Abstract

A molecular approach was developed to address the relationship between the diversity of ciliates in the plankton and those of the underlying benthos in the same locations. The clustering of phylogenetic types revealed that benthic assemblages of oligotrichs and choreotrichs seem to be more like those from spatially remote benthic communities than the ciliate communities.

Supplemental Digital Content is available in the text.Primary breast diffuse large B-cell lymphoma (PB-DLBCL) is a rare disease comprising <3% of extranodal lymphomas. It frequently reveals an activated B-cell (ABC)-like phenotype. ABC-like DLBCL was reported to have gain-of-function mutations in MYD88, CD79B, CARD11, and TNFAIP3, resulting in constitutive activation of the NFκB pathway. Because of the rare occurrence of PB-DLBCL, the frequency of MYD88and CD79Bmutations is still unknown. We used Sanger sequencing to study these mutations from 46 breast DLBCL cases and also investigated the associated clinicopathologic factors. MYD88L265P was confirmed by allele-specific polymerase chain reaction and compared with the Sanger sequencing results. MYD88L265P and CD79Bmutations were detected in 27/46 (58.7%) and 11/33 (33.3%) cases, respectively. Twenty-eight of 46 cases met the criteria for PB-DLBCL, and the latter 18 cases were further classified as clinical breast DLBCL (CLB-DLBCL). The frequency of MYD88L265P and CD79Bmutations was 16/28 (57.1%) and 9/23 (39.1%), respectively, in PB-DLBCL and 11/18 (61.1%) and 2/10 (20%), respectively, in CLB-DLBCL. When the cutoff value was set at ΔCt≤1, the result of allele-specific polymerase chain reaction for MYD88corresponded to those of the Sanger sequence at 92.6% sensitivity and 100% specificity. According to Choi’s algorithm, 16/27 (59.3%) demonstrated an ABC-like phenotype in PB-DLBCL, and 15/18 (83.3%) demonstrated an ABC-like phenotype in CLB-DLBCL. In conclusion, MYD88L265P and CD79Bmutations were frequently detected in PB-DLBCL, and they may be key molecules associated with PB-DLBCL lymphomagenesis. Further analysis will be required to clarify the mechanism of its pathogenesis.

Abstract

A new sand-dwelling dinoflagellate, Pileidinium ciceropse Tamura et Horiguchi gen. et sp. nov., is described from a sandy beach on Mecherchar Island, Republic of Palau, using light and electron microscopy. The cell is laterally compressed and consists of a small epitheca and a large hypotheca. It has a yellow-brown chloroplast and a spherical pyrenoid. This dinoflagellate possesses ornamented thecal plates and an unusual thecal plate arrangement of 1′, 5′′, 4c, 4s, 5′′′, 1′′′′. The cingulum is incomplete and does not encircle the entire cell. Transmission electron microscopy reveals that the cell possesses typical dinoflagellate organelles. Phylogenetic analysis based on SSU rDNA did not demonstrate a consistent phylogenetic position of Pileidinium ciceropse within the Dinophyceae and therefore affinities with existing families could not be determined. Abstract

A new sand-dwelling dinoflagellate from Palau,Galeidinium rugatumTamura et Horiguchi gen. et sp. nov., is described. The life cycle of this new alga consists of a dominant nonmotile phase and a brief motile phase. The motile cell transforms itself directly into the nonmotile cell after swimming for a short period, and cell division takes place in the nonmotile phase. The nonmotile cell possesses a dome-like cell covering, which is wrinkled and equipped with a transverse groove on the surface. The cell has 10–20 chloroplasts and a distinct eyespot. The motile cell isGymnodinium-like in shape. The dinoflagellate possesses an endosymbiotic alga to which the chloroplasts belong and which is separated from the host (dinoflagellate) cytoplasm by a unit membrane. The endosymbiont cytoplasm also possesses its own eukaryotic nucleus and mitochondria. The eyespot is surrounded by triple membranes and is located in the host cytoplasm. Photosynthetic pigment analysis, using HPLC, revealed thatG. rugatumpossesses fucoxanthin as the principal accessory pigment instead of peridinin. TherbcL tree showed thatG. rugatumis monophyletic withDurinskia baltica(Levander) Carty et Cox andKryptoperidinium foliaceum(Stein) Lindemann and that this clade is closely related to the pennate diatom,Cylindrothecasp. The endosymbiont ofG. rugatumis therefore shown to be a diatom. Phylogenetic analysis based on small subunit rDNA sequences demonstrated thatG. rugatum,D. baltica, andK. foliaceum, all of which are known to harbor an endosymbiont of diatom origin, are closely related.
