Anaplastic thyroid carcinoma is arguably the most lethal human malignancy. It often co-occurs with differentiated thyroid cancers, yet the molecular origins of its aggressivity are unknown. We sequenced tumor DNA from 329 regions of thyroid cancer, including 213 from patients with primary anaplastic thyroid carcinomas. We also whole genome sequenced 9 patients using multi-region sequencing of both differentiated and anaplastic thyroid cancer components. Using these data, we demonstrate thatanaplastic thyroid carcinomas have a higher burden of mutations than other thyroid cancers, with distinct mutational signatures and molecular subtypes. Further, different cancer driver genes are mutated in anaplastic and differentiated thyroid carcinomas, even those arising in a single patient. Finally, we unambiguously demonstrate that anaplastic thyroid carcinomas share a genomic origin with co-occurring differentiated carcinomas and emerge from a common malignant field through acquisition of characteristic clonal driver mutations.

Lineage transitions are a central feature of prostate development, tumourigenesis and treatment resistance. While epigenetic changes are well known to drive prostate lineage transitions, it remains unclear how upstream metabolic signalling contributes to the regulation of prostate epithelial identity. To fill this gap, we developed an approach to perform metabolomics on primary prostate epithelial cells. Using this approach, we discovered that the basal and luminal cells of the prostate exhibit distinct metabolomes and nutrient utilization patterns. Furthermore, basal-to-luminal differentiation is accompanied by increased pyruvate oxidation. We establish the mitochondrial pyruvate carrier and subsequent lactate accumulation as regulators of prostate luminal identity. Inhibition of the mitochondrial pyruvate carrier or supplementation with exogenous lactate results in large-scale chromatin remodelling, influencing both lineage-specific transcription factors and response to antiandrogen treatment. These results establish reciprocal regulation of metabolism and prostate epithelial lineage identity.

Red Culture is one of the symbols of Chinese national spirit in China. With the enhancement of China's international status, the world gets to know the Red Culture stage by stage. And yet, in the process of Red Culture's foreign publicity, the depth and breadth of cultural diffusion have been affected by different factors. This paper objectively analyzes the current situation of foreign publicity translation about Red Culture and then focuses on the Red tourism industry to explore the characteristics and advantages of foreign-oriented publicity translation under the intercultural communicative perspective. In the end, we are trying to come up with some methods to increase the accuracy of Red Culture publicity translation.

Using dense global navigation satellite system data and brightness temperature data across the New Zealand and Australia regions, we tracked the propagation of traveling ionospheric disturbances (TIDs) associated with the 15 January 2022 Tonga volcanic eruptions. We identified two shock wave-related TIDs and two Lamb wave-related TIDs following the eruptions. The two shock wave-related TIDs, propagating with velocities of 724–750 and 445–471 m/s, respectively, were observed around New Zealand and Australia within a distance of 3500–6500 km from the eruptive center. These shock wave-related TIDs suffered severe attenuation during the propagation and disappeared more than 6500 km from the eruptive center. Based on the TEC data from the nearest ground-based receivers, we estimated the onset times of two main volcanic explosions at 04:20:54 UT ± 116 s and 04:24:37 UT ± 141 s, respectively. The two shock wave-related TIDs were most likely generated by these two main volcanic eruptions. The two Lamb wave-related TIDs propagated with velocities of 300–370 and 250 m/s in the near-field region. The Lamb wave-related TIDs experienced minimal attenuation during their long-distance propagation, with only a 0.17% decrease observed in the relative amplitudes of the Lamb wave-related TIDs from the near-field to far-field regions.

With the advent of the energy Internet and the swift growth of unified energy systems, the comprehensive energy demand of users has gradually become a problem that cannot be ignored for the planning of integrated energy systems. Aiming at this problem, this paper suggests a multi-agent planning approach for electricity and gas, considering users’ holistic energy consumption behavior. First, utilizing a combined subjective and objective weighting method, this study establishes a utility model for users’ energy consumption characteristics. The analysis of comprehensive energy consumption behavior is conducted through an evolutionary game. On this basis, the planning revenue model for electricity grid and gas network investors is formulated, and the game mechanism of different investors is analyzed. A dynamic game model of electricity–gas multi-agent planning considering comprehensive energy consumption behavior is proposed. Ultimately, the model is resolved using an iterative exploration approach. The validity and efficacy of the proposed method are confirmed through a simulation example.

Abstract Background Home confinement during the epidemic has a significant impact on the lifestyle and behavior of school-aged children, who have exhibited an increase in the prevalence and development of myopia. Our research will look at if home confinement will affect school-aged children on myopia control with orthokeratology. Method Data on axial length was gathered from school-aged children who had received OK lenses treatment. The entire data was separated into subgroups based on gender, age, and initial refraction, and the AL changes for each period were calculated using the formula defined in our study. Finally, the acquired data will be examined using various statistical approaches, and the ideas of slow, moderate, and rapid myopia progression will be applied to our study. Result A total of 258 study subjects met the requirements to be included in the study. We discovered that the percentage of rapid myopia growth increased during the epidemic. In addition, the AL changes before and during the epidemic were found to be statistically significant in 171 subjects in the overall data. (P = 0.041) In the high age group, the AL changes before and during the epidemic、(P = 0.033) before and after the epidemic (P = 0.023) were found to be statistically significant. The AL changes before and during the epidemic (P = 0.035) were shown to be statistically significant in the moderate myopia group. Finally, we did not find statistically significant results for other groups. Conclusion We cannot conclude that home confinement did have a negative impact on myopia control with orthokeratology in school-aged children. But we found there was an increase in the percentage of patients with OK treatment that had fast myopia progression during the confinement. We also observed that older children with higher initial refraction were more likely to be affected by home confinement.

The universality of load subjects in distribution network brings challenges to the reliability of distribution network planning results. In this paper, a two-stage dynamic robust distribution network planning method considering correlation is proposed. The method evaluates the correlation between random variables using the Spearman rank correlation coefficient, and converts the correlated random variables into mutually independent random variables by Cholesky decomposition and independent transformation; expresses the source-load uncertainty by a bounded interval without distribution, and describes the active distribution network planning as a dynamic zero-sum game problem by combining with the two-phase dynamic robust planning; use the Benders decomposition approach to tackle the issue; mathematical simulation is used to confirm the accuracy and efficacy of the method. The results show that the dynamic robustness planning method of active distribution network taking into account the correlation can accurately simulate the operation of active distribution network with uncertain boundaries, which enhances the reliability and economy of the active distribution network planning results.

Abstract The Multiband Ultraviolet Spectrum Imager (MUSI) is an optical remote sensing instrument scheduled to launch on the Fengyun‐4C meteorological satellite in 2024. MUSI is designed to measure the airglow emissions between ∼120 and 160 nm above the East Asia and Pacific region from a geostationary orbit to infer ionospheric parameters. Here we develop a lookup‐table algorithm for retrieving the peak electron density (nmF2) and the Total Electron Content (TEC) from nighttime observations of the OI 135.6 nm emission. The algorithm takes into account the north‐south asymmetry of the equatorial ionization anomaly and the atmospheric variations with solar activity and local time. Analysis of synthetic observations shows that the assumptions made in the algorithm only lead to ∼2%–4% uncertainties in the retrievals under the modeled ionospheric conditions. The algorithm is verified by retrieving nmF2 and TEC from the Global‐Scale Observations of the Limb and Disk (GOLD) mission and comparing with coincided radio measurements. The comparison indicates almost negligible systematic differences (typically less than ∼3%) between the GOLD and the ionosonde nmF2, which are within the error limit of the GOLD retrievals. The accuracy of the GOLD TEC retrievals is shown to be comparable to (or in some sense even better than) that of the measurements from the Global Navigation Satellite System. Several simplified yet robust retrieval methods are also developed, which can be readily implemented for both existing and upcoming missions. Our results promote far ultraviolet (FUV) remote sensing to be a key technology for accurate determination of the ionospheric state.

Given the recent escalation of the energy and environmental crises, it is crucial to consider the evolution of various energy systems from a macro perspective in order to increase the overall efficiency of energy use and improve the environment. In order to tackle this issue, a wide-area integrated energy system planning method that takes into account consumers' comprehensive energy consumption behavior (CECB) and policy recommendations is provided in this study. First of all, according to the analysis of users’ CECB, the decision-making model of regional users' energy supply transformation is established. A game model is constructed to explore cooperative planning of an integrated electricity-natural gas energy system based on the profit models of various market participants. The government is considered as the main body of planning and decision-making, and the evaluation model of government’s pollution emissions from both supply and demand sides is established. A two-level dynamic game planning model for the wide-area IES is then created as a result of the examination of the game mechanism between users, the government, the integrated energy system (IES), and other market players. Finally, the model is solved using an iterative search approach. Results from simulations are used to validate the suggested method’s accuracy and efficacy.

We develop a preliminary interplanetary disturbance index ( J ${}_{{\rm{s}}}^{{\rm{W}}}$ ) by applying the spectral whitening method to an energy coupling function with solar wind measurements during the years 1998–2014 as the input, which can be used as an indicator of perturbations in the near-Earth solar wind. The correlation and temporal variation between J ${}_{{\rm{s}}}^{{\rm{W}}}$ and the geomagnetic disturbance index ( J ${}_{{\rm{p}}}^{{\rm{G}}}$ ) constructed from the same method have been analyzed in detail for 167 geomagnetic storms with the minimum D _st index less than or equal to −50 nT. The time delay between J ${}_{{\rm{s}}}^{{\rm{W}}}$ and J ${}_{{\rm{p}}}^{{\rm{G}}}$ is clearly observable and varies for different events, according to which J ${}_{{\rm{s}}}^{{\rm{W}}}$ is shifted backward with respect to J ${}_{{\rm{p}}}^{{\rm{G}}}$ . We obtain a fairly good negative correlation between the shifted J ${}_{{\rm{s}}}^{{\rm{W}}}$ and the J ${}_{{\rm{p}}}^{{\rm{G}}}$ indices for the majority of events, and the significance level for 88% of the events (i.e., 147 events) does not exceed 0.05. A statistical analysis of the shifted J ${}_{{\rm{s}}}^{{\rm{W}}}$ and the J ${}_{{\rm{p}}}^{{\rm{G}}}$ indices for 147 selected events reveals that larger values of J ${}_{{\rm{s}}}^{{\rm{W}}}$ and smaller magnitudes of J ${}_{{\rm{p}}}^{{\rm{G}}}$ are commonly accompanied by enhanced southward magnetic fields, which implies that more solar wind energy is entering the magnetosphere and thus causing strong geomagnetic storms. Furthermore, a linear fit of the two indices suggests that the evolution of J ${}_{{\rm{p}}}^{{\rm{G}}}$ can be predicted about 2 hr in advance based on J ${}_{{\rm{s}}}^{{\rm{W}}}$ , indicating that J ${}_{{\rm{s}}}^{{\rm{W}}}$ can provide early warnings of possible disturbances in the geomagnetic fields, which is crucial for space weather monitoring and operational forecasting.

Objective To explore the function and mechanism of tamoxifen, an estrogen receptor modulator, in the pathogenesis of hyperplatic scar. Methods Primary human fibroblasts were isolated and cultured for 4 generations and then aliquated into 3 groups: control group, TGF-β1 group and TGF-β1+ tamoxifen group. The effects of tamoxifen on human fibrogenic factor production, cell proliferation, migration and differentiation of TGF-β1-induced skin fibroblasts were detected by real-time quantitative polymerase chain reaction (RT-qPCR), MTS, cell scratch test, Western blot and immunofluorescence assay. Results Tamoxifen (10 μmol/L) inhibited the production of collagen 1, collagen 3 and α-SMA induced by TGF-β1 in fibroblasts. MTS results showed that the proliferation activity of TGF-β1 group was higher than that of tamoxifen group (P＜0.05). Wound healing test verified that the migration ability of cells was reduced by tamoxifen intervention. Tamoxifen inhibited collagen secretion and differentiation of skin fibroblasts into muscle fibroblasts. Conclusions The estrogen receptor modulator tamoxifen inhibits pathological proliferation of fibroblasts, so it may be a potential novel strategy for the treatment of hyperplastic scar.

Objective: The influence of continuous renal replacement therapy (CRRT) on the steady-state plasma concentration of high-dose tigecycline was investigated in septic shock patients to provide references for drug dosing.Methods: In this prospective observational study, 17 septic shock patients presenting with severe infections needing a broad-spectrum antibiotic therapy with high-dose tigecycline (100 mg per 12 h) in the intensive care unit were included and divided into CRRT group (n = 6) or non-CRRT group (n = 11). The blood samples were collected and plasma drug concentration was determined by SHIMADZU LC-20A and SHIMADZU LCMS 8040. The steady-state plasma concentration was compared between groups using unpaired t-test. Furthermore, between-groups comparisons adjusted for baseline value was also done using multivariate linear regression model.Results: Peak concentration (Cmax) of tigecycline was increased in CRRT group compared to non-CRRT group, but there were no statistical differences (505.11 ± 143.84 vs. 406.29 ± 108.00 ng/mL, p-value: 0.129). Trough concentration (Cmin) of tigecycline was significantly higher in CRRT group than in non-CRRT group, with statistical differences (287.92 ± 41.91 vs. 174.79 ± 33.15 ng/mL, p-value: 0.000, adjusted p-value: 0.000). In safety, Cmin was reported to be a useful predictor of hepatotoxicity with a cut-off of 474.8 ng/mL. In our studies, Cmin of all patients in CRRT group was lower than 474.8 ng/mL.Conclusion: The plasma concentration of tigecycline was increased in septic shock patients with CRRT treatment and only Cmin shown statistical differences. Lead (Pb2+) pollution in the soil sub-ecosystem has been a continuously growing problem due to economic development and ever-increasing anthropogenic activities across the world. In this study, the photosynthetic performance and antioxidant capacity of Triticeae cereals (rye, wheat and triticale) were compared to assess the activities of antioxidants, the degree of oxidative damage, photochemical efficiency and the levels of photosynthetic proteins under Pb stress (0.5 mM, 1 mM and 2 mM Pb (NO3)2). Compared with triticale, Pb treatments imposed severe oxidative damage in rye and wheat. In addition, the highest activity of major antioxidant enzymes (SOD, POD, CAT, and GPX) was also found to be elevated. Triticale accumulated the highest Pb contents in roots. The concentration of mineral ions (Mg, Ca, and K) was also high in its leaves, compared with rye and wheat. Consistently, triticale showed higher photosynthetic activity under Pb stress. Immunoblotting of proteins revealed that rye and wheat have significantly lower levels of D1 (photosystem II subunit A, PsbA) and D2 (photosystem II subunit D, PsbD) proteins, while no obvious decrease was noticed in triticale. The amount of light-harvesting complex II b6 (Lhcb6; CP24) and light-harvesting complex II b5 (Lhcb5; CP26) was significantly increased in rye and wheat. However, the increase in PsbS (photosystem II subunit S) protein only occurred in wheat and triticale exposed to Pb treatment. Taken together, these findings demonstrate that triticale shows higher antioxidant capacity and photosynthetic efficiency than wheat and rye under Pb stress, suggesting that triticale has high tolerance to Pb and could be used as a heavy metal-tolerant plant.

In this paper, the thermoviscoelastic response of skin tissue, as a typical class of viscoelastic biological tissue, subject to thermal shock is studied based on the fractional Kelvin-Voigt model and fractional Pennes bioheat conduction. A time-fractional biothermoviscoelastic governing equation is derived and initial-boundary value conditions are given for monolayer skin tissue of finite thickness. The temperature change and thermal stress of viscoelastic skin tissue are obtained by the Laplace transform and its inversion using matlab software. The influences of viscosity coefficient, relaxation times, fractional-order viscosity/heat parameter, and blood perfusion rate of skin tissue on the temperature distribution and the elastic displacement are analyzed. The numerical results show that fractional-order viscosity does not affect temperature, whereas fractional-order heat flow does. Moreover, fractional-order heat flow affects the sensitivity of blood perfusion rate to the thermodynamic response. [ABSTRACT FROM AUTHOR]

In real applied fields such as clinical medicine, environmental sciences, psychology as well as economics, we often encounter the task of conducting statistical inference for longitudinal data with ordinal responses. The traditional methods of longitudinal data analysis are often inclined to model continuous responses, which are no longer suitable for such ordinal data. Logistic regression and probit regression are two considerable methods which are frequently used to model ordinal longitudinal responses. However, such modelling methods just depict the mean feature of latent outcome variable and may produce non-robust results when encountering nor-normal errors or outliers. As a proper alternative of mean regression models, composite quantile regression (CQR) method is usually employed to derive robust estimation. The target of this paper is to investigate the CQR estimation approach for ordinal latent longitudinal model. The joint Bayesian hierarchical model is established and a relative CQR estimation approach is suggested to conduct posterior inference for the considered model. Further, in longitudinal data modelling, excessive predictors may be brought into in the models which result in the decrease of the model prediction precision. Bayesian $ L_{1/2} $ L 1 / 2 regularized prior is incorporated into ordinal longitudinal CQR model to conduct variable selection simultaneously. As the amount of distributed energy storage (DES) in a power system continues to increase, it will not be long before there are multiple DES aggregators participating in frequency regulation, and the realization of their coordinated control is a critical topic of current research. This study focused on the primary frequency regulation (PFR) power allocation strategy among multiple DES aggregators participating in PFR. This study first calculated the PFR demand according to a system frequency response model of the power system with DESs. Next, a PFR power allocation model of DES aggregators was developed based on model predictive control. The objective of this model was to minimize the overall frequency regulation cost while satisfying all of the constraints of DESs. Finally, the distributed interior point method was used to solve the model rapidly. The correctness and effectiveness of the proposed model and algorithm were verified on two unified transmission and distribution systems with DES aggregators used to supply the PFR service. The results revealed that the proposed model could effectively allocate PFR power to the various types of energy storage, with the additional benefits of slowing down the shift in the state of charge for energy storage units and ensuring the continuity of energy storage participation in frequency regulation.

Heavy metal air pollution poses a serious threat to human health and the environment in Chinese tourist cities. In this study, we investigated the temporal and spatial variations of atmospheric heavy metal pollution using moss bags in Xichang, a tourist destination in Southwest China. The biomonitoring investigation used an indigenous moss (Taxiphyllum taxirameum) transplanted into bags. Moss bags were exposed to 22 sites including industrial, agricultural, urban/residential, tourist, and high-traffic sites, across four different seasons in 2019–2020. The results showed that T. taxirameum was a good biomonitor of air pollution in Xichang. Among the 22 sample points, air pollution was the worst along the G102 motorway. Heavy metal emissions varied in different regions and directions. Temporal changes significantly influenced the heavy metals accumulated in moss bags, with low deposition of most elements observed at nearly all sampling sites in summer. Different seasons and regions were important factors affecting atmospheric heavy metal pollution. Based on the correlation analysis and the positive matric factorization model, the results revealed that heavy metals in moss bags in Xichang were mainly derived from anthropogenic sources and atmospheric deposition. Overall, this research provides an important reference for air pollution monitoring in urban areas.

Photosynthetic capacity is usually affected by light intensity in the field. In this study, photosynthetic characteristics of four different Triticeae crops (wheat, triticale, barley, and highland barley) were investigated based on chlorophyll fluorescence and the level of photosynthetic proteins under high light. Compared with wheat, three cereals (triticale, barley, and highland barley) presented higher photochemical efficiency and heat dissipation under normal light and high light for 3 h, especially highland barley. In contrast, lower photoinhibition was observed in barley and highland barley relative to wheat and triticale. In addition, barley and highland barley showed a lower decline in D1 and higher increase in Lhcb6 than wheat and triticale under high light. Furthermore, compared with the control, the results obtained from PSII protein phosphorylation showed that the phosphorylation level of PSII reaction center proteins (D1 and D2) was higher in barley and highland barley than that of wheat and triticale. Therefore, we speculated that highland barley can effectively alleviate photodamages to photosynthetic apparatus by high photoprotective dissipation, strong phosphorylation of PSII reaction center proteins, and rapid PSII repair cycle under high light.

Objective To investigate the association between triglyceride glucose index (TyG) combined with waist circumference (WC) and the risk of new-onset coronary heart disease (CHD) in the middle-aged and elderly people in 10 years of non-interventional follow-up in Luzhou area. Methods A total of 8 647 eligible individuals participated in epidemiological survey in Luzhou area were selected. The subjects were divided into different groups according to the quartiles of the TyG-WC and followed for ten years. The correlation between different TyG-WC quartile group and 10-year new CHD in middle-aged and elderly population was analyzed by using univariate analysis and Cox regression analysis. Results Of the 8 647 participants in the study, 484 had new CHD, accounting for 5.5%. The comparison of the number of newly diagnosed CHD subjects grouped by TyG-WC quartiles indicated the 4th quartile group > the 3rd quartile group > the 2nd quartile group > the 1st quartile group, showing statistical significance (P < 0.001). Cox regression analysis showed that without adjusting for any variables, the risk of new CHD in 10 years increased with the increase of TyG-WC quartile group, and the differences between the second, third and fourth quartile array and the first quartile array were statistically significant (P < 0.05). After adjusting for age and sex and further adjusting for the influence of various influencing factors, the risk of new CHD in 10 years increased with the increase of TyG-WC quartile group. The difference between the third and fourth quartile array and the first quartile array was statistically significant (P < 0.05). Conclusion TyG-WC is a risk factor for new-onset CHD in the middle-aged and elderly population in Luzhou area in 10 years of non-interventional follow-up, and TyG-WC may be a predictor of the risk of CHD in the middle-aged and elderly population.
