[ES] La berenjena (Solanum melongena L.) es uno de los cultivos comerciales de hortalizas solanáceas más importantes que se cultiva ampliamente en Asia y la región del Mediterráneo. A pesar de su importancia económica, la disponibilidad de poblaciones experimentales y herramientas genómicas para el mejoramiento es aún muy limitada en comparación con otros cultivos importantes. Debido a la alteración progresiva del ecosistema global por el cambio climático, las plantas están constantemente expuestas a condiciones ambientales estresantes que impactan negativamente en su productividad. El cuello de botella genético ocurrido durante la domesticación de la berenjena, que limita la disponibilidad de recursos genéticos para su mejoramiento genético, hace que este cultivo sea extremadamente vulnerable al cambio climático, por lo que se requieren nuevas estrategias para reducir su erosión genética. En este contexto, los parientes silvestres de los cultivos (CWRs) han demostrado ser un recurso genético válido para la mejora vegetal, ya que su uso permite ampliar la diversidad genética de los cultivos y, en paralelo, desarrollar variedades mejoradas adaptadas al cambio climático. Para lograr este objetivo, en esta tesis doctoral informamos sobre el desarrollo y la evaluación de materiales avanzados de berenjena obtenidos mediante el uso de parientes silvestres. En el primer capítulo, realizamos una evaluación fenotípica en dos ambientes de un conjunto de 16 ILs de berenjena con introgresión de S. incanum, un pariente silvestre. Se evaluaron diecisiete caracteres agronómicos para comparar el rendimiento de las ILs con el parental recurrente e identificar QTLs para los caracteres investigados. Encontramos diferencias morfológicas significativas entre los parentales, y el híbrido resultó heterótico para los caracteres de vigor. A pesar de que la interacción entre genotipo y ambiente (G x E) resultó significativa para la mayoría de los caracteres, en general las ILs mostraron pocas diferencias fenotípicas con el progenitor receptor, incluso en presencia de grandes fragmentos de introgresión del progenitor silvestre. Se encontraron valores de heredabilidad bajos a moderados para los caracteres agronómicos. En total, detectamos diez QTLs estables, dos de los cuales estaban relacionados con caracteres de planta y cuatro para caracteres de flor y fruto. En general, las introgresiones de S. incanum mejoraron los valores medios de la mayoría de los caracteres de planta y flor, y disminuyeron el de los caracteres de fruto. Para tres QTLs relacionados con la longitud del pedicelo del fruto y con el peso del fruto, encontramos evidencia de sintenia con otros QTLs identificados previamente en poblaciones de berenjena. Siete QTLs eran nuevos, de los cuales cuatro relacionados con la altura de la planta, con la espinosidad del cáliz de la flor y con la longitud del pedicelo del fruto no colocalizaron con ningún QTL previamente identificado en las poblaciones de berenjena, y tres relacionados con el diámetro del tallo, con la longitud del pedúnculo y del estigma, fueron los primeros identificados en berenjena para estos caracteres. En el segundo capítulo, el conjunto de IL de berenjena con introgresiones de S. incanum se evaluó para la forma del fruto en dos ambientes. Específicamente, realizamos un fenotipado detallado de los frutos de los parentales, del híbrido y de las ILs utilizando 32 descriptores morfológicos de la herramienta fenómica Tomato Analyzer. Se encontraron grandes diferencias morfológicas en los frutos de los parentales, y el híbrido presentó valores negativos de heterosis para muchos de los caracteres de forma del fruto, siendo fenotípicamente más cercano al parental S. incanum. Para la mayoría de los descriptores de forma del fruto observamos diferencias significativas entre las ILs y el parental receptor, incluso en presencia de pequeños fragmentos de introgresión del parental silvestre. En el segon capítol, el conjunt de IL d'albergínia amb introgresions de S. incanum es va avaluar per a la forma del fruit en dos ambients. Despite its economic importance, the availability of experimental populations and genomic tools for breeding is still very limited compared to other major crops. Due to the progressive alteration of global ecosystem by climate change, plants are constantly exposed to stressful environmental conditions that impact negatively on their productivity. The genetic bottleneck occurred during eggplant domestication, which limits the availability of genetic resources for its genetic improvement, makes this crop extremely vulnerable to climate change, and, therefore, new strategies are needed for reducing its genetic erosion. In this context, crop wild relatives (CWRs) have demonstrated to be a valid genetic resources for plant breeding, as their use allows to broaden the genetic diversity of the crop and, in parallel, develop improved varieties adapted to climate change. To achieve this objective, in this doctoral thesis we reported on the development and evaluation of eggplant advanced materials obtained by using crop wild relatives. In the first chapter, we have conducted a phenotypic evaluation in two environments of a set of 16 eggplant ILs with introgression from S. incanum, a close wild relative. Seventeen agronomic traits were scored to test the performance of ILs compared to the recurrent parent and identify QTLs for the investigated traits. We found significant morphological differences between parents, and the hybrid was heterotic for vigour related traits. Although significant genotype x environment interaction (G x E) was detected for most traits, the ILs generally exhibited few phenotypic differences with recipient parent, even in the presence of large introgression fragments from the wild parent. Low to moderate heritability values were found for the agronomic traits. In total, we detected ten stable QTLs, two of which were for plant-related traits and four for both flower- and fruit-related traits. In general, S. incanum introgressions improved the performance of most plant- and flower-related traits and decreased that of fruit-related traits. For three QTLs related to fruit pedicel length and fruit weight, we found evidence of synteny to other QTLs previously reported in eggplant populations. Seven QTLs were new, of which four related to plant height, flower calyx prickles, and fruit pedicel length, did not colocalized with any previous identified QTLs in eggplant populations, and three related to stem diameter, peduncle length, and stigma length, were the first reported in eggplant for these traits. In the second chapter, the set of eggplant ILs with introgression from S. incanum was evaluated for fruit shape in two environments. Specifically, we performed a detailed phenotyping of the fruits of the parents, hybrid, and ILs using 32 morphological descriptors of the phenomics tool Tomato Analyzer. Large differences in fruit morphology were found between ILs parents, and the hybrid exhibited negative values of heterosis for many fruit shape traits, being phenotypically closer to S. incanum parent. For most fruit shape descriptors, we observed significant differences between ILs and recipient parent, even in the presence of small wild donor fragments. Although the contribution of the environment and G × E interaction were significant for almost all descriptors, we found that their effects on fruit shape were relatively low, and the observed variations in fruit shape was mainly genetically regulated. Hierarchical clustering revealed nine clusters of highly correlated traits and six ILs groups. A total of 41 QTLs were mapped. [EN] Eggplant fruit shape is an important quantitative agronomic trait. The use of introgression lines (ILs) for QTLs identification is a powerful tool for the elucidation of the genetic control of eggplant fruit shape. In the present study, a set of 16 eggplant ILs, each harboring a single marker-defined chromosomal segment from the wild eggplant relative S. incanum in the genetic background of S. melongena, was evaluated for fruit shape in two environments (open field and screenhouse). A detailed phenotyping of the fruits of the two parents, hybrid and ILs was performed using 32 morphological descriptors of the phenomics tool Tomato Analyzer. Several morphological differences were found between parents, and the hybrid displayed negative heterosis for many fruit shape traits, being more similar to the S. incanum parent. Significant differences for most fruit shape descriptors were found between ILs and the recipient parent. For many descriptors, the genotype factor had the highest contribution to the percentage of the sum of squares. Although the contributions of the environment and the G x E interaction were significant for almost all descriptors, their effects on fruit shape were relatively low. Hierarchical clustering revealed nine clusters of highly correlated traits and six ILs groups. A total of 41 stable QTLs spread over ten chromosomes were detected. Of these, twenty QTLs associated to Basic Measurement and Fruit Shape Index descriptors were syntenic to other previously reported in several intraspecific and interspecific eggplant populations, while twenty-one QTLs, including Blockiness, Homogeneity, Asymmetry and Internal Eccentricity, were new. In addition, mutations associated to genes belonging to SUN, OVATE and YABBY families described in tomato were reported in the QTLs genomic regions identified in eggplant. Eleven SUN and YABBY genes were proposed as potential candidate controlling fruit shape variations in eggplant. [EN] Spain is a relevant secondary centre of diversity for Capsicum annuum peppers, especially for the bell types known as Pimiento Morron or Pimiento de Morro. Thus, a myriad of highly regarded landraces adapted to a wide range of conditions can be found throughout the country, as a result of centuries of farmers breeding. Despite that, these materials lack of proper characterization, of paramount importance for farmers, breeders and germplasm management. In this regard, in addition to internationally accepted conventional descriptors, high-throughput digital phenotyping tools like Tomato Analyzer, a software originally developed to process scanned images of cut tomato fruits and to record a range of morphological parameters, may provide an important help towards exhaustive germplasm characterization. With this aim, 32 conventional and 35 Tomato Analyzer digital traits were used herein to characterize a large collection of C. annuum accessions from all Spanish regions, including Protected Designations of Origin and Protected Geographical Indications, with emphasis on Marron peppers, in order to assess the diversity within Spanish elite germplasm and to test the efficiency of those methods to differentiate varietal types and closely related materials. A considerable amount of variation was found using both conventional and digital traits, even within Marron pepper groups, reflecting the diversity of Spanish peppers in terms of plant and fruit morphology, essential for future breeding programs. Both conventional descriptors and digital parameters were able to distinguish varietal groups. However, on the whole, digital phenotyping was able to discriminate in a more accurate way. Most digital parameters were able to discriminate varietal groups into higher numbers of categories (>= 4) than conventional traits (usually 2-4). In addition, the number of significant pairwise differences among varietal groups was considerably higher for digital parameters than for conventional descriptors, enabling a powerful separation, particularly relevant for closely related groups such as Morron peppers. Likewise, as revealed by Principal Components Analysis, digital phenotyping allowed a more powerful intra-varietal separation compared to conventional descriptors. Finally, a subset of 4 conventional descriptors and 13 Tomato Analyzer traits were identified as the most discriminant to distinguish among closely related C. annuum accessions, explaining 81.81 % of total variance found by Principal Components Analysis. Bell pepper is an important vegetable crop containing lots of bioactive compounds. The present study was designed to improve the productivity and quality of bell pepper with the application of arbuscular mycorrhizal fungi (Rhizophagus irregularis) and plant growth-promoting bacteria (Azotobacter chroococcum) in a combination of chemical fertilizer. Five treatments consisted of 75% chemical fertilizer (T1), 100% chemical fertilizer (T2), 75% chemical fertilizer + R. irregularis (T3), 75% chemical fertilizer + A. chroococcum (T4) and 75% chemical fertilizer + R. irregularis + A. chroococcum (T5). Out of 18 morphological parameters, 11 morphometric fruit parameters were recorded in detail by a tomato analyzer. The morphological and biochemical (TSS, ascorbic acid and capsaicin content) attributes of bell pepper were recorded higher in the case of a mixed consortium of chemical fertilizers having R. irregularis and A. chroococcum. Similarly, the amount of mineral content recorded was highest after 75% chemical fertilizer + R. irregularis + A. chroococcum, followed by the treatment with only 100% chemical fertilizer. The root mycorrhization (%) and the number of spores were observed highest in 75% chemical fertilizer + R. irregularis + A. chroococcum, and there was no mycorrhization and spore formation in 75% CF, 100% CF and 75% CF+AC. The treatment involving 75% chemical fertilizer + R. irregularis + A. chroococcum proved better for pepper’s growth, yield and yield-related traits.

[Abstract] The main goal of this work was to investigate the relationship between the fruit morphology and biochemical composition of peppers (Capsicum spp.). For that purpose, one hundred native varieties from the Andean region, where the genus Capsicum has its origin, were analysed for different phytochemical compounds. In addition, pepper fruits were assessed with the highly precise phenomics tool Tomato Analyzer. The collection showed a broad variability which was more evident within the C. annuum group. On average, C. frutescens accessions displayed the highest levels of solid soluble content, pH, polyphenols and antioxidant activity. The Tomato Analyzer descriptors under the categories of size, shape index, and latitudinal section, mostly contributed to the variance among Capsicum groups. C. annuum hold the larger fruits, whereas C. frutescens comprised fruits of smaller sizes. The correlation analysis revealed that biochemical traits were negatively correlated with the fruit parameters related to size, suggesting that huger fruits contain lower amounts of chemical metabolites. The multivariate approximations demonstrated that Andean peppers assorted according to morphometric and colorimetric characteristics, but independently of their species or geographical origin. Groups of valuable native varieties carrying promising traits were identified. Anther or microspore culture induced haploid and double haploids (DH) are increasingly being utilized by breeders since it can shorten breeding time by achieving complete homozygosity within a single generation. Pepper (Capsicum annuum L.) is one of the most important vegetables, distinguished by its high level of heterozygosity, making the breeding process very laborious and long. Fourteen different DH lines were obtained as a result of anther culture of four parental genotypes. Data from different plant and fruit morphological traits as well as quality traits including vitamin C, dry matter content, total polyphenols and antioxidant activity were collected. A total of 47 different descriptors for fruit morphology and color were characterized using Tomato Analyzer v. 3 software. Findings from this research revealed significant variation of fruit morphology, quality and productivity traits between DH lines and their respective parental genotypes. Among these studied 14 DH lines, 42.9% were superior to the parental genotypes for fruit weight, width, fruit wall thickness, and usable part of the fruits. As compared to parental genotypes, DH lines exhibited higher values for tested fruit quality traits and dry matter content. Multivariate analysis allowed us to identify the trait combination that contributed to the total variation. A total of eight principle components (PCs) explained 95.4% variation with PC1 and PC2 contributing 32.4% and 21.1%, respectively. Pepper androgenesis clearly indicates its usefulness as a well established technique that can allow pepper breeders to save the time and breeding resources by expediting the breeding process. Our research findings prove the advantages of pepper androgenesis to utilize the diversity of pepper genetic resources and development of novel pepper breeding lines to utilize in future breeding.

[EN] An essential step in many plant breeding programmes is the morphoagronomic phenotyping of the materials that are being developed, using standardized descriptors or, when not available, as it is the case of new crops, consensus guidelines to characterize them. Although many of the traits must be evaluated in the field, those related to the shape and size of leaves and fruits can be easily measured with specific software using digital pictures. This tool provides several advantages, including: 1) more accurate and objective measurements; 2) the possibility of measuring other traits of interest such as area or perimeter, that in other case would not be easily evaluated; or even 3) the possibility of delaying the analysis of pictures in order to focus on field traits. Therefore, the knowledge and management of this tool can become of high usefulness. The Plant Breeding Master offered by our institute is aimed at training professionals able to improve crops and develop new ones according to market trends, in addition to improve adaptation and resistance to biotic and abiotic stresses. Due to the importance of phenotyping considering any of the above breeding goals, in this paper we propose the introduction of a practical session in the mandatory subject "Instrumental Techniques": the management and comparison of adequacy of two specific software tools considering the material to be analyzed. As material, we propose a collection of pepper varieties for analysis of fruits, and a collection of rocket (Eruca and Diplotaxis spp.) germplasm for analysis of leaves, due to the broad variability observed in those materials. The MorphoLeaf v1.0 software will be used for the analysis of leaves, and the Tomato Analyzer 3.0 software will be used for the analysis of fruits and also for leaves. According to the design of the practical lesson, only one session of three hours is required, in which students will learn to properly digitalize materials considering the restrictions of each software and material used, and to obtain all the information needed in each case. In addition, there will be one hour of autonomous work in which both tools and the results obtained will be compared by the students, and a report will be prepared. This practical lesson allows students to acquire the competence for the correct use of different computer tools in the phenotyping task. The comparison of analyses using different scan adjustments and software will allow students to understand the disadvantages of each option and problems that could occur, in order to minimize them. [ES] El trabajo realizado en la presente tesis pretende contribuir a la puesta en valor de las variedades tradicionales valencianas de tomate. En primer lugar se aborda la caracterización de una colección de 69 entradas de variedades valencianas de tomate de ocho tipos varietales ('De Borseta', 'Cherry', 'Cor¿, 'De Penjar', 'Plana', 'De Pruna', 'Redona' y 'Valenciana') con 64 descriptores convencionales y 38 características del Tomato Analyzer. Se ha puesto de manifiesto que el uso de forma complementaria de la herramienta fenómica del Tomato Analyzer es una nueva estrategia para la caracterización y clasificación de las variedades locales de tomate, así como para la distinción entre tipos varietales relacionados. A pesar del uso generalizado de los descriptores estandarizados de tomate, hay poca información sobre los efectos ambientales sobre los valores de los descriptores y sobre su herencia. La evaluación de 12 entradas de tomate procedentes de siete tipos varietales en tres ambientes diferentes (cultivo convencional aire libre, cultivo ecológico aire libre e invernadero) mediante el uso de 36 descriptores, ha permitido establecer la influencia del sistema de cultivo sobre estos descriptores, encontrándose una amplia gama de variación, y demostrando su utilidad para describir los materiales de tomate, su diversidad y relaciones. Por otra parte, puede ser muy interesante asociar características saludables como un elevado contenido en vitaminas o sustancias antioxidantes beneficiosas para la salud. El análisis de 69 entradas locales valencianas de tomate procedentes de ocho tipos varietales para caracteres como contenido en distintos azúcares, ácidos y antioxidantes puso de manifiesto diferencias significativas entre los tipos varietales ensayados para la mayoría de las características. Los tipos varietales 'Cherry' y 'De Penjar' presentaban mayor materia seca, contenido de sólidos solubles, acidez titulable, índice de sabor, ß-caroteno, ácido ascórbico, fenólicos totales y actividad antioxidante que los otros grupos. Los resultados obtenidos serán útiles para la diferenciación, mejora y selección de variedades locales de tomate con propiedades organolépticas mejoradas. Los estudios de caracterización y tipificación de las variedades tradicionales de tomate son imprescindibles para detectar factores que dificultan su puesta en valor y fomento de la explotación comercial. En este sentido, dentro de esta tesis se han caracterizado 3 poblaciones de tomate 'Valenciana d'El Perelló'. La menor productividad de las selecciones ensayadas en comparación con el control comercial parece ser debida a una concentración de la producción en los 2-3 primeros ramilletes de la planta. En el programa de selección y mejora del tomate 'Valenciana d¿El Perelló' que se ha realizado se dispone de selecciones con una distribución de cuajado mucho más uniforme. Por otra parte, una forma muy interesante de potenciar su cultivo, es obtener variedades resistentes al ToMV, derivándose un conjunto de linajes de este tomate con el gen Tm22 que confiere resistencia al ToMV. En cuanto a la adaptación a las condiciones agroclimáticas locales de las variedades tradicionales, esta característica es fundamental de este tipo de variedades. Así, las plantas de tomate del tipo 'De Penjar', han sido cultivadas tradicionalmente en la región mediterránea. Estos materiales están adaptados a condiciones de campo abierto bajo condiciones de bajo consumo de recursos. Sin embargo, el cultivo bajo invernadero se está expandiendo debido al aumento de su demanda. En primer lloc s'aborda la caracterització d'una col·lecció de 69 entrades de varietats valencianes de tomàquet de vuit tipus varietals ( 'De Borseta', 'Cherry', 'Cor',' De penjar ',' Plana ',' De Pruna ', 'Redona' i 'Valenciana') amb 64 descriptors convencionals i 38 característiques del Tomato Analyzer. S'ha posat de manifest que l'ús de forma complementària de l'eina fenómica del Tomato Analyzer és una nova estratègia per a la caracterització i classificació de les varietats locals de tomàquet, així com per la distinció entre tipus varietals relacionats. Malgrat l'ús generalitzat dels descriptors estandarditzats de tomàquet, hi ha poca informació sobre els efectes ambientals sobre els valors dels descriptors i sobre la seva herència. Els estudis de caracterització i tipificació de les varietats tradicionals de tomàquet són imprescindibles per detectar factors que en dificulten la posada en valor i foment de l'explotació comercial. En aquest sentit, dins d'aquesta tesi s'han caracteritzat març poblacions de tomàquet 'Valenciana d'El Perelló'. First, the characterization of a collection of 69 entries of Valencian tomato varieties of eight varietal types ('De Borseta', 'Cherry', 'Cor',' De Penjar ',' Plana ',' De Pruna ', 'Redona' and 'Valenciana') with 64 conventional descriptors and 38 characteristics of the Tomato Analyzer. It has been shown that the complementary use of the Tomato Analyzer's phenomic tool is a new strategy for the characterization and classification of local tomato varieties, as well as for the distinction between related varietal types. Despite the widespread use of standardized tomato descriptors, there is little information about the environmental effects on the values of the descriptors and their inheritance. The evaluation of 12 entries of tomato from seven varietal types in three different environments (conventional open air cultivation, open air ecological cultivation and greenhouse) through the use of 36 descriptors, has allowed to establish the influence of the cultivation system on these descriptors, being a wide range of variation, and demonstrating its usefulness to describe tomato materials, their diversity and relationships. On the other hand, it can be very interesting to associate healthy characteristics such as a high content of vitamins or antioxidant substances beneficial to health. The analysis of 69 Valencian local tomato entries from eight varietal types for characters as content in different sugars, acids and antioxidants revealed significant differences between the varietal types tested for most of the characteristics. The varietal types 'Cherry' and 'De Penjar' had higher dry matter, content of soluble solids, acidity, flavor index, ß-carotene, ascorbic acid, total phenolic and antioxidant activity than the other groups. The results obtained will be useful for the differentiation, improvement and selection of local tomato varieties with improved organoleptic properties. Characterization and typification studies of traditional tomato varieties are essential to detect factors that make it difficult to value and promote commercial exploitation. In this sense, within this thesis 3 populations of tomato 'Valenciana d'El Perelló' have been characterized. The lower productivity of the selections tested compared to the commercial control seems to be due to a concentration of the production in the first 2-3 bunches of the plant. In the program of selection and improvement of the tomato 'Valenciana d'El Perelló' that has been made, selections are available with a much more uniform fruit set distribution. On the other hand, a very interesting way to enhance their crop is to obtain varieties resistant to ToMV, deriving a set of lineages of this tomato with the Tm22 gene that confers resistance to ToMV. Regarding the adaptation to the local agroclimatic conditions of the traditional varieties, this characteristic is fundamental of this type of varieties. Thus, tomato plants of the 'De Penjar' type have traditionally been grown in the Mediterranean region. These materials are adapted to open field conditions under conditions of low resource consumption. However, greenhouse cultivation is expanding due to the increase in demand. The evaluation of 12 varieties (seven traditional entrances, three selections and two hybrids) of the 'De Penjar' type in outdoor cultivation and in greenhouse cultivation has revealed a high impact of the cultivation environment on the morphological, agronomic and chemical properties of varieties of tomato 'De Penjar' and suggests the need to develop specific varieties adapted to greenhouse cultivation.

[ES] Hemos utilizado descriptores convencionales y digitales para caracterizar germoplasma español, con el fin de evaluar la variabilidad y probar la eficiencia de esos métodos para diferenciar los materiales. Los resultados muestran gran diversidad dentro de la colección. Además, el fenotipado digital fue capaz de proporcionar una separación más potente. Finalmente, se seleccionó un subconjunto 17 descriptores capaz de distinguir entre accesiones, explicando el 81.81% de la varianza total. Los rasgos del fruto son los más relevantes para la separación entre variedades. Estos hallazgos serán útiles para la recuperación de las variedades tradicionales y el manejo del germoplasma. Las variedades tradicionales más apreciadas de México poseen una aptitud extraordinaria para ser usados como materiales de pre-mejora. Por lo tanto, nos propusimos estudiar la diversidad fenotípica y genotípica de estos materiales, así como el efecto de la polinización abierta en la fijación de las características. La caracterización morfológica y genética de la progenie mostró niveles similares de uniformidad y heterocigocidad a aquellos mostrados por las líneas parentales, sugiriendo que el sistema de reproducción por polinización abierta es eficiente en términos de alcanzar uniformidad agronómica, preservando un cierto grado de diversidad genética. Los estudios genéticos del germoplasma aportan información de gran utilidad. Se utilizó el GBS para estudiar una colección de Capsicum spp. española con el fin de arrojar luz sobre las relaciones filogenéticas de las variedades locales y evaluar su diversidad. Las accesiones europeas mostraron estar estrechamente relacionadas. Además, el origen y tipo de fruto fueron los principales factores que determinaron estructura genética. Los estudios de filogenia también mostraron una estrecha relación entre las accesiones española y mexicana. Los valores Tajima D fueron consistentes con selección positiva en los grupos de C. annuum relacionados con la domesticación. C. baccatum surge como una importante fuente de variabilidad genética y proporcionando a los investigadores la oportunidad de seleccionar individuos para ser introducidos en programas de mejora para el contenido en compuestos bioactivos y resilientes a las condiciones del cambio climático. Por ello, decidimos caracterizar una colección de C. baccatum y C. annuum. Se observó una gran influencia del sistema de cultivo sobre el contenido nutricional donde las concentraciones vieron favorecidas bajo cultivo al aire libre. Asimismo, las accesiones control de C. annuum mostraron concentraciones superiores para la mayoría de compuestos para los dos ambientes. Por otro lado, la buena respuesta C. baccatum demuestra que existe la posibilidad de mejorar estos materiales bajo las condiciones mediterráneas y un alto contenido en compuestos bioactivos. Por último, bajo nuestras condiciones, una ración de pimiento puede aportar entre un 70% y un 120% de la dosis diaria recomendada de ácido ascórbico y entre un 10% y un 60% de la dosis recomendada de minerales. La mejora de las variedades de pimiento para la absorción y uso del fósforo es de suma importancia en la tarea actual de reducir significativamente la necesidad de fertilizantes. El objetivo de este trabajo fue caracterizar una colección de pimiento frente a bajos insumos de fósforo. En general, las condiciones de estrés condujeron a una reducción significativa de la biomasa. Además, el tratamiento de bajo fósforo estimuló el desarrollo de la raíz lateral y pelos radiculares. Además, la concentración de fósforo en los tejidos vegetales disminuyó significativamente. Esta respuesta fue notoriamente superior en las raíces, mostrando la capacidad de movilizar el fósforo hacia otros órganos. Esto proporciona evidencias de la existencia de variabilidad dentro de Capsicum para la eficiencia del uso del fósforo susceptible de ser utilizada en lo, [CA] Hem utilitzat descriptors convencionals i digitals per a caracteritzar una col·lecció de varietats espanyoles, amb la finalitat d'avaluar la variabilitat i provar l'eficiència d'aqueixos mètodes per a diferenciar els materials. Els resultats mostren una gran diversitat dins de la col·lecció. A més, el fenotipat digital va permetre una separació més potent. Finalment, es va seleccionar un subconjunt de 17 descriptors digitals per a distingir entre accessions, explicant el 81.81% de la variància total. Els trets del fruit són els més rellevants per a la separació entre varietats. Aquestes troballes seran útils per a la recuperació de les varietats tradicionals i el maneig del germoplasma. Moltes de les varietats tradicionals més benvolgudes es poden trobar a Mèxic y posseeixen una aptitud extraordinària per a ser usats com a materials de pre-millora. Per tant, ens vam proposar estudiar la diversitat fenotípica i genotípica d'aquests materials, així com l'efecte de la pol·linització oberta en la fixació de les característiques morfològiques i en la fixació genètica. La caracterització morfològica y genètica va mostrar nivells similars d'uniformitat de la progènie a aquells mostrats per les línies parentals, sugerint que la reproducció de pol·linització oberta és eficient en termes d'aconseguir uniformitat agronòmica i alhora preservant un cert grau de diversitat genètica, un fet d'enorme rellevància per a l'adaptació a les condicions de canvi climàtic. Els estudis genètics del germoplasma existent aporten informació de gran utilitat. En la present tesi es va utilitzar el GBS per a estudiar una col·lecció de Capsicum spp. amb la finalitat de llançar llum sobre les relacions filogenètiques de les varietats locals i avaluar la seua diversitat i estructura. Les accessions europees estaven estretament relacionades. A més, les característiques dels fruits i l'origen van ser els principals factors que van determinar la l'estructura genètica. Els estudis de filogènia també van mostrar una estreta relació entre les accessions espanyola i mexicana. Els valors del mètode estadístic Tajima D van ser consistents amb la selecció positiva en els grups de C. annuum relacionats amb la domesticació. C. baccatum sorgeix com una important font de variabilitat i proporcionant l'oportunitat de seleccionar individus per a programes de millora per al contingut en compostos bioactius i condicions del canvi climàtic. Ací es va caracteritzar una col·lecció de C. baccatum i C. annuum. Es va observar una gran influència del sistema de cultiu, on les compostos es van veure afavorides baix cultiu a l'aire lliure. C. annuum van mostrar concentracions superiors per a la majoria de compostos. La bona resposta de C. baccatum demostra que existeix la possibilitat de millorar aquests materials de cara a obtindre individus adaptats a les condicions mediterrànies i un alt contingut en compostos bioactius. Per ultime, sota les nostres condicions, una ració de pimentó pot aportar entre un 70% i un 120% de la dosi diària recomanada d'acidifique ascòrbic i entre un 10% i un 60% de la dosi recomanada de minerals. La millora de les varietats de pimentó per a l'absorció i ús del fòsfor és de suma importància en la tasca actual de reduir la necessitat de fertilitzants. L'objectiu va ser caracteritzar una col·lecció de pimentó enfront de baixos inputs de fòsfor. En general, les condicions d'estrés van conduir a una reducció de la biomassa. A més, va estimular el desenvolupament de l'arrel lateral i de pèls radiculars. A més, la concentració de fòsfor en els teixits vegetals va disminuir significativament. Aquesta resposta va ser notòriament superior en les arrels, mostrant la capacitat de mobilitzar el fòsfor acumulat cap a altres òrgans. Això proporciona evidències de l'existència de variabilitat dins de Capsicum per a l'eficiència de l'ús del fòsfor susceptible de ser utilitzada en, [EN] Herein we used conventional and digital descriptors to characterize a collection Spanish landraces in order to assess the diversity and to test the discriminating ability of said methods. A considerable variation was found for the collection. Digital phenotyping enabled a more powerful separation. We conclude by selecting a subset of 17 descriptors which enable to distinguish among closely related C. annuum accessions. Finally, fruit traits explained the highest percentage of variance for our collection. These findings will be useful to the recovery of heirloom peppers and will boost germplasm characterization and management in seed banks. Some of the most known landraces from Mexico encompass a remarkable aptitude to be used as pre-breeding materials. Hence, we studied the phenotypic and genotypic diversity within these materials and the open-pollination effect on the fixation of morphological characteristics and on the genetic fixation. Morphological and genetic analysis of the progeny showed similar or lower levels of genetic and morphological uniformity than those from progenitors, suggesting that open-pollinated program is efficient in terms of reaching enough agronomic uniformity, while preserving a certain degree of genetic diversity, of paramount importance for the adaptation to climate change. Germplasm genetic studies provide vital information. Herein we used GBS to study a Spanish collection of Capsicum spp. to shed light into phylogenetic relationships and to evaluate their diversity and structure. European accessions showed a close relationship. Furthermore, fruit traits and region of origin were the main factors defining population structure. Spanish and Mexican accessions showed a close phylogenetic relationship; Finally, Tajima's D statistic values were consistent with positive selection in the C. annuum clusters related to domestication. These findings provide relevant information on the origin and relationships of Spanish landraces and for future association mapping studies in pepper. C. baccatum represents a remarkable genetic pool that provides opportunity to select superior individuals to be used in breeding programs for improved bioactive compounds content and resilience to climate change materials. Hence, we characterized a collection of C. baccatum and C. annuum materials. Cultivation system had a major effect controlling fruit's nutrient profile. C. annuum controls presented higher concentrations for most compounds under both conditions. The good performance of C. baccatum accessions shows that there are opportunities to breed materials adapted to the Mediterranean conditions and with interesting properties, especially under open-field conditions. Finally, under our conditions, a serving of pepper cultivated could provide between 70% and 120% of the recommended dietary allowance for ascorbic acid and between 10% and 60% for minerals. Improving pepper varieties for their uptake and use of phosphorus would significantly reduce the need for fertilizer applications. Hence, we characterized the main adaptations, of a Capsicum spp. collection, to low phosphorus inputs. Overall, stress conditions lead to significant reduction of biomass. Stress treatment stimulated lateral root length and root hairs growth. Furthermore, concentration of this mineral in plant tissues decreased significantly. This response was notoriously higher in the roots, demonstrating a high ability to mobilise accumulated phosphorus to other plant organs, providing evidence that within the Capsicum genus there is usable variability for phosphorus use efficiency for breeding programs for low input adaptation.

[EN] Conventional tomato (Solanum lycopersicum L.) descriptors are of great utility for gross morphological characterization but may not be practical for the precise fruit description required for distinguishing closely related cultivar groups. Tomato Analyzer is a new phenomics tool that provides multiple fruit morphology data from scanned images of fruit sections. We characterized 69 accessions of local tomato varieties from the region of Vale`ncia (Spain) corresponding to eight cultivar groups (Borseta, Cherry, Cor, Penjar, Plana, Pruna, Redona, and Valenciana) with 64 conventional and 38 Tomato Analyzer descriptors. Significant differences were found among accessions for all traits except for five monomorphic conventional descriptors, revealing a large diversity in the collection. Significant differences were also found among cultivar groups for 36 conventional and 37 Tomato Analyzer descriptors. The groups Borseta, Cherry, Penjar, Plana, and Pruna were clearly distinct and each of them presented many significant differences with the rest of groups. Conventional descriptors did not differentiate well the Cor, Redona, and Valenciana cultivar groups, but Tomato Analyzer descriptors clearly distinguish Valenciana from Cor and Redona groups. A multivariate principal components analysis (PCA) showed that with the exception of six (8.7 %) accessions, the different cultivar groups (including the very similar Cor and Redona) plotted in separate areas of the PCA graph. The results have shown that combined conventional and Tomato Analyzer descriptors in conjunction with PCA analysis are a powerful tool for characterization and classification of local tomato varieties, as well as for distinguishing between related cultivar groups. This has important implications for the enhancement and protection of local tomato varieties

[EN] Eggplant (Solanum melongena) is related to a large number of wild species that are a source of variation for breeding programmes, in particular for traits related to adaptation to climate change. However, wild species remain largely unexploited for eggplant breeding. Detailed phenotypic characterization of wild species and their hybrids with eggplant may allow identifying promising wild species and information on the genetic control and heterosis of relevant traits. We characterizated six eggplant accessions, 21 accessions of 12 wild species (the only primary genepool species S. insanum and 11 secondary genepool species) and 45 interspecific hybrids of eggplant ith wild species (18 with S. insanum and 27 with secondary genepool species) using 27 conventional morphological descriptors and 20 fruit morphometric descriptors obtained with the phenomics tool Tomato Analyzer. Significant differences were observed among cultivated, wild and interspecific hybrid groups for 18 conventional and 18 Tomato Analyzer descriptors, with hybrids generally having intermediate values. Wild species were generally more variable than cultivated accessions and interspecific hybrids displayed intermediate ranges of variation and coefficient of variation (CV) values, except for fruit shape traits in which the latter were the most variable. The multivariate principal components analysis (PCA) reveals a clear separation of wild species and cultivated accessions. Interspecific hybrids with S. insanum plotted closer to cultivated eggplant, while hybrids with secondary genepool species generally clustered together with wild species. Many differences were observed among wild species for traits of agronomic interest, which allowed identifying species of greatest potential interest for eggplant breeding. Heterosis values were positive for most vigor-related traits, while for fruit size values were close to zero for hybrids with S. incanum and highly negative for hybrids with secondary genepool species. Our results allowed the identification of potentially interesting wild species and interspecific hybrids for introgression breeding in eggplant. Il pomodoro (Solanum lycopersicum L.) è tra gli ortaggi più consumati nell'Unione Europea, e costituisce una preziosa fonte alimentare di nutrienti, vitamine e antiossidanti che sono riconosciuti importanti per una dieta sana. Nel corso dei secoli, numerose varietà di pomodoro tradizionali sono state selezionate dagli agricoltori per soddisfare le esigenze locali. Per contro, le cultivar moderne sono principalmente il risultato del miglioramento genetico focalizzato sulla produttività e sulla resistenza agli stress biotici; questo ha comportato una perdita progressiva della biodiversità della coltura, ed, in generale, un peggioramento delle qualità sensoriali. Nell'ambito del programma Horizon 2020, il progetto TRADITOM "Traditional tomato varieties and cultural practices: a case for agricultural diversification with impact on food security and health of European population" (http://traditom.eu/) punta alla conservazione, caratterizzazione e valorizzazione della diversità genetica presente nelle varietà tradizionali, anche al fine di migliorare la qualità e la sostenibilità delle varietà moderne di pomodoro. A tale scopo un'ampia collezione di varietà tradizionali europee (circa 1400) è oggetto di caratterizzazione a livello genomico (mediante Genotyping by Sequencing) e fenotipico. Il nostro gruppo ha reperito circa 240 varietà italiane tradizionali di pomodoro di diversa tipologia commerciale e forma del frutto (tondino, piennolo, tondeggiante, a cuore di bue, allungata). La collezione è stata allevata secondo pratiche colturali tradizionali ed è stata caratterizzata attraverso l'impiego di descrittori qualitativi e biometrici sia a livello di pianta che di frutto, anche mediante l'utilizzo del software Tomato Analyzer. Inoltre, una parte della collezione, ascrivibile alla tipologia da serbo, è stata caratterizzata per il comportamento dei frutti in post-raccolta.

Las frutas y hortalizas presentan compuestos bioactivos beneficiosos para la salud humana. El desarrollo de variedades con un mayor contenido en este tipo de compuestos es de interés, ya que contribuye a satisfacer una demanda creciente por parte de los consumidores por productos con propiedades funcionales. Dentro de las hortalizas, la berenjena (Solanum melongena) presenta una alta actividad antioxidante, fundamentalmente derivada de su alto contenido en polifenoles, y se ha demostrado que presenta efectos beneficiosos para la salud humana. Entre los compuestos fenólicos de la berenjena destaca el ácido clorogénico, ya que se trata del más abundante en este cultivo y presenta múltiples propiedades beneficiosas para la salud. Esta Tesis Doctoral trata de la caracterización y mejora de la berenjena para obtener información relevante y material vegetal para el desarrollo de variedades de berenjena con un mayor contenido en compuestos bioactivos, en particular polifenoles. Para ello utilizamos la variación intraespecífica e interespecífica. Por otra parte, en una mejora integral se debe tener en cuenta no solo el carácter a mejorar, sino también aquellos caracteres de interés para el éxito de una variedad, por lo que también hemos estudiado otros caracteres relacionados con el incremento del contenido en polifenoles, como puede ser el pardeamiento del fruto, y así como otros caracteres de interés general en mejora. En una primera parte de esta Tesis Doctoral nos centramos en el estudio de la diversidad en berenjena común y especies relacionadas para los caracteres objeto de esta tesis y también para caracteres de interés agronómico. El objetivo es evaluar la diversidad, identificar fuentes de variación y estudiar las relaciones entre caracteres. En un primer estudio, evaluamos una colección de variedades tradicionales de berenjena, en la cual hemos encontrado una alta diversidad para caracteres de calidad funcional y pardeamiento. En este estudio encontramos que el contenido en ácido clorogénico está correlacionado positivamente con la actividad antioxidante y que la correlación con el pardeamiento es baja, demostrando que es posible seleccionar variedades de berenjena con alto contenido en ácido clorogénico y pardeamiento moderado. También comprobamos que incluso con baja actividad polifenol oxidasa (PPO) se puede producir pardeamiento significativo, sugiriendo que la actividad PPO no es el factor limitante para el pardeamiento en la colección estudiada. Con objeto de ampliar la diversidad genética de la berenjena para la mejora de compuestos bioactivos y otros caracteres de importancia hemos estudiado la diversidad en una colección de berenjenas escarlata (S. aethiopicum) y gboma (S. macrocarpon). La caracterización morfológica mediante descriptores convencionales y herramientas fenómicas (Tomato Analyzer) nos ha permitido estudiar las relaciones entre los distintos grupos de cultivares y especies silvestres relacionadas y determinar que los complejos berenjena escarlata y gboma son hipervariables. En esta colección hemos estudiado también la actividad reductora y el contenido en ácido clorogénico, encontrado una enorme variabilidad. En general, la berenjena escarlata presenta contenidos relativamente bajos, mientras que la berenjena gboma, en particular su ancestro silvestre S. dasyphyllum, presentan valores muy elevados. También hemos comprobado en cultivos celulares de macrófagos que las variedades con mayor contenido en ácido clorogénico presentan una mayor inhibición de la producción de óxido nítrico (NO) indicando propiedades beneficiosas para la salud. En la segunda parte de la Tesis Doctoral hemos evaluado el interés de la hibridación interespecífica para la mejora de la berenjena, en particular para el contenido en compuestos bioactivos. Hemos obtenido dos familias, incluyendo retrocruzamientos, entre la berenjena común (S. melongena) por una parte y la berenjena escarlata cultivada (S. aethiopicum) y la especie silvestre S. incanum por otra. Los resultados muestran que la fertilidad de los materiales derivados de la hibridación entre S. melongena y S. aethiopicum es baja y que se obtiene una baja eficiencia en los retrocruzamientos hacia S. melongena. Además, el bajo contenido en polifenoles de S. aethiopicum se comporta como dominante. En cambio el retrocruzamiento hacia S. aethiopicum proporciona muchas plantas con mayores niveles de fertilidad. Sugerimos, por tanto, que S. melongena puede ser una fuente de variación para la mejora en contenido en polifenoles de la berenjena escarlata. La familia obtenida por hibridación interespecífica entre S. melongena y S. incanum mostró altos niveles de fertilidad y en el primer retrocruce hacia S. melongena se encuentran individuos morfológicamente similares a la Resumen berenjena cultivada. El estudio de los compuestos fenólicos mostró que S. incanum es una buena fuente de variación para la mejora de la berenjena común, con valores muy superiores a los de la especie cultivada. En el primer retrocruce se encuentran ya individuos con alto contenido en ácido clorogénico y pardeamiento moderado, lo cual sugiere que es posible introgresar exitosamente el alto contenido en ácido clorogénico de S. incanum en el fondo genético de la berenjena cultivada. En definitiva, los trabajos realizados en esta Tesis Doctoral aportan nuevo conocimiento sobre la diversidad y relaciones entre caracteres implicados en la calidad funcional de la berenjena y otros caracteres de interés en la mejora genética de este cultivo. [EN] The vegetable crop gboma eggplant (Solanum macrocarpon) and its wild ancestor S. dasyphyllum are native to Africa. Although it is an important crop in certain areas, there are few studies on gboma eggplant diversity and breeding. We have studied the morphological diversity of a collection of 11 accessions of S. macrocarpon and 1 accession of S. dasyphyllum using conventional descriptors for common eggplant (S. melongena) as well as the phenomics tool Tomato Analyzer. Results revealed that the gboma eggplant collection studied is highly diverse, with signi􀏐�icant differences among accessions for 27 traits. Also, signi􀏐�icant differences were found between S. macrocarpon and S. dasyphyllum for 14 traits, revealing that both species present considerable morphological differences. The large diversity found in the collection found indicates that selection and breeding can result in considerable improvement in this crop. [EN] Traditional descriptors for characterization of fruit shape of eggplant (Solanum melongena L.) provide limited information and do not allow a detailed characterization of this complex trait. New image analysis software tools like Tomato Analyzer, allow obtaining quantitative and objective data for a large number of fruit shape parameters. The Tomato Analyzer software tool was used for the evaluation of fruit shape in a collection of traditional varieties of eggplant from four varietal groups (Round, Semi-long, Egg Shaped, and Long). A total of 26 fruit shape parameters were obtained using the Tomato Analyzer software. Significant differences were found among varieties for all traits, except for shoulder height. In addition, considerable differences were identified among varietal groups for many parameters. Values for broad-sense heritability were high for most of the traits. The principal components analysis clearly separated the four varietal groups. Discriminant analysis allowed the correct classification to their actual variety of almost 60 % of the individual fruits. The wide variation observed and high values for heritability for most fruit shape parameters indicated that the image analysis software tools like Tomato Analyzer are of great utility for the phenotypic characterization of fruit shape in eggplant germplasm.
