activity is lower or similar to that of ERalpha, depending on the cell type. Deletion of the amino-terminal domain in both ER subtypes resulted in no or a lower decrease of transcriptional activity of ERbeta compared with ERalpha, suggesting that the ERbeta amino-terminal domain contains a weaker transcriptional activation function-1. Using ERalpha and ERbeta deletion mutants, we showed that the amino-terminal transcriptional activity of ERbeta maps to amino acids 1-31. Interestingly, this domain contains a six amino-acid motif (amino acids 5-10 in human ERbeta) that is part of the ERalpha-activation function-1 region (amino acids 49-54 in human ERalpha) and highly conserved among all mammalian ERalpha amino-terminal domains. Despite this similarity between the two ER subtypes, no autonomous and ligand-independent activity of the ERbeta-amino-terminal domain was observed in yeast and mammalian cells in contrast to ERalpha. This study provides a molecular basis for the difference in transcriptional activity between ERalpha and ERbeta and establishes that ERbeta contains a structurally and functionally restricted amino-terminal transcriptional activity.

The full length cDNA encoding a rainbow trout glucocorticoid receptor (rtGR) has been obtained from rainbow trout liver and intestine libraries. Northern blot analysis showed that the corresponding messengers are detected in the brain of trout with a size 7.5 kb similar to the size of rtGR mRNA in other target tissues. The distribution of the rtGR mRNA and protein was studied in the forebrain of the trout by means of both in situ hybridization and immunohistochemistry and compared with that of the oestrogen receptor (rtER). The GR and ER mRNAs and proteins were detected with a strong overlapping mainly in the: (a) preoptic region; (b) mediobasal hypothalamus; and (c) anterior pituitary, confirming their implication in the neuroendocrine control of pituitary functions. In both diencephalon and pituitary, the peptidergic phenotype of some neuron or cell categories expressing either type of receptors could be determined by double staining. Furthermore, double staining studies have demonstrated colocalization of the two receptors in the same neurons or pituitary cells. The rtER and rtGR were found to be co-expressed in the dopaminergic neurons inhibiting GTH2 secretion and in pituitary cells of the anterior lobe--notably the gonadotrophs. Given that the promoter of the ER gene contains several potential glucocorticoid-responsive elements (GRE) and that cortisol inhibits the oestradiol-stimulated ER expression in the liver, the possibility exists for modulation of ER gene expression by GR in the hypothalamo-pituitary complex. This could explain some of the well documented effects of stress on the reproductive performance in salmonids.

Fragments of cDNA that encode the N-terminal and DNA-binding domains (DBD) of the rainbow trout glucocorticoid receptor (rtGR) were expressed in Escherichia coli as fusion proteins with glutathione-S-transferase (GST). The fusion proteins induced by IPTG could readily be detected as 45- and 40-kDa bands, respectively, in crude extracts, as well as in proteins purified on glutathione-agarose. These purified hybrid proteins were used to immunize rabbits. The antisera produced were tested for specificity by Western blot analysis using extracts from COS-1 cells transfected with an rtGR expression vector and from trout liver cells. The antisera raised against the DBD domain did not detect any bands on Western blots, even at low antiserum dilution. However, the purified DBD fusion protein specifically bound GRE-containing DNA fragments in gel-shift assays, and the retarded complexes were supershifted by these antibodies. The antisera raised against the N-terminal domain consistently detected two protein bands at 104 and 100 kDa in the two cell extracts and allowed specific immunohistochemical staining in fish brain and pituitary. For the first time in fish, these antibodies will allow analysis of GR expression in different cortisol target tissues.

The expression and distribution of glucocorticoid receptor mRNA was studied in the forebrain of mature female and immature undifferentiated rainbow trout (Oncorhynchus mykiss) by means of Northern blotting and in situ hybridization. A single mRNA species of 7.5 kb was detected in mRNA polyA+ prepared from the anterior brain. In situ hybridization was carried out using a 35S-labelled riboprobe corresponding to the A/B-domain (between nucleotides 1224 and 1763) of the recently cloned rainbow trout glucocorticoid receptor cDNA. Comparison of adjacent sections hybridized with the sense and antisense probes allowed detection of a specific signal with a similar distribution pattern in all animals studied. In the telencephalon, a specific hybridization was detected in scattered cells of the dorsal telencephalic hemisphere, but the stronger signal was consistently observed in the dorsal nucleus, and to a lesser degree in the ventral nucleus of the ventral telencephalon. Heavy hybridization staining was consistently observed in all subdivisions of the preoptic nucleus and the nucleus lateralis tuberis, which are the main hypophysiotrophic regions in fish. A weaker signal was detected in the nucleus anterioris periventricularis, nucleus suprachiasmaticus and thalamic region. The presence of a strong signal in virtually all magnocellular neurons of the nucleus preopticus, known for producing vasotocin, isotocin and corticotropin-releasing factor favors a direct function of cortisol in regulating its own secretion.

The molecular basis for the initial specification of dorsoventral polarity in the Amphibian egg prior to the mid-blastula transition still remains an open question. Regional differences in the protein pattern of Pleurodeles egg were investigated during early cleavage (8- and 512-cell stages, prior to the mid-blastula transition). Animal-dorsal, animal-ventral, vegetal-dorsal and vegetal-ventral quarters were separated and proteins were analyzed by 2D-electrophoresis. The comparison of acidic protein patterns from dorsal and ventral quarters revealed differences between vegetal cells but no difference was detected between animal cells. One protein (p11, 30 kDa) was characterized in the dorsal side as early as the 8-cell st. and two dorsal spots were detected at the 512-cell st. (p11 and p5, 65 kDa). Similarly one protein (p7b, 46 kDa) appears to be ventral-specific from the 8-cell st. The p11 spot was shown to appear in ventral cells as a consequence of a dorsalizing LiCl-treatment at the 32-cell stage. Conversely, p11 disappeared from dorsal cells and p5 did not appear at 512-cell stage after UV-irradiation of the uncleaved egg, which results in the expression of the ventral-specific protein p7b in the dorsal part of the egg. Therefore differential protein expression is already present at very early cleavage stages. Its significance needs further investigation.
