Abstract

In this article, we consider nonlocal Hamilton-Jacobi Equations on networks with Kirchhoff type conditions for the interior vertices and Dirichlet boundary conditions for the boundary ones: our aim is to provide general existence and comparison results in the case when the integro-differential operators are of order strictly less than 1. The main originality of these results is to allow these nonlocal terms to have contributions on several different edges of the network. The existence of Lipschitz continuous solutions is proved in two ways: either by using the vanishing viscosity method or by the usual Perron's method. The comparison proof relies on arguments introduced by Lions and Souganidis. We also introduce a notion of flux-limited solution, nonlocal analog to the one introduced by Imbert and Monneau, and prove that the solutions of the Kirchhoff problem are flux-limited solutions for a suitable flux-limiter. After treating in details the case when we only have one interior vertex, we extend our approach to treat general networks.

Abstract

This document is comprised of a collection of updated preliminary parameters for the key parts of the muon collider. The updated preliminary parameters follow on from the October 2023 Tentative Parameters Report. Particular attention has been given to regions of the facility that are believed to hold greater technical uncertainty in their design and that have a strong impact on the cost and power consumption of the facility. The data is collected from a collaborative spreadsheet and transferred to overleaf.

3. Comparison principle for general nonlocal Hamilton-Jacobi equations with superlinear gradient

Abstract

We obtain the comparison principle for discontinuous viscosity sub- and supersolutions of nonlocal Hamilton-Jacobi equations, with superlinear and coercive gradient terms. The nonlocal terms are integro-differential operators in L\'evy form, with general measures: $x$-dependent, possibly degenerate and without any restriction on the order. The measures must satisfy a combined Wasserstein/Total Variation-continuity assumption, which is one of the weakest conditions used in the context of viscosity approach for this type of integro-differential PDEs. The proof relies on a regularizing effect due to the gradient growth. We present several examples of applications to PDEs with different types of nonlocal operators (measures with density, operators of variable order, L\'evy-It\^o operators).

Abstract

In recent years, Multi-Agent Reinforcement Learning (MARL) has found application in numerous areas of science and industry, such as autonomous driving, telecommunications, and global health. Nevertheless, MARL suffers from, for instance, an exponential growth of dimensions. Inherent properties of quantum mechanics help to overcome these limitations, e.g., by significantly reducing the number of trainable parameters. Previous studies have developed an approach that uses gradient-free quantum Reinforcement Learning and evolutionary optimization for variational quantum circuits (VQCs) to reduce the trainable parameters and avoid barren plateaus as well as vanishing gradients. This leads to a significantly better performance of VQCs compared to classical neural networks with a similar number of trainable parameters and a reduction in the number of parameters by more than 97 \% compared to similarly good neural networks. We extend an approach of K\"olle et al. by proposing a Gate-Based, a Layer-Based, and a Prototype-Based concept to mutate and recombine VQCs. Our results show the best performance for mutation-only strategies and the Gate-Based approach. In particular, we observe a significantly better score, higher total and own collected coins, as well as a superior own coin rate for the best agent when evaluated in the Coin Game environment.

Abstract

A key challenge in particle accelerators is to achieve high peak intensity. Space charge is particularly strong at lower energy such as during injection and typically limits achievable peak intensity. The beam stacking technique can overcome this limitation by accumulating a beam at high energy where space charge is weaker. In beam stacking, a bunch of particles is injected and accelerated to high energy. This bunch continues to circulate, while a second and subsequent bunches are accelerated to merge into the first. It also allows the user cycle and acceleration cycles to be separated which is often valuable. Beam stacking is not possible in a time varying magnetic field, but a fixed field machine such as an Fixed Field Alternating Gradient Accelerator (FFA) does not sweep the magnetic field. In this paper, we describe experimental demonstration of beam stacking of two beams at KURNS FFA in Kyoto University. The momentum spread and intensity of the beam was analysed by study of the Schottky signal, demonstrating stacking with only a slight increase of momentum spread of the combined beams. The intensity of the first beam was, however, significantly reduced. RF knock-out is the suspected source of the beam loss., Comment: 14 pages, 16 figures

Abstract

The International Muon Collider Collaboration (IMCC) [1] was established in 2020 following the recommendations of the European Strategy for Particle Physics (ESPP) and the implementation of the European Strategy for Particle Physics-Accelerator R&D Roadmap by the Laboratory Directors Group [2], hereinafter referred to as the the European LDG roadmap. The Muon Collider Study (MuC) covers the accelerator complex, detectors and physics for a future muon collider. In 2023, European Commission support was obtained for a design study of a muon collider (MuCol) [3]. This project started on 1st March 2023, with work-packages aligned with the overall muon collider studies. In preparation of and during the 2021-22 U.S. Snowmass process, the muon collider project parameters, technical studies and physics performance studies were performed and presented in great detail. Recently, the P5 panel [4] in the U.S. recommended a muon collider R&D, proposed to join the IMCC and envisages that the U.S. should prepare to host a muon collider, calling this their "muon shot". Environmental pollution is escalating due to rapid global development that often prioritizes human needs over planetary health. Despite global efforts to mitigate legacy pollutants, the continuous introduction of new substances remains a major threat to both people and the planet. In response, global initiatives are focusing on risk assessment and regulation of emerging contaminants, as demonstrated by the ongoing efforts to establish the UNs Intergovernmental Science-Policy Panel on Chemicals, Waste, and Pollution Prevention. This review identifies the sources and impacts of emerging contaminants on planetary health, emphasizing the importance of adopting a One Health approach. Strategies for monitoring and addressing these pollutants are discussed, underscoring the need for robust and socially equitable environmental policies at both regional and international levels. Urgent actions are needed to transition toward sustainable pollution management practices to safeguard our planet for future generations.

The hydrodynamics of thin films is typically described using phenomenological models whose connection to the microscopic particle dynamics is a subject of ongoing research. Existing methods based on density functional theory provide a good description of static thin films, but are not sufficient for understanding nonequilibrium dynamics. In this work, we present a microscopic derivation of the thin film equation using the Mori-Zwanzig projection operator formalism. This method allows to directly obtain the correct gradient dynamics structure along with microscopic expressions for the mobility and the free energy. Abstract

Abstract Phenotyping methods for seed morphology are mostly limited to two-dimensional imaging or manual measures. In this study, we present a novel seed phenotyping approach utilizing lab-based X-ray microscopy (XRM) to characterize 3D seed morphology, internal structures, and cellular analysis from a single scan. Seeds of pennycress (Thlaspi arvense L.) an oilseed cover crop, were scanned and segmented using a machine learning model. Seed morphological analysis and a coat thickness map was applied to compare seed volumes of four genotypes. Notably, the 3D seed volume measurement alone was not enough to reveal differences in seed coat between the genotypes. Applying a seed coat thickness map revealed that the Large-Golden genotype had a thinner seed coat compared to wildtype despite a greater seed coat volume. Cellular analysis showed that cotyledon cell size was a driving factor for larger seeds. XRM was compared to traditional seed morphology traits and positive correlations were observed. Between Large-Golden and wildtype, differences could only be resolved by XRM highlighting the limitations of 2D seed area measures. These results demonstrate that XRM can provide quantitative measures extracted from seeds for enhancing our understanding of seed structure, development, and facilitate breeding efforts for enhanced seed quality and crop performance.

Abstract

Multi-Agent Reinforcement Learning is becoming increasingly more important in times of autonomous driving and other smart industrial applications. Simultaneously a promising new approach to Reinforcement Learning arises using the inherent properties of quantum mechanics, reducing the trainable parameters of a model significantly. However, gradient-based Multi-Agent Quantum Reinforcement Learning methods often have to struggle with barren plateaus, holding them back from matching the performance of classical approaches. While gradient free Quantum Reinforcement Learning methods may alleviate some of these challenges, they too are not immune to the difficulties posed by barren plateaus. We build upon an existing approach for gradient free Quantum Reinforcement Learning and propose three genetic variations with Variational Quantum Circuits for Multi-Agent Reinforcement Learning using evolutionary optimization. We evaluate our genetic variations in the Coin Game environment and also compare them to classical approaches. We showed that our Variational Quantum Circuit approaches perform significantly better compared to a neural network with a similar amount of trainable parameters. Compared to the larger neural network, our approaches archive similar results using $97.88\%$ less parameters.

Abstract

Multiple adjustment difficulties have been associated with childrens exposure to recent parental wartime military deployments, but long-term consequences have not yet been systematically studied. This investigation will assess direct and indirect relationships between exposures to parental deployments early in life and later youth adjustment. Parents psychological health and family processes will be examined as mediators, and parents and childrens vulnerability and support will be examined as moderators. Archival data will be combined with new data gathered from two children and up to two parents in families where children will be aged 11 to 16 at the first data collection and will have experienced at least one parental deployment, for at least one child prior to age 6. Data are being gathered via telephone interviews and web-based surveys conducted twice one year apart. Outcomes are indicators of childrens social-emotional development, behavior, and academic performance. Notable features of this study include oversampling of female service members, inclusion of siblings, and inclusion of families of both veterans and currently serving members. This study has potentially important implications for schools, community organizations and health care providers serving current and future cohorts of military and veteran families.

Here we report a multi-messenger platform using velocimetry and \textit{in situ} angularly and spectrally resolved X-ray scattering to measure the thermodynamic conditions and ion structure factor of materials at extreme pressures. We document the pressure, density, and temperature of shocked silicon near 100 GPa with uncertainties of 6%, 2%, and 20%, respectively. In this work, we present a systematic approach to investigate the existence, multiplicity, and local gradient regularity of solutions for nonlocal quasilinear equations with local gradient degeneracy. Our method involves an interactive geometric argument that interplays with uniqueness property for the corresponding homogeneous problem, leading with gradient H\"older regularity estimates. This approach is intrinsically developed for nonlocal scenarios, where uniqueness holds for the local homogeneous problem. We illustrate our results by showing classes of exterior data that exhibit multiple solutions, while also highlighting relevant cases where uniqueness is confirmed.

We show that the critical $\lambda=\lambda^*$, defined as the infimum of all $\lambda$ attaining a non-negative supersolution, attains a nonnegative solution $u$. Under suitable conditions, it is also shown that $\lambda^*$ is the supremum of all $\lambda$ for which a non-positive subsolution is possible. Moreover, uniqueness of the solution $u$, corresponding to $\lambda^*$, is also established. Furthermore, we provide a probabilistic characterization that determines the uniqueness of the pair $(u, \lambda^*)$ in the class of all solution pair $(u, \lambda)$ with $u\geq 0$. Our proof technique involves both analytic and probabilistic methods in combination with a new local Lipschitz estimate obtained in this article.

Abstract

The LHCb upgrade represents a major change of the experiment. The detectors have been almost completely renewed to allow running at an instantaneous luminosity five times larger than that of the previous running periods. Readout of all detectors into an all-software trigger is central to the new design, facilitating the reconstruction of events at the maximum LHC interaction rate, and their selection in real time. The experiment's tracking system has been completely upgraded with a new pixel vertex detector, a silicon tracker upstream of the dipole magnet and three scintillating fibre tracking stations downstream of the magnet. The whole photon detection system of the RICH detectors has been renewed and the readout electronics of the calorimeter and muon systems have been fully overhauled. The first stage of the all-software trigger is implemented on a GPU farm. The output of the trigger provides a combination of totally reconstructed physics objects, such as tracks and vertices, ready for final analysis, and of entire events which need further offline reprocessing. Abstract

Hidradenitis suppurativa is a chronic skin disease associated with significant disease burden. To assess patients’ treatment benefits, disease-specific outcomes measurement is needed. This study aimed to develop a questionnaire called Patient Benefit Index for hidradenitis suppurativa (PBI-HS). After an open-item survey, the items were condensed into a 26-item questionnaire on patient needs and benefits, with a 5-point Likert scale. In the validation study, construct and content validity, responsiveness, and feasibility of the questionnaire were assessed at 2 time points. A 26-item questionnaire was created following open-item generation by n = 72 patients. In the validation study, 3 items perceived as most relevant by patients were: “to be free of pain” (mean: 3.6 on a scale of 0–4), “to be free of inflammation” (mean: 3.6), “to have no more scars” (mean: 3.1). Significant correlations of PBI-HS at follow-up visit were found with the physical global assessment for hidradenitis suppurativa (HS-PGA) (r = –0.471; p = 0.000), number of inflammatory lesions (r = –0.359; p = 0.005), and DLQI (r = –0.383; p = 0.003), indicating less disease burden in patients with higher treatment benefits. The PBI-HS is a feasible and valid instrument to assess patient-reported treatment benefits in hidradenitis suppurativa.

Abstract

Wastewater treatment plants (WWTPs) represent one of biotechnology's largest and most critical applications, playing a pivotal role in environmental protection and public health. In WWTPs, activated sludge (AS) plays a major role in removing contaminants and pathogens from wastewater. While metagenomics has advanced our understanding of microbial communities, it still faces challenges in revealing the genomic heterogeneity of cells, uncovering the microbial dark matter, and establishing precise links between genetic elements and their host cells as a bulk method. These issues could be largely resolved by single-cell sequencing, which can offer unprecedented resolution to show the unique genetic information. Here we show the high-throughput single-cell sequencing to the AS microbiome. The single-amplified genomes (SAGs) of 15,110 individual cells were clustered into 2,454 SAG bins. We find that 27.5% of the genomes in the AS microbial community represent potential novel species, highlighting the presence of microbial dark matter. Furthermore, we identified 1,137 antibiotic resistance genes (ARGs), 10,450 plasmid fragments, and 1,343 phage contigs, with shared plasmid and phage groups broadly distributed among hosts, indicating a high frequency of horizontal gene transfer (HGT) within the AS microbiome. Complementary analysis using 1,529 metagenome-assembled genomes from the AS samples allowed for the taxonomic classification of 98 SAG bins, which were previously unclassified. Our study establishes the feasibility of single-cell sequencing in characterizing the AS microbiome, providing novel insights into its ecological dynamics, and deepening our understanding of HGT processes, particularly those involving ARGs. Additionally, this valuable tool could monitor the distribution, spread, and pathogenic hosts of ARGs both within AS environments and between AS and other environments, which will ultimately contribute to developing a health risk evaluation system for diverse environments within a One Health framework.

Abstract

We study existence, uniqueness and boundary blow-up profile for fractional harmonic functions on a bounded smooth domain $\Omega \subset \mathbb R^N$. We deal with harmonic functions associated to uniformly elliptic, fully nonlinear nonlocal operators, including the linear case $$ (-\Delta)^s u = 0 \quad \mbox{in} \ \Omega, $$ where $(-\Delta)^s$ denotes the fractional Laplacian of order $2s \in (0,2)$. We use the viscosity solution's theory and Perron's method to construct harmonic functions with zero exterior condition in $\bar \Omega^c$, and boundary blow-up profile $$ \lim_{x\to x_0, x \in \Omega}\mathrm{dist}(x, \partial \Omega)^{1-s}u(x)=h(x_0), \quad \mbox{for all} \quad x_0\in \partial \Omega, $$ for any given boundary data $h \in C(\partial \Omega)$. Our method allows us to provide blow-up rate for the solution and its gradient estimates. Results are new even in the linear case.

Abstract

Given $\epsilon \in (0,1)$ and $\lambda > 1$, we address the existence of solutions for the Sinh-Poisson equation with Robin boundary value condition $$ \begin{cases} \Delta u+\epsilon^2 (e^{u} - e^{-u})=0 &\mbox{in }\Omega\\ \frac{\partial u}{\partial\nu}+\lambda u=0 &\mbox{on }\partial\Omega, \end{cases} $$ where $\Omega\subset\mathbb{R}^2$ is a bounded smooth domain. We prove two existence results under a suitable relation between $\epsilon$ small and $\lambda$ large. When $\Omega$ is symmetric with respect to an axis, we prove the existence of a family of solutions $u_{\epsilon,\lambda}$ concentrating at two points with different spin, both located on the symmetry line and close to the boundary. In the second result, we assume $\Omega$ is not simply connected and we construct sign-changing solutions concentrating at points located close to the boundary, each of them on a different connected component of the boundary.

Abstract

BACKGROUND: Brexucabtagene autoleucel (brexu-cel) is an autologous anti-CD19 chimeric antigen receptor (CAR) T-cell therapy approved in the USA for adults with relapsed or refractory (R/R) B-cell acute lymphoblastic leukemia (B-ALL) and in the European Union for patients ≥26 years with R/R B-ALL. After 2 years of follow-up in ZUMA-3, the overall complete remission (CR) rate (CR+CR with incomplete hematological recovery (CRi)) was 73%, and the median overall survival (OS) was 25.4 months in 78 Phase 1 and 2 patients with R/R B-ALL who received the pivotal dose of brexu-cel. Outcomes by prior therapies and subsequent allogeneic stem cell transplantation (alloSCT) are reported. METHODS: Eligible adults had R/R B-ALL and received one infusion of brexu-cel (1×10⁶ CAR T cells/kg) following conditioning chemotherapy. The primary endpoint was the CR/CRi rate per central review. Post hoc subgroup analyses were exploratory with descriptive statistics provided. RESULTS: Phase 1 and 2 patients (N=78) were included with median follow-up of 29.7 months (range, 20.7-58.3). High CR/CRi rates were observed across all prior therapy subgroups examined: 1 prior line of therapy (87%, n=15) and ≥2 prior lines (70%, n=63); prior blinatumomab (63%, n=38) and no prior blinatumomab (83%, n=40); prior inotuzumab (59%, n=17) and no prior inotuzumab (77%, n=61); and prior alloSCT (76%, n=29) and no prior alloSCT (71%, n=49). The frequency of Grade ≥3 cytokine release syndrome, neurological events, and treatment-related Grade 5 adverse events were largely similar among prior therapy subgroups.Median duration of remission (DOR) in responders with (n=14) and without (n=43) subsequent alloSCT was 44.2 (95% CI, 8.1 to not estimable (NE)) and 18.6 months (95% CI, 9.4 to NE); median OS was 47.0 months (95% CI, 10.2 to NE) and not reached (95% CI, 23.2 to NE), respectively. Median DOR and OS were not reached in responders without prior or subsequent alloSCT (n=22). CONCLUSIONS: In ZUMA-3, adults with R/R B-ALL benefited from brexu-cel, regardless of prior therapies and subsequent alloSCT status, though survival appeared better in patients without certain prior therapies and in earlier lines of therapy. Additional studies are needed to determine the impact prior therapies and subsequent alloSCT have on outcomes of patients who receive brexu-cel.

Abstract

Abstract Background When managed effectively, oil wealth can drive economic development and improve wellbeing. Conversely, as has been the experience in Ogoniland in the Niger Delta, the presence of the oil industry can lead to environmental degradation and social and economic vulnerabilities with negative impacts on health and wellbeing. Few studies have explored the experiences and perceptions of these impacts among residents of host communities themselves. Methods This qualitative study explored Ogoni residents’ experiences and perceptions of oil-industry related economic exclusion and associated impacts on health and wellbeing. In-depth interviews with 22 participants from four Local Government Areas (LGAs) were analyzed with open (inductive) coding. Guided by constructivist philosophy, interpretation of emerging themes was informed by the concept of social exclusion which recognizes that multi-dimensional processes can deprive individuals or groups of resources, rights, goods, and services, thus limiting broader societal participation. Results Findings highlight the exclusionary impacts of the oil industry at the intersection of i) damaged livelihoods and family income, that increased economic vulnerability and reduced participants’ ability to meet basic needs including ability to pay for healthcare; ii) lack of progress on environmental remediation and non-transparent benefit sharing schemes that exacerbate economic displacement and contribute to ongoing exposures to air and water pollution; and iii) insufficient of investment in economic development and essential health services, limiting employment opportunities and ability to access adequate healthcare. Conclusion Addressing these issues requires integrated policy interventions focusing on equitable resource distribution, environmental restoration, and inclusive economic development to promote sustainable community resilience.

Abstract

Abstract Phase Ib trials are common in oncology development but often are not powered for statistical significance. Go/no‐go decisions are largely driven by observed trends in response data. We applied a bootstrapping method to systematically compare tumor dynamic end points to historical control data to identify drugs with clinically meaningful efficacy. A proprietary mathematical model calibrated to phase Ib anti–PD‐1 therapy trial data (KEYNOTE‐001) was used to simulate thousands of phase Ib trials (n = 30) with a combination of anti–PD‐1 therapy and four novel agents with varying efficacy. A redacted bootstrapping method compared these results to a simulated phase III control arm (N = 511) while adjusting for differences in trial duration and cohort size to determine the probability that the novel agent provides clinically meaningful efficacy. Receiver operating characteristic (ROC) analysis showed strong ability to separate drugs with modest (area under ROC [AUROC] = 83%), moderate (AUROC = 96%), and considerable efficacy (AUROC = 99%) from placebo in early‐phase trials (n = 30). The method was shown to effectively move drugs with a range of efficacy through an in silico pipeline with an overall success rate of 93% and false‐positive rate of 7.5% from phase I to phase III. This model allows for effective comparisons of tumor dynamics from early clinical trials with more mature historical control data and provides a framework to predict drug efficacy in early‐phase trials. We suggest this method should be employed to improve decision making in early oncology trials.
