The methylation- demethylation dynamics of RNA plays major roles in different biological functions, including stress responses, in plants. m6A methylation in RNA is orchestrated by a coordinated function of methyl transferases (writers) and demethylases (Erasers). Genome-wide analysis of genes involved in methylation and demethylation was performed in pigeon pea. Blast search, using Arabidopsis gene sequences, resulted in the identification of two methylation genes (CcMTA70 , CcMTB70), two genes encoding adaptor proteins for methylation (CcFIPA and CcFIPB) and 10 demethylase (ALKBH) genes (CcALKBH1A , CcALKBH1B , CcALKBH1C , CcALKBH2 , CcALKBH8 , CcALKBH8A , CcALKBH8B , CcALKBH9 , CcALKBH10A and CcALKBH10B) in the pigeon pea genome. The identified genes were analyzed through phylogenetic relationship, chromosomal position, gene structure, conserved motif, domain and subcellular location prediction etc. These structural analyses resulted in categorization of MTs and FIPs into one group, i.e. , CcMTA/B and CcFIPA/B , respectively; and ALKBHs into four groups, viz. CcALKBH1/ 2, CcALKBH 8, CcALKBH 9 and CcALKBH 10. Relative expression analysis of the identified genes in various tissues at different developmental stages revealed the highest level of expression in leaf and the least in root. CcMT s and CcFIP s had similar patterns of expression, and CcALKBH10B demonstrated the highest and CcALKBH2 the lowest level of expression in all the tissues analyzed. CcALKBH8 showed the highest induction in expression upon exposure to heat stress, and CcALKBH10B demonstrated the highest level of induction in expression during drought, salt and biotic (Helicoverpa armigera infestation) stresses. Diurnal regulation of expression of the gene encoding the dominant isoform of UDP-Glucose Pyrophosphorylase, StUGPase2, was studied in potato leaves at different developmental stages (pre-tuberization, tuber initiation, and tuber bulking). Across the developmental stages, the StUGPase2 steady-state mRNA level increased gradually from morning (6 AM) to noon (12 PM), which was followed by a transient drop at 2 PM. The mRNA level increased thereafter, and the highest transcript abundance was recorded at 8:00 PM. A sharp decline in StUGPase2 transcript abundance at 2 PM in all the stages studied is believed to be associated with the previous reports of diurnal behaviour of sucrose concentration and light responsiveness of sucrose synthase enzyme. Moreover, in all three stages, a steady state increase of StUGPase2 transcript despite a consistent decrease in irradiance after 12 PM, suggests the presence of more complex transcriptional regulation. Comparatively higher StUGPase2 transcript abundance at the tuberization initiation stage as compared to the pre-and-post-tuberization stages indicates the prominent role of the gene during tuber development. Bioinformatics analysis of the StUGPase2 promoter region predicted the presence of various light-responsive cis-regulatory nucleotide sequences, which are considered to be associated with the light-mediated diurnal change in StUGPase2 gene expression. The population of the world is increasing nearly exponentially over time. To feed this population following the environment conservation protocol, it is essential to enhance the agricultural productivity even in the synchronizing agrarian land use pattern. To enhance the quality and productivity in agriculture sector, introducing the cutting edge technology is need of the hour. From ancient times, traditional approaches like selective breeding, adoption of agronomic management practices and application of indigenous technical knowledge have been used to attain resilience against various abiotic and biotic stresses. However, these traditional approaches are not sufficient to tackle the increasing repercussions of climate change and feed quality food to the expanding population. Therefore, in order to address these issues of climate change, population explosion and malnutrition, biotechnological interventions can be a promising approach. In the past, biotechnology based approaches have given successful products like Herbicide-resistant Soybean, Pusa Basmati 1, Bt Cotton, Bt Brinjal, Flavr-Savr tomato, a therapeutically significant product of Lithospermum erythrorhizon and Panax ginseng. Besides that many more need based products are in pipeline which is under scrutiny of regulatory bodies, policymakers and environmentalists. It is profoundly expected that in the coming day's agricultural biotechnology applications will bring revolutionary changes to existing agricultural scenario. Therefore, in this review, we have summarized the achievement of agricultural biotechnology that is assisting to enhance the agricultural produce to double the income of farmers. However, this much is not enough; hence full utilization of all the sustainable agricultural biotechnological tools must come into the existence that definitely will boost the agricultural productivity.
