Abstract Separating the dam body, spillway, and other structures from the point cloud in the dam area is an important step in dam deformation monitoring. Manual segmentation is time consuming and inaccurate. This study proposes a point cloud segmentation neural network model based on normal vector optimization suitable for dam environment: (1) This model utilizes the voxel uniform sampling method of equal length cubes to solve the problem of uneven point cloud density caused by wide range and long-distance measurement during point cloud measurement in dam areas. (2) Designed block input and combined output modules in the model, achieving efficient input of large volume point cloud and eliminating the impact of interpolation points offset during seq2seq model decoding process. (3) In response to the diverse characteristics of point cloud normal vectors presented by vegetation, rock mass, and complex dam structures in the dam area, this paper proposes an adaptive radius plane fitting vector estimation method based on eigenvalue method to improve the accuracy of segmentation. The experiment on the prototype arch dam shows that the proposed normal estimation method improves the classification accuracy of PointNet + + from 96.26 to 98.27%. Compared with the other three normal estimation methods (2-jets, Hough CNN, iterative PCA), the overall accuracy is improved by 0.82%, 1.22%, and 0.22%, and the joint average intersection is improved by 0.0293, 0.0325, and 0.0104. The prototype arch dam experiment shows that our proposed method has a segmentation accuracy of 98.27%. Compared with 2-jets, Hough CNN, and iterative PCA, the overall accuracy has been improved by 0.82%, 1.22%, and 0.22%. This study provides a high-precision segmentation scheme for applications such as deformation detection of dam components based on point clouds.

Abstract Osteoblasts and osteoclasts play an important role in maintaining the structural integrity of bone tissue, in which osteoclasts degrade bone structure and osteoblasts restore bone tissue. The imbalance of osteoblast and osteoclast function can lead to many bone-related diseases, such as osteoporosis and inflammatory osteolysis. The drug that can both promote bone formation and inhibit bone loss will be able to treat those diseases. In this study, it was found that LMK-235, an selective HDAC4/5 inhibitor, inhibited the differentiation and maturation of osteoclasts by regulating NF-κB and p-Smad2/3 signaling pathways via inhibition of HDAC4. At the same time, we found that LMK-235 promoted osteoblast mineralization by upregulating Runx2 expression via inhibition of HDAC4. In vivo, LMK-235 was able to alleviate lipopolysaccharide (LPS)-induced calvarial osteolysis and promote the repair of bone defects. Taken together, LMK-235 suppresses osteoclast differentiation and promotes osteoblast formation by inhibiting HDAC4. This may provide a valuable treatment for bone diseases caused by abnormal osteoclast bone resorption and osteoblast bone regeneration.

Heterogeneous karst surfaces exerted scaling effects whereby specific runoff decrease with increasing area. The existing RUSLE-L equations are limited by the default implicit assumption that the surface-runoff intensity is constant at any slope length. The objective of this study was to modify the L-equation by establishing the functional relationship between surface-runoff intensity and karst slope length, and to evaluate its predictive capability at different resolution DEMs. Transfer grid layers were generated based on the area rate of surface karstification and considered the runoff transmission percentage at the exposed karst fractures or conduits to be zero. Using the multiple flow direction algorithm united with the transfer grid (MFDTG), the flow accumulation of each grid cell was simulated to estimate the average surface-runoff intensity over different slope lengths. The effectiveness of MFDTG algorithm was validated by runoff plot data in Southwestern China. The simulated results in a typical peak-cluster depression basin with an area rate of surface karstification of 6.5% showed that the relationship between surface-runoff intensity and slope length was a negative power function. Estimated by the proposed modified L-equation ((alx(b+1)/22.13)m), the L-factor averages of the study basin ranged from 0.35 to 0.41 at 1, 5, 25 and 90 m resolutions respectively. This study indicated that the modified L-equation enables an improved prediction of the much smaller L-factor and the use of any resolution DEMs on karst landscapes. Particular attention should be given to the variation of surface-runoff intensity with slope length when predicting L-factor on hillslopes with runoff scale effect.

High-resolution remote sensing imagery, reaching meter or sub-meter levels, provides essential data for extracting and identifying road information. However, rural roads are often narrow, elongated, and have blurred boundaries, with textures that resemble surrounding environments such as construction sites, vegetation, and farmland. These features often lead to incomplete extraction and low extraction accuracy of rural roads. To address these challenges, this study introduces the RC-MSFNet model, based on the U-Net architecture, to enhance rural road extraction performance. The RC-MSFNet model mitigates the vanishing gradient problem in deep networks by incorporating residual neural networks in the downsampling stage. In the upsampling stage, a connectivity attention mechanism is added after dual convolution layers to improve the model’s ability to capture road completeness and connectivity. Additionally, the bottleneck section replaces the traditional dual convolution layers with a multi-scale fusion atrous convolution module to capture features at various scales. The study focuses on rural roads in the Xiong’an New Area, China, using high-resolution imagery from China’s Gaofen-2 satellite to construct the XARoads rural road dataset. Roads were extracted from the XARoads dataset and DeepGlobe public dataset using the RC-MSFNet model and compared with some models such as U-Net, FCN, SegNet, DeeplabV3+, R-Net, and RC-Net. Experimental results showed that: (1) The proposed method achieved precision (P), intersection over union (IOU), and completeness (COM) scores of 0.8350, 0.6523, and 0.7489, respectively, for rural road extraction in Xiong’an New Area, representing precision improvements of 3.8%, 6.78%, 7.85%, 2.14%, 0.58%, and 2.53% over U-Net, FCN, SegNet, DeeplabV3+, R-Net, and RC-Net. (2) The method excelled at extracting narrow roads and muddy roads with unclear boundaries, with fewer instances of omission or false extraction, demonstrating advantages in complex rural terrain and areas with indistinct road boundaries. Accurate rural road extraction can provide valuable reference data for urban development and planning in the Xiong’an New Area.

Intelligent control algorithms have been extensively utilized for adaptive controller parameter adjustment. While the Particle Swarm Optimization (PSO) algorithm has several issues: slow convergence speed requiring a large number of iterations, a tendency to get trapped in local optima, and difficulty escaping from them. It is also sensitive to the distribution of the solution space, where uneven distribution can lead to inefficient contraction. On the other hand, the Beetle Antennae Search (BAS) algorithm is robust, precise, and has strong global search capabilities. However, its limitation lies in focusing on a single individual. As the number of iterations increases, the step size decays, causing it to get stuck in local extrema and preventing escape. Although setting a fixed or larger initial step size can avoid this, it results in poor stability. The PSO algorithm, which targets a population, can help the BAS algorithm increase diversity and address its deficiencies. Conversely, the characteristics of the BAS algorithm can aid the PSO algorithm in finding the optimal solution early in the optimization process, accelerating convergence. Therefore, considering the combination of BAS and PSO algorithms can leverage their respective advantages and enhance overall algorithm performance. This paper proposes an improved algorithm, W-K-BSO, which integrates the Beetle Antennae Search strategy into the local search phase of PSO. By leveraging chaotic mapping, the algorithm enhances population diversity and accelerates convergence speed. Additionally, the adoption of linearly decreasing inertia weight enhances algorithm performance, while the coordinated control of the contraction factor and inertia weight regulates global and local optimization performance. Furthermore, the influence of beetle antennae position increments on particles is incorporated, along with the establishment of new velocity update rules. Simulation experiments conducted on nine benchmark functions demonstrate that the W-K-BSO algorithm consistently exhibits strong optimization capabilities. It significantly improves the ability to escape local optima, convergence precision, and algorithm stability across various dimensions, with enhancements ranging from 7 to 9 orders of magnitude compared to the BAS algorithm. Application of the W-K-BSO algorithm to PID optimization for the Pointing and Tracking System (PTS) reduced system stabilization time by 28.5%, confirming the algorithm’s superiority and competitiveness.

Background: Cognitive impairment, increasingly recognized as a major social burden, is commonly found in chronic kidney disease (CKD) patients. Summary: Vascular damage, uremic toxicity, oxidative stress, and peripheral/central inflammation induced by CKD might be involved in brain lesions and ultimately result in cognitive decline. Uncovering the pathophysiology of CKD-associated cognitive impairment is important for early diagnosis and prevention, which undoubtedly prompts innovative pharmacological treatments. Key Messages: Here, we sequentially review the current understanding and advances in the epidemiology, risk factors, and pathological mechanisms of cognitive impairment in CKD. Furthermore, we summarize the currently available therapeutic strategies for cognitive impairment in CKD.

Abstract Background New therapies are urgently needed for Alzheimer’s disease (AD). Sodium oligomannate (GV-971) is a marine-derived oligosaccharide with a novel proposed mechanism of action. The first phase 3 clinical trial of GV-971 has been completed in China. Methods We conducted a phase 3, double-blind, placebo-controlled trial in participants with mild-to-moderate AD to assess GV-971 efficacy and safety. Participants were randomized to placebo or GV-971 (900 mg) for 36 weeks. The primary outcome was the drug-placebo difference in change from baseline on the 12-item cognitive subscale of the Alzheimer’s Disease Assessment Scale (ADAS-cog12). Secondary endpoints were drug-placebo differences on the Clinician’s Interview-Based Impression of Change with caregiver input (CIBIC+), Alzheimer’s Disease Cooperative Study-Activities of Daily Living (ADCS-ADL) scale, and Neuropsychiatric Inventory (NPI). Safety and tolerability were monitored. Results A total of 818 participants were randomized: 408 to GV-971 and 410 to placebo. A significant drug-placebo difference on the ADAS-Cog12 favoring GV-971 was present at each measurement time point, measurable at the week 4 visit and continuing throughout the trial. In this study, the degree of bleaching of multi-grain coarse quartz optically stimulated luminescence (OSL) signal of near-surface aeolian samples collected around the Tengger Desert is assessed. The single-aliquot regenerative dose (SAR) protocol and the standardized growth curve (SGC) method are applied to measure the equivalent dose (De) of these samples. The bleaching degree of the samples is assessed by investigating 1) the relationship between Ln/Tn and SAR De and 2) the SGC De distribution. Various degrees of heterogeneously-bleached multi-grain dose distributions synthetized with a numerical simulation method is further used to validate the bleaching performance of the samples. It demonstrates that the investigated samples are characterized by tight De distributions and the maximum De estimate is smaller than 1.1 Gy. The numerical simulation method which uses as input a large proportion of fully-bleached grains and a small baseline dose is able to reproduce multi-grain De distributions similar to the measured ones. We conclude that OSL signals of multi-grain coarse quartz extracted from most of the investigated aeolian samples are fully bleached before deposition.

The precise fabrication of artificially designed molecular complexes into ordered structures resembling their natural counterparts would find broad applications but remains a major challenge in the field. Here we genetically design chitin-binding domain (CBD)-containing amyloid proteins, and rationally fabricate well-ordered CBD-containing functional amyloid-chitin complex structures by coupling a top-down manufacturing process with a bottom-up self-assembly. Our fabrication approach starts with the dissolution of both CBD-containing functional amyloid and chitin in hexafluoroisopropanol (HFIP) to make a hybrid ink. This hybrid ink platform, coupled with multiple fabrication methods including airbrushing, electrospinning and soft-lithography, produces a series of unique freestanding structures. The structural features of the products, such as the ability to direct the light path and mimicking of the extracellular matrix enable applications in functional light gratings and cell culture, respectively. Further genetic engineering of the protein component allowed tunable functionalization of these materials, including nanoparticle immobilization and protein conjugation, resulting in broad applications in electronic devices and enzyme immobilization. Our technological platform can drive new advances in biocatalysis, tissue engineering, biomedicine, photonics and electronics.

The nitrogen isotope composition of plants (δ15N) can comprehensively reflect information on climate change and ecosystems’ nitrogen cycle. By collecting common herbs and soil samples along the 400 mm isoline of mean annual precipitation (MAP) in the agro-pastoral zone of North China (APZNC) and measuring their δ15N values, the statistical characteristics of foliar δ15N of herbs and the responses of foliar δ15N to the MAP and mean annual temperature (MAT) were analyzed. The results showed that: (1) the δ15N values of all herbs investigated varied from −5.5% to 15.25%. Among them, the δ15N value range of C3 herbs (−5.5~15.00%) was wider than that of C4 herbs (−2.17~15.25%), but the average value (3.27%) of C3 herbs was significantly lower than that of C4 herbaceous plants (5.55%). This difference provides an important method for identifying plants of different photosynthetic types by nitrogen isotope technology. (2) Along the transect from northeast to southwest, the δ15N of both C3 and C4 herbs decreased with the increase in the MAP, but not significantly for C3 herbs. The inverse relationship between the nitrogen isotopic signatures of herbs and MAP is consistent with previous studies. However, the MAP in the APZNC is found to only explain a small amount of the observed variance in the δ15N herbs (C3 herbs: 10.40%; C4 herbs: 25.03%). (3) A strong negative relationship was found between δ15N of herbs and MAT across the transect (C3 herbs: −0.368%/°C; C4 herbs: −0.381%/°C), which was contrary to the global pattern and some regional patterns. There was no significant difference in the δ15N responses of two different photosynthetic herbs to temperature, but the effect of temperature on the variances of δ15N of C3 and C4 herbs was significantly greater than that of precipitation. This suggests that temperature is a key factor affecting foliar δ15N of herbs in this transect. The above findings may be of value to global change researchers studying the processes of the nitrogen cycle and gaining an insight into climate dynamics of the past.

The advantages of the all-fiber optical current transformer include but are not limited to being small in size, having no magnetic saturation, exhibiting high measurement accuracy, and boasting strong electromagnetic interference resistance. However, the high cost of the all-fiber optical transformer limits its promotion and application in engineering. This paper proposes a design scheme of an independent double acquisition loop for the all-fiber optical current transformer based on the single optical path. Firstly, based on the closed-loop control mode and open-loop control mode, the two- channel sampling signal demand for relay protection, and the independent dual-acquisition loop design scheme of the all-fiber optical current transformer are proposed. Secondly, the reliability and economic feasibility of the scheme are demonstrated by an analysis of system failure and cost. The results show that the scheme can actualize the acquisition function of two independent all-fiber optical current transformer products on a single all-fiber current transformer in an integrated manner, which greatly reduces the cost of the all-fiber optical current transformer in engineering applications. BackgroundThe immunotherapy of Glioma has always been a research hotspot. Although tumor associated microglia/macrophages (TAMs) proves to be important in glioma progression and drug resistance, our knowledge about how TAMs influence glioma remains unclear. The relationship between glioma and TAMs still needs further study.MethodsWe collected the data of TAMs in glioma from NCBI Gene Expression Omnibus (GEO) that included 20 glioma samples and 15 control samples from four datasets. Six genes were screened from the Differential Expression Gene through Gene ontology (GO) analysis, Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis, protein–protein interaction (PPI) network and single-cell sequencing analysis. A risk score was then constructed based on the six genes and patients’ overall survival rates of 669 patients from The Cancer Genome Atlas (TCGA). The efficacy of the risk score in prognosis and prediction was verified in Chinese Glioma Genome Atlas (CGGA).ResultsSix genes, including CD163, FPR3, LPAR5, P2ry12, PLAUR, SIGLEC1, that participate in signal transduction and plasma membrane were selected. Half of them, like CD163, FPR3, SIGLEC1, were mainly expression in M2 macrophages. FPR3 and SIGLEC1 were high expression genes in glioma associated with grades and IDH status. The overall survival rates of the high risk score group was significantly lower than that of the low risk score group, especially in LGG.ConclusionJoint usage of the 6 candidate genes may be an effective method to diagnose and evaluate the prognosis of glioma, especially in Low-grade glioma (LGG).

Half-wavelength AC transmission (HWACT) is an ultra-long distance AC transmission technology, whose electrical distance is close to half-wavelength at the system power frequency. It is very important for the construction and operation of HWACT to analyze its fault features and corresponding protection technology. In this paper, the steady-state voltage and current characteristics of the bus bar and fault point and the steady-state overvoltage distribution along the line will be analyzed when a three-phase symmetrical short-circuit fault occurs on an HWACT line. On this basis, the three-phase fault characteristics for longer transmission lines are also studied. Organic modified kaolinite-urea intercalation complex (KUIC) was prepared using dimethyl sulfoxide (DMSO) as the precursor of kaolinite intercalation. Its structure was characterized by Fourier transform infrared (FTIR) and X-ray diffraction (XRD). Subsequently, as a synergistic agent, KUIC was combined with flame retardant ammonium polyphosphate (APP) to improve the flame retardant and smoke suppression performance of unsaturated polyester (UP) resin. A cone calorimeter (CONE) was used to study its flame retardancy and smoke suppression, and a scanning electron microscope (SEM) and thermogravimetry (TG) were used to study the micro morphology of the char and flame retardant mechanism. The results show that 12 phr of APP and 3 phr of KUIC were doped into UP to obtain a 28.0% limiting oxygen index (LOI) value. Compared with UP, the heat release rate and smoke production of UP/APP/KUIC composites were greatly decreased. Meanwhile, KUIC indeed enhanced the mechanical properties of UP.

The importance of autonomy learning is widely acknowledged in teaching English as a Foreign Language (EFL) in Asian contexts. The present study attempted to report low proficiency level students’ autonomous vocabulary learning. The first part of the present study focused on exploring how learners perceived and experienced their autonomous vocabulary learning, with a questionnaire and group discussion applied to collect data. During the second part of the study, the data were discussed collaboratively by six teachers to identify the different approaches utilized by learners. Priorities included setting weekly goals, self-planning and self-monitoring the process of learning vocabulary. Results also revealed that a weak ‘top-down’ approach in implementing learner autonomy is inadequate. Teachers need to provide scaffolding skills tailored to learners’ context-specific needs, for which a strong ‘bottom-up’ approach is more appropriate in supporting autonomous learning. In this case, classroom time should be allocated to demonstrate the strategies and teachers should work collaboratively to develop resources and materials for learners’ autonomous vocabulary learning. The present study also addressed the need for establishing an effective intervention for supporting learner autonomy. Keywords: learner autonomy, vocabulary learning, self-plan, self-monitor

Abstract The photovoltaic properties of solar cell based on the blends of poly[2-methoxy-5-(2-ethylhexoxy-1,4-phenylenevinylene) (MEH-PPV), fullerene (C60), and ZnCdTe-alloyed nanocrystals were investigated. Comparing the spectral response of photocurrent of the MEH-PPV:C60(+ZnCdTe) nanocomposite device with that of the devices based on MEH-PPV:C60and pristine MEH-PPV, one can find that the nanocomposite device exhibits an enhanced photocurrent. In comparing the composite devices with different ZnCdTe:[MEH-PPV + C60] weight ratios of 10 wt% (D1–1), 20 wt% (D1–2), 40 wt% (D1–3), and 70 wt% (D1–4), it was found that the device D1–3exhibits the best performance. The power conversion efficiency (η) is improved doubly compared with that of the MEH-PPV:C60device.

Abstract In this paper, we synthesized a novel type II cuprous sulfide (Cu2S)–indium sulfide (In2S3) heterostructure nanocrystals with matchstick-like morphology in pure dodecanethiol. The photovoltaic properties of the heterostructure nanocrystals were investigated based on the blends of the nanocrystals and poly(2-methoxy-5-(2′-ethylhexoxy)-p-phenylenevinylene) (MEH-PPV). In comparison with the photovoltaic properties of the blends of Cu2S or In2S3nanocrystals alone and MEH-PPV, the power conversion efficiency of the hybrid device based on blend of Cu2S–In2S3and MEH-PPV is enhanced by ~3–5 times. This improvement is consistent with the improved exciton dissociation or separation and better charge transport abilities in type II heterostructure nanocrystals.

Hepatocellular carcinoma (HCC) is a major public health problem due to increased incidence, late diagnosis and limited treatment options. TGF-β is known to provide cytostatic signals during early stages of liver damage and regeneration, but exerts tumor promoting effects in onset and progression of liver cancer. To understand the mechanistic background of such a switch, we systematically correlated loss of cytostatic TGF-β effects with strength and dynamics of its downstream signaling in 10 HCC cell lines. We demonstrate that TGF-β inhibits proliferation and induces apoptosis in cell lines with low endogenous levels of TGF-β and Smad7 and strong transcriptional Smad3 activity (PLC/PRF/5, HepG2, Hep3B, HuH7), previously characterized to express early TGF-β signatures correlated with better outcome in HCC patients. TGF-β dependent cytostasis is blunted in another group of cell lines (HLE, HLF, FLC-4) expressing high amounts of TGF-β and Smad7 and showing significantly reduced Smad3 signaling. Of those, HLE and HLF exhibit late TGF-β signatures, which is associated with bad prognosis in HCC patients. RNAi with Smad3 blunted cytostatic effects in PLC/PRF/5, Hep3B and HuH7. HCC-M and HCC-T represent a third group of cell lines lacking cytostatic TGF-β signaling despite strong and prolonged Smad3 phosphorylation and low Smad7 and TGF-β expression. Inhibitory linker phosphorylation, as in HCC-T, may disrupt C-terminally phosphorylated Smad3 function. In summary, we assort 10 HCC cell lines in at least two clusters with respect to TGF-β sensitivity. Cell lines responsive to the TGF-β cytostatic program, which recapitulate early stage of liver carcinogenesis exhibit transcriptional Smad3 activity. Those with disturbed TGF-β/Smad3 signaling are insensitive to TGF-β dependent cytostasis and might represent late stage of the disease. Regulation of this switch remains complex and cell line specific. These features may be relevant to discriminate stage dependent TGF-β functions for the design of efficient TGF-β directed therapy in liver cancer.

Abstract Background Hepatocellular carcinoma (HCC) still is a big burden for China. In recent years, the third-generation platinum compounds have been proposed as potential active agents for HCC. However, more experimental and clinical data are warranted to support the proposal. In the present study, the effect of lobaplatin was assessed in five HCC cell lines and the underlying molecular mechanisms in terms of cell cycle kinetics were explored. Methods Cytotoxicity of lobaplatin to human HCC cell lines was examined using MTT cell proliferation assay. Cell cycle distribution was determined by flow cytometry. Expression of cell cycle-regulated genes was examined at both the mRNA (RT-PCR) and protein (Western blot) levels. The phosphorylation status of cyclin-dependent kinases (CDKs) and retinoblastoma (Rb) protein was also examined using Western blot analysis. Results Lobaplatin inhibited proliferation of human HCC cells in a dose-dependent manner. For the most sensitive SMMC-7721 cells, lobaplatin arrested cell cycle progression in G1 and G2/M phases time-dependently which might be associated with the down-regulation of cyclin B, CDK1, CDC25C, phosphorylated CDK1 (pCDK1), pCDK4, Rb, E2F, and pRb, and the up-regulation of p53, p21, and p27. Conclusion Cytotoxicity of lobaplatin in human HCC cells might be due to its ability to arrest cell cycle progression which would contribute to the potential use of lobaplatin for the management of HCC.
