Procedures and mechanisms of bamboo cell wall digestion in the giant panda were studied, in which several indirect methods were combined to avoid spoiling the intactness of the experimental animal. Examinations with surface scanning electron microscopy showed less extensive degradation of bamboo leaf tissues in panda feces than in the residue of sheep rumen digestion. Amounts of remaining fermentable organic matter, as estimated from the amount of volatile fatty acid produced during rumen fermentation in vitro, were much higher in the panda feces than in the residue of sheep rumen digestion. Incubation of fresh excreted panda feces for 12 hr at 37°C simulating extension of the digestion time caused no further progress of digestion. In vitro digestion with gastric juice of the panda showed some decrease of substrate cellulose at lower pH without corresponding reducing sugar production, although neither apparent cellulose digestion nor reducing sugar production was found with pressed fluid of the ...

Stillbirth and neonatal mortality are significant problems in captive breeding of dolphins, however, the causes of these problems are not fully understood. Here, we report a case of meconium aspiration syndrome (MAS) in a male neonate of bottlenose dolphin (Tursiops truncates) who died immediately after birth. At necropsy, a true knot was found in the umbilical cord. The lungs showed diffuse intraalveolar edema, hyperemic congestion and atelectasis due to meconium aspiration with mild inflammatory cell infiltration. Although the exact cause of MAS in this case was unknown, fetal hypoxia due possibly to the umbilical knot might have been associated with MAS, which is the first report in dolphins. MAS due to perinatal asphyxia should be taken into account as a possible cause of neonatal mortality and stillbirth of dolphin calves.

A 20-year-old Gentoo penguin was found dead with a clinical history of inappetence and dyspnoea. At necropsy, the lungs showed severe congestion/hemorrhage and atelectasis. Histopathologically, fibrosis was observed exclusively around parabronchi with severe collagen deposition. In fibrotic lesions, there were numerous depositions of crystalline structures accompanied by epithelioid cells and multinucleated giant cells (foreign body type). In addition to irregularly lamellar structures as the morphology, the crystals were demonstrated calcium oxalate (CaOx) by the Alizarin red S staining with and without polarized light and von Kossa's staining. Myocobacteria and fungi were not found by special and immuohistochemical stainings. Pulmonary dystrophic oxalosis is a very rare lesion in Gentoo penguin.

A 10-year-old female false killer whale (Pseudorca crassidens) developed skin lesions in the left breast fin. Histopathologically, the lesions consisted of multiple granulomas spread diffusely into the deep dermis and bone; characteristically, each granuloma had septate, branching fungal hyphae and chlamydospores surrounded by eosinophilic Splendore-Hoeppli materials. Macrophages, epithelioid cells and multinucleated giant cells in the granulomas reacted mainly to anti-SRA-E5 antibody against human macrophage scavenger receptor type I. Fusarium solani was isolated and its gene was detected from the skin samples. Mycotic skin lesions by Fusarium spp. reported so far in marine mammals were regarded as superficial dermatitis; therefore, the present case is very uncommon in that the lesions spread deeper into the skin.

Two digestion trials by total feces collection method were conducted, using a male adult giant panda in captivity fed feedstuffs including bamboo. Collected pieces of feces were assorted according to undigested feedstuff mainly forming each piece of feces, and analyzed separately. The data were converted into undigested nutrients of each feedstuff with a calculating procedure devised to assess respective digestibility. Cell wall constituents of bamboo were apparently digested to a certain extent, although the digestibility was low (24.1% and 15.0% in Experiment 1 and 2, respectively, in the case of NDF (neutral detergent fiber)). Cellular contents of the bamboo were also less digestible compared with those of the other feedstuffs, while nutrients in less fibrous feedstuffs (concentrates) were digested fairly well (more than 90%). A 10-year-old female sea otter exhibited convulsions, arrhythmia, hyperthermia, forced breathing and anorexia and died after a week. Histopathological examination revealed neoplastic proliferation of small round cells with scant cytoplasm and round or oval nuclei distributed mainly in the thalamus. The proliferation of neoplastic cells was observed in the cerebral parenchyma and perivascular areas. The neoplastic cells were immunopositive for CD3, but not CD20. No neoplastic proliferation of T-cells was found in other organs. Taken together, we diagnosed this case as a primary cerebral T-cell lymphoma. To our knowledge, this is the first case of primary cerebral T-cell lymphoma in a sea otter.

A milk sample from a captive giant panda (Ailuropoda melanoleuca), obtained at 13 days postpartum, contained 7.1% protein, 1.6% carbohydrate, 10.4% lipid and 0.9% ash. The ratio of casein to whey proteins was 5.0:2.1. Sodium dodecyl sulfate polyacrylamide electrophoresis (SDS-PAGE) of the whey protein fraction showed the presence of at least two major proteins other than alpha-lactalbumin and beta-lactoglobulin. SDS-PAGE and urea-gel electrophoresis showed that alphas-casein is not a major component. The proportions of triacylglycerol, cholesterol, cholesterol esters and phospholipid were 90.5, 5.3, 0.96 and 3.1%, of the total lipid, respectively. The dominant saccharide in the panda milk was Gal(alpha1-3)Gal(beta1-4)Glc (isoglobotriose). The milk contained, in addition, lesser amounts of lactose, Gal(alpha1-3)Gal(beta1-4)[Fuc(alpha1-3)]Glc (fucosyl isoglobotriose), Neu5Ac(alpha2-3)Gal(beta1-4)Glc (3'-N-acetylneuraminyl-lactose), Neu5Ac(alpha2-6)Gal(beta1-4)Glc (6'-N-acetylneuraminyl-lactose) and Neu5Ac(alpha2-3)Gal(beta1-4)[Fuc(alpha1-3)]Glc.
