Abstract

The TRPA1 ion channel is activated by electrophilic compounds through the covalent modification of intracellular cysteine residues. How non-covalent agonists activate the channel and whether covalent and non-covalent agonists elicit the same physiological responses are not understood. Here, we report the discovery of a non-covalent agonist, GNE551, and determine a cryo-EM structure of the TRPA1-GNE551 complex, revealing a distinct binding pocket and ligand-interaction mechanism. Unlike the covalent agonist allyl isothiocyanate, which elicits channel desensitization, tachyphylaxis, and transient pain, GNE551 activates TRPA1 into a distinct conducting state without desensitization and induces persistent pain. Furthermore, GNE551-evoked pain is relatively insensitive to antagonist treatment. Thus, we demonstrate the biased agonism of TRPA1, a finding that has important implications for the discovery of effective drugs tailored to different disease etiologies.

Abstract

Development corridors are linear programmes of infrastructure and agriculture aiming to facilitate rapid socio-economic development. In Africa, they are a major development activity, with 88 underway or planned corridors. Drawing from extensive literature and insights gleaned from a 4 year research programme, this review scrutinizes the impacts of development corridors on people, wildlife and ecosystems in Kenya and Tanzania, proposing solutions to achieve better outcomes. The overarching goal was to discern the principle challenges emerging from the practical execution of the prevailing corridor model. The holistic approach taken, assessing the development corridors paradigm through an integrated ecological, social, and economic lens, provides novel insights that have not been possible using more traditional—siloed—research approaches. Eight key challenge areas are identified: impact assessments processes; coherence across international, national and local planning; governance; inclusivity; equality; impacts on biodiversity and ecosystem services; incorporation of future climate risks; and integrated water resource management. Poorly planned and implemented corridors detrimentally impact livelihoods and ecosystems. They lack a sustainable development vision, detailed social, environmental or climate risk assessments, and develop incrementally in policy and corporate spaces. There is also often a disconnect between investors and recipient governments, with some investors funding what governments request without applying internationally-recognised safeguards, and governments lacking capacity and resources to enforce regulations. We make recommendations for addressing these challenge areas. 28. Mitigating the impacts of development corridors 1 on biodiversity: a global review

Abstract

Development corridors are extensive, often transnational and linear, geographical areas targeted for investment to help achieve sustainable development. They often comprise the creation of hard infrastructure (i.e., physical structures) and soft infrastructure (i.e., policies, plans, and programmes) involving a variety of actors. They are globally widespread, and likely to be a significant driver of habitat loss. Here, we describe the development corridors phenomenon from a biodiversity perspective and identify the elements of best practice in biodiversity impact mitigation. We use these to carry out a review of the peer reviewed literature on corridors to respond to three questions: (i) how impacts on biodiversity and ecosystem services are assessed; (ii) what mitigation measures are discussed to manage these impacts; and (iii) to what extent do these measures approximate to best practice. We found that of 271 publications on development corridors across all continents (except for Antarctica) mentioning biodiversity or ecosystem services, only 100 (37%) assessed impacts on biodiversity and 7 (3%) on ecosystem services. Importantly, only half of these (52, 19% of the total 271 articles) discussed mitigation measures to manage these impacts. These measures focused on avoidance and minimisation and there was scant mention of restoration or ecological compensation illustrating a deficient application of the mitigation hierarchy. We conclude that the academic literature on corridors does not give sufficient consideration to comprehensive mitigation of biodiversity impacts. To change this, impact assessment research needs to acknowledge the complexity of such multiproject and multi-stakeholder initiatives, quantify biodiversity losses due to the full suite of their potential direct, indirect and cumulative impacts, and follow all the steps of the mitigation hierarchy impact framework. We suggest a series of research avenues and policy recommendations to improve impact assessments of corridors towards achieving better biodiversity outcomes

We found that of 271 publications on development corridors across all continents (except for Antarctica) mentioning biodiversity or ecosystem services, only 100 (37%) assessed impacts on biodiversity and 7 (3%) on ecosystem services. Importantly, only half of these (52, 19% of the total 271 articles) discussed mitigation measures to manage these impacts. These measures focused on avoidance and minimisation and there was scant mention of restoration or ecological compensation illustrating a deficient application of the mitigation hierarchy. We conclude that the academic literature on corridors does not give sufficient consideration to comprehensive mitigation of biodiversity impacts. To change this, impact assessment research needs to acknowledge the complexity of such multi-project and multi-stakeholder initiatives, quantify biodiversity losses due to the full suite of their potential direct, indirect and cumulative impacts, and follow all the steps of the mitigation hierarchy impact framework. Abstract

Unlike classical voltage-gated sodium (NaV) channels, NaX has been characterized as a voltage-insensitive, tetrodotoxin-resistant, sodium (Na+)-activated channel involved in regulating Na+ homeostasis. However, NaX remains refractory to functional characterization in traditional heterologous systems. Here, to gain insight into its atypical physiology, we determine structures of the human NaX channel in complex with the auxiliary β3-subunit. NaX reveals structural alterations within the selectivity filter, voltage sensor-like domains, and pore module. We do not identify an extracellular Na+-sensor or any evidence for a Na+-based activation mechanism in NaX. Instead, the S6-gate remains closed, membrane lipids fill the central cavity, and the domain III-IV linker restricts S6-dilation. We use protein engineering to identify three pore-wetting mutations targeting the hydrophobic S6-gate that unlock a robust voltage-insensitive leak conductance. This constitutively active NaX-QTT channel construct is non-selective among monovalent cations, inhibited by extracellular calcium, and sensitive to classical NaV channel blockers, including tetrodotoxin. Our findings highlight a functional diversity across the NaV channel scaffold, reshape our understanding of NaX physiology, and provide a template to demystify recalcitrant ion channels. NaX is an atypical member of the voltage-gated sodium channel family that may contribute to Na+ homeostasis.
