Abstract: In HF/SbF5 at −78°C, quinidinone 1 yields fluoroketone 3 (50% yield). The reaction implies a cyclic carboxonium ion as an intermediate, which reacts through a concerted rearrangement and fluorination to yield ketone 3. [Copyright &y& Elsevier]

Abstract: In HF-SbF5, quinine 1a or its dihydrochloride rearranges into compound 3 (89%), the preferred conformation of the substrate favouring the observed cyclization. Under similar conditions epiquinine 2a dihydrochloride yields in equal amounts two 10,10-difluoro derivatives, epimeric at C-3. In this case, the more stable conformation of the substrate in which the benzylic hydroxyl group is ‘exo’ to the quinuclidyl moiety, prevents the cyclisation. Similarly acetates 1b and 2b give the corresponding 10,10-difluoro derivatives epimeric at C-3. Formation of gem-difluoro compounds implies the formation of chloro intermediates at C-10 followed by an hydride abstraction, yielding an α-chloronium ion. This one is trapped by a fluoride ion and leads to the product by halogen exchange. [Copyright &y& Elsevier]

Reaction of cyclic ketones 2–6 and imines 15–17 in HF/SbF5 in presence of CCl4 yield hydroxy or fluoroderivatives, hydride abstraction occurring at a site located far from the functional group. Whereas ketones 2–5 yield only hydroxy derivatives, through cyclic carboxonium ion, imines 15, 16 N-protonated in the media conditions give only fluoroderivatives 18, 19, respectively, after quenching with HF–pyridine. Ring contraction is operative when starting from large membered ring ketones 24 and 25, and imine 26, leading to a mixture of hydroxy or fluoro cyclohexanones and/or cycloheptanones. Since the pioneer reports of the groups of Akiyama and Terada on Brønsted acid organocatalysis, this field never stopped growing with the development of ingenious strategies for the activation of challenging poorly reactive substrates. The development of superacidic organocatalysts is an important way to selectively functionalize reluctant electrophiles and other approaches have also emerged such as the combination of Lewis and Brønsted acids as well as the consecutive organocatalysis and superacid activation. The field of medicinal chemistry is currently witnessing a deuterium rush owing to the remarkable properties of this element as bioisoster of hydrogen atom. Aromatic hydrogen isotope exchange (HIE) is one of the most studied strategies nowadays as it promises to access deuterium‐modified drugs directly from their non‐labeled parents. While most of the recent studies focus on metal‐catalyzed C−H activation strategy, the use of superacidic conditions has been largely overlooked. Taking advantage of the wide range of data coming from the quest for carbonic anhydrase (CA) fluorinated inhibitors, this Minireview attempts to give "general guidelines" on how to wisely insert fluorine atom(s) within an inhibitor moiety to precisely enhance or disrupt ligand–protein interactions, depending on the target location of the fluorine substitution in the ligand. Multiple approaches such as ITC, kinetic and inhibition studies, X‐ray crystallography, and NMR spectroscopy are useful in dissecting single binding contributions to the overall observed effect. Exploiting superacid activation, the reactivity of aminonitriles was enhanced through the transient formation of highly reactive ammonium–nitrilium superelectrophiles. Exploiting trimethylsilyltrifluoromethanesulfonate as Lewis acid, (hetero)aromatics underwent regioselective and direct carbamoylation. The method is based on the in situ generation of a highly electrophilic carbamoyl triflate active species. TMSOTf acts as Lewis acid to activate carbamoyl chlorides and generates highly electrophilic carbamoyl triflates in situ; these are active species for late‐stage aromatic carbamoylation. After hydrofluorination of ynesulphonamides in superacid or in the presence of hydrofluoric acid/base reagents, a series of α-fluoroenamides has been synthesised and tested for the inhibition of carbonic anhydrase (CA, EC 4.2.1.1) isoforms. This study reveals a new, highly selective family of cancer-related transmembrane human (h) CA IX/XII inhibitors. These original fluorinated ureido isosters do not inhibit the widespread cytosolic isoforms hCA I and II and selectively inhibit the transmembrane cancer-related hCA IX and XII, offering interesting new leads for future studies. Dodoneine (Ddn) is one of the active compounds identified from Agelanthus dodoneifolius, which is a medicinal plant used in African pharmacopeia and traditional medicine for the treatment of hypertension. In the context of a scientific program aiming at discovering new hypotensive agents through the original combination of natural product discovery and superacid chemistry diversification, and after evidencing dodoneine's vasorelaxant effect on rat aorta, superacid modifications allowed us to generate original analogues which showed selective human carbonic anhydrase III (hCA III) and L-type Ca2+ current inhibition. Internal C-aryl and C-alkyl glycosides derived from 2-aminoglycopyranoses have been synthesized, exploiting a HF-mediated stereoselective intramolecular glycosylation. These conditions are compatible with acetate protecting groups and allow introduction of aromatics with various electronic distributions at the anomeric position. This strategy also provides straightforward entry to original fluorinated sugar-azacycle hybrids via a tandem internal C-glycosylation/fluorination reaction starting from 2-N-allyl/propargyl glycopyranoses. Rate constants between ozone and triketone herbicides, tembotrione and sulcotrione, were determined between pH 2 and 12. Both compounds are completely transformed within seconds by ozone under real water treatment conditions. A reaction pathway was proposed based on the identification of transformation products. Toxicity evaluation usingVibrio fischeritest showed that a residual toxicity remained even after degradation of the major by-products, but biodegradation significantly increased from 50% to 70% for molar ozone to pesticide ratios from 0 to 2. An efficient and straightforward regio-controlled route to β-fluoro-γ-chlorinated amines was developed via an ammonium–chloronium superelectrophilic activation in superacid HF/SbF 5 . Through halogen directing effect the method was extended to a variety of (poly)halofluorinated building blocks amenable for further synthetic modifications. A series of substituted pyrrolidines and piperidines were synthesized using superacid HF/SbF5chemistry. Investigated as inhibitors of several human carbonic anhydrase (hCA, EC 4.2.1.1) isoforms, i.e. the cytosolic hCA I and II as well as the tumor-associated transmembrane isoforms hCA IX and XII, these compounds showed a never yet reported selectivity toward the human carbonic anhydrase hCA II. Ethnopharmacological relevance Dodoneine (Ddn) is one of the active compounds identified from Agelanthus dodoneifolius ( DC .) Polhill and Wiens, a medicinal plant used in traditional medicine for the treatment of hypertension. This dihydropyranone exerts hypotensive and vasorelaxant effects on rats, and two molecular targets have been characterized: the carbonic anhydrase and the L-type calcium channel in cardiomyocytes with biochemical and electrophysiological techniques, respectively. To further evaluate the involvement of these two molecular targets in vasorelaxation, the effect of Ddn on rat vascular smooth muscle was investigated. Material and methods The effects of Ddn on L-type calcium current and on resting membrane potential were characterized in A7r5 cell line using the whole-cell patch-clamp configuration. The molecular identities of carbonic anhydrase isozymes in smooth muscle cells were examined with RT-PCR. Vascular response was measured on rat aortic rings in an organ bath apparatus and the effect of Ddn on intracellular pH was determined by flow cytometry using the pH-sensitive fluorescent probe BCECF-AM [2,7-Bis-(2-Carboxyethyl)-5-(and-6)-Carboxyfluorescein, Acetoxymethyl Ester]. Results 100 µM Ddn reduced calcium current density of about 30%. In addition, carbonic anhydrase II, III, XIII and XIV were shown to be expressed in rat aorta and inhibited in smooth muscle cells by Ddn. This inhibition resulted in a rise in pH i of about 0.31, leading to K Ca channel activation, thereby inducing membrane hyperpolarization and vasorelaxation. The results of vascular reactivity experiments obtained with pharmacological tools acting on the L-type calcium current and carbonic anhydrase suggest that Ddn produces its vasorelaxant effect via the inhibition of these two molecular targets. Conclusion This study demonstrates that Ddn induced vasorelaxation by targeting two proteins involved in the modulation of excitation-contraction coupling: L-type calcium channels and carbonic anhydrase. (E)- and (Z)-α-fluoroenamides could be easily prepared with high levels of chemo- and regioselectivities by hydrofluorination of readily available ynamides with HF/pyridine. The scope and limitations of this new process for the hydrofluorination of ynamides, as well as the stability of the resulting α-fluoroenamides, have been extensively studied. Theoretical calculations at the MP2 and B3LYP levels of theory showed that the resulting fluoroenamides exhibit geometrical and electronic properties that partially mirror those of ureas, therefore demonstrating that the hydrofluorination of ynamides provides a general, straightforward, and user-friendly approach to bioisosteres of ureas, potent building blocks for biological studies and medicinal chemistry. A novel and efficient keteniminium-initiated cationic polycyclization is reported. Agelanthus dodoneifolius is one of the medicinal plants used in African pharmacopeia and traditional medicine for the treatment of cardiovascular diseases. A chemical analysis has identified one of the active principles: Dodoneine (Ddn). It is a new dihydropyranone which exerts hypotensive and vasorelaxant effects on rat. Since the mechanism of the hypotensive effect is unknown, we performed a variety of preclinical and mechanistic studies to characterize the specific cardiac effect of Ddn at tissue (ex-vivo) and cellular levels (in-vitro) in order to determine a molecular target. Ddn effects were evaluated in an isolated rat heart preparation using Langendorff retrograde perfusion and then, the effects of Ddn were characterized in freshly dissociated cardiac ventricular myocytes using the whole-cell patch-clamp configuration. Ex-vivo, Ddn produced a dose-dependent negative inotropic effect with an IC50 value of 10µM without changed heart rate. 100µM Ddn decreased left ventricular developed pressure of about 40%. In isolated cardiac myocytes, Ddn reduced I Ca,L density of about 30% with an IC50 value estimated at 3µM. Ddn did not change current–voltage relation but it shifted the inactivation curve toward negative potentials and modified the half inactivation potentials. Furthermore, Ddn induced a phasic-dependent blocking on I Ca,L. This study demonstrates that the hypotensive property of dodoneine is likely associated with a negative inotropic effect and the blockade of the L-type calcium channels. [ABSTRACT FROM AUTHOR]

Small chemical modifications of the basic scaffold revealed strong changes in the selectivity profile against different CA isoforms. These new compounds selectively inhibited isoforms CA I (K Is in the range of 0.13–0.76μM), III (K Is in the range of 5.13–10.80μM), XIII (K Is in the range of 0.34–0.96μM) and XIV (K Is in the range of 2.44–7.24μM), and can be considered as new leads, probably acting as non-zinc-binders, similar to other phenols/lactones investigated earlier. Abstract: The reactivity of aliphatic amides/ketones/imines and nitriles was studied in superacid HF/SbF5 in the presence of CCl4. After the identification of the reaction intermediates by in situ NMR experiments, we propose that the sp3C–H bond functionalization (fluorination) by the superacid system is strongly dependant on the superelectrophilic character of the distonic dicationic intermediates. Abstract: A novel series of 4-aminobenzofused sultams was prepared using an efficient synthetic procedure based on superacid mediated reactions followed either by copper-catalyzed coupling or SNAr reactions. Abstract: In HF/SbF5, in the presence of CCl4 or NBS, unsaturated or hydroxylated piperidines yield difluoroanalogues in good yields. The ability to perform hydride abstraction on chloroderivatives, and the ability to form bridged bromonium ions from unsaturated substrates seem to completely influence the reaction course. Tertiary substituted (fluorinated) benzenesulfonamides were synthesized in superacid HF/SbF5 and tested as inhibitors of human carbonic anhydrases (hCAs, EC 4.2.1.1).
