5. Semiotic Language Use in Schoolscapes on the Arctic Borderland

Abstract

The article explores the visible, semiotic use of languages in two schoolscapes on the Arctic borderland and how these schoolscapes stimulate the learning of languages and cultures. The schools are situated in a historically multilingual area with several languages present. However, since the 17th century, the area has undergone Swedification, resulting in the current situation where all languages other than Swedish are endangered minority languages. The schoolscapes were studied through visual ethnography, and 229 photographs were analysed by qualitative content analysis. The results show that in one school in the middle of Sápmi, the Sámi languages were almost entirely excluded, despite their relevance and importance in maintaining bilingualism. Conversely, in a school where some pupils were from the Finnish side of the national border, the Finnish language was present in the form of various subject-relevant books. However, in both schools, all formal information is given to pupils in Swedish, with only a few exceptions permitted by the school management. Overall, the visual use of semiotic language is mainly teacher-produced, and the pupils’ opportunities to use the schoolscape as an affordance for active bilingualism through social participation are minimal.

Abstract

Genetic factors underlying coronary artery disease (CAD) have been widely studied using genome-wide association studies (GWASs). However, the functional understanding of the CAD loci has been limited by the fact that a majority of GWAS variants are located within non-coding regions with no functional role. High cholesterol and dysregulation of the liver metabolism such as non-alcoholic fatty liver disease confer an increased risk of CAD. Here, we studied the function of non-coding single-nucleotide polymorphisms in CAD GWAS loci located within liver-specific enhancer elements by identifying their potential target genes using liver cis-eQTL analysis and promoter Capture Hi-C in HepG2 cells. Altogether, 734 target genes were identified of which 121 exhibited correlations to liver-related traits. To identify potentially causal regulatory SNPs, the allele-specific enhancer activity was analyzed by (1) sequence-based computational predictions, (2) quantification of allele-specific transcription factor binding, and (3) STARR-seq massively parallel reporter assay. Altogether, our analysis identified 1,277 unique SNPs that display allele-specific regulatory activity. Among these, susceptibility enhancers near important cholesterol homeostasis genes (APOB, APOC1, APOE, and LIPA) were identified, suggesting that altered gene regulatory activity could represent another way by which genetic variation regulates serum lipoprotein levels. Using CRISPR-based perturbation, we demonstrate how the deletion/activation of a single enhancer leads to changes in the expression of many target genes located in a shared chromatin interaction domain. Our integrative genomics approach represents a comprehensive effort in identifying putative causal regulatory regions and target genes that could predispose to clinical manifestation of CAD by affecting liver function.

Turkulainen sairasti 1600-luvulla tautia, sjuka, eikä lähteissä kerrota useinkaan sairauden tarkempaa laatua. Sairaita ja heidän parantajiaan mainitaan silti jatkuvasti kaupunkia koskevissa lähteissä, sillä sairaus esti saapumasta oikeuden eteen tai matkustamasta, aiheutti perheelle ja naapureille vaikeuksia, tai viranomaisten oli mietittävä sopivia torjuntakeinoja epidemian uhatessa. Kodeissa harjoitetun parantamisen rinnalla Turussa oli vuosisadan aikana vähintään viitisenkymmentä ammattimaista parantajaa sekä lisäksi puoliammattimaisia kansanparantajia, jotka tekivät sairaanhoitotyötä korvausta vastaan. Keskityn tässä artikkelissa lähiluvun ja mikrohistoriallisen näkökulman kautta Turun 1600-luvun sairaanhoitoon liittyviin tiloihin ja parantajiin, sekä sairaiden kohteluun paikallisyhteisössään. Tässä artikkelissa esitetyt toimintamallit olivat pääosin käytössä myös muualla Ruotsin valtakunnassa, sillä samat hoitotyötä harjoittaneet ammattiryhmät toimivat koko sen alueella, ja niitä koskevat säännöt ja ohjeet annettiin keskushallinnon toimesta Tukholmassa. Along with the healing practiced at homes, there were at least fifty professional healers in Turku during the seventeenth century, as well as semi-professional folk healers who practised medical treatment for compensation. Entries regarding medical treatment can be found on the court records of the Turku Magistrate. Depending on wealth, one could rely on bathers, barbers, farmacists, army barbers and medical professors of the Academy of Turku. At least some of the patients belonging to all social groups allowed the use of white magic in healing work. Healing was a comprehensive process that used both the various skills of healers and the possibilities offered by religion and magic. The operational models presented in this article were mainly used in other parts of the Swedish realm too, as the same professional groups practiced nursing throughout its territory, and the rules and instructions concerning them were issued by the central administration in Stockholm. Medical care throughout the seventeenth century was marked by the emergence of professional groups according to different professional association rules. They were closely monitored by both the crown and the church. However, this did not mean the disappearance of more traditional ways, but institutional improvement, i.e. side by side with folk healing.

Recently, many preservative-free prostaglandin analogs have been marketed to increase their tolerance in chronic use. However, potentially safer formulations have been reported to induce inflammation within ocular surface and adnexa, associated with pronounced activation of tissue macrophages.Aim: We aimed to evaluate the effect of a Stearoyl-CoA desaturase-1 (SCD1) inhibitor, MF-438, on the differentiation of monocytes exposed to eye drop detergents, representing saturated fatty acid derivatives.Methods: A culture of human peripheral blood monocytes was exposed to eye drops containing fatty acid derivatives (eye drop detergents), pf-latanoprost (Monoprost®, hydroxystearate macrogolglycerol – MGHS40) or pf-tafluprost (Taflotan®, polysorbate 80 – PS80), as well as pf-latanoprost+MF-438, MGHS40, and PS80. For the negative control C(-), monocytes were cultured in basal medium, and for the positive controls, monocytes were stimulated with Lipopolysaccharide (LPS) and Interferon γ (IFNγ) (M1 macrophages) or Interleukin-4 (IL-4) (M2 macrophages). The concentration of desaturase in the cell homogenates was determined by ELISA. The number of cells was counted under a microscope at 20x magnification.Results: The following concentrations of SCD1 (ng/mL) were measured: 7.8± 0.3 – pf-latanoprost group; 1.5± 0.4 – pf-tafluprost group; 6.8± 0.7 – MGHS40 group; 0.4± 0.002 – PS80 group; 0.9± 0.02 – pf-latanoprost+MF-438 group; 5.4± 1.6 – C(-) control; 0.5± 0.04 – M1 control; 2.2± 0.13 – M2 control. The percentages of macrophages in culture were 33.6%, 17.6%, 33%, 0%, 13.5%, 18.6%, 36.3%, and 39.3% for the pf-latanoprost, pf-tafluprost, MGHS40, PS80, pf-latanoprost+MF-438, C(-), M1, and M2 cultures, respectively. There was a strong correlation between SCD1 concentration and macrophage count in the culture (r=0.8, p< 0.05).Conclusion: Inhibition of SCD1 in monocytes prevents their transformation into macrophages after exposure to saturated fatty acid derivatives contained in eye drops, which may contribute to the limitation of latent inflammation within ocular adnexa and could possibly translate into better tolerability of the topical treatment.Keywords: prostaglandin analogs, Stearoyl-CoA desaturase-1, latent inflammation, macrophages

Abstract

Inom ramen för en kontextualiserad grammatikundervisning utforskar och analyserar den här artikeln svenska gymnasieelevers metalingvistiska aktiviteter där de resonerar om ett skönlitterärt textutdrag. En induktiv innehållsanalys genomfördes i syfte att belysa på vilka sätt tre elevgrupper resonerar om huvudsatsformade och icke huvudsatsformade meningar i ett textutdrag ur Stina Jacksons roman Silvervägen. Resultatet visar, i likhet med tidigare studier, att elevernas resonemang i första hand stödjs av minnesregler och betydelser, vilket kan försvåra igenkännandet av den syntaktiska (o)fullständigheten. När eleverna kopplar samman meningarna till kringliggande meningar lyckas de emellertid känna igen den syntaktiska (o)fullständigheten. Även om ett betydelsemässigt fokus kan förhindra eleverna att identifiera och förklara grammatiska strukturer visar resultatet att utgångspunkten i att textuellt knyta an meningar, såväl syntaktiskt som semantiskt, kan stötta eleverna i resonemang rörande syntaktisk (o)fullständighet. Resultatet pekar vidare på att en utgångspunkt i skönlitterära texter och syntaktisk (o)fullständighet kan fungera som en deskriptiv ingång för att synliggöra och diskutera språkliga strukturer och konventioner i förstaspråksundervisningen.
