This account presents information on all aspects of the biology of Salvia pratensis L. (Meadow Clary) that are relevant to understanding its ecological characteristics and behaviour. The main topics are presented within the standard framework of the Biological Flora of the British Isles: distribution, habitat, communities, responses to biotic factors, responses to environment, structure and physiology, phenology, floral and seed characters, herbivores and disease, history and conservation.Salvia pratensis is an erect, rosette-forming, perennial herb with a broad native distribution covering much of Europe—from the British Isles, Spain and Morocco in the west, across Europe into Asia, as far east as the Urals. In the British Isles, the species is nationally scarce, confined to a few south- to west-facing sites with calcareous soils in Southern England and one site in Wales. It is predominately found in unimproved pasture, hay meadows and grassy verges, but can occur on the fringes of scrub or woodland. Although the species is abundant in central Europe, changes to land management since the mid-20th century have resulted in fragmented and threatened populations in several European countries. It is cultivated as an ornamental, as is S. × sylvestris, the hybrid with S. nemorosa.Populations are typically gynodioecious, having both female (male-sterile) and hermaphrodite individuals at variable proportions. The species has a mixed mating system and is self-compatible via insect pollination, but predominantly outcrosses. Honeybees and bumblebees are abundant pollinators, but a diverse range of bee species and other insect species visit S. pratensis flowers. Inbreeding depression has been documented, presenting a conservation concern for small, fragmented populations.The species is the focus of conservation efforts and has been reintroduced to sites where it had become locally extinct in Britain. To sustain favourable habitat, site management should maintain low soil nutrient levels, and prevent scrub encroachment and the dominance of coarse grasses. The removal of sward by hay cutting or grazing after plants have flowered and set seed is advised, in addition to maintaining a degree of disturbance to provide bare patches of soil for seedling recruitment.

Societal Impact Statement We rely on plants and fungi for most aspects of our lives. Yet plants and fungi are under threat, and we risk losing species before we know their identity, roles, and potential uses. Knowing names, distributions, and threats are first steps toward effective conservation action. Accessible products like field guides and online resources engage society, harnessing collective support for conservation. Here, we review current knowledge of the plants and fungi of the UK and UK Overseas Territories, highlighting gaps to help direct future research efforts toward conserving these vital elements of biodiversity. Summary This review summarizes current knowledge of the status and threats to the plants and fungi of Great Britain and the UK Overseas Territories (UKOTs). Although the body of knowledge is considerable, the distribution of information varies substantially, and we highlight knowledge gaps. The UK vascular flora is the most well studied and we have a relatively clear picture of its 9,001 native and alien taxa. We have seedbanked 72% of the native and archaeophyte angiosperm taxa and 78% of threatened taxa. Knowledge of the UKOTs flora varies across territories and we report a UKOTs flora comprising 4,093 native and alien taxa. We have conserved 27% of the native flora and 51% of the threatened vascular plants in Kew's Millennium Seed Bank, UK. We need a better understanding of the conservation status of plants in the wild, and progress toward completion or updating national red lists varies. Site‐based protection of key plant assemblages is outlined, and progress in identifying Important Plant Areas analyzed. Knowledge of the non‐vascular flora, especially seaweeds remains patchy, particularly in many UKOTs. The biggest gaps overall are in fungi, particularly non‐lichenized fungi. Considerable investment is needed to fill these knowledge gaps and instigate effective conservation strategies.

This study focuses on the frequency of reproductive modes and the role of reproductive mode variation in diversification of the genus Sorbus s.l. and evolutionary success of individual taxa. Our sampling covers all four ploidies known in Sorbus and includes stabilized taxa and recently formed biotypes. We infer reproductive modes of 3660 seeds using the flow cytometric seed screen method; we also calculated ploidy of contributing pollen. Sorbus shows high variability in reproduction pathways (sexuality, apomixis, formation of BIII hybrids, haploid parthenogenesis). Diploids are sexual, whereas polyploids show predominantly pseudogamous apomixis. In half of the polyploids studied, several reproductive modes were present and even intra-individual variation was detected. Diploids, tetraploids and pentaploids are pollinated by conspecific pollen, whereas triploids are largely, but not fully, dependent on the pollen of other species. In pseudogamous seeds, fertilization of the central cell by both sperms is a dominant pathway of endosperm origin. Flow cytometry of pollen did not detect unreduced pollen at any ploidy. The principal mechanisms of diversification in Sorbus are residual sexuality in apomicts, production of triploid taxa from diploid–tetraploid crosses and production of BIII hybrids. Evolutionary success of Sorbus taxa is largely dependent on the breeding system and is determined by self-(in)compatibility and the interplay between stabilizing apomixis and variation-generating sexuality.

DNA barcoding and metabarcoding provide new avenues for investigating biological systems. These techniques require well-curated reference libraries with extensive coverage. Generating an exhaustive national DNA barcode reference library can open up new avenues of research in ecology, evolution and conservation, yet few studies to date have created such a resource. In plant DNA barcoding, herbarium collections provide taxonomically robust material but also pose challenges in lab processing. Here, we present a national DNA barcoding resource covering all of the native flowering plants and conifers of the United Kingdom. This represents 1,482 plant species, with the majority of specimens (81%) sourced from herbaria. Using Sanger sequencing of the plant DNA barcode markers, rbcL, matK, and ITS2, at least one DNA barcode was retrieved from 98% of the UK flora. We sampled from multiple individuals, resulting in a species coverage for rbcL of 96% (4,477 sequences), 90% for matK (3,259 sequences) and 75% for ITS2 (2,585 sequences). Sequence recovery was lower for herbarium material compared to fresh collections, with the age of the specimen having a significant effect on the success of sequence recovery. Species level discrimination was highest with ITS2, however, the ability to successfully retrieve a sequence was lowest for this region. Analyses of the genetic distinctiveness of species across a complete flora showed DNA barcoding to be informative for all but the most taxonomically complex groups. The UK flora DNA barcode reference library provides an important resource for many applications that require plant identification from DNA.

In plants, apomixis results in the production of clonal offspring via seed and can provide reproductive assurance for isolated individuals. However, many apomicts require pollination to develop functional endosperm for successful seed set (pseudogamy) and therefore risk pollination-limitation, particularly in self-incompatible species that require heterospecific pollen. We used microsatellite paternity analysis and hand pollinations to investigate pollen-limitation in Sorbus subcuneata, a threatened endemic tree that co-occurs with its congener, S. admonitor. We confirmed that S. subcuneata is an obligate pseudogamous apomict, but open-pollinated flowers rarely produced seed (flower-to-seed conversion

Atmospheric nitrogen (N) deposition has been identified as a major threat to biodiversity, but field surveys of its effects have rarely focussed on sites which are actively managed to maintain characteristic species. We analysed permanent quadrat data from 106 plots in nature reserves on calcareous grassland sites in the United Kingdom collected during a survey between 1990 and 1993 and compared the data with the results from resurvey of 48 of these plots between 2006 and 2009. N deposition showed no significant spatial association with species richness, species diversity, or the frequency of species adapted to low nutrient conditions in the 1990–1993 dataset. However, temporal analysis showed that N deposition was significantly associated with changes in Shannon diversity and evenness. In plots with high rates of N deposition, there was a decrease in species diversity and evenness, a decline in the frequency of characteristic calcareous grassland species, and a lower number of rare and scarce species. As all sites had active management to maintain a high diversity and characteristic species, our results imply that even focussed management on nature conservation objectives cannot prevent adverse effects of high rates of N deposition. Structural equation modelling was used to compare different causal mechanisms to explain the observed effects. For change in Shannon diversity, direct effects of N deposition were the dominant mechanism and there was an independent effect of change in grazing intensity. In contrast, for change in herb species number, indirect effects on soil acidity, linked to both N and S deposition, were more important than direct effects of N deposition.

Background: Pollen stainability is often used in hybridisation studies as a proxy for true pollen viability. Its value for assessing fertility and ploidy level was assessed in British Sorbus L. Aims: To assess variation in pollen stainability within and between Sorbus taxa, and its relationship to ploidy level. Methods: Stainability of pollen was examined in herbarium specimens using Alexander's Stain. Results: The diploid species (S. aria, S. aucuparia, S. torminalis) have the highest pollen stainability, followed by tetraploids (S. croceocarpa, S. devoniensis, S. eminens, S. intermedia, S. lancastriensis, S. latifolia, S. leptophylla, S. pseudofennica, S. rupicola). The diploid hybrids (S. × thuringiaca, S. × vagensis) have lower stainability than their parents. Triploids (S. arranensis, S. bristoliensis, S. decipiens, S. leyana, S. minima, S. pseudomeinichii, S. whiteana, S. wilmottiana) usually have the lowest stainability. Hieracium cyathis (Ley) W. R. Linton (Asteraceae), Chalice Hawkweed, is a rare species endemic to Britain. It is a distinct member of Hieracium Section Oreadea, characterized by a short acladium and out-curved tips to the phyllaries. Historic records and current field survey indicate it has occurred in two localities in SW England and seven in South Wales. It is still present in all sites except for one Welsh locality, is significantly threatened at Cheddar Gorge, and two Welsh sites have only three plants each. Comparative morphology, cultivation experiments and amplified fragment length polymorphism (AFLP) analysis do not support the reported differences between material from SW England and Wales. AFLP analysis showed that every plant in the field was genetically similar but different, and minor genetic variation was found in one progeny array. It is a triploid (2n = 27) with apomictic seed production, and 26% potentially viable pollen. It qualifies as ‘Near Threatened’ under the 2001 IUCN threat criteria. The parentage of polyploid Sorbus species in the British Isles was investigated using plastid DNA microsatellites. Four hundred and fifty-three samples from 30 taxa were screened using six microsatellite fragments, which gave 28 haplotypes. The haplotypes formed groups clearly related to the ancestral diploids Sorbus aria, Sorbus aucuparia, and Sorbus torminalis. Species in the Sorbus aria group all had Aria haplotypes (with the exception of one English S. aria), species in the Sorbus anglica group had an Aucuparia haplotype, and species in the Sorbus latifolia group had a Torminalis haplotype. Sorbus intermedia had an Aucuparia haplotype. This indicated that the hybridization events that led to the formation of species in the S. anglica and S. latifolia groups usually did so with S. aria s.l. as the pollen-donating (paternal) parent. The polyploids S. anglica, Sorbus bristoliensis, Sorbus croceocarpa, Sorbus decipiens, Sorbus devoniensis, Sorbus hibernica, Sorbus lancastriensis, Sorbus leptophylla, Sorbus leyana, Sorbus minima, Sorbus rupicola, Sorbus subcuneata, Sorbus vexans, Sorbus whiteana, Sorbus wilmottiana, and three unnamed taxa may each be derived from a single maternal lineage. The polyploids Sorbus eminens, Sorbus porrigentiformis, and S. latifolia have multiple maternal lineages. The temporal and geographical variation in the history of botanical recording in the British Isles between 1836 and 1988 has been reconstructed from literature, herbarium, and field records, and related to the number and distribution of botanists. The number of botanists increased steadily to the 1930s and then five-fold after the Second World War, and is a consistent proportion of the local population. The amount of variation in the literature, assessed from the number of pages published, number of publications, and number of floras, has changed from a low rate between 1836 and 1880, to twice that between 1890 and 1940, and increased steadily to 1988. The number of herbarium specimens collected was also low up to the 1870s and then increased three-fold between 1870 and 1914, decreased between the World Wars, increased in the 1950s, and then decreased as conservation concerns came into play. The number of field records, now on computer, has risen dramatically since the 1930s, especially associated with major recording projects. The Botanical Society of the British Isles (BSBI) Monitoring Scheme data show a strong relationship between the number of hours spent recording at a site and the number of species recorded. The production of literature, collection of herbarium specimens, and field records also vary geographically, being concentrated in the areas in which botanists live. General patterns of recording activity are presented from a combination of the data that follow the general trends shown by the sources. There has been an increase in recording activity, with a low level between 1836 and 1870, followed by a large increase, peaking between 1890 and 1910, followed by decreases during the World Wars, with a recovery between, and a sustained increase since 1950. There is a clear trend from a high recording activity in south-east England to low levels in Scotland and, especially, Ireland, high levels being associated with the main population areas. Botanical recording data are often used to assess changes in the frequencies of plant species over time, but are subject to marked variations in recording activity. We compare and evaluate some general methods that can be used to detect changes in species’ frequencies taking into account the recording variations. Models for 15 species that have been studied in detail previously were compared using the numbers of individual records, sites, hectads, or vicecounties at different time scales (year, decade, moving averages, and pre-/post- specific dates), with or without correction for recording variation. The best methods had a correction for the amount of recording over time, summarized records by decade or moving average, and used an extrapolation between first and last records for sites or hectads. Increasing the geographical and temporal scales can decrease the influence of recording variations, but leads to a loss of sensitivity and under-estimates the true extent of change. Teodor Hedlund was a Swedish botanist who published nearly 50 new names or combinations in Sorbus L. (Rosaceae). Lectotypes are designated for Sorbus arranensis Hedlund, S. minima (Ley) Hedlund, S. quercifolia Hedlund and S. subsimilis Hedlund. The lectotypification of S. anglica Hedlund by A. J. Wilmott is rejected and a new lectotype selected. Notes are given on typification of S. rupicola (Syme) Hedlund. "Sorbus globulifera Hedlund ex Riddelsdell" is not validly published and the material it is based on is part of the variation in S. aria (L.) Crantz.

Summary. The taxonomic history, distribution and ecology of Hymenophyllum tunbrigense (L.) Sm. (Hymenophyllaceae) are discussed with special reference to the sandrocks in south-east England, and the species is illustrated.

In order to create high quality databases, each plant that is DNA barcoded needs to have a herbarium voucher that accompanies the rbcL and matK DNA sequences. The quality of the DNA sequences, the primers used, and trace files should also be accessible to users of the data. Multiple individuals should be DNA barcoded for each species in order to check for errors and allow for intraspecific variation. The world's herbaria provide a rich resource of already preserved and identified material and these can be used for DNA barcoding as well as by collecting fresh samples from the wild. These protocols describe the whole DNA barcoding process, from the collection of plant material from the wild or from the herbarium, how to extract and amplify the DNA, and how to check the quality of the data after sequencing.

Asparagus prostratus Dumort(wild asparagus) and A. officinalis L. (cultivated asparagus), often regarded as subspecies or varieties of a single species, have been confused for historical and nomenclatural reasons. A taxonomic review was carried out, and they were found to be distinct species which differ in morphology (characters retained in cultivation), cytology, distribution and ecology, and they are reproductively isolated. The tetraploid A. prostratus is unlikely to be the taxon from which the diploid A. officinalis evolved. Morphological descriptions are presented, and A. prostratus is lectotypified. A. prostratus is a western European endemic of coasts of Belgium, Britain, the Channel Islands, France, Germany, Ireland, Spain and The Netherlands.

The uncommon spontaneous hybrid between Sorbus aucuparia L. and S. intermedia (Ehrh.) Pers (Rosaceae) is described and named as Sorbus×liljeforsii.

We compare the ecological and habitat characteristics of alien and native vascular plants which have recently expanded in range m England, Scotland, the Republic of Ireland and the Netherlands In the great majority of respects, expanding aliens and natives are functionally indistinguishable However, there are a few consistent differences aliens are more likely than natives to be clonal, polycarpic perennials with erect, leafy stems, and to have transient seed banks We discuss these trends m the context of the difficulties faced by aliens in invading mostly closed plant communities in relatively cool, damp climates Our results are consistent with some predictions of the attributes of‘ideal’invaders, but contradict others We argue that the ecological attributes of successful alien invaders are strongly habitat-dependent

We present the first national DNA barcode resource that covers the native flowering plants and conifers for the nation of Wales (1143 species). Using the plant DNA barcode markers rbcL and matK, we have assembled 97.7% coverage for rbcL, 90.2% for matK, and a dual-locus barcode for 89.7% of the native Welsh flora. We have sampled multiple individuals for each species, resulting in 3304 rbcL and 2419 matK sequences. The majority of our samples (85%) are from DNA extracted from herbarium specimens. Recoverability of DNA barcodes is lower using herbarium specimens, compared to freshly collected material, mostly due to lower amplification success, but this is balanced by the increased efficiency of sampling species that have already been collected, identified, and verified by taxonomic experts. The effectiveness of the DNA barcodes for identification (level of discrimination) is assessed using four approaches: the presence of a barcode gap (using pairwise and multiple alignments), formation of monophyletic groups using Neighbour-Joining trees, and sequence similarity in BLASTn searches. These approaches yield similar results, providing relative discrimination levels of 69.4 to 74.9% of all species and 98.6 to 99.8% of genera using both markers. Species discrimination can be further improved using spatially explicit sampling. Mean species discrimination using barcode gap analysis (with a multiple alignment) is 81.6% within 10×10 km squares and 93.3% for 2×2 km squares. Our database of DNA barcodes for Welsh native flowering plants and conifers represents the most complete coverage of any national flora, and offers a valuable platform for a wide range of applications that require accurate species identification.

Sagina boydii F. B. White (Caryophyllaceae), Boyd's pearlwort, is an enigmatic taxon of unknown geographical origin which is only known in cultivation and is of uncertain taxonomic status. Morphological, cytological and geographical evidence suggests that it is most similar to S. procumbens L. and S. saginoides (L.) H. Karst., but it differs in its compact growth form. Genetically, AFLP analysis shows that it can be included as part of the variation in S. procumbens. In cultivation, it will grow true from seed. Other compact growth forms occur in S. procumbens but do not grow true from seed. As S. boydii is part of the variation in S. procumbens, is of uncertain geographical origin, is known only in cultivation, and grows true from seed, it is best treated as a new cultivar, Sagina procumbens‘Boydii’.
