Objective: The aim of study was to characterize Giardia isolates genetically among patients in Chandigarh region, India. For this, nested PCR targeting fragment of the glutamate dehydrogenase (GLUD1 earlier named as GDH) gene was used. Phylogenetic analysis was done by constructing neighbor-joining tree made out of the nucleotide sequences of G. intestinalis isolates obtained in this study and with the known sequences published in GenBank. Results: Out of 40 samples, GLUD1 gene was amplified in 33 samples (82.5%). The product of GLUD1 gene was successfully sequenced only in 32 samples. In these samples, assemblage B was found in 27 (84.37%) samples whereas 5 (15.6%) samples had assemblage A. Among assemblage B most of them were of BIII. Therefore, genotyping of Giardia would be helpful in conducting epidemiological studies. [ABSTRACT FROM AUTHOR]

Background & objectives: Present treatment strategies for kala-azar (visceral leishmaniasis, VL) include use of first line drug sodium antimony gluconate (SAG) to all patients but a large number of patients do not get relief with this drug. If a patient does not respond to a full course of SAG, a second or third line drug is given. We undertook this study to test whether an improved outcome can be achieved by employing a strategy of treatment based on culture and sensitivity of amastigotes to SAG compared with conventional empirical treatment. Methods: In a double-blind, randomized, controlled trial done in Balaji Utthan Sansthan, Patna, of the 181 patients screened, 140 were finally randomly allocated to two groups A and B; group A patients were treated with SAG if their amastigotes were sensitive to SAG, and all patients in group B were treated with SAG to start with. Primary outcome measured was as no relapse within 6 months of follow up after cure and other outcomes measured were period of stay of patients in hospital, expenditure involved in the treatment, and infectivity periods of two groups, two-third of treatment period and whole of untreated period were taken as infectivity period. SAG was used at a dosage of 20 mg/kg given deep intramuscular injections in buttock for 28 days, amphotericin B (AMB) given at a dose of 1 mg/kg body wt daily for 20 days as a slow intravenous infusion in 5 per cent dextrose. Results: Of the 70 patients in group A, 29 patients whose amastigotes were sensitive to SAG were treated with SAG, 2 patients were withdrawn due to drug toxicity; and 2 relapsed within 6 months of follow up and ultimate cure occurred in 25 (86.2%) patients only. Of the 70 patients in group B treated with SAG, 5 (7.1%) patients withdrew due to drug toxicity, 35 patients (50%) did not respond to treatment, 5 (7.1%) relapsed during 6 months of follow up and thus only 25 patients (35.7%) were ultimately cured. The difference between the two groups was significant (P<0.001). No patient died during treatment due to any toxicity because of early withdrawl of patients from treatment apprehending toxicity. Patients whose amastigotes were resistant to SAG, withdrawn from the study due to SAG toxicity, relapsed after cure with SAG, and who did not respond to SAG in both the groups were treated with AMB and all were cured. Groups B and A patients spent 3065 and 2340 days respectively in hospital group B 1.3 times more than group A. The likely period of spread of parasites in society was 1965 days in group B and 1644 days in group A, group B 1.4 times more than group A. The total expenditure on treatment in groups B and A was $ 65,575 and $ 50,590 respectively; group B patient had to spend 1.3 times more than group A. Interpretation & conclusions: A new strategy for treatment of kala-azar based on culture and sensitivity of amastigotes improved the cure rate, saved expenditure on the patient's treatment, patients had to stay for shorter periods in hospital and reduced the chance of spread of SAG resistant disease in society. Till the government opts for better drugs, the treatment based on culture and sensitivity of the parasites to SAG may be a better method. [ABSTRACT FROM AUTHOR]

• Bark of populus ciliata of the salicaceae family is traditionally used for blood purification, treatment of rheumatism, fatigue, and menstrual cramps pain. • HPTLC analysis revealed the higher concentrations of gallic acid, salicin, and quercetin in the ethyl acetate fraction of P. ciliata bark. • In-vitro studies revealed the strong antioxidant, antimicrobial, anti-inflammatory, and anticancer properties of the ethyl acetate fraction of P. ciliata bark. • The findings of the current study validate the ethnomedicinal use of bark of P. ciliata. Populus ciliata Wall. ex Royle (Salicaceae family) is a fast-growing, multipurpose native tree of the Himalayas. Traditionally, the tree's bark is used for blood purification, treatment of rheumatism, and fatigue, and to relieve the pain of menstrual cramps. The present study deals with screening crude methanolic extracts and fractions (n-hexane, chloroform, ethyl acetate, and n-butanol) of P. ciliata bark and leaves for total polyphenolic contents, antioxidant, antimicrobial, anti-inflammatory, and cytotoxic activities. Results showed that among all tested samples, the bark ethyl acetate fraction had the highest polyphenolic contents and strong antioxidant potential (DPPH 25.75±3.60 µg/mL; ABTS 33.55±2.96 µg/mL; FRAP 70.39±1.75 µM of Fe equivalents). The HPTLC analysis confirmed the presence of gallic acid, salicin, and quercetin molecules in the extracts and different fractions of P. ciliata whereas ethyl acetate fractions showed their maximum quantity. The ethyl acetate fraction of bark also revealed higher antibacterial activity against Pseudomonas aeruginosa (MIC 0.0156 mg/mL), Klebsiella pneumoniae and Staphylococcus aureus (MIC 0.0312 mg/mL), Bacillus subtilis and Escherichia coli (MIC 0.0625 mg/mL), and antifungal activity against Fusarium oxysporum (MIC 0.125 mg/mL) and Rosellinia necatrix (0.250 mg/mL), whereas butanol fraction of bark showed maximum antifungal activity against Candida auris (MIC 1.25 mg/mL). The highest anti-inflammatory activity was observed in bark ethyl acetate fraction (IC 50 0.113±0.017 mg/mL for egg albumin and 0.160±0.014 mg/mL for bovine serum albumin assay) in comparison with other extracts and fractions. Additionally, bark crude extract and its ethyl acetate fraction were observed more effective against colon cancer cell lines (IC 50 values <0.170 mg/mL for HCT-116 and <0.340 mg/mL for SW-620). The study concludes that the bark ethyl acetate fraction is the most biologically active fraction due to a higher number of polyphenolic contents (salicin, gallic acid, and quercetin) and among both plant parts, bark has a higher medicinal value than leaves. Rubus ellipticus fruits aqueous extract derived ZnO-nanoparticles (NPs) were synthesized through a green synthesis method. The structural, optical, and morphological properties of ZnO-NPs were investigated using XRD, FTIR, UV-vis spectrophotometer, XPS, FESEM, and TEM. The Rietveld refinement confirmed the phase purity of ZnO-NPs with hexagonal wurtzite crystalline structure and p-63-mc space group with an average crystallite size of 20 nm. XPS revealed the presence of an oxygen chemisorbed species on the surface of ZnO-NPs. In addition, the nanoparticles exhibited significant in vitro antioxidant activity due to the attachment of the hydroxyl group of the phenols on the surface of the nanoparticles. Among all microbial strains, nanoparticles' maximum antibacterial and antifungal activity in terms of MIC was observed against Bacillus subtilis (31.2 µg/mL) and Rosellinia necatrix (15.62 µg/mL), respectively. The anticancer activity revealed 52.41% of A549 cells death (IC50: 158.1 ± 1.14 µg/mL) at 200 μg/mL concentration of nanoparticles, whereas photocatalytic activity showed about 17.5% degradation of the methylene blue within 60 min, with a final dye degradation efficiency of 72.7%. All these results suggest the medicinal potential of the synthesized ZnO-NPs and therefore can be recommended for use in wastewater treatment and medicinal purposes by pharmacological industries. The present report aims to synthesize the zinc oxide nanoparticles (ZnO NPs) employing Myrica esculenta fruits aqueous extract as a reducing and/or capping agent. Different techniques viz., X-ray powder diffraction (XRD), X-ray photoelectron spectroscopy (XPS), field emission scanning electron microscope (FESEM), and ultraviolet–visible spectroscopy (UV–vis), were used to characterize the formation of ZnO NPs. The results revealed the formation of thin pellets-like ZnO NPs with an average crystallite size of 31.67 nm. Further, the aqueous extract of fruits and fabricated ZnO NPs were investigated for their antimicrobial [disc diffusion assay, poisoned food technique, and Minimum Inhibitory Concentration (MIC) assay], antioxidant [2,2-diphenyl-1-picrylhydrazyl (DPPH), and Ferric ion reducing antioxidant power (FRAP) assays], and photocatalytic activities. The study showed that ZnO NPs have higher antimicrobial activity (MIC- Bacillus subtilis : 0.031 mg/mL, Staphylococcus aureus : 0.062 mg/mL, Fusarium oxysporum : 0.250 mg/mL, and 0.125 mg/mL for Pseudomonas aeruginosa, Escherichia coli , Rosellinia necatrix) as compared to pure fruits' aqueous extract. Additionally, they also showed strong antioxidant activity (IC 50 -DPPH: 182.63 ± 3.21 µg/mL; FRAP: 129.44 µM Ferric sulphate (FeSO 4) equivalents). The XPS analysis revealed that a high percentage of surface chemisorbed species could be accountable for the excellent biological activities of ZnO NPs. Furthermore, photocatalytic activity showed 91% photodegradation of methylene blue dye in 180 min. Carissa, a genus of the Apocynaceae family, consists of evergreen species, such as shrubs as well as small trees that are native to Asia, Africa, and Oceania's subtropical and tropical regions. Most of the Carissa species are traditionally used to treat various diseases, such as chest pain, headaches, gonorrhoea, rheumatism, syphilis, oedema, rabies, stomach pain, hepatitis, cardiac diseases, and asthma. The pharmacological studies on Carissa species revealed its antioxidant, antimicrobial, anticancer, cardioprotective, antipyretic, analgesic, wound healing, anticonvulsant, antiarthritic, adaptogenic, anti-inflammatory, and antidiabetic activities, thus validating its use in indigenous medicine systems. The review article summarised the comprehensive literature available, including morphology, indigenous uses, bioactive composition, nutraceutical, and pharmacological activities of Carissa species. A total of 155 research papers were cited in this review article. The Carissa fruits are rich in dietary fibre, lipids, proteins, carbohydrates, vitamin C, and macro- and micro-elements. A total of 121 compounds (35 polyphenols (flavonoids and phenolic acids), 30 lignans, 41 terpenoids, 7 steroids, 2 coumarins, and 6 cardiac glycosides) have been extracted from C. spinarum, C. carandas, and C. macrocarpa. Among all chemical constituents, lupeol, carissol, naringin, carisssone, scopoletin, carissaeduloside A, D, J, carandinol, sarhamnoloside, carissanol, olivil, carinol, 3β-hydroxyolean-11-en-28,13β-oilde, ursolic acid, and carissone are the key bioactive constituents responsible for pharmacological activities of genus Carissa. The gathered ethnopharmacological information in the review will help to understand the therapeutic relevance of Carissa as well as paving a way for further exploration in the discovery of novel plant-based drugs. Background & objectives: Of the two reservoirs of infection of kala-azar Le., patients of kala-azar and post kala-azar dermal leishmaniasis (PKDL), PKDL provides easy access for the sandfly to pick up the parasites. In the last epidemic of 1977 in India, the importance of PKDL as a potential cause of increase in number of kala-azar cases was ignored. During recent years, we found an increase in the cases of kalaazar whereas cases of PKDL were decreasing in Bihar. We undertook this study to find out reasons for this phenomenon. Methods: These three different settings were selected to study the trends of the disease. (i) Cases of PKDL registered in the Dermatology Department of Patna Medical College Hospital (PMCH), one of the largest and oldest teaching hospital in Bihar, between 1970 and 2005; (ii) Rajendra Memorial Research Institute of Medical Sciences, Patna (RMRIMS), a research institute exclusively devoted to kala-azar (2000 and 2005); and (iii) interviews with two leading dermatologists of Patna selected by lottery on the incidence of PKDL and possible causes of its decrease, if any. The number of eases of kala-azar (visceral leishmaniasis, VL) from Bihar was studied from Malaria Departments of the Government of Bihar and Government of India, the two nodal departments dealing with the kala-azar. Results: Analysis of data from Dermatology Department of PMCH showed increase in number of cases of PKDL from two in 1970 to 12 in 1976, a year before the first epidemic of kala-azar in 1977 with 100,000 cases. Kala-azar cases decreased to 11,120 in 1982 due to control measures taken between 1977- 1979 but cases of PKDL reached 28 and kept on increasing. During 1950 to 1977, low dose and short duration regimen of sodium antimony gluconate (SAG) was mainly used in the treatment of kala-azar. Between 1977-1991 increasing incidence of unresponsiveness to SAG, led to the usage of longer duration and higher dose regimen of SAG, more use of amphotericin B (AMB) for SAG resistant cases and also as a first line drug for kala-azar and PKDL. The number of kala-azar cases started decreasing after control measures taken during 1992-1994 but cases of PKDL continued decreasing. The effect of control measures on the incidence of kala-azar was visible upto 2002, but decrease in number of PKDL cases continued. In 2005 the number of PKDL cases was 14 but number of kala-azar cases reached 21,177 in Bihar. In the interview, the two dermatologists also opined that PKDL was decreasing due to increased use of amphotericin B in the treatment of kala-azar. Trend analysis done on the data of PMCH and RMRIMS showed that PKDL will decrease in coming years and kala-azar will increase. Interpretation & conclusion: Incidence of PKDL decreased in PMCH and RMRIMS and also suggested by two dermatologists that extensive use of amphotericin B in the treatment of kala-azar might be responsible for decrease in number of cases of PKDL. The use of metallic nanoparticles in engineering and biomedicine disciplines has gained considerable attention. Scientists are exploring new synthesis protocols of these substances considering their small size and lucrative antimicrobial potential. Among the most economical techniques of synthesis of metallic nanoparticles via chemical routes, which includes the use of chemicals as metal reducing agents, is considered to generate nanoparticles possessing toxicity and biological risk. This limitation of chemically synthesized nanoparticles has engendered the exploration for the ecofriendly synthesis process. Biological or green synthesis approaches have emerged as an effective solution to address the limitations of conventionally synthesized nanoparticles. Nanoparticles synthesized via biological entities obtained from plant extracts exhibit superior effect in comparison to chemical methods. Recently, conifer extracts have been found to be effective in synthesizing metallic nanoparticles through a highly regulated process. The current review highlights the importance of conifers and its extracts in synthesis of metallic nanoparticles.
