Background: The apicomplexan parasites Eimeria spp. are the causative agents of coccidiosis, a disease with a significant global impact on the poultry industry. The complex life cycle of Eimeria spp. involves exogenous (sporogony) and endogenous (schizogony and gametogony) stages. Unfortunately, the genetic regulation of these highly dynamic processes, particularly for genes involved in specific developmental phases, is not well understood. Methods: In this study, we used RNA sequencing (RNA-Seq) analysis to identify expressed genes and differentially expressed genes (DEGs) at seven time points representing different developmental stages of Eimeria tenella. We then performed K-means clustering along with co-expression analysis to identify functionally enriched gene clusters. Additionally, we predicted apicomplexan AP2 transcription factors in E. tenella using bioinformatics methods. Finally, we generated overexpression and knockout strains of ETH2_0411800 to observe its impact on E. tenella development. Results: In total, we identified 7329 genes that are expressed during various developmental stages, with 3342 genes exhibiting differential expression during development. Using K-means clustering along with co-expression analysis, we identified clusters functionally enriched for oocyte meiosis, cell cycle, and signaling pathway. Among the 53 predicted ApiAP2 transcription factors, ETH2_0411800 was found to be exclusively expressed during sporogony. The ETH2_0411800 overexpression and knockout strains did not exhibit significant differences in oocyst size or output compared to the parental strain, while the resulting ETH2_0411800 knockout parasite showed a relatively small oocyst output. Conclusions: The findings of our research suggest that ETH2_0411800 is not essential for the growth and development of E. tenella. Our study provides insights into the gene expression dynamics and is a valuable resource for exploring the roles of transcription factor genes in regulating the development of Eimeria parasites. As an important means of improving positioning accuracy, block adjustment has been used in the improvement and assessment of accuracy for the Chinese Gaofen-7 (GF-7) satellite. However, there is little research on what factors affect accuracy without ground control points (GCPs). The correlation between accuracy and the images participating in the adjustment is not clear. This paper proposes the correlation coefficients and canonical correlation analysis between five accuracy indicators and three sets of ten adjustment factors, including topographic factors, participating image factors, and tie points (TPs) factors, to quantify the influence of adjustment factors on accuracy. Block adjustment without GCPs for GF-7 stereo imagery is verified in three study areas to evaluate the relationship between accuracy and adjustment factors. The results show that block adjustment without GCPs can improve direct positioning accuracy with an average improvement of 1.27 m in the planar direction and 0.13 m in the elevation direction. Moreover, plane accuracy is more easily affected by three sets of factors, while the influence on elevation accuracy is more balanced. The set of TP factors has the greatest influence on accuracy, and the image overlap is more critical than the image coverage area, number, and time periods. Topographic factors also play an important role, and the influence of the elevation factor with the highest canonical correlation coefficient (−0.71) is more significant than the other two factors, roughness, and slope. The results provide a reference for the improvement of adjustment accuracy without GCPs, the reasonable selection of adjustment images, the optimization of TPs, and the strategy of the partition processing of large-area block adjustment for GF-7 stereo imagery. At present, some mapping satellites, such as GaoFen-7 and ZiYuan3-03, are equipped with both optical stereo cameras and laser altimeters that can synchronously obtain stereo images and sparse ground laser altimetry points (LAPs). To effectively improve the geometric accuracy of these satellite stereo images, this study proposed an integrated processing method for LAPs and stereo images derived from the same satellite. This method makes full use of the advantages of synchronously obtaining stereo images and LAPs, and designs measurement technology for accurate pixel coordinates of LAPs in stereo images, which works toward solving a technical difficulty that has restricted their integration to achieve higher accuracy. The method also constructs a combined block adjustment model of LAPs and stereo images. We selected 70 GaoFen-7 stereo images and 463 GaoFen-7 LAPs from Hebei Province, China, and 12 ZiYuan3-03 stereo images and 81 ZiYuan3-03 LAPs from Heilongjiang Province, China, to conduct integrated processing experiments. The vertical accuracy of the GaoFen-7 images in all types of terrain were improved substantially and reached the accuracy requirements of 1:10,000 (even 1:5000) scale mapping in China. The vertical accuracy of the ZiYuan3-03 images in various terrain areas were also improved markedly, satisfying the accuracy requirement of 1:50,000 scale mapping. These experimental results indicate that the working mode of synchronously obtaining LAPs and stereo images using the same satellite is advanced, and the proposed method is correct and effective. To meet the demands of natural resource monitoring, land development supervision, and other applications for high-precision and high-frequency information extraction from constructed land change, this paper focused on automatic feature extraction and data processing optimization methods for newly constructed bare land based on remote sensing images. A generalized deep convolutional neural network change detection model framework integrating multi-scale information was developed for the automatic extraction of change information. To resolve the problems in the automatic extraction of new bare land parcels, such as mis-extractions and parcel fragmentation, a proximity evaluation model that integrates the confidence-based semantic distance and spatial distance between parcels and their overlapping area is proposed to perform parcel aggregation. Additionally, we propose a complete set of optimized processing techniques from pixel pre-processing to vector post-processing. The results demonstrated that the aggregation method developed in this study is more targeted and effective than ArcGIS for the automatically extracted land change parcels. Additionally, compared with the initial parcels, the total number of optimized parcels decreased by more than 50% and the false detection rate decreased by approximately 30%. These results indicate that this method can markedly reduce the overall data volume and false detection rate of automatically extracted parcels through post-processing under certain conditions of the model and samples and provide technical support for applying the results of automatic feature extraction in engineering practices. Very-high-resolution (VHR) remote sensing images contain various multiscale objects, such as large-scale buildings and small-scale cars. However, these multiscale objects cannot be considered simultaneously in the widely used backbones with a large downsampling factor (e.g., VGG-like and ResNet-like), resulting in the appearance of various context aggregation approaches, such as fusing low-level features and attention-based modules. To alleviate this problem caused by backbones with a large downsampling factor, we propose a feature-selection high-resolution network (FSHRNet) based on an observation: if the features maintain high resolution throughout the network, a high precision segmentation result can be obtained by only using a $1\times $ 1 convolution layer with no need for complex context aggregation modules. Specifically, the backbone of FSHRNet is a multibranch structure similar to HRNet where the high-resolution branch is the principal line. Then, a lightweight dynamic weight module, named the feature-selection convolution (FSConv) layer, is presented to fuse multiresolution features, allowing adaptively feature selection based on the characteristic of objects. Finally, a specially designed $1\times $ 1 convolution layer derived from hypersphere embedding is used to produce the segmentation result. Experiments with other widely used methods show that the proposed FSHRNet obtains competitive performance on the ISPRS Vaihingen dataset, the ISPRS Potsdam dataset, and the iSAID dataset. The GF-7 satellite is China's first civil sub-meter resolution stereo mapping satellite, aiming at 1:10,000-scale mapping. To achieve this goal, apart from the stereo optical cameras that reach sub-meter resolution, the GF-7 satellite is equipped with a laser altimetry system capable of obtaining three-dimensional laser points (LPs) with high elevation accuracy. However, the combination of laser altimetry data and optical stereo images has not been thoroughly studied. In this paper, we exploit the images recorded by the highly integrated laser footprint cameras and propose a hierarchical phase correlation method based on a geographic pyramid for the registration of laser altimetry data and high-resolution optical stereo images, which lays a solid foundation for the following combined adjustment. Experiments show that the proposed registration method can automatically locate the LPs on high-resolution stereo images and meet the requirements of bundle adjustment. A series of bundle adjustment experiments were carried out, showing that laser altimetry data can significantly enhance the vertical accuracy of optical image stereo mapping and that elevation accuracy can reach roughly 1.0 m (RSME) without ground control points. In November 2019, the GaoFen‐7(GF‐7) satellite was equipped with China's first laser altimeter with full waveform recording capability, which obtains high‐precision long‐range three‐dimensional coordinates. The influence of clouds is noticeable for laser transmission, and a footprint camera is used to determine laser pointing and to image the ground. However, the cloud inevitably appears in the laser footprint image. In this study, the authors propose a cloud detection scheme for footprint images based on deep learning. First, an adaptive pooling model is proposed according to the characteristics of the cloud region. Next, model fusion was performed based on the SegNet and U‐Net training results. Finally, test time augmentation was used to enhance the data and to improve cloud detection accuracy. The experimental results show that the fusion result of the model was approximately 5% better than that of the traditional cloud detection algorithm, which improved the shortcomings of the traditional algorithm, such as poor detection effect for thin clouds and complex underlying cloud surfaces. Very high resolution (VHR) satellite and aerial images often suffer from scene occlusion caused by redundant objects. The task of removing these redundant objects can be solved by missing data reconstruction technology. However, when dealing with VHR images with large-scale missing regions, existing spatial-based methods often destroy the structural information of ground objects. To alleviate this problem, this paper proposes a novel missing data reconstruction method based on deep learning. The reconstruction process is divided into two parts: structure prediction and texture generation. First, a progressive edge generation network (PEGN) is designed to predict the edges of objects in missing regions in a progressive manner. Then, the edge map predicted by PEGN is input to a texture generation network (TGN) as structural information to produce the reconstruction results. This is a spatial-based method that can produce realistic and reasonable results without any need for auxiliary spectral or temporal data. Experiments demonstrate that our model can better restore the structure of ground objects in VHR images than other spatial-based methods and outperform them in SSIM and PSNR indices. The main objective of this study is to develop a robust soil moisture retrieval approach using multi-sensor remote sensing data. Firstly, the water cloud model was employed to eliminate the vegetation effects on SAR observations over vegetated areas, thus to obtain the bare soil backscatter associated with soil moisture. Then, against the underdetermined system for soil moisture retrieval, the advanced integral equation model and calibrated integral equation model were integrated to construct soil moisture retrieval scheme in combination with multi-sensor SAR observations. Through the above processing the influences of vegetation and surface roughness can be minimized in the developed soil moisture retrieval scheme. Finally, the least square based iteration approach was applied to derive soil moisture with multi-sensor SAR observations as inputs. Based on the field measurements and multi-sensor SAR data (C-band and X-band). acquired on June 2015 and October 2015 over Hebei agricultural areas, quantitative evaluation of the developed approach was implemented. The results indicate that accurate soil moisture was obtained by the developed approach over corn covered area and bare agricultural area. This article proposes a digital elevation model (DEM) generation approach using the Shuttle Radar Topography Mission (SRTM) DEM as the elevation constraint without ground control points. First, during the process of image block adjustment, we took advantage of the relatively high vertical accuracy of the SRTM-DEM in flat terrain regions and applied effective constraints on the object-space elevation-corrected value of tie points using the SRTM-DEM, achieving improved vertical accuracy for large-scale block adjustments. Subsequently, for the DEM matching process, multiple two-linear array stereo image pairs were obtained from along-track and across-track images with different look angles over the same region after the block adjustment. Then, the matching result of each stereo image pair underwent weighted fusion, before being used to generate the final DEM product. This approach can effectively enhance the matching quality and grid density of the final DEM product. The DEM generation experiment, using Ziyuan-3 images covering 186,000 km2 of Hubei Province, China, showed that the matching quality of the 10 m grid DEM was excellent. The vertical root mean square errors were 1.5 m in the flat regions and 2.96 m in the mountainous regions, thus achieving China’s 1:25,000 scale specification requirement for DEM products. Abstract: ZY3‐02 is the first Chinese earth‐observation satellite equipped with a laser altimeter, designed to assist stereo‐image mapping elevations. Launch vibrations, the space environment and other factors, both expected and unexpected, cause bias in the designed ranging and pointing of the laser altimeter, influencing its height measurement accuracy. This paper proposes an in‐orbit geometric calibration model which considers this bias as unknown systematic errors. The unknown parameters were calibrated by the minimum ranging error principle using laser spot geolocation data captured by a ground‐based laser detector array. Based on precise orbit and attitude data, the precise geolocation information of the laser footprints could be obtained. The objective of this research is to optimize the Alpha approximation model for soil moisture retrieval using multitemporal SAR data. The Alpha model requires prior knowledge of soil moisture range to constrain soil moisture estimation. The solution of the Alpha model is an undetermined problem due to the fact that the number of observation equations is less than the number of unknown parameters. This research primarily focused on the optimization of Alpha model by employing multisensor and multitemporal SAR data. The disadvantage of the Alpha model can be eliminated by the combination of multisensor SAR data. The optimized Alpha model was evaluated on the basis of a comprehensive campaign for soil moisture retrieval, which acquired multisensor time series SAR data and coincident field measurements. The agreement between the estimated and measured soil moisture was within a root mean square error of 0.08 cm3/cm3 for both methods. The optimized Alpha model shows an obvious improvement for soil moisture retrieval. The results demonstrated that multisensor and multitemporal SAR data are favorable for time series soil moisture retrieval over bare agricultural areas. [ABSTRACT FROM AUTHOR]

Abstract: This paper proposes a stereo‐image block‐adjustment algorithm without ground control points (GCPs) by adopting a Shuttle Radar Topography Mission (SRTM) digital elevation model (DEM) as a height assistant. Using dense, evenly distributed tie points on images used in the block adjustment, the initial height values of tie points are derived directly from the SRTM DEM. In the subsequent block‐adjustment iterative computation, the height correction value for tie points is rigorously restricted by the SRTM DEM elevation. A total of 509 ZY‐3 satellite stereo‐images across 190 000 km2 of China's Hubei Province were used for this block‐adjustment experiment. The results show that the vertical root mean square errors (RMSEs) are improved from 7·2 to 2·3 m without GCPs, while the RMSEs in flat and mountainous terrain are improved to 1·5 and 2·8 m, respectively, fulfilling the vertical accuracy requirement of China's 1:25 000 scale mapping. There are many problems in existing reconstruction-based super-resolution algorithms, such as the lack of texture-feature representation and of high-frequency details. Multi-scale detail enhancement can produce more texture information and high-frequency information. Therefore, super-resolution reconstruction of remote-sensing images based on adaptive multi-scale detail enhancement (AMDE-SR) is proposed in this paper. First, the information entropy of each remote-sensing image is calculated, and the image with the maximum entropy value is regarded as the reference image. Subsequently, spatio-temporal remote-sensing images are processed using phase normalization, which is to reduce the time phase difference of image data and enhance the complementarity of information. The multi-scale image information is then decomposed using the L0 gradient minimization model, and the non-redundant information is processed by difference calculation and expanding non-redundant layers and the redundant layer by the iterative back-projection (IBP) technique. The different-scale non-redundant information is adaptive-weighted and fused using cross-entropy. Finally, a nonlinear texture-detail-enhancement function is built to improve the scope of small details, and the peak signal-to-noise ratio (PSNR) is used as an iterative constraint. Ultimately, high-resolution remote-sensing images with abundant texture information are obtained by iterative optimization. Real results show an average gain in entropy of up to 0.42 dB for an up-scaling of 2 and a significant promotion gain in enhancement measure evaluation for an up-scaling of 2. The experimental results show that the performance of the AMED-SR method is better than existing super-resolution reconstruction methods in terms of visual and accuracy improvements. ZY-3 is the first Chinese civilian high-resolution stereo-mapping satellite, designed for 1:50 000 scale mapping. Although ZY-3 has a very high accuracy for direct image geolocation without ground control points ( GCPs), the use of some GCPs is still indispensable for stereo mapping. A novel method is proposed whereby spaceborne Geoscience Laser Altimeter System ( GLAS) laser altimetry datasets are viewed as generalised height control points to support a ZY-3 block adjustment. This method separates planimetric and height control, integrating the latter with GLAS data such that, without other GCPs, the vertical accuracy of ZY-3 images can be improved significantly. Both flat and mountainous terrain were selected to evaluate the proposed method. The results show that by adding two GLAS height control points for a single image, or spaced every 50 km in the along-track (orbital) direction for block images, the vertical accuracy can satisfy the requirements of 1:50 000 scale mapping, thus reducing the need for field surveying. This paper details a meticulous ground model using triangular networks and furring texture images as well as imaging simulations of TDI CCD cameras for surveying and mapping. The sight target index was built by projecting the ground triangular network to the image, and then evaluated to see if a ground point was visible in the camera and sun direction by "imaging" in these two directions, and creating a shadow. The TDI CCD plane was divided into small CCDs and the irradiance of every small CCD was computed to get the "continuous focal plane irradiance image". The static MTF was simulated by filtering the "continuous focal plane irradiance image" using PSF. The integration time was divided and the filtered static image computed for every sub-time, then the images added for every sub-time to get the "time mean static image". Then, graupel noise was added to every "time mean static image" of every line CCD, and then, adding the "time mean static image" of the same target by different CCD lines to get the average energy image, thus realizing TDI CCD multistage dynamic imaging simulation. In this manner, it was possible to derive the camera setting angle and optical panel point transfer function from platform coordinates to object coordinates, realizing an imaging simulation for any incline angle and setting angle by integrating attitude and CCD distortion parameters. The Ziyuan3-01 (ZY3-01) satellite is China's first civilian stereo surveying and mapping satellite to meet the 1:50,000 scale mapping requirements, and has been operated in orbit for 10 years. The boresight misalignment of the three-line camera (TLC) is an essential factor affecting the geolocation accuracy, which is a principal concern for stereo mapping satellites. However, the relative relationships of TLC are often regarded as fixed for the same ground scene in most traditional geometric calibrations, without considering the on-orbit long-periodic changes. In this paper, we propose a long-periodic method to analyze and estimate the boresight misalignments between three cameras, with the attitude estimation of a nadir (NAD) camera as the benchmark. Offsets and drifts of the three cameras were calculated and calibrated with different compensation models using scale invariant feature transform (SIFT) points as the ground control. Ten simultaneous NAD–Forward (FWD)–Backward (BWD) imagery of the ZY3-01 satellite acquired from 2012 to 2020 were selected to verify the long-periodic changes in TLC boresight misalignments. The results indicate that the boresight alignment angles of ZY3-01 TLC are dynamic during the long-periodic flight, but the structure of TLC is stable for the misalignments of both FWD and BWD within only 7 arc seconds, which can provide a positive reference for subsequent satellite design and long-periodic on-orbit geometric calibration. Slope failures, subsidence, earthworks, consolidation of waste dumps, and erosion are typical active deformation processes that pose a significant hazard in current and abandoned mining areas, given their considerable potential to produce damage and affect the population at large. This work proves the potential of exploiting space-borne InSAR and airborne LiDAR techniques, combined with data inferred through a simple slope stability geotechnical model, to obtain and update inventory maps of active deformations in mining areas. The proposed approach is illustrated by analyzing the region of Sierra de Cartagena-La Union (Murcia), a mountainous mining area in southeast Spain. Firstly, we processed Sentinel-1 InSAR imagery acquired both in ascending and descending orbits covering the period from October 2016 to November 2021. The obtained ascending and descending deformation velocities were then separately post-processed to semi-automatically generate two active deformation areas (ADA) maps by using ADATool. Subsequently, the PS-InSAR LOS displacements of the ascending and descending tracks were decomposed into vertical and east-west components. Complementarily, open-access, and non-customized LiDAR point clouds were used to analyze surface changes and movements. Furthermore, a slope stability safety factor (SF) map was obtained over the study area adopting a simple infinite slope stability model. Finally, the InSAR-derived maps, the LiDAR-derived map, and the SF map were integrated to update a previously published landslides' inventory map and to perform a preliminary classification of the different active deformation areas with the support of optical images and a geological map. Complementarily, a level of activity index is defined to state the reliability of the detected ADA. A total of 28, 19, 5, and 12 ADAs were identified through ascending, descending, horizontal, and vertical InSAR datasets, respectively, and 58 ADAs from the LiDAR change detection map. The subsequent preliminary classification of the ADA enabled the identification of eight areas of consolidation of waste dumps, 11 zones in which earthworks were performed, three areas affected by erosion processes, 17 landslides, two mining subsidence zone, seven areas affected by compound processes, and 23 possible false positive ADAs. The results highlight the effectiveness of these two remote sensing techniques (i.e., InSAR and LiDAR) in conjunction with simple geotechnical models and with the support of orthophotos and geological information to update inventory maps of active deformation areas in mining zones. Satellite laser altimetry can obtain sub-meter or even centimeter-scale surface elevation data over large areas, but it is inevitably affected by scattering caused by clouds, aerosols, and other atmospheric particles. This laser ranging error caused by scattering cannot be ignored. In this study, we systematically combined existing atmospheric scattering identification technology used in satellite laser altimetry and observed that the traditional algorithm cannot effectively estimate the laser multiple scattering of the GaoFen-7 (GF-7) satellite. To solve this problem, we used data from the GF-7 satellite to analyze the importance of atmospheric scattering and propose an identification scheme for atmospheric scattering data over land and water areas. We also used a look-up table and a multi-layer perceptron (MLP) model to identify and correct atmospheric scattering, for which the availability of land and water data reached 16.67% and 26.09%, respectively. After correction using the MLP model, the availability of land and water data increased to 21% and 30%, respectively. These corrections mitigated the low identification accuracy due to atmospheric scattering, which is significant for facilitating satellite laser altimetry data processing. [ABSTRACT FROM AUTHOR]

Digital Surface Model (DSM) derived from high resolution satellite imagery is important for various applications. GFDM is China's first civil optical remote sensing satellite with multiple agile imaging modes and sub-meter resolution. Its panchromatic resolution is 0.5 m and 1.68 m for multi-spectral images. Compared with the onboard stereo viewing instruments (0.8 m for forward image, 0.65 m for back image, and 2.6 m for back multi-spectrum images) of GF-7, a mapping satellite of China in the same period, their accuracy is very similar. However, the accuracy of GFDM DSM has not yet been verified or fully characterized, and the detailed difference between the two has not yet been assessed either. This paper evaluates the DSM accuracy generated by GFDM and GF-7 satellite imagery using high-precision reference DSM and the observations of Ground Control Points (GCPs) as the reference data. A method to evaluate the DSM accuracy based on regional DSM errors and GCPs errors is proposed. Through the analysis of DSM subtraction, profile lines, strips detection and residuals coupling differences, the differences of DSM overall accuracy, vertical accuracy, horizontal accuracy and the strips errors between GFDM DSM and GF-7 DSM are evaluated. The results show that the overall accuracy of both is close while the vertical accuracy is slightly different. When regional DSM is used as the benchmark, the GFDM DSM has a slight advantage in elevation accuracy, but there are some regular fluctuation strips with small amplitude. When GCPs are used as the reference, the elevation Root Mean Square Error (RMSE) of GFDM DSM is about 0.94 m, and that of GF-7 is 0.67 m. GF-7 DSM is more accurate, but both of the errors are within 1 m. The DSM image residuals of the GF-7 are within 0.5 pixel, while the residuals of GFDM are relatively large, reaching 0.8 pixel. ZiYuan-3 (ZY-3) surveying satellite is China's first civilian high-resolution stereo mapping satellite. The satellite's objective is oriented towards plotting 1:50,000 and 1:25,000 topographic maps. Compared with plotting using foreign commercial mapping satellite imagery, establishment of the image geometry model is the core technical problem for different products and various applications of ZY-3 surveying satellite. This article analyses key problems of high-precision geometry processing based on the overall design, and proposes ZY-3 surveying satellite image geometry model with the technology of virtual CCD line-array imaging. In addition, this article utilises the first orbit imagery of ZY-3 satellite with coverage of Dalian region in Liaoning Province, and produces forward, backward and nadir camera calibrated products. Different ground control points are selected for the block adjustment experiment and the digital surface model (DSM), digital ortho-map (DOM) are generated. The accuracy is validated by check points. As the important infrastructures for land mapping and resource monitoring, high-resolution remote sensing satellites (HRSS) are urgently demanded for the development of China. In this article, the key technologies of the main HRSS are summarized, and these technologies include sensor design, attitude and orbit determination, geometric calibration, imaging model construction, and block adjustment, etc., which involve the mapping accuracy of HRSS. Finally, the system design of the ZY-3 Satellite (China's first civil stereoscopic surveying and mapping satellite, to be launched in 2012) is introduced, which mainly include satellite technical specifications and strategies design based on these key technologies research. Rabbit coccidiosis is caused by infection with one or, more commonly, several Eimeria species that parasitize the hepatobiliary ducts or intestinal epithelium of rabbits. Currently, there are eleven internationally recognized species of rabbit coccidia, with the complete mitochondrial (mt) genomes of six species commonly infecting rabbits having been sequenced and annotated. Eimeria kongi was initially discovered in 2011 and prompted a preliminary study on this species. Through traditional morphological analysis, E. kongi was identified as a novel species of rabbit coccidia. To further validate this classification, we sequenced and annotated its mitochondrial genome. The complete mt genome of E. kongi spans 6258 bp and comprises three cytochrome genes (cytb, cox1, cox3), fourteen gene fragments for the large subunit (LSU) rRNA, and nine gene fragments for the small subunit (SSU) rRNA, lacking transfer RNA (tRNA) genes. Moreover, phylogenetic analysis of the mitochondrial genome sequence of E. kongi revealed its clustering with six other species of rabbit coccidia into a monophyletic group. Additionally, E. irresidua and E. flavescens were grouped within the lineage lacking oocyst residuum, consistent with their morphological characteristics. Consistent with multiple molecular phylogenies, in this investigation, E. kongi was further confirmed as a new species of rabbit coccidia. Slope and roughness are basic geophysical properties of terrain surface, and also sources of error in satellite laser altimetry systems. The full-waveform satellite laser altimeter records the complete echo waveform backscattered from the target surface worldwide, so it may be used for both range measurement and inversion analysis of geometric parameters of the target surface. This paper proposes a new method for inversion of slope and roughness of the bare or near-bare terrain within laser footprint using full-waveform satellite laser altimeter data, Shuttle Radar Topographic Mission (SRTM) and topographic prior knowledge. To solve the non-uniqueness of the solution to the inversion problem, this paper used the SRTM and airborne Light Detection and Ranging (LiDAR) data in North Rhine-Westphalia, Germany, to establish a priori hypothesis about real information of topographic parameters. Then, under the constraints of prior hypothesis, the theoretical formulas and rules for slope and roughness inversion using the pulse-width broadening knowledge of satellite laser altimeter echo full-waveform were developed. Finally, based on the full-waveform data from the Geoscience Laser Altimeter System (GLAS) that was borne on ICE, Cloud, and Land Elevation Satellite (ICESat) and SRTM in the West Valley City, Utah and Jackson City, Wyoming, United States of America, the inversion was carried out. The experiment compares the results of proposed method with those of existing ones and evaluates the inversion results using high precision terrain slope and roughness information, which indicates that our proposed method is superior to the state-of-the-art methods, and the inversion accuracy for slope is 0.667 ° (Mean Absolute Error, MAE) and 1.054 ° (Root Mean Square Error, RMSE), the inversion accuracy for roughness is 0.171 m (MAE) and 0.250 m (RMSE). Intensive and large-scale underground coal mining has caused geological disasters such as local ground subsidence, cracks and collapse in the Datong coalfield, China, inducing serious threats to local residents. Interferometric synthetic aperture radar (InSAR) has the capability of surface deformation detection with high precision in vast mountainous areas. DInSAR, stacking-InSAR and SBAS-InSAR are commonly used InSAR-related deformation analysis methods. They can provide effective support for mine ecological security monitoring and prevent disasters. We use the three methods to conduct the deformation observation experiments in the Datong coalfield. Sentinel-1A data from November 2020 to October 2021 are used. As a result, a total of 256 deformations in the Datong coalfield were successfully detected by the three methods, of which 218 are mining deformations, accounting for 85% of the total deformations. By comparing the results of the three methods, we found that DInSAR, stacking-InSAR, and SBAS-InSAR detected 130, 256, and 226 deformations in the Datong coalfield, respectively, while the deformations caused by coal mining were 128, 218, and 190. DInSAR results with long spatiotemporal baselines are seriously incoherent. SBAS-InSAR results of displacement rate are more precise than stacking-InSAR, and the mean standard deviation is 1.0 mm/a. However, for areas with lush vegetation or low coherence, SBAS-InSAR has poor performance. The detection deformation area results of DInSAR and SBAS-InSAR are subsets of stacking-InSAR. The displacement rates obtained by stacking-InSAR and SBAS-InSAR are consistent; the mean difference in the displacement rate between the two methods is 2.7 mm/a, and the standard deviation is 5.1 mm/a. The mining deformation locations and their shapes in the study area can be identified with high efficiency and power by stacking-InSAR. Therefore, with a comprehensive understanding of the advantages and limitations of the three methods, stacking-InSAR can be an effective and fast method to identify the level, location and range of mining deformation in lush mountainous areas. The Gaofen-7 (GF-7) satellite uses a two-beam laser altimetry system in which each beam is equipped with a laser footprint camera (LFC) to provide geometric processing of the laser footprint images that assist in optical image stereo mapping. Because of the violent vibrations during launch and the difference in the environment before and after entering orbit, the key parameters for geometric processing of the laser footprint images may change, which will cause large geolocation errors. Therefore, it is essential to carry out on-orbit calibration and validation for the laser footprint cameras. This study first constructs a rigorous geometric positioning model for the LFC of the GF-7 satellite and analyses various error sources that affect the geometric positioning accuracy of laser footprint images. Then, a comprehensive calibration method, which effectively eliminates the distortion of the LFC optical system, and the positioning error caused by the long-period jitter of the satellite platform, is proposed based on the multi-scene images combined with image simulation. The proposed method can effectively eliminate various errors that affect the geometric positioning accuracy of the GF-7 laser footprint image. The internal geometric positioning accuracy of the calibrated LFC is better than 0.7 pixels, and the absolute geometric positioning accuracy is within 6.0 m after using precise post-processing orbital and attitude data. Satellite platform microvibration is a common phenomenon in earth observation satellite orbits that directly affects the imaging quality and accuracy of surveying and mapping. With the continuous improvement in the spatial resolution, the influence of satellite platform microvibration on image geometric accuracy is becoming increasingly significant. High-precision microvibration detection and compensation are key technologies for eliminating image distortion and location deviation caused by satellite platform microvibration. In this paper, the microvibration detection methods of different satellite platforms are summarized, and the verification and analysis are performed on the data downloaded from the Resource-3 satellite (ZY-3) platform, to provide technical support for subsequent refined processing of satellite attitude. The Gao Fen 7 (GF‐7) satellite is China's first submeter, high‐resolution Earth observation and remote sensing satellite for natural resource monitoring, land surveying, and other industrial applications in China. The GF‐7 laser altimeter system is the successor of the ZiYuan3‐02 (ZY3‐02 satellite) laser altimeter. The key objective of the GF‐7 laser altimetry system is to provide high‐precision ground elevation control points that assist in optical imaging to achieve 1:10000 mapping in China. This paper introduces the GF‐7 laser altimeter system payload specifications, mission objectives, in‐orbit calibration plan, data products, and service policy. In the future, the GF‐7 laser altimeter system will also serve multidisciplinary and professional applications through its precise elevation measurement capabilities. Chicken coccidiosis is an intestinal disease caused by the parasite Eimeria, which severely damages the growth of chickens and causes significant economic losses in the poultry industry. Improvement of the immune protective effect of antigens to develop high efficiency subunit vaccines is one of the hotspots in coccidiosis research. Sporozoite‐specific surface antigen 1 (SAG1) of Eimeria tenella (E. tenella) is a well‐known protective antigen and is one of the main target antigens for the development of subunit, DNA and vector vaccines. However, the production and immunoprotective effects of SAG1 need to be further improved. Here, we report that both SAG1 from E. tenella and its fusion protein with the xylanase XynCDBFV‐SAG1 are recombinant expressed and produced in Pichia pastoris (P. pastoris). The substantial expression quantity of fusion protein XynCDBFV‐SAG1 is achieved through fermentation in a 15‐L bioreactor, reaching up to about 2 g/L. Moreover, chickens immunized with the fusion protein induced higher protective immunity as evidenced by a significant reduction in the shedding of oocysts after E. tenella challenge infection compared with immunized with recombinant SAG1. Our results indicate that the xylanase enhances the immunogenicity of subunit antigens and has the potential for developing novel molecular adjuvants. Eimeria species serve as promising eukaryotic vaccine vectors. And that the location of heterologous antigens in the subcellular components of genetically modified Eimeria may determine the magnitude and type of immune responses. Therefore, our study aimed to target a heterologous fluorescent protein to the cell surface or microneme, two locations where are more effective in inducing protective immunity, of Eimeria tenella and E. acervulina sporozoites. We used an enhanced yellow fluorescent protein (EYFP) as a tagging biomarker, fusing variously with some localization or whole sequences of compartmental proteins for targeting. After acquiring stable transgenic Eimeria populations, we observed EYFP expressing in expected locations with certain strategies. That is, EYFP successfully localized to the surface when it was fused between signal peptides and mature products of surface antigen 1 (SAG1). Furthermore, EYFP was efficiently targeted to the apical end, an optimal location for secretory organelle known as the microneme, when fused to the C terminus of microneme protein 2. Unexpectedly, EYFP exhibited dominantly in the apical end with only weak expression on the surface of the transgenic sporozoites when the parasites were transfected with plasmid with EYFP fused between signal peptides and mature products of E. tenella SAG 13. These strategies worked in both E. tenella and E. acervulina, laying a solid foundation for studying E. tenella and E. acervulina-based live vaccines that can be further tailored to the inclusion of cargo immunogens from other pathogens. Coccidiosis, an intestinal disease caused by Eimeria protozoan parasites, affects various animal species, and especially poses a significant threat to the poultry industry. The current primary control methods include anticoccidial drugs and vaccines. However, emerging challenges such as drug resistance and vaccine efficacy issues are rooted in the complex life cycle and species diversification of Eimeria. In this review, we first consolidate recent breakthroughs in understanding Eimeria biology, focusing on the parasite development and its intricate interactions with the host, notably its relationships with host immune cells and the gut microbiota. Furthermore, we provide an extensive summary of current control strategies for Eimeria infections. This includes an in-depth analysis of anticoccidial drugs, their mechanisms of resistance, and the increasing utilization of diverse anticoccidial vaccines to combat these challenges. Finally, we highlight the latest innovative strategies leading the way in coccidiosis control. Through an exploration of cutting-edge techniques, we also provide insights into future directions for effectively combating this disease. In conclusion, the future of coccidiosis control lies in the use of a multifaceted approach, integrating advanced biological insights with innovative therapeutic strategies. Background: Live anticoccidial vaccines have been a tremendous success for disease prevention. The establishment of the reverse genetic manipulation platform has enabled the development of Eimeria parasites, the live anticoccidial vaccine strains, as vaccine vectors. In our previous study, recombinant E. tenella expressing a single immunodominant antigen of E. maxima (Et-EmIMP1) was able to protect chickens against challenge infection with E. maxima. This promising result encouraged us to further explore strategies to improve the protection efficacy of recombinant Eimeria and develop it as a vaccine vector. Results: We constructed a novel recombinant Eimeria line expressing apical membrane antigen 1 of E. maxima (Et-EmAMA1) and then immunized chickens with Et-EmAMA1 and/or Et-EmIMP1. We found that the E. maxima soluble antigen-specific cell-mediated immunity was much stronger in the birds that were co-immunized with Et-EmAMA1 and Et-EmIMP1 than in those that were immunized with Et-EmAMA1 or Et-EmIMP1 alone. The oocyst production after E. maxima infection was significantly reduced in the recombinant Eimeria-immunized birds compared with the wild-type-immunized and naïve birds. The oocyst production in the birds co-immunized with Et-EmAMA1 and Et-EmIMP1 was consistently the lowest among the treatment groups after E. maxima infection. Conclusions: These results demonstrated that Eimeria is an effective vaccine vector that can carry and deliver heterologous Eimeria antigens to the host immune system and trigger specific immune responses. Our results also suggested that increasing the number of recombinant Eimeria lines is an effective approach to enhance protective immunity against infections with heterologous pathogens. Toxoplasma gondii rapidly propagates through endodyogeny of tachyzoites, a process in which daughter parasites divide within the cell of the mother parasite. Recent studies have revealed that transcription factors with AP2‐domain participate in the process of cell division in T. gondii. However, the concise regulation of the division cycles by AP2 proteins is poorly understood. In this study, we evaluated the effect of the transcription factor TgAP2IX‐5 on the daughter cell formation in T. gondii. TgAP2IX‐5 is a nuclear protein and is highly expressed during the S phase of the cell cycle of tachyzoites. TgAP2IX‐5‐disrupted strain showed a severe defect in replication and completely blocked lytic parasite growth. Following 3‐indoleacetic acid treatment or without treatment of AP2IX‐5‐AID‐3HA tagged strain for 30 min, 1 and 2 hr, the differentially expressed genes were 8, 54 and 202, respectively. Among these genes, the significantly downregulated ones were AP2 proteins, inner membrane complex (IMC) proteins and SAG‐related proteins. Interestingly, loss of TgAP2IX‐5 leads to a defect in internal daughter IMC formation and abnormalities in the morphology of organelles during cell division. Background: Protozoan parasites of the genus Eimeria are the causative agents of chicken coccidiosis. Parasite resistance to most anticoccidial drugs is one of the major challenges to controlling this disease. There is an urgent need for a molecular marker to monitor the emergence of resistance against anticoccidial drugs, such as decoquinate. Methods: We developed decoquinate-resistant strains by successively exposing the Houghton (H) and Xinjiang (XJ) strains of E. tenella to incremental concentrations of this drug in chickens. Additionally, we isolated a decoquinate-resistant strain from the field. The resistance of these three strains was tested using the criteria of weight gain, relative oocyst production and reduction of lesion scores. Whole-genome sequencing was used to identify the non-synonymous mutations in coding genes that were highly associated with the decoquinate-resistant phenotype in the two laboratory-induced strains. Subsequently, we scrutinized the missense mutation in a field-resistant strain for verification. We also employed the AlphaFold and PyMOL systems to model the alterations in the binding affinity of the mutants toward the drug molecule. Results: We obtained two decoquinate-resistant (DecR) strains, DecR_H and XJ, originating from the original H and XJ strains, respectively, as well as a decoquinate-resistant E. tenella strain from the field (DecR_SC). These three strains displayed resistance to 120 mg/kg decoquinate administered through feed. Through whole-genome sequencing analysis, we identified the cytochrome b gene (cyt b; ETH2_MIT00100) as the sole mutated gene shared between the DecR_H and XJ strains and also detected this gene in the DecR_SC strain. Distinct non-synonymous mutations, namely Gln131Lys in DecR_H, Phe263Leu in DecR_XJ, and Phe283Leu in DecR_SC were observed in the three resistant strains. Notably, these mutations were located in the extracellular segments of cyt b, in close proximity to the ubiquinol oxidation site Qo. Drug molecular docking studies revealed that cyt b harboring these mutants exhibited varying degrees of reduced binding ability to decoquinate. Conclusions: Our findings emphasize the critical role of cyt b mutations in the development of decoquinate resistance in E. tenella. The strong correlation observed between cyt b mutant alleles and resistance indicates their potential as valuable molecular markers for the rapid detection of decoquinate resistance. Abstract Eimeria spp., the causative agents of coccidiosis, are the most common protozoan pathogens of chickens. Infection with these parasites can result in poor development or death of animals leading to a devastating economic impact on poultry production. The establishment of transfection protocols for genetic manipulation of Eimeria species and stable expression of genes would help advance the biology of these parasites as well as establish these organisms as novel vaccine delivery vehicles. Here, we report the selection of the first stable transgenic E. necatrix population, EnHA1 , consitutively expressing the EYFP reporter following transfection of the 2nd generation merozoites with a linear DNA fragment harboring the EYFP reporter gene, the HA1 gene from the avian influenza virus H9N2 and the TgDHFR-TS selectable marker, which confers resistance to pyrimethamine. Transfected merozoites were inoculated into chickens via the cloacal route, and feces from 18 h to 72 h post inoculation were collected and subjected to subsequent serial passages, FACS sorting and pyrimethamine selection. A gradual increase in the number of EYFP-expressing sporulated oocysts was noticed with more than 90% EYFP + oocysts obtained after five passages. Immunofluorescence assay confirmed successful expression of the HA1 antigen in the EnHA1 population. The ability to genetically manipulate E. necatrix merozoites and express heterologous genes in this parasite will pave the way for possible use of this organism as a vaccine-delivery vehicle. Eimeria species are apicomplexan parasites with a direct life cycle consisting of a replicative phase involving multiple rounds of asexual replication in the intestine or other organs including kidneys, liver, and gallbladder, depending on the species, followed by a sexual phase or gamogony involving the development and fertilization of gametes, an essential process for Eimeria transmission. Recent advances in the genetic manipulation of these parasites made it possible to conduct genetic crosses combined with genomic approaches to elucidate the genetic determinants of Eimeria development, virulence, drug resistance, and immune evasion. Here, we employed genetic techniques to generate two transgenic Eimeria acervulina lines, EaGAM56 and EaHAP2, each expressing two unique fluorescent proteins, with one controlled by a constitutive promotor for cross-efficiency analysis and the other by a male or female gametocyte stage-specific promoter to observe sexual development. The expression of fluorescent proteins in the transgenic lines was analyzed in different developmental stages of the E. acervulina life cycle by immunoblotting and by examination of frozen sections using fluorescence microscopy. The effect of infective doses on cross-fertilization was further investigated by conducting several genetic crosses between the two transgenic lines at different doses and ratios. Two transgenic lines expressing constitutive and gametocyte-specific fluorescence proteins were generated and characterized. These transgenic parasites display synchronous development in chickens, comparable with that of the wild type. Genetic crosses between the two transgenic parasites showed that a high rate of oocysts co-expressing the two reporters could be achieved following inoculation with high doses of infective oocysts. We further showed that the proportion of co-transfected oocysts can be modulated by altering the ratio of the transgenic parental lines. Higher infective doses and similar numbers of functional gametocytes from the parents increase the rate of cross-fertilization. Our data highlight the usefulness of genetic manipulation and fluorescently-labeled transgenic gametocytes as tools to study Eimeria development and to elucidate the factors that modulate sexual development. The atmospheric refraction effect seriously restricts the precision of satellite measurements. Various atmospheric refraction models have been proposed to solve the bending effect due to refraction. But the atmosphere shows a complicated spatio-temporal distribution due to turbulences, which affects the precision of the atmosphere models. In this study, we proposed a novel method to detect the atmospheric refraction effect according to the dispersion of the electromagnetic wave. Depending on one satellite image only, we can invert the displacement of atmospheric refraction effect. And this method makes it a reality without constructing the atmosphere model and monitoring a series of meteorological data. In the experiment, two high-resolution images were used to validate the method, an inclined shooting scene at zenith angle of 30° and a vertical shooting scene as a control. The result is close to the value calculated by the atmospheric refraction model. So the proposed method is effective to detect the atmospheric refraction effect using satellite remote sensing. GaoFen 7 (GF-7) is China's first submeter high-resolution stereo mapping satellite with dual-linear-array cameras and a laser altimeter system onboard for high-precision mapping. To further take advantage of the very high elevation accuracy of laser altimetry data and the high relative accuracy with stereo images, an innovative combined adjustment method for GF-7 stereo images with laser altimetry data is presented in this paper. In this method, two flexible and effective schemes were proposed to extract the elevation control point according to the registration of footprint images and stereo images and then utilized as vertical control in the block adjustment to improve the elevation accuracy without ground control points (GCPs). The validation experiments were conducted in Shandong, China, with different terrains. The results demonstrated that, after using the laser altimetry data, the root mean square error (RMSE) of elevation was dramatically improved from the original 2.15 m to 0.75 m, while the maximum elevation error was less than 1.6 m. Moreover, by integrating a few horizontal control points, the planar and elevation accuracy can be simultaneously improved. Abstract: Topological relations are one of the most important aspects in GIS modeling. The topological relations between crisp spatial objects have been well identified. However, the topological relations between fuzzy spatial objects need more investigation. This paper deals with building a special fuzzy topological space for fuzzy sets. Based on it, a formal definition of simple fuzzy region is given. A 4×4-intersection and furthermore a 5×5-intersection approach are proposed for the formalism of topological relations. This study described the on-orbit vicarious radiometric calibration of Chinese civilian high-resolution stereo mapping satellite ZY3-02 multispectral imager (MUX). The calibration was based on gray-scale permanent artificial targets, and multiple radiometric calibration tarpaulins (tarps) using a reflectance-based approach between July and September 2016 at Baotou calibration site in China was described. The calibration results reveal a good linear relationship between DN and TOA radiances of ZY3-02 MUX. The uncertainty of this radiometric calibration was 4.33%, indicating that radiometric coefficients of ZY3-02 MUX are reliable. A detailed discussion on the validation analysis of the comparison results between the different radiometric calibration coefficients is presented in this paper. To further validate the reliability of the three coefficients, the calibrated ZY3-02 MUX was compared with Landsat-8 Operational Land Imager (OLI). The results also indicate that radiometric characteristics of ZY3-02 MUX imagery are reliable and highly accurate for quantitative applications. [ABSTRACT FROM AUTHOR]

China's first civilian, sub-meter, high-resolution stereo mapping satellite, GF-7, launched on 3 November 2019. Radiometric characterization of GF-7 multispectral imagery has been performed in this study. A relative radiometric accuracy evaluation of the GF-7 multispectral imagery was performed using several large uniform scenes, and the results showed that the accuracy is better than 2%. The absolute radiometric evaluation of the GF-7 satellite sensor was conducted at the Baotou and Dunhuang calibration sites, using the reflectance-based vicarious approach. The synchronous measurements of surface reflectance and atmospheric parameters were collected as the input for the radiative transfer model. The official radiometrically calibrated coefficient of the GF-7 multispectral imagery was evaluated with the predicted top-of-atmosphere (TOA) radiance from the radiative transfer model. The results indicated that the absolute radiometric accuracy of GF-7 multispectral imagery is better than 5%. In order to monitor the radiometric stability of the GF-7 satellite multispectral sensor, a relative and absolute radiometric accuracy assessment campaign should be performed several times a year. The increasing frequency of human activities has accelerated changes in land use types and consequently affected the atmospheric environment. In this manuscript, we analyze the relationships between the particulate matter concentration and land use changes in the Beijing–Tianjin–Hebei (BTH) region, China, from 2015 to 2018. The experimental results indicate that (1) an improved sine function model can suitably fit the periodic changes in the particulate matter concentration, with the average R2 value increasing to 0.65 from the traditional model value of 0.49, while each model coefficient effectively estimates the change characteristics of each stage. (2) Among all land use types, the particulate matter concentrations in construction land and farmland are high, with a large annual difference between high and low values. The concentration decreases slowly in spring and summer but increases rapidly in autumn and winter. The concentrations in forestland and grassland are the lowest; the difference between high and low values is small for these land use types, and the concentration fluctuation pattern is relatively uniform. Natural sources greatly influence the concentration fluctuations, among which frequent dusty weather conditions in spring impose a greater influence on forestland and grassland than on the other land use types. (3) The landscape pattern of land use exerts a significant influence on the particulate matter concentration. Generally, the lower the aggregation degree of patches is, the higher the fragmentation degree is, the more complex the shape is, the higher the landscape abundance is, and the lower the particulate matter concentration is. The higher the construction land concentration is, the more easily emission sources can be aggregated to increase the particulate matter concentration. However, when forestland areas are suitably connected, this land use type can play a notable role in inhibiting particulate matter concentration aggravation. [Display omitted] • Six isolates of Cryptosporidium cuniculus were isolated from rabbit farms, all belonging to the Ⅴb subtype. • Two isolates (DF12 and CC1) were characterized for their infectivity in immunocompetent rabbits. • The DF12 isolate was used to establish a rabbit infection model for successful evaluation of anti-cryptosporidial efficacy. Cryptosporidium is one of the leading causes of diarrheal disease in humans and animals, which can be severe and deadly in neonates and immunocompromised hosts. Studies on the biology of Cryptosporidium and drug discovery efforts have been hindered by a number of factors including the limited availability of animal models. Here, we report the establishment and characterization of an immunocompetent rabbit model for infection with Cryptosporidium cuniculus. By testing four known anti-cryptosporidial compounds (nitazoxanide, baicalein, curcumin and matrine), we showed that the rabbit could be used as an alternative animal model for evaluating anti-cryptosporidial drug efficacy in vivo. GF-6 is the first optical remote sensing satellite for precision agriculture observations in China. Accurate identification of the cloud in GF-6 helps improve data availability. However, due to the narrow band range contained in GF-6, Fmask version 3.2 for Landsat is not suitable for GF-6. Hence, this paper proposes an improved Fmask based on the spectral-contextual information to solve the inapplicability of Fmask version 3.2 in GF-6. The improvements are divided into the following six aspects. The shortwave infrared (SWIR) in the "Basic Test" is replaced by blue band. The threshold in the original "HOT Test" is modified based on the comprehensive consideration of fog and thin clouds. The bare soil and rock are detected by the relationship between green and near infrared (NIR) bands. The bright buildings are detected by the relationship between the upper and lower quartiles of blue and red bands. The stratus with high humidity and fog_W (fog over water) are distinguished by the ratio of blue and red edge position 1 bands. Temperature probability for land is replaced by the HOT-based cloud probability (LHOT), and SWIR in brightness probability is replaced by NIR. Pneumocystis pneumonia (PCP) and pulmonary toxoplasmosis (PT) are caused by Pneumocystis jirovecii and Toxoplasma gondii. The clinical symptoms and imaging of PCP and PT are indistinguishable. A duplex qPCR was developed to differentiate between these two pathogens. In testing 92 clinical samples to validate the performance of this method for P. jirovecii detection, it identified 31 positive samples for P. jirovecii infection, consistent with clinical diagnosis. Among the remainder of the 61 clinical samples with suspected PCP, yet showing as negative by the conventional PCR diagnosis approach, 6 of them proved positive using our new assay. Our new approach also produced similar results in identification of T. gondii infections, giving a result of 2 positive and 20 negative in clinical samples. An investigation was undertaken on the prevalence of P. jirovecii and T. gondii infections using 113 samples from lung infection patients. 9% (10/113) were shown to be positive with infections of P. jirovecii, 2% with T. gondii (2/113) and 5% (6/113) were co-infected with both pathogens. Although this duplex qPCR can detect individual P. jirovecii and T. gondii infection, and co-infection of both pathogens, further large-scale investigations are needed to validate its performance, especially in T. gondii detection. Our assay provides a rapid and accurate tool for PCP and PT diagnosis in immunocompromised population and clinical surveillance of these infections in patients with no immune defects. The GF-7 satellite is China's first high-resolution stereo mapping satellite that reaches sub-meter resolution, equipped with new-type payloads, such as an area array footprint camera that can achieve synchronization acquisition of laser spots. When the satellite is in space, the variation of camera parameters may occur due to launch vibration and environmental changes, and on-orbit geometric calibration thereby must be made. Coupled with the data from the GF-7 satellite, this paper constructs a geometric imaging model of the area array footprint camera based on the two-dimensional direction angle, and proposes a coarse-to-fine "LPM-SIFT + Phase correlation" matching strategy for the automatic extraction of calibration control points. The single-image calibration experiment shows that the on-orbit geometric calibration model of the footprint camera constructed in this paper is correct and effective. The matching method proposed is used to register the footprint images with the DOM (Digital Orthophoto Map) reference data to obtain dense control points. Compared with the calibration result using a small number of manually collected control points, the root mean square error (RMSE) of the residual of the control points is improved from half a pixel to 1/3, and the RMSE of the same orbit checkpoints in the image space is improved from 1 pixel to 0.7. It can be concluded that using the coarse-to-fine image matching method proposed in this paper to extract control points can significantly improve the on-orbit calibration accuracy of the footprint camera on the GF-7 satellite. Avian coccidiosis caused by Eimeria is a serious parasitic disease that poses a threat to the poultry industry. Currently, prevention and treatment mainly rely on the administration of anticoccidials and live oocyst vaccines. However, the prevalence of drug resistance and the inherent limitations of live vaccines have driven the development of novel vaccines. In this study, the surface protein (Et-SAG14), a previously annotated rhoptry protein (Eten5-B), and a gametocyte phosphoglucomutase (Et-PGM1) were characterized and the vaccine potential of the recombinant proteins were evaluated. Et-SAG14 was dispersed in the form of particles in the sporozoite and merozoite stages, whereas Et-PGM1 was distributed in the apical part of the sporozoite and merozoite stages. The previously annotated rhoptry Eten5-B was found not to be located in the rhoptry but distributed in the cytoplasm of sporozoites and merozoites. Immunization with rEten5-B significantly elevated host interferon gamma (IFN-γ) and interleukin 10 (IL-10) transcript levels and exhibited moderate anticoccidial effects with an anticoccidial index (ACI) of 161. Unexpectedly, both recombinant Et-SAG14 and Et-PGM1 immunization significantly reduced host IFN-γ and IL-10 transcription levels, and did not show protection against E. tenella challenge (ACI < 80). These results suggest that the rEten5-B protein can trigger immune protection against E. tenella and may be a potential and effective subunit vaccine for the control of coccidiosis in poultry.
