Abstract

Transient receptor potential vanilloid 4 (TRPV4) plays an important role in chondrocytes via Ca2+ signaling. However, its role in the progression of osteoarthritis is unclear. This study aimed to evaluate the effects of TRPV4 activation on articular cartilage and chondrocytes stimulated with interleukin (IL)-1β. Bovine and human articular chondrocytes were stimulated with various agents, including IL-1β, GSK1016790A (GSK101; a TRPV4 agonist), Compound C (an AMP-activated protein kinase (AMPK) inhibitor), and STO-609 (a calmodulin-dependent protein kinase kinase (CaMKK) inhibitor), and were processed for Western blot analysis and real-time PCR. The dimethylmethylene blue (DMMB) assay and Safranin O staining were also performed. GSK101 reversed the IL-1β-induced increase in expression of matrix metalloproteinase (MMP)-13 and decrease in expression of aggrecan. GSK101 also decreased proteoglycan release in the DMMB assay and retained Safranin O staining of articular cartilage tissue. Furthermore, GSK101 increased AMPK phosphorylation and decreased IL-1β-induced nuclear factor kappa B (NF-κB) phosphorylation. Compound C and STO-609 reversed the suppressive effects of GSK101 on NF-κB activation and MMP-13 expression. In conclusion, TRPV4 activation had chondroprotective effects on articular cartilage stimulated with IL-1β by activating CaMKK/AMPK and suppressing the NF-κB pathway. TRPV activators may offer a promising therapeutic option for preventing the progression of osteoarthritis.

Abstract

Herein we report the manipulation of perpendicular magnetic anisotropy (PMA), a very important technique for spintronics devices, which is achieved in an all-solid-state redox (reduction/oxidation) device. The device consisted of NiCo2O4 (NCO) thin film with a c -axis orientation and a proton-conducting polymer (Nafion). The PMA of NCO can be manipulated in situ with low voltage (V ≤ 0.7 V) due to the utilization of a proton-conducting solid electrolyte. The magnetic anisotropy variation was quantitatively evaluated by an anomalous Hall measurement. The magnetic anisotropy field of the NCO thin film was successfully manipulated in the range of 5%, although the rotation of the easy axis of magnetization was not observed. This modulation is attributed to the strain and electronic configuration variation by proton insertion. This manipulation method for PMA with the solid electrolyte enables a variety of reservoir states and contributes to lower power consumption and high-precision machine learning. Abstract

There is no assistive device for extramedullary surgery coordinated with 3D surgical assistive software for the total knee arthroplasty (TKA). We developed a novel extramedullary universal guide coordinated with 3D surgical assistive software and a novel extramedullary patient-specific assistive guide for the placement of femoral components by referring to an area not affected by cartilage or bone spurs, and filed a patent application. In this study, we visualize and reconstruct the total alignment of the lower extremity in TKA using these surgical devices, and validate their precision. A report releasing study results will be submitted in an appropriate journal.

Abstract

An ionic decision-maker (IDM), with the remarkable ability to solve multi-armed bandit problems (MBPs), has been developed as a new artificial intelligence technology, which have humanlike decision-making capabilities. Owing to their low durability, the Nafion polymer electrolytes and electrode materials normally found within IDMs do not meet the high-level integration and repetitive operation requirements of such devices. Therefore, in this study, mesoporous SiO2, which is a proton conducting inorganic oxide, has been utilized as the IDM electrolyte. The oxide-based IDMs developed showed good solvability for MBPs and excellent adaptability to dramatic environmental shifts, which were both at similar levels to those found in Nafion-based IDMs. While the adaptation exhibited by the oxide-based IDMs was slightly inferior to that found in Nafion-based IDMs, fluctuations in the correct selection rate (CSR) were significantly improved over those observed in Nafion-based IDMs. Further, the average of reached CSR was enhanced to 0.96 in comparison to 0.92 for Nafion-based IDMs. [ABSTRACT FROM AUTHOR]

Abstract

We have developed a nanostructuring method using the solid electrochemical reaction induced by a scanning tunneling microscope (STM). This method has some distinctive features that have not previously been obtained by conventional nanostructuring STM methods. The formation and disappearance of the nanostructure are reversible, and the rates can be controlled using STM. These features are realized via a local oxidation/reduction reaction of mobile metal ions in an ionic/electronic mixed conductor. In this study, a crystal of silver sulfide (Ag[sub 2]S), a mixed conductor, was used as the material for the STM tip. A nanoscale Ag cluster was formed at the apex of the Ag[sub 2]S tip when a negative bias voltage was applied to the sample. The Ag ions in the Ag[sub 2]S tip are reduced to Ag atoms by the tunneling electrons from the sample, and the Ag cluster is formed by the precipitation of the Ag atoms at the apex of the tip. The Ag cluster shrank gradually and disappeared when the polarity of the sample bias voltage was switched to positive. Ag atoms in the Ag cluster are oxidized to Ag ions, and the Ag ions redissolve into the Ag[sub 2]S tip.
