Abstract

The spread of e-commerce has transformed consumer habits and distribution. There is a need to improve the location of pickup points, starting from small inland areas to areas with particular travel problems, such as the one chosen, i.e. Enna, in the centre of Sicily. The following research focuses on a two-level analysis of the accessibility perceived by users and commercial vehicles. A series of indicators were defined to evaluate the supply in terms of accessibility, the construction of isochrones, and the selection of the walking mode of travel (by demand-user). Comparing the results obtained with the values of the resident population in the different areas adjacent to the sampling points, it emerged that, at present, the 12 presented sampling points do not have an optimised spatial distribution. The overall coverage is about 60% of the population, while all sampling points are generally easily accessible. The results form the basis for more in-depth analyses to improve urban logistics and last-mile services. [ABSTRACT FROM AUTHOR]

7. Perceived Pedestrian Level of Service : The case of Thessaloniki, Greece

Abstract

Level of Service (LOS) is one of the most crucial components for the assessment of pedestrian facilities by mainly concerning the effective width as well as pedestrians’ flows. However, current research reveals that qualitative characteristics can also contribute to LOS estimation as perceived by pedestrians. Specifically, socio-demographic characteristics (i.e., age, gender) as well as characteristics related with perceived comfort and safety can be related with perceived LOS. A Revealed Preference (RP) face to face questionnaire-based survey (including 301 interviewees) was realized during October 2019 at a central pedestrian facility in the city of Thessaloniki, Greece. RP questionnaire survey assisted in gaining valuable knowledge concerning the factors that mainly affect pedestrians’ perceived LOS across the pedestrian facility. The examined pedestrian facility is one of the most important in Thessaloniki since it facilitates high pedestrian flows within the city center daily. The present survey considered pedestrians’ general mobility characteristics such as walking frequency along the facility and trip purpose. Additionally, the evaluation of the greater facility’ area in terms of land use attractiveness, comfort, personal and road safety, public transport, parking conditions and traffic delays, accessibility, pedestrians and bicycles were concerned as well. Ordinal regression was the main tool for the development of the ordinal regression model, and therefore, for the conclusions’ drawing of the present research. Abstract

Recently, the field of earthquake engineering experienced substantial enhancements due to numerous results of experimental and theoretical researches. Modern technical codes recognized the last advances in this field and provided new solutions for the structural assessment and design of buildings. However, the need for large scale testing is still taking several open questions, especially for the design of civil infrastructures, and experimental activity on full scale specimens has become necessary. This paper presents the main testing facilities of a new research centre, namely the Laboratory of Earthquake engineering and Dynamic Analysis (LEDA) of the University of Enna “Kore”. In particular, the design procedures of facilities for dynamic and pseudo-dynamic testing, built in order to perform tests on specimens of large dimensions, are described and discussed. The LEDA centre, includes several laboratories operating in the fields of structural engineering and dynamics. The most important features of LEDA are the broad band of tests that can be performed and the presence of a high capacity strong floor - reaction wall system and of two six-degrees-of-freedom shaking tables. The capabilities of the laboratory are highlighted, stressing as the complementarity of both traditional techniques (pseudo-dynamic and dynamic tests) can be fully studied and advanced hybrid testing method can be investigated.

16. The synergistic effect exerted by the HDAC inhibitor SAHA and the sesquiterpene lactone parthenolide on triple negative breast canc er cells

Abstract

Triple-negative breast cancer (TNBC) is a subtype o f breast cancer, insensitive to endocrine therapy. Chemotherapy is the main form of treatment, but is accompanied by a high rate of recidivism. The sesquiterpene lactone Parthenolide (PN) exerts a cy totoxic effect on MDA-MB231 cells, a TNBC cell line (1), but was ineffective at low doses (2-5μM). This repr esents an obstacle for a therapeutic utilization of PN. We supposed, in line with other authors (2), that PN c auses a protective response, which at low doses pre vails on the cytotoxic effect. With the aim of inhibiting this protective effect we have shown that pre-trea tment of MDA-MB231 cells with SAHA (2-5μM), an histone deace tylates inhibitor, synergistically sensitizes the c ells to the cytotoxic effect of PN, also at low doses of th is compound. SAHA/PN combination induced hyperacetylation of his tones H3 and H4 and hypomethylation of DNA. These changes cause epigenetic effects, which can be resp onsible for the increased expression of tumour suppressors p21 and p27 and decreased levels of Bcl 2 and p65, a component of NFkB. Moreover SAHA alone induced ROS generation as well as autophagy, which favours cell survival, and apoptosis. The addition of PN (8 μ M) to SAHA reduced production of ROS and autophagy, while increased the apoptotic process. Interestingly PN activates Akt, mTOR, phospho-p70S6 kinase and ULK1/2, a factor that inhibits autophagy . In addition PN caused nuclear accumulation of Nrf2 wit h stimulates antioxidant genes. SAHA prevented thes e effects. In conclusion SAHA/PN stimulated cytotoxicity throu gh many mechanisms: (i) induces epigenetic events w ith changes in gene expression, (ii) PN prevents SAHA e ffect on autophagy and (iii) SAHA suppresses the protective response exerted by PN through inactivat ion of m-TOR. Taken together our results suggest th at combination SAHA/PN can be a candidate for TNBC the rapy.

Abstract

C2-ceramide, a cell-permeable analogue of ceramide, induced significant, dose- and time-dependent death in human retinoblastoma Y79 cells. Dying cells strongly displayed the morphology of apoptosis as characterized by microscopic evidence of cell shrinkage, membrane blebbing, nuclear and chromatin condensation and degeneration of the nucleus into membrane-bound apoptotic bodies. Upon induction of apoptosis Y79 cells evidence early phosphatidylserine externalization, as shown by annexin V-FITC. Apoptosis was also assessed by monitoring changes in cell granularity by staining with the combined fluorescent dyes acridine orange and ethidium bromide. C2-ceramide induced these morphological changes without a concomitant production of oligonucleosomal fragments responsible for the DNA ladder and without changes in p53 protein level. Apoptosis was accompanied by accumulation of a modified Bcl-2 protein with a slower-mobility form, and by proteolytic cleavage of PARP. The effect seemed to be specific for C2-ceramide, as C2-dihydroceramide, or other amphiphilic lipid analogues, or products of ceramide hydrolysis were ineffective. The effect also depended on mRNA and protein synthesis as it was markedly inhibited by actinomycin D and cycloheximide. Sphingomyelinase and interleukin-1beta, which are known to activate the sphingomyelin turnover leading to ceramide generation, also induced apoptosis mimicking the effects of ceramide. These findings propose ceramide as an activator of the suicidal program in Y79 cells.

Abstract

PURPOSE:To examine the apoptotic effect induced in human retinoblastoma Y79 cells by camptothecin, etoposide, and amsacrine, to examine the effect of these drugs on the expression of many apoptosis-related modulators, and to test the antiapoptotic effect exerted by insulin-like growth factor-I (IGF-I). METHODS:Morphologic features of apoptosis were demonstrated using acridine orange- ethidium bromide staining and electron microscopy. DNA fragmentation was determined by means of an in situ cell detection procedure (TdT-dUTP terminal nick-end labeling [TUNEL]) or by electrophoresis on agarose gels and was quantified by enzyme-linked immunosorbent assay. The expression of apoptosis-related modulators was studied by western blot analysis. The processing of latent p53 was examined by means of pulse- chase analysis. RESULTS:Camptothecin, etoposide, and amsacrine induced apoptosis in Y79 cells in a dose-dependent manner; camptothecin was the most efficacious compound. The effect, which was dependent on macromolecular synthesis, appeared after a lag of 8 hours and increased for as long as 24 hours. It was lower in cells treated with IGF-I, a potent mitogenic factor. Camptothecin and etoposide increased the p53 level after 4 hours of treatment, before the onset of apoptosis. This effect seemed to be a consequence of the conversion of latent p53 to one that is transcriptionally active. The drugs also induced an increase in p53-related proteins, such as p21, Bax, and IGF binding protein-3 (IGF-BP3), and caused a significant reduction of the Bcl-2 level. The latter effect was less evident in cells pretreated with IGF-I. CONCLUSIONS:Topoisomerase inhibitors induce apoptosis in Y79 cells. This event is accompanied by a decrease in the expression of Bcl-2, a death antagonist, and an increase in that of Bax, a death agonist. A probable consequence of these modifications is the activation of ICE-like activity with degradation of poly-(adenosine diphosphate [ADP] ribose)-polymerase. Insulin-like growth factor-I exerts an antiapoptotic action in Y79 cells, and this function is most likely reduced by the overexpression of IGF-BP3 that is induced by drug treatment.

Abstract

Finding new treatments targeting cancer stem cells (CSCs) within a tumor seems to be critical to halt cancer and improve patient survival. Osteosarcoma is an aggressive tumor affecting adolescents, for which there is no second-line chemotherapy. Uncovering new molecular mechanisms underlying the development of osteosarcoma and origin of CSCs is crucial to identify new possible therapeutic strategies. Here, we aimed to characterize genetically and molecularly the human osteosarcoma 3AB-OS CSC line, previously selected from MG63 cells and which proved to have both in vitro and in vivo features of CSCs. Classic cytogenetic studies demonstrated that 3AB-OS cells have hypertriploid karyotype with 71-82 chromosomes. By comparing 3AB-OS CSCs to the parental cells, array CGH, Affymetrix microarray, and TaqMan® Human MicroRNA array analyses identified 49 copy number variations (CNV), 3,512 dysregulated genes and 189 differentially expressed miRNAs. Some of the chromosomal abnormalities and mRNA/miRNA expression profiles appeared to be congruent with those reported in human osteosarcomas. Bioinformatic analyses selected 196 genes and 46 anticorrelated miRNAs involved in carcinogenesis and stemness. For the first time, a predictive network is also described for two miRNA family (let-7/98 and miR-29a,b,c) and their anticorrelated mRNAs (MSTN, CCND2, Lin28B, MEST, HMGA2, and GHR), which may represent new biomarkers for osteosarcoma and may pave the way for the identification of new potential therapeutic targets
