Abstract

Several trematodes including Opisthorchis viverrini utilize Bithynia siamensis goniomphalos as a snail intermediate host in their life cycles. In order to capture a comprehensive range of host–parasite interactions and their transmission dynamic patterns, B. s. goniomphalos were sampled monthly over 4 consecutive years in an irrigated paddy-field habitat in northeast Thailand. Using a standard cercarial shedding method, a high diversity of trematodes (17 types) was recovered. Virgulate xiphidiocercariae were the most prevalent (7.84%) followed by O. viverrini (0.71%). In addition to seasonal and environmental factors, the quantity of irrigation water for rice cultivation correlated with transmission dynamics of trematodes in B. s. goniomphalos. The peak prevalence of all trematode infections combined in the snails shifted from the cool-dry season in 2010–2012 to the hot-dry season in 2013 associated with an increasing quantity of water irrigation. A low frequency of mixed trematode infections was found, indicating that the emergence of virgulate cercariae, but not of O. viverrini, was negatively impacted by the presence of other trematodes in the same snail. Taken together, the observed results suggest that interactions between host and parasite, and hence transmission dynamics, depend on specific characteristics of the parasite and environmental factors including irrigated water for rice cultivation. Abstract

The human liver fluke Opisthorchis viverrini (Ov), the primary risk factor for cholangiocarcinoma (CHCA), is a parasite endemic to southeast Asian countries. With no effective treatments for CHCA currently available, early diagnosis and treatment of Ov infection remains the only practical method for the prevention of CHCA. In this study, plasma phosphoproteomes of patients in the non-Ov infection, non-cholangiocarcinoma subject group (non-OVCCA), the asymptomatic Ov infected group (OV), and the CHCA group (CCA), were investigated to identify potential biomarkers for Ov infection and CHCA. The AKT signalling pathway was found to be up-regulated. Phosphatidylinositol 4,5-bisphosphate 3-kinase catalytic subunit beta isoform (PIK3CB), an upstream signalling molecule, was selected as a potential biomarker and evaluated using indirect enzyme-linked immunosorbent assay (ELISA). Results demonstrated evidence that levels of PIK3CB in both the OV group and CCA group was statistically different compared to the non-OVCCA group (P < 0.01). However, the levels of PIK3CB between the OV group and the CCA group were found not to be statistically different. Sensitivity and specificity for OV using OD450 cut-off at >1.570 was 76 and 72%, respectively. For CCA, sensitivity and specificity using OD450 cut-off at >1.398 was 68 and 76%, respectively. Abstract

The Plumbago indica roots have been used as traditional medicines against many maladies; however, the information of toxicity has been needed when consumed in the high volume or for a long period. The objective of this study was to investigate the toxicity of crude extract from P. indica root (cPI) in hamsters by mortality, behavior changes, histochemical and histopathological changes. Golden Syrian hamsters were randomly divided into 5 groups and administered with cPI at 0, 100, 400, 1,000, and 3,000 mg/kg orally for 24 h and every day for 7 days for studying acute and subacute toxicity assessments, respectively. Neither mortality nor behavior changes of animals were found during acute and subacute toxicity tests. Weights of liver and kidney in cPI treated at 3,000 mg/kg were significantly increased after oral administration at 24 h and 7 days. The biochemical parameters revealed insignificant differences in the liver function tests (ALT and AST) and kidney function tests ( creatinine and BUN) . The morphological hepatic lobules in the subacute assessment were better than acute assessment. Although glomeruli swelling and angiectasis in the kidney were found in cPI treated group at 3,000 mg/kg for both the 24 h and 7 days, the renal tubular compartments had become similar to the control groups. The LD50 of cPI is classified as category 5 (2,000-5,000 mg/kg). These results suggest that the precaution of users should be concern harmful to hepatotoxic property and renal function in early acute and subacute periods at the high dose of cPI extract. Abstract

Objective: To investigate the maturity development of miracidia in uterine eggs from four portions of the Opisthorchis viverrini uterus and environmental factors possibly affected in maturation and infectivity of distal part uterine eggs. Methods: Uteri of adult worms were divided into 4 equal parts. Development of eggs was determined under light microscope. Only embryonated eggs were used to evaluate the effects of physicochemical factors: temperature, salinity, acidity, ultraviolet A, B, C. Infection success was evaluated by feeding treated eggs to intermediate host snails and determining by using a PCR approach. Results: Eggs obtained from the uterus closest to the ovary (regions 1 and 2) failed to develop in vitro. Eggs from region 4 of the uterus (close to the genital pore) were used to study effects of physicochemical factors. The highest survival and infection success was in groups of eggs kept at 30 °C (95.20%). The calculated period of loss infection success (LI50 and LI95) on miracidia in distal uterine eggs by exposure to UV-A, UV-B and UV-C were 73 and 1 523 d; 8 and 20 d; 1 and 2 d, respectively. Lethal concentrations (LC50 and LC95) of salinity, HCl and NaOH on miracidia in distal uterine eggs were 45.43 and 120.09 ppt, 0.01 and 0.25 M; 0.01 and 0.11 M, respectively, after 24 h exposure. Abstract

Four isoforms of calcium binding proteins containing 2 EF hand motifs and a dynein light chain-like domain in the human liver fluke Opisthorchis viverrini, namely OvCaBP1, 2, 3, and 4, were characterized. They had molecular weights of 22.7, 21.6, 23.7, and 22.5 kDa, respectively and showed 37.2-42.1% sequence identity to CaBP22.8 of O. viverrini. All were detected in 2- and 4-week-old immature and mature parasites. Additionally, OvCaBP4 was found in newly excysted juveniles. Polyclonal antibodies against each isoform were generated to detect the native proteins in parasite extracts by Western blot analysis. All OvCaBPs were detected in soluble and insoluble crude worm extracts and in the excretory-secretory product, at approximate sizes of 21-23 kDa. The ion-binding properties of the proteins were analyzed by mobility shift assays with the divalent cations Ca, Mg, Zn, and Cu. All OvCaBPs showed mobility shifts with Ca and Zn. OvCaBP1 showed also positive results with Mg and Cu. As tegumental proteins, OvCaBP1, 2, and 3 are interesting drug targets for the treatment of opisthorchiasis. [ABSTRACT FROM AUTHOR]

Abstract

To determine the current reservoir status and prevalence of Opisthorchis viverrini infection in endemic areas, 78 dogs and 22 cats were sampled for fecal examination in 3 villages along the Chi River in Khon Kaen Province, northeastern Thailand. Sex, age, color, identifiable markings, subjective body condition scores (BCS) and diet were recorded and analyzed. Fecal samples were evaluated using direct smear and modified formalin-ethyl acetate fecal assays. All parasites found were identified. In dogs and cats, the prevalences of O. viverrini infection were 3.8% and 36.4%, respectively. The highest prevalences of parasitic infection in both dogs and cats were Ancylostoma spp, in 64.1% and 77.3%, respectively. These results provide insight into the role of the canines and felines in maintaining the presence of O. viverrini eggs in the environment. If similar patterns occur on a broader scale, the zoonotic role of dogs and cats in the epidemiology of this disease should be considered in the development of improved control and education programs.
