Abstract

Investigating the age at sea departure of returning Atlantic salmon is critical to better understand the role of emigration timing in survival. Among the methods available, the use of otolith chemistry can be challenging in anadromous fish, as the elements frequently used (i.e., magnesium, manganese, and zinc) can jointly reflect individual metabolism and environmental conditions. Here, we present a new unsupervised and robust method for successfully estimating the outmigration age of Atlantic salmon in the Bay of Biscay, based on otolith zinc concentrations. Abstract

Although otoliths are widely used as archives to infer life‐history traits and habitat use in fishes, their biomineralization process remains poorly understood. This lack of knowledge is problematic as it can lead to misinterpretation of the different types of signals (e.g., optical or chemical) that provide basic data for research in fish ecology, fisheries management, and species conservation. Otolith calcification relies on a complex system involving a pericrystalline fluid, the endolymph, whose organic and inorganic compositions are spatially heterogeneous for some constituents. This property stems from the particular structure of the calcifying saccular epithelium. In this study, we explored the spatial heterogeneity of elemental incorporation in otoliths for two species of high economic interest, European hake Merluccius merluccius (L. 1758) and European sea bass Dicentrarchus labrax (L. 1758). Two‐dimensional mappings of chemical elements were obtained using UV high‐repetition‐rate femtosecond laser ablation (fs‐LA) system coupled to a high‐resolution inductively coupled plasma sector field mass spectrometer analyses on transverse sections of sagittae. Results highlighted a clear asymmetry between proximal (sulcus) and distal (antisulcus) concentrations for elements such as magnesium (Mg), phosphorus (P), manganese (Mn), and potassium (K) with concentration gradient directions that varied depending on the element. Strontium (Sr) and barium (Ba) did not show a proximo‐distal gradient. These results are discussed in light of current knowledge on the endolymph composition and the mechanisms that lead to its compartmentalization, highlighting the need for further research on otolith biomineralization. Operational implications for studies based on otolith chemical composition are also discussed with emphasis on advice for sampling strategies to avoid analytical biases and the need for in‐depth analyses of analytical settings before comparing otolith signatures between species or geographical areas. Sur l’échantillon total de brochets adultes analysés (n=40, dont 17 individus prélevés en 2018-2019), 25% (IC95% 14.2-40.2%) des individus proviennent de la pisciculture et 75% (IC95% 59.8-85.8%) de la reproduction naturelle. Bien qu’une incertitude demeure en raison de la taille de l’échantillon, la cohérence des récents résultats avec les analyses réalisées en 2015-2016 (Richard et al., 2018) confirme la robustesse de ces proportions et la part majoritaire de la reproduction naturelle dans le stock de brochet du lac de Serre-Ponçon. Ce résultat questionne sur les sites et les périodes de reproduction dans le lac de Serre-Ponçon au vu de l’important marnage saisonnier.

Abstract

Dans la perspective de mieux comprendre le rôle de la dispersion dans la conservation des populations de saumon Atlantique face aux pressions environnementales et anthropiques qu’ils subissent, il est vital de quantifier la part relativedes migrants des populations. Pour répondre à cet enjeu, ce projet a eu pour objectif d’évaluer le potentiel de la microchimie des écailles comme outil non-léthal permettant de déterminer l’origine spatiale (rivière natale) des individus. Une telle avancée doit permettre un retour historique sur les larges banques de données scalimétriques existantes alors que les banques d'otolithes sont beaucoup plus restreintes, tant spatialement que temporellement.Durant ce projet, nous avons évalué le potentiel de l’analyse microchimique élémentaire (Sr, Ba, Mn, Mg, Zn, …) des écailles de saumon Atlantique (Salmo salar) pour déterminer l’origine spatiale des individus. Pour ce faire, nous effectuerons une analysecomparative des éléments microchimiques entre otolithes (méthode de référence) et écailles dans le but d’évaluer 1) l’importance des effets de l’altération des signatures géochimiques contenues dans les écailles lors du séjour en mer (contrairement à l’otolithe dans lequel les signatures sont inertes, celles des écailles pourraient subir des modifications dans le temps), et 2) la pertinence de l’écaille pour déterminer l’origine spatiale des individus. Abstract

International audience; The specific stock composition and dispersion of anadromous fish species aggregations in the marine environment are poorly known, while they can play a major role in the metapopulation dynamics. Otolith microchemistry has proven to be a powerful tool to address natal origins of anadromous fish. We used archived otolith microchemistry to investigate the population-specific composition of European shads (Alosa alosa and Alosa fallax) subadults in the ocean during the 80’s. The allocation of natal origin was addressed relying on contemporary water and juveniles signatures within a Bayesian model. A great discrimination of natal origin was obtained at the Biscay Gulf scale. However, the discrimination of 80’s natal origin for the southern rivers with similar geology based on 2013 water and juveniles baselines was doubtful. Our results showed that the most abundant southern populations were dominant, suggesting that population-specific composition was related to population relative abundance. The dispersion in the marine environment was plastic; alternatively shads were found large distances away from their natal rivers, while others remained in the vicinity of their natal river plume.

Abstract

Phytoplankton dynamics in coastal ecosystems is increasingly altered by land‐based human activities. Yet, this global vision conceals major disparities, among sites and through time. As conventional monitoring time series are quite sparse and relatively short, biological records of environmental variability appear as relevant tools to gain insights into phytoplankton dynamics over larger temporal and spatial scales. Here, we present results of an interdisciplinary project dealing with chemical information archived in shells of Pecten maximus (Bivalvia; Pectinidae), known to form daily growth striae on its shell surface. Several individuals were collected in the Bay of Brest (France) in 2011 and 2012, and analyzed for the molybdenum and lithium concentrations in their soft tissues and the element‐to‐calcium ratios (Mo : Ca and Li : Ca) in their calcitic striae. All shells revealed high synchrony and reproducibility in their Mo : Ca and Li : Ca profiles, characterized with a major peak at the end of May and in mid‐June 2011, respectively. Detailed analysis of physical, chemical, and biological variables measured in seawater during an extensive 9‐month environmental survey enabled a meticulous description of phytoplankton dynamics in 2011 and its impact on shell geochemistry. Main findings strongly suggest that (1) the timing of Mo : Ca peaks reflects the occurrence of silicon limitation and diatom aggregation periods, (2) the height of these peaks relates to the amplitude of the first spring diatom bloom, and (3) Li : Ca serves as a proxy for the temporal dynamics of diatom biovolume and of biogenic silica recycling at the sediment–water interface. Abstract

All native fishes living in the Caribbean island rivers are considered diadromous but few species have been validated as diadromous. In Guadeloupe (Lesser Antilles) as well as in the other tropical islands, the riverine habitats are subject to strong anthropogenic pressure due to resource modifications or dams. The management of fish biodiversity requires knowledge concerning their life history traits and behaviour that are still unknown for most species at a local scale and at the whole distribution area of the species. In a Guadeloupe river, we inferred the migratory patterns of four freshwater fishes common to Caribbean rivers by the analysis of otolith microchemistry. We found two species Eleotris perniger (Eleotridae) and Sicydium plumieri (Gobiidae) to be completely amphidromous with a marine larval phase and a freshwater adult phase. Agonostomus monticola (Mugilidae) was also found to be amphidromous in this system. The fourth species Gobiesox cephalus (Gobiesocidae) showed elemental ratios suggesting an estuarine larval phase and was defined as "limited amphidromous". The identification of such life history traits, with limited dispersion abilities, argues for specific management measures in tropical island rivers at a basin scale for the conservation of biodiversity. Azkenik, teknika honen egungo egoeraz jabetzeko, jatorri askotariko laginen analisi elementala laburbildu eta berrikusten da biologian eta ingurumenean, osasunean, geokimikan zein auzitegi zientzien alorretan.; Since the early 80s, laser-ablation inductively-coupled-plasma mass spectrometry (LA-ICPMS) has been widely explored and nowadays is considered to be one of the most versatile analytical techniques for the direct trace elemental and isotopic analyses of solid samples. The most remarkable features are ease of use, fast sample throughput, limited sample damages, high sensitivity and wide dynamic range which allows the simultaneous acquisition of major and trace elements, as well as the measurement of isotope ratios. Throughout this manuscript, the principles and general setup of LA-ICPMS will be extensively explained and the performance of femtosecondLA in comparison to nanosecond-LA discussed. In order to show the state of the art in this field a variety of examples for elemental analysis of solid samples in biological/environmental science, geochemistry and forensic science applications will be presented.

32. Larval traits of the Caribbean amphidromous goby Sicydium punctatum ( Gobioidei: Sicydiinae) in Guadeloupe.

Abstract

Amphidromous fish are the biggest contributor to the diversity of fish communities in river systems of Caribbean islands. Among them, Sicydium punctatum Perugia, 1896, which is endemic to the West Indies, represents the vast majority of fish in these rivers. The otolith microstructure and the biometry of S. punctatum postlarvae from Guadeloupe were investigated to explore the dispersal modalities of this species through an appreciation of the growth pattern, the pelagic larval duration ( PLD) and the size-at-recruitment. The study was made on one cohort of 83 recruited postlarvae, fished at the Capesterre River's mouth on 2 November 2011. The mean (± SD) size-at-recruitment of the postlarvae was 24.6 ± 1.3 mm (range of 20.5-28.1 mm, n = 83). We found a mean (± SD) PLD of 72.2 ± 10.5 days (range of 54-101.5 days, n = 67). The growth rate estimated from the otolith increments showed a globally decreasing pattern during the marine larval phase. Growth rates at the beginning of the larval stage were significantly different between hatching periods, suggesting a relation between the hatching period and the growth rate in S. punctatum. This study adds on to the general understanding of the life cycle of S. punctatum in Guadeloupe that will help implement strategies to manage amphidromous fish populations in the Caribbean region. Abstract

Eleotris fusca is a widespread amphidromous fish found in the Indo-Pacific area. Its life cycle is characterised by a marine pelagic larval phase allowing the species to disperse in the ocean and then to recruit to remote island rivers. In order to examine its migratory history and pelagic larval duration (PLD), otolith microchemical and microstructural analyses were undertaken on 100 specimens collected in the rivers of four areas: Mayotte, Reunion Island (Indian Ocean), Vanuatu and French Polynesia (Pacific Ocean). Variations of Sr:Ca ratios were observed along the otoliths using a femtosecond laser ablation-inductively coupled plasma spectrometer (fs-LA-ICP-MS). The variation of the Sr:Ca ratio allowed us to reconstruct fish migratory movements between sea water (high Sr:Ca) and freshwater (low Sr:Ca), and led us to assume that Eleotris fusca is an amphidromous species. Using microstructural analysis, we observed that the PLD depended on the sampling site. In the Pacific Ocean, the PLD is similar in Vanuatu (mean ± SD: 90 ± 19.2 days) and French Polynesia (96.2 ± 17.5 days) whereas a significant difference emerged in the Indian Ocean between Mayotte (63.3 ± 11 days) and Reunion Island (114.8 ± 21.8 days). Abstract

A methodology to simultaneously determine mercury (MeHg, IHg) and butyltin (TBT, DBT, MBT) compounds in eel samples was assessed and validated using multiple isotopically enriched species. The developed methodology was able to analyse simultaneously the organometal species accurately and precisely and to correct for the potential transformations/degradations of the different species during the various steps of the analytical procedure. Low detection limits were achieved (0.007–0.17 µg Hg kg−1for mercury (Hg) species and 0.42–0.71 µg Sn kg−1for tin (Sn) species) allowing analysis of low-mass samples and thus the analysis at the individual organism scale, including glass eels for which samples dry weight ranged from 60 to 100 mg. The methodology was validated with certified reference materials (BCR-464, BCR-477, BCR-710, DOLT-4 and NIST-2977) and applied to the analysis of these pollutants in two developmental stages of the European eel,Anguilla anguilla: individual whole glass eels and muscle tissue from yellow eels. The Adour estuary (South-west France) was selected to monitor the bioaccumulation of organometal species in these organisms, according their developmental stage, their morphological parameters and the sampling site. The results suggest that the accumulation of methylmercury in glass eel tissue is related to weight, with higher concentrations in smaller individuals. Butyltin concentrations were very close to the limit of detection, and no significant differences were detected between glass and yellow eels. [ABSTRACT FROM AUTHOR]

The amphidromous life cycle and morphological characteristics of Sicydiinae species allow them to colonise tropical freshwater habitats from the river mouth to the upper reaches of the river basin. Otolith microchemistry of 62 Sicydium punctatum from the Pérou River, Guadeloupe (French West Indies) was investigated with a femtosecond laser ablation-inductively coupled plasma mass spectrometer (fs-LA-ICP-MS) in order to reconstruct individual life history and follow the possible movements of Sicydium punctatum after the recruitment. Sr:Ca fingerprint confirmed the amphidromous cycle of this species. Variations of Ba:Ca in some individuals indicated changes in environmental conditions after the recruitment in the river. Even if results did not allow concluding to the specific origin of these variations, the hydrological regime and the biofilm composition may have a non negligible influence on the Ba availability. Results supported the use of multi-elemental signatures in otoliths and highlight the need for a large geographical and temporal sampling of Ba and Sr in freshwater systems for a better understanding of amphidromous fish species.
