8. OrgoPrep: A Remote Peer-Led Summer Program Preparing Students for Organic Chemistry

Abstract

The transition from general chemistry to organic chemistry is challenging for many students. To succeed, students must remember their foundational chemistry skills and effectively transfer that knowledge into the context of organic chemistry. However, this transition is often hindered by many obstacles: (1) lack of prerequisite knowledge, (2) lack of understanding how foundational chemistry knowledge is applicable to organic chemistry, and (3) high levels of anxiety and low levels of self-efficacy toward organic chemistry. To address these challenges, a peer-led summer program that leverages question-embedded videos, worksheets, and synchronous remote webinars to enhance students' transition into organic chemistry was developed and assessed. This six-week program, OrgoPrep, covers a wide set of introductory organic chemistry topics: resonance structures, hybridization and molecular shape, molecular orbitals, acid--base chemistry, limiting reagents, and more. Substantial benefits, such as decreasing student anxiety and doubling student self-efficacy, were observed for students who completed OrgoPrep. These students also significantly outperformed their peers who did not participate in the program, despite having no statistical difference in prior chemistry abilities--OrgoPrep students had a 3-fold decrease in failing grades and a 2-fold increase in A grades. Overall, OrgoPrep successfully serves as a bridge between general and organic chemistry, while facilitating knowledge transfer and enhancing student self-efficacy.

18. Corticosteroids for the Treatment of Internal Temporomandibular Joint Disorders: A Systematic Review and Network Meta-Analysis.

Abstract

Background: We evaluated the comparative effectiveness of all intra-articular injection corticosteroids for treating internal temporomandibular joint (TMJ) disorders. Methods: We searched MEDLINE, CENTRAL, EMBASE, SCOPUS, and LILACS through December 2023. We included randomized clinical trials (RCTs) enrolling patients with symptomatic internal disorders of the TMJ comparing any type of intra-articular corticosteroid therapy against another or to another minimally invasive therapy. The outcomes of interest were pain, range of mandibular motion (RoM), quality of life (QoL) and adverse effects at 1, 3, 6, and 12 months. We assessed the risk of bias using the Cochrane Collaboration's tool. We conducted a frequentist network meta-analysis and assessed the certainty of the evidence (CoE) using GRADE. Results: We included 20 RCTs enrolling 810 participants, which assessed five corticosteroids alone or combined with arthrocentesis or hyaluronic acid. Based on moderate CoE, betamethasone is among the most effective corticosteroids for reducing pain at one (mean difference compared to arthrocentesis [MD], −3.80; 95% confidence interval [CI], −4.55 to −3.05) and three months (MD, −2.74; 95%CI, −3.42 to −2.06), and arthrocentesis plus dexamethasone at six months (MD, −0.80; 95%CI, −1.57 to −0.03). There was no convincing evidence that any intervention was better than arthrocentesis for improving the RoM and QoL at any follow-up time. Methylprednisolone may be more harmful than arthrocentesis for adverse effects. Discussion: Betamethasone and arthrocentesis plus dexamethasone are the most effective in managing pain in the short and medium term compared to arthrocentesis (moderate CoE). Decisions about their use should consider other factors, such as costs, feasibility, and acceptability. 27. Investigating Attitudinal Profiles and Disparities in Attitudes among Historically Marginalized Undergraduate Chemistry Students

Abstract

Self-efficacy holds important implications for attitudes toward learning and course performance. This study explores the configurations of self-reported sources of self-efficacy in a sample of general and organic chemistry students, as well as disparities in these configurations for historically marginalized students. A latent profile analysis classified 687 students into groups labeled as Multi-Source, Low Social Persuasions, Physiological State Dependent, or At-Risk based on the four sources of self-efficacy (i.e., mastery experiences, vicarious experiences, social persuasions, and physiological state). Profile membership was shown to be related to important attitudes toward the subject of chemistry (i.e., intellectual accessibility, emotional satisfaction, and anxiety) and was predictive of exam performance later in the semester. Students belonging to the Multi-Source profile, with high levels of all sources of self-efficacy, were found to have scored the highest on exams, one standard deviation above those with the At-Risk profile. Profile membership was not homogeneous among groups of students as a result of racism, sexism, and classism; rather, general chemistry, female, and first-generation students were each more likely to belong to the At-Risk group compared to organic chemistry, male, and continuing-generation students. Similarly, Black students were five times more likely to belong to the At-Risk group than White students. These findings indicate that self-efficacy profiles may serve as early screening tools for helping to identify at-risk chemistry students needing support, allowing instructors to proactively implement interventions to better support all students and mitigate the inequity of outcomes experienced by students from historically marginalized groups.

Abstract

Genomic alterations in GNAS, the gene coding for the Gαs heterotrimeric G protein, are associated with a large number of human diseases. Here, we explored the role of Gαs on stem cell fate decisions by using the mouse epidermis as a model system. Conditional epidermal deletion of Gnas or repression of PKA signalling caused a remarkable expansion of the stem cell compartment, resulting in rapid basal-cell carcinoma formation. In contrast, inducible expression of active Gαs in the epidermis caused hair follicle stem cell exhaustion and hair loss. Mechanistically, we found that Gαs-PKA disruption promotes the cell autonomous Sonic Hedgehog pathway stimulation and Hippo signalling inhibition, resulting in the non-canonical activation of GLI and YAP1. Our study highlights an important tumour suppressive function of Gαs-PKA, limiting the proliferation of epithelial stem cells and maintaining proper hair follicle homeostasis. These findings could have broad implications in multiple pathophysiological conditions, including cancer.
