13. Peroxide removal from organic solvents and vegetable oils.

Abstract

Peroxide contamination of organic solvents and related reagents, e.g. vegetable oils, has created potential problems for the toxicologist, biochemist and industrial hygienist. Because of tedious or complicated procedures to test for and remove, or prevent the formation of peroxides, the tendency of laboratory investigators has been to ignore the problem. In this study, straight‐forward procedures for the testing and removal, and/or prevention of formation of peroxides were investigated. Total peroxides were assayed by measuring FeIII(SCN)3 formed by FeII oxidation and reaction with KSCN. Most solvents and reagents, as received from the manufacturer, were found to have low levels of peroxides. After opening the container, peroxides were formed rapidly in many solvents. Techniques for the complete retardation of peroxide formation in fresh solvents or reagents are described. For peroxides already present in organic solvents or vegetable oils, a novel technique for easy removal without Introducing contaminants 1s described. [ABSTRACT FROM PUBLISHER]

Abstract

Previous studies in guinea pigs showed that DTDM, MMBT and MBT were sensitizing agents. The purpose of this study was to investigate the biochemical basis of this sensitization reaction. The reactions of representative amino acids, Cys, Lys & Gly, with morpholine, DTDM, MMBT & MBT were examined. New compounds were found between MMBT and all 3 amino acids. Additional new compounds were seen for Cys/MBT, Cys/DTDM, Lys/MBT & Gly/DTDM. No reaction was observed with morpholine. The new compounds were examined spectroscopically. Results suggest that the sensitizing nature of DTDM is probably a reaction of its disulfide bond with the -SH & -NH[SUB2] groups of amino acid moieties. The conjugation of Cys/MMBT may be via the the same reaction mechanism. The -SH group of MBT may react either via oxidation and/or thioester formation with the carboxylate group of amino acids. These results also suggest that in vitro tests may predict the sensitizing potential of certain compounds. [ABSTRACT FROM AUTHOR]

17. Exogenous Iron Overload in Perinatal Hemochromatosis: A Case Report

Abstract

We report an infant with fatal neonatal liver disease in whom efforts to correct an associated unremitting anemia resulted in massive exogenous iron overload that was expressed as perinatal hemochromatosis (PH). Levels of iron and copper were elevated in multiple tissues. Echovirus subtype 9, recovered from the urine at age 3 weeks, may have been etiologic in the liver failure. PH is best viewed as a definable phenotype with an undefined genetic and/or environmental basis that emerges only in the context of severe perinatal liver disease. The absence of hemosiderin in splenic and bone marrow reticuloendothelial (RE) cells of our patient suggests an important role for RE cell dysfunction.

Abstract

Blood levels, excretion and tissue distribution of WR-158,122 14C were studied in 10 bile duct ligated rats, 10 bile duct cannulated rats, 8 control rats and in 2 control and one bile duct cannulated monkeys. In 72 hours bile duct ligated rats excreted about 16.3 + or - 3.8% as 14C in the urine and the remaining 14C was recovered primarily in the feces. About 2% was recovered in the carcass and tissues at necropsy. Bile duct cannulated rats excreted about 10.9 + or - 6.1% in the bile, 2.3 + or - 1.3% in the urine and the remainder in the feces. Only about 1.3 + or - 1.0% was recovered in the carcass and tissues. Control rats excreted the compound rather rapidly; 1.5 + or - 0.8% of the 14C was recovered in the urine, 0.3 + or - 0.2 in the carcass and other tissues and the remainder in the feces. Total average recovery (as 14C) in the three groups of rats varied from 94 to 100% of the dose. Two experiments were conducted in a single bile duct cannulated monkey. In the first the blood and plasma levels were highest at two hours after dosing. Plasma levels routinely exceeded levels in whole blood indicating little sequestration of WR-158,122 (as 14C) in the red cells. Both levels were low at 24 hours and below detection at 48 hours. In this bile duct cannulated monkey biliary excretion over a 96 h. period accounted for 0.6 to 0.9% of the dose (as 14C), urinary excretion for 3.1% and 87 to 95% of the dose was recovered in the feces. In two control monkeys urinary excretion over a 96 hour period amounted to 14 and 18%; 70% of the dose (as 14C) was recovered in the feces. Excretion was essentially complete after 96 hours. In both rats and monkeys WR-158,122 appears to be rapidly but incompletely absorbed., Revision of report dated 12 Feb 81.
