Abstract Natalisin (NTL) is a conserved neuropeptide, only present in insects, that has been reported to regulate their sexual activity. In this study, we investigated the involvement of NTL in the reproductive behaviors of a major invasive pest, Spodoptera frugiperda. We identified NTL precursor-encoded transcripts, and evaluated their transcript levels in different stages and tissues of S. frugiperda. The results showed that the NTL transcript level was expressed in both male and female pupae and both male and female adults in the later stage. It was highly expressed in male pupae, 3-day-old male and female adults, and 5-day-old male adults. In different tissues, the expression level is higher in the male and female adult brain and male testis. Immunohistochemical staining of the brain of S. frugiperda female and male adults revealed that three pairs of brain neurons of S. frugiperda adults of both sexes secreted and expressed NTL. To study the role of NTL in reproductive behaviors, NTL was silenced in S. frugiperda male and female adults by RNA interference (RNAi) technology, the results showed that silencing NTL could significantly affect the sexual activity behavior of the adults, reducing the calling rate of females, the courtship rate of males, and the mating rate. In summary, this study emphasizes the important role of NTL in regulating the mating behavior and sexual activity of S. frugiperda in both male and female adults, potentially laying a foundation to employ NTL as a new insect-specific target to control populations of pest insects.

Parasitoids have the potential to alter the gut microbiota of their host insects post-parasitization, thereby influencing the host’s physiological functions and creating a more favorable environment for the survival of the parasitoid’s progeny. Cotesia ruficrus is a native enemy of the important invasive fall armyworm (FAW) pest, Spodoptera frugiperda, in China, exhibiting significant pest control capabilities. To investigate the impact of C. ruficrus on the gut bacteria of FAW caterpillars following parasitism, we used 16S rRNA sequencing technology to analyze the diversity and richness of gut bacteria in both long-term laboratory and short-term laboratory FAW caterpillars. The results revealed Enterococcus as the predominant bacteria across all treatments, while no significant differences were observed in the diversity and richness of gut bacteria between non-parasitized and parasitized long-term laboratory FAW caterpillars. Similarly, while the diversity of gut bacteria in non-parasitized and parasitized short-term laboratory FAWs showed no significant variance, a marked discrepancy in richness was noted. Moreover, the richness of gut bacteria in short-term laboratory FAW caterpillars surpassed that of their long-term laboratory counterparts. In addition, it was found that Corynebacterium existed only in the intestinal tract of FAW caterpillars that were parasitized by C. ruficrus. These results substantiate that C. ruficrus parasitization can alter the gut microbiota of FAW caterpillars, providing valuable insights into the interplay between gut microbiota and the dynamics of parasitoid–host interactions.

Chemosensory proteins (CSPs) are a class of soluble proteins that facilitate the recognition of chemical signals in insects. While CSP genes have been identified in many insect species, studies investigating their function remain limited. Cotesia ruficrus (Hymenoptera: Braconidae) holds promise as an indigenous biological control agent for managing the invasive pest Spodoptera frugiperda (Lepidoptera: Noctuidae) in China. This study aimed to shed light on the gene expression, ligand binding, and molecular docking of CrufCSP1 in C. ruficrus. A RT-qPCR analysis revealed that the expression of CrufCSP1 was higher in the wings, with male adults exhibiting significantly higher relative expression levels than other developmental stages. A fluorescence competitive binding analysis further demonstrated that CrufCSP1 has a high binding ability with several host-related volatiles, with trans-2-hexenal, octanal, and benzaldehyde showing the strongest affinity to CrufCSP1. A molecular docking analysis indicated that specific amino acid residues (Phe24, Asp25, Thr53, and Lys81) of CrufCSP1 can bind to these specific ligands. Together, these findings suggest that CrufCSP1 may play a crucial role in the process of C. ruficrus locating hosts. This knowledge can contribute to the development of more efficient and eco-friendly strategies for protecting crops and managing pests.

The green peach aphid, Myzus persicae (Sulzer), is a significant global pest in horticultural and field crops. Afidopyropen is a novel systemic insecticide with high efficacy against sucking pests, and it is suitable for the management of M. persicae. However, the persistent toxicity and dissipation dynamics of afidopyropen in vegetables remain unknown. In this study, we determined the residual activity and dissipation dynamics of afidopyropen against M. persicae on cabbage and chili. The data showed that the toxicity of afidopyropen against M. persicae lasted more than 30 days; the corrected mortality was greater than 80% 10 days after application and was 50–60% 30 days post-application. The afidopyropen residues on cabbage and chili plants were quantified using ultrahigh-pressure liquid chromatography–tandem mass spectrometry. The dissipation half-lives of afidopyropen on cabbage and chili plants ranged from 1.45 to 2.34 days and 3.98–5.98 days at different recommended dosages, respectively. Our findings provide valuable data for the maximum residue limits of afidopyropen on vegetables and will help growers determine the frequency and timing of its application on cabbage and chili.

ABSTRACT Aphids display wing polyphenism, and the mother can produce a wingless morph for reproduction and a winged morph for dispersal. It is believed that the wingless morph is an adaptive status under favorable conditions and is determined prenatally. In this study, we have found that winged nymphs of the pea aphid, Acyrthosiphon pisum, can change from winged to wingless during normal development. Our results showed that winged nymphs could become the wingless morph by apterization in response to changes from stressful to favorable conditions. The acquired wingless aphids had higher fecundity than the winged morph. However, this process of regression from winged to wingless morph was inhibited by Serratia symbiotica. The existence of the symbiont did not affect the body mass and fecundity of adult aphids, but it increased the body weight of nymphs and temporally increased the quantity of a primary symbiont, Buchnera aphidicola. Our results showed that despite temporal improvement of living conditions causing the induction of apterization of winged nymphs, the inhibition effect of S. symbiotica on this process was activated simultaneously. This finding, for the first time, reveals that the wingless morph can be changed postnatally, which explains a novel regulating mechanism of wing polyphenism driven by external abiotic stimuli and internal biotic regulation together in aphids. IMPORTANCE Wing polyphenism is an important adaptative response to environmental changes for aphids. Endosymbionts are widespread in aphids and also confer the ability to withstand unfavorable conditions. However, little is known about whether endosymbionts are involved in the wing polyphenism. In this study, we report a new finding that winged nymphs of the pea aphid could turn into adults without wings or wing-related structures through apterization when winged nymphs escaped from stressful to favorable environments. Further analysis revealed that the facultative symbiont S. symbiotica could prevent the temporal determination of the host in wing suppression by inhibiting apterization, to enhance its spread. Our findings provide a novel angle to understanding the wing polyphenism regulation of aphids.

Concerns regarding the ecological and health risks posed by synthetic insecticides have instigated the exploration of alternative methods for controlling insects, such as entomopathogenic fungi (EPF) as biocontrol agents. Therefore, this review discusses their use as a potential alternative to chemical insecticides and especially focuses on the two major ones, Beauveria bassiana and Metarhizium anisopliae, as examples. First, this review exemplifies how B. bassiana- and M. anisopliae-based biopesticides are used in the world. Then, we discuss the mechanism of action by which EPF interacts with insects, focusing on the penetration of the cuticle and the subsequent death of the host. The interactions between EPF and the insect microbiome, as well as the enhancement of the insect immune response, are also summarized. Finally, this review presents recent research that N-glycans may play a role in eliciting an immune response in insects, resulting in the increased expression of immune-related genes and smaller peritrophic matrix pores, reducing insect midgut permeability. Overall, this paper provides an overview of the EPF in insect control and highlights the latest developments relating to the interaction between fungi and insect immunity.

Gut-associated microbes can influence insect health and fitness. Understanding the structure of bacterial communities provides valuable insights on how different species may be selected and their functional characteristics in their hosts. The neutral model is powerful in predicting the structure of microbial communities, but its application in insects remains rare. Here, we examined the contribution of neutral processes to the gut-associated bacterial communities in Helicoverpa armigera caterpillars collected from different maize varieties at four locations. The gut-associated bacteria can be assigned to 37 Phyla, 119 orders, and 515 genera, with each individual gut containing 17–75% of the OTUs and 19–79% of the genera in the pooled samples of each population. The distribution patterns of most (75.59–83.74%) bacterial taxa were in good agreement with the neutral expectations. Of the remaining OTUs, some were detected in more individual hosts than would be predicted by the neutral model (i.e., above-partition), and others were detected in fewer individual hosts than predicted by the neutral model (i.e., below-partition). The bacterial taxa in the above-partitions were potentially selected by the caterpillar hosts, while the bacteria in the below-partitions may be preferentially eliminated by the hosts. Moreover, the gut-associated microbiota seemed to vary between maize varieties and locations, so ecological parameters outside hosts can affect the bacterial communities. Therefore, the structure of gut microbiota in the H. armigera caterpillar was mainly determined by stochastic processes, and the bacteria in the above-partition warrant further investigation for their potential roles in the caterpillar host.

Chemosensory genes play important roles in insect behaviors and have thus become potential molecular targets for pest control based on the manipulation of chemoreception-driven behaviors. The great gray weevil Sympiezomias velatus (Chevrolat) (Coleoptera: Curculionidae) is an important agricultural pest that causes serious economic losses to many crops in China, but its chemosensory genes have not been reported. Here we assembled the antennal transcriptomes of female and male adult S. velatus and revealed the major chemosensory genes necessary for olfaction. A total of 138 candidate chemosensory genes in six families were identified, including 41 encoding odorant-binding proteins (OBPs), 11 encoding chemosensory proteins (CSPs), 62 encoding odorant receptors (ORs), 15 encoding gustatory receptors (GRs), six encoding ionotropic receptors (IRs), and three encoding sensory neuron membrane proteins (SNMPs). We analyzed their phylogenetic relationship based on the amino acid sequences of these chemosensory-related protein families in S. velatus and other insects, and the expression profiles based on their antennal transcriptomes. Chemosensory genes that show antenna-abundant/specific or sex-biased expression were observed, suggesting that these genes might have functions in olfaction. Furthermore, we chose an antenna-abundant OBP belonging to ABPX subfamily, SvelOBP15, to investigate its binding property. The results showed that among 33 tested compounds, SvelOBP15 displayed high binding affinities (Ki = 7.36–12.94 μmol/L) with farnesol, nerolidol, limonene and diisobutyl phthalate, indicating that SvelOBP15 plays olfactory roles by binding and transporting specific plant volatiles. These findings will help us better understand the olfactory systems of S. velatus, and provide a basis for functional elucidation of these chemosensory genes.

Plant resistance against insects mainly depends on nutrient restriction and toxic metabolites, but the relative importance of nutrition and toxins remains elusive. We examined performance, nutrition ingestion, and transcriptome response of the English grain aphid, Sitobion avenae, feeding on resistant Xiaoyan 22 (XY22) and susceptible Xinong 979 (XN979) wheat cultivars. Aphids had lower body weight and fecundity when feeding on XY22 than on XN979, although the phloem sap of XY22 had a higher nutritive quality (in terms of amino acid:sucrose ratio). Aphids feeding on XY22 also had a lower honeydew excretion rate than those on XN979, suggesting that aphids ingested less phloem sap from XY22. The transcriptome data showed 600 differentially expressed genes (DEGs), and 11 of the top 20 KEGG pathways significantly enriched in DEGs were involved in nutrient metabolism. We found 81 DEGs associated with the metabolism of sugars, lipids, and amino acids, 59 of which were significantly downregulated in aphids feeding on XY22. In contrast, there were 18 DEGs related to detoxifying metabolism, namely eight UDP-glucuronosyltransferases, six cytochromes P450 monooxygenases, one glutathione S-transferase, two ATP-binding cassette transporters, and one major facilitator superfamily transporter; 12 of these were upregulated in the aphids feeding on XY22. Our results indicated that both the quantity and quality of phloem nutrition available to aphids are critical for the growth and development of aphids, and the higher resistance of XY22 is mainly due to the reduction in phloem sap ingested by aphids, rather than toxic metabolites.

In insects, male accessory gland proteins (ACPs) are important reproductive proteins secreted by male accessory glands (MAGs) of the internal male reproductive system. During mating, ACPs are transferred along with sperms inside female bodies and have a significant impact on the post-mating physiology changes of the females. Under sexual selection pressures, the ACPs exhibit remarkably rapid and divergent evolution and vary from species to species. The diamondback moth, Plutella xylostella (L.) (Lepidoptera: Plutellidae), is a major insect pest of cruciferous vegetables worldwide. Mating has a profound impact on the females’ behavior and physiology in this species. It is still unclear what the ACPs are in this species. In this study, two different proteomic methods were used to identify ACPs in P. xylostella. The proteins of MAGs were compared immediately before and after mating by using a tandem mass tags (TMT) quantitative proteomic analysis. The proteomes of copulatory bursas (CB) in mated females shortly after mating were also analyzed by the shotgun LC-MS/MS technique. In total, we identified 123 putative secreted ACPs. Comparing P. xylostella with other four insect ACPs, trypsins were the only ACPs detected in all insect species. We also identified some new insect ACPs, including proteins with chitin binding Peritrophin-A domain, PMP-22/ EMP/ MP20/ Claudin tight junction domain-containing protein, netrin-1, type II inositol 1,4,5-trisphosphate 5-phosphatase, two spaetzles, allatostatin-CC, and cuticular protein. This is the first time that ACPs have been identified and analyzed in P. xylostella. Our results have provided an important list of putative secreted ACPs, and have set the stage for further exploration of the functions of these putative proteins in P. xylostella reproduction.

The green peach aphid, Myzus persicae (Sulzer) is a generalist pest of various host plants, whose feeding preference and growth performance mainly depends on the quantity and quality of nutrients and defensive metabolites in host plants. Here, we studied the preference and performance of M. persicae on three major Brassicaceae vegetables in China and measured nutrient (amino acids) and defensive metabolites (glucosinolates) in these plants. We found that M. persicae preferred and performed better on Chinese cabbage than cabbage and radish, which may be due to the relatively higher concentration of amino acids and lower levels of indole glucosinolates in their leaves. The glucosinolates level in cabbage leaves was ten times higher than the other two plants, while the amino acid concentration in radish was only half of the cabbage or Chinese cabbage. The higher concentration of indole glucosinolates in cabbage and lower levels of amino acids in radish may account for the poorer preference and growth of M. persicae on these two plants. These results suggest that both amino acids and glucosinolates in plants may play important roles in the preference and performance of M. persicae, which provide new knowledge for the cultivation and breeding of Brassicaceae vegetables.

Cereal aphids are major insect pests of wheat, which cause significant damages to wheat production. Previous studies mainly focused on the resistance of different wheat varieties to one specific aphid species. However, reports on the physiology and defense responses of wheat to different cereal aphids are basically lacking. In this work, we studied the feeding behavior of three cereal aphids: the grain aphid, Sitobion avenae (Fabricius), the greenbugs, Schizaphis graminum (Rondani), and the bird cherry-oat aphid, Rhopalosiphum padi (Linnaeus) on winter wheat, and the physiology and defense responses of wheat to the infestation of these cereal aphids with focus on how these cereal aphids utilize divergent strategies to optimize their nutrition requirement from wheat leaves. Our results indicated that S. graminum and R. padi were better adapted to penetrating phloem tissue and to collect more nutrition than S. avenae. The harm on wheat physiology committed by S. graminum and R. padi was severer than that by S. avenae, through reducing chlorophyll concentration and interfering metabolism genes. Furthermore, cereal aphids manipulated the plant nutrition metabolism by increasing the relative concentration of major amino acids and percentage of essential amino acids. In addition, different cereal aphids triggered specific defense response in wheat. All of these results suggested that different cereal aphids utilize divergent strategies to change the physiological and defense responses of their host plants in order to optimize their nutrition absorption and requirement. These findings not only extend our current knowledge on the insect–plant interactions but also provide useful clues to develop novel biotechnological strategies for enhancing the resistance and tolerance of crop plants against phloem-feeding insects.

The oriental armyworm, Mythimna separata (Lepidoptera: Noctuidae) is a harmful pest which causes heavy loss in agricultural crops. Sex identification of M. separata is very important for understanding the biology and management of this pest. Development of fast and convenient methods to identify M. separata female and male is the necessary prerequisite for conducting gender-related studies. In this study, new methods to differentiate sex of M. separata at pupal and adult stages were investigated. Our results showed that posterior abdominal segments containing genital organs which are morphological traits to differentiate female and male. Genital openings are important characters for the female and male pupae differentiation. For female pupa, it locates on the ventral side of eighth and ninth abdominal segments and appears as a visible longitudinal suture; for male pupa, it locates on the ventral side of the ninth abdominal segment and appears as a short slit surrounded by a pair of visible semicircular lumps. Bristle numbers are another indicator for sex differentiation of adults. Each female has three-bristled frenulum and each male has one bristled-frenulum near the base of hind wing of each side of moth body. We confirmed the two sex differentiation methods manually by dissecting genitalia of adults and found their genger was as same as what we examined the sex of pupae and adults using the morphological methods.

Aphelinus asychis is an important aphid endoparasitoid. Under field and greenhouse conditions, high temperature is one of the factors limiting the application of A. asychis for biological pest control. To explore the potential role of transient receptor potential (TRP) channels and heat shock proteins (HSPs) in this process, we identified 11 genes encoding TRP channels and nine genes encoding HSPs. Three proteins (AasyTRPA5, AasyPyrexia, AasyPainless) that belong to transient receptor potential ankyrin (TRPA) subfamily and nine HSPs are involved in the response to high temperature. We also investigated the survival of A. asychis and the response of the identified TRP channels and HSPs to high temperature. The results showed that the maximum temperature that allowed A. asychis survival was approximately 41°C; females had higher survival rates than that of the males at 40 and 41°C. Short-term heat-shock resulted in increased expression of Aasyshsp in males, and Aasyhsp40, Aasyhsp68, Aasyhsp70-4, Aasyhsp70-5 and Aasyhsp90 were upregulated and then downregulated, whereas Aasyhsp70-3 was upregulated at 41°C. Moreover, Aasyhsp40 and Aasyhsp90 showed higher expression levels in females, while Aasyshsp and Aasyhsp70-3 presented opposite expression patterns. At temperature above 35°C, expression of AasyPyrexia in females was significant higher than that in males, whereas AasyPainless and AasyTRPA5 presented higher expression in males at 40 and 41°C, respectively. Altogether, these results indicate that protection against thermal stress in A. asychis is coordinated by TRP channels and HSPs. These findings provide a basis for understanding the potential mechanism of A. asychis in response to high temperatures.

Background: Flower thrips (Frankliniella intonsa, IFT) and west flower thrips (Frankliniella occidentalis, WFT) are often found together on the host plant in China. WFT is an important invasive species that seems to outcompete the native IFT. In order to clarify the interspecific competitions between the two thrips, this study measured the population development of IFT and WFT under sexual and parthenogenetic reproductive modes on two hosts (fresh lentil bean pods with/without honey and pepper plants at seedling/flowering stages) in the laboratory. Results: When reared on fresh lentil bean pods (with/without honey), WFT population size was lower in mixed species populations compared to single species populations but the presence of WFT had nor negative effect on IFT population size. These results were dependent of the reproductive mode. When honey was supplied, the ratio of female-to-male in the progeny of WFT produced under sexual reproductive mode increased significantly in the presence of IFT. On pepper seedlings, mixed populations were more favorable to the population development of IFT at the seedling stage, but more favorable to WFT at the flowering stage. Conclusions: In the early stage of WFT invasion and colonization, the emergence of flowering and honey (nectar) sources may have a positive effect on the population development of WFT.

The Hessian fly, Mayetiola destructor (Say) (Diptera: Cecidomyiidae), is a destructive wheat pest worldwide and an important alien species in China. Based on 258 distribution records and nine environmental factors of the Hessian fly, we predicted the potential distribution area in China under three current and future (2050s and 2070s) climate change scenarios (RCP2.6, RCP4.5, and RCP8.5) via the optimized MaxEnt model. Under the current climate conditions, the suitable distribution areas of the Hessian fly in China were 25–48° N, 81–123° E, and the total highly suitable distribution area is approximately 9.63 × 105 km2, accounting for 9.99% of the total national area. The highly suitable areas are mainly located in northern Xinjiang and central and eastern China. With the rising global temperatures, except for the high-suitable areas under the RCP8.5 scenario, most potential geographic distribution areas would expand in the future. The minimum temperature in February (tmin-2), precipitation in March (prec-3), maximum temperature in November (tmax-11), and precipitation seasonality (bio-15) are important factors that affect the potential geographic distribution of the Hessian fly. This study provides an important reference and empirical basis for management of the Hessian fly in the future.

Crop resistance and biological control are both considered efficient and environmentally friendly methods of sustainable pest control. In this study, we aimed at investigating the direct influence of four wheat lines with varying resistance level on the life-history traits of the greenbug, Schizaphis graminum, and the mediational effect on the functional response of a predatory ladybird, Propylaea japonica, under laboratory conditions. Results showed that the aphid fitness was the lowest for aphids that had been feeding on wheat line ‘98-10-19’ for one year. These aphids had the longest development time, and least adult mass, minimal mean relative growth rate, and lowest reproductive fitness. In contrast, the aphids that fed on wheat line ‘98-10-30’ were the fittest, with the shortest development time and highest levels of reproductive fitness. The predatory activities of the ladybeetle, especially the adult male significantly decreased following the consumption of aphids belonging to the ‘98-10-19’-acclimated population. However, there were no significant differences in predatory efficiency (net attack frequency) among the four aphid acclimated populations. Our results showed that the wheat line ‘98-10-19’ has a relative higher resistance to S. graminum than the other three wheat lines, which could further decrease the amount of prey available for consumption. However, the ecological effect of the resistance of ‘98-10-19’ to S. graminum posed no negative influence on the biocontrol potential of P. japonica to these aphids, as their predatory efficiency increases at the fourth instar larvae phase.

Insect cuticular hydrocarbons (CHCs) serve as important intersexual signaling chemicals and generally show variation between the sexes, but little is known about the generation of sexually dimorphic hydrocarbons (SDHCs) in insects. In this study, we report the molecular mechanism and biological significance that underlie the generation of SDHC in the German cockroach Blattella germanica. Sexually mature females possess more C29 CHCs, especially the contact sex pheromone precursor 3,11-DimeC29. RNA interference (RNAi) screen against the fatty acid elongase family members combined with heterologous expression of the genes in yeast revealed that both BgElo12 and BgElo24 were involved in hydrocarbon (HC) production, but BgElo24 is of wide catalytic activities and is able to provide substrates for BgElo12, and only the female-enriched BgElo12 is responsible for sustaining female-specific HC profile. Repressing BgElo12 masculinized the female CHC profile, decreased contact sex pheromone level, and consequently reduced the sexual attractiveness of female cockroaches. Moreover, the asymmetric expression of BgElo12 between the sexes is modulated by sex differentiation cascade. Specifically, male-specific BgDsx represses the transcription of BgElo12 in males, while BgTra is able to remove this effect in females. Our study reveals a novel molecular mechanism responsible for the formation of SDHCs and also provide evidences on shaping of the SDHCs by sexual selection, as females use them to generate high levels of contact sex pheromone.

The fall armyworm, Spodoptera frugiperda (Lepidoptera: Noctuidae), is a major invasive pest that seriously threatens world agricultural production and food security. Microorganisms play a crucial role in the growth and development of insects. However, the diversity and dynamics of gut microbes with different developmental stages, environmental habitats, and diets in S. frugiperda remain unclear. In this study, we found the changes of the microbiome of S. frugiperda across their life stages, and the bacteria were dominated by Firmicutes and Proteobacteria. The community composition of the egg stage was quite different from other developmental stages, which had the highest community diversity and community richness, and was dominated by Proteobacteria. The bacterial community compositions of male and female adults were similar to those of early larvae stage (L1–L2), and operational taxonomic units (OTUs) with abundant content were Enterococcus and Enterobacteriaceae bacteria, including Enterobacteria, Klebsiella, Pantoea, and Escherichia. The third instar larvae (L3) mainly consist of Enterococcus. The late stage larvae (L4–L6) harbored high proportions of Enterococcus, Rhodococcus, and Ralstonia. There was no significant difference in gut microbial composition between field populations and laboratory populations in a short period of rearing time. However, after long-term laboratory feeding, the gut microbial diversity of S. frugiperda was significantly reduced. Enterococcus and Rhodococccus of S. frugiperda feeding on maize showed higher relative proportion, while the microbial community of S. frugiperda feeding on artificial diet was composed mainly of Enterococcus, with a total of 98% of the gut microbiota. The gene functions such as metabolism, cell growth and death, transport and catabolism, and environmental adaptation were more active in S. frugiperda feeding on corn than those feeding on artificial diet. In short, these results indicate that developmental stage, habitat, and diet can alter the gut bacteria of S. frugiperda, and suggest a vertical transmission route of bacteria in S. frugiperda. A comprehensive understanding of gut microbiome of S. frugiperda will help develop novel pest control strategies to manage this pest.

The epidemiology of Tomato chlorosis virus (ToCV) in China is closely associated with its vector whitefly, Bemisia tabaci (Gennadius) MED. However, the transmission characteristics of ToCV by B. tabaci MED remain poorly understood. In this study, we analyzed: 1) the horizontal and vertical transmission of ToCV by B. tabaci MED whiteflies; 2) the acquisition of ToCV by male and female B. tabaci MED whiteflies after different feeding durations; 3) the transmission efficacy of viruliferous male and female B. tabaci MED whiteflies after different inoculation access periods (IAPs); 4) the retention of ToCV by viruliferous male and female B. tabaci MED whiteflies after a 48 h acquisition access period (AAP); and 5) the effects of ToCV on host choice of healthy or ToCV-infected tomato plant of viruliferous and non-viruliferous B. tabaci MED at different time points. Our results showed that: 1) viruliferous males could not transfer ToCV to non-viruliferous females, and vice versa, viruliferous females could not pass on ToCV to non-viruliferous males. ToCV could not be detected in the F1 generation adults; 2) ToCV could be detected within 4.0% of females or males after a 20 min AAP; 3) ToCV could be detected in 33.3% of tomato plants inoculated by 10 viruliferous males or females with IAPs of 20 or 30 min; 4) the maximum retention time in females was 7 and 5 days in males; and 5) non-viruliferous B. tabaci MED did not show a preference for ToCV-infected tomato plants or healthy tomato plants. However, viruliferous B. tabaci MED whiteflies did prefer to settle on healthy tomato plants over ToCV-infected tomato plants. These findings will be helpful to better understand the epidemiology of the recently emerged plant virus, ToCV, in tomato fields in China.

The thermal tolerance of Bemisia tabaci MED, an invasive whitefly species with worldwide distribution, plays an important role in its ecological adaptation during the invasion process. Heat-shock proteins (HSPs) are closely related to heat resistance. In this study, 33 Hsps (BtaHsps) were identified based on sequenced genome of B. tabaci MED belonging to six HSP families, among which 22 Hsps were newly identified. The secondary structures of a further 22 BtaHsps were also predicted. The results of RT-qPCR showed that heat shock could affect the expression of 14 of the 22 Hsps newly identified in this study. Among them, the expression level of six Hsps increased under 42 °C treatment. As the unstudied gene, BtaHsp90A3 had the highest increase rate. Therefore, BtaHsp90A3 was chosen for the RNAi test, and silencing BtaHsp90A3 by RNAi decreased the survival rate of adult B. tabaci at 42 °C. The results indicated that only a few Hsps were involved in the thermal tolerance of host whitefly although many Hsps would response under heat stress. This study conducted a more in-depth and comprehensive identification that demonstrates the evolutionary relationship of BtaHsps and illustrates the response of BtaHsps under the influence of thermal stress in B. tabaci MED.

Plants have evolved a number of different chemical defenses, covering nearly all classes of (secondary) metabolites, that represent a major barrier to herbivory: some are constitutive; others are induced after attacks from herbivores (HIPVs) and may elicit the attraction of predators and parasitoids. Here, we studied how the female solitary endoparasitoid Aphelinus varipes responds to plant and host aphid volatiles in a series of experiments on five commercially important vegetables that were either healthy or infested with the aphid Myzus persicae: chili pepper, eggplant, crown daisy, Chinese cabbage and cabbage. The results for the olfactory responses of A. varipes showed that the presence of M. persicae increased the attraction of the endoparasitoid to the infested plants. In a second experiment, volatiles from highly attractive and repellent plants were obtained via headspace collection to investigate volatiles from healthy and aphid-damaged plants. The results for the differences in volatile profiles in response to aphid infestation in chili pepper cultivar were dominated by the volatile blends, including α-pinene, decanal and phthalic acid, while in cabbage they were dominated by isophorone. Moreover, when HIPVs with different concentrations were compared, α-pinene at a dose rate of 100 ng/μL attracted more parasitoids, and the comparison was useful to understand the mechanisms of plant secondary volatiles during aphid infestation and to provide new resources to control this insect pest. Overall our study shows how HIPVs can bolster tritrophic interactions by enhancing the attractiveness of parasitoids.

Cuticle sclerotization is critical for insect survival. Laccase2 (Lac2) is a phenol oxidase that plays a key role in cuticle formation and pigmentation in a variety of insects. However, the function of Lac2 in whitefly, Bemisia tabaci, remains unclear. In this study, we identified a BtLac2 gene in B. tabaci MEAM1 and found that BtLac2 was expressed in all stages. It was highly expressed in the egg stage, followed by nymph and adult. Moreover, the expression of BtLac2 was higher in the cuticle than in other tissues. Knockdown of BtLac2 in nymphs produced thinner and fragile cuticles, which significantly increased the mortality rate, extended the development duration of nymphs, and decreased the emergence rate of adults. This result demonstrates that BtLac2 plays an important role in the cuticle hardening of B. tabaci and suggests a potential management strategy using RNAi to knock down BtLac2 expression.

It is well known that high temperature, a typically negative environmental factor, reduces the parasitism of a parasitoid. Generally, high temperature causes the rapid overproduction of reactive oxygen species (ROS) in organisms, and antioxidative enzymes participate in the process of resisting environmental stress by eliminating excess ROS. In the present study, we identify two superoxide dismutase (SOD), one catalase (CAT), six peroxidases (POD), and five glutathione-S-transferase (GST) genes; and the survival rate and antioxidative enzyme patterns under short-term high temperature exposure of the parasitoid wasp, A.asychis, are examined. Survival results of A.asychis reveal that females show higher thermal tolerance than males. Under short-term high-temperature exposure, in females, the expression levels of most antioxidant enzyme genes decreased first and then increased to a peak at 41 °C, while only the expression of AasyGST4 showed a continuous increase. In males, the expression patterns of most antioxidant enzyme genes fluctuated and reached a maximum at 41 °C. Moreover, the expression levels of the majority of antioxidant enzyme genes were higher in females than in males. In addition, at temperatures of and above 35 °C, the activities of these four antioxidant enzymes were induced. The results show that the antioxidant enzymes confer thermo-tolerance to A. asychis against lethal thermal stress. Our observations enrich the understanding of the response mechanism to high-temperature assaults of A. asychis.

The decision to delay or cancel spraying insecticides against pest aphids is dependent on the ratio of prey/predator, which reflects how well the predator can suppress the aphid population increase in the field. It is challenging to estimate the ratio of prey/predator due to the multiple factors involved in the interaction between prey and predator. Cotton aphid (Aphis gossypii Glover) is a serious pest, widely distributed in cotton-growing areas around the world. We combined different ratios of aphids with aphid oligophagous ladybird beetles (Coccinella septempunctata Linnaeus) under laboratory and garden conditions to investigate the critical threshold for prey/predator which effectively reduced the cotton aphid population increase. Two kinds of modeling were developed to understand the relationships between the ratio of prey/predator and the PGR (population growth rate), and with the effectiveness of biocontrol (EBC). We found the critical values of PGR should be less than −0.0806 (predators artificially released after 5 days) and then less than −0.075 (predators released after 10 days) if EBC is less than 50%. We recommend that the ratio of prey/predator should be less than 450 for the effective biocontrol of cotton aphids at the cotton seedling stage. These values can be reference indices for the management of aphids in mid-summer.

Praying mantises are distributed all over the world. Though some Mantodea mitogenomes have been reported, an evolutionary genomic and phylogenetic analysis study lacks the latest taxonomic system. In the present study, four new mitogenomes were sequenced and annotated. Deroplatys truncate, D. lobate, Amorphoscelis chinensis and Macromantis sp. belong to Deroplatyidae, Amorphoscelidae and Photinaidae family, respectively. Our results indicated that the ATP8 gene may be lost in D. truncate and D. lobata mt genome, and four tRNA genes have not been found in D. truncate, D. lobata and Macromantis sp. A dN/dS pair analysis was conducted and it was found that all genes have evolved under purifying selection. Furthermore, we tested the phylogenetic relationships between the eight families of the Mantodea, including 35 species of praying Mantis. Based on the complete mitochondrial genome data, it was also suggested as sister to Deroplatyidae + Mantidae, Metallyticus sp., the only representative of Metallyticidae, is sister to the remaining mantises. Our results support the taxonomic system of Schwarz and Roy and are consistent with previous studies.

The diamondback moth Plutella xylostella is a serious pest of crucifers. It has high reproductive potential and is resistant to many insecticides. Typically, the last-instar larvae of P. xylostella, before pupation, move to the lower or outer plant leaves to make a loose silk cocoon and pupate inside for adult formation. To better understand this pivotal stage we studied the cocoon-spinning behavior of P. xylostella and measured three successive phases by video-recording, namely the selection of a pupation site, spinning a loose cocoon and padding the scaffold cocoon. Subsequently, we cloned three fibroin genes related to cocoon production, i.e., fibroin light chain (Fib-L), fibroin heavy chain (Fib-H), and glycoprotein P25. A spatio-temporal study of these three fibroin genes confirmed a high expression in the silk glands during the final larval instar silk-producing stage. In parallel, we did an exogenous treatment of the insect molting hormone 20-hydroxyecdysone (20E), and this suppressed fibroin gene expression, reduced the normal time needed for cocoon spinning, and we also observed a looser cocoon structure under the scanning electron microscope. Hence, we demonstrated that the expression levels of key genes related to the synthesis of 20E [the three Halloween genes Spook (Spo), Shadow (Sad), and Shade (Shd)] decreased significantly during spinning, the expression of the 20E receptor (EcR and USP) was significantly lower during spinning than before spinning, and that the expression levels of CYP18-A1 related to 20E degradation were significantly up-regulated during spinning. The significance of the cocoon and the effects of 20E on the cocoon-spinning behavior of P. xylostella are discussed.

Insect short neuropeptide F (sNPF), an ortholog of prolactin-releasing peptide of invertebrates, regulates diverse biological processes, including feeding, olfaction, locomotion, and sleep homeostasis in insects. However, its function is still unclear in an important model insect and agricultural pest, the pea aphid (Acyrthosiphon pisum). Here, we investigated short neuropeptide F (ApsNPF) and its receptor (ApsNPFR) in A. pisum. The sNPF gene contains three exons and two long introns. In addition, the genome contains a single sNPF receptor with seven transmembrane domains. Stage- and tissue-specific transcript profiling by qRT-PCR revealed that ApsNPF and ApsNPFR were mainly expressed in the central nervous system. The receptor was also detected in antennae, midgut, and integument. The highest expression levels were found in first instar nymphs compared to other developmental stages. Besides, the starvation-induced pattern indicated that the sNPF network depends on the nutritional state of the insect. An electrical penetration graph showed that probing time and phloem duration of A. pisum on broad bean plants decreased in response to dssNPF and dssNPFR in RNAi assays. sNPF silencing reduced the number of nymphs per female but not aphid survival. We believe that our results advance in-depth knowledge of the sNPF/sNPFR signaling cascade and its place in regulating feeding behavior in insects. In turn, it may contribute to the potential design of new strategies to control aphids, with a focus on the sNPF system.

Bemisia tabaci and the diamondback moth (DBM), Plutella xylostella, are two major cosmopolitan pests that often occur together and cause severe economic losses to cruciferous crops. However, little is known about how they interact with each other. To determine the effects of defense responses induced by the two pests on the biology and population dynamics of the herbivores, we studied the performance and fitness of B. tabaci and DBM when they damaged Chinese kale simultaneously and in different orders. The results showed that DBM pre-infestation shortened the developmental duration, increased longevity, oviposition days, and fecundity of B. tabaci. Meanwhile, the intrinsic rate of increase (r), net reproductive rate (R0) and finite rate of increase (λ) of B. tabaci increased significantly with dual infection as compared with only B. tabaci infestation. In contrast, B. tabaci pre-infestation reduced the longevity and oviposition days of DBM, but the population parameters r, R0, and λ did not vary significantly compared with only DBM infestation. Thus, co-infestation of B. tabaci and DBM was beneficial to the performance of the B. tabaci population. The present findings highlight that B. tabaci has become a dominant competitor when mixing with DBM on the same host plant.

Neuropeptides are the most abundant and diverse signal molecules in insects. They act as neurohormones and neuromodulators to regulate the physiology and behavior of insects. The majority of neuropeptides initiate downstream signaling pathways through binding to G protein-coupled receptors (GPCRs) on the cell surface. In this study, RNA-seq technology and bioinformatics were used to search for genes encoding neuropeptides and their GPCRs in the cowpea aphid Aphis craccivora. And the expression of these genes at different developmental stages of A. craccivora was analyzed by quantitative real-time PCR (qRT-PCR). A total of 40 candidate genes encoding neuropeptide precursors were identified from the transcriptome data, which is roughly equivalent to the number of neuropeptide genes that have been reported in other insects. On this basis, software analysis combined with homologous prediction estimated that there could be more than 60 mature neuropeptides with biological activity. In addition, 46 neuropeptide GPCRs were obtained, of which 40 belong to rhodopsin-like receptors (A-family GPCRs), including 21 families of neuropeptide receptors and 7 orphan receptors, and 6 belong to secretin-like receptors (B-family GPCRs), including receptors for diuretic hormone 31, diuretic hormone 44 and pigment-dispersing factor (PDF). Compared with holometabolous insects such as Drosophila melanogaster, the coding genes for sulfakinin, corazonin, arginine vasopressin-like peptide (AVLP), and trissin and the corresponding receptors were not found in A. craccivora. It is speculated that A. craccivora likely lacks the above neuropeptide signaling pathways, which is consistent with Acyrthosiphon pisum and that the loss of these pathways may be a common feature of aphids. In addition, expression profiling revealed neuropeptide genes and their GPCR genes that are differentially expressed at different developmental stages and in different wing morphs. This study will help to deepen our understanding of the neuropeptide signaling systems in aphids, thus laying the foundation for the development of new methods for aphid control targeting these signaling systems.

Maternal phenotypic regulations between different generations of aphid species help aphids to adapt to environmental challenges. The pea aphid Acyrthosiphon pisum has been used as a biological model for studies on phenotypic regulation for adaptation, and its alternative phenotypes are typically and physiologically based on maternal effects. We have observed an artificially induced and host-related maternal effect that may be a new aspect to consider in maternal regulation studies using A. pisum. Marked phenotypic changes in the cuticular melanization of daughter A. pisum were detected via tyrosine hydroxylase knockdown in the mothers during their period of host plants alternations. This phenotypic change was found to be both remarkable and repeatable. We performed several studies to understand its regulation and concluded that it may be controlled via the dopamine pathway. The downregulation and phenotypes observed were verified and described in detail. Additionally, based on histological and immunofluorescence analyses, the phenotypic changes caused by cuticular dysplasia were physiologically detected. Furthermore, we found that this abnormal development could not be reversed after birth. Transcriptome sequencing confirmed that this abnormal development represents a systemic developmental failure with numerous transcriptional changes, and chemical interventions suggested that transgenerational signals were not transferred through the nervous system. Our data show that transgenerational regulation (maternal effect) was responsible for the melanization failure. The developmental signals were received by the embryos from the mother aphids and were retained after birth. APTH RNAi disrupted the phenotypic determination process. We demonstrate that non-neuronal dopamine regulation plays a crucial role in the transgenerational phenotypic regulation of A. pisum. These results enhance our understanding of phenotyping via maternal regulation in aphids.

Oviposition is an important reproductive behavior that is triggered by mating in insects, and biogenic amines might be involved in its regulation. The effects of biogenic amines on oviposition have only been studied in a few insect species, and the findings to date have not been conclusive. In addition, there are few studies on the effects of biogenic amines on oviposition of the diamondback moth, Plutella xylostella L. Here, we tested how mating and biogenic amines regulate oviposition of P. xylostella by injecting amines and amine receptor antagonists into virgin and mated females and counting the number of eggs laid afterward. Biogenic amines of octopamine and tyramine could induce virgin adults of P. xylostella to lay eggs, while dopamine and serotonin had no such effect on oviposition. Furthermore, the octopamine antagonists mianserin, epinastine, and phentolamine inhibited oviposition by mated females. The tyramine antagonist yohimbine, dopamine antagonist SCH23390, and serotonin antagonist ketanserin did not block oviposition by mated females, and octopamine and tyramine-inducing oviposition by virgin females could be inhibited by the octopamine antagonists mianserin and epinastine instead of the tyramine antagonist yohimbine. We conclude that octopamine and its receptors are involved in mating-triggered oviposition in P. xylostella, while tyramine acts as a subsidiary. Further, the inducing effect of tyramine on oviposition is achieved via octopamine receptors instead of tyramine receptors. This experiment is helpful to further understand the role of biogenic amines in mating regulation and to provide a new strategy for controlling P. xylostella.

Free amino acids in the phloem sap are the dominant nitrogen source for aphids, but their availability is usually poor. Although some studies have explored the effect of dietary amino acid restriction on aphid performance, little is known about the molecular basis of these effects. Here, we examined the performance and transcriptome of the green peach aphid, Myzus persicae, fed a standard diet (Control diet) or a diet containing 50% of the total amino acids of the Control diet (Half diet). Aphid weight and fecundity were significantly reduced in the Half diet group. Transcriptomic analysis showed that a total of 1460 genes were differentially expressed between the groups were fed on the two diets, which many of them were associated with nutrient and energy metabolism. When feeding on the Half diet, aphids upregulated genes associated with the amino acid biosynthetic pathway (predominantly amino acid biosynthesis genes and some amino acid transporter genes) as well as the cysteine and serine protease genes. Furthermore, these aphids displayed increased expression of genes associated with glycolysis, which could generate intermediates for de novo amino acid biosynthesis. Consistent with this, elevated glucose levels were observed in aphids in the Half diet group. Additionally, the expression levels of several genes associated with hormonal signaling pathway were altered. Several genes related to juvenile hormone and insulin-like peptide (ILP) signaling were downregulated, including Krüppel homolog 1 (Kr-h1) and insulin-like peptide 5 (Ilp5), respectively. In contrast, several genes related to ecdysone signaling were upregulated including broad-complex core protein (Br-c) and shade (Shd). Despite their poor performances, M. persicae adapted to dietary restriction of amino acids, through upregulation of genes involved in amino acid biosynthesis, glycolysis, and protein degradation, as well as by altering the expression level of genes involved in hormone signaling pathways.

Animals have developed numerous strategies to contend with environmental pressures. We observed that the same adaptation strategy may be used repeatedly by one species in response to a certain environmental challenge. The ladybird Harmonia axyridis displays thermal phenotypic plasticity at different developmental stages. It is unknown whether these superficially similar temperature-induced specializations share similar physiological mechanisms. We performed various experiments to clarify the differences and similarities between these processes. We examined changes in the numbers and sizes of melanic spots in pupae and adults, and confirmed similar patterns for both. The dopamine pathway controls pigmentation levels at both developmental stages of H. axyridis. However, the aspartate-β-alanine pathway controls spot size and number only in the pupae. An upstream regulation analysis revealed the roles of Hox genes and elytral veins in pupal and adult spot formation. Both the pupae and the adults exhibited similar morphological responses to temperatures. However, they occurred in different body parts and were regulated by different pathways. These phenotypic adaptations are indicative of an effective thermoregulatory system in H. axyridis and explains how insects contend with certain environmental pressure based on various control mechanisms.

Plutella xylostella (L.) (Plutellidae) is an important agricultural pest throughout the world. In this study, the morphology of antennal and mouthpart sensilla in the larvae and adults of P. xylostella (L.) was observed by using a scanning electron microscope. The larval antennae possess six sensilla basiconica, two sensilla chaetica and one sensillum styloconicum. Larval mouthparts possess six types of sensilla: sensilla chaetica, sensilla digitiformia, sensilla epipharyngeal, sensilla basiconica, sensilla styloconica and sensilla placodea. In the adult, seven types of sensilla are found on the antennae in males and six types of sensilla (sensilla basiconica absent) occur in females. Sexual dimorphism is also found in the number and size of these sensilla on the antennae of adults. We describe for the first time the five types of sensilla on the mouthparts of the adult of P. xylostella. This study provides useful information for further research into the function of these sensilla, and better understanding the behavioral mechanisms involved in pest control.

Abstract The maternal effects of the English grain aphid, Sitobion avenae on offspring phenotypes and performance on wheat varieties with different resistance traits were examined. We found that both conditioning wheat varieties(the host plant for over 3 months) and transition wheat varieties affected the biological parameters of aphid offspring after they were transferred between wheat varieties with different resistance traits. The conditioning varieties affected weight gain, development time (DT), and the intrinsic rate of natural increase (rm), whereas transition varieties affected the fecundity, rm, net reproductive rate, and fitness index. The conditioning and transition wheat varieties had significant interaction effects on the aphid offspring’s DT, mean relative growth rate, and fecundity. Our results showed that there was obvious maternal effects on offspring when S. avenae transferred bwteen wheat varieties with different resistance level, and the resistance traits of wheat varieties could induce an interaction between the conditioning and transition wheat varieties to influence the growth, development, reproduction, and even population dynamics of S. avenae. The conditioning varieties affected life-history traits related to individual growth and development to a greater extent, whereas transition varieties affected fecundity and population parameters more.

Benzoxazinoids are important secondary metabolites in gramineae plants and have inhibitory and toxic effects against a wide range of herbivore pests. However, the relationship between benzoxazinoid level and plant resistance to aphids remains controversial. In this study, we investigated the relationship between benzoxazinoids composition and concentration in wheat leaves and the resistance to the grain aphid Sitobion avenae. Overall, six benzoxazinoids were detected and identified by mass spectrometry based metabolites profiling, including three lactams, two hydroxamic acids, and one methyl derivative. The constitutive levels of these benzoxazinoids were significantly different among the wheat varieties/lines. However, none of these benzoxazinoids exhibited considerable correlation with aphid resistance. S. avenae feeding elevated the level of 2-O-β-D-glucopyranosyloxy-4,7-dimethoxy-(2H)-1,4-benzoxazin-3(4H)-one (HDMBOA-Glc) and reduced the level of 2-O-β-D-glucopyranosyloxy-4-hydroxy-7-(2H)-methoxy-1,4-benzoxazin-3(4H)-one (DIMBOA-Glc) in some of the wheat varieties/lines. Moreover, aphid-induced level of DIMBOA-Glc was positively related with callose deposition, which was closely associated with aphid resistance. Wheat leaves infiltrated with DIMBOA-Glc caused a noticeable increase of callose deposition and the effect was in a dose dependent manner. This study suggests that the constitutive level of benzoxazinoids has limited impact on S. avenae. Aphid feeding can affect the balance of benzoxazinoids metabolism and the dynamic level of benzoxazinoids can act as a signal of callose deposition for S. avenae resistance. This study will extend our understanding of aphid–wheat interaction and provides new insights in aphid-resistance wheat breeding.

In insects, neuropeptides and their receptors not only play a critical role in insect physiology and behavior but also are the potential targets for novel pesticide discoveries. Aphidius gifuensis is one of the most important and widespread aphid parasitoids, and has been successfully used to control aphid. In the present work, we systematically identified neuropeptides and their receptors from the genome and head transcriptome of A. gifuensis. A total of 35 neuropeptide precursors and 49 corresponding receptors were identified. The phylogenetic analyses demonstrated that 35 of these receptors belong to family-A, four belong to family-B, two belong to leucine-rich repeat-containing GPCRs, four belong to receptor guanylyl cyclases, and four belong to receptor tyrosine kinases. Oral ingestion of imidacloprid significantly up-regulated five neuropeptide precursors and four receptors whereas three neuropeptide precursors and eight receptors were significantly down-regulated, which indicated that these neuropeptides and their receptors are potential targets of some commercial insecticides. The RT-qPCR results showed that dopamine receptor 1, dopamine receptor 2, octopamine receptor, allatostatin-A receptor, neuropeptides capa receptor, SIFamide receptor, FMRFamide receptor, tyramine receptor and short neuropeptide F predominantly were expressed in the head whilst the expression of ion transport peptide showed widespread distribution in various tissues. The high expression levels of these genes suggest their important roles in the central nervous system. Taken together, our study provides fundamental information that may further our understanding of neuropeptidergic signaling systems in the regulation of the physiology and behavior of solitary wasps. Furthermore, this information could also aid in the design and discovery of specific and environment-friendly insecticides.

Insect cuticular hydrocarbons (CHCs) are organic compounds of the surface lipid layer, which function as a barrier against water loss and xenobiotic penetration, while also serving as chemical signals. Plasticity of CHC profiles can vary depending upon numerous biological and environmental factors. Here, we investigated potential sources of variation in CHC profiles of Nilaparvata lugens, Laodelphax striatellus and Sogatella furcifera, which are considered to be the most important rice pests in Asia. CHC profiles were quantified by GC/MS, and factors associated with variations were explored by conducting principal component analysis (PCA). Transcriptomes were further compared under different environmental conditions. The results demonstrated that CHC profiles differ among three species and change with different developmental stages, sexes, temperature, humidity and host plants. Genes involved in cuticular lipid biosynthesis pathways are modulated, which might explain why CHC profiles vary among species under different environments. Our study illustrates some biological and ecological variations in modifying CHC profiles, and the underlying molecular regulation mechanisms of the planthoppers in coping with changes of environmental conditions, which is of great importance for identifying potential vulnerabilities relating to pest ecology and developing novel pest management strategies.

The green peach aphid, Myzus persicae Sulzer (Hemiptera: Aphididae), a polyphagous insect pest is a major threat to a wide range of crops worldwide. Aiming to evaluate the life history traits of M. persicae, feeding on different host plants, we used five vegetables: cabbage, Brassica oleracea (Brassicaceae); chinese cabbage, B. rapa (Brassicaceae); chili pepper, Capsicum annum (Solanaceae); crown daisy, Chrysanthemum coronarium (Asteraceae); and eggplant, Solanum melongena (Solanaceae). TWOSEX-MSchart software was used for the statistical analysis about the age-stage, two-sex life table theory. The highest fecundity (69.65 individuals) rate of M. persicae, intrinsic rate of increase (r = 0.425 d−1), finite rate of increase (λ = 1.531 d−1), net reproductive rate (R0 = 69.65 offspring), and shortest mean generation time (T = 9.964 d) were recorded on the chili pepper plant. Whereas, lower fitness occurred on cabbage. The findings attained from population growth parameters indicate that chili pepper is the most susceptible plant, while cabbage is resistant to aphids. Population projection results also supported this statement, as the final total population size on cabbage was significantly lower than other plants. The reported information would be useful for devising integrated pest management programs, particularly those involving M. persicae. This information also suggests the adaptability of M. persicae causing economic damage to these vegetable cultivars.

Abstract Olfaction and gustation play critical roles during the host-location search process of insects. Several chemosensory receptor genes are thought to be involved in providing specificity to the olfactory sensory neuron responses. The aphid endoparasitoid, Aphidius gifuensis, has been used as a biological control agent against a variety of aphid species; this parasitoid is able to detect its target host(s) effectively during the parasitic process. To understand the mechanism of host detection in A. gifuensis, we assembled specific antennal transcriptomes of each sex through next generation sequencing technology to identify the major chemosensory receptor genes. Using a bioinformatics screen, we identified 100 olfactory receptors candidates (62 odorant receptors, 15 gustatory receptors, and 23 ionotropic receptors) from the sex-specific antennal transcriptome. In addition, combining with the demonstrated functions of chemosensory genes in other insects, the sex-, tissue-, and host-specific expression profile of chemosensory genes potentially revealed the candidate physiological functions. The identification and expression profile of chemosensory receptor genes in A. gifuensis provide valuable information for understanding and investigating the intraspecific or interspecific chemical communications in the solitary parasitic wasps.

The impact of vector density and timing of infestation on potato were investigated. Healthy potato plants at different growth stages (4, 5, and 7 weeks after germination) were exposed separately to four different B. cockerelli densities (0, 5, 20, and 40 psyllids per cage) in field cages and Zebra chip (ZC) symptoms, leaf photosynthetic rates, tuber yield, and total nonstructural carbohydrate accumulation in leaves and tubers of healthy and B. cockerelli-infested plants were monitored. Potato psyllid nymph and egg populations reached a seasonal peak at 6 weeks after the exposure to insect. Younger plants at 4-week growth stage after germination were more susceptible to B. cockerelli infestation and ZC expression than older plants. As few as five B. cockerelli adults were enough to transmit the ZC pathogen and cause ZC expression both in foliage and tuber. At the density of 20 psyllids per cage, more than 50% of plants showed ZC symptoms in tubers. Furthermore, B. cockerelli infestation reduced leaf photosynthesis rates (Pn), resulting in less starch and more reducing sugars in tubers, and hence reduced tuber weight and yield, especially when psyllid infestation occurred at the early growth stages. The results indicate that early B. cockerelli infestation of younger plants was associated with more severe ZC expression in both foliage and tubers, leading to earlier dead plants. The data suggest that strategies for controlling B. cockerelli during early potato crop development could thus lessen the severity of ZC development.

Wingless forms of aphids are relatively sedentary, and have a limited ability to migrate or disperse. However, they can drop off hosts or walk away if disturbed, or their food quality or quantity become deteriorated. Earlier, we found that the pea aphid, Acyrthosiphon pisum (Harris, 1776), could use differed strategies to escape danger and locate new host plants. To determine the mechanisms behind the different strategies, we undertook a series of studies including the aphids' host location, energy reserves under starvation, glycogenesis, sugar assimilation, olfactory and probing behaviors. We found that in our controlled laboratory conditions, one strain (local laboratory strain) moved longer distances and dispersed wider ranges, and correspondingly these aphids assimilated more sugars, synthesized more glycogen, and moved faster than another strain (collected from Gansu Province, northwestern China). However, the latter strain could locate the host faster, probed leaves more frequently, and identified plant leaves more accurately than the former strain after they were starved. Our results explained how flightless or wingless insects adapt to fit biotic and abiotic challenges in the complex processes of natural selection.

Plants emit volatile organic compounds (VOCs) in response to herbivore attack. VOCs emitted from the Chinese cabbage cultivars in response to the damage by the green peach aphid, Myzus persicae, were unknown. Using a solid-phase microextraction-based headspace collection method, we investigated and compared the emissions of VOCs from seven Chinese cabbage cultivars (Qibao, Qingan 80, Lvlong, Yuanbao, Qingan 70, Jinlv, and Lvqiu 66) in response to M. persicae attack. Our results showed that the VOCs emitted from the cultivars Qingan 80 and Yuanbao differed significantly from the other cultivars in response to the attraction of wingless M. persicae. Most importantly, out of the 27 detected compounds, α-caryophyllene was detected only in Qingan 80 and Qibao, but not in the other five cultivars. Among the compounds detected, 2 monoterpene and 12 terpenes were predominant in all cabbage cultivars. Furthermore, the wingless M. persicae showed preference to Qingan 80 while it had the highest nitrogen content among the tested cultivars. Moreover, we found a remarkable relationship among M. persicae attraction, plant nitrogen content, and total volatile emissions. Nitrogen content of the plants has a significant impact on volatile emission and preference behavior of M. persicae. Our results indicate that the wingless M. persicae were efficient in their interspecific host selection with an ability to distinguish plant cultivar differences by leaf nitrogen content. This study will be helpful in understanding aphid host selection, and sets a stage to further study the attractant-based integrated aphid management programs.

The pea aphid, Acyrthosiphon pisum (Harris), shows body color shifting from red to pale under starvation in laboratory conditions. These body color changes reflect aphid’s adaptation to environmental stress. To understand the color-shifting patterns, the underlying mechanism and its biological or ecological functions, we measured the process of A. pisum’s body color shifting patterns using a digital imagery and analysis system; we conducted a series of biochemical experiments to determine the mechanism that causes color change and performed biochemical and molecular analyses of the energy reserves during the color shifting process. We found that the red morph of A. pisum could shift their body color to pale red, when starved; this change occurred rapidly at a certain stress threshold. Once A. pisum initiated the process, the shifting could not be stopped or reversed even after food was re-introduced. We also discovered that the orange-red pigments may be responsible for the color shift and that the shift might be caused by the degradation of these pigments. The carbohydrate and lipid content correlated to the fading of color in red A. pisum. A comparative analysis revealed that these reddish pigments might be used as backup energy. The fading of color reflects a reorganization of the energy reserves under nutritional stress in A. pisum; surprisingly, aphids with different body colors exhibit diverse strategies for storage and consumption of energy reserves.

The integrated pest management (IPM) strategy was developed and used in combination with pesticides and beneficial biological control agents. To further develop IPM efficiency, it is important to evaluate the side effects of pesticides on biological control agents. Aphidius gifuensis is one of the most important aphid natural enemies and has been successfully used to control Myzys persicae and other aphid species. Imidacloprid (IMD) is a popular pesticide used worldwide and is highly toxic to non-target arthropods. Here, we investigated the short-term sublethal toxicity of IMD in Aphidius gifuensis and its impact on the biological performance and gene expression of this parasitoid. We found that sublethal IMD doses had a significant negative effect on the life history traits of female A. gifuensis, including shortening the lifespan and lowering parasitic capacity. Moreover, exposure to sublethal IMD also adversely affected the response of A. gifuensis to aphid-infested plant volatiles. Based on the transcriptome analysis, we found that the exposure to sublethal IMD doses significantly affected expression of genes involved in the central nervous system, energy metabolism, olfactory, and detoxification system of A. gifuensis. RT-qPCR also revealed that short term expose to sublethal IMD doses significantly induced the gene expression of genes related to the central nervous system (nAChRa7, nAChRa9, TbH, OAR1, NFR, TYR, and DAR1), olfactory system (OR28 and IR8a1), and detoxification system (CYP49p3, CYP6a2, and POD), while it suppressed the expression of genes involved in the central nervous system (nAChRa4 and nAChRb1), olfactory system (Orco1, IR8a2, and GR1), and detoxification system (GST2). Furthermore, exposure to sublethal doses of IMD also significantly increased the activities of CarEs and POD, whereas we observed no influence on the activities of CAT, GST, and SOD. Our results indicate that sublethal IMD doses might adversely affect the biological performance of A. gifuensis by altering gene expression related to the function of olfactory, nervous, energy metabolism, and detoxification systems. Thus, how the use of pesticides directly affect insect population should be considered when used in conjunction with natural pest parasitoids in IPM strategies.

Herbivores respond differently to the level of plant diversity encountered. Bemisia tabaci Gennadius (Hemiptera: Aleyrodidae) are highly polyphagous herbivores which cause considerable damage to various crops. Herein, we reared this species both in polyculture and monoculture, including preferred and less preferred host plants such as Chinese cabbage (Brassica rapa L.), tomato (Solanum lycopersicum L.), kidney bean (Phaseolus vulgaris L.) and summer squash (Cucurbita pepo L.). Trends in survival and oviposition were recorded, and impact of plants on growth and development of B. tabaci were studied, particularly in terms of detoxification and digestive enzymatic activity in the insects. We found that the survival rate was the highest in Chinese cabbage monoculture treatment. Further, the egg numbers on individual species in the polyculture generally reflected numbers on the same plant species in monoculture. However, more eggs were observed in each of the four plant species tested in the context of polyculture. The activity of superoxide dismutases (SOD) and alkaline phosphatase (AKP) in B. tabaci fed in a choice situation were significantly lower than those fed with tomato monoculture, indicating a dilution of toxicity with a multi-plant diet compared with less preferred host plant diet. Also, the survival rate of B. tabaci in monoculture was negatively correlated with SOD amount of whitefly. In the plants attacked by whiteflies, the activity of polyphenol oxidase (PPO) and catalase (CAT) in Chinese cabbage was lower in polyculture than in the monoculture. These results implied that multi-plant treatments contained fewer secondary metabolite substances and might be less toxic to polyphagous herbivores. As such, the work herein contributes knowledge relevant for more effective control and management of B. tabaci.

Symbionts contribute nutrients that allow insects to feed on plants. The whitefly Bemisia tabaci Middle East-Asia Minor 1 (MEAM1) is a polyphagous pest that depends on symbionts to provide key nutrients that are deficient in the diet. Here, we established three whitefly populations on eggplants, cucumbers, and tomatoes and observed that they harbored the same symbiont taxa in different quantities. The amount of the primary symbiont, Portiera, decreased with increasing concentrations of host-plant essential amino acids (EAAs). Whitefly populations transferred to different plant species exhibited fluctuations in Portiera amounts in the first three or four generations; the amount of Portiera increased when whitefly populations were transferred to plant species with lower EAAs proportions. As for the secondary symbionts, the whitefly population of eggplants exhibited lower quantities of Hamiltonella and higher quantities of Rickettsia than the other two populations. The changes of both symbionts’ abundance in whitefly populations after host-plant-shifting for one generation showed little correlation with the EAAs’ proportions of host plants. These findings suggest that host-plant nitrogen nutrition, mainly in the form of EAAs, influences the abundance of symbionts, especially Portiera, to meet the nutritional demands of whiteflies. The results will inform efforts to control pests through manipulating symbionts in insect–symbiont associations.

Natural enemy guilds normally forage for prey that is patchily distributed simultaneously. Previous studies have investigated the influence of conspecific interactions and prey distribution on the functional response of natural enemies. However, little is known about how prey distribution and heterospecific interactions between natural enemies could affect their foraging efficiency. We examined the effects of prey distribution (aggregate and uniform) and heterospecific interactions on the functional response of a predator, Harmonia axyridis (Pallas) (Coleoptera: Coccinellidae) and a parasitoid, Aphidius gifuensis Ashmead (Hymenoptera: Braconidae) to the green peach aphid, Myzus persicae (Sulzer) (Hemiptera: Aphididae). Type II functional responses were observed in all experiments. Functional response curves of single H. axyridis or A. gifuensis were higher in the aggregate treatment than in the uniform treatment when aphid densities were between 40–180 or 70–170, respectively. When comparing between aggregate and uniform treatments with the heterospecific enemy occurrence, no differences were found in the parasitism efficiency of A. gifuensis, while H. axyridis consumed more aphids in the aggregate treatment than in the uniform treatment when aphid densities were between 50–230. The functional response of individual H. axyridis was not affected by A. gifuensis under two aphid distributions. However, the functional response of a single A. gifuensis and the treatment when A. gifuensis concurrently with H. axyridis overlapped in uniform treatment of above approximately 150 aphids. Our results indicate that the predation rate of H. axyridis was affected by aphid distribution, but was not affected by heterospecific interactions. The parasitism rate of A. gifuensis was affected by aphid distribution, and by heterospecific interactions in both the aggregate and uniform treatments. Thus, to optimize the management efficiency of M. persicae, the combined use of H. axyridis and A. gifuensis should be considered when M. persicae is nearly uniformly distributed under relatively high density.

In natural ecological systems, plants are often simultaneously attacked by both insects and pathogens, which can affect each other’s performance and the interactions can be extended to higher trophic levels, such as parasitoids. The English grain aphid (Sitobion avenae) and powdery mildew (Blumeria graminis f. sp. tritici) are two common antagonists that pose a serious threat to wheat production. Numerous studies have investigated the effect of a single factor (insect or pathogen) on wheat production. However, investigation on the interactions among insect pests, pathogens, and parasitoids within the wheat crop system are rare. Furthermore, the influence of the fungicide, propiconazole, has been found to imitate the natural ecosystem. Therefore, this study investigated the effects of B. graminis on the biological performance of grain aphids and the orientation behavior of its endoparasitic wasp Aphidius gifuensis in the wheat system. Our findings indicated that B. graminis infection suppressed the feeding behavior, adult and nymph weight, and fecundity and prolonged the developmental time of S. avenae. We found that wheat host plants had decreased proportions of essential amino acids and higher content of sucrose following aggravated B. graminis infection. The contents of Pro and Gln increased in the wheat plant tissues after B. graminis infection. In addition, B. graminis infection elicited immune responses in wheat: increase in the expression of defense genes, content of total phenolic compounds, and activity of three related antioxidant enzymes. Moreover, co-infection of B. graminis and S. avenae increased the attraction to A. gifuensis compare to that after infestation with aphids alone. In conclusion, our results indicated that B. graminis infection adversely affected the performance of S. avenae in wheat through restricted nutrition and induced defense response. Furthermore, the preference of parasitoids in such an interactive environment might provide an important basis for pest management control.
