The laccase activity of three cultivated Pleurotus strains was investigated using different lignocellulosic biomasses for solid-state fermentation. The lignocellulosic biomasses were Sophora japonica, Salix babylonica, Populus beijingensis, and Pinus tabuliformis, which were selected because of their laccase producing ability and low cost. The maximum laccase activities from P. ostreatus Han 1189, P. citrinopileatus Han 1192, and P. eryngii Han 1205 using Sophora japonica were 768.74 U/L ± 11.25 U/L, 144.77 U/L ± 9.70 U/L, and 200.42 U/L ± 6.85 U/L, respectively. The three species of Pleurotus showed consistency in having a preference of hardwood to secret laccase under solid-state fermentation when facing hardwood and softwood as substrates. Furthermore, the presence of Sophora japonica was beneficial to improving the laccase activity for three Pleurotus strains. The capacity of the laccase secretion of P. ostreatus Han 1189 was superior to the capacity of P. citrinopileatus Han 1192 and P. eryngii Han 1205. The discovery was helpful for obtaining new high-producing strains and suitable wood materials, as well as expanding these high-producing strains for industrial applications to obtain high laccase activity, high laccase yield, and low cost laccase.

Laccase has huge potential application in all aspects of biotechnology due to the ability of laccase to oxidize a wide range of phenolic and non-phenolic compounds. The future success of such applications requires large amounts of laccase with low costs. White-rot fungi are important groups of laccase production. In this study, three white-rot fungi, Phlebia acerina Han 618, Trametes hirsuta Han 726, and Coriolopsis trogii Han 751, isolated from a native North China habitat, were identified by the method of molecular biology and preliminary screening of the ability of laccase-production by guaiacol selection medium. Then they were fermented on different lignocellulosic biomass. Three species showed consistency in preference of lignocellulosic biomass, and the presence of stalk of Sorghum bicolor was more suitable for secreting laccase. The capacity of laccase secretion from different species was significantly different. The capacity of secreting laccase of C. trogii Han 751 was superior to that of P. acerina Han 618 and T. hirsuta Han 726. The discovery of a new strain with superior capacity of secreting laccase and suitable lignocellulosic biomass were helpful for laying a foundation for the optimization of the fermentation conditions for the highest laccase production, the isolation and purification of laccase, and the industrial application of laccase.

Assembly of the adenovirus capsid protein hexon depends on the assistance of the molecular chaperone L4-100K. However, the chaperone mechanisms remain unclear. In this study, we found that L4-100K was involved in the hexon translation process and could prevent hexon degradation by the proteasome in cotransfected human cells. Two nonadjacent domains, 84-133 and 656-697, at the N-terminal and C-terminal regions of human adenovirus type 5 L4-100K, respectively, were found to be crucial and cooperatively responsible for hexon trimer expression and assembly. These two chaperone-related domains were conserved in the sequence of L4-100K and in the function of hexon assembly across different adenovirus serotypes. Different degrees of cross-activity of hexon trimerization with different serotypes were detected in subgroups B, C, and D, which were proven to be controlled by the interaction between the C-terminal chaperone-related domain of L4-100K and hypervariable regions (HVR) of hexon. Additionally, HVR-chimeric hexon mutants were successfully assembled with the assistance of the 1-697 mutant. Structural analysis of 656-697 by nuclear magnetic resonance and structural prediction of L4-100K using Robetta showed that the two conserved domains are mainly composed of α-helices and are located on the surface of the highly folded core region. Our research provides a more complete understanding of hexon assembly and guidance for the development of hexon-chimeric adenovirus vectors that will be safer, smarter, and more efficient. IMPORTANCE Adenovirus vectors have been widely used in clinical trials of vaccines and gene therapy, although some deficiencies remain. Chimeric modification of the hexon was expected to improve the potency of preexisting immune evasion and targeting, but in many cases, viral packaging is prevented by the inability of the chimeric hexon to assemble correctly. So far, few studies have examined the mechanisms of hexon trimer assembly. Here, we show how the chaperone protein L4-100K contributes to the assembly of the adenovirus capsid protein hexon, and these data will provide a guide for novel adenovirus vector design and development, as we desired.

As a common abiotic stress, drought severely impairs the growth, development and even sur-vival of plants. Here we report a transcription factor, Caragana korshinskii REVOLUTA(CkREV), which can bidirectionally regulate the expression of the critical enzyme gene CkYUC5 in auxin synthesis according to external environment changes, so as to control the biosynthesis of auxin and further enhance drought resistance of plants. Quantitative analysis reveals that the expression level of CkYUC5 and AtYUC5 is both down-regulated after C. korshinskii and Arabidopsis thaliana are exposed to drought. Functional verification of CkREV reveals that CkREV up-regulates the expression of AtYUC5 in transgenic A. thaliana under common conditions, while down-regulates it under drought conditions. Meanwhile, the expression of CkYUC5 is also down-regulated in C. korshinskii leaves instantaneously overexpressing CkREV. We apply dual-luciferase reporter system to discover that CkREV can bind to the promoter of CkYUC5 to regulate its expression, which is further proved by EMSA and Y1H experiments. Functional verification of CkREV in C. korshinskii and transgenic A. thaliana shows that CkREV can regulate the expression of CkYUC5 and AtYUC5 in a contrary way, maintaining the equilibrium of plants between growth and drought resisting. CkREV can positively regulate the expression of CkYUC5 to promote auxin synthesis in favor of growth under normal development. However, CkREV can also respond to external signals and negatively regulate the expression of CkYUC5, which inhibits auxin synthesis in order to reduce growth rate and lower water demands and eventually improves drought resistance of plants.

The classical four-equation [Formula: see text] transition model has presented excellent accuracy in low-speed boundary layer transition prediction. However, once the incoming flow reaches hypersonic speed, the original model is no longer applicable due to the compressibility problem and the appearance of multiple instability modes. Recently, there has been widespread interest in data-driven modeling for quantifying uncertainty or improving model prediction accuracy. In this paper, a data-driven framework based on field inversion and machine learning is performed to extend the prediction capability of the original [Formula: see text] transition model for the hypersonic boundary layer transition. First, the iterative regularized ensemble Kalman filter method is applied to obtain the spatial distribution of the perturbation correction term β for the switching function [Formula: see text], and the effectiveness of this method is initially verified in the hypersonic flat plate case. Then, the random forest algorithm is adopted to construct a mapping from the average flow features to β. The generalizability of the well-trained learning model is fully validated in the blunt cone cases with different unit Reynolds numbers, free-stream flow temperature, and bluntness. The simulation results indicate that the performance of the original [Formula: see text] transition model in the hypersonic boundary layer transition prediction is significantly improved, and the boundary layer transition onset location and the length of transition zone can be correctly obtained. In addition, the machine learning model investigates the importance of the input features and confirms that the effective length scale plays a significant role in the numerical simulation of the hypersonic boundary layer transition.

The paper systematically investigates the drag reduction of aerodisk fixed on the non-axisymmetric blunt body with an ellipsoid nose, different from conventional axisymmetric blunt body. The effects of spike length, aerodisk size and shape on drag reduction performance are numerically examined. Based on the analysis, a well-designed aerodisk configuration is determined and a maximum drag reduction of 41.9% is obtained for the whole blunt body. Meanwhile, some meaningful phenomena are discussed: (i) As the spike length increases, the drag of forebody and whole blunt body decreases first, then increases, and then decreases again. (ii) The total drag of the blunt body decreases first and then increases with the increase of aerodisk size. (iii) An elliptic disk with aspect ratio approaching that of the ellipsoid nose is superior to a round disk with the same size. This numerical study is expected to illustrate the significance of aerodisk to drag reduction over the non-axisymmetric blunt body and provide a reference for the design of aerodisk in hypersonic field.

Polydatin (PD), a natural precursor of resveratrol, has been used to treat several diseases, such as cardiovascular diseases, hepatic diseases and various cancers. In this study, we aimed to investigate the protective effects and underlying mechanisms of PD on non-alcoholic fatty liver disease (NAFLD) using a high fat induced obese mice model. The studied subjects were randomly divided into a lean group, a high fat diet (HFD) group, and a high fat diet with PD (HFD + PD) group. The results showed that PD reduced the body weights in HFD mice. PD also downregulated the serum levels of triglyceride (TG), low density lipoprotein (LDL), aspartate aminotransferase (AST) and alanine aminotransferase (ALT), and upregulated high density lipoprotein (HDL). Moreover, PD significantly alleviated hepatocyte steatosis and reduced Gr-1+ cells in the liver tissues of HFD mice. The mRNA levels of pro-inflammatory factors, such as monocyte chemoattractant protein-1 (MCP-1), tumor necrosis factor alpha (TNF-α), interleukin-6 (IL-6), S100A8 and S100A9 were significantly decreased in the liver tissues of HFD mice with PD treatment, and the downregulation of MCP-1 and S100A9 protein expressions was also observed. In conclusion, PD had beneficial roles in suppressing lipid accumulation in hepatocytes and anti-inflammatory responses in the liver tissue of obese associated NAFLD.

Magnetic perturbations characterize the solar wind interaction of the Moon. The solar wind plasma absorption on the dayside surface produces large-scale field perturbations behind, i.e., the field enhancement in the central wake and reduction on the wake boundary. The solar wind repellence over local lunar magnetic anomalies (LMAs) leads to small-scale magnetic compressions ahead. In this study, the magnetic perturbations around the Moon are examined by using the observations from a near-Moon satellite mission, the Lunar Prospector, and they exhibit a clear left–right asymmetry in a coordinate system related to the solar wind convection electric field ( ). The magnetic field is observed to enhance before the left terminator that points to, while on the opposite side, it is not. The test particle simulations show that can divert the particles reflected over the LMAs to the left and then the solar wind pickup of these particles leads to the field enhancement observed before the left terminator. Behind the lunar terminator, the wake field reduction is also asymmetric. On the left, the field reduction is more remarkable and located closer to the central wake. The denser plasma, consisting of the background as well as the reflected solar wind particles, may produce a stronger diamagnetic current and thus more significant field reduction there. The asymmetric plasma and magnetic perturbations associated with the reflected particles may be a common and nonnegligible element during the solar wind interaction of a small-scale magnetic field, such as that of an asteroid or a comet.

The paper aims to research on online mind map as interface of learning resource integration and sharing. We developed a web application that provides a interface to allow users to create a knowledge mind map. With the online mind mapping tool, the electronic learning resource will be discovered and shared in the form of subject-oriented package, it is more flexibly than the other E-Learning systems, and the interaction and cooperation of the users are strengthened to some extent.

To screen out the suitable erythrocytes compatible to the results from the routine blood matching for the patients suffered from the relapse of acute hemolytic transfusion reaction.The in vitro hemolysis test was used to screen out the erythrocytes from the non-hemolytic donors for the transfusion of erythrocytes into the patients.Three U of the non-hemolytic erythrocytes were obtained by using hemolytic test in vitro, the post-transfusion effects were good, and the hemolytic reaction will not occure once more.When it is compatible to the results obtained from the routine blood matching and the post-transfusion hemolytic reaction appeared. The blood matching by using in vitro hemolytic test can be used to screen out the non-hemolytic erythrocytes for transfusion of the patients.

In order to screen the compatible red cells by using extracorporal hemolysis test for acute autoimmune hemolytic anemia (AIHA) patients who were difficult to be matched by automatic microcolumn gel indirect antiglobulin test. Twenty-six cases of AIHA were chosen as control group, to whom the same type of donor red blood cells were infused with the weakest blood agglutination; 12 cases of acute AIHA patients were chosen as test group, these patients were difficult to be matched by automatic microcolumn gel indirect antiglobulin test, and the donor red cells without hemolysis by extracoral hemolysis test were transfused for them. The results showed that compared with the control group,the effect of transfusion was better in test group (P0.01), with 2.26 U leukocyte-depleted erythrocyte suspension in average, whose hemoglobin, reticulocyte and total bilirubin levels were changed significantly compared with those before blood transfusion (P0.01) . It is concluded that the compatible red blood cells for the acute AIHA patients can be screened by the extracorporal hemolysis test, when it is difficult to screen by the automatic microcolumn gel indirect antiglobulin test.
