Purpose: To establish a highly sensitive and specific approach for the detection of circulating PIK3CA H1047R mutation in breast cancer (BC) patients and to investigate the association between the prevalence of PIK3CA H1047R mutation and clinical presentations., Methods: A proper blocker was designed in an allele-specific manner and optimized for PCR-based identification of the PIK3CA H1047R mutation. The established technique was validated in cell-free DNA samples from 196 recruited BC patients., Results: The allele-specific PCR assay with a properly designed blocker was able to detect the H1047R mutant variant with 0.01%. By applying the newly established assay, 62 cases (31.6% of the total recruited cases) were found to carry a blood-circulating H1047R mutant. Wherein, the detected mutant rates increased with disease stages from 2/18 (11.1%) of stage I to 17/71 (23.9%) of stage II, 20/53 (37.7%) of stage III, and 23/31 (42.6%) of stage IV (p = 0.025), respectively. Higher frequencies of H1047R mutation were associated with late-stage (p = 0.033) or recurrence (p = 0.045) or metastatic patients (p = 0.049) as well as radiation-treated human epidermal growth factor receptor 2 (HER2) positive BC (p = 0.004). PIK3CA mutant carriers were frequently observed in patients under the age of 50 who had liver-metastasized or brain metastases or lymph node-invaded (p < 0.05)., Conclusion: A novel allele-specific PCR assay with high sensitivity was established successfully for the detection of the PIK3CA H1047R mutation in clinical practice., Competing Interests: The authors have declared that no competing interests exist., (Copyright: © 2024 Thao et al. Overuse of seed and chemical inputs is a major constraint for sustainable rice production in Vietnam. In this study, two seasons of field trials were conducted to compare different crop establishment practices for rice production in the Mekong River Delta using environmental and economic sustainability performance indicators. The indicators including energy efficiency, agronomic use efficiency, net income, and greenhouse gas emissions (GHGEs) were quantified based on four treatments including manual broadcast-seeding, blower seeding, drum seeding, and mechanized transplanting. Across the four treatments, yields ranged from 7.3-7.5 Mg ha -1 and 6.2-6.8 Mg ha -1 in the Winter-Spring (WS) and Summer-Autumn (SA) seasons, respectively. In comparison with direct seeding methods, mechanized transplanting decreased the seed rate by 40%. It also led to a 30-40% reduction in pesticide use during the main crop season (WS). Mechanized transplanting required higher inputs, including machine depreciation and fuel consumption, but its net energy balance, net income and GHGE were at a similar level as the other non-mechanized planting practices. Objectives: The aim of this study was to evaluate the difference between anti-cariogenic biofilm activities of glass ionomers (G-Is) during the initial and second fluoride release phases and to define relationships between the anti-biofilm activities and fluoride release., Methods: Fluoride release of three commercially available G-Is in a buffer was evaluated for 770 h, and then 70-h-old Streptococcus mutans UA159 biofilms were formed on the G-Is that had been immersed in the buffer for 0, 100, 200, or 700 h. The dry weight, bacterial cell number, water-insoluble extracellular polysaccharides (EPSs), and accumulated fluoride concentration of the 70-h-old biofilms and fluoride release and acid production rates during biofilm formation were determined. Relationships between the experimental variables and fluoride release rate were also evaluated using linear regression analysis., Results: In this study, fluoride release of the tested G-Is did not exhibit a biphasic pattern during biofilm formation. The release was sustained or did not rapidly decrease even over long immersion periods and was strongly correlated with an increase in accumulated fluoride concentration of the biofilms (R=0.99, R(2)=0.98) and reductions in dry weight, water-insoluble EPSs, and acid production rate of the biofilms (R=-0.99 to -0.96, R(2)=0.92-0.98)., Conclusions: This study suggests that G-Is can effectively affect acid production, EPS formation, and accumulation of cariogenic biofilms even during the second fluoride release phase, and that the anti-cariogenic biofilm activity is strongly correlated with fluoride release, which may be enhanced by acid production of cariogenic biofilms., Clinical Significance: G-Is can affect cariogenic biofilm formation even during the second fluoride release phase., (Copyright © 2016 Elsevier Ltd. The aim of this study was to analyze all new pulmonary tuberculosis cases and to assess the frequency and consequences of associated diabetes mellitus. Although apparently distinct, these two diseases could be linked, as suspected since decades. The context is the persistent endemicity of tuberculosis and the recent emergence of diabetes, two major health events for developing countries. The study was conducted at National TB hospital in Hanoï, Vietnam, and retrospectively included tuberculosis patients, cases with diabetes and controls without diabetes, collected during a three-year period 2006-2008. Cases associated with HIV infection or other comorbidities potentially affecting mortality were excluded. Among 2867 new pulmonary TB cases, 254 (8.8%) had associated diabetes. Eighty-six cases and 86 matched controls were included. Among cases mean blood glucose at admission was 14.4 mmol/dl, and patients were mostly urban (50% cases vs 32.5% controls, p=0.03). Clinical symptoms were similar in both groups. However, chest X-Ray cavitary images were significantly more frequent among cases (38.3% vs 16.3%, p=0.002, OR=4.1), as did infiltrative images (78% vs 62%, p=0.017, OR=2.55) and positive sputum smears (67.4% vs 47.8%, p=0.014, OR=2.21). In Vietnam, DM is found in quasi 9% of patients with a new diagnosis of primary pulmonary TB. The diabetes-TB patients may have more severe radiologic findings and a higher smear positivity rate. We suggest that in addition to the systematic HIV screening, all new TB patients should also be screened for diabetes.

Objectives: The aim of this study was to evaluate acidogenicity and composition of Streptococcus mutans biofilms on glass ionomer cements (GICs) and then to determine the relationship between the anti-S. mutans biofilm activity and fluoride release rate of the GICs., Methods: S. mutans biofilms were formed on discs prepared using five commercial GICs. Acid production and fluoride release rates of the biofilms on the GIC discs during biofilm formation (0-94 h) were determined. Next, 94-h-old S. mutans biofilms on GIC discs were analyzed to evaluate the biofilm composition (dry weight, bacterial cell number, and extra-cellular polysaccharide (EPS) amount) using microbiological, biochemical, and confocal laser scanning microscopy (CLSM) methods. Lastly, relationships between the fluoride release rate and changes in acidogenicity and composition of the biofilms were determined using a linear-fitting procedure., Results: All of the tested GICs released fluoride ions. Of the GICs, the two that showed the highest fluoride release rates strongly affected acidogenicity, dry weight, and EPS formation of the biofilms. Furthermore, they reduced the bacterial and EPS bio-volumes and EPS thickness. However, the number of colony forming units (CFUs) of the biofilms was higher than that of the control. Generally, changes in the acidogenicity and composition (except for CFU count) of the biofilms on the GICs followed a negative linear-pattern of fluoride release rate-dependence (R=-0.850 to -0.995, R(2)=0.723-0.990)., Significance: These results suggest that the anti-cariogenic biofilm activity of GICs is closely correlated with their fluoride release rate during biofilm formation., (Copyright © 2014 Academy of Dental Materials. Objectives: The aim of this study was to evaluate Streptococcus mutans adhesion to fluoride varnishes and subsequent change in biofilm accumulation and acidogenicity., Methods: After producing fluoride varnish-coated hydroxyapatite discs using Fluor Protector (FP), Bifluorid 12 (BIF), Cavity Shield (CASH), or Flor-Opal Varnish White (FO), S. mutans biofilms were formed on the discs. To assess S. mutans adhesion to the discs, 4-h-old biofilms were analysed. To investigate the change in biofilm accumulation during subsequent biofilm formation, the biomass, colony forming units (CFU), and water-insoluble extracellular polysaccharides (EP) of 46-, 70-, and 94-h-old biofilms were analysed. To investigate the change in acidogenicity, pH values of the culture medium were determined during the experimental period. The amount of fluoride in the culture medium was also determined during the experimental period., Results: BIF, CASH, and FO affected S. mutans adhesion (67-98% reduction) and subsequent biofilm accumulation in 46-, 70-, and 94-h-old biofilms. However, the reducing effect of the fluoride varnishes on the biomass, CFU count, water-insoluble EP amount, and acid production rate of the biofilms decreased as the biofilm age increased. These results may be related to the fluoride-release pattern of the fluoride varnishes. Of the fluoride varnishes tested, FO showed the highest reducing effect against the bacterial adhesion and subsequent biofilm accumulation., Conclusions: Our findings suggest that if the results of these experiments are extrapolable to the in vivo situation, then reduced clinical benefit of using fluoride varnishes may occur with time., Clinical Significance: Fluoride varnish application can affect cariogenic biofilm formation but the anti-biofilm activity may be reduced with time., (Copyright © 2014 Elsevier Ltd. In Vietnam, most of municipal solid waste (MSW) is disposed of at open dumping and landfill sites, and the methane gas from waste is the un-ignorable source of greenhouse gas (GHG) emission. It is indispensable to explore the possibility for GHG mitigation in MSW management. The objective of this study was to estimate alternative waste treatment practices towards the GHG emission mitigation, energy consumption and generation, reduction of landfill volume, and various benefits for proposing the appropriate selection by scenario analyses for representative Vietnam's cities. Impacts were calculated by utilizing life cycle assessment (LCA) method. A literature review survey on the current applicability of LCA database for assessing impacts from waste sector in developing countries, especially for Vietnam, was carried out. This study assessed the contribution of alternative solid waste treatment practices. The result showed that, except investment and operation costs, incineration with energy recovery seems the suitable alternative for treating waste from representative cities of Vietnam according to reduction of GHG emission and waste burden to landfill sites and energy recovery and generation. Besides, MSW composition was identified as an important factor directly influencing to impacts as well as other products and benefits of waste treatment alternatives. Reliable data on waste composition are indispensable for assessing to choose, improve, or plan the waste treatment practices towards sustainable development.

Waste disposal is an important part of the life cycle of a product and is associated with environmental burdens like any other life-cycle stages. In this study, an integrated assessment for solid waste treatment practices, especially household solid waste, was undertaken to evaluate the impact contribution of household solid waste treatment alternatives towards the sustainable development by using Life Cycle Inventory Assessment method. A case study has been investigated under various possible scenarios, such as (1) landfill without landfill gas recovery, (2) landfill with landfill gas recovery and flaring, (3) landfill with landfill gas recovery and electric generation, (4) composting, and (5) incineration. The evaluation utilized the Life Cycle Inventory Assessment method for multiple assessments based on various aspects, such as greenhouse gas emission/reduction, energy generation/consumption, economic benefit, investment and operating cost, and land use burden. The results showed that incineration was the most efficient alternative for greenhouse gas emission reduction, economic benefit, energy recovery, and land use reduction, although it was identified as the most expensive for investment and operating cost, while composting scenario was also an efficient alternative with quite economic benefit, low investment and operating cost, and high reduction of land use, although it was identified as existing greenhouse gas emission and no energy generation. Furthermore, the aim of this study was also to establish localized assessment methods that waste management agencies, environmental engineers, and environmental policy decision makers can use to quantify and compare the contribution to the impacts from different waste treatment options.

Plastic solid waste has become a serious problem when considering the disposal alternatives following the sequential hierarchy of sound solid waste management. This study was undertaken to assess the quantity and composition of household solid waste, especially plastic waste to identify opportunities for waste recycling. A 1-month survey of 130 households was carried out in Can Tho City, the capital city of the Mekong Delta region in southern Vietnam. Household solid waste was collected from each household and classified into ten physical categories; especially plastic waste was sorted into 22 subcategories. The average household solid waste generation rate was 281.27 g/cap/day. The compostable and recyclable shares respectively accounted for high percentage as 80.74% and 11%. Regarding plastic waste, the average plastic waste generation rate was 17.24 g/cap/day; plastic packaging and plastic containers dominated with the high percentage, 95.64% of plastic waste. Plastic shopping bags were especially identified as the major component, accounting for 45.72% of total plastic waste. Relevant factors such as household income and household size were found to have an existing correlation to plastic waste generation in detailed composition. The household habits and behaviors of plastic waste discharge and the aspects of environmental impacts and resource consumption for plastic waste disposal alternatives were also evaluated.

This study was undertaken to evaluate the quantity and composition of household solid waste to identify opportunities for waste recycling in Can Tho city, the capital city of the Mekong Delta region in southern Vietnam. Two-stage survey of 100 households was conducted for dry season and rainy season in 2009. Household solid waste was collected from each household and classified into 10 physical categories and 83 subcategories. The average household solid waste generation rate was 285.28 g per capita per day. The compostable and recyclable shares respectively accounted for 80.02% and 11.73%. The authors also analyzed the relations between some socioeconomic factors and household solid waste generation rates by physical categories and subcategories. The household solid waste generation rate per capita per day was positively correlated with the population density and urbanization level, although it was negatively correlated with the household size. The authors also developed mathematical models of correlations between the waste generation rates of main physical categories and relevant factors, such as household size and household income. The models were proposed by linear models with three variables to predict household solid waste generation of total waste, food waste, and plastic waste. It was shown that these correlations were weak and a relationship among variables existed. Comparisons of waste generation by physical compositions associated with different factors, such as seasonal and daily variation were conducted. Results presented that the significant average differences were found by the different seasons and by the different days in a week; although these correlations were weak.
