Abstract Background Pudendal neuralgia is a chronic and debilitating condition. Its prevalence ranges from 5 to 26%. Currently, therapeutic approaches to treat pudendal neuralgia include patient education, medication management, psychological and physical therapy, and procedural interventions, such as nerve block, trigger point injections, and surgery. Drug therapy has a limited effect on pain relief. A pudendal nerve block may cause a significant decrease in pain scores for a short time; however, its efficacy significantly decreases over time. In contrast, pudendal nerve pulsed radiofrequency can provide pain relief for 3 months, and ganglion impar block has been widely used for treating chronic perineal pain and chronic coccygodynia. This study aimed to determine the efficacy and safety of monotherapy (pudendal nerve pulsed radiofrequency) and combination therapy (pudendal nerve pulsed radiofrequency plus ganglion impar block) in patients with pudendal neuralgia. Methods This randomized, controlled clinical trial will include 84 patients with pudendal neuralgia who failed to respond to drug or physical therapy. Patients will be randomly assigned into one of the two groups: mono or combined treatment groups. The primary outcome will be a change in pain intensity measured using the visual analog scale. The secondary outcomes will include a Self-Rating Anxiety Scale score, Self-Rating Depression Scale score, the use of oral analgesics, the Medical Outcomes Study Health Survey Short Form-36 Item score, and the occurrence of adverse effects. The study results will be analyzed using intention-to-treat and per-protocol analyses. Primary and secondary outcomes will be evaluated between the mono and combined treatment groups. Subgroup analyses will be conducted based on the initial ailment, age, and baseline pain intensity. The safety of the treatment will be assessed by monitoring adverse events, which will be compared between the two groups. Discussion This study protocol describes a randomized, controlled clinical trial to determine the efficacy and safety of mono and combination therapies in patients with pudendal neuralgia. The study results will provide valuable information on the potential benefits of this combination therapy and contribute to the development of more effective and safer treatments for patients with pudendal neuralgia. In this study, the microstructural evolution and mechanical properties of a series of FeCrNiCuTix (x = 0, 0.2, 0.4, 0.6, 0.8, and 1, in mole ratio) high entropy alloys (HEAs) were analyzed in detail to study the influence of Ti content on the HEAs. The microstructures of the alloys transformed from single-phase face-centered cubic (FCC) phase structure to FCC and body-centered cubic (BCC) diphase, finally to FCC, BCC and intermetallic compound (Fe2Ti) phase with the increase of Ti content. The addition of Ti element significantly refined the grain of the alloys. The hardness, strength and tribology tests of the alloys were also carried out. The results indicated that the Vickers hardness, yield strength, and fracture strength increased first and then decreased, the average friction coefficient variation law was just the opposite, and the plastic strain reduced monotonically, with the increase of x values for the alloys. At x = 0.4, the maximum yield strength and fracture strength of the alloy were 1646.89 MPa and 1947.09 MPa, respectively, and the average friction coefficient was a minimum of 0.47, with a hardness value of HV 489.68 and a plastic strain of 13.98%. The maximum hardness value of the alloy was HV 656.19 at x = 0.8. The fracture mechanism of the alloys was from ductile fracture to brittle fracture and the wear mechanism was form of abrasive wear. Accordingly, the FeCrNiCuTi0.4 alloy obtained the optimal mechanical properties overall. This research is of great significance for the development of engineering and structural materials with excellent mechanical properties.

ABSTRACT: Wenchang chicken, a prized local breed in Hainan Province of China renowned for its exceptional adaptability to tropical environments and good meat quality, is deeply favored by the public. However, an insufficient understanding of its population architecture and the unclear genetic basis that governs its typical attributes have posed challenges in the protection and breeding of this precious breed. To address these gaps, we conducted whole-genome resequencing on 200 Wenchang chicken samples derived from 10 distinct strains, and we gathered data on an array of 21 phenotype traits. Population genomics analysis unveiled distinctive population structures in Wenchang chickens, primarily attributed to strong artificial selection for different feather colors. Selection sweep analysis identified a group of candidate genes, including PCDH9, DPF3, CDIN1, and SUGCT, closely linked to adaptations that enhance resilience in tropical island habitats. Genome-wide association studies (GWAS) highlighted potential candidate genes associated with diverse feather color traits, encompassing TYR, RAB38, TRPM1, GABARAPL2, CDH1, ZMIZ1, LYST, MC1R, and SASH1. Through the comprehensive analysis of high-quality genomic and phenotypic data across diverse Wenchang chicken resource groups, this study unveils the intricate genetic backgrounds and population structures of Wenchang chickens. Additionally, it identifies multiple candidate genes linked to environmental adaptation, feather color variations, and production traits. These insights not only provide genetic reference for the purification and breeding of Wenchang chickens but also broaden our understanding of the genetic basis of phenotypic diversity in chickens.

Y-type zeolites are widely used in the carbon dioxide (CO2) adsorption applications due to their exceptional ion exchange performance and high stability. However, the pelletization of Y powder by adding binders such as kaolin often results in performance drops, which has limited its industrial application. This study employs a zeolite-kaolin blend to produce cylindrical pelletized Y zeolite, in which the kaolin is then transformed to zeolites via hydrothermal crystallization, resulting in binder-free Y pellets. The Design-Expert software and the response surface method (RSM) are employed to optimize the hydrothermal crystallization conditions for binder transformation, with the objective of attaining a maximized capacity for CO2 adsorption. Results demonstrate that optimized synthesis conditions yield cylindrical Y zeolite with a notable CO2 adsorption capacity of 5.52 mmol/g at 298 K and 1 bar, surpassing that of the initial Y powder. The average crushing strength is 110 N per particle (Diameter × Height = 3 mm × 3 mm). The IAST selectivity of CO2/N2 (15/85) is 824 at 298 K and 1 bar. The reaction mechanisms of the binder transformation are also investigated via experiments and DFT simulation. This study presents a simple and reliable method for manufacturing binder free Y zeolite, which is promising for carbon capture applications.

Abstract Due to the tolerance of mismatches between gRNA and targeting sequence, base editors frequently induce unwanted Cas9-dependent off-target mutations. Here, to develop models to predict such off-targets, we design gRNA-off- target pairs for adenine base editors (ABEs) and cytosine base editors (CBEs) and stably integrate them into the human cells. After five days of editing, we obtain valid efficiency datasets of 54,663 and 55,727 off-targets for ABEs and CBEs, respectively. We use the datasets to train deep learning models, resulting in ABEdeepoff and CBEdeepoff, which can predict off-target sites. We use these tools to predict off-targets for a panel of endogenous loci and achieve Spearman correlation values varying from 0.710 to 0.859. Finally, we develop an integrated tool that is freely accessible via an online web server http://www.deephf.com/#/bedeep/bedeepoff . These tools could facilitate minimizing the off-target effects of base editing.

In order to cleanly and efficiently extract zirconium from zircon sand (the main component is ZrSiO4), sodium hydroxide sub-molten salt was used to decompose ZrSiO4 in this study. When ZrSiO4 reacts with sodium hydroxide sub-molten salt, the formation of Na2ZrSiO5 (a water-insoluble product) considerably affects the separation efficiency of Zr and Si and increases production cost. Thus, it is necessary to control the formation of Na2ZrSiO5. The influence of NaOH content, reaction temperature, reaction time, and NaOH/ore mass ratio on the formation of Na2ZrSiO5 were systematically investigated. The optimum reaction parameters for the inhibition of Na2ZrSiO5 formation were as follows: 80% NaOH content, 245 °C reaction temperature, 4:1 NaOH/ore mass ratio, 10 h reaction time, and 400 r/min agitation speed. These results indicate that ZrSiO4 is decomposed to Na2ZrO3 and Na2SiO3 by reacting with NaOH, realizing the separation of Zr and Si, and then the reactions between Na2ZrO3 and Na2SiO3 result in the formation of Na2ZrSiO5, during the decomposition of ZrSiO4 using NaOH sub-molten salt. The sub-molten salt decomposition process can realize the clean extraction of zirconium, which is conducive to the sustainable development of zirconium resources.

Due to the coexistence of Al3+ and RE3+ and their similar properties, the separation of aluminum from rare earths is difficult. In this study, selective precipitation was used to separate aluminum from rare earth chloride solution via electrochemical regulated hydrolysis. By controlling the current density and electrolytic time, the rate of hydroxyl ion production was regulated, and the selective separation of rare earth and aluminum was realized according to the different precipitation sequences. By altering the temperature, current density, pH value, and other parameters, the separation performance of aluminum from rare earth in mixed rare earth chloride systems was systematically investigated. The removal rate of aluminum reached 88.35%, and the loss rate of rare earth was only 5.99% under optimized conditions. Compared with traditional neutralization hydrolysis, the new process showed higher efficiency and lower rare earth loss rate. Furthermore, a kinetic analysis of aluminum precipitation revealed that the reaction adhered to pseudo-first order kinetics. Additionally, the precipitate obtained via separation and filtration was amorphous alumina hydroxide with a small amount of rare earth attached. No reagent was consumed for the new process, which was more efficient and cleaner, providing a new idea for removing aluminum impurities from rare earth solutions.

The natural habitats of the Qinghai-Tibet Plateau (QTP) are fragile and vulnerable to human activities. Moreover, human activities in the QTP have increased rapidly over the past few decades. Thus, it is essential to effectively evaluate the impacts of human activities on the natural habitats in the QTP. However, existing studies have not adequately quantified the comprehensive impacts of human activities. Moreover, there is a lack of analysis of the direct and indirect impacts of human activities. In this study, we used the human footprint index to reflect comprehensive human activities and quantitatively analysed both direct and indirect impacts in the QTP at multiple scales. The results indicated that the human footprint in the QTP increased rapidly by 6.08% from 2000 to 2020. The increased human footprint resulted in habitat quality losses of 0.75%, including 0.41% of direct impacts and 0.34% of indirect impacts. The high-intensity human footprint had the greatest impacts on natural habitats, resulting in a 0.43% reduction in habitat quality, which accounted for 57.40% of the total losses in the QTP. The adverse impacts of human activities on natural habitats further hampered the achievement of biodiversity conservation targets on the QTP. Therefore, we recommend that future efforts in the QTP should focus on the impacts of human activities on natural habitats, especially the impacts of high-intensity human activities.

Abstract Extracting useful knowledge from big data is important for machine learning. When data is privacy-sensitive and cannot be directly collected, federated learning is a promising option that extracts knowledge from decentralized data by learning and exchanging model parameters, rather than raw data. However, model parameters may encode not only non-private knowledge but also private information of local data, thereby transferring knowledge via model parameters is not privacy-secure. Here, we present a knowledge transfer method named PrivateKT, which uses actively selected small public data to transfer high-quality knowledge in federated learning with privacy guarantees. We verify PrivateKT on three different datasets, and results show that PrivateKT can maximally reduce 84% of the performance gap between centralized learning and existing federated learning methods under strict differential privacy restrictions. PrivateKT provides a potential direction to effective and privacy-preserving knowledge transfer in machine intelligent systems.

The extraction of zirconium (Zr) and hafnium (Hf) in methyl isobutyl ketone (MIBK)—thiocyanic acid (HSCN) system has been widely used in the production of nuclear-grade zirconium and hafnium in industry, while the extraction mechanism was not adequately studied. In this study, the extraction and stripping equilibrium of Zr and Hf in the MIBK-HSCN system was studied. The results showed that elevated HCl concentration can increase the distribution ratio of SCN− and decrease that of Zr/Hf in organic phase. In the stripping process, HCl concentration and the Organic/Aqueous (O/A) phase ratio played important roles. The mechanism of the extraction reaction was discussed by considering the stoichiometric relationship of possible reaction equations and corresponding equilibrium constants. The results indicated that SCN− could be extracted into MIBK as HSCN·MIBK. Meanwhile, SCN− could also be extracted into MIBK by complexing with metal (Zr or Hf). The molar ratios of MIBK to the complexes of Zr and Hf have been found to be 5.34 and 5.03, respectively. With the increase in the initial concentration of HCl in the aqueous phase, the complexation molar ratios of SCN− to Zr and Hf increased first and then decreased, and so do the extraction equilibrium constants, which might be due to the extraction competition of HSCN and metal complexes.
