Abstract Behavioral and physiological synchrony facilitate emotional closeness in attachment relationships. The aim of this pseudorandomized cross-over study was to investigate the emotional and physiological link, designated as co-modulation, between dogs and their owners. We measured the heart rate variability (HRV) and physical activity of dogs belonging to co-operative breeds (n = 29) and their owners during resting baselines and positive interaction tasks (Stroking, Training, Sniffing, Playing) and collected survey data on owner temperament and dog–owner relationship. Although overall HRV and activity correlated between dogs and their owners across tasks, task-specific analyses showed that HRV of dogs and owners correlated during free behaving (Pre- and Post-Baseline), whereas the activity of dogs and owners correlated during predefined interaction tasks (Stroking and Playing). Dog overall HRV was the only predictive factor for owner overall HRV, while dog height, ownership duration, owner negative affectivity, and dog–owner interaction scale predicted dog overall HRV. Thus, the characteristics of dog, owner, and the relationship modified the HRV responses in dog–owner dyads. The physiology and behavior of dogs belonging to co-operative breeds and their owners were therefore co-modulated, demonstrating physiological and emotional connection comparable to those found in attachment relationships between humans.

Abstract It is well known that dogs are capable of following human verbal instructions. However, very little is known about the equivalent ability in cats. In this study, we used a switched stimuli task to examine whether cats rapidly form picture-word association, which is a fundamental ability for word learning. We presented cats with two meaningless picture-word combinations, in the habituation phase. Then, on half of the trials we switched the combination (switched condition), but the other half of the trials remained as before (non-switched condition). If cats rapidly form picture-word association, they were expected to look at the monitor for longer in the switched condition, reflecting detection of the change. We used human speech as stimuli in Exp.1, and mechanical sounds (electronic sounds) in Exp.2. Cats expressed detection of the switched combination in Exp.1, where human speech and objects were paired. However, in Exp.2 where non-social sounds and objects were paired, there was no statistical difference between switched and non-switched conditions, although there was a main effect of condition when the data from the two experiments were pooled. These results demonstrate that cats can rapidly form picture-word association. Further research should investigate whether domestication has played a role in this ability.

Abstract Background The gut microbiota plays an important role in the development of behavior and immunity in infants and juveniles. Early weaning (EW), a form of social stress in mice, leads to increased anxiety and an enhanced stress response in the hypothalamic-pituitary-adrenal axis during adulthood. Early life stress also modulates the immune system and increases vulnerability to infection. However, studies investigating the causal relationships among juvenile stress, microbiota changes, and immune and behavioral deficits are limited. Therefore, we hypothesized that EW alters gut microbiota composition and impairs the development of the nervous and immune systems. Results EW mice moved longer distances in the marble-burying test and had longer immobility times in the tail suspension test than normal weaning (NW) mice. In parallel, the gut microbiome composition differed between NW and EW mice, and the abundance of Erysipelotrichacea in EW mice at 8 weeks of age was lower than that in NW mice. In an empirical study, germ-free mice colonized with the gut microbiota of EW mice (GF-EW mice) demonstrated higher depressive behavior than GF mice colonized with normal weaning microbiota (GF-NW mice). Immune cell profiles were also affected by the EW microbiota colonization; the number of CD4 + T cells in the spleen was reduced in GF-EW mice. Conclusion Our results suggest that EW-induced alterations in the gut microbiota cause depressive behaviors and modulate the immune system.

IntroductionThe One Health concept is a comprehensive understanding of the interaction between humans, animals, and the environment. The cohabitation of humans and pets positively affects their physical, mental, and social well-being. It is recognized as an essential factor from the One Health perspective. Furthermore, a healthy balance in the gut microbiome is essential for good health, and the changes in the gut microbiome associated with cohabitation between humans and pets could potentially affect various aspects of the health of both hosts. Therefore, elucidating the sharing of gut bacteria between humans and pets associated with cohabitation is important for understanding One Health. However, most studies have examined sharing at the taxonomic level, and it remains unclear whether the same bacteria are transferred between humans and pets, and whether they mutually influence each other.MethodsHere, microbiome analysis and shared 16S rRNA gene amplicon sequence variant (ASV) analysis were conducted before the start of cohabitation between humans and dogs, as well as at 2 weeks, 1 month, and 3 months after cohabitation.Results16S rRNA gene ASVs analysis indicated that gut microbes have been transferred between humans and dogs. The overall structure of the gut microbiota within human–dog pairs remained unchanged after 3 months of adaptation. However, 11ASVs were shared within human–dog pairs. Many shared ASVs were highly abundant within each host, and this high abundance may be considered a factor that influences bacterial transfer between hosts.DiscussionOur results provide important insights into the potential for the transfer of gut bacteria between humans and dogs. These findings are considered crucial for understanding the impact of human–dog cohabitation on various aspects of health.

Abstract Many animal species, including mice, form societies of numerous individuals for survival. Understanding the interactions between individual animals is crucial for elucidating group behavior. One such behavior in mice is huddling, yet its analysis has been limited. In this study, we propose a cost‐effective method for monitoring long‐term huddling behavior in mice using online image processing with OpenCV. This method treats a single mouse or a group of mice as a cluster of pixels (a ‘blob’) in video images, extracting and saving only essential information such as areas, coordinates, and orientations. This approach reduces data storage needs to 1/200000th of what would be required if the video were recorded in its compressed form, thereby enabling long‐term behavioral analysis. To validate the performance of our algorithm, ~2000 video frames were randomly chosen. We manually counted the number of clusters of mice in these frames and compared them with the number of blobs automatically detected by the algorithm. The results indicated a high level of consistency, exceeding 90% across the selected video frames. Initial observations of both male and female groups suggested some variations in huddling behavior among male and female groups; however, these results should be interpreted cautiously due to a small sample. Group behavior is known to be disrupted in several neuropsychiatric disorders, such as autism. Various mouse models of these disorders have been proposed. Our measurement system, when combined with drug or genetic modification screening, could provide a valuable tool for high‐throughput analyses of huddling behavior.

Abstract The number of mothers suffering from mental illness is increasing steadily, particularly under conditions of the coronavirus pandemic. The identification of factors that contribute to resilience in mothers is urgently needed to decrease the risks of poor physical and psychological health. We focused on the risk of parenting stress and psychological resilience in healthy mothers with no psychiatric and physical disorders and conducted two studies to examine the relationships between intestinal microbiota, physical condition, and psychological state. Our results showed that alpha diversity and beta diversity of the microbiome are related to high parenting stress risk. Psychological resilience and physical conditions were associated with relative abundances of the genera Blautia, Clostridium, and Eggerthella. This study helps further understand the gut–brain axis mechanisms and supports proposals for enhancing resilience in mothers.

Abstract The affiliative relationship between humans and dogs is compared to a mother–infant attachment relationship. We hypothesized that dog’s attachment behavior in negative emotional state aroused the owner’s attention toward the dog, that is, reduced parasympathetic activity. We measured heart rate variability in both dogs and humans during the Strange Situation Test to examine whether the owners' parasympathetic activity was decreased by being gazed at by their dogs. Our results in a short-term of 6 s before and after the moment the dog gazed at the human face indicated that dogs’ parasympathetic activity was lower when the dogs were gazing at their owners than when it was gazing at unfamiliar persons. Dog’s autonomic activity was also lower when the dogs were living with their owners for a longer period. However, we could not determine whether gaze from the dog affected the autonomic activity in humans as attachment behavior.

The matrilineal transmission of maternal behavior has been reported in several species. Studies, primarily on rats, have suggested the importance of postnatal experience and the involvement of epigenetic mechanisms in mediating these transmissions. This study aims to determine whether the matrilineal transmission of maternal behavior occurs in mice and whether the microbiota is involved. We first observed that early weaned (EW) female mice showed lower levels of maternal behavior, particularly licking/grooming (LG) of their own pups, than normally weaned (NW) female mice. This difference in maternal behavioral traits was also observed in the second generation, even though all mice were weaned normally. In the subsequent cross-fostering experiment, the levels of LG were influenced by the nurturing mother but not the biological mother. Finally, we transplanted the fecal microbiota from EW or NW mice into germ-free (GF) mice raising pups. The maternal behaviors that the pups exhibited toward their own offspring after growth were analyzed, and the levels of LG in GF mice colonized with microbiota from EW mice were lower than those in GF mice colonized with microbiota from NW mice. These results clearly indicate that, among maternal behavioral traits, LG is intergenerationally transmitted in mice and suggest that the vertical transmission of microbiota is involved in this process. This study demonstrates the universality of the intergenerational transmission of maternal behavioral traits and provides new insights into the role of microbiota.

Summary: Building affiliative relationships with others is important for mental health. Recently, robots have been expected to play a role in improving mental health, but there is little scientific evidence as to whether they can build affiliative relationships with humans. To investigate that, we conducted studies combining behavior, physiology and questionnaires for companion robot Owners and Non-Owners. The results reveal that the steady-state concentration of oxytocin, a hormone related to affiliative relationships, was significantly higher in Owners than in Non-Owners. In addition, the Owners showed more behaviors indicative of intimacy than the Non-Owners. These results suggest that humans can build affiliative relationships with robots. Fifteen minutes of contact with the robot decreased the concentration of cortisol in both groups, suggesting that even a brief contact can contribute to improving mental health. Therefore, relationships between humans and robots may be one option to improve mental health and enhance well-being.

IntroductionLate adolescence is a crucial period during which individuals connect with new communities. Furthermore, their mental health has lasting effects on their overall well-being. Involvement with family and the local community plays a significant role in shaping adolescents’ personalities and well-being. Additionally, pets, such as dogs and cats, function as social catalysts and increase interactions with family and the local community. We hypothesized that pet ownership would increase involvement with family and the local community and thereby impact adolescents’ personalities and well-being.MethodsTherefore, this study investigated whether owning dogs or cats was related to well-being through increased involvement with family and local community members in late adolescence. Data were collected via a questionnaire administered to high school and university students. The questionnaire included questions on basic information about adolescents and their families, pet ownership experience, family and local community involvement, well-being, cultural estrangement inventory, and general trust.ResultsStructural equation modeling revealed that adolescent women who owned dogs or cats had higher well-being and general trust through their involvement with their families. Although previous research reported that men who had experienced pet ownership in childhood were more sociable in old age, the effect of pet ownership on men was not observed in this study.DiscussionDuring late adolescence, when individuals experience many connections with new communities, the effects of pets may temporarily decrease. Therefore, future cohort studies should examine the effects of pets on each age group.

Abstract The dog (Canis familiaris) was the first domesticated animal and hundreds of breeds exist today. During domestication, dogs experienced strong selection for temperament, behaviour, and cognitive ability. However, the genetic basis of these abilities is not well-understood. We focused on ancient dog breeds to investigate breed-related differences in social cognitive abilities. In a problem-solving task, ancient breeds showed a lower tendency to look back at humans than other European breeds. In a two-way object choice task, they showed no differences in correct response rate or ability to read human communicative gestures. We examined gene polymorphisms in oxytocin, oxytocin receptor, melanocortin 2 receptor, and a Williams–Beuren syndrome-related gene (WBSCR17), as candidate genes of dog domestication. The single-nucleotide polymorphisms on melanocortin 2 receptor were related to both tasks, while other polymorphisms were associated with the unsolvable task. This indicates that glucocorticoid functions are involved in the cognitive skills acquired during dog domestication.

Abstract Humans communicate with each other through language, which enables us talk about things beyond time and space. Do non-human animals learn to associate human speech with specific objects in everyday life? We examined whether cats matched familiar cats’ names and faces (Exp.1) and human family members’ names and faces (Exp.2). Cats were presented with a photo of the familiar cat’s face on a laptop monitor after hearing the same cat’s name or another cat’s name called by the subject cat’s owner (Exp.1) or an experimenter (Exp.2). Half of the trials were in a congruent condition where the name and face matched, and half were in an incongruent (mismatch) condition. Results of Exp.1 showed that household cats paid attention to the monitor for longer in the incongruent condition, suggesting an expectancy violation effect; however, café cats did not. In Exp.2, cats living in larger human families were found to look at the monitor for increasingly longer durations in the incongruent condition. Furthermore, this tendency was stronger among cats that had lived with their human family for a longer time, although we could not rule out an effect of age. This study provides evidence that cats link a companion's name and corresponding face without explicit training.

In order to validate a previously inferred function of the lateral prefrontal cortex (LPFC)-caudate nucleus (CdN) pathway in humans in inhibitory control and working memory, Oguchi et al used pathway-selective chemogenetic manipulation of neural activity in macaques. By suppressing pathway-specific neural activity and observing resultant behavioural changes, they provided evidence that the LPFC-CdN pathway does indeed play a role in inhibitory control.

Abstract In vivo calcium imaging with genetically encoded indicators has recently been applied to macaque brains to monitor neural activities from a large population of cells simultaneously. Microendoscopic calcium imaging combined with implantable gradient index lenses captures neural activities from deep brain areas with a compact and convenient setup; however, this has been limited to rodents and marmosets. Here, we developed miniature fluorescent microscopy to image neural activities from the primary visual cortex of behaving macaques. We found tens of clear fluorescent signals from three of the six brain hemispheres. A subset of these neurons showed clear retinotopy and orientation tuning. Moreover, we successfully decoded the stimulus orientation and tracked the cells across days. These results indicate that microendoscopic calcium imaging is feasible and reasonable for investigating neural circuits in the macaque brain by monitoring fluorescent signals from a large number of neurons.

Sensory signals are critical to perform adaptive social behavior. During copulation, male mice emit ultrasonic vocalizations (USVs). Our previous studies have shown that female mice exhibit approach behavior toward sound sources of male USVs and that, after being exposed to a male pheromone, exocrine gland-secreting peptide 1 (ESP1), female mice exhibited a preference toward a particular type of male USVs. These findings suggest that male USVs modulate female courtship behavior. However, it remains unclear which brain regions and what cell types of neurons are involved in neuronal processing of male USVs. To clarify this issue, immediate early gene analysis, behavioral analysis, and neurochemical analysis were performed. The in situ hybridization analysis of c-fos mRNA in multiple brain regions showed that neurons in the prelimbic cortex were responsive to presentation of male USVs in the presence of ESP1. Furthermore, this study found that activity of prelimbic cortex was correlated with the duration of female exploration behavior toward a sound source of the USVs. Finally, by using double immunohistochemistry, the present study showed that the prelimbic neurons responding to the presentation of male USVs were presumably excitatory glutamatergic neurons. These results suggest that the prelimbic cortex may facilitate female courtship behavior in response to male USVs.

Summary: The human vesicular monoamine transporter 1 (VMAT1) harbors unique substitutions (Asn136Thr/Ile) that affect monoamine uptake into synaptic vesicles. These substitutions are absent in all known mammals, suggesting their contributions to distinct aspects of human behavior modulated by monoaminergic transmissions, such as emotion and cognition. To directly test the impact of these human-specific mutations, we introduced the humanized residues into mouse Vmat1 via CRISPR/Cas9-mediated genome editing and examined changes at the behavioral, neurophysiological, and molecular levels. Behavioral tests revealed reduced anxiety-related traits of Vmat1Ile mice, consistent with human studies, and electrophysiological recordings showed altered oscillatory activity in the amygdala under anxiogenic conditions. Transcriptome analyses further identified changes in gene expressions in the amygdala involved in neurodevelopment and emotional regulation, which may corroborate the observed phenotypes. This knock-in mouse model hence provides compelling evidence that the mutations affecting monoaminergic signaling and amygdala circuits have contributed to the evolution of human socio-emotional behaviors.

Physical and mental effects stemming from COVID-19 have impacted not only people’s lives but also the lives of their pets, which in recent years are often seen as members of the family. This study aimed to explore the impact of the COVID-19 pandemic in Japan on pets and their owners. Participants reported changes in physical activity and mental health, as well as working conditions and frequency of interaction with pets, before and after behavioural restrictions due to the COVID-19 pandemic. We also asked about their pets’ behaviours using the Feline Behavioural Assessment and Research Questionnaire (Fe-BARQ) and the Canine Behavioural Assessment and Research Questionnaire (C-BARQ). This study showed that most cat and dog owners spent more time at home due to the COVID-19 behavioural restrictions and that the frequency of contact interaction with their pets increased. However, this study showed higher stress-related behaviours (e.g., cats: excessive grooming; dogs: aggression towards owners) among pets whose owners increased contact interaction. Furthermore, owners’ low mental health was correlated with high stress-related behaviours (e.g., touch sensitivity) in pets. The results of this study indicate that the lockdown caused by the COVID-19 pandemic in Japan may have affected not only the lives of owners but also the interaction between owners and their pets, and consequently their pets’ behaviours. Therefore, there is also concern that changes in lifestyle patterns caused by pandemics could form a negative feedback loop between the health status of both owners and their pets.

We examined whether dogs show emotional response to social stimuli played on videos. Secondary, we hypothesized that if dogs recognize themselves in videos, they will show a different emotional response to videos of self and other dogs. We compared heart rate variability among four video stimuli: a video of the owner ignoring another dog (OW-A-IGN), a video of a non-owner interacting with another dog (NOW-A-INT), a video of the owner interacting with another dog (OW-A-INT), and a video of the owner interacting with the dog subject (OW-S-INT). The results showed that root mean square of the difference between adjacent R-R Intervals (RMSSD) and standard deviation of the R-R Interval (SDNN) were lower in NOW-A-INT and OW-S-INT than in OW-A-IGN. There was no statistical difference in the responses to OW-S-INT and OW-A-INT, suggesting that dogs did not distinguish themselves and other dogs in videos. On the other hand, the difference in mean R-R Interval between OW-S-INT and OW-A-INT showed positive correlation with the score of attachment or attention-seeking behavior. Therefore, this study does not completely rule out self-recognition in dogs and there remains the possibility that the more attached a dog to its owner, the more distinct the dog's emotional response to the difference between the self-video stimulus and the video stimulus of another dog. Further studies are needed to clarify this possibility.

Domestic cats (Felis silvestris catus) can live in high densities, although most feline species are solitary and exclusively territorial animals; it is possible that certain behavioral strategies enable this phenomenon. These behaviors are regulated by hormones and the gut microbiome, which, in turn, is influenced by domestication. Therefore, we investigated the relationships between the sociality, hormone concentrations, and gut microbiome of domestic cats by conducting three sets of experiments for each group of five cats and analyzing their behavior, hormone concentrations (cortisol, oxytocin, and testosterone), and their gut microbiomes. We observed that individuals with high cortisol and testosterone concentrations established less contact with others, and individuals with high oxytocin concentrations did not exhibit affiliative behaviors as much as expected. Additionally, the higher the frequency of contact among the individuals, the greater the similarity in gut microbiome; gut microbial composition was also related to behavioral patterns and cortisol secretion. Notably, individuals with low cortisol and testosterone concentrations were highly tolerant, making high-density living easy. Oxytocin usually functions in an affiliative manner within groups, but our results suggest that even if typically solitary and territorial animals live in high densities, their oxytocin functions are opposite to those of typically group-living animals.

We aimed to clarify the changes in facial morphology of cats in relation to their interactions with humans. In Study 1, we compared the facial morphology of cats (feral mixed breed, owned domestic mixed breed, and owned domestic purebreds) with that of African wildcats. After collecting 3295 photos, we found that owned domestic cats’ noses were significantly shorter than those of African wildcats and feral mixed breed, and there were no significant differences between the latter two. The eye angles were significantly more gradual in owned domestic purebreds than in the other groups. In Study 2, we examined the correlation between facial morphology and years with the owner, and found that the former is not affected by the latter. This suggests that changes in facial morphology are possibly transgenerational changes. The difference in facial morphology between wildcats and owned cats might be caused by domestication, and that between feral cats and owned cats might be due to feralization. In Study 3, we investigated whether cats’ facial features affect cuteness ratings. We asked human participants to evaluate the cuteness of cats’ face images and found that faces with shorter nose lengths were considered cuter. This suggests that owned domestic cats’ facial morphology is preferred by humans.

Individual differences in the sociality of cats are influenced by inherited and environmental factors. We recently revealed that hormones can make a difference in intraspecies social behavior. It remains unclear whether cat behavior toward humans is modulated by hormones. Therefore, we analyzed the relationship between cat behavior and their basal hormone concentrations after spending time together with human experimenters. In addition, we analyzed the relationship between cat behavior and the timing of when the individual cats began living with a human because the sociality of cats could be dependent on their developmental experiences. The results showed that male cats that began living with humans earlier had more contact with an experimenter. In addition, individual male cats with low testosterone levels were more likely to interact with an experimenter. These findings of this pilot study suggest that the sociality of male cats toward humans is affected by testosterone and the age at which they begin to live with humans.

Testosterone masculinizes male sexual behavior through an organizational and activational effects. We previously reported that the emission of ultrasonic vocalizations (USVs) in male mice was dependent on the organizational effects of testosterone; females treated with testosterone in the perinatal and peripubertal periods, but not in adults, had increased USV emissions compared to males. Recently, it was revealed that male USVs have various acoustic characteristics and these variations were related to behavioral interactions with other mice. In this regard, the detailed acoustic characteristic changes induced by testosterone have not been fully elucidated. Here, we revealed that testosterone administered to female and male mice modulated the acoustic characteristics of USVs. There was no clear difference in acoustic characteristics between males and females. Call frequencies were higher in testosterone propionate (TP)-treated males and females compared to control males and females. When the calls were classified into nine types, there was also no distinctive difference between males and females, but TP increased the number of calls with a high frequency, and decreased the number of calls with a low frequency and short duration. The transition analysis by call type revealed that even though there was no statistically significant difference, TP-treated males and females had a similar pattern of transition to control males and females, respectively. Collectively, these results suggest that testosterone treatment can enhance the emission of USVs both in male and female, but the acoustic characteristics of TP-treated females were not the same as those of intact males.

In mice, early exposure to environmental odors affects social behaviors later in life. A signaling molecule, Semaphorin 7A (Sema7A), is induced in the odor-responding olfactory sensory neurons. Plexin C1 (PlxnC1), a receptor for Sema7A, is expressed in mitral/tufted cells, whose dendrite-localization is restricted to the first week after birth. Sema7A/PlxnC1 signaling promotes post-synaptic events and dendrite selection in mitral/tufted cells, resulting in glomerular enlargement that causes an increase in sensitivity to the experienced odor. Neonatal odor experience also induces positive responses to the imprinted odor. Knockout and rescue experiments indicate that oxytocin in neonates is responsible for imposing positive quality on imprinted memory. In the oxytocin knockout mice, the sensitivity to the imprinted odor increases, but positive responses cannot be promoted, indicating that Sema7A/PlxnC1 signaling and oxytocin separately function. These results give new insights into our understanding of olfactory imprinting during the neonatal critical period.

The human parenting brain network mediates caregiving behaviors. When exposed to the stimuli of their infants, compared with non-parents, both fathers and mothers exhibit distinct patterns of neural activation. As human males, relative to females, do not undergo robust physiological changes during pregnancy, when and how the paternal brain networks begin to form remains unclear. Thus, using functional MRI, we examined brain activation in response to infant-interaction videos in two groups, childless males and first-time expectant fathers during their partners’ early pregnancy before remarkable changes in their partners’ appearances commenced. Multivoxel pattern analysis revealed that expectant fathers’ left anterior insula and inferior frontal gyrus showed incipient changes in response to parenthood during early pregnancy. Furthermore, these changes were associated with several paternal traits, such as a negative image toward parenting. Such external factors might influence the paternal brain's development during early pregnancy.

Summary: Defensive behaviors are evolved responses to threat stimuli, and a potential threat elicits risk assessment (RA) behavior. However, neural mechanisms underlying RA behavior are hardly understood. Urocortin-3 (Ucn3) is a member of corticotropin-releasing factor peptide family and here, we report that Ucn3 neurons in the hypothalamic perifornical area (PeFA) are involved in RA of a novel object, a potential threat stimulus, in mice. Histological and in vivo fiber photometry studies revealed that the activity of PeFA Ucn3 neurons was associated with novel object investigation involving the stretch-attend posture, a behavioral marker for RA. Chemogenetic activation of these neurons increased RA and burying behaviors toward a novel object without affecting anxiety and corticosterone levels. Ablation of these neurons caused the abnormal behaviors of gnawing and direct contacts with novel objects, especially in a home-cage. These results suggest that PeFA Ucn3 neurons modulate defensive responses to a potential threat stimulus.

Dogs (Canis familiaris) and cats (Felis silvestris catus) have been domesticated through different processes. Dogs were the first domesticated animals, cooperating with humans by hunting and guarding. In contrast, cats were domesticated as predators of rodents and lived near human habitations when humans began to settle and farm. Although the domestication of dogs followed a different path from that of cats, and they have ancestors of a different nature, both have been broadly integrated into—and profoundly impacted—human society. The coexistence between dogs/cats and humans is based on non-verbal communication. This review focuses on “gaze,” which is an important signal for humans and describes the communicative function of dogs’ and cats’ eye-gaze behavior with humans. We discuss how the function of the gaze goes beyond communication to mutual emotional connection, namely “bond” formation. Finally, we present a research approach to multimodal interactions between dogs/cats and humans that participate in communication and bond formation.

The olfactory bulb is arranged in glomeruli defined by their olfactory receptor expression. The authors identify an olfactory receptor for fox odour, TMT, and show that activation of the glomerulus expressing that receptor in mice leads to immobility, but does not lead to other fear behaviours.

Neuropilin 2 (Nrp2) is expressed by a subset of mitral cells in the postero-ventral olfactory bulb in mice. Here the authors show that Nrp2 is sufficient to instruct mitral cell targeting to the anterior part of medial amygdala that modulates olfactory-driven attractive social behaviour.

Abstract Background The common marmoset has been used as an experimental animal for various purposes. Because its average weight ranges from 250 to 500 g, weight loss quickly becomes critical for sick animals. Therefore, effective and non-stressful treatment for chronic diseases, including diarrhoea, is essential. Case presentation We report a case in which faecal microbiota transplantation (FMT) led to immediate recovery from chronic and recurrent diarrhoea caused by Clostridium difficile infection. A male common marmoset experienced chronic diarrhoea after antibiotic treatments. The animal experienced severe weight loss, and a faecal sample was confirmed to be C. difficile-positive but was negative for protozoa. Metronidazole was partially effective at the first administration but not after the recurrence of the clinical signs. Then, oral FMT was administered to the subject by feeding fresh faeces from healthy individuals mixed with the marmoset’s usual food. We monitored the faeces by categorization into four groups: normal, loose, diarrhoea, and watery. After the first day of FMT treatment, the marmoset underwent a remarkable recovery from diarrhoea, and after the fourth day of treatment, a test for C. difficile was negative. The clinical signs did not recur. The marmoset recovered from sinusitis and bilateral dacryocystitis, which also did not recur, as a by-product of the improvement in its general health caused by the cessation of diarrhoea after the FMT. Conclusion This is the first reported case of successful treatment of a marmoset using oral FMT. As seen in human patients, FMT was effective for the treatment of recurrent C. difficile infection in a captive marmoset.

Emotional contagion is a primitive form of empathy that does not need higher psychological functions. Recent studies reported that emotional contagion exists not only between humans but also among various animal species. The dog (Canis familiaris) is a unique animal and the oldest domesticated species. Dogs have coexisted with humans for more than 30,000 years and are woven into human society as partners bonding with humans. Dogs have acquired human-like communication skills and, likely as a result of the domestication process, the ability to read human emotions; therefore, it is feasible that there may be emotional contagion between human and dogs. However, the higher time-resolution of measurement of emotional contagion between them is yet to be conducted. We assessed the emotional reactions of dogs and humans by heart rate variability (HRV), which reflects emotion, under a psychological stress condition on the owners. The correlation coefficients of heart beat (R-R) intervals (RRI), the standard deviations of all RR intervals (SDNN), and the square root of the mean of the sum of the square of differences between adjacent RR intervals (RMSSD) between dogs and owners were positively correlated with the duration of dog ownership. Dogs’ sex also influenced the correlation coefficients of the RRI, SDNN, and RMSSD in the control condition; female showed stronger values. These results suggest that emotional contagion from owner to dog can occur especially in females and the time sharing the same environment is the key factor in inducing the efficacy of emotional contagion.

Deprivation of maternal care via lack of somatosensory input causes offspring to experience adverse consequences, especially in the central nervous system. However, little is known about the developmental effect of maternal care on peripheral tissues such as the skin, which includes cutaneous sensory neurons. In the present study, we examined the involvement of maternal care in the development of the skin. We investigated offspring reared by early-weaned mother mice who spontaneously showed lower frequency of licking/grooming on nursing. Offspring of early-weaned mothers showed higher resistance against skin barrier disruption than did offspring of normally-weaned mothers, and had normal skin barrier function in the intact trunk skin. In the dorsal root ganglion of early-weaned mother offspring, we also found up-regulation of mRNA levels of the Mas-related G-protein coupled receptor B4 (MrgprB4), which is a marker of sensory neurons that detect gentle stroking. We further found that levels of MrgprB4 mRNA were correlated with the enhancement of skin resistance. The present findings suggest that maternal somatosensory inputs have a developmental impact on the cutaneous sensory neurons of the skin in offspring. Interestingly, the present results suggest that lower maternal care has a benefit on the skin resistance. This provides important information for understanding the development of peripheral tissues in offspring reared under severe conditions such as lower maternal care in the wild.

Oxytocin appears to link social interaction and cell aging in rats, especially in females.

Dogs acquired unique cognitive abilities during domestication, which is thought to have contributed to the formation of the human-dog bond. In European breeds, but not in wolves, a dog’s gazing behavior plays an important role in affiliative interactions with humans and stimulates oxytocin secretion in both humans and dogs, which suggests that this interspecies oxytocin and gaze-mediated bonding was also acquired during domestication. In this study, we investigated whether Japanese breeds, which are classified as ancient breeds and are relatively close to wolves genetically, establish a bond with their owners through gazing behavior. The subject dogs were treated with either oxytocin or saline before the starting of the behavioral testing. We also evaluated physiological changes in the owners during mutual gazing by analyzing their heart rate variability (HRV) and subsequent urinary oxytocin levels in both dogs and their owners. We found that oxytocin treatment enhanced the gazing behavior of Japanese dogs and increased their owners’ urinary oxytocin levels, as was seen with European breeds; however, the measured durations of skin contact and proximity to their owners were relatively low. In the owners’ HRV readings, inter-beat (R-R) intervals (RRI), the standard deviation of normal to normal inter-beat (R-R) intervals (SDNN), and the root mean square of successive heartbeat interval differences (RMSSD) were lower when the dogs were treated with oxytocin compared with saline. Furthermore, the owners of female dogs showed lower SDNN than the owners of male dogs. These results suggest that the owners of female Japanese dogs exhibit more tension during interactions, and apart from gazing behavior, the dogs may show sex differences in their interactions with humans as well. They also suggest that Japanese dogs use eye-gazing as an attachment behavior toward humans similar to European breeds; however, there is a disparity between the dog sexes when it comes to the owners’ oxytocin secretion. Japanese dogs also showed different attachment behaviors from both European breeds and wolves, and they likely use additional strategies to substitute gaze when forming the human–dog bond.

To precisely understand how higher cognitive functions are implemented in the prefrontal network of the brain, optogenetic and pharmacogenetic methods to manipulate the signal transmission of a specific neural pathway are required. The application of these methods, however, has been mostly restricted to animals other than the primate, which is the best animal model to investigate higher cognitive functions. In this study, we used a double viral vector infection method in the prefrontal network of the macaque brain. This enabled us to express specific constructs into specific neurons that constitute a target pathway without use of germline genetic manipulation. The double-infection technique utilizes two different virus vectors in two monosynaptically connected areas. One is a vector which can locally infect cell bodies of projection neurons (local vector) and the other can retrogradely infect from axon terminals of the same projection neurons (retrograde vector). The retrograde vector incorporates the sequence which encodes Cre recombinase and the local vector incorporates the "Cre-On" FLEX double-floxed sequence in which a reporter protein (mCherry) was encoded. mCherry thus came to be expressed only in doubly infected projection neurons with these vectors. We applied this method to two macaque monkeys and targeted two different pathways in the prefrontal network: The pathway from the lateral prefrontal cortex to the caudate nucleus and the pathway from the lateral prefrontal cortex to the frontal eye field. As a result, mCherry-positive cells were observed in the lateral prefrontal cortex in all of the four injected hemispheres, indicating that the double virus vector transfection is workable in the prefrontal network of the macaque brain.

BACKGROUND: Sexual imprinting is important for kin recognition and for promoting outbreeding, and has been a driving force for evolution; however, little is known about sexual imprinting by auditory cues in mammals. Male mice emit song-like ultrasonic vocalizations that possess strain-specific characteristics. OBJECTIVES: In this study, we asked whether female mice imprint and prefer specific characteristics in male songs. METHODS AND FINDINGS: We used the two-choice test to determine the song preference of female C57BL/6 and BALB/c mice. By assessing the time engaged in searching behavior towards songs played back to females, we found that female mice displayed an innate preference for the songs of males from different strains. Moreover, this song preference was regulated by female reproductive status and by male sexual cues such as the pheromone ESP1. Finally, we revealed that this preference was reversed by cross-fostering and disappeared under fatherless conditions, indicating that the behavior was learned by exposure to the father's song. CONCLUSIONS: Our results suggest that female mice can discriminate among male song characteristics and prefer songs of mice from strains that are different from their parents, and that these preferences are based on their early social experiences. This is the first study in mammals to demonstrate that male songs contribute to kin recognition and mate choice by females, thus helping to avoid inbreeding and to facilitate offspring heterozygosity.

BACKGROUND: Vocal learning is a central functional constituent of human speech, and recent studies showing that adult male mice emit ultrasonic sound sequences characterized as "songs" have suggested that the ultrasonic courtship sounds of mice provide a mammalian model of vocal learning. OBJECTIVES: We tested whether mouse songs are learned, by examining the relative role of rearing environment in a cross-fostering experiment. METHODS AND FINDINGS: We found that C57BL/6 and BALB/c males emit a clearly different pattern of songs with different frequency and syllable compositions; C57BL/6 males showed a higher peak frequency of syllables, shorter intervals between syllables, and more upward frequency modulations with jumps, whereas BALB/c males produced more "chevron" and "harmonics" syllables. To establish the degree of environmental influences in mouse song development, sons of these two strains were cross-fostered to another strain of parents. Songs were recorded when these cross-fostered pups were fully developed and their songs were compared with those of male mice reared by the genetic parents. The cross-fostered animals sang songs with acoustic characteristics--including syllable interval, peak frequency, and modulation patterns--similar to those of their genetic parents. In addition their song elements retained sequential characteristics similar to those of their genetic parents' songs. CONCLUSION: These results do not support the hypothesis that mouse "song" is learned; we found no evidence for vocal learning of any sort under the conditions of this experiment. Our observation that the strain-specific character of the song profile persisted even after changing the developmental auditory environment suggests that the structure of these courtship sound sequences is under strong genetic control. Thus, the usefulness of mouse "song" as a model of mammalian vocal learning is limited, but mouse song has the potential to be an indispensable model to study genetic mechanisms for vocal patterning and behavioral sequences.

Male mice emit ultrasonic vocalizations (USVs) towards females during male-female interaction. It has been reported that USVs of adult male mice have the capability of attracting females. Although the waveform pattern of USVs is affected by genetic background, differences among strains with respect to USV and the effects of these differences on courtship behavior have not been analyzed fully. We analyzed USV patterns, as well as actual social behavior during USV recording, in 13 inbred mouse strains, which included laboratory and wild-derived strains. Significant effects of strain were observed for the frequency of USV emission, duration, and frequency of the waveform category. Principal component (PC) analysis showed that PC1 was related to frequency and duration, and PC2-4 were related to each waveform. In the comparison of USV patterns and behaviors among strains, wild-derived KJR mice displayed the highest scores for PC2-4, and female mice paired with KJR males did not emit rejection-related click sounds. It is assumed that the waveforms emitted by KJR males have a positive effect in male-female interaction. Therefore, we extracted waveforms in PC2-4 from the USV recordings of KJR mice to produce a sound file, "HIGH2-4". As a negative control, another sound file ("LOW2-4") was created by extracting waveforms in PC2-4 from strains with low scores for these components. In the playback experiments using these sound files, female mice were attracted to the speaker that played HIGH2-4 but not the speaker that played LOW2-4. These results highlight the role of strain differences in the waveforms of male USVs during male-female interaction. The results indicated that female mice use male USVs as information when selecting a suitable mate.

The sleep-wake cycle represents a crucial physiological process essential for maintaining homeostasis and promoting individual growth. In dogs, alterations in sleep patterns associated with age and dog's correlation with temperament factors, such as nervousness, have been reported, and there is an increasing demand for precise monitoring of sleep and physical activity in dogs. The present study aims to develop an analysis method for measuring sleep-wake patterns and physical activity in dogs by utilizing an accelerometer and a smartphone. By analyzing time series data collected from the accelerometer attached to the dog's collar, a comprehensive sleep and activity analysis model was constructed. This model classified the activity level into seven classes and effectively highlighted the variations in sleep-activity patterns. Two classes with lower activity levels were considered as sleep, while other five levels were regarded as wake based on the rate of occurrence. This protocol of data acquisition and analysis provides a methodology that enables accurate and extended evaluation of both sleep and physical activity in dogs. [ABSTRACT FROM AUTHOR]
