Accurate measurements of above ground biomass are important to evaluate its contribution as a CO2 absorption source. If global canopy height model can be obtained, we can make an appropriate evaluation. This is MOLI’s mission. Moreover, MOLI can improve the accuracy of digital terrain model. In this paper, we will report on the development status of MOLI for LIDAR observation from space by mounting on the Japanese Experiment Module with International Space Station.

Many organisms exhibit phenotypic plasticity that changes their traits in response to their environment. Although whether or not this plasticity contributes to adaptive evolution is a fundamental question in evolutionary biology, various studies report that natural populations adapt to rapid environmental changes via plasticity, which leads to novel adaptive traits as “triggers.” Namely, phenotypic plasticity has considered allowing an accumulation of genetic mutations to fix the alternative phenotypes induced by nongenetic perturbations that include gene expression noise or epigenetic modification caused by environmental change. However, because the molecular mechanism of phenotypic plasticity is unknown, verification of the process from phenotypic plasticity to genetic fixation remains insufficient. Here we show that decrease in methylated CpG sites leads to loss of plasticity, which triggers genetic fixation of novel traits, in medaka fish (Oryzias latipes). We found that the gut length was correlated with the number of methylated CpG sites upstream of the Plxnb3 gene, which is involved in the developmental process of nerve axons. The medaka, in which the methylated DNA region is deleted by CRISPR/Cas9, showed a loss of plasticity in gut length and a lower survival rate caused by nonoptimal feeding environments. Moreover, standing variation in the promoter region of another gene, Ppp3r1, which is also related to nerve axon development, raised the gene expression and made a longer gut stably in wild medaka groups that lost the gut-length plasticity. Furthermore, our phylogenetic analysis revealed the timing of these evolutionary events, indicating that the loss of phenotypic plasticity by nucleotide substitutions initiates the process of genetic fixation of the novel trait. That is, while phenotypic plasticity plays a role as a buffer of evolution and contributes to environmental adaptation, as previously thought, our molecular data suggest that mutation on CpG site causing the loss of phenotypic plasticity, is the trigger for a generation of novel traits.

MOLI stands for Multi-footprint Observation Lidar and Imager. The feature of MOLI is to set multi-footprints for improving the precision of canopy height, and we can find out whether ground surface is flat or slope because an angle of inclination affects the estimation of canopy height. MOLI is going to be mounted on the Exposed Facility (EF) of the Japanese Experiment Module (JEM, also known as “Kibo”) on the International Space Station (ISS). Now, we are carrying out a feasibility study and some experiments. We introduce an overview and a status of MOLI.

We carried out various tests of 800-mm-diameter aperture, lightweight optics that consisted wholly of carbon fiber-reinforced SiC composite, called HB-Cesic. A cryogenic optical test was performed on the primary mirror to examine any CTE irregularity as a surface change, and only small deformations were observed. The primary mirror was assembled with a convex secondary mirror into an optical system and tested under vacuum at the 6-m-diameter radiometer space chamber at Tsukuba Space Center of JAXA, where we have prepared interferometric metrological facilities to establish techniques to test large optical systems in a horizontal light-axis configuration. The wavefront difference between under vacuum and under atmosphere was confirmed to be less than 0.1 λ at λe633 nm, if we realigned the optical axis of the interferometer and flat mirror under vacuum. We also demonstrated a stitching interferometry using the Φ800-mm optics by rotating a mask wheel of subapertures in front of the optical reference flat. The wavefront stitched from eight individual measurements of Φ275-mm subapertures differs from the full-aperture measurement without the mask by about 0.1 λ nm RMS, which showed the technique could able to be applied to test large telescopes especially for infrared wavelength region.

Canopy height is a factor to evaluate the above-ground biomass of a forest area by remote sensing techniques. Measurements of canopy height from space progress to understand the carbon budget. JAXA has studied a space-borne vegetation lidar (the Multi-footprint Observation Lidar and Imager :MOLI) that observes canopy height on the new exposed facility of the Japanese Experiment Module (JEM), the IVA-replaceable Small Exposed Experiment Platform (i-SEEP) on the International Space Station (ISS). The MOLI will be the first space lidar for earth environmental observation in Japan; a high-energy pulsed laser with a long operational life is key to the operation of the MOLI mission. The concepts underlying the MOLI and vacuum test of functional model of pulsed laser transmitter will be discussed herein.

A large-scale lightweight mirror that is made of silicon carbide-based material is required for the coming astronomical and earth observation missions. The influence of the inhomogeneity of the coefficient of thermal expansion (CTE) on specular surface accuracy was studied as an important technological issue for such a large optical component. At first, a systematic case study for the series of CTE’s main factors was conducted using the finite element method, and consequently a comprehensive equation to calculate the amount of surface deviation was derived. Based on that technology, finite element analysis to simulate the surface accuracy profile that a test mirror sample showed during cryogenic measurement was carried out using experimentally obtained CTE data from cutout test pieces, and the profile was successfully reproduced.

Chinese grass shrimp, Palaemonetes sinensis (Sollaud, 1911) were collected from inland waters of Nao-shima Island and Awa-shima Island, Seto Inland Sea, western Japan. The appearance of P. sinensis is thought to be due to the escape of imported shrimps used as sport fishing bait. P. sinensis looks conspicuously like the native freshwater shrimp, Palaemon paucidens De Haan, 1844. However, the two species have different eye sizes and differences in three diagonal stripes on the carapace. We provide a simple method using eye size and carapace color pattern to enable preliminary differentiation of the two species in the field.

The Japan Aerospace Exploration Agency has studied large-scale, lightweight mirrors constructed of SiC-based materials as a key technology for future earth observations and astronomical missions. One of the most important technical issues for large-scale ceramic components is their quality stability (viz., differences in material properties depending on the part and the processing), which might influence the structural and/or thermal reliability through unforeseen deformation and breakage. In this study, the authors used a simple, low-cost method for evaluating the properties of SiC mirror materials. The Japan Aerospace Exploration Agency has studied a large-scale lightweight mirror constructed of reaction-bonded silicon carbide-based material as a key technology in future astronomical and earth observation missions. The authors selected silicon carbide as the promising candidate due to excellent characteristics of specific stiffness and thermal stability. One of the most important technical issues for large-scale ceramic components is the uniformity of the material's property, depending on part and processing. It might influence mirror accuracy due to uneven thermal deformation. The authors conducted systematic case studies for the conditions of CTE by finite element analysis to know the typical influence of material property nonuniformity on mirror accuracy and consequently derived a comprehensive empirical equation for the series of CTE's main factors. Multi-footprint Observation LIDAR and Imager (MOLI) is a candidate mission for International Space Station – Japanese Experiment Module. The mission objective MOLI is to manage forest and to be a good calibrator for evaluation of forest biomass using satellite instrument such as L-band SAR. SAR is the powerful tool to evaluate biomass globally. However it has some signal saturation over 100 t/ha biomass measurement, whereas Vegetation LIDAR is expected to measure higher mass precisely. MOLI is designed to evaluate forest biomass with high accuracy. An imager, that is equipped together in good registration with LIDAR, will help to understand the situation of target forest. Also two simultaneous Laser beams from MOLI will calibrate the relief effect, which affects the precision of canopy height extremely. Using together with L-band SAR observation data or multispectral image, it is expected to have a good “wall to wall” biomass map with its phonological information. Such MOLI observation capability is so important, because both quantity and quality evaluation of biomass are essential for carbon circulation system understandings. Currently, as a key technical development, LASER Transmitters for MOLI is under test in vacuum condition. Its power is 40mJ and PRF is 150Hz. Pressure vessel design for LIDAR transmitter is supressing Laser induced contamination effect. MOLI is now under study towards around 2020 operation.

The demand for outpatient management of spontaneous pneumothorax (SP) has been increased, therefore we evaluated the effectiveness of Thoracic Egg(TE), a portable thoracic drainage kit for SP. We studied 43 SP patients who had received TE treatment between May 2008 and October 2010. Ages were ranged 12~73 years (mean:29.1), with 39 males, 4 females, 25 had primary, 18 had recurrent, pneumothorax. Of the 43 patients, 23 were treated outpatient therapy with TE only, 20 were required hospitalization for persistent air leakage, poor expansion or intent to surgery. Surgical intervention was undergone in 18 patients for persistent air leaks or recurrent pneumothorax. The average length of treatment was 8.4 days for outpatient therapy only cases. Of 25 patients who had primary SP, 18( 72%) were not required hospitalization. Outpatient therapy using TE was considered very useful for SP, especially for primary cases.

The aim of this study was to evaluate the hypothesis that cerebral hemoglobin (Hb) oxygenation is related to phosphorylation potential during primary and secondary cerebral energy failure in newborn infants who have experienced birth asphyxia. We subjected newborn piglets to severe transient cerebral hypoxic-ischemia followed by resuscitation and examined cerebral energy metabolism by 31P-magnetic resonance spectroscopy and evaluated changes in cerebral Hb oxygen saturation (ScO2) using full-spectrum near-infrared spectroscopy before, during, and up to 54 h after the hypoxic-ischemic insult. ScO2 was significantly decreased during the hypoxic-ischemic insult compared with baseline values. During secondary energy failure, piglets were separated based on the relationship between the ratio of phosphocreatine to inorganic phosphate and ScO2; those with a negative correlation were less injured than those with a positive correlation. These results indicate that changes in ScO2 as measured by near-infrared spectroscopy are related to phosphorylation potential during secondary energy failure in asphyxiated infants.

Using optical topography, changes in the cerebral oxygenation were compared in the parieto-temporal lobe of preterm and term infants of equal postconceptional age in response to verbal stimulation. Eight preterm infants of gestational age 23-34 weeks were studied at postconceptional term age (38-46 weeks). Ten term infants were studied at 2-11 days after birth. Twenty-four-channel near-infrared optical topography (NIOT) was used to measure changes in concentration of oxyhemoglobin ([oxyHb]), deoxyhemoglobin ([deoxyHb]) and total hemoglobin ([totalHb]) in the bilateral temporal cortices. Verbal stimulation was provided by a recording of a Japanese fairy tale. The latency in response to verbal stimulation was significantly shorter in the preterm infants than in the term infants. This time is thought to reflect brain development, particularly the development of the neuro-vascular coupling mechanisms in the cerebral cortex. The present results indicate that the number of days after birth is more closely related to development of auditory system and neuro-vascular coupling than is postconceptional age. Thus, this suggests that early extrauterine environment affects the cortical responses to verbal stimulation in preterm infants.

Evaluation of the lung function involves the measurement of many factors. The closing volume (CV) curve is clinically important as an index of uneven alveolar ventilation and airway closure. Although conventional methods for CV measurement are usually based on the pattern of the exhaled nitrogen (N2) concentration curve with respect to the lung volume, it is often difficult to measure the steep pattern of patients with chronic obstructive pulmonary disease (COPD). In this paper, we proposed a new method called the "N2 first derivative (fdN) wave method" for measuring CV. The N2 concentration of the CV curve was transformed to a derivative with respect to the lung volume, which revealed the existence of cardiogenic oscillations. Discrimination between phases III and IV was straightforward based on the difference in the slope or in the amplitude of oscillations of the fdN wave. Our new method was able to distinguish phase IV from phase III using the difference in amplitude of the oscillation of the fdN wave even in the presence of COPD with steep patterns of the CV curve. Close relationships were seen among normal subjects including COPD patients in both the slope of the alveolar plateau (ΔN2) and the CV values measured with the conventional and new methods. Background: The light-emitting diode is used as one of the new light sources for phototherapy. NeoBLUE (Atom Medical, Tokyo, Japan) incorporates blue light-emitting diodes for the treatment of neonatal hyperbilirubinemia. The authors compared the in vitro efficacy of neoBLUE with conventional phototherapy devices. Methods: The three light devices used included neoBLUE and two conventional phototherapy devices with six blue–white (BW) or six green (GR) fluorescent tubes. A bilirubin/human serum albumin solution (15 mg/dL) in 200 × 300 mm elliptical bag was irradiated with each three light device. The average light intensity of neoBLUE, BW and GR was 22.5, 10.2 and 2.6 μW/cm2 per nm, respectively, for the irradiated area. Bilirubin photoisomers and native bilirubin were measured by high-performance liquid chromatography. Results: In neoBLUE, BW and GR, the respective production rate of cyclobilirubin was 6.0, 3.7 and 3.9 × 10−2 mg/dL/min, and the respective (4Z, 15E)-bilirubin/(4Z, 15Z)-bilirubin ratio after irradiation was 0.44, 0.33 and 0.12; the (4Z, 15Z)-bilirubin reduction rate at 20 min after irradiation was 60, 68 and 82%, respectively. The reduction rate of (4Z, 15Z)-bilirubin correlated with the (4Z, 15E)-bilirubin/(4Z, 15Z)-bilirubin ratio. Conclusion: Phototherapy using the neoBLUE under high level may be clinically more effective than therapy using the conventional light source from the results of the production rate of cyclobilirubin.

Background Cyclooxygenase-2 (COX-2) is known to play a role in carcinogenesis and tumor progression. The aim of this study was to evaluate the relationship between COX-2 expression and clinicopathologic features, and to define the importance of COX-2 expression alone and in combination with p53 and Ki-67 expression in the clinical outcome of NSCLC. Methods A total of 219 patients with stage I-IIIB nonsmall cell lung cancer (NSCLC) who previously underwent surgery were analyzed in this study. The COX-2 expression was evaluated by means of immunohistochemistry; p53 and Ki-67 immunoreactivity were also studied. Results The COX-2 expression was observed in 137 patients (63%) and was significantly associated with lymph node metastasis and the histological grade of those with adenocarcinoma ( p = 0.02 and 0.04, respectively). Kaplan-Meier analyses revealed that COX-2 expression was correlated with poor survival ( p = 0.005), whereas multivariate survival analysis did not reveal COX-2 expression to be an independent prognostic factor. When the patients were stratified according to gender, age, tumor histology, and disease stage, COX-2 expression was significantly associated with unfavorable prognosis in males, younger patients (≤ 65 years), and those with adenocarcinoma and stage I tumors. The prognosis of patients with tumors negative for both COX-2 and p53 expression was significantly favorable, whereas those with tumors positive for COX-2 expression and with a high Ki-67 labeling index had a significantly unfavorable prognosis. Conclusions These findings indicate that combined immunohistochemical analysis of COX-2 with p53 and Ki-67 can be useful for identifying the prognosis of NSCLC patients.

This report is of an embryonal rhabdomyosarcoma presenting in the mandible of a 12-year-old boy. The tumour exhibited rapid growth and resistance to chemotherapy. The clinical findings, diagnostic course, and treatment are described in detail.

Summary Background: Estimation of hemodynamics is important for critically ill infants. Pulse dye densitometry (PDD) using indocyanine green (ICG), which enables measurements of circulating blood volume at the bedside, has recently been developed for adults. Methods: We conducted a basic investigation to determine whether this method can be applied to infants and measured circulating blood volume in 25 infants whose gestational ages ranged from 24 to 40 weeks (median, 32 weeks). At first, to validate the accuracy of measurements, arterial ICG concentrations determined by blood sample measurements were compared using a spectrophotometer ([ICG blood]) and by noninvasive measurement using PDD ([ICG pdd]) in seven infants. Background: The developmental changes of the umbilical cortisol levels in neonates at gestational age of 23–41 weeks were studied and the effect of antenatal steroid administration on the umbilical cortisol levels were examined. Methods: Cortisol levels in the umbilical vein (UV) and the umbilical artery (UA) were studied in 35 neonates at the gestational age (GA) of 23–41 weeks with or without antenatal administration of corticosteroids. Serum cortisol concentrations were measured by the high performance liquid chromatography method. Results: The correlation between cortisol levels in UV and birthweight (BW) was weak and negative in premature infants. UV cortisol levels in the neonates with antenatal corticosteroid were lower than those in the neonates without antenatal corticosteroid, but the relation was not significant. The developmental changes of UV cortisol levels were the same as those in Murphy's study (spontaneous-onset labor). The cortisol levels in UV and UA had a significantly positive correlation and both had almost equal concentrations. There were no correlations between cortisol levels in UV and placental weight, Apgar Score at 1 and 5 min. Conclusions: In the neonates whose birthweight was less than 2000 g without antenatal corticosteroid, there was a negative correlation between cortisol levels in UV and BW but there was no correlation between cortisol levels in UV and GA. That the neonates with antenatal corticosteroid would have a suppressed adrenocortical function after birth could not be proved.

Summary Fibrosing mediastinitis is an extremely rare disease in which fibrous tissue is formed progressively within the mediastinum due to various causes. Here, we report two cases of the idiopathic form of fibrosing mediastinitis. We investigated the pathogenesis of fibrosing mediastinitis by immunohistochemical methods, and demonstrated the expression of transforming growth factor β in this disease. These results suggest that transforming growth factor β plays an important role in the pathogenesis of fibrosing mediastinitis.

Background: The clinical effect of phototherapy for neonatal hyperbilirubinemia is based on the production and elimination of cyclobilirubin. Generally, the clinical effect of light sources is estimated by the reduction in the total serum bilirubin level. One procedure with less invasiveness than blood collecting is urine collection. Whether the effectiveness of light sources used for phototherapy could be assessed using measurements of bilirubin photoisomers in urine was studied. Methods: This study was a retrospective analysis of 38 term infants with hyperbilirubinemia who underwent phototherapy. Bilirubin fractions in serum and urine before and 24 h after the phototherapy were measured by high-performance liquid chromatography. The light sources used for the phototherapy were blue-white light (n = 11), Biliblanket plus high output (n = 13) or green light (n = 14). The relationships between serum and urine bilirubin photoisomers after phototherapy and whether the levels of urine bilirubin photoisomer are affected by the light sources with different wavelength characteristic were analyzed. Results: There was no correlation between serum (ZE)-bilirubin and urine configurational isomers, but a weak positive correlation between serum (EZ)-cyclobilirubin and urine structural isomers after phototherapy. Although serum (ZE)-bilirubin levels depended on the wavelength characteristic of each light source during phototherapy, the urine configurational isomer levels did not depend on it. The increase in serum (EZ)-cyclobilirubin levels and the urine structural isomer levels were mostly in agreement. Conclusions: The urine bilirubin structural isomers may be used to estimate the serum (EZ)-cyclobilirubin levels and to evaluate the clinical effects of light sources.

Proton NMR was measured in two Nb–H samples prepared so as to introduce vacancy–hydrogen (vac–H) clusters and a reference sample not containing vac–H clusters. A motionally narrowed signal persisting to very low temperatures was observed only in the former samples, and was assigned to protons executing a cage motion in Nb-atom vacancies. From the temperature dependence of the linewidth between 7 and 30 K, an apparent activation energy of 3.8 meV was deduced.

During the developmental stage, the brain undergoes anatomic, functional, and metabolic changes necessary to support the complex adaptive behavior of a mature individual. Estimation of developmental changes occurring in different regions of the brain would provide a means of relating various behavioral phenomena to maturation‐specific brain structures, thereby providing useful information on structure‐function relationships in both normal and disease states. We used multichannel near‐infrared spectroscopy (MNIRS), a new noninvasive imaging technique for revealing the course of neural activity in selected brain regions, to monitor the activities of the visual cortex as mirrored by hemodynamic responses in infants subjected to photostimulation during natural sleep. In the infants, oxyhemoglobin and total hemoglobin decreased and deoxyhemoglobin increased in the visual cortex with photostimulation. This pattern of responses was different from the response pattern in adults reported previously. The different patterns of responses to photostimulation in the visual cortices of infants and adults might reflect developmental and behavioral differences. It may reflect a different functional organization of the visual cortex in infants or ongoing retinal development. Our results demonstrated that regional hemodynamic change could be detected in a small area around the visual cortex. Near-infrared spectroscopy (NIRS) has been used for measurement of changes in cerebral hemoglobin concentrations in neonates to study cerebral oxygenation and hemodynamics. In this study, measurements by time-resolved reflectance spectroscopy (TRS) were performed in a piglet model with various degrees of cerebral oxygenation to estimate the differential pathlength factor (DPF). A portable three-wavelength TRS system (TRS-10, Hamamatsu Photonics K.K.) with a probe attached to the head of a piglet was used. Eleven newborn piglets were anesthetized and respired by a ventilator to induce stepwise hypoxia loading. The DPF showed positive linear relationship with arterial hemoglobin (Hb) oxygen saturation and sagittal sinus venous Hb oxygen saturation at 761 and 795 nm. The DPF at 835 nm also showed very slight positive linear relationship with arterial hemoglobin oxygen saturation. The DPF values obtained in this study should contribute to a better understanding of noninvasive measurements by NIRS in neonates.
