Introduction: Paclitaxel (PTX) cannot effectively treat glioma because it cannot cross the bloodbrain barrier (BBB). A specific mode electroacupuncture stimulation (SMES) can temporarily open the BBB, thereby improving drug delivery to the brain. This study aimed to observe SMES-mediated accumulation of PTX in the brain and its anti-glioma effect and explore the role of the Hedgehog pathway. Methods: The acupoint selectivity of SMES in opening the BBB was examined in normal rats. The penetration and anti-glioma activity were determined in a C6-Luc glioma rat model. SMES was performed using 2/100 Hz, 3 mA, 6–6 s, and 40 min The survival curve was analysed by the KaplanMeier method, brain tumour pathology and size was observed by HE staining, and in vivo imaging system respectively. Results: SMES-induced BBB opening had acupoint selectivity. SMES could improve PTX accumulation in brain and SMES-mediated PTX delivery showed enhanced anti-glioma activity due to better brain penetration. Hedgehog pathway was involved in SMES-mediated PTX delivery by regulating Occludin expression. Conclusion: SMES at the head acupoints to deliver PTX is a feasible and effective method for treating glioma. The Hedgehog pathway may play a key role in SMES-mediated PTX delivery across the BBB.

Abstract Background In cancer patients receiving immune checkpoint inhibitors (ICIs), there is emerging evidence suggesting a correlation between gut microbiota and immune-related adverse events (irAEs). However, the exact roles of gut microbiota and the causal associations are yet to be clarified. Methods To investigate this, we first conducted a univariable bi-directional two-sample Mendelian randomization (MR) analysis. Instrumental variables (IVs) for gut microbiota were retrieved from the MiBioGen consortium (18,340 participants). GWAS summary data for irAEs were gathered from an ICIs-treated cohort with 1,751 cancer patients. Various MR analysis methods, including inverse variance weighted (IVW), MR PRESSO, maximum likelihood (ML), weighted median, weighted mode, and cML–MA–BIC, were used. Furthermore, multivariable MR (MVMR) analysis was performed to account for possible influencing instrumental variables. Results Our analysis identified fourteen gut bacterial taxa that were causally associated with irAEs. Notably, Lachnospiraceae was strongly associated with an increased risk of both high-grade and all-grade irAEs, even after accounting for the effect of BMI in the MVMR analysis. Akkermansia, Verrucomicrobiaceae, and Anaerostipes were found to exert protective roles in high-grade irAEs. However, Ruminiclostridium6, Coprococcus3, Collinsella, and Eubacterium (fissicatena group) were associated with a higher risk of developing high-grade irAEs. RuminococcaceaeUCG004, and DefluviitaleaceaeUCG011 were protective against all-grade irAEs, whereas Porphyromonadaceae, Roseburia, Eubacterium (brachy group), and Peptococcus were associated with an increased risk of all-grade irAEs. Conclusions Our analysis highlights a strong causal association between Lachnospiraceae and irAEs, along with some other gut microbial taxa. These findings provide potential modifiable targets for managing irAEs and warrant further investigation.

In this study, we examined the relationship between the sensitivity of interdigitated electrode (IDE) impedimetric biosensors and the gap between the IDEs. Our aim is to find an optimal design to maximize sensitivity. A three-dimensional COMSOL model was constructed for determining the effects of electrode gap, width, and height on impedance sensitivity, revealing a singular linear correlation with the inner gap. Considering both the simulation results and fabrication processes, we have developed three IDE prototype chips with electrode gaps of 3 μm, 4 μm, and 5 μm, respectively. For empirical validation, human anti-SARS-CoV-2 monoclonal antibody (mAb) was utilized, with immobilization of the SARS-CoV-2 spike protein on the chip's surface for mAb capture. This interaction, further amplified by Protein G conjugation, induced shifts in the impedance spectrum. The sensitivity of each prototype chip was evaluated across mAb concentrations ranging from 50 ng/mL to 500 ng/mL. The 3 μm configuration emerged as the most sensitive, demonstrating the ability to detect mAb concentrations as low as 50 ng/mL, a threshold unattainable by the other designs. This outcome underscores the critical influence of reduced inter-electrode gap on enhancing biosensor detection limits. The findings from this investigation offer a foundational approach for advancing biosensor sensitivity via electrode geometric optimization, with broad potential applications extending beyond COVID-19 diagnostics to a wide spectrum of clinical and research contexts.

Introduction Whether gastric cancer (GC) patients with deficient mismatch repair or microsatellite instability-high (dMMR/MSI-H) benefit from perioperative (neoadjuvant and/or adjuvant) chemotherapy is controversial. This protocol delineates the planned scope and methods for a systematic review and meta-analysis that aims to compare the efficacy of perioperative chemotherapy with surgery alone in resectable dMMR/MSI-H GC patients.Methods and analysis This study protocol is reported according to the Preferred Reporting Items for Systematic Reviews and Meta-Analysis Protocols-P guideline. PubMed, Embase, Cochrane (CENTRAL), and the Web of Science databases will be searched, supplemented by a secondary screening of relevant records. Both randomised controlled trials and non-randomised studies will be included in this study. The primary and secondary outcomes under scrutiny will be overall survival, disease-free survival and progression-free survival. Two reviewers will independently screen studies, extract data and assess the risk of bias. We will analyse different treatment settings (eg, neoadjuvant or adjuvant or combined as perioperative chemotherapies) separately and conduct sensitivity analyses.Ethics and dissemination No ethics approval is required for this systematic review and meta-analysis, as no individual patient data will be collected. The findings of our study will be published in a peer-reviewed journal.Prospero registration number CRD42023494276.

Abstract Scintillators, which can convert high‐energy particles (X‐rays) into detectable low‐energy ultraviolet–visible–near‐infrared photons, are essential components of X‐ray detectors and show extensive practical applications in nondestructive detection and medical imaging. Traditionally, inorganic scintillators represented by CsI:Tl have achieved definite progress. However, the harsh preparation conditions, high production cost, and poor mechanical properties impede their potential development in the high‐end X‐ray imaging field. Organic–inorganic hybrid metal complexes could be excellent alternatives, by virtue of their structural and spectral tunability, good solution processability, and excellent photophysical properties. This review mainly focuses on eco‐friendly lead‐free metal (Mn2+, Cu+, Sb3+, Sn2+, Ge2+, Ln3+, etc.) complex scintillators. The luminescence mechanisms are introduced and the scintillation performance, such as light yield, limit of detection, imaging resolution, etc., is highlighted. Moreover, the current challenges and perspectives in this emerging field are described. It is hoped to provide some theoretical guidance for the continuous development of the new scintillator systems.

Objectives: The objective of this study was to translate the Intensive Care Oral Care Frequency and Assessment Scale into Chinese and to evaluate its reliability and validity in Chinese ICU patients. Methods: This study was conducted using a cross-sectional survey design in ICUs of three tertiary hospitals in Huai'an and Taizhou from October 2022 to April 2023. The Chinese version of the Intensive Care Oral Care Frequency and Assessment Scale (C-ICOCFAS) was developed by expert consultation and cultural adaptation according to the two-person verbatim translation-back translation criteria described in the Brislin model. Item analysis was conducted using correlation analysis, and validity analysis included content validity, construct validity, and criterion validity. Reliability analyses included Cronbach's alpha coefficient, Guttman split-half reliability, and interrater reliability. Results: The Chinese version of the scale consisted of one dimension and nine items, consistent with the original version. Exploratory factor analysis showed KMO = 0.891, and the cumulative variance contribution reached 65.534 %. The confirmatory factor analysis indicated a good fit, χ2/df = 2.124, NFI = 0.950, GFI = 0.942, IFI = 0.973, CFI = 0.973, SRMR = 0.037, and RMSEA = 0.073. The content validity of the scale was 0.97, and the content validity of the items ranged from 0.83 to 1.00; the criterion validity was 0.969; the Cronbach's alpha coefficient was 0.919; the total item correlation coefficient was 0.725–0.831; the Guttman split-half reliability was 0.919; and the interrater reliability was 0.885. Conclusion: The C-ICOCFAS has good reliability and validity and can effectively guide nurses in the frequency of oral care for ICU patients. Implications for clinical practice: This tool can significantly improve the level of oral care among ICU patients and further promote the health and safety of patients. These findings can help clinical nursing experts to better understand and master the use of scales and standards to improve nursing.

Objectives: Assess the prevalence, mortality, and disability-adjusted life years (DALYs) of vascular intestinal disorders (VID) from 1990 to 2019.Methods: This study conducted a secondary data analysis utilizing the Global Burden of Diseases Study 2019. The prevalence, mortality and DALYs of VID were analyzed by sex, age and socio-demographic index (SDI), respectively. Analyses were performed by using R software.Results: Globally, the number of prevalent VID cases increased from 100,158 (95% uncertainty interval: 89,428–114,013) in 1,990–175,740 (157,941–198,969) in 2019. However, the age-standardized rates (ASR) of VID prevalence declined from 2.47 (95% uncertainty interval: 2.24–2.76) per 100,000 population to 2.21 (1.98–2.48) per 100,000 population between 1990 and 2019. Furthermore, the ASR of mortality also decreased from 1990 to 2019. Between 1990 and 2019, the regions with high and high-middle level exhibited the highest diseases burden.Conclusion: Globally, the diseases burden associated with VID demonstrated a decline from 1990 to 2019. However, concerted efforts are still required to enhance measures to combat VID within countries categorized as high and high-middle SDI.

BackgroundTo date, several studies have compared the surgical and oncological outcomes of local excision (LE) and radical excision (RE) for rectal gastrointestinal stromal tumors (GISTs), but some have limited numbers of small series. This protocol outlines the planned scope and methods for a systematic review and meta-analysis that will compare the surgical and oncological outcomes of LE and RE in patients with rectal GISTs.MethodsThis protocol is presented in accordance with the PRISMA-P guideline. PubMed, Embase, Web of Science, Cochrane Library and Wanfang database will be systematically searched. Furthermore, reference lists of all included articles will be screened manually to add other eligible studies. We will include randomized controlled trials (RCTs) and non-randomized studies (NRS) in this study. The primary outcomes evaluated will be R0 resection rate and disease-free survival, while the secondary outcomes will contain overall survival, length of stay, tumor rupture rate and complications. Two reviewers will independently screen and select studies, extract data from the included studies, and assess the risk of bias of the included studies. Preplanned subgroup analyses and sensitivity analyses are detailed within this protocol. The strength of the body of evidence will be assessed using GRADEDiscussionThis review and meta-analysis will provide a comprehensive evaluation of the current evidence concerning the application of LE and RE in patients with rectal GISTs. BackgroundMachine learning radiomics models are increasingly being used to predict gastric cancer prognoses. However, the methodological quality of these models has not been evaluated. Therefore, this study aimed to evaluate the methodological quality of radiomics studies in predicting the prognosis of gastric cancer, summarize their methodological characteristics and performance.MethodsThe PubMed and Embase databases were searched for radiomics studies used to predict the prognosis of gastric cancer published in last 5 years. The characteristics of the studies and the performance of the models were extracted from the eligible full texts. The methodological quality, reporting completeness and risk of bias of the included studies were evaluated using the RQS, TRIPOD and PROBAST. The discrimination ability scores of the models were also compared.ResultsOut of 283 identified records, 22 studies met the inclusion criteria. The study endpoints included survival time, treatment response, and recurrence, with reported discriminations ranging between 0.610 and 0.878 in the validation dataset. The mean overall RQS value was 15.32 ± 3.20 (range: 9 to 21). The mean adhered items of the 35 item of TRIPOD checklist was 20.45 ± 1.83. The PROBAST showed all included studies were at high risk of bias.ConclusionThe current methodological quality of gastric cancer radiomics studies is insufficient. Large and reasonable sample, prospective, multicenter and rigorously designed studies are required to improve the quality of radiomics models for gastric cancer prediction.Study registrationThis protocol was prospectively registered in the Open Science Framework Registry (https://osf.io/ja52b).

BackgroundHepatic hemangioma is among the most common benign liver lesions. However, giant pedunculated hepatic hemangiomas are exceptionally rare and associated with additional risks, such as torsion.Case presentationWe present the case of a 63-year-old female patient who presented with abdominal distension and pain. Barium meal examination and gastroscopy revealed a large, smooth-surfaced submucosal bulge located at the fundus of the stomach. Subsequent MRI examination identified a mass measuring approximately 6.4 x 7 cm in the left upper abdomen. Surgical intervention was planned for mass removal. However, intraoperative exploration revealed the origin of the mass to be the liver, and subsequent histopathological examination confirmed it as a hemangioma.ConclusionWe systematically summarized the characteristics of our case along with 31 previously reported cases. Giant pedunculated hepatic hemangiomas typically occur in the left lobe of the liver. Due to their atypical presentation, a combination of imaging methods such as ultrasound, CT, and/or MRI is essential for accurate diagnosis. Furthermore, surgical intervention is recommended due to the potential risks of bleeding, rupture, and torsion.

This paper presents a portable, fast and accurate electrochemical impedance spectroscopy (EIS) device with 8-well interdigitated electrode chips for biomarker detection. The design adopts low crest factor multisine signal synthesis at low frequencies (1 kHz), which significantly increases measurement speed without sacrificing accuracy. In addition, the low excitation amplitude of 10 mV preserves impedance linearity and protects the biosamples. The system achieved an average magnitude accuracy error of 0.30% in the frequency range of interest and it requires only 0.46 s to scan 28 frequency points from 10 Hz to 1 MHz. Experiments were conducted to test the capability to detect antibodies against SARS-CoV-2. Gold nanoparticles bound with protein G (GNP-G) were employed as the conjugated secondary antibody probe to detect anti-SARS-CoV-2 IgG in serum. A highly statistical significance (p = 7×10−6) could be found in the impedance data at 10 kHz. The impedance magnitude alteration caused by the GNP-G of the positive and negative groups were 27.2%±13.6% and 4.1%±1.7%, respectively. The results imply that the proposed system enables rapid COVID-19 antibody biomarker detection. Moreover, the EIS system and GNPs have the potential to be modified to detect other biomarkers.

Background: The global burden of digestive cancer is expected to increase. Therefore, crucial for the prognosis of patients with these tumors is to identify early diagnostic markers or novel therapeutic targets. There is accumulating evidence connecting histone chaperones to the pathogenesis of digestive cancer. Histone chaperones are now broadly defined as a class of proteins that bind histones and regulate nucleosome assembly. Recent studies have demonstrated that multiple histone chaperones are aberrantly expressed and have distinct roles in digestive cancers. Objective: The purpose of this review is to present the current evidence regarding the role of histone chaperones in digestive cancer, particularly their mechanism in the development and progression of esophageal, gastric, liver, pancreatic, and colorectal cancers. In addition, the prognostic significance of particular histone chaperones in patients with digestive cancer is discussed. Methods: According to PRISMA guidelines, we searched the PubMed, Embase, and MEDLINE databases to identify studies on histone chaperones and digestive cancer from inception until June 2022. Results: A total of 104 studies involving 21 histone chaperones were retrieved. Conclusions: This review confirms the roles and mechanisms of selected histone chaperones in digestive cancer and suggests their significance as potential prognostic biomarkers and therapeutic targets. However, due to their non-specificity, more research on histone chaperones should be conducted in the future to elucidate novel strategies of histone chaperones for prognosis and treatment of digestive cancer.

Gastrointestinal stromal tumors (GISTs) are considered insensitive to radiotherapy. However, a growing number of case reports and case series have shown that some lesions treated by radiotherapy achieved an objective response. The aim of the study was to perform a systematic review of all reported cases, case series, and clinical studies of GISTs treated with radiotherapy to reevaluate the role of radiotherapy in GISTs. A systematic search of the English-written literature was conducted using PubMed, Web of Science, and Embase databases. Overall, 41 articles describing 112 patients were retrieved. The included articles were of low to moderate quality. Bone was the most common site treated by radiotherapy, followed by the abdomen. In order to exclude the influence of effective tyrosine kinase inhibitors (TKIs), a subgroup analysis was conducted on whether and which TKIs were concurrently applied with radiotherapy. Results showed that radiotherapy alone or combined with resistant TKIs could help achieve objective response in selected patients with advanced or metastatic GISTs; however, survival benefits were not observed in the included studies. Pain was the most common symptom in symptomatic GISTs, followed by neurological dysfunction and bleeding. The symptom palliation rate was 78.6% after excluding the influence of effective TKIs. The adverse reactions were mainly graded 1–2. Radiotherapy was generally well-tolerated. Overall, radiotherapy may relieve symptoms for GIST patients with advanced or metastatic lesions and even help achieve objective response in selected patients without significantly reducing the quality of life. In addition to bone metastases, fixed abdominal lesions may be treated by radiotherapy. Publication bias and insufficient quality of included studies were the main limitations in this review. Further clinical studies are needed and justified.

Scintillators can convert high-energy X-rays into low-energy detectable ultraviolet-visible-near-infrared photons, which show extensive practical applications in X-ray detection and imaging. Metal-based organic-inorganic hybrid halides (OIHHs) are expected to become substitutes for commercial scintillators due to their flexible tunability of structure and spectra, good solution processability, and excellent photophysical properties. Among them, antimony-based (Sb3+) OIHH scintillators have exhibited highly efficient photophysical properties and scintillation performance due to the high stereo active lone pairs of the ns² electron configuration. Herein, we synthesized a series of Sb3+-based (TEBA)2SbCl5-xBrx (x = 0, 1, 2, 3, 4, 5) metal halides (TEBA: benzyltriethylammonium chloride) by regulating the ratio of halogen atoms (Cl/Br). (TEBA)2SbCl5-xBrx scintillators all show broadband emission, large Stokes shifts and microsecond lifetimes. With the increase of the Br- ion concentration, the photophysical properties and scintillation performance of (TEBA)2SbCl5-xBrx have been tuned. Among the six Sb3+-based OIHHs, (TEBA)2SbCl3Br2 exhibits the best scintillation performance with a high photoluminescent quantum yield of 73.83%, a high light yield of 15 000 photons MeV-1, and a low detection limit of 50.1 nGyair s-1. Besides that, X-ray imaging based on a (TEBA)2SbCl3Br2 scintillator screen presents a high spatial resolution of 10.4 lp mm-1. [ABSTRACT FROM AUTHOR]

According to the abnormal fracture phenomenon of 87MnSi steel wire rod for bridge cable， the evolution of microstructure and mechanical properties during production processing were systematically analyzed. The results show that the microstructure of the rod is composed of pearlite and a small amount of proeutectoid ferrite， and the cementite lamellae are randomly distributed. After cold drawing， the cementite lamellae are rotated or fibrotic， and some cementite are broken and dissolved， the dissolution amount is 1.40wt.%（9.69%）. After ho-dip galvanizing， the dissolved cementite is spheroidized. In the process of cold drawing， hot-dip galvanizing and stabilization treatment， the microhardness of wire rod （329.8HV） increases （450.2HV） firstly， then decreases （447.9HV） and increases （474.8HV） finally. The cementite dissolved in the cold drawing process increases the hardness of the steel wire to 468.3HV， while the spheroidized cementite in the hot galvanizing process decreases the hardness to 439.8HV， and deteriorates tensile property. When the degree of spheroidization of cementite on the edge is obvious， the microstructure and mechanical properties of the steel wire inside and outside are greatly different， and the fracture is eventually caused by uneven force during stabilizing treatment.
