Impaired socioemotional functioning characterizes autistic children, but does weak inhibition control underlie their socioemotional difficulty? This study addressed this question by examining whether and, if so, how inhibition control is affected by face realism and emotional valence in school-age autistic and neurotypical children. Fifty-two autistic and 52 age-matched neurotypical controls aged 10-12 years completed real and cartoon emotional face Go/Nogo tasks while event-related potentials (ERPs) were recorded. The analyses of inhibition-emotion components (i.e., N2, P3, and LPP) and a face-specific N170 revealed that autistic children elicited greater N2 while inhibiting Nogo trials and greater P3/LPP and late LPP for real but not cartoon emotional faces. Moreover, autistic children exhibited a reduced N170 to real face emotions only. Furthermore, correlation results showed that better behavioral inhibition and emotion recognition in autistic children were associated with a reduced N170. These findings suggest that neural mechanisms of inhibitory control in autistic children are less efficient and more disrupted during real face processing, which may affect their age-appropriate socio-emotional development., (© 2024 The Author(s). - Abstract
This study investigated the neural dynamics underlying the interplay between emotion and inhibition in Chinese autistic children. Electroencephalography (EEG) signals were recorded from 50 autistic and 46 non-autistic children during an emotional Go/Nogo task. Based on single-trial ERP analyses, autistic children, compared to their non-autistic peers, showed a larger Nogo-N170 for angry faces and an increased Nogo-N170 amplitude variation for happy faces during early visual perception. They also displayed a smaller N200 for all faces and a diminished Nogo-N200 amplitude variation for happy and neutral faces during inhibition monitoring and preparation. During the late stage, autistic children showed a larger posterior-Go-P300 for angry faces and an augmented posterior-Nogo-P300 for happy and neutral faces. 4. Unraveling the complex interplay between statistical learning and working memory in Chinese children with and without dyslexia across different ages.

- Abstract
The relation between statistical learning and working memory in children with developmental dyslexia (DD) remains unclear. This study employed a distributional and a conditional statistical learning experiment and a working memory task to examine this relation in 651 Chinese 6- to 12-year-olds with and without DD (N
DD = 199, 101 females; NwoDD = 452, 227 females; participated 2014-2019). Results showed working memory positively associated with recognizing high-predictable and familiar items in both groups, but negatively associated with recognizing unfamiliar items in the DD group only. These findings uncovered the complex interplay between statistical learning and working memory, demonstrating how different working memory abilities in children with and without DD relate to various statistical learning mechanisms at the item level., (© 2024 The Author(s). 5. DysDiTect: Dyslexia Identification Using CNN-Positional-LSTM-Attention Modeling with Chinese Dictation Task.

- Abstract
Handwriting difficulty is a defining feature of Chinese developmental dyslexia (DD) due to the complex structure and dense information contained within compound characters. Despite previous attempts to use deep neural network models to extract handwriting features, the temporal property of writing characters in sequential order during dictation tasks has been neglected. By combining transfer learning of convolutional neural network (CNN) and positional encoding with the temporal-sequential encoding of long short-term memory (LSTM) and attention mechanism, we trained and tested the model with handwriting images of 100,000 Chinese characters from 1064 children in Grades 2-6 (DD = 483; Typically Developing [TD] = 581). Using handwriting features only, the best model reached 83.2% accuracy, 79.2% sensitivity, 86.4% specificity, and 91.2% AUC. With grade information, the best model achieved 85.0% classification accuracy, 83.3% sensitivity, 86.4% specificity, and 89.7% AUC. These findings suggest the potential of utilizing machine learning technology to identify children at risk for dyslexia at an early age., Competing Interests: The authors declare no conflicts of interest. 6. Unraveling Temporal Dynamics of Multidimensional Statistical Learning in Implicit and Explicit Systems: An X-Way Hypothesis.

- Abstract
Statistical learning enables humans to involuntarily process and utilize different kinds of patterns from the environment. However, the cognitive mechanisms underlying the simultaneous acquisition of multiple regularities from different perceptual modalities remain unclear. A novel multidimensional serial reaction time task was developed to test 40 participants' ability to learn simple first-order and complex second-order relations between uni-modal visual and cross-modal audio-visual stimuli. Using the difference in reaction times between sequenced and random stimuli as the index of domain-general statistical learning, a significant difference and dissociation of learning occurred between the initial and final learning phases. Furthermore, we used a negative and positive occurrence-frequency-and-reaction-time correlation to indicate implicit and explicit learning, respectively, and found that learning simple uni-modal patterns involved an implicit-to-explicit segue, while acquiring complex cross-modal patterns required an explicit-to-implicit segue, resulting in a X-shape crossing of regularity learning. Thus, we propose an X-way hypothesis to elucidate the dynamic interplay between the implicit and explicit systems at two distinct stages when acquiring various regularities in a multidimensional probability space., (© 2024 The Authors. 7. Cue predictiveness and uncertainty determine cue representation during visual statistical learning.

- Abstract
This study investigated how humans process probabilistic-associated information when encountering varying levels of uncertainty during implicit visual statistical learning. A novel probabilistic cueing validation paradigm was developed to probe the representation of cues with high (75% probability), medium (50%), low (25%), or zero levels of predictiveness in response to preceding targets that appeared with high (75%), medium (50%), or low (25%) transitional probabilities (TPs). Experiments 1 and 2 demonstrated a significant negative association between cue probe identification accuracy and cue predictiveness when these cues appeared after high-TP but not medium-TP or low-TP targets, establishing exploration-like cue processing triggered by lower-uncertainty rather than high-uncertainty inputs. Experiment 3 ruled out the confounding factor of probe repetition and extended this finding by demonstrating (1) enhanced representation of low-predictive and zero-predictive but not high-predictive cues across blocks after high-TP targets and (2) enhanced representation of high-predictive but not low-predictive and zero-predictive cues across blocks after low-TP targets for learners who exhibited above-chance awareness of cue-target transition. These results suggest that during implicit statistical learning, input characteristics alter cue-processing mechanisms, such that exploration-like and exploitation-like mechanisms are triggered by lower-uncertainty and higher-uncertainty cue-target sequences, respectively., (© 2023 Zhang et al.; Published by Cold Spring Harbor Laboratory Press.)

8. Artificial intelligence technology and ultrasound-guided nerve block for analgesia in total knee arthroplasty.

- Abstract
Background: Knee diseases are more common in middle-aged and elderly people, so artificial knee replacement is also more used in middle-aged and elderly people. Although the patient's pain can be reduced through surgery, often accompanied by moderate pain after surgery and neutralization, which not only increases the psychological burden of the patient, but also greatly reduces the postoperative recovery effect, and may also lead to the occurrence of postoperative adverse events in severe cases., Aim: To investigate the analgesic effect of artificial intelligence (AI) and ultrasound-guided nerve block in total knee arthroplasty (TKA)., Methods: A total of 92 patients with TKA admitted to our hospital from January 2021 to January 2022 were opted and divided into two groups according to the treatment regimen. The control group received combined spinal-epidural anesthesia. The research group received AI technique combined with ultrasound-guided nerve block anesthesia. The sensory block time, motor block time, visual analogue scale (VAS) at different time points and complications were contrasted between the two groups., Results: The time of sensory block onset and sensory block perfection in the research group was shorter than those in the control group, but the results had no significant difference ( P > 0.05). Duration of sensory block in the research group was significantly longer than those in the control group ( P < 0.05). The time of motor block onset and motor block perfection in the research group was shorter than those in the control group, but the results had no significant difference ( P > 0.05). Duration of motor block in the research group was significantly longer than those in the control group. The VAS scales of the research group were significantly lower than that of the control group at different time points ( P < 0.05). The postoperative hip flexion and abduction range of motion in the research group were significantly better than those in the control group at different time points ( P < 0.05). Purpose: Although children's prosodic sensitivity links with their reading comprehension, the factors affecting this link remain unclear. By simultaneously measuring first language (L1) Chinese and second language (L2) English prosodic sensitivity and reading comprehension, this study examined the mediating role of syntactic awareness on prosody-reading comprehension among Hong Kong Chinese-English bilingual children., Method: A group of 227 Hong Kong Chinese-English bilingual fourth graders completed L1 and L2 prosodic sensitivity (Cantonese lexical tone awareness and English prosodic sensitivity), syntactic awareness, and reading comprehension and control measures of cognitive (nonverbal IQ, short-term memory, and working memory), metalinguistic (phonological awareness and morphological awareness), linguistic (vocabulary knowledge), and word reading skills., Results: The within-language analyses showed a partial mediation effect of Chinese syntactic awareness on the relation between Cantonese lexical tone awareness and Chinese reading comprehension, but a full mediation effect of English syntactic awareness on the relation between English prosodic sensitivity and English reading comprehension. The cross-language analyses revealed a significant direct effect of Cantonese lexical tone awareness on English reading comprehension and a significant indirect effect of English prosodic sensitivity on Chinese reading comprehension via Chinese syntactic awareness., Conclusion: These findings suggest that, despite the language-independent mediating role of syntactic awareness in bridging prosody and reading comprehension, the degree of this mediation is shaped by language-specific prosody and its relations with other linguistic structures, including semantics.

Using event-related potentials (ERPs), this study investigated how the brains of Chinese children of different ages extract and encode relational patterns contained in orthographic input. Ninety-nine Chinese children in Grades 1-3 performed an artificial orthography statistical learning task that comprised logographic components embedded in characters with high (100%), moderate (80%), and low (60%) positional consistency. The behavioral results indicated that across grades, participants more accurately recognized characters with high rather than low consistency. The neurophysiological results revealed that in each grade, the amplitude of some ERP components differed, with a larger P1 effect in the high consistency condition and a larger N170 and left-lateralized P300 effect in the low consistency condition. A smaller N170 effect occurred in Grade 3 than in Grade 1, and a larger P300 effect occurred in Grade 1 than in either Grade 2 or 3. 12. Psychosocial factors, but not professional practice skills, linked to self-perceived effectiveness of telepractice in school-based speech and language therapists during COVID-19 pandemic.

Background: Face-to-face class suspensions during the coronavirus disease (COVID-19) pandemic in 2019 increased telepractice in speech and language therapy. However, little is known about speech and language therapists' (SLTs) perceived effectiveness of telepractice and its antecedents., Aims: To examine the use of telepractice and the factors affecting its perceived effectiveness in Hong Kong mainstream schools during COVID-19., Methods & Procedures: Seventy-two school-based Hong Kong SLTs completed a 110-item online survey with six structural components: (1) concerns, (2) adoption, (3) student selection criteria, (4) perceived effectiveness, (5) continuous professional development and (6) attitudes/beliefs., Outcomes & Results: Over 90% of respondents adopted telepractice during the pandemic. Confirmatory factor analysis identified reliable constructs from their component measures. These participants reported great telepractice difficulties (especially in therapy preparation and managing students' attention and/or communication). Mixed-response analysis revealed that psychosocial factors (i.e., students' engagement and SLTs' attitudes towards telepractice) but not professional practice skill factors (i.e., student client selection criteria and SLTs' years of experience in school settings) contributed to SLTs' self-perceived effectiveness of telepractice., Conclusions & Implications: Our findings suggest that psychosocial factors play a more important role than professional practice skill factors in the self-perceived effectiveness of telepractice., What This Paper Adds: What is already known on the subject Previous surveys reported that although SLTs were interested in using telepractice before the COVID-19 pandemic, they showed concern about its effectiveness, resulting in a low adoption rate. One critical question naturally arises: What factors may affect the perceived effectiveness of telepractice by SLTs? What this paper adds to existing knowledge This study demonstrates for the first time that despite the high adoption rate of telepractice during the COVID-19 pandemic, school-based SLTs exhibited great difficulties, and the SLTs' self-perceived effectiveness of telepractice was related to psychosocial factors instead of professional practice skill factors. 13. Development and validation of the online learning process questionnaire (OLPQ) at home for primary-school children and their caregivers.

Despite the increasing use of virtual modalities in schools since the COVID-19 pandemic, no systematic tools exist to evaluate the process of online learning. We developed and validated an Online Learning Process Questionnaire (OLPQ) for assessing online at-home learning among 219 Hong Kong primary-school students and 474 caregivers. Exploratory and confirmatory factor analyses of caregivers' data classified the 58-item OLPQ into 11 subscales: (1) learning aims, (2) environmental structuring, (3) learning environment, (4) time management, (5) engagement in learning activities, (6) persistence, (7) interaction between teachers and students, (8) interaction among students, (9) feedback from the interface, (10) application of learning, and (11) meaning of learning under three learning phases. Confirmatory factor analysis of students' data further categorized the 11-subscale framework into three learning phases: preparatory, performance, and transfer. The OLPQ demonstrated excellent reliability and discriminant validity across caregiver (Cronbach's alpha = 0.98) and student samples (alpha = 0.98). 14. Syntactic awareness matters: uncovering reading comprehension difficulties in Hong Kong Chinese-English bilingual children.

This study examined whether syntactic awareness was related to reading comprehension difficulties in either first language (L1) Chinese or second language (L2) English, or both, among Hong Kong Chinese-English bilingual children. Parallel L1 and L2 metalinguistic and reading measures, including syntactic word-order, morphological awareness, phonological awareness, vocabulary, word reading, reading comprehension, and cognitive measures of nonverbal intelligence and working memory, were administered to 224 fourth-graders. Five groups of comprehenders were identified using a regression approach: (1) 12 poor in Chinese-only (PC), (2) 18 poor in English-only (PE), (3) six poor in both Chinese and English (PB), (4) 14 average in both Chinese and English (AB), and (5) seven good in both (GB). The results of multivariate analyses of covariance showed that (1) the PB group performed worse than the AB and GB groups in both L1 Chinese and L2 English syntactic awareness; (2) the PC and PE groups performed worse than the AB and GB groups in Chinese syntactic awareness; (3) the PE group had lower performance than the PC, AB, and GB groups in English syntactic awareness; and (4) no significant group difference was found in L2 morphological awareness or vocabulary across both languages. By suggesting that weakness in syntactic awareness can serve as a universal indicator for identifying poor comprehenders in either or both L1 Chinese and L2 English among Hong Kong Chinese-English bilingual children, these findings demonstrate the fundamental role of syntactic awareness in bilingual reading comprehension., (© 2022. - Abstract
Background: Pulsed radiofrequency (PRF) of the Gasserian ganglion is a common surgical intervention used to treat trigeminal postherpetic neuralgia (PHN). Dexamethasone has been reported to possess anti-inflammatory effects and potential analgesic benefits., Objectives: The primary objective of our study was to compare the therapeutic efficacies of PRF alone versus a combination of PRF and dexamethasone for trigeminal PHN., Study Design: A prospective, double-blind, randomized controlled trial., Setting: Department of Pain Management, Wuhan First Hospital., Methods: A total of 103 patients diagnosed with trigeminal PHN were randomly assigned into 2 groups (the PRF group and PRF plus dexamethasone [PRF+D] group). Digital subtraction angiography-guided puncture of the Gasserian ganglion was performed. All patients received PRF of the Gasserian ganglion first, and then a local injection was administered into the Gasserian ganglion. Patients in the PRF+D group received PRF therapy and one mL of 5 mg dexamethasone in the Gasserian ganglion, while patients in the PRF group received PRF therapy and one mL of normal saline in the Gasserian ganglion. The primary outcome was pain intensity, measured by the visual analog scale (VAS). The secondary outcome was quality of life, assessed by the Short Form-36 questionnaire (SF-36). The dosage of pregabalin administered was recorded to assess treatment effectiveness., Results: Compared with the PRF group in this study, the PRF+D group showed more promising outcome results in pain relief as measured by the VAS; quality of life enhancement, as measured by the SF-36; and a reduced requirement for antiepileptic drugs (P < 0.01)., Limitations: Single center study, relatively small number of patients., Conclusions: The therapeutic efficacy of PRF combined with a dexamethasone injection into the Gasserian ganglion was superior to that of PRF{and saline injection} alone of the Gasserian ganglion for trigeminal PHN.

16. Drawing a New Picture: Children with Developmental Dyslexia Exhibit Superior Nonverbal Creativity.

Background: Creativity and literacy are essential skills that today's children are expected to develop. However, the question of whether children with developmental dyslexia, a severe difficulty in reading and writing, exhibit any intact creativity strengths remains controversial. This study examined creativity strengths across verbal, figural, and nonverbal modalities, and the relations among creativity, nonverbal intelligence, and literacy skills, in younger and older Chinese children with and without developmental dyslexia., Methods: Two age groups of Chinese children with developmental dyslexia (Grade 2: N = 62; Grades 4-5: N = 62) and their age matched controls (Grade 2: N = 61; Grades 4-5: N = 61) were assessed on fluency and originality of verbal, figural, and nonverbal creativity, as well as on nonverbal intelligence, vocabulary, working memory, Chinese word reading accuracy and fluency, and word dictation., Results: Multifactorial analysis of variance demonstrated that regardless of grade level, children with developmental dyslexia exhibited higher nonverbal creativity than, and comparable figural creativity to, their typically developing peers. Moreover, the partial correlation analysis revealed creativity was not significantly correlated with nonverbal intelligence and literacy skills., Conclusions: These findings suggest that children with dyslexia possess strengths and even advantages in nonverbal creativity and that the relationship between intelligence and/or literacy and creativity is negligible., (Copyright © 2021 Elsevier Ltd. This study examined the reading comprehension profiles, and the related linguistic and cognitive skills, of 146 Chinese students in Grades 3-9 who are deaf or hard of hearing (d/Dhh). Employing a rigorous regression approach, the current study identified 19 unexpected poor comprehenders, 24 expected average comprehenders, and 16 unexpected good comprehenders. Compared to the expected average and unexpected good comprehenders, the unexpected poor comprehenders performed worse in broad linguistic skills (i.e., Chinese sign language comprehension, vocabulary, and segmental and suprasegmental phonological awareness), but their weaknesses in cognitive skills (i.e., working memory and executive function) were less severe. This study examined whether, and if so how, L1 and L2 segmental and suprasegmental phonological awareness is longitudinally related to L1 and L2 reading comprehension difficulties among Hong Kong Chinese-English bilingual children. Using a regression approach, we identified five types of comprehenders, i.e., 11 poor-Chinese/average-English comprehenders, 19 poor-English/average-Chinese comprehenders, six poor-Chinese/poor-English comprehenders, 12 average-Chinese/average-English comprehenders, and seven good-Chinese/good-English comprehenders among 223 Grade 4 Chinese-English bilingual children who were comparable in age, nonverbal IQ, and word reading, but differed in reading comprehension. These children were compared retrospectively on segmental and suprasegmental phonological awareness in both Chinese and English for three consecutive years from Grade 2 to Grade 4. The results revealed that only Cantonese lexical tone awareness distinguished poor comprehenders from typically developing comprehenders. Specifically, the poor-English/average-Chinese comprehenders performed worse than the average-Chinese/average-English and good-Chinese/good-English comprehenders in Grades 4 and 3, but not in Grade 2; and the poor-Chinese/average-English comprehenders performed worse than the good-Chinese/good-English comprehenders in Grades 4 and 3, but not in Grade 2. These findings suggest that suprasegmental phonological awareness, especially Cantonese lexical tone awareness, is critical for both Chinese and English reading comprehension development among Hong Kong bilingual children.

The authors investigated the roles of Chinese segmental (i.e., onset and rime) and suprasegmental (i.e., lexical tone) phonological awareness in Chinese text reading comprehension among 146 Chinese students in grades 3-9 who were d/Deaf and hard of hearing. All participants completed tasks involving onset and rime detection, lexical tone identification, sight word reading, text reading comprehension, nonverbal intelligence, and working memory. Path analyses showed that after controlling for age, grade, nonverbal intelligence, and working memory, segmental phonological awareness contributed both directly and indirectly (via sight word reading) to text reading comprehension, whereas suprasegmental lexical tone awareness only exerted only an indirect effect on text reading comprehension via sight word reading. These findings underscore the importance of phonological awareness in text reading comprehension for Chinese students who are d/Dhh.

This study examined distributional statistical learning of positional, phonetic, and semantic regularities of an artificial orthography in Chinese children aged 8-10 years: 29 with dyslexia, 29 age-matched controls, and 30 reading-level matched controls. Despite having positional regularity learning performance comparable to the controls, the children with dyslexia were poorer at learning left-right structured characters than top-bottom structured characters in high- and low-consistency conditions. Moreover, they showed difficulties in mapping a given sound or meaning to a specific character compared with the typically developing controls. These findings suggest that children with dyslexia have deficits in some, though not all, aspects of statistical learning of character orthography, which may reflect their difficulties in coping with distractors and inconsistency of orthographic input., (© 2020 Society for Research in Child Development.)

21. Lexical processing in sign language: A visual mismatch negativity study.

- Abstract
Event-related potential studies of spoken and written language show the automatic access of auditory and visual words, as indexed by mismatch negativity (MMN) or visual MMN (vMMN). The present study examined whether the same automatic lexical processing occurs in a visual-gestural language, i.e., Hong Kong Sign Language (HKSL). Using a classic visual oddball paradigm, deaf signers and hearing non-signers were presented with a sequence of static images representing HKSL lexical signs and non-signs. When compared with hearing non-signers, deaf signers exhibited an enhanced vMMN elicited by the lexical signs at around 230 ms, and a larger P1-N170 complex evoked by both lexical sign and non-sign standards at the parieto-occipital area in the early time window between 65 ms and 170 ms. These findings indicate that deaf signers implicitly process the lexical sign and that neural response differences between deaf signers and hearing non-signers occur at the early stage of sign processing., (Copyright © 2020 Elsevier Ltd. 22. Neurocognitive Correlates of Statistical Learning of Orthographic-Semantic Connections in Chinese Adult Learners.

- Abstract
We examined the neural correlates of the statistical learning of orthographic-semantic connections in Chinese adult learners. Visual event-related potentials (ERPs) were recorded while participants were exposed to a sequence of artificial logographic characters containing semantic radicals carrying low, moderate, or high levels of semantic consistency. The behavioral results showed that the mean accuracy of participants' recognition of previously exposed characters was 63.1% that was significantly above chance level (50%), indicating the statistical learning of the regularities of semantic radicals. The ERP data revealed a temporal sequence of the neural process of statistical learning of orthographic-semantic connections, and different brain indexes were found to be associated with this processing, i.e., a clear N170-P200-N400 pattern. For N170, the larger negative amplitudes were evoked by the high and moderate consistency than the low consistency. For P200, the mean amplitudes elicited by the moderate and low consistency were larger than the high consistency. In contrast, a larger N400 amplitude was observed in the low than moderate and high consistency; and more negative amplitude was elicited by the moderate than high consistency. We propose that the initial potential shifts (N170 and P200) may reflect orthographic or graphic form identification, while the later component (N400) may be associated with semantic information analysis.

Combining eye-tracking technique with a revised visual world paradigm, this study examined how positional, phonological, and semantic information of radicals are activated in visual Chinese character recognition. Participants' eye movements were tracked when they looked at four types of invented logographic characters including a semantic radical in the legal (e.g., [Formula: see text]) and illegal positions ([Formula: see text]), a phonetic radical in the legal (e.g., [Formula: see text]) and illegal positions (e.g., [Formula: see text]). These logographic characters were presented simultaneously with either a sound-cued (e.g., /qiao2/) or meaning-cued (e.g., a picture of a bridge) condition. Participants appeared to allocate more visual attention towards radicals in legal, rather than illegal, positions. In addition, more eye fixations occurred on phonetic, rather than on semantic, radicals across both sound- and meaning-cued conditions, indicating participants' strong preference for phonetic over semantic radicals in visual character processing. These results underscore the universal phonology principle in processing non-alphabetic Chinese logographic characters.

24. The Neural Signature of Statistical Learning of Orthography.

- Abstract
While an increasing number of behavioral studies suggest the importance of statistical learning in acquiring orthographic regularity across writing systems, no direct neural evidence supports this claim. The present study used event-related potentials (ERPs) to investigate the time course and the neural correlate of statistical learning of positional consistency in Chinese orthography. Visual ERPs were recorded, while Chinese adults performed an orthographic statistical learning task involving artificial characters varying in high, moderate, and low levels of positional consistency. The negative ERP deflection at the N1 time window, typically linked with orthographic regularity processing, was found in orthographic statistical learning with the low and moderate consistencies eliciting larger neural responses than the high consistency in the time window of 150-210 ms over occipital-temporal brain areas. - Abstract
Previously disclosed benzimidazole-based DGAT1 inhibitors containing a cyclohexane carboxylic acid moiety suffer from isomerization at the alpha position of the carboxylic acid group, generating active metabolites which exhibit DGAT1 inhibition comparable to the corresponding parent compounds. In this report, we describe the design, synthesis and profiling of benzimidazole-based DGAT1 inhibitors with a [3.1.0] bicyclohexane carboxylic acid moiety. Our results show that single isomer 3A maintains in vitro and in vivo inhibition against DGAT1. Purpose: The present study investigated the onset of statistical learning and examined whether the number of exposures to a repeated sequence influences the learning performance of children with dyslexia on a serial reaction time task., Method: Three groups of children (29 with dyslexia, 29 age-matched controls, and 30 reading level-matched controls) were administered a serial reaction time task, and their statistical learning performances after a small and a large number of exposures (40 vs. 180 exposures) were recorded and compared., Results: Children with dyslexia showed impaired statistical learning after a small number of exposures to a sequence, but intact statistical learning after a large number of exposures. In contrast, the age-matched and reading level-matched control groups showed intact statistical learning after both small and large numbers of exposures. Children with dyslexia also exhibited a slower learning rate than either control group., Conclusion: These results suggest that the amount of exposure to statistical patterns influences statistical learning performance in children with dyslexia.

Objective: To analyze the epidemiological characteristics of imported malaria in Xinjiang., Methods: The information of reported malaria cases and epidemiological records from various sources in Xinjiang Uygur Autonomous Region from 2004 to 2015 was collected. The epidemiological characteristics of malaria cases were analyzed., Results: Seventy-seven malaria cases were reported in Xinjiang from 2004 to 2015, and all were imported cases. In detail, 42 cases (54.5%) were reported during the time period of 2004-2009, with 24 laboratory diagnosed and 18 clinically diagnosed; 35 cases (45.5%) were reported from 2010 to 2015, with 27 laboratory diagnosed and 8 clinically diagnosed. Among the 51 cases with laboratory diagnosis, 18 were falciparum malaria, 31 were vivax malaria, and 2 unidentified. The male-to-female ratio was 6.7 ∶ 1(P<0.05). The age range of the patients was 14-75 years (39.7 ± 13.3 years), comprising 26.0% (20/77) for the range of 31-40 years and 24.7% (19/77) for the range of 41-50(P>0.05 among the age groups). From the perspective of case sources, 28 cases (36.4%) were imported from Africa and 49 cases(63.6%) from Asia (including other provinces of China). The interval from onset to final diagnosis ranged 1-320 days. Only 3 patients(3.9%) were diagnosed within 24 h, and 59 patients(76.6%) were not diagnosed until or over one week. In addition, 28 cases(36.4%) were reported by medical institutions at the provincial level, 15 cases(19.5%) were reported by medical institutions at the prefecture level, 5 cases(6.5%) were reported by the Center for Disease Control and Prevention, and the rest 29 cases(37.7%) were reported at the county level or below. No secondary transmission was found., Conclusion: Most of the imported malaria cases during 2004-2015 were diagnosed with laboratory tests, reported by medical institutions, and were from Africa and Asia.

MK-4256, a tetrahydro-β-carboline sstr3 antagonist, was discontinued due to a cardiovascular (CV) adverse effect observed in dogs. Additional investigations revealed that the CV liability (QTc prolongation) was caused by the hERG off-target activity of MK-4256 and was not due to sstr3 antagonism. In this Letter, we describe our extensive SAR effort at the C3 position of the tetrahydro-β-carboline structure. Background: Kashi Prefecture of Xinjiang is one of the most seriously affected areas with anthroponotic visceral leishmaniasis in China. A better understanding of space distribution features in this area was needed to guide strategies to eliminate visceral leishmaniasis from highly endemic areas. We performed a spatial analysis using the data collected in Bosh Klum Township in Xinjiang China., Methods: Based on the report of endemic diseases between 1990 and 2005, three villages with a high number of visceral leishmaniasis cases in Bosh Klum Township were selected. We conducted a household survey to collect the baseline data of kala-azar patients using standard case definitions. The geographical information was recorded with GIS equipment. A binomial distribution fitting test, runs test, and Scan statistical analysis were used to assess the space distribution of the study area., Results: The result of the binomial distribution fitting test showed that the distribution of visceral leishmaniasis cases in local families was inconsistent (χ(2) = 53.23, P < 0.01). The results of runs test showed that the distribution of leishmaniasis infected families along the channel was not random in the group of more than five infected families. The proportion of this kind of group in all infected families was 63.84 % (113 of 177). In the Scan statistical analysis, spatial aggregation was analyzed by poisson model, which found 3 spatial distribution areas 1) Zone A was located in a center point of 76.153447°E, 39.528477°N within its 1.11 mile radius, where the cumulative life-incidence of leishmaniasis was 1.95 times as high as that in surrounding areas (P < 0.05); 2) Zone B was located in a center point of 76.111968°E, 39.531895°N within its 0.54 mile radius, where the cumulative life-incidence of leishmaniasis was 1.82 times as high as that in surrounding areas (P < 0.01); and 3) Zone C was located in a center point of 76.195427°E, 39.563835°N within its 0.68 mile radius, where the cumulative life-incidence of leishmaniasis was 1.31 times as high as that in surrounding areas (P < 0.05)., Conclusions: The spatial distribution of visceral leishmaniasis-infected families was clustered. Thus, the proper use of this finding would be an improvement in highly endemic areas, which could help identify the types of endemic areas and population at high risk and carry out appropriate measures to prevent and control VL in this area as well.

- Abstract
Objective: To explore the effect of dexmedetomidine combined electrical stimulation on cognitive function of neurosurgical diseases patients treated by extracerebral intervention., Methods: Totally 122 patients with neurosurgical diseases who underwent selective intervention were randomly assigned to the observation group and the control group, 61 cases in each group. Patients in the control group recieved anesthesia by dexmedetomidine. Those in the observation group received electrical stimulation at Baihui (DU20), Yintang ( EX-HN3), and Neiguan (PC6) before dexmedetomidine anesthesia. The cognitive function of patients at preoperative day 1 and postoperative day 1 was respectively evaluated by Mini-Mental State Examinations (MMSE). Serum NSE, S-100β, IL-1β, IL-6, and TNF-α levels were detected in the two groups before intervention and immediately after intervention using ELISA., Results: MMSE scores of two groups were significantly reduced at post-intervention day 1, as compared with one day before intervention. MMSE score of the observation group at post-intervention day 1 was (23.15 ± 1.87) points, significantly higher than that of the control group [ (19.34 ± 1.64) points , (P < 0.05)]. The postoperative cognitive dysfunction (POCD) incidence rate of the observation group was 16.4% (10/61), significantly lower than that of the control group [39.3% (24/61); P < 0.05]. Compared with before intervention, NSE and S-100β protein levels, IL-1β, IL-6 and α-TNF levels of the two groups increased (P < 0.05). Post-intervention NSE and S-100β protein levels, IL-1β, IL-6 and α-TNF levels were significantly lower in the observation group than in the control group (P < 0.05)., Conclusion: Dexmedetomidine combied electrical stimulation could effectively prevent the occurrence of postoperative cognition, and reduce levels of NSA, S-100β, IL-1β, IL-6 and TNF-α.

The imidazolyl-tetrahydro-β-carboline class of sstr3 antagonists have demonstrated efficacy in a murine model of glucose excursion and may have potential as a treatment for type 2 diabetes. The first candidate in this class caused unacceptable QTc interval prolongation in oral, telemetrized cardiovascular (CV) dogs. Herein, we describe our efforts to identify an acceptable candidate without CV effects. We report the discovery of a novel series of DGAT1 inhibitors in the benzimidazole class with a piperdinyl-oxy-cyclohexanecarboxylic acid moiety. This novel series possesses significantly improved selectivity against the A2A receptor, no ACAT1 off-target activity at 10 μM, and higher aqueous solubility and free fraction in plasma as compared to the previously reported pyridyl-oxy-cyclohexanecarboxylic acid series. In particular, 5B was shown to possess an excellent selectivity profile by screening it against a panel of more than 100 biological targets. Compound 5B significantly reduces lipid excursion in LTT in mouse and rat, demonstrates DGAT1 mediated reduction of food intake and body weight in mice, is negative in a 3-strain Ames test, and appears to distribute preferentially in the liver and the intestine in mice. We believe this lead series possesses significant potential to identify optimized compounds for clinical development.

- Abstract
Antagonism of somatostatin subtype receptor 3 (sstr3) has emerged as a potential treatment of Type 2 diabetes. Unfortunately, the development of our first preclinical candidate, MK-4256, was discontinued due to a dose-dependent QTc (QT interval corrected for heart rate) prolongation observed in a conscious cardiovascular (CV) dog model. As the fate of the entire program rested on resolving this issue, it was imperative to determine whether the observed QTc prolongation was associated with hERG channel (the protein encoded by the human Ether-à-go-go-Related Gene) binding or was mechanism-based as a result of antagonizing sstr3. We investigated a structural series containing carboxylic acids to reduce the putative hERG off-target activity. A key tool compound, 3A, was identified from this SAR effort. As a potent sstr3 antagonist, 3A was shown to reduce glucose excursion in a mouse oGTT assay. Consistent with its minimal hERG activity from in vitro assays, 3A elicited little to no effect in an anesthetized, vagus-intact CV dog model at high plasma drug levels. These results afforded the critical conclusion that sstr3 antagonism is not responsible for the QTc effects and therefore cleared a path for the program to progress.

We report the design and synthesis of a series of novel DGAT1 inhibitors in the benzimidazole class with a pyridyl-oxy-cyclohexanecarboxylic acid moiety. In particular, compound 11A is a potent DGAT1 inhibitor with excellent selectivity against ACAT1. Compound 11A significantly reduces triglyceride excursion in lipid tolerance tests (LTT) in both mice and dogs at low plasma exposure. An in vivo study in mice with des-fluoro analogue 10A indicates that this series of compounds appears to distribute in intestine preferentially over plasma. The propensity to target intestine over plasma could be advantageous in reducing potential side effects since lower circulating levels of drug are required for efficacy. However, in the preclinical species, compound 11A undergoes cis/trans epimerization in vivo, which could complicate further development due to the presence of an active metabolite.

An outbreak of fever and meningitis/encephalitis occurred in Xinjiang, China, from August 5 to September 3, 2004. In preliminary diagnostic testing, several cerebrospinal fluid (CSF) and serum samples showed positive immunoglobulin M (IgM) antibody to Japanese encephalitis virus. Here, the CSF and serum samples of 6 cases collected at that time were tested by immunofluorescence assay (IFA), enzyme-linked immunosorbent assay (ELISA), and plaque reduction neutralization assay (PRNT) for the existence of IgM antibody or neutralization antibody against West Nile virus (WNV) or other arboviruses. The results demonstrate the evidence of West Nile infection in Xinjiang, China.

A structure-activity relationship study of the imidazolyl-β-tetrahydrocarboline series identified MK-4256 as a potent, selective SSTR3 antagonist, which demonstrated superior efficacy in a mouse oGTT model. MK-4256 reduced glucose excursion in a dose-dependent fashion with maximal efficacy achieved at doses as low as 0.03 mg/kg po. As compared with glipizide, MK-4256 showed a minimal hypoglycemia risk in mice.

- Abstract
Liao ning virus (LNV) is related to Banna virus, a known human-pathogen present in south-east Asia. Both viruses belong to the genus Seadornavirus, family Reoviridae. LNV causes lethal haemorrhage in experimentally infected mice. Twenty seven isolates of LNV were made from mosquitoes collected in different locations within the Xinjiang province of north-western China during 2005. These mosquitoes were caught in the accommodation of human patients with febrile manifestations, or in animal barns where sheep represent the main livestock species. The regions where LNV was isolated are affected by seasonal encephalitis, but are free of Japanese encephalitis (JE). Genome segment 10 (Seg-10) (encoding cell-attachment and serotype-determining protein VP10) and Seg-12 (encoding non-structural protein VP12) were sequenced for multiple LNV isolates. Phylogenetic analyses showed a less homogenous Seg-10 gene pool, as compared to segment 12. However, all of these isolates appear to belong to LNV type-1. These data suggest a relatively recent introduction of LNV into Xinjiang province, with substitution rates for LNV Seg-10 and Seg-12, respectively, of 2.29×10(-4) and 1.57×10(-4) substitutions/nt/year. These substitution rates are similar to those estimated for other dsRNA viruses. Our data indicate that the history of LNV is characterized by a lack of demographic fluctuations. However, a decline in the LNV population in the late 1980s-early 1990s, was indicated by data for both Seg-10 and Seg-12. Data also suggest a beginning of an expansion in the late 1990s as inferred from Seg-12 skyline plot.

In 2006, the first Chinese Tahyna virus isolate (XJ0625) was obtained in Xinjiang province and human infection were found in the same region. In this study, cell culture, animal experiments, electron microscopy, immunofluorescence assay and cross neutralization tests were performed to see the cell susceptibility, animal pathogenicity, morphology and antigenic and other biological characteristics of XJ0625. In addition, molecular biology software was used to analyze the characteristics of molecular evolution. The results showed that BHK-21 cell line was susceptible to XJ0625 and the virus was lethal to suckling mice when injected by intracranial ways. Similar to the other Bunyavirus, Tahyna virus is spherical enveloped virus under electron microscopy. XJ0625 infected cells showed strong fluorescent signal and could be neutralized by immune asities fluid with immnity to protype Tahyna virus Bardos 92. The sequence of the S and M segments showed 91.8% and 81.9% homology with Bardos 92.

41. Gallstone in jejunal limb with jejunocolonic fistula 10 years after Roux-en-Y choledochojejunostomy.

- Abstract
Roux-en-Y choledochojejunostomy is a common biliary reconstruction procedure. The collection of gallstones in the jejunal limb is a rare complication. Here we present a case of a 61-year-old Chinese female who received Roux-en-Y choledochojejunostomy 10 years ago. Diagnosis of recurrent bile duct stones accompanying infection was made before operation. She also had an abdominal mass which was possibly an intussuscepted colon or a huge fecolith. At laparotomy, an oval stone (5 cm in diameter) and 3 smaller multifaceted stones (2 cm in diameter) were found in the jejunal limb. A fistula between this jejunum and colon was also found. Although the typical manifestations of diarrhea were present, the diagnosis of a biliary colonic fistula was missed before operation. Partial colectomy was performed with the fistulous opening repaired. A T-tube was left in the jejunal limb and the mesocolon aperture was enlarged and revised. Her postoperative convalescence was uneventful. We report this case hoping to sharpen our diagnostic acumen.

Few outbreaks of the desert sub-type of zoonotic visceral leishmaniasis (VL) have been described worldwide. In 2008, the incidence rate of VL in Jiashi County, Xinjiang Uygur Autonomous Region in the western part of the People's Republic of China, increased more than twenty-folds compared to the average annual incidence rate. The majority of the cases (96.6%) occurred among <2 year-old infants. For the first time in the desert area of Xinjiang, the parasites were isolated from bone marrow aspirates, using the NNN medium culture approach. The genetic analysis of the ITS-1 nucleotide sequence indicated that three isolates from eastern Jiashi County were genetically closely related and belonged to the Leishmaniainfantum group. However, they differed from an isolate from Kashi city which was classified as a member of the Leishmaniadonovani group.

Objective: To investigate the epidemiological status of visceral leishmaniasis in Hamangou coal mine area of Korla City of Xinjiang Uygur Autonomous Region., Methods: Based on a hint of possible existence of patients, a retrospective survey was carried out house by house to find cases with suspected signs/symptoms of the disease. Meanwhile, a survey on current status was conducted, including physical examination (liver and spleen palpation) to those less than 15 years-old, leishmanin skin test and rk39 immunochromatographic strip test for part of the residents. Bone marrow smears were examined for the cases with clinical signs/symptoms and positive rk39 strip test. Sandflies were collected using routine methods in and around the area, identified, and dissected to find infection of promastigotes., Results: Leishmanin skin test was performed in 185 people with a positive rate of 21.1% (39/185), 39 out of 140 local residents who have lived there for more than 6 years showed positive (27.9%), while all residents who have lived less than 6 years and children under 5 years old were negative. Of the 81 children under 15 years old with a negative skin test, one showed positive for rk39 strip test, and leishmania body was found in the bone marrow smear of this case, so confirmed as visceral leishmaniasis. 12 sandflies were identified as Phlebotomus alexandri, and natural infection with promastigotes was found in one sandly., Conclusion: The investigation confirms that visceral leishmaniasis is endemic in the Hamangou coal mine area.

Objective: To probe the primary characteristic of 0507JS11 virus isolated from Culex sp. and determine the classification of 0507JS11 virus in taxonomy., Methods: 0507JS11 virus was cultured in Aedes albopictus C6/36 cells and cytopathic effects (CPEs) were recorded. Electro-microscopic morphology of 0507JS11 virus was observed. Total DNA extract of 0507JS11 virus was detected by 1% Agarose Gel Electrophoresis. Complete genomic sequence of 0507JS11 virus was sequenced and then made phylogenetic analysis., Results: 0507JS11 virus could cause CPEs in Aedes albopictus C6/36 cells. Viral particles have no envelope and appear icosahedron symmetry with diameter of 20 nm. The genome of 0507JS11 virus was positive single strand DNA (ssDNA) with full length of 3977 nt. However, a DNA band about 4 kbp was observed in the electrophoresis of total DNA extract of 0507JS11 virus. The coding region of the genome included three ORFs, ORF1 and ORF2 code NSP1 and NSP2, ORF3 codes VP. Phylogenetic analysis of the complete genomic sequence of 0507JS11 virus indicated an independent linear in Brevidensovirus., Conclusion: 0507JS11 virus is a new member in Brevidensovirus.

Objective: To isolate viruses from mosquitoes in the south of Xinjiang and identify these viruses primarily., Methods: A total of 13 491 mosquitoes were collected in the south of Xinjiang from Jul to Aug, 2005. These mosquitoes were divided into 130 groups and grinded respectively. The supernates were inoculated in C6/36 and Vero cells. Viruses isolated were detected, the genomic nucleic types by electrophoresis of viral genomes and the morphologies observed under electronmicroscope., Results: All 42 viruses were isolated, which caused CPEs on C6/36 but not on Vero cells. 27 viruses showed similar genomic profiles with 12 dsRNA segments. 1 virus displayed genomic profile with 10 dsRNA segments. 5 viruses took on similar genomic profiles with about 4 kbp DNA band. 9 viruses did not get any taxonomy information. Electromicroscopic pictures of these viruses revealed that above four types of viruses had distinguished morphologies indicating different virus species., Conclusion: There should be several virus species in the mosquitoes in the south of Xinjiang. dsRNA virus with 12 genomic segments should play analysis a predominant role in the south of Xinjiang.

Objective: To make an investigation on echinococcosis among animals in Gannan Tibetan Autonomous Prefecture., Methods: 21 villages from Maqu and Luqu counties were selected for the survey in August of 2004-September of 2007. Rodents were trapped in the field. Sheep and yak livers, hearts and lungs were collected from the local slaughterhouses for pathological examination. Domestic dogs (shepherd dogs) were de-wormed by 15% arecoline to receive adult worms and stray dogs were shot for dissection., Results: The prevalence of alveolar echinococcosis (AE) in Ochotona dahurica was 1.2% (1/87), and 2.3% (3/132) in Myospalax fontaniere, but no infection was found in Marmota himalayana, Ochotona tibetana and Mus musculus. 113 out of 1021 (11.1%) sheep were found infected with cystic echinococcosis (CE), and 3 (0.3%) with AE. 126 out of 634 (19.9%) yaks were infected with CE, and 2 yaks (0.3%) with AE. 17 out of 74 (23.0%) dogs were infected with Echinococcus granulosus (Eg), and 4 (5.4%) with Echinococcus multilocularis (Em)., Conclusion: The results showed that there is a widespread endemic of Echinococcus granulosus in dogs and wild animals in Gannan Tibetan Autonomous Prefecture, with less Echinococcus multilocularis infection.

Objective: To probe time-space clustering on the distribution of visceral leishmaniasis (VL) in Kashgar Region., Methods: Based on the geographic information system, a Poisson model of time-space statistical software was applied to analyze data over the past 11 years in the Kashgar Region. Zones with clustering phenomenon were conformed by geographic location and remote sensing images., Results: There existed three high risk clustering zones and corresponding time frames of VL in Kashgar Region. The center location of zone A was located in E 76.08 degrees, N 39.52 degrees, with radius as 6.58 km. The high risk time frame was from January 1st of 1999 to December 31st of 2003. Within the zone and time frame, the relative risk (RR) of VL incidence was 45.98 times higher than those outside the scope (P < 0.0001). Zone B' s center location was at E 79.20 degrees, N 39.91 degrees, with the radius as 4.93 km. Its high risk time frame was from January 1st of 2002 to December 31st of 2006. Within the zone and time frame, the RR of VL incidence was 9.58 times higher than those outside of the scope (P < 0.0001). Zone C' s center location was in E 76.23 degrees, N 39.40 degrees, and the radius was 7.63 km, with the high risk time frame from January 1st of 2000 to December 31st of 2004. Within the zone and time frame, the RR of VL incidence was 5.18 times higher than the one from outside of the scope (P < 0.0001)., Conclusion: The incidence of VL in Kashgar Region was non-randomly distributed while there existed obvious time-space clustering, with all of three high risk clustering zones located in oasis area where appeared the focus area for control and surveillance of VL.

0507JS60 virus was isolated from a pool of Culex sp. collected in Kashi, Xinjiang, which could be propagated stably on C6/36 cells and caused cytopathic effects continuously. Viral particles had no envelope and appeared round with diameter of about 55nm (n = 10). Capsomeres on the surface of capsid were clearly visible. Electrophoresis of viral genome showed a profile of 12 double stranded RNA (dsRNA) segments. Sequencing of the twelfth segment revealed the length of 760bp (GenBank ID: FJ157354). A single open reading frame (ORF) was found and encoded a protein of 174 amino acids with a molecular mass of 18.9kD. The nucleotide sequence had similarity over 89% with that of LNV, but the deduced amino acid sequence had similarity over 91% with that of LNV. A phylogenetic tree was constructed to compare the corresponding genetic sequences in Seadornavirus. The tree demonstrated that 0507JS60 virus lied in the same branch with LNV and more closely related to LNV-NE9712. 0507JS60 virus was identified as LNV, which was firstly isolated outside the Northeast of China.

0507BS3 virus was isolated from a mixed pool of Culex sp. and Anopheles sp. collected in Kashi, Xinjiang, China. 0507BS3 virus could cause cytopathic effects on C6/36 cells but not on Vero and BHK-21 cells. Viral particles had no envelope and appeared round with diameter of about 60 nm (n = 20). Viral capsid was composed of a single layer and a central core. Capsomeres on the surface of capsid were clearly visible. Electrophoresis of viral genome showed a profile of 10 double stranded RNA (dsRNA) segments. Sequencing of the tenth segment revealed the length of 964bp (GenBank ID: FJ150869). The nucleotide sequence had no similarity with any other viral genomic sequences, but the deduced amino acid sequence significantly matched the polyhedrin genes of cytoplasmic polyhedrosis virus (CPV) in some sections. A phylogenetic tree was constructed to compare the polyhedrin gene sequences of all CPV types in GenBank. The tree demonstrated that 0507BS3 virus was only distantly related to the other CPV types and belonged to a new CPV type.

- Abstract
Health education activities including distribution of brochures, posters, and lectures were conducted in selected villages of a kala-azar endemic-township, Kashi city of Xinjiang in 2004-2006. Questionnaire was carried out before and after health education in residents and students. The percentage of correctly answering all 5 questions on leishmaniasis in residents and primary/secondary school students increased from 0.7% and 0 to 54.2% and 70.7% respectively, indicating that health education can raise public awareness for the disease prevention and control.
