Abstract

A novel 99mTc generator was proposed herein using nanoparticles consisting of natural Mo and an alumina column for 99mTc scintigraphy. 99mTc was obtained from 99Mo decay produced via the irradiation of nanoparticles consisting of natural Mo with bremsstrahlung γ-rays using a 30 MeV electron beam. The irradiated nanoparticles were placed in an alumina column, and the 99mTc daughter from 99Mo decay was separated by the removal of the ion-exchanged water or saline. This separation procedure was repeated at regular intervals for one week. The maximum separation yield of 99mTc was 14.4 ± 0.7%, which is sufficiently high to produce 99mTc suitable for medical use. Abstract

In order to establish a self-sufficient supply of (99m)Tc, we studied feasibilities to produce its parent nucleus, (99)Mo, using Japanese accelerators. The daughter nucleus, (99m)Tc, is indispensable for medical diagnosis. (99)Mo has so far been imported from abroad, which is separated from fission products generated in nuclear reactors using enriched (235)U fuel. We investigated (99m)Tc production possibilities based on the following three scenarios: (1) (99)Mo production by the (n, 2n) reaction by spallation neutrons at the J-PARC injector, LINAC; (2) (99)Mo production by the (p, pn) reaction at Ep = 50-80 MeV proton at the RCNP cyclotron; (3) (99m)Tc direct production with a 20 MeV proton beam from the PET cyclotron. Among these three scenarios, scenario (1) is for a scheme on a global scale, scenario (2) works in a local area, and both cases take a long time for negotiations. Scenario (3) is attractive because we can use nearly 50 PET cyclotrons in Japan for (99m)Tc production. We here consider both the advantages and disadvantages among the three scenarios by taking account of the Japanese accelerator situation.

Abstract

Chalcogenidecapped octahedral hexatechnetiumIII complexes, Cs4[Tc6S8Br6]·CsBr Cs4[1]·CsBr and PPh44[Tc6S8Br6] PPh44[1], PPh44[Tc6S8I6] PPh44[2], [Tc6Se8I2] [3], and [Tc6Te15] [4] were synthesized. Singlecrystal Xray structural studies were carried out for Cs4[1]·CsBr, [3], and [4]. Sulfidecapped octahedral hexatechnetiumIII clusters [1]4–and [2]4–are discrete complex anions, whereas the selenide and telluridecapped hexatechnetiumIII complexes adopt extended polymeric structures. A cyclic voltammogram of [1]4–in CH3CN showed two oneelectron redox waves: Tc624e23e and Tc623e22e. The potential of the Tc624e23e process is more positive than that of the Re624e23e process for the hexarhenium analog [Re6S8X6]4–X = Br, I, whereas that of the Tc623e22e process shifts to negative values compared to the potential of the Re623e22e process. Thus, the Tc623e mixedvalence state is thermodynamically less stable than Re623e.

Abstract

Abstract: Positron-emission tomography (PET) can be used to visualize active stage cancer. Fluorine-18 ([18F])-labeled 2-([18F])2-deoxy-2-fluoroglucose (([18F])-FDG), which accumulates in glucose-dependent tissues, is a good cancer-targeting tracer. However, ([18F])-FDG is obscured in glucose-dependent normal tissues. In this study, we assessed the cancer-selective accumulation of zinc-labeled glycoconjugated 5,10,15,20-tetrakis(pentafluorophenyl)porphyrin (ZnGlc1–4), both in vitro and in vivo. Experiments using both normal and cancer cells confirmed the relationship between cancer cell-selective accumulation and the substitution numbers and orientations of glycoconjugated porphyrins. ZnGlctrans-2 accumulated at greater levels in cancer cells compared with other glycoconjugated porphyrins. PET imaging showed that ZnGlctrans-2 accumulated in tumor. [Copyright &y& Elsevier]
