1. Effects of the consumption of algal biomass versus protein concentrate on postprandial satiety and metabolism

Abstract

Algae are promising sources of nutritious and sustainable protein, but little is known about their metabolic health impact and acceptability as meal ingredients. This acute, randomized, controlled, five-way crossover trial compared whole algal biomasses and their corresponding protein concentrates to soy protein concentrate in terms of palatability, appetite, satiety, and metabolic response. Nineteen healthy Chinese males (21–50 years, 18.5–25.0 kg/m2) consumed noodle meals supplemented with 10 g of nori biomass/protein concentrate (NB/NC), Chlorella vulgaris biomass/protein concentrate (CB/CC) or soy protein concentrate control (CON) in randomized order. At regular intervals, blood samples were collected to measure biochemical markers, while gastrointestinal tolerance, palatability, and appetite were assessed using questionnaires and visual analog scales (VAS). Results indicated that algae-enriched meals were well-tolerated and comparable to soy in both visual appeal and smell, with NB and CC outperforming soy in aftertaste (p < 0.05). There were no significant differences between treatments in glucose, insulin, C-peptide, appetite/satiety, plasma ghrelin, and GLP-1. However, exploratory analysis of serum triglycerides revealed significant time × treatment effects (p < 0.004) and differences in incremental area under the curve (iAUC0–120, p = 0.0249). Our findings reveal that algal biomasses and protein concentrates are as comparable to soy protein concentrate in palatability, satiety, and metabolic outcomes, highlighting their potential as practical, sustainable, and nutritious ingredients.

Abstract

BackgroundPelvic organ prolapse (POP) refers to symptomatic descent of the vaginal wall. To reduce surgical failure rates, surgical correction can be augmented with the insertion of polypropylene mesh. This benefit is offset by the risk of mesh complication, predominantly mesh exposure through the vaginal wall. If mesh placement is under consideration as part of prolapse repair, patient selection and counseling would benefit from the prediction of mesh exposure; yet, no such reliable preoperative method currently exists. Past studies indicate that inflammation and associated cytokine release is correlated with mesh complication. While some degree of mesh-induced cytokine response accompanies implantation, excessive or persistent cytokine responses may elicit inflammation and implant rejection. ObjectiveHere, we explore the levels of biomaterial-induced blood cytokines from patients who have undergone POP repair surgery to (1) identify correlations among cytokine expression and (2) predict postsurgical mesh exposure through the vaginal wall. MethodsBlood samples from 20 female patients who previously underwent surgical intervention with transvaginal placement of polypropylene mesh to correct POP were collected for the study. These included 10 who experienced postsurgical mesh exposure through the vaginal wall and 10 who did not. Blood samples incubated with inflammatory agent lipopolysaccharide, with sterile polypropylene mesh, or alone were analyzed for plasma levels of 13 proinflammatory and anti-inflammatory cytokines using multiplex assay. Data were analyzed by principal component analysis (PCA) to uncover associations among cytokines and identify cytokine patterns that correlate with postsurgical mesh exposure through the vaginal wall. Supervised machine learning models were created to predict the presence or absence of mesh exposure and probe the number of cytokine measurements required for effective predictions. ResultsPCA revealed that proinflammatory cytokines interferon gamma, interleukin 12p70, and interleukin 2 are the largest contributors to the variance explained in PC 1, while anti-inflammatory cytokines interleukins 10, 4, and 6 are the largest contributors to the variance explained in PC 2. Additionally, PCA distinguished cytokine correlations that implicate prospective therapies to improve postsurgical outcomes. Among machine learning models trained with all 13 cytokines, the artificial neural network, the highest performing model, predicted POP surgical outcomes with 83% (15/18) accuracy; the same model predicted POP surgical outcomes with 78% (14/18) accuracy when trained with just 7 cytokines, demonstrating retention of predictive capability using a smaller cytokine group. ConclusionsThis preliminary study, incorporating a sample size of just 20 participants, identified correlations among cytokines and demonstrated the potential of this novel approach to predict mesh exposure through the vaginal wall following transvaginal POP repair surgery. Further study with a larger sample size will be pursued to confirm these results. If corroborated, this method could provide a personalized medicine approach to assist surgeons in their recommendation of POP repair surgeries with minimal potential for adverse outcomes.

Abstract

Primary penile urethral cancer is a rare genitourinary malignancy arising from the epithelial lining of the urethra. Our patient is a 63-year-old male with newly diagnosed penile urethral carcinoma who presented with headache and was found to have leptomeningeal disease on imaging and cerebral spinal fluid analysis. He was treated with systemic and intrathecal chemotherapy with some response and improvement in symptoms. This is the first reported case of leptomeningeal disease as a complication of penile urethral carcinoma. Recognition and prompt treatment are important; however, overall prognosis of this entity remains poor.

7. Stopping School Shootings: Lessons from Parkland, Oxford, Uvalde, and the Federal Commission on School Safety

Key PointsAfter the school shooting in Parkland, Florida, President Donald Trump tasked a multiagency effort to review best practices for school safety, which issued a landmark Federal Commission on School Safety (FCSS) report containing practical and actionable advice to keep students safe.After the school shooting in Oxford, Michigan, which—judging by a lawsuit filed against the school district—appeared to parallel the Parkland shooting with red flags missed and administrative missteps made, the Biden administration declared that the FCSS no longer reflected federal policy.There is a willful campaign from social justice advocacy organizations to fight against some of the key best practices FCSS identified. An examination of the alleged circumstances of the Oxford shooting suggests that school leaders should take their cues on school safety not from advocacy organizations or the Biden administration but from a careful evaluation of previous tragedies.

8. Data from Hippo Pathway Effector Yap Is an Ovarian Cancer Oncogene

Abstract

The Hippo pathway regulates organ size and tumorigenesis in Drosophila and mammals and is altered in a variety of human cancers, yet it remains unclear if the Hippo pathway is of prognostic significance to cancer patients. Here we show that the key targets of Hippo signaling, transcriptional coactivators Yki and Yap, play a conserved role in promoting ovarian cancer in flies and humans, respectively. Whereas studies linking Yap to cancer in other tissues have focused on overall Yap levels, we show for the first time that subcellular levels of Yap show an exceptionally strong association with poor patient survival. Specifically, high levels of nuclear Yap (nYap), or low levels of cytoplasmic phosphorylated Yap (cpYap), associated with poor survival from ovarian cancer. Patients with both high nYap and low cpYap had ∼50% lower 5-year survival, and this combination is an independent prognostic marker for survival, with an exceptionally high hazard ratio of 7.8. We find that Yap2 is the predominantly expressed Yap isoform in both the ovarian surface epithelium (OSE) and epithelial ovarian cancers. Overexpression of Yap2 and phosphorylation-defective Yap2-5SA in immortalized OSE cells resulted in increased cell proliferation, resistance to cisplatin-induced apoptosis, faster cell migration, and anchorage-independent growth, whereas Yap knockdown resulted in increased sensitivity to cisplatin-induced apoptosis. Abstract

Uveal Melanoma (UM) is the most common primary intra-ocular tumor in adults. New diagnostic procedures and basic science discoveries continue to change our patient management paradigms. A recent meeting of the European Vision Institute (EVI) special interest focus group was held on “Outcome Measures of New Technologies in Uveal Melanoma”, addressing the latest advances in UM, starting with genetic developments, then moving on to imaging and treatment of the primary tumor, as well as to investigating the most recent developments in treating metastases, and eventually taking care of the patient’s wellbeing. This review highlights the meeting’s presentations in the context of the published literature.

Abstract

This paper characterizes the actual science performance of the James Webb Space Telescope (JWST), as determined from the six month commissioning period. We summarize the performance of the spacecraft, telescope, science instruments, and ground system, with an emphasis on differences from pre-launch expectations. Commissioning has made clear that JWST is fully capable of achieving the discoveries for which it was built. Moreover, almost across the board, the science performance of JWST is better than expected; in most cases, JWST will go deeper faster than expected. Abstract

To develop and validate a new tissue-based biomarker that improves prediction of outcomes in localized prostate cancer by quantifying the host response to tumor. We use digital image analysis and machine learning to develop a biomarker of the prostate stroma called quantitative reactive stroma (qRS). qRS is a measure of percentage tumor area with a distinct, reactive stromal architecture. Kaplan Meier analysis was used to determine survival in a large retrospective cohort of radical prostatectomy samples. qRS was validated in two additional, distinct cohorts that include international cases and tissue from both radical prostatectomy and biopsy specimens. In the developmental cohort (Baylor College of Medicine, n=482), patients whose tumor had qRS > 34% had increased risk of prostate cancer-specific death (HR 2.94; p=0.039). This result was replicated in two validation cohorts, where patients with qRS > 34% had increased risk of prostate cancer-specific death (MEDVAMC; n=332; HR 2.64; p=0.02) and also biochemical recurrence (Canary; n=988; HR 1.51; p=0.001). By multivariate analysis, these associations were shown to hold independent predictive value when compared to currently used clinicopathologic factors including Gleason score and PSA. qRS is a new, validated biomarker that predicts prostate cancer death and biochemical recurrence across three distinct cohorts. It measures host-response rather than tumor-based characteristics, and provides information not represented by standard prognostic measurements.

Abstract

Biconformal gravity, based on gauging of the conformal group to 2n dimensions, reproduces n-dim scale-covariant general relativity on the co-tangent bundle in any dimension. We generalize this result to include Yang-Mills matter sources formulated as SU\left(N\right) gauge theories with a twisted action on the full 2n-dimensional biconformal space. We show that the coupling of the sources to gravity does not stop the reduction of effective dimension 2n\rightarrow n of the gravity theory, and instead forces the Yang-Mills source to reduce to ordinary n-dimensional Yang-Mills theory on the gravitating cotangent bundle, with the usual Yang-Mills energy tensor as gravitational source. Abstract

Accurate diagnosis of cribriform Gleason pattern 4 (CrP4) prostate adenocarcinoma (PCa) is important due to its independent association with adverse clinical outcomes and as a growing body of evidence suggests that it impacts clinical decision making in PCa management. To identify reproducible features for diagnosis of CrP4, we assessed interobserver agreement among 27 experienced urologic pathologists of 60 digital images from 44 radical prostatectomies (RP) that represented a broad spectrum of potential CrP4. The following morphologic features were correlated with the consensus diagnosis (defined as 75% agreement) for each image: partial vs. transluminal glandular bridging, intraglandular stroma, 50% of luminal space) vs. loose, and regular vs. irregular contour. Interobserver reproducibility for the overall diagnostic agreement was fair (k=0.40). Large CrP4 had better agreement (k=0.49) compared to small CrP4 (k=0.40). Transluminal bridging, dense cellular proliferation, a clear luminal space along the periphery of gland occupying 50% of the glandular circumference were associated with consensus against CrP4. In summary, we identified reproducible morphological features for and against CrP4 diagnosis, which could be used to refine and standardize the diagnostic criteria for CrP4.

We study the relaxation dynamics of quantum turbulence in a two-component Bose-Einstein condensate containing half-quantum vortices. We find a temporal scaling regime for the number of vortices and the correlation lengths that at early times is strongly dependent on the relative strength of the inter-species interaction. Abstract

In this paper we consider infinite horizon discounted dynamic programming problems with finite state and control spaces, and partial state observations. We discuss an algorithm that uses multistep lookahead, truncated rollout with a known base policy, and a terminal cost function approximation. This algorithm is also used for policy improvement in an approximate policy iteration scheme, where successive policies are approximated by using a neural network classifier. A novel feature of our approach is that it is well suited for distributed computation through an extended belief space formulation and the use of a partitioned architecture, which is trained with multiple neural networks. Abstract

Em uma primeira etapa foi determinado o Centro de Massa do Corpo de cães mediante a aplicação de um método experimental, a prancha de reação e um método analítico baseado em equações de regressão. Esta avaliação básica forneceu parâmetros para, numa segunda etapa, efetuar um estudo cinemático da deambulação de cães submetidos a artroplastia capsular da articulação coxofemoral. O exame biomecânico, comparado à avaliação clínica, demonstrou ser um método objetivo (quantitativo) para avaliação de alterações na locomoção.

RESUMO A técnica de artroplastia capsular de Colonna padronizada para cães por WHEELER et al (1991) foi adaptada em 10 cães: cinco portadores de displasia, quatro de luxação e um com displasia e luxação coxofemoral. No pré e pós-operatório os animais foram avaliados clinica e radiologicamente. Um deles foi sacrificado e submetido a estudo anátomo e histopatológico. A técnica adotada proporcionou alívio da dor. A recuperação funcional foi excelente em 50% dos casos e boa nos demais.

RESUMO Em oito cães sadios, considerados livres de displasia procurou-se adaptar a técnica de artroplastia capsular de Colonna, utilizada em humanos para o tratamento da luxação coxofemoral congênita. Os animais foram submetidos às avaliações clínica e radiológica pré e pós-operatório e após período de observação variável, sacrificados para estudo anátomo-histopatológico. A técnica cirúrgica consistiu em abordagem crâneo-dorsal da articulação coxofemoral, aprofundamento do acetábulo, cobrimento da cabeça femoral com pericárdio homogêneo, reconstituição da cápsula articular e estruturas abordadas. O período de recuperação funcional variou de 20 a 45 dias. Os resultados obtidos mostraram melhora na congruência entre a cabeça femoral e fossa acetabular.

Abstract

Identifying the provenance of uranium-rich materials is a critical objective of nuclear forensic analysis. Rare earth element (REE) distributions within uranium ores are well-established forensic indicators, but quantifying and correlating trace element signatures for U ores to known deposits has thus far involved intricate statistical analyses. This study reports average chondrite normalized (CN)-REE signatures for important U deposit types worldwide, which are then employed to evaluate U ore paragenesis using a simple linear regression analysis. This technique provides a straightforward method that can aid in determining the deposit type of U ores based on their REE abundances, and combined with other forensic indicators (e.g. radiogenic isotope signatures) can provide essential provenance information for nuclear materials.

Abstract

Securing multi-robot teams against malicious activity is crucial as these systems accelerate towards widespread societal integration. This emerging class of “physical networks” requires research into new methods of security that exploit their physical nature. This paper derives a theoretical framework for securing multi-agent consensus against the Sybil attack by using the physical properties of wireless transmissions. Our frame-work uses information extracted from the wireless channels to design a switching signal that stochastically excludes potentially untrustworthy transmissions from the consensus. Intuitively, this amounts to selectively ignoring incoming communications from untrustworthy agents, allowing for consensus to the true average to be recovered with high probability if initiated after a certain observation time T 0 that we derive. This work is different from previous work in that it allows for arbitrary malicious node values and is insensitive to the initial topology of the network so long as a connected topology over legitimate nodes in the network is feasible. We show that our algorithm will recover consensus and the true graph over the system of legitimate agents with an error rate that vanishes exponentially with time.

Abstract

We consider the conformal group of a space of dim n=p+q, with SO(p,q) metric. The quotient of this group by its homogeneous Weyl subgroup gives a principal fiber bundle with 2n-dim base manifold and Weyl fibers. The Cartan generalization to a curved 2n-dim geometry admits an action functional linear in the curvatures. Because symmetry is maintained between the translations and the special conformal transformations in the construction, these spaces are called biconformal; this same symmetry gives biconformal spaces overlapping structures with double field theories, including manifest T duality. We establish that biconformal geometry is a form of double field theory, showing how general relativity with integrable local scale invariance arises from its field equations. While we discuss the relationship between biconformal geometries and the double field theories of T-dual string theories, our principal interest is the study of the gravity theory. We show that vanishing torsion and vanishing co-torsion solutions to the field equations overconstrain the system, implying a trivial biconformal space. Wih co-torsion unconstrained, we show that (1) the torsion-free solutions are foliated by copies of an n-dim Lie group, (2) torsion-free solutions generically describe locally scale-invariant general relativity with symmetric, divergence-free sources on either the co-tangent bundle of n-dim (p,q)-spacetime or the torus of double field theory, and (3) torsion-free solutions admit a subclass of spacetimes with n-dim non-abelian Lie symmetry. These latter cases include the possibility of a gravity-electroweak unification. It is notable that the field equations reduce all curvature components to dependence only on the solder form of an n-dim Lagrangian submanifold, despite the increased number of curvature components and doubled number of initial independent variables., 56 pages

Abstract

We develop the properties of Weyl geometry, beginning with a review of the conformal properties of Riemannian spacetimes. Decomposition of the Riemann curvature into trace and traceless parts allows an easy proof that the Weyl curvature tensor is the conformally invariant part of the Riemann curvature, and shows the explicit change in the Ricci and Schouten tensors required to insure conformal invariance. We include a proof of the well-known condition for the existence of a conformal transformation to a Ricci-flat spacetime. We generalize this to a derivation of the condition for the existence of a conformal transformation to a spacetime satisfying the Einstein equation with matter sources. Then, enlarging the symmetry from Poincar\'e to Weyl, we develop the Cartan structure equations of Weyl geometry, the form of the curvature tensor and its relationship to the Riemann curvature of the corresponding Riemannian geometry. We present a simple theory of Weyl-covariant gravity based on a curvature-linear action, and show that it is conformally equivalent to general relativity. Abstract

After two years of discussion, UN member states have finally agreed on a new set of Sustainable Development Goals (SDGs), which will replace the Millennium Development Goals (MDGs) and establish a global development framework for the next fifteen years. The SDGs which were formally adopted at the UN Substainable Development Summit on 25 September 2015. The Common African Position (CAP) calls for the SDGs to give adequate attention to peace and security, acknowledging the inextricable links between peace, security, stability, and development. While China has acknowledged the importance of peace for development, it initially voiced some skepticism over whether the issue should be addressed explicitly through the SDGs. Nonetheless, China has committed to coordinating its position with that of African countries and has now accepted the inclusion of Goal 16, which aims to promote peaceful and inclusive societies. Indeed, China has already expressed its strong support for African peace and security as an enabler for development in other forums. This paper demonstrates how many of the commitments made in the last Forum on China-Africa Cooperation (FOCAC) Action Plan (2013–2015) in fact overlap with many of the peace-related targets in the draft SDGs. This suggests that the upcoming 6th FOCAC meeting in South Africa in December 2015 could be an opportunity to discuss how the next Action Plan can serve as an implementation mechanism for the SDGs, particularly with regard to their focus on peace.

This paper provides a working example of an SoS architecture in Systems Modeling Language (SysML) using IBM Rational Rhapsody that was developed in support of the Defense Advanced Research Projects Agency (DARPA) Systems of Systems (SoS) program Cross-Domain Maritime Surveillance and Targeting (CDMaST). One goal of this effort was to provide the CDMaST program industry participants with an example of an executable SysML model that represented an SoS architecture and supported the DARPA desire for SoS documentation through a model-based implementation approach. The SoS architecture was defined as a selection of platforms, weapons, sensors, and mission systems (elements) operating collectively in the maritime environment, the assignment of weapons, sensors, and mission systems to platforms, the interfaces between elements, and the services SoS elements provide. An architecture element is a distinct component in the overall SoS architecture. Elements include platforms, weapons, communications and data links, sensors, battle managers (manned and unmanned), and systems that support mission processing (e.g. data fusion, targeting etc.). An SoS architecture model provides the framework for two lines of reasoning about the SoS. First, the model reflects the results of physics-based trades conducted to inform architecture decisions. Second, the model defines the systems and behaviors which will be analyzed to demonstrate their operational effectiveness in mission and campaign modeling analyses. The model aims to capture information that is important for analysis, implementation and testing activities.

Abstract

According to the Coleman-Mandula theorem, any gauge theory of gravity combined with an internal symmetry based on a Lie group must take the form of a direct product in order to be consistent with basic assumptions of quantum field theory. However, we show that an alternative gauging of a simple group can lead dynamically to a spacetime with compact internal symmetry. The biconformal gauging of the conformal symmetry of n-dim Euclidean space doubles the dimension to give a symplectic manifold. Examining one of the Lagrangian submanifolds in the flat case, we find that in addition to the expected SO(n) connection and curvature, the solder form necessarily becomes Lorentzian. General coordinate invariance gives rise to an SO(n-1,1) connection on the spacetime. The principal fiber bundle character of the original SO(n) guarantees that the two symmetries enter as a direct product, in agreement with the Coleman-Mandula theorem., Comment: Added section to discuss full space as cotangent bundle

Of those gauge theories of gravity known to be equivalent to general relativity, only the biconformal gauging introduces new structures - the quotient of the conformal group of any pseudo-Euclidean space by its Weyl subgroup always has natural symplectic and metric structures. Using this metric and symplectic form, we show that there exist canonically conjugate, orthogonal, metric submanifolds if and only if the original gauged space is Euclidean or signature 0. In the Euclidean cases, the resultant configuration space must be Lorentzian.
