2. On the Representation of Partially Specified Implementations and its Application to the Optimization of Linear Algebra Kernels on GPU

Abstract

Traditional optimizing compilers rely on rewrite rules to iteratively apply program transformations. This iterative approach hides optimization opportunities behind intermediate transformation steps. For instance, vectorization can only be applied to the innermost loop in a nest: one must first perform a loop interchange before even considering vectorization of an outer loop. In contrast, we propose an implementation framework representing programs as sets of possible implementation decisions. Specifying one decision can have an impact on others in a bidirectional manner: specifying that a loop must be vectorized prevents other loops from being nested inside it; conversely, specifying a loop as an outer loop will prevent it from being vectorized. These optimization decisions commute, obviating the pass ordering problem. We present a constraint programming system to formally define, represent and explore such implementation spaces. We also propose an exploration strategy combining tree search and branch-and-bound; the strength and novelty of this strategy reside in an analytical model of the lower bound on the execution time of a set of possible implementations. We showcase our approach on the construction and exploration of an implementation space for linear algebra kernels running on GPUs. We show this search space is expressive enough to represent complex decisions that fundamentally change the structure of the generated code. We also present preliminary results competitive with the performance of native GPU libraries.

12. Do KM and TQM have an impact on employee effectiveness and supply chain performance?

Abstract

Purpose: Companies should enhance their market position and competitiveness by improving staff effectiveness, skills, resource commitment, and applying relevant managerial methods. This study aims to examine the impact of knowledge management (KM) and total quality management (TQM) on employee effectiveness (EE) and supply chain performance (SCP) in emerging economies. Design/methodology/approach: The used methodology consists on conducting a survey within Tunisian companies, where the authors gathered 206 responses. Collected data was analyzed using statistical package for the social sciences (SPSS) software, enabling the authors to establish a conceptual model. This model was further examined through structural equation modeling, using analysis of moment structures (AMOS) software for hypothesis validation. Additionally, the authors’ research aimed to enhance SCP and boost EE while minimizing costs through a nonlinear mathematical model and the quality function deployment method. Findings: The results indicate that TQM and KM positively impact EE, and KM and EE positively impact SCP. However, the significance of employee performance on SCP varies depending on company location and industry sector studied. Originality/value: This work emphasized the involvement of small- and medium-sized enterprise managers from emerging economies in the studied concepts and confirmed the effects of KM and TQM practices on EE and SCP.

Les patients en insuffisance rénale terminale hémodialysés avec pour voie d’abord une fistule artérioveineuse (FAV) sont exposés au risque de sténoses au niveau de la veine de drainage de la FAV, ce qui peut conduire à une thrombose de cette dernière. Cependant, les critères échographiques utilisés pour évaluer ces sténoses sont nombreux, et les seuils à utiliser varient largement dans la littérature. L’objectif est d’évaluer la performance de plusieurs critères écho-Doppler indirects pour la détection de sténoses des veines de drainage, définies comme sévères ou significatives.

Abstract

AbstractSupply Chain Performance (SCP) is a critical issue in most industries. Organizations expect to enhance their SCP because it allows better competitiveness through minimizing useless resources activities, unsatisfactory delivery times, noncompliant quality, overproduction, etc. In this context, this paper provides an overview of the methods in use in Supply Chain Performance Measurement (SCPM) and investigates the most used methods for data collection, approaches, and data analysis techniques in order to measure SCP. This study was based on 180 of 682 related articles, published from 2008 to 2019, which were collected from the Crossref and Sciencedirect databases. Articles were reviewed in order to update methods and techniques which are the most used to define and analyze this performance, both in academic research or industrial case studies. Findings of this work can be very helpful to researchers and practitioners to know similitude and difference of each method, approach and tool exploited in the field of SCPM, in their particular area of activity.

41. Blood‐based biomarkers for Alzheimer's Disease in Down Syndrome.

Abstract

Background: People with DS are at high risk to develop Alzheimer's disease dementia (AD) (Lott & Head, 2001). Predicting and monitoring decline and onset of dementia is a diagnostic challenge and of essence in daily care and support for people with DS. Behavioral and Psychological Symptoms of Dementia (BPSD) are an easily accessible tool for the prediction of dementia onset in the DS population. The BPSD‐DS II scale was developed and validated by our team to identify behavioral changes linked to AD between the last six months and pre‐existing behavior in an individual with DS (Dekker et al., 2021). This project is based on a cohort of DS individuals without AD (n = 94), with AD (n = 43), or with questionable AD (qAD) (n = 54) aged >35 years. At inclusion, the BPSD‐DS II was applied to assess AD‐linked behavioral changes, and biofluid sampling was performed as well. Changes in cerebrospinal fluid (CSF) levels of monoaminergic neurotransmitters and metabolites have been associated with BPSD in AD (Dekker et al., 2015). However, CSF sampling procedures remain difficult in DS, illustrating the need for blood biomarkers. BPSD‐DS II results will be correlated with the serum levels of monoaminergic and classical AD biomarkers. Three years post‐inclusion, a follow‐up study was conducted to assess the evolution of possible AD dementia within this group and to determine the predictive values of the BPSD‐DS II scale and the different analytes. Method: We will use reversed‐phase ultra‐high performance liquid chromatography with electrochemical detection to determine the levels of (nor)epinephrine, dopamine, serotonin and their metabolic products in serum. We will use the Simoaâ platform on the serum of clients with DS without or with (q)AD to detect the levels of Ab40, Ab42, total tau, phosphorylated tau, Glial Fibrillary Acidic Protein and Neurofilament Light chain. Result: We expect a correlation between the results of the BPSD‐DS II and the levels of the monoamines and classical biomarkers in patients with DS without or with (q)AD. Conclusion: The diagnosis predicted by the results of the BPSD‐DS II correlates with the levels of monoamines and classical biomarkers in individuals with DS without or with (q)AD. Abstract

Here, Caron et al. show that the HECT E3 ubiquitin ligase WWP2 associates with components of the DNA-PK and RNAPII complexes and is recruited to DSBs at RNAPII transcribed genes. Their findings suggest that WWP2 operates in a DNA-PK-dependent shutoff circuitry for RNAPII clearance that promotes DSB repair by protecting the NHEJ machinery from collision with the transcription machinery.

47. Large-Scale Phosphoproteomics Reveals Shp-2 Phosphatase-Dependent Regulators of Pdgf Receptor Signaling

Abstract

Despite its low cellular abundance, phosphotyrosine (pTyr) regulates numerous cell signaling pathways in health and disease. We applied comprehensive phosphoproteomics to unravel differential regulators of receptor tyrosine kinase (RTK)-initiated signaling networks upon activation by Pdgf-ββ, Fgf-2, or Igf-1 and identified more than 40,000 phosphorylation sites, including many phosphotyrosine sites without additional enrichment. The analysis revealed RTK-specific regulation of hundreds of pTyr sites on key signaling molecules. We found the tyrosine phosphatase Shp-2 to be the master regulator of Pdgfr pTyr signaling. Application of a recently introduced allosteric Shp-2 inhibitor revealed global regulation of the Pdgf-dependent tyrosine phosphoproteome, which significantly impaired cell migration. In addition, we present a list of hundreds of Shp-2-dependent targets and putative substrates, including Rasa1 and Cortactin with increased pTyr and Gab1 and Erk1/2 with decreased pTyr. Our study demonstrates that large-scale quantitative phosphoproteomics can precisely dissect tightly regulated kinase-phosphatase signaling networks.

48. Genome-wide identification and comparative analysis of NBS-LRR resistance genes in Brassica napus

Abstract

Plant disease-resistance genes play a critical role in providing resistance against pathogens. The largest family of resistance genes are the nucleotide-binding site (NBS) and leucine-rich repeat (LRR) genes. They are classified into two major subfamilies, toll/interleukin-1 receptor (TIR)-NBS-LRR (TNL) and coiled-coil (CC)-NBS-LRR (CNL) proteins. We have identified and characterised 641 NBS-LRR genes in Brassica napus, 249 in B. rapa and 443 in B. oleracea. A ratio of 1:2 of CNL:TNL genes was found in the three species. Domain structure analysis revealed that 57% of the NBS-LRR genes are typical resistance genes and contain all three domains (TIR/CC, NBS, LRR), whereas the remaining genes are partially deleted or truncated. Of the NBS-LRR genes, 59% were found to be physically clustered, and individual genes involved in clusters were more polymorphic than those not clustered. Of the NBS-LRR genes in B. napus, 50% were identified as duplicates, reflecting a high level of genomic duplication and rearrangement. Comparative analysis between B. napus and its progenitor species indicated that >60% of NBS-LRR genes are conserved in B. napus. This study provides a valuable resource for the identification and characterisation of candidate NBS-LRR genes.

Abstract

Twelve patients underwent cardiac surgical procedures after having previously received irradiation of the mediastinum for various types of malignancies (Hodgkin's disease, carcinoma of the breast and seminoma). The patients' ages ranged from 39 to 69 years (mean 57 years); nine patients were female and three male. The average length of time from radiation to surgery was 15 years (3-24 years interval). The patients were divided into two groups according to the type of heart disease: Group I included seven patients who developed coronary artery disease (CAD) despite minimal risk factors. Three asymptomatic patients developed sudden myocardial infarction. In two patients the myocardial infarction was complicated (with ventricular septal defect in one and ventricular septal defect plus ventricular aneurysm in the other). All seven patients were surgically managed. Internal thoracic artery (ITA) was used in three patients as a conduit of myocardial revascularization. There were two hospital deaths in this group. The five patients in Group II underwent aortic valve replacement for aortic stenoses. Prior to the irradiation, none of the patients had a documented history of rheumatic fever, bacterial endocartitis, significant murmur, etc. There were no deaths in this group. Intraoperative findings for both groups included thickening and/or fibrosis of the ascending aortic wall, coronary arteries and aortic valve tissue. Epicardium was whitish colored and thickened.

50. Tralokinumab Effectively Disrupts the IL-13/IL-13Rα1/IL-4Rα Signaling Complex but Not the IL-13/IL-13Rα2 Complex

Abstract

Tralokinumab, a fully human mAb specifically targeting the IL-13 cytokine, has demonstrated clinical efficacy and safety in patients with moderate-to-severe atopic dermatitis. Tralokinumab binds IL-13 with high affinity, which prevents the interaction of IL-13 with IL-13Rα1 and subsequent signaling. Similarly, tralokinumab-bound IL-13 cannot bind to IL-13Rα2, a proposed decoy receptor that is reported to bind IL-13 with extraordinarily high affinity. It has however not been fully elucidated to what extent tralokinumab interferes with the endogenous regulation of IL-13 through IL-13Rα2. In this mechanistic study, we used biophysical, biochemical, and cellular assays to investigate the effect of tralokinumab on the interaction between IL-13 and IL-13Rα1 and IL-13Rα2, respectively, as well as the effects on IL-13Rα2–mediated IL-13 internalization. We demonstrate that IL-13Rα2 binds IL-13 with exceptionally high affinity and that tralokinumab is unable to displace IL-13 from IL-13Rα2. In contrast to this, tralokinumab is able to disrupt the IL-13/IL-13Rα1 and IL-13Rα1/IL-13/IL-4Rα complex. Furthermore, we demonstrate that whereas the IL-13/tralokinumab complex is unable to bind IL-13Rα2, any IL-13 that is not bound by tralokinumab (i.e., free IL-13) can be bound by IL-13Rα2 and subsequently internalized, regardless of the presence of tralokinumab. In summary, our study indicates that tralokinumab does not interfere with endogenous IL-13Rα2–mediated regulation of free IL-13.
