Titanium carbide is used as reinforcement particles in composites due to its hardness, wear resistance and stability. This work is a part of the study in which titanium carbides are formed in stainless steel castings in the mold to improve the wear resistance of a certain surface of the casting. Such local reinforcement is a very potential method but it is a quite demanding task requiring profound knowledge of interfacial phenomena in the system, wettability, stability, dissolution and precipitation of new phases in production of these materials. Good wetting between different constituents in the material is a key factor to attain maximal positive effects. Mo is used with TiC or Ti(C,N) reinforcement in composites to improve wettability. In this work the effect of Mo on the phenomena in Fe/stainless steel–TiC systems was examined by wetting experiments between the substrate and the alloy. Wetting was not significantly improved by adding Mo to the systems. Core-rim type carbides as well as more homogenous carbide particles were observed. Overall the carbide particles are very complex regarding to their chemistry, size and shape which aspects have to be taken into account in the development of these materials and manufacturing processes.

The authors have investigated metastable phase separation in multi-component silicate glass for the fabrication of porous glass from multi-component slag. Spinodal decomposition forms an interconnected microstructure in glass spontaneously, and porous glass is obtained by leaching one of the decomposed phases with an acid solution. This porous glass can be used for a filter to remove impurities in polluted water or air. In this study, the metastable miscibility gap was predicted in multi-component silicate glass using thermodynamic analyses where glass was regarded as a super-cooled liquid phase. Occurrence of spinodal decomposition was observed in annealed glass, and it corresponded to the predicted miscibility gap. Then, we fabricated porous glass using spinodal decomposition in multi-component borosilicate glass and by removing one of the decomposed phases. Furthermore, for the creation of functional porous glass applicable for environmental purification, the spinodal decomposition was prepared in multi-component borosilicate glass containing titanium oxide based on the predicted metastable miscibility gap in multi-component silicate glass.

This work is part of the study in which Ti and C are added in the mould during casting of stainless steel to form TiC at a specific surface. Wettability, stability and dissolution of the reinforcement particles are key phenomena in producing these composites in liquid state. Wetting experiments between TiC and Fe as well as FeNi and FeCr alloys were performed in order to get information about wettability, dissolution and formation of TiC and other phases related to the systems. Contact angles for the systems were attained without any reactions between the substrate and the alloy during heating. Wetting was not significantly improved by adding Ni or Cr. Dissolution of TiC was very strong into all the alloys. No new phases at the interface were formed in the experiments except for Cr-carbides in Cr-containing alloys. These results gave important information about the interfacial phenomena needed in developing the production methods.

Electromotive force method with solid electrolyte is briefly explained, and a thermodynamic study of semi conducting compound solid solution ZnTe-CdTe is picked up to show the way how thermodynamic functions of this system are obtained by several experimental methods based on our published papers and recently published data are added for comparison.

Abstract Background The strong linkage disequilibrium (LD) recently found in genic or exonic regions of the human genome demonstrated that LD can be increased by evolutionary mechanisms that select for functionally important loci. This suggests that LD might be stronger in regions conserved among species than in non-conserved regions, since regions exposed to natural selection tend to be conserved. To assess this hypothesis, we used genome-wide polymorphism data from the HapMap project and investigated LD within DNA sequences conserved between the human and mouse genomes. Results Unexpectedly, we observed that LD was significantly weaker in conserved regions than in non-conserved regions. To investigate why, we examined sequence features that may distort the relationship between LD and conserved regions. We found that interspersed repeats, and not other sequence features, were associated with the weak LD tendency in conserved regions. To appropriately understand the relationship between LD and conserved regions, we removed the effect of repetitive elements and found that the high degree of sequence conservation was strongly associated with strong LD in coding regions but not with that in non-coding regions. Conclusion Our work demonstrates that the degree of sequence conservation does not simply increase LD as predicted by the hypothesis. Rather, it implies that purifying selection changes the polymorphic patterns of coding sequences but has little influence on the patterns of functional units such as regulatory elements present in non-coding regions, since the former are generally restricted by the constraint of maintaining a functional protein product across multiple exons while the latter may exist more as individually isolated units.

Kawasaki disease (KD) is a pediatric vasculitis caused by an unknown trigger in genetically susceptible children. The incidence varies widely across genetically diverse populations. Several associations with HLA Class I alleles have been reported in single cohort studies. Using a genetic approach, from the nine single nucleotide variants (SNVs) associated with KD susceptibility in children of European descent, we identified SNVs near the HLA-C (rs6906846) and HLA-B genes (rs2254556) whose association was replicated in a Japanese descent cohort (rs6906846 p = 0.01, rs2254556 p = 0.005). The risk allele (A at rs6906846) was also associated with HLA-C*07:02 and HLA-C*04:01 in both US multi-ethnic and Japanese cohorts and HLA-C*12:02 only in the Japanese cohort. The risk A-allele was associated with eight non-conservative amino acid substitutions (amino acid positions); Asp or Ser (9), Arg (14), Ala (49), Ala (73), Ala (90), Arg (97), Phe or Ser (99), and Phe or Ser (116) in the HLA-C peptide binding groove that binds peptides for presentation to cytotoxic T cells (CTL). This raises the possibility of increased affinity to a "KD peptide" that contributes to the vasculitis of KD in genetically susceptible children.

A major problem in autoimmunity has been identification of the earliest events that lead to breach of tolerance. Although there have been major advances in dissecting effector pathways and the multilineage immune responses to mitochondrial self-antigens in primary biliary cholangitis, the critical links between environmental factors and tolerance remain elusive. We hypothesized that environmental xenobiotic modification of the E2 subunit of the pyruvate dehydrogenase (PDC-E2) inner lipoyl domain can lead to loss of tolerance to genetically susceptible hosts. Previously we demonstrated that serum anti-PDC-E2 autoantibodies cross-react with the chemical xenobiotics 2-octynoic acid and 6,8-bis (acetylthio) octanoic acid and further that there is a high frequency of PDC-E2-specific peripheral plasmablasts. Herein we generated 104 recombinant monoclonal antibodies (mAbs) based on paired heavy-chain and light-chain variable regions of individual plasmablasts derived from primary biliary cholangitis patients. We identified 32 mAbs reactive with native PDC-E2, including 20 specific for PDC-E2 and 12 cross-reactive with both PDC-E2 and 2-octynoic acid and 6,8-bis (acetylthio) octanoic acid. A lower frequency of replacement somatic hypermutations, indicating a lower level of affinity maturation, was observed in the complementarity-determining regions of the cross-reactive mAbs in comparison to mAbs exclusively recognizing PDC-E2 or those for irrelevant antigens. In particular, when the highly mutated heavy-chain gene of a cross-reactive mAb was reverted to the germline sequence, the PDC-E2 reactivity was reduced dramatically, whereas the xenobiotic reactivity was retained. Importantly, cross-reactive mAbs also recognized lipoic acid, a mitochondrial fatty acid that is covalently bound to PDC-E2.ConclusionOur data reflect that chemically modified lipoic acid or lipoic acid itself, through molecular mimicry, is the initial target that leads to the development of primary biliary cholangitis. Large-scale meta-analyses of genome-wide association studies (GWAS) have identified >175 loci associated with fasting cholesterol levels, including total cholesterol (TC), high-density lipoprotein cholesterol (HDL-C), low-density lipoprotein cholesterol (LDL-C), and triglycerides (TG).
