Abstract

The primary pre-neoplastic lesion of the lower esophagus in the vicinity of the gastroesophageal junction (GEJ) is any Barrett's esophageal lesions (BE), and esophageal neoplasia has increased in the US population with predispositions (Caucasian males, truncal obesity, age, and GERD). The responses to BE are endoscopic and screening cytologic programs with endoscopic ablation of various forms. The former have not been proven to be cost-effective and there are mixed results for eradication. A fresh approach is sorely needed. We prospectively followed 2229 mostly male veterans at high risk for colorectal cancer in a 27-year longitudinal long-term study, collecting data on colorectal neoplasia development and other preneoplastic lesions, including BE and spontaneous regression (SR). Another cross-sectional BE study at a similar time period investigated antigenic changes at the GEJ in both BE glandular and squamous mucosa immunohistochemistry and the role of inflammation. Ten of the prospective cohort (21.7%) experienced SR out of a total of forty-six BE patients. Significant differences between SR and stable BE were younger age (p < 0.007); lower platelet levels (p < 0.02); rectal p87 elevation in SR (p < 0.049); a reduced innate immune system (InImS) FEREFF ratio (ferritin: p87 colonic washings) (p < 0.04). Ancillary testing showed a broad range of neoplasia biomarkers. InImS markers may be susceptible to intervention using commonplace and safe medical interventions and encourage SR. Abstract

LPS activated macrophages require metabolic reprogramming and glucose uptake mediated by hypoxia inducible factor (HIF-1) α and glucose transporter (Glut)1 expression for proinflammatory cytokine production, especially IL-1β. This process is tightly regulated through activation of mitogen-activated protein kinases (MAP) kinases including the MEK/ERK pathway as well as several transcription factors including HIF-1α. While Mitogen-Activated Protein Kinase Kinase (MEK) 2 deficiency had no significant effect on nitric oxide, TNF-α, and IL-12 production in response to LPS challenge, MEK2 deficient murine bone marrow derived macrophages (BMDMs) exhibited lower IL-10 production. Importantly, MEK2 deficient BMDMs exhibited a preserved ERK1/2 phosphorylation, higher HIF-1α, Glut1 levels and substantially increased IL-1β as well as IL-6 production in response to LPS stimulation. Knockdown of HIF-1α expression via small interfering RNA (siRNA) decreased the level of HIF-1α expression in MEK2 deficient BMDMs and decreased IL-1β production in response to LPS treatment. Furthermore, we performed gain of function experiments by overexpressing MEK2 protein in RAW264.7 cells. LPS stimulation of MEK2 overexpressed in RAW264.7 cells led to a marked decreased IL-1β production. Finally, we investigated the role of Mek1 and Mek2 double and triple mutation on ERK phosphorylation, HIF-1α expression and IL-1β production. We found that MEK2 is the major kinase, which inversely proportionally regulates HIF-1α and IL-1β expression independent of ERK activation. Our findings demonstrate a novel regulatory function for MEK2 in response to TLR4 activation in IL-1β production through modulating HIF-1α expression.

Abstract

Long-term exposure to sunlight results in skin collagen and elastin damages that are believed to be the cause of wrinkles. It is shown that mere hours of exposure to solar ultraviolet irradiation can stimulate collagen- and elastin-damaging metalloproteinase messenger RNAs, proteins and activities in the human skin in vivo. If the resulting degradation of the extracellular matrix is not properly repaired, it will create solar scars that may eventually cause the skin to age prematurely.

45. Stay-green quantitative trait loci's effects on water extraction, transpiration efficiency and seed yield depend on recipient parent background

Abstract

A stay-green phenotype enhances the adaptation of sorghum (Sorghum bicolor (L.) Moench) to terminal drought, although the mechanisms leading to its expression remain unclear. Differences in tillering and leaf area at anthesis, transpiration efficiency (TE), water extraction, harvest index (HI) and yield under terminal drought and fully irrigated conditions were assessed in 29 introgression lines (IL) targeting stay-green quantitative trait loci (QTLs) Stg1, Stg2, Stg3, Stg4, StgA and StgB in an S35 background, and 16 IL targeting Stg1, Stg3, Stg4 and StgB in an R16 background. TE was increased by StgB in the R16 background, whereas there was no effect in the S35 background. Water extraction was increased by Stg1 in the S35 background but not in R16. StgB modified the proportion of water extracted before and after anthesis in the S35 background. While tillering and leaf area at anthesis were decreased by Stg1 and Stg3 in S35, there was no effect in R16. Yield data under fully irrigated conditions showed higher tiller grain yield in Stg1, Stg2 and Stg3 ILs. Although yield differences were mostly explained by HI variation, the yield variation unexplained by HI was closely related to TE in S35 (R2 = 0.29) and R16 (R2 = 0.72), and was closely related to total water extracted in S35 (R2 = 0.41) but not in R16. These data indicate the potential for several stay-green QTLs to affect traits related to plant water use. However, these effects depend on the interaction between the genetic background and individual QTLs.

Abstract

The BCL2 (B cell lymphoma/leukemia-2) proto-oncogene encodes a 26-kDa protein that has been localized to the inner mitochondrial membrane and that has been shown to enhance the survival of some types of hematopoietic cells. Here we show that NIH3T3 fibroblasts stably transfected with a BCL2 expression plasmid exhibit reduced dependence on competence-inducing growth factors (platelet-derived growth factor, PDGF; epidermal growth factor, EGF) for initiation of DNA synthesis. The importance of BCL2 for growth factorinduced proliferation of these cells was further confirmed by the useage of BCL2 antisense oligodeoxynucleotides. The mechanisms by which overexpression of p26 BCL2 contributes to fibroblast proliferation are unknown, but do not involve alterations in: (a) the production of inositol triphosphates (IP3), (b) PDGF-induced transient elevations in cytosolic Ca2+ ions, or (c) the activity of protein kinase C enzymes in these transfected cells. The results imply that changes in mitochondrial functions play an important role in the early stages of the cell cycle that render 3T3 cells competent to respond to the serum progression factors that stimulate entry into S-phase.
