Rita Muzzalupo, Lorena Tavano Department of Pharmacy and Health and Nutrition Science, University of Calabria, Edificio Polifunzionale, Cosenza, Italy Abstract: Niosomes are vesicular nanocarriers and are receiving much attention as potential transdermal drug delivery systems due to properties such as enhanced drug penetration, local depot for sustained drug release, and a rate-limiting membrane for modulation of systemic absorption of drugs via the skin. Several mechanisms have been proposed to explain the ability of niosomes to increase drug transfer through the skin. Niosomal carriers are suitable for the transdermal delivery of numerous pharmacological agents, including antioxidant, anticancer, anti-inflammatory, antimicrobial, and antibacterial molecules, and this review attempts to provide an exhaustive collection of recent investigations in this interesting field, with special emphasis on the strategies used to enhance the potential of niosomes. Abstract

In recent years, significant efforts have been devoted to the application of nanotechnology for the development of devices to be employed as drug delivery systems. The synergic combination of polymeric science, drug delivery concepts and material technology offers challenging and precious opportunities for the achievement of new useful materials. In particular surfactants, lipids and polymeric systems play an important role in modern drug delivery, where they may allow control of the drug release rate, enhance effective drug solubility, minimize its degradation, contribute to reduced its toxicity and facilitate control of its uptake. In all, they contribute significantly to therapeutic efficiency. Abstract

Physicochemical characteristics of cationic vesicular systems prepared from biocompatible diacyl glycerol-arginine surfactants are investigated. These systems form stable cationic vesicles by themselves and the average diameter of the vesicles decreases as the alkyl chain length of the surfactant increases. The addition of DPPC also modifies the physicochemical properties of these vesicles. Among the drugs these cationic formulations can encapsulate, we have considered Ciprofloxacin and 5-Fluorouracil (5-FU). We show that the percentage of encapsulated drug depends on both the physicochemical properties of the carrier and the type of drug. The capacity of these systems to carry different molecules was evaluated performing in vitro drug release studies. Finally, the antimicrobial activity of empty and Ciprofloxacin-loaded vesicles against Gram-positive and Gram-negative bacteria has been determined. Three bacteria were tested: Escherichia coli, Staphylococcus aureus and Klebsiella pneumoniae. The in vitro drug release from all formulations was effectively delayed. Empty cationic vesicles showed antimicrobial activity and Ciprofloxacin-loaded vesicles showed similar or higher antimicrobial activity than the free drug solution. These results suggest that our formulations represent a great innovation in the pharmaceutical field, due to their dual pharmacological function: one related to the nature of the vehiculated drug and the other related to the innate antibacterial properties of the surfactant-based carriers. © 2014 Elsevier B.V.

40. Role of aggregate size in the hemolytic and antimicrobial activity of colloidal solutions based on single and gemini surfactants from arginine

Abstract

Cationic colloidal systems composed of arginine based surfactants (single or gemini structures) and membrane additive compounds such as DLPC or cholesterol have been characterized by means of size distribution and zeta-potential measurements. The single or monocatenary surfactant (LAM) as well as the gemini with the shortest spacer chain (C6(LA)2) formed micelles, while aqueous solutions of pure gemini surfactants with longer spacers (C9(LA)2 and C12(LA)2) formed very big aggregates. The addition of phospholipids or cholesterol changed drastically the aggregation behaviour. In the case of LAM and C 6(LA)2, the incorporation of additives gave rise to the formation of cationic vesicles. For C9(LA)2 and C 12(LA)2, this type of additives promoted the formation of smaller aggregates. All the formulations had positive zeta-potential values and in general exhibited high colloidal stability. We also evaluated the hemolysis and the antimicrobial activity of these systems. The capability of disrupting erythrocyte membranes depends on the hydrophobicity of the molecules and the size of aggregates in the solution. Gemini surfactants with short spacer chains are more hemolytic than their single chain homologue, while gemini surfactants with long spacers are much less hemolytic than their single chain counterpart. Moreover, for the same formulation, the hemolysis depends on the initial concentration of the stock solution used to set up the hemolysis/concentration curve. Results show that small aggregates interact easily with these biological membranes. The alkyl spacer chain and the presence of additives also play an important role in the antimicrobial activity, and, in general, the interaction with bacteria and erythrocytes is affected by the same parameters. The physico-chemical and biological characterization of these systems might be important for several biotechnological applications in which cationic vesicular systems are involved. © The Royal Society of Chemistry 2013.

44. Spontaneous temperature-sensitive Pluronic® based niosomes: Triggered drug release using mild hyperthermia.

Abstract

Inclusion of lipids or polymers with a transition temperature closer to physiological body temperature (40–42 °C) is a strategy used in tumor therapy for more than 30 years, because it allows induction of drug release from delivery systems by mild hyperthermia. Unfortunately, most of these thermo-sensitive carriers are removed from circulation before completion of their function. Thus, novel multi-functional niosomes possessing spontaneous stealth and thermo-sensitive properties were developed from L64 Pluronic ® and L64ox as its derivative, in presence or absence of cholesterol. The use of L64 both as amphiphilic constituent and thermo-sensitive molecule, gave the possibility to bypass the use of additional excipients and increased the system biocompatibility. Niosomes diameter ranged from 400 to 750 nm and were long term stable. Calcein and 5-FU possess great affinity to niosomal matrices rich in PEO groups. Negative Z-potential values were attributed to the negative charges onto the niosomes surface and generally change according to the temperature. The in vitro drugs release studies were performed at 25 °C, 37 °C and 42 °C, that are representative of certain conditions (storage, physiological condition and mild hyperthermia, respectively). Results showed that L64-based niosomes possess spontaneous thermo-sensitive properties: drugs releases were found to be more pronounced at 42 °C. These early results are a promising first step for the development of multi-functional devices that combine several advantages such as stealth properties and temperature controllability at the desired location and time, for a more specific and efficient pharmacological therapy. 45. Nanovesicular Formulations for Cancer Gene Therapy

Abstract

In the last decades, gene therapy has become a novel therapeutic strategy for cancer treatment, including immunologic and molecular approaches. Among molecular avenue, the design of efficient and effective gene delivery systems, like cationic liposomes and niosomes, has been widely investigated and proposed as the most promising research area. The advantages of cationic vesicles rely on their natural ability to form complexes with anionic genetic molecules and deliver them into the cells via the endosomal pathway. Obviously, cationic vesicles- mediated gene delivery is affected by numerous factors, in particular composition, that strongly affects vesicle physical-chemistry characteristics and transfection effectiveness. This review will analyse the potential of cationic nanocarriers in cancer gene therapy, focusing on the role of liposomes and niosomes as vesicular devices and giving an exhaustive collection of the most representative investigations.

Abstract

In this study, we report the results of our investigations on the percutaneous permeation profiles of Diclofenac sodium, Paracetamol, Propanolol hydrochloride, and α-Tocopherol from the different lyotropic liquid crystalline phases obtained by Pluronic P105/water mixtures, in order to understand if the particular assembly shown in the formulations could influence the delivery across the skin. Recent studies have focused on the Pluronic liquid crystalline phases to evaluate the potential use of these phases in drug delivery, but no comparative investigation has been yet performed on the drug permeation from the different liquid crystalline phases obtained by the same Pluronic surfactant. The cubic, hexagonal, and lamellar mesophases (loading the above-mentioned drug) were characterized by Deuterium Nuclear Magnetic Resonance spectroscopy and Polarized Optical Microscopy observations. Results revealed that the liquid crystalline gel microscopic structure obtained in the different formulations drastically affects the drug percutaneous availability. Abstract

In the present work, we report the synthesis of cellulose hydrogel containing ferulic moieties and the evaluation of its antioxidant and scavenger activity. Acrylic groups were inserted onto cellulose backbone by a heterogeneous synthesis to produce two cellulose monomers with different degree of substitution (DS). The radical copolymerization of acrylcellulose (AcrC) with N,N-dimethylacrylamide (DMAA) was carried out in NH3/Urea aqueous solution, in a range of composition between 24 and 60 wt% of AcrC. The obtained hydrogels were characterized by infrared spectroscopy (FT-IR). Their equilibrium swelling degree (α%) was evaluated. They showed good swelling behavior in simulating gastric, intracellular and intestinal fluids and no more different at various pH. The ferulic moieties were directly grafted on the free hydroxylic groups of cellulose hydrogel by acylation, using dicyclohexylcarbodiimide (DCC) and 4-hydroxybenzotriazole (HBT) as condensation agents. Finally, the antioxidant activity in inhibiting the lipid peroxidation, in rat-liver microsomal membranes, induced in vitro by two different sources of free radicals, 2,2′-azobis (2-amidinopropane) (AAPH) and tert-butyl hydroperoxide (tert-BOOH), was evaluated. The effects of scavenging DPPH (1,1-diphenyl-2-picrylhydrazyl) radicals were also investigated. Hydrogel was found to be very efficient scavengers of DPPH radicals. The results strongly suggested that the antioxidant hydrogel neutralize free radicals. This biomaterial could be successfully applied in pharmaceutical field both as prodrug of trans-ferulic acid than as carrier for photo and thermo-degradable drugs to improve their stability.
