An important task of natural resource management is deciding amongst alternative policy options, including how interventions will affect the dynamics of resource exploitation. Yet predicting the behaviour of natural resource users in complex, changeable systems presents a significant challenge for managers. Scenario planning, which involves thinking creatively about how a socio-ecological system might develop under a set of possible futures, was used to explore uncertainties in the future of the Indian Ocean tuna purse seine fishery. This exercise stimulated thinking on how key social, economic and environmental conditions that influence fleet behaviour may change in the future, and how these changes might affect the dynamics of fishing effort. Three storylines were explored: an increase in marine protection, growing consumer preference for sustainable seafood, and depletion of tuna stocks. Comparing across several possible future scenarios, a number of critical aspects of fleet behaviour were identified that should be important considerations for fishery managers, but which are currently poorly understood. These included a switch in fishing practices, reallocation of effort in space, investment in new vessels and exit from the fishery. Recommendations for future management interventions in the Indian Ocean were offered, along with suggestions for research needed to reduce management uncertainty.

The use of drifting fish aggregating devices (FADs) has become the dominant practice in tropical tuna purse seine fishing. However, just as FADs can increase fishing efficiency, their use has been associated with several negative ecosystem impacts, and moves are being made to manage the use of FADs. In the evaluation of potential management options it is important to consider how fishers will respond to the introduction of control measures, which first requires an understanding of fishery and fleet dynamics. This paper addresses this need by characterising the past and present use of FADs in the Indian Ocean tropical tuna purse seine fishery. The paper describes historical trends in fishing practices, summarises spatiotemporal patterns in the use of FADs and establishes and attributes variation in FAD fishing strategies within the fleet. It also provides an overview of current FAD management policies in the Indian Ocean and examines the observed effects of existing measures on the behaviour of the purse seine fleet. Using this comprehensive understanding, the potential impact on the purse seine fleet of a number of plausible FAD management options are discussed and inferences are drawn for the future sustainability of tropical tuna purse seine fishing in the Indian Ocean.

Spatial closures are widely used in marine conservation and fisheries management and it is important to understand their contribution to achieving management objectives. Many previous evaluations of closed area effects have used before-after comparisons, which, without controlling for a full range of factors, cannot ascribe changes in fleet behaviour to area closures per se. In this study we used a counterfactual approach to disentangle the effect of two closed areas on fishing location from other competing effects on the behaviour of the Indian Ocean tuna purse seine fishery. Our results revealed an inconsistent effect of the one of the closed areas between years, after taking into account the influence of environmental conditions on fleet behaviour. This suggests that the policy of closing the area per se was not the main driver for the fleet allocating its effort elsewhere. We also showed a marked difference in effect between the two closed areas resulting from their different locations in the fishery area. These findings highlight the need to account for other key fleet behavioural drivers when predicting or evaluating the contribution of area closures to achieving conservation and fishery management objectives.

Plasmonic nanoparticles influence the absorption and emission processes of nearby emitters due to local enhancements of the illuminating radiation and the photonic density of states. Here, we use the plasmon resonance of metal nanoparticles in order to enhance the stimulated depletion of excited molecules for super-resolved nanoscopy. We demonstrate stimulated emission depletion (STED) nanoscopy with gold nanorods with a long axis of only 26 nm and a width of 8 nm. These particles provide an enhancement of up to 50% of the resolution compared to fluorescent-only probes without plasmonic components irradiated with the same depletion power. The nanoparticle-assisted STED probes reported here represent a ∼2 × 103 reduction in probe volume compared to previously used nanoparticles. Finally, we demonstrate their application toward plasmon-assisted STED cellular imaging at low-depletion powers, and we also discuss their current limitations.

Glioblastomas (GBM) are aggressive and therapy-resistant brain tumours, which contain a subpopulation of tumour-propagating glioblastoma stem-like cells (GSC) thought to drive progression and recurrence. Diffuse invasion of the brain parenchyma, including along preexisting blood vessels, is a leading cause of therapeutic resistance, but the mechanisms remain unclear. Here, we show that ephrin-B2 mediates GSC perivascular invasion. Intravital imaging, coupled with mechanistic studies in murine GBM models and patient-derived GSC, revealed that endothelial ephrin-B2 compartmentalises non-tumourigenic cells. In contrast, upregulation of the same ephrin-B2 ligand in GSC enabled perivascular migration through homotypic forward signalling. Surprisingly, ephrin-B2 reverse signalling also promoted tumourigenesis cell-autonomously, by mediating anchorage-independent cytokinesis via RhoA. In human GSC-derived orthotopic xenografts, EFNB2 knock-down blocked tumour initiation and treatment of established tumours with ephrin-B2-blocking antibodies suppressed progression. Thus, our results indicate that targeting ephrin-B2 may be an effective strategy for the simultaneous inhibition of invasion and proliferation in GBM. On average, patients with glioblastoma only survive 15 months even with aggressive treatment. One of the main reasons that therapy fails is the strong tendency of the tumor cells in this form of cancer to spread into the normal brain tissue. This makes it impossible for surgeons to completely remove the tumor, which means that the disease will almost always recur. Within the brain, invading glioblastoma tumor cells spread along pre-existing structures, like blood vessels. When the tumors use blood vessels as a highway to the rest of the brain it is called “perivascular invasion”. Scientists do not know exactly how glioblastoma cells move along the blood vessels. Learning more about this type of tumor cell movement could help scientists develop treatments to stop the tumor cells from spreading. Now, Krusche et al. show that the glioblastoma tumor cells highjack the system that normally helps keep brain cells in place. Experiments with mouse and human tumor cells grown in the laboratory and injected in mice to produce glioblastoma tumors showed that a family of proteins called ephrins determines whether perivascular invasion occurs. Ephrins are found on the surface of both tumor cells and blood vessels. Normally, the blood vessels use these proteins to block the spread of normal brain cells. However, tumor cells override this normal anti-tumor mechanism and become able to spread along the blood vessels. Specifically, Krusche et al. showed that an increase in the levels of ephrin-B2 in tumor cells allows them to move along the blood vessels. Ephrin-B2 was also found to drive the multiplication of the tumor cells independently of the protein’s interactions with the blood vessels. Using antibodies to block ephrin-B2 in tumors greatly reduced tumor size and extended survival in mice with glioblastoma tumors. The experiments suggest the blocking ephrin-B2 might be a therapy that both stops the glioblastoma cells from spreading and prevents the tumor cells from multiplying. Background The seasonal aggregations of whale sharks that occur at coastal sites throughout the tropics are the focus of growing ecotourism industries. Genetic and modelling studies suggest that these aggregations may be linked by migration, although the temporal and spatial scales at which this occurs is unknown. Here, we utilized a continuously expanding photoidentification database collected by citizen scientists and researchers to assess connectivity and residency patterns of five whale shark aggregation sites across the entire Indian Ocean at timescales of up to a decade. We also investigated the effectiveness of such an approach in detecting different levels of migration given the population size at each aggregation site. Approach We used the semi-automated program I3S (Individual Interactive Identification System) to compare 6,519 photographs of the unique natural marking patterns of individual whale sharks collected from aggregations at Mozambique, the Seychelles, the Maldives, Christmas Island (Australia) and Ningaloo Reef (Australia). We searched the database for matches between aggregations (migration) and matches between years within aggregations (residency). A Monte Carlo simulation approach utilizing population sizes of aggregations reported by previous studies was used to examine the sample sizes that would be required to reliably detect varying levels of migration rates among four of these aggregations. Results We found no evidence of connectivity of whale shark aggregations at ocean-basin scales within the time frame of the study, and evidence for only limited connectivity at regional (100s–1000s km) scales. A male whale shark sampled in January 2010 at Mozambique was resighted eight months later in the Seychelles and was the only one of 1,724 individuals to be photographed at more than one site. On average, 35% of individuals were re-sighted at the same site in more than one year. The Monte Carlo simulation study showed that the power of this photo-identification approach was strongly dependent on both the number of individuals identified in aggregations each year and the size of resident populations. The simulation also indicated that there was a higher chance of detecting migrants (if they were present) in aggregations at Ningaloo Reef and the Maldives than the Seychelles and Mozambique given the current numbers of individuals identified each year. Conclusions The weight of evidence from both our photo-identification study and previous tagging studies suggests regional or small-scale movements (10–100s km) may be more common in whale sharks than movement at ocean-basin scales. We recommend that the management of whale sharks in the Indian Ocean occur at regional scales, and photo-identification databases are expanded to include additional sampling sites within each region.

The News Focus story "Giant marine reserves pose vast challenges" (C. Pala, 8 February, p. [640][1]) discusses the potential role of large marine reserves in conserving pelagic species. We highlight three points that are misrepresented in the article. First, the story describes tropical tuna stocks

Context The conservation status of the whale shark, Rhincodon typus, remains uncertain. Throughout their range, whale sharks are a focus for wildlife tourism and many identification photographs taken by tourists have become available online, presenting a potentially valuable source of data for monitoring populations using mark–recapture techniques. However, the suitability of these photographs for mark–recapture models has yet to be investigated. Aims We explore the suitability of identification-photographs available from online databases to produce assessments of life-history parameters and conservation status of whale sharks in the Maldives. Methods To test the validity of using publically sourced images, we used photo-identification images collected from both experienced researchers and tourists between 2003 and 2008 to construct two databases. Images taken by tourists were compiled from online databases. Researcher and public databases were analysed separately and the results of mark–recapture models then compared. Key results The dataset constructed from online public databases did not violate the assumptions of mark–recapture modelling. Estimates of parameters and abundance obtained from models using these data were similar to those produced using data provided by experienced researchers. Conclusions Publically sourced data allowed for the accurate estimation of abundance of whale sharks. These estimates were not confounded by the suitability of photographs, probably because of the high encounter rate in the aggregation, the high residency rate of sharks and the retrospective nature of photo-identification, which limited heterogeneity in capture probability between marked and unmarked sharks. Implications Our findings support the use of publically sourced data for use in mark–recapture studies of whale sharks, at least in situations where sharks are resident to the location. This approach will be useful in regions where data collected by tourists are available online, and research funding is limited.
