Abstract

The inhibitory effect of growth hormone (GH) on adipose tissue growth is well known, but the underlying mechanism is not fully understood. In this study, we determined the possibility that GH inhibits adipose tissue growth by inhibiting adipogenesis, the process of formation of adipocytes from stem cells, in the lit/lit mice. The lit/lit mice are GH deficient because of a spontaneous mutation to the GH releasing hormone receptor (ghrhr) gene, and they have more subcutaneous fat despite being smaller than the lit/+ mice at the same age. We found that cells of the stromal vascular fraction (SVF) of subcutaneous fat from the lit/lit mice had greater adipogenic potential than those from the lit/+ mice, as evidenced by forming greater numbers of lipid droplets-containing adipocytes and having greater expression of adipocyte marker genes during induced adipocyte differentiation in culture. However, addition of GH to the culture did not reverse the superior adipogenic potential of subcutaneous SVF from the lit/lit mice. Through florescence-activated cell sorting and quantification of mRNAs of preadipocyte markers, including CD34, CD29, Sca-1, CD24, Pref-1, and PPAR gamma, we found that subcutaneous SVF from the lit/lit mice contained more preadipocytes than that from the lit/+ mice. These results support the notion that GH inhibits adipose tissue growth in mice at least in part by inhibiting adipogenesis. Furthermore, these results suggest that GH inhibits adipogenesis in mice not by inhibiting the terminal differentiation of preadipocytes into adipocytes, rather by inhibiting the formation of preadipocytes from stem cells or the recruitment of stem cells to the fat depot. 15. Shifted frequency‐based electromagnetic transient simulation for AC power systems in symmetrical component domain.

Abstract

Power systems are becoming more and more complicated. Traditional simulation tools such as transient stability programs and electromagnetic transients (EMT) programs are unable to meet the accuracy or efficiency requirements. This paper proposes a shifted frequency‐based EMT (SFEMT) simulation in the symmetrical component‐domain (abbreviated as SCSFEMT simulation) for the three‐phase transmission system. The sequence‐decoupled SCSFEMT modeling for the power system components is first presented based on the symmetrical component transformation. Then, the solution procedure of the presented SCSFEMT simulation algorithm is elaborated in detail based on the EMTP‐type solution and multi‐sequence Thevenin equivalence (MSTE). With the MSTE, the three sequence networks can be decoupled during the whole simulation. Thus, the dimension of the system is greatly reduced. Last, the accuracy and efficiency of the SCSFEMT simulation are validated on the 10‐machine 39‐bus New England system by comparing it with traditional EMT and SFEMT simulation. Test results show that the SCSFEMT simulation algorithm is much more efficient than traditional SFEMT simulation. [ABSTRACT FROM AUTHOR]

22. Accuracy Enhancement of Shifted Frequency-Based Simulation Using Root Matching and Embedded Small-Step

Abstract

Electromagnetic transients (EMT) simulation is fundamental in power system design and operation. To improve the computational efficiency of EMT simulation, the shifted frequency-based EMT (SFEMT) simulation with large time-steps has been proposed in the literature recently. Nevertheless, the existing SFEMT simulation methods with a large time-step have accuracy issues when there is a sudden change in the power system. In this paper, the causes of the poor accuracy of traditional SFEMT simulation in sudden-change scenarios are first analyzed. Then, it proposes a highly accurate SFEMT simulation algorithm based on the root matching (RM) and embedded small-step (ESS) techniques. Next, the RM-based SFEMT (RM-SFEMT) models of the power system components are derived and the flowchart of RM-SFEMT simulation with ESS is presented. Test results on three power systems of different scales demonstrate the improved accuracy of the proposed SFEMT simulation algorithm.

Abstract

A series of single-phase Mg x Zn 1 − x O films with different Mg contents were prepared on quartz substrates by RF magnetron sputtering technique using different MgZnO targets, and annealed under the atmospheric environment. The absorption edges of Mg x Zn 1 − x O films can cover the whole near ultraviolet and even the whole solar-blind spectra range, and the solar-blind wurtzite/cubic Mg x Zn 1 − x O films have been realized successfully by the same method. In addition, the absorption edges of annealed films shift to a long wavelength, which is caused by the diffusion of Zn atoms gathering at the surface during the thermal treatment process. Finally, the truly solar-blind metal-semiconductor-metal structured photodetectors based on wurtzite Mg 0.445 Zn 0.555 O and cubic Mg 0.728 Zn 0.272 O films were fabricated. The corresponding peak responsivities are 17 mA/W at 275 nm and 0.53 mA/W at 250 nm under a 120 V bias, respectively. [ABSTRACT FROM AUTHOR]

39. Coat colour phenotype of Qingyu pig is associated with polymorphisms of melanocortin receptor 1 gene.

Abstract

Objective: Qingyu pig, a Chinese indigenous pig breed, exhibits two types of coat colour phenotypes, including pure black and white with black spotting respectively. Melanocortin receptor 1 ( MC1R ) and agouti signaling protein ( ASIP ) are two widely reported pivotal genes that significantly affect the regulation of coat colour. The objectives of this study were to investigate whether the polymorphisms of these two genes are associated with coat colour and analyze the molecular mechanism of the coat colour separation in Qingyu pig., Methods: We studied the phenotype segregation and used polymerase chain reaction amplification and Sanger sequencing to investigate the polymorphism of MC1R and ASIP in 121 Qingyu pigs, consisting of 115 black and 6 white with black spotted pigs., Results: Coat colour of Qingyu pig is associated with the polymorphisms of MC1R but not ASIP . We only found 2 haplotypes, E QY and E qy , based on the 13 observed mutations from MC1R gene. Among which, E qy presented a recessive inheritance mode in black spotted Qingyu pigs. Further analysis revealed a g.462-463CC insertion that caused a frameshift mutation and a premature stop codon, thus changed the first transmembrane domain completely and lost the remaining six transmembrane domains. Altogether, our results strongly support that the variety of Qingyu pig's coat colour is related to MC1R ., Conclusion: Our findings indicated that black coat colour in Qingyu pig was dominant to white with black spotted phenotype and MC1R gene polymorphism was associated with coat colour separation in Qingyu pig.
