Abstract

The Space Domain Awareness (SDA) community routinely tracks satellites in orbit by fitting an orbital state to observations made by the Space Surveillance Network (SSN). In order to fit such orbits, an accurate model of the forces that are acting on the satellite is required. Over the past several decades, high-quality, physics-based models have been developed for satellite state estimation and propagation. These models are exceedingly good at estimating and propagating orbital states for non-maneuvering satellites; however, there are several classes of anomalous accelerations that a satellite might experience which are not well-modeled, such as satellites that use low-thrust electric propulsion to modify their orbit. Physics-Informed Neural Networks (PINNs) are a valuable tool for these classes of satellites as they combine physics models with Deep Neural Networks (DNNs), which are highly expressive and versatile function approximators. By combining a physics model with a DNN, the machine learning model need not learn astrodynamics, which results in more efficient and effective utilization of machine learning resources. This paper details the application of PINNs to estimate the orbital state and a continuous, low-amplitude anomalous acceleration profile for satellites. The PINN is trained to learn the unknown acceleration by minimizing the mean square error of observations. We evaluate the performance of pure physics models with PINNs in terms of their observation residuals and their propagation accuracy beyond the fit span of the observations. For a two-day simulation of a GEO satellite using an unmodeled acceleration profile on the order of $10^{-8} \text{ km/s}^2$, the PINN outperformed the best-fit physics model by orders of magnitude for both observation residuals (123 arcsec vs 1.00 arcsec) as well as propagation accuracy (3860 km vs 164 km after five days).

Abstract

BackgroundThe etiology of frontotemporal dementia (FTD) is poorly understood. To identify genes with predicted expression levels associated with FTD, we integrated summary statistics with external reference gene expression data using a transcriptome-wide association study approach.MethodsFUSION software was used to leverage FTD summary statistics (all FTD: n = 2154 cases, n = 4308 controls; behavioral variant FTD: n = 1337 cases, n = 2754 controls; semantic dementia: n = 308 cases, n = 616 controls; progressive nonfluent aphasia: n = 269 cases, n = 538 controls; FTD with motor neuron disease: n = 200 cases, n = 400 controls) from the International FTD-Genomics Consortium with 53 expression quantitative loci tissue type panels (n = 12,205; 5 consortia). Significance was assessed using a 5% false discovery rate threshold.ResultsWe identified 73 significant gene-tissue associations for FTD, representing 44 unique genes in 34 tissue types. Most significant findings were derived from dorsolateral prefrontal cortex splicing data (n = 19 genes, 26%). The 17q21.31 inversion locus contained 23 significant associations, representing 6 unique genes. Other top hits included SEC22B (a gene involved in vesicle trafficking), TRGV5, and ZNF302. A single gene finding (RAB38) was observed for behavioral variant FTD. For other clinical subtypes, no significant associations were observed.ConclusionsWe identified novel candidate genes (e.g., SEC22B) and previously reported risk regions (e.g., 17q21.31) for FTD. Most significant associations were observed in dorsolateral prefrontal cortex splicing data despite the modest sample size of this reference panel. This suggests that our findings are specific to FTD and are likely to be biologically relevant highlights of genes at different FTD risk loci that are contributing to the disease pathology.

Abstract

There is no effective treatment for amyotrophic lateral sclerosis (ALS), a devastating motor neuron disease. However, discovery of a G4C2 repeat expansion in the C9ORF72 gene as the most common genetic cause of ALS has opened up new avenues for therapeutic intervention for this form of ALS. G4C2 repeat expansion RNAs and proteins of repeating dipeptides synthesized from these transcripts are believed to play a key role in C9ORF72-associated ALS (c9ALS). Therapeutics that target G4C2 RNA, such as antisense oligonucleotides (ASOs) and small molecules, are thus being actively investigated. A limitation in moving such treatments from bench to bedside is a lack of pharmacodynamic markers for use in clinical trials. We explored whether poly(GP) proteins translated from G4C2 RNA could serve such a purpose. Poly(GP) proteins were detected in cerebrospinal fluid (CSF) and in peripheral blood mononuclear cells from c9ALS patients and, notably, from asymptomatic C9ORF72 mutation carriers. Moreover, CSF poly(GP) proteins remained relatively constant over time, boding well for their use in gauging biochemical responses to potential treatments. Treating c9ALS patient cells or a mouse model of c9ALS with ASOs that target G4C2 RNA resulted in decreased intracellular and extracellular poly(GP) proteins. This decrease paralleled reductions in G4C2 RNA and downstream G4C2 RNA-mediated events. These findings indicate that tracking poly(GP) proteins in CSF could provide a means to assess target engagement of G4C2 RNA-based therapies in symptomatic C9ORF72 repeat expansion carriers and presymptomatic individuals who are expected to benefit from early therapeutic intervention.

The article presents the results of a survey ranking the best academic programs in international relations for undergraduate, master's, and doctoral degrees. Ivy League schools, Stanford University, and the University of Chicago were among the top choices for undergraduate programs, with Georgetown University being favored by policymakers and think tank staff. East Coast schools, particularly Georgetown University, were preferred for master's programs. The top five Ph.D. programs were Harvard University, Princeton University, Stanford University, Columbia University, and the University of Chicago. The rankings have remained relatively stable, and policymakers and think tank staff value academic Ph.D. programs in Washington, D.C., and outside the United States more than U.S.-based scholars do. Abstract

BackgroundFrontotemporal dementia (FTD) is a complex disorder characterised by a broad range of clinical manifestations, differential pathological signatures, and genetic variability. Mutations in three genes-MAPT, GRN, and C9orf72--have been associated with FTD. We sought to identify novel genetic risk loci associated with the disorder.MethodsWe did a two-stage genome-wide association study on clinical FTD, analysing samples from 3526 patients with FTD and 9402 healthy controls. To reduce genetic heterogeneity, all participants were of European ancestry. In the discovery phase (samples from 2154 patients with FTD and 4308 controls), we did separate association analyses for each FTD subtype (behavioural variant FTD, semantic dementia, progressive non-fluent aphasia, and FTD overlapping with motor neuron disease [FTD-MND]), followed by a meta-analysis of the entire dataset. Abstract

As the national war on drugs continues, children and youth are told to "just say no" but are seldom given the information and skills to sort out the mixed signals coming from peers, adults, and the mass media. This guidebook presents examples of three approaches to community "storytelling" projects through which children and youth can explore conflicting information and feelings about the use of drugs and alcohol. These examples are drawn from projects in rural, American Indian, and Alaska Native communities. Participatory research is a process in which the people affected by an issue or problem develop the information to support action. They learn new skills, undertake the necessary research, and come to their own conclusions. Activities for applying participatory research to substance abuse study include brainstorming, researching drug effects, simple surveys, community surveys, and developing a local resource guide. Cultural journalism can contribute to understanding by looking for both the roots and the cure of substance abuse within personal and collective histories. Activities include private reflection, group discussion, oral history interviews, and shaping the story in written form. Experiential writing activities encourage students to express their own experiences and must take place in a safety zone that guards confidentiality and supports the generation of ideas. Abstract

Over the past fifteen years, a narrative has developed that IR scholars have become a "cult of the irrelevant," with declining influence on and engagement with policy debates. Despite these assertions, the evidence for limited policy engagement has been anecdotal. We investigate the extent of policy engagement—the ways in which IR scholars participate in policy-making processes and/or attempt to shape those processes—by surveying IR scholars directly about their engagement activities. We find policy engagement is pervasive among IR scholars. We draw on theories of credit-claiming to motivate expectations about how and when scholars are likely to engage with practitioners. Consistent with our expectations, much of this engagement comes in forms that involve small time commitments and provide opportunities for credit-claiming, such as media appearances and short-form, bylined op-eds and blog posts. However, sizable minorities report engaging in consulting activities not for attribution/publication and writing policy briefs, and a majority of respondents indicate they engaged in these activities several times a year or more. We find only small differences in engagement across gender and rank. Our results demonstrate that, for IR scholars, some form of policy engagement is the norm. [ABSTRACT FROM AUTHOR]

Abstract

Introduction: This study aims to measure frequency and correlates of initial idiopathic psychiatric diagnosis in a cohort of 147 patients with Frontotemporal Dementia (FTD)-spectrum disorders. Methods: Participants were evaluated at the National Institutes of Health in Bethesda, Maryland. Initial participant diagnoses were determined by chart review and patient and informant interviews. Logistic regression was used to assess the relationships between diagnosis and age of symptom onset, gender, education, family history of psychiatric illness, and family history of dementia. Additional exploratory analyses investigated patients' first symptom type. Results: 25% (n=43) of all the patients reviewed were initially misdiagnosed with an idiopathic psychiatric illness, which is less than half the commonly cited 50% rate.3 Depression was the most common misdiagnosis (46.5%). Family history of dementia, family history of mental illness and an exploratory analysis of behavioral first symptoms suggested significant association with a greater likelihood of initial idiopathic psychiatric diagnosis in FTD patients. Discussion: This data confirms patterns of initial idiopathic psychiatric diagnosis in FTD and elucidates potential factors underlying misdiagnosis.
