Abstract

ABSTRACT Impact microindentation (IMI) is a technique to assess bone material properties of the cortical bone at the tibia in a transcutaneous, microinvasive, way. IMI is increasingly used in studies evaluating the contribution of tissue material properties to bone fragility in humans, and is approved for use in the clinic in Europe and the United States. Previous data show that IMI is well tolerated during and immediately after the procedure. The aim of this prospective observational study was to evaluate the longer‐term safety and acceptability of an IMI measurement using the handheld OsteoProbe device®. Included were patients who were scheduled for a measurement at the Leiden University Medical Center from September 2019 to December 2020 and willing to participate. Patients were asked to review the procedure right after the measurement, and by telephone interviews 1 week and 1 month thereafter. The primary outcome was the 30‐day complication rate after the measurement. Included were 106 patients (71 women) with a median age of 59 years (range, 20 to 86 years). Only three minor events were reported by 1‐week follow‐up, with an overall 30‐day event rate of 2.8%. These were a very small hematoma in two patients, and a small bruise in one patient, all of which resolved without medical intervention. No other safety‐related concerns were observed, and all 106 patients would undergo the measurement again if needed. The vast majority had no pain at baseline, 1‐week and 1‐month follow‐up (80.2%, 88.4% and 94.3%, respectively). In this first and large longitudinal study we demonstrated that although minimally‐invasive, IMI using the OsteoProbe® device at the tibia did not lead to any complications, and was well accepted by patients. Results strongly suggest that IMI can be safely used in studies as well as in the clinic in the hands of an experienced operator. © 2023 The Authors. This article reviews existing evidence on etiology, associated symptoms, and management of sleep problems associated with chemotherapy treatment during cancer. It also discusses limitations and methodological issues of current research. The existing literature suggests that subjectively and objectively measured sleep problems are the highest during the chemotherapy phase of cancer treatments. A possibly involved mechanism reviewed here includes the rise in the circulating proinflammatory cytokines and the associated disruption in circadian rhythm in the development and maintenance of sleep dysregulation in cancer patients during chemotherapy. Various approaches to the management of sleep problems during chemotherapy are discussed with behavioral intervention showing promise. Exercise, including yoga, also appear to be effective and safe at least for subclinical levels of sleep problems in cancer patients. Numerous challenges are associated with conducting research on sleep in cancer patients during chemotherapy treatments and they are discussed in this review. Dedicated intervention trials, methodologically sound and sufficiently powered, are needed to test current and novel treatments of sleep problems in cancer patients receiving chemotherapy. Optimal management of sleep problems in patients with cancer receiving treatment may improve not only the well-being of patients, but also their prognosis given the emerging experimental and clinical evidence suggesting that sleep disruption might adversely impact treatment and recovery from cancer.Keywords: cancer, sleep, chemotherapy, intervention, circadian rhythm, cognitive behavioral therapy

Abstract

Importance: Nonclinical studies suggest that the combination of poly(ADP-ribose) polymerase and programmed cell death 1/programmed cell death-ligand 1 inhibitors has enhanced antitumor activity; however, the patient populations that may benefit from this combination have not been identified. Objective: To evaluate whether the combination of avelumab and talazoparib is effective in patients with pathogenic BRCA1/2 or ATM alterations, regardless of tumor type. Design, Setting, and Participants: In this pan-cancer tumor-agnostic phase 2b nonrandomized controlled trial, patients with advanced BRCA1/2-altered or ATM-altered solid tumors were enrolled into 2 respective parallel cohorts. The study was conducted from July 2, 2018, to April 12, 2020, at 42 institutions in 9 countries. Interventions: Patients received 800 mg of avelumab every 2 weeks and 1 mg of talazoparib once daily. Main Outcomes and Measures: The primary end point was confirmed objective response (OR) per RECIST 1.1 by blinded independent central review. Results: A total of 200 patients (median [range] age, 59.0 [26.0-89.0] years; 132 [66.0%] women; 15 [7.5%] Asian, 11 [5.5%] African American, and 154 [77.0%] White participants) were enrolled: 159 (79.5%) in the BRCA1/2 cohort and 41 (20.5%) in the ATM cohort. The confirmed OR rate was 26.4% (42 patients, including 9 complete responses [5.7%]) in the BRCA1/2 cohort and 4.9% (2 patients) in the ATM cohort. In the BRCA1/2 cohort, responses were more frequent (OR rate, 30.3%; 95% CI, 22.2%-39.3%, including 8 complete responses [6.7%]) and more durable (median duration of response: 10.9 months [95% CI, 6.2 months to not estimable]) in tumor types associated with increased heritable cancer risk (ie, BRCA1/2-associated cancer types, such as ovarian, breast, prostate, and pancreatic cancers) and in uterine leiomyosarcoma (objective response in 3 of 3 patients and with ongoing responses greater than 24 months) compared with non-BRCA-associated cancer types. 5. Effects of green light supplementation with red and blue combinations of LED light spectrums on the growth and transcriptional response of Haematococcus pluvialis.

Abstract

Light management strategy is crucial for improving microalgal production in terms of higher biomass and economically valuable bioactive molecules. However, green light has received less attention in developing light managements for algae and higher plant due to its low absorption rate by chlorophyll. In this study, the effects of green light supplementation, in the combination with red and blue light were investigated in Haematococcus pluvialis. 10% and 20% of green light supplementations were applied in 3:2 ratios of red and blue LED light combinations as an expense of red‐light. Growth rates, chlorophyll concentration, and dry weight were measured to assess the growth kinetics of H. pluvialis along with the relative transcript accumulations of four mRNAs: Rubisco, PTOX2, PsaB, and PsbS. Growth rates, chlorophyll concentrations and dry weight were found significantly higher in presence of 10% green light supplementation compared to red and blue light combinations. The relative transcript accumulations of Rubisco and PsbS genes showed significant upregulation at the end of the experiments (with the fold change of 42.91 ± 12.08 and 98.57 ± 27.38, respectively, relative to the beginning of the experiments) compared to combinations of red and blue light (fold change of 19.09 ± 3.0 and 47.77 ± 14.21, respectively, relative to beginning of the experiments). PsaB and PTOX2 transcripts did not show significant accumulation differences between treatments. It seems that green light has a dose dependent additive effect on the growth rate of H. pluvialis. The upregulation of Rubisco and PsbS may indicate green light dependent carbon assimilation and light‐harvesting response in H. pluvialis. [ABSTRACT FROM AUTHOR]

Abstract

Octahedral tilt transitions in epitaxial AgTa0.5Nb0.5O3 (ATN) films grown on (001)p (where p=pseudocubic) oriented SrRuO3/LaAlO3 and LaAlO3 substrates were characterized by electron diffraction and high resolution x-ray diffraction. It was found that the ATN films exhibited octahedral rotations characteristic of the Pbcm space group, similar to those seen in bulk materials; however, the temperature of the M3↔M2 phase transition has been suppressed by ∼250 K due to the fact that the correlation length for rotations about cp was significantly reduced. The average off-center B-cation displacements, which signify the degree of long-range order for these local cation positions, were negligibly small compared to bulk materials, as inferred from the near-zero intensity of the 1/4(00l)p-type reflections. On cooling, pronounced ordering of B-cation displacements occurred at ≈60 K which is significantly lower compared to bulk (≈310 K). The onset of this ordering coincides with a broad maximum in relative permittivity as a function of temperature. It is believed that point and planar defects in thin ATN films disrupt the complex sequence of in-phase and antiphase rotations around cp thereby reducing the effective strength of interactions between the tilting and cation displacements. Abstract

Epitaxial silver tantalate niobate, Ag(Ta0.5Nb0.5)O3 (ATN), thin films on (001)SrRuO3/(001)LaAlO3 substrates were deposited by a chemical solution deposition route. The solution was prepared using niobium ethoxide, tantalum ethoxide, and silver nitrate precursors in a 2-methoxyethanol and pyridine solvents. Films were crystallized between 450 and 800 °C in O2 ambient. X-ray diffraction data demonstrated cube-on-cube epitaxy, with a limited reaction between the ATN thin films and the SrRuO3. The dielectric constants of the epitaxial films improved from 260 to 360 at 100 kHz with an increase in the crystallization temperatures from 500 to 800 °C. It is speculated that the phase transition between the orthorhombic M2 and M3 phases (which occurs at ∼330 K in ceramics) is shifted to lower temperatures (<=99 K) for the films. For a film that was crystallized at 750 °C, the frequency dependence was small (<=20.3%) within the range of 1 Hz–100 kHz, the capacitance change, ΔC/C, was small (<=12.1%) over the temperature range of -20–120 °C with a corresponding temperature coefficient of capacitance of -0.11%/°C at room temperature at 100 kHz, and the dc field tunability was not hysteretic, but was limited to 4.8% with a 150 kV/cm bias. Abstract

Glass-metal joining is important for improving efficiencies of heat collecting units employed in linear parabolic sun collector systems. Soda lime silicate glass tube-titanium inset joining was made by using propane/O2 flame working in air. Optic and scanning electron microscope investigations of joined samples showed oxidation of titanium in air and its reaction with glass leading bubble formation. ANSYS14 Multiphysics modeling of residual joining stress levels suggested that titanium inset and soda lime silicate glass had rather low residual stress levels due to close thermal expansion coefficients. For the used sample dimensions with uniform glass tube shape, highest residual maximum principal stress was ~60 MPa and minimum principal stress was ~-40 MPa for the glass at joining interface. However, glass shape was found to be affecting residual stresses during bonding and shape defects like groove formation increased residual minimum principal stress levels to -90 MPa near joining interface. Some microcracking of glass was also observed in groove region. Bubble formation in glass near reaction interface was also found to be involving crack formation and its propagation as well. Therefore, controlling glass shape and interfacial reaction was found to be important for successful titanium inset-soda lime silicate glass tube joining by flame working in air. Abstract

In this study, the presence of Campylobacter jejuni in water lines of commercial broiler house and its role in the epidemiology of the infection of broiler flocks was investigated. The study was done in three sequential commercial broiler flocks previously known to be infected with C. jejuni in two poultry houses with different water sources. C. jejuni was identified in drinking water and drinking nipple swab samples in water-line samples from both houses. Fresh fecal dropping samples were taken from broiler flocks for determination of C. jejuni-carriage. Twenty and 130 C. jejuni isolates were recovered from water-line system and fecal dropping samples, respectively. A total of 150 C. jejuni isolates were genotyped by pulsed-field gel electrophoresis (PFGE) with SmaI digestion and 9 distinct PFGE patterns were identified. Six and 5 different PFGE types were identified in houses 1 and 2, respectively. C. jejuni isolates, recovered from water lines samples, were genotypically similar to the isolates from fresh fecal dropping in both houses. These results showed that C. jejuni water-line contamination was related to flock contamination and could help to continuously make it infected with C. jejuni.

Abstract

The purpose of this study was to investigate the prevalence and aetiology of carpal tunnel syndrome (CTS) and the effect of open carpal tunnel decompression (CTD) in a group of patients on renal haemodialysis. In 91 patients attending a Renal Unit who were assessed only by clinical means, the prevalence of CTS was 7.1%. CTS development was not significantly correlated with known risk factors, in particular duration of dialysis and presence of an artero-venous fistula on the symptomatic limb. CTD led to a mean symptom-free period of 12 years. In summary, CTS is a frequent and serious disease affecting renal dialysis patients, but its aetiology remains uncertain. Nevertheless, CTD is an effective and lasting treatment for this condition.

Abstract

Glass-to-metal joining is important for improving efficiencies of heat collecting units, employed in parabolic solar collecting systems. In this study, soda lime silicate glass was joined to titanium in air at 700, 800 and 900 °C for 5 min by controlled heat treatments. Glass-to-titanium bonding was good and no delamination or macro cracking in glass was observed. During heat treatment, titanium is oxidized in air prior to the reaction with glass. The reaction between oxidized titanium and glass results in bubble formation at joining interface. Bubble formation was found to be decreased with decreasing joining temperature. Thermal stresses developed at glass-titanium junction were modeled by employing ANSYS 14 multiphysics simulation software. For the studied sample profile, soda lime silicate glass, close to the joining interface, experienced rather low residual stresses of the order of 10 MPa maximum and -40 MPa minimum principal stress levels. Good glass-to-titanium bonding and low thermal residual stress are mainly due to similar thermal expansion coefficients of these materials, which was found to be important for achieving successful joining in air.
