16. Metabolic Analysis of Three-Dimensional Cultured Gastrointestinal Cancer Cells Suggests that l-Arginine Inhibits Tumor Growth by Affecting the Urea Cycle.

Abstract

Introduction: There is evidence for the anticancer effects of l-arginine (arginine); however, the direct effects on cancer cells and mechanism of action are unclear. Methods: Various upper gastrointestinal cancer cells (OE19, OE33, MKN1, MKN45, MKN74, and AGS) were divided into arginine-treated and -untreated groups and cultured using two-dimensional and three-dimensional culture systems. Proliferation was evaluated using the MTT assay to identify arginine-sensitive (OE33) and arginine-insensitive (OE19) strains. Furthermore, the effects of arginine were evaluated using a mitochondrial stress test, cell cycle assay, comprehensive metabolic analysis, and tracer study using (13C6) l-arginine. Results: In OE33 (but not in OE19), the maximal respiratory capacity of mitochondria was lower in the treated group than in the control group. In OE33, S phase cells (determined using BrdU) were significantly reduced. In a comprehensive metabolic analysis of OE33, citrulline/ornithine levels were significantly lower in arginine-treated than in untreated cells. Using OE33, carbamoyl aspartic acid (CAA) levels were significantly lower in arginine-treated than in untreated cells. A tracer study suggested that arginine promotes the urea cycle. Conclusion: Arginine affected urea cycle metabolism, thereby decreasing CAA, which is required for pyrimidine nucleotide synthesis. Abstract

To reduce CO2 emissions and save grey energy, natural materials like wood and wooden materials are becoming more and more important. However, these products are particularly sensitive to moisture, as they can be attacked by mould or decay fungi. In contrast to mould growth, which typically is associated with visual impairment and health problems, the growth of decay fungi may result in structural defects which clearly must be excluded. Up to now it is mostly assumed that wooden materials are more sensitive to such attack than solid wood. Therefore, different wood fibre insulation materials were inoculated with decay fungi and exposed to different climates to determine the requirements for the decay process and to compare them with the requirements of decay by the same fungi of solid wood. The results prove that some natural fibre materials are equally or even more resistant to decay fungi than solid wood, while others are less. The resistant products can therefore be assessed like solid wood – for which already temperature dependent thresholds and in part also transient decay prediction models are available. Maybe even specific higher moisture levels can be acceptable. However, the results also suggest a differentiated view on natural fibre insulations, as they have a very different susceptibility to wood decay. Uniform and significantly lower limits than for solid wood are not justified.

Abstract

Drought severely damages crop production, even under conditions so mild that the leaves show no signs of wilting. However, it is unclear how field-grown plants respond to mild drought. Here, we show through six years of field trials that ridges are a useful experimental tool to mimic mild drought stress in the field. Mild drought reduces inorganic phosphate levels in the leaves to activate the phosphate starvation response (PSR) in soybean plants in the field. Using Arabidopsis thaliana and its mutant plants grown in pots under controlled environments, we demonstrate that PSR occurs before abscisic acid response under progressive mild drought and that PSR plays a crucial role in plant growth under mild drought. Our observations in the field and laboratory using model crop and experimental plants provide insight into the molecular response to mild drought in field-grown plants and the relationship between nutrition and drought stress response.

Abstract

DNA metabarcoding on a single organism is a promising approach to clarify the biological interactions (e.g., predator–prey relationships and symbiosis, including parasitism) of difficult‐to‐culture protists. To evaluate the effectiveness of this method, Radiolaria and Phaeodaria, which are ecologically important protistan groups, were chosen as target taxa. DNA metabarcoding on a single organism focused on the V9 region of the 18S rRNA gene revealed potential symbionts, parasites and food sources of Radiolaria and Phaeodaria. Previously reported hosts and symbionts (parasites) were detected, and newly recognized combinations were also identified. The contained organisms largely differed between Radiolaria and Phaeodaria. In Radiolaria, members of the same order tended to contain similar organisms, and the taxonomic composition of possible symbionts, parasites, and food sources was fixed at the species level. Members of the same phaeodarian family, however, did not contain similar organisms, and body part (i.e., the central capsule or the phaeodium) was the most important factor that divided the taxonomic composition of detected organisms, implying that the selection of appropriate body part is important when trying to ascertain contained organisms, even for unicellular zooplankton.
