Abstract

Epub ahead of print; Nowadays, high-resolution mass spectrometry is widely used for metabolomic studies. Thanks to its high sensitivity, it enables the detection of a large range of metabolites. In metabolomics, the continuous quest for a metabolite identification as complete and accurate as possible has led during the last decade to an ever increasing development of public MS databases (including LC-MS data) concomitantly with bioinformatic tool expansion. To facilitate the annotation process of MS profiles obtained from biological samples, but also to ease data sharing, exchange and exploitation, the standardization and harmonization of the way to describe and annotate mass spectra seemed crucial to us. Indeed, under electrospray (ESI) conditions, a single metabolite does not produce a unique ion corresponding to its protonated or deprotonated form but could lead to a complex mixture of signals. These MS signals result from the existence of different natural isotopologues of the same compound and also to the potential formation of adduct ions, homo and hetero-multimeric ions, fragment ions resulting from "prompt" in-source dissociations. As a joint reflection process within the French Infrastructure for Metabolomics and Fluxomics (MetaboHUB) and with the purpose of developing a robust and exchangeable annotated MS database made from pure reference compounds (chemical standards) analysis, it appeared to us that giving the metabolomics community some clues to standardize and unambiguously annotate each MS feature, was a prerequisite to data entry and further efficient querying of the database. The use of a harmonized notation is also mandatory for inter-laboratory MS data exchange. Additionally, thorough description of the variety of MS signals arising from the analysis of a unique metabolite might provide greater confidence on its annotation.

Abstract

The coexistence of two gas-phase structures of CHS2(super -) produced in a chemical ionization source and their regioselective reactions with CS2 has been reported. The results showed that CS2 gives its radical anion by electron capture and CHS2(super -), by hydride attachment, in a chemical ionization source with CH4/N2O. Mass spectrum analysis and ab initio calculations showed that hydride attachments can produce either thioformate anion or thiohydroxythiocarbonyl carbanion without interconversion.

Abstract

Characterization of synthetic polymers by Matrix assisted laser desorption (MALDI) is limited by the solubility of different oligomers in a suitable solvent, and the finger-print of the mass spectrum is affected by the properties of solvents employed (eg., pH, secondary solvents, evaporation) during sample target preparation. If solvents are not used during sample target preparation, then solvent properties should not play an important role in determining the quality of the MALDI mass spectrum. We report here two solventless approaches for sample target preparation. It was observed that Poly(ethylene glycol) 6000 (PEG) showed the same molecular mass distribution in different modes of sample target preparation. Fluorinated polymer used in these studies was affected by sample target preparation protocol and by target surface. Pyrolysis of PEG oligomers was observed in all the methods of sample target preparation. The desorbed high mass neutral oligomers fragment to give small oligomers which are then cationized by the desolvation of the cationized matrix clusters. Moreover, the origin of the matrix clusters (i.e., formed in the condensed phase or in the gas phase) determined the relative intensities of PEG oligomers cationized by sodium potassium.

Abstract

Negative ion chemical ionization was carried out using a quadrupole ion trap mass spectrometer with selected reactant negative ions, primarily injected from a homemade dual EI/CI external ion source. Hence, selective ion/molecule reactions were provided according to the reaction time, which induce a greater control over bimolecular ionization mechanisms than in conventional a high-pressure ion source combined with beam instruments, where several competitive ionization processes take place mainly due to source conditions (e.g., temperature, pressure, and repeller). By selecting the reactant ions, ion/molecule reactions were specifically produced (i.e., charge exchange, proton transfer, nucleophilic substitution, and/or [Alpha]-[Beta] elimination) with several organic target compounds. Gas-phase reactivity of phosphorus-and nitrogen-containing compounds (such as phosphonates as representative for chemical warfare agents and phosphorothionates, phosphorodithionates, and triazines for pesticides) as well as dinitro aromatic compounds (for pesticides) has been explored, in the present work, to ensure further unambiguous detection.

Abstract

A new combination of a dual EI/CI ion source with a quadrupole ion trap mass spectrometer has been realized in order to efficiently produce negative ions in the reaction cell. Analysis of volatile compounds was performed under negative ion chemical ionization (NICI) during a reaction period where selected reactant negative ions, previously produced in the external ion source, were allowed to interact with molecules, introduced by hyphenated techniques such as gas chromatography. The [MATHEMATICAL EXPRESSION NOT REPRODUCIBLE IN ASCII], [CH.sub.3][O.sup.-], and [Cl.sup.-] reactant ions were used in this study to ensure specific ion/molecule interactions such as proton transfer, nucleophih'c displacement, or charge exchange processes, respectively leading to even-electron species, i.e., deprotonated [[M - H].sup.-] molecules, diagnostic [[M - R].sup.-] ions, or odd-electron [[M.sup.-][multiplied by]] molecular species. The reaction orientation depends on the thermochemistry of reactions within kinetic controls. First analytical results are presented here for the trace-level detection of several contaminants under NICI/[Cl.sup.-] conditions. Phosphoruscontaining compounds (malathion, ethyl parathion, and methyl parathion as representative for pesticides) and nitro-containing compounds (2,4,6-trinitrotoluene for explosive material) have been chosen in order to explore the analytical ability of this promising instrumental coupling.

Abstract

Proteomic approach in combination with mass spectrometry demonstrates a great potential for identification of proteinaceous materials in works of art. In this study we used a linear trap quadrupole Orbitrap (LTQ-Orbitraph a state-of-the-art mass spectrometer for parts per million accuracy analyses of peptides behind tryptic hydrolysis. After the efficiency of the proteomic method was confirmed for reference and model samples, micro-samples from historical paintings were for the first time analysed using this technique. Superior performances of the liquid chromatography-mass spectrometry approach using a LTQ-Orbitrap mass spectrometer allowed identification of egg yolk peptides in two samples from nineteenth-century Orthodox icons, indicating egg tempera as the painting technique. Accurate precursor ion masses, in the range of 2 ppm, and retention times of tryptic peptides strengthen protein identification. Additionally, in all historical samples the presence of animal glues suggested that the ground layer was likely bound using bovine collagen. Comparing to results acquired using matrix-assisted laser desorption/ionization time-of-flight tandem mass spectrometry in our previous study, here we achieved higher ion scores and protein scores, better sequence coverage and more identified proteins. In fact, a combination of the two mass spectrometric techniques provided overlapping and complementary data, related to the detection of peptides with different physicochemical properties.

Abstract

Ephrin B2, one of the ligand of the EphB receptors, is involved in a complex signaling pathway regulating the development of the nervous system, neuronal migration, erythropoiesis and vasculogenesis. We report a patient with a de novo variant in EFNB2 and a family in which segregates a 610‐kb deletion at chromosome 13q33 encompassing only ARGLU1 and EFNB2 genes. The de novo variant was observed in a patient with anal stenosis, hypoplastic left ventricle and mild developmental delay. The deletion was identified in 2 sibs with congenital heart defect and mild developmental delay. One of the affected sibs further had myoclonic epilepsy and bilateral sensorineural hearing loss. The carrier mother was apparently asymptomatic. Because EFNB2 is located in the subtelomeric region of 13q chromosome, we reviewed the previous reports of terminal 13q deletion. We suggest that haploinsufficiency of the EFNB2 could be at the origin of several clinical features reported in 13qter deletions, including intellectual disability, seizures, congenital heart defects, anorectal malformation and hearing loss. Abstract

Oral‐facial‐digital (OFD) syndromes are a subgroup of ciliopathies distinguished by the co‐occurrence of hamartomas and/or multiple frenula of the oral region and digital anomalies. Several clinical forms of OFD syndromes are distinguished by their associated anomalies and/or inheritance patterns, and at least 20 genetic types of OFD syndromes have been delineated. We describe here a child with preaxial and postaxial polydactyly, lingual hamartoma, a congenital heart defect, delayed development and cerebellar peduncles displaying the molar tooth sign. Whole‐exome sequencing and SNP array identified compound heterozygous variants in the INTU gene, which encodes a protein involved in the positioning of the ciliary basal body. INTU is a subunit of the CPLANE multiprotein complex essential for the assembly of IFT‐A particles and intraflagellar transport. This report of a second patient with INTU‐related OFD syndrome and the further delineation of its neuroimaging and skeletal phenotype now allow INTU‐related OFD syndromes to be classified within the OFD syndrome type VI group. Patients display a phenotype similar to that of mice with a hypomorphic mutation of Intu, but with the addition of a heart defect. [ABSTRACT FROM AUTHOR]

Abstract

Covalent peptide-nucleotide conjugates are an original class of compounds that play a role in different steps of protein biosynthesis. The structural analysis of such compounds by electrospray mass spectrometry (ESI-MS) is not usual and their fragmentation mechanisms are not well known. Microcin C7-C51 (MccC7-C51) is an antimicrobial peptide that targets the aspartyl-tRNA synthetase and inhibits translation. Its structure consists of an N-formylated heptapeptide MRTGNAD with the C-terminus covalently bound to an n-aminopropanol-substituted adenosine monophosphate group. Characterization of MccC7-C51 by ESI-MS combined with collision-induced dissociation experiments is reported. The fragmentation patterns of the positively and negatively charged microcin were compared to the corresponding unconjugated heptapeptide, and provide a comprehensive approach to analyze the structure of peptide-nucleotide conjugates.
