PURPOSE: Patients' values and priorities in their lives should be appreciated from an early phase of incurable diseases such as advanced cancer. However, studies examining these characteristics have been lacking. This study attempted to determine what patients with advanced lung cancer valued most, once they had been diagnosed, and any associated factors. METHODS: Patients with newly diagnosed advanced lung cancer (N = 248) were enrolled in a questionnaire survey conducted at 16 hospitals in Japan. Their priorities were assessed using a free-text response to the question what is the most important thing to you now? at the time of diagnosis and 3 months after diagnosis. The free-text responses were classified into 10 categories for quantification. The clinical characteristics associated with the category describing daily life were further examined. RESULTS: Free-text comments were obtained from 103 (44.0%) and 66 (42.6%) patients at the time of diagnosis and at 3 months, respectively. The most frequent categories were family (at diagnosis: 50.5%; at 3 months: 50.0%) and daily life (at diagnosis: 33.0%; at 3 months: 36.4%), followed by health (at diagnosis: 32.0%; at 3 months: 27.3%) at both time points. The patients mentioning daily life, the issues related to how to spend daily life, showed significantly higher total scores and functional well-being subscale scores on the Functional Assessment of Cancer Therapy-Lung scale at both time points and lower depression scores at diagnosis and lower anxiety scores at 3 months on the Hospital Anxiety and Depression Scale. CONCLUSION: Family and daily life were highly valued by patients with advanced lung cancer at diagnosis. A better quality of life and better mood were associated with mentioning daily life, which should be taken into account in care planning to maintain patients' involvement in daily life even with incurable diseases.

Multiple epidermal growth factor receptor (EGFR) tyrosine kinase inhibitors (EGFR-TKI) have been developed to effectively inhibit EGFR-derived signals in non–small cell lung cancer (NSCLC). In this study, we assessed the efficacy of EGFR-TKIs, including a novel third-generation inhibitor naquotinib (ASP8273), in clinically relevant EGFR mutations, including L858R, exon 19 deletion, L858R+T790M, exon 19 deletion+T790M with or without a C797S mutation, and several exon 20 insertion mutations. Using structural analyses, we also elucidated the mechanism of activation and sensitivity/resistance to EGFR-TKIs in EGFR exon 20 insertion mutations. The efficacy of naquotinib in cells with L858R, exon 19 deletion and exon 19 deletion+T790M was comparable with that of osimertinib. Interestingly, naquotinib was more potent than osimertinib for L858R+T790M. Additionally, naquotinib and osimertinib had comparable efficacy and a wide therapeutic window for cells with EGFR exon 20 insertions. Structural modeling partly elucidated the mechanism of activation and sensitivity/resistance to EGFR-TKIs in two EGFR exon 20 insertion mutants, A767_V769dupASV and Y764_V765insHH. In summary, we have characterized the efficacy of EGFR-TKIs for NSCLC using in vitro and structural analyses and suggested the mechanism of activation and resistance to EGFR-TKIs of EGFR exon 20 insertion mutations. Our findings should guide the selection of appropriate EGFR-TKIs for the treatment of NSCLC with EGFR mutations and help clarify the biology of EGFR exon 20 insertion mutations. High cell density Ni monolithic catalysts coated with a series of Ni–Re bimetallic surface layers (Re 0–38 at%) were synthesized and examined for application in methane steam reforming (MSR) under a steam-to-carbon ratio of 1.36 and a gas hourly space velocity of 6400 h−1 in the temperature range of 773–1173 K. The influence of mass and heat transport was verified to be minor below 973 K under the present experimental conditions. The catalytic activities of the Ni monolithic catalysts were effectively improved in the presence of a Ni–Re bimetallic surface layer coating, and the stability in terms of the catalyst activity was significantly enhanced below 973 K upon increasing the Re content to >24 at%. The Ni–Re surface layer was analyzed by using in situ X-ray absorption fine structure spectroscopy, synchrotron radiation X-ray diffraction, scanning electron microscopy, and density functional theory calculations. These results revealed that a Ni–Re alloy consisting of a Ni(Re) solid solution phase and a Re phase was formed during hydrogen pre-reduction and the subsequent MSR reaction. The Re atoms in the Ni–Re alloy promoted hydrogen adsorption on the surface, thereby suppressing the oxidation of the surface Ni atoms during MSR, and enhancing the stability of the monolithic catalysts.

Coronavirus disease 2019 (COVID-19) is a recently-emerged infectious disease that has caused millions of deaths, where comprehensive understanding of disease mechanisms is still unestablished. In particular, studies of gene expression dynamics and regulation landscape in COVID-19 infected individuals are limited. Here, we report on a thorough analysis of whole blood RNA-seq data from 465 genotyped samples from the Japan COVID-19 Task Force, including 359 severe and 106 non-severe COVID-19 cases. We discover 1169 putative causal expression quantitative trait loci (eQTLs) including 34 possible colocalizations with biobank fine-mapping results of hematopoietic traits in a Japanese population, 1549 putative causal splice QTLs (sQTLs; e.g. two independent sQTLs at TOR1AIP1), as well as biologically interpretable trans-eQTL examples (e.g., REST and STING1), all fine-mapped at single variant resolution. We perform differential gene expression analysis to elucidate 198 genes with increased expression in severe COVID-19 cases and enriched for innate immune-related functions. Finally, we evaluate the limited but non-zero effect of COVID-19 phenotype on eQTL discovery, and highlight the presence of COVID-19 severity-interaction eQTLs (ieQTLs; e.g., CLEC4C and MYBL2). In molecular rotors, a type of molecular machine, spontaneous rotors without the need for an external stimulus are promising because conventional molecular rotors require a continuous energy supply. In this study, we demonstrate spontaneous transformation from kinetically favored metastable right-handed helical aggregates to thermodynamically stable left-handed helical aggregates after an evaporation procedure. In addition, we propose the conditions for preparation of metastable right-handed helical aggregates, whose chirality can be spontaneously inverted, based on kinetic analysis. This molecular power spring will be useful for designing new types of molecular machines.

To elucidate the host genetic loci affecting severity of SARS-CoV-2 infection, or Coronavirus disease 2019 (COVID-19), is an emerging issue in the face of the current devastating pandemic. Here, we report a genome-wide association study (GWAS) of COVID-19 in a Japanese population led by the Japan COVID-19 Task Force, as one of the initial discovery GWAS studies performed on a non-European population. Enrolling a total of 2,393 cases and 3,289 controls, we not only replicated previously reported COVID-19 risk variants (e.g., LZTFL1, FOXP4, ABO, and IFNAR2), but also found a variant on 5p35 (rs60200309-A at DOCK2) that was significantly associated with severe COVID-19 in younger (-8 (odds ratio = 2.01, 95% confidence interval = 1.58-2.55). This risk allele was prevalent in East Asians, including Japanese (minor allele frequency [MAF] = 0.097), but rarely found in Europeans. Cross-population Mendelian randomization analysis made a causal inference of a number of complex human traits on COVID-19. In particular, obesity had a significant impact on severe COVID-19. The presence of the population-specific risk allele underscores the need of non-European studies of COVID-19 host genetics.

The canonical molecular orbitals (CMO) calculation provides useful knowledge such as molecular orbitals, energy level, charge density and so on. As one of the properties obtained from the CMO calculation of protein results, the interaction energy analysis between amino acid residues and/or substrates is useful as molecular design and functional analysis. The energy decomposition analysis approaches are the well-known calculation methods for interaction energy analysis, however, the electronic structure calculation of large molecule is very expensive and hard to achieve. Therefore, we have implemented the interaction analysis method by using the energy density analysis (EDA) method. The calculation for the EDA method is as simple as the Mulliken population method but requires matrix elements for energy calculation. By adopting the grid-free method for the exchange-correlation term, we could apply the EDA method in the DFT scheme.

Streptomyces incarnatus NRRL8089 produces the antiviral, antifungal, antiprotozoal nucleoside antibiotic sinefungin. To enhance sinefungin production, multiple mutations were introduced to the rpoB gene encoding RNA polymerase (RNAP) β-subunit at the target residues, D447, S453, H457, and R460. Sparse regression analysis using elastic-net lasso-ridge penalties on previously reported H457X mutations identified a numeric parameter set, which suggested that H457R/Y/F may cause production enhancement. H457R/R460C mutation successfully enhanced the sinefungin production by 3-fold, while other groups of mutations, such as D447G/R460C or D447G/H457Y, made moderate or even negative effects. To identify why the rif cluster residues have diverse effects on sinefungin production, an RNAP/DNA/mRNA complex model was constructed by homology modeling and molecular dynamics simulation. The 4 residues were located near the mRNA strand. Density functional theory–based calculation suggested that D447, H457, and R460 are in direct contact with ribonucleotide, and partially positive charges are induced by negatively charged chain of mRNA.

The catalysis of methane steam reforming (MSR) by pure Ni honeycombs with high cell density of 2300 cells per square inch (cpsi) was investigated to develop efficient and inexpensive catalysts for hydrogen production. The Ni honeycomb catalyst was assembled using 30-μm-thick Ni foils, and showed much higher activity than that of a Ni honeycomb catalyst with cell density of 700 cpsi at a steam-to-carbon ratio of 1.36 and a gas hourly space velocity of 6400 h−1 in a temperature range of 873–1173 K. Notably, the activity increased approximately proportional to the increasing geometric specific surface area of the honeycombs. The turnover rate of the Ni honeycomb catalyst was higher than that of supported Ni catalysts. The changes in chemical state of the Ni catalyst during hydrogen reduction and MSR reaction were analyzed by in situ X-ray absorption fine structure spectroscopy, which revealed that deactivation was mainly due to oxidation of the surface Ni atoms. These results demonstrated that the high-cell-density Ni honeycomb catalyst exhibits good performance for MSR reaction, and easy regeneration of the deactivated Ni honeycomb catalyst is possible only via hydrogen reduction.

The effect of steam and hydrogen treatments on the catalytic properties of a high-cell-density pure Ni honeycomb catalyst for methane steam reforming was investigated. A pretreatment consisting of steam treatment and subsequent hydrogen reduction at 1173 K significantly enhanced the catalytic activity of the Ni honeycomb catalyst at low temperatures below 1073 K. In situ X-ray absorption fine structure spectroscopy analysis showed that during the steam treatment, the surface Ni atoms were oxidized, and then these oxides were completely reduced to metallic Ni during the subsequent hydrogen reduction. The evolution analysis in surface morphology during pretreatment showed that many fine Ni particles formed on the surface. This surface structure was considered to be catalytically active for the methane steam reforming which is known to be structure sensitive.

IntroductionThe rapid spread of COVID-19 posed a global burden. Substantial number of people died of the disease in the acute phase of infection. In addition, a significant proportion of patients have been reported to suffer from post-acute phase symptoms, sequelae of COVID-19, which may negatively influence the quality of daily living and/or socioeconomic circumstances of the patients. However, no previous study has comprehensively and objectively assessed the quality of life of patients by using existing international scales. Further, evidence of socioeconomic consequences among patients with COVID-19 is scarce. To address the multidimensional issues from sequelae of COVID-19, evidence from comprehensive surveys beyond clinical perspectives is critical that investigates health, and social determinants of disease progression as well as socioeconomic consequences at a large scale.Methods and analysisIn this study, we plan to conduct a nationwide and comprehensive survey for the sequelae of COVID-19 in a total of 1000 patients diagnosed at 27 hospitals throughout Japan. This study will evaluate not only the health-related status of patients from clinical perspectives but also the Health-related Quality of Life (HRQoL) scores, socioeconomic status and consequences to discuss the sequelae of the disease and the related risk factors. The primary endpoint is the frequency of long-term complications of COVID-19 infection. The secondary endpoints are risk factors for progression to sequelae of COVID-19 infection. The study will provide robust and important evidence as a resource to tackle the issues from the sequelae of COVID-19 from the multi-dimensional perspectives.Ethics and disseminationThis trial was approved by the Keio University School of Medicine Ethics Committee (20200243, UMIN000042299). The results of this study will be reported at a society meeting or published in a peer-reviewed journal.

A tightly rolled pure Ni coil catalyst was assembled to ensure a stable catalytic performance in a wide range of conditions for methane steam reforming. With a high geometric specific surface area of 88.1 cm2/cm3, the coil catalyst achieved the objective of this study in the range of the space velocity (455-2800 h−1), steam-to-carbon ratio (0.62–2.48), and temperature (973–1073 K), which was hard to achieve on our previously developed Ni honeycomb catalyst with a geometric specific surface area of 59.4 cm2/cm3. Methane steam reforming reaction was slightly accelerated by the reactant flow because of the mass transfer enhancement of H2, CO, and CO2. The conversion, selectivity, and yield of the products were evaluated as a function of the conditions mentioned above. The high H2 production rate per unit catalyst volume demonstrated the high potential of the Ni coil catalyst for applications to small-scale hydrogen production system.

Third-generation epidermal growth factor receptor (EGFR) tyrosine kinase inhibitors (EGFR-TKIs) were developed to overcome EGFR T790M-mediated resistance to first- and second-generation EGFR-TKIs. Third-generation EGFR-TKIs, such as osimertinib and nazartinib, are effective for patients with the EGFR T790M mutation. However, there are no direct comparison data to guide the selection of a third-generation EGFR-TKI for patients with different EGFR mutations. We previously established an in vitro model to estimate the therapeutic windows of EGFR-TKIs by comparing their relative efficacies against cells expressing mutant or wild type EGFRs. The present study used this approach to characterize the efficacy of third-generation EGFR-TKIs and compare them with that of other EGFR-TKIs. Treatment efficacy was examined using human lung cancer-derived cell lines and Ba/F3 cells, which were transduced with clinically relevant mutant EGFRs. Interestingly, mutation-related differences in EGFR-TKI sensitivity were observed. For classic EGFR mutations (exon 19 deletion and L858R, with or without T790M), osimertinib showed lower IC50 values and wider therapeutic windows than nazartinib. For less common EGFR mutations (G719S or L861Q), afatinib showed the lowest IC50 values. For G719S+T790M or L861Q+T790M, the IC50 values of osimertinib and nazartinib were around 100 nM, which was 10- to 100-fold higher than those for classic+T790M mutations. On the contrary, osimertinib and nazartinib showed similar efficacies in cells expressing EGFR exon 20 insertions. The findings highlight the diverse mutation-related sensitivity pattern of EGFR-TKIs. These data may help in the selection of EGFR-TKIs for non-small cell lung cancer patients harboring EGFR mutations.

Epidermal growth factor receptor (EGFR)-tyrosine kinase inhibitors (EGFR-TKIs) are widely used for the treatment of non-small cell lung cancers (NSCLCs) harboring EGFR-activating mutations. However, lung cancer cells inevitably acquire resistance to these EGFR-TKIs. The majority of patients whose lung cancer acquires resistance to EGFR-TKIs are subjected to treatment using cytotoxic agents. The present study aimed to determine if lung cancer cells acquiring resistance to EGFR-TKIs also develop altered sensitivity to cytotoxic agents. It was revealed that lung cancer cells that had developed resistance to EGFR-TKIs had increased sensitivity to gemcitabine and vinorelbine compared with EGFR-TKI naïve cells. The expression levels of ATP-binding cassette (ABC) transporter genes, including ABCC3, ABCC5 and ABCG2, were observed to be commonly repressed in EGFR-TKI-resistant cells. In addition, two cases were identified in which gemcitabine and vinorelbine exerted marked responses to lung cancers that had acquired resistance to EGFR-TKIs, even with late-line treatment. Therefore, it was proposed that gemcitabine and vinorelbine may be effective agents for patients with lung cancer previously treated with EGFR-TKIs.

Ni-25Fe and Ni-15Fe-13Mg (at.%) alloy nanoparticles were fabricated by a thermal plasma method, and their catalytic properties for methanol decomposition were examined in the 513–793 K temperature range. Both nanoparticles started showing activity for methanol decomposition above 593 K, with activity rapidly increasing with temperature. Ni-15Fe-13Mg showed much higher activity and less carbon deposition than Ni-25Fe. These results indicated that the addition of Mg in the Ni-Fe alloy significantly increased the activity, but suppressed carbon deposition. Characterization of the nanoparticles revealed that the Mg was segregated and a thin layer of MgO was formed on the surface of the nanoparticles. It is likely that the formation of MgO suppressed carbon deposition, but enhanced the activity by delaying the coarsening of nanoparticles during the reaction.

In order to establish the design methodology of a small axial fan, the axial fan with impeller diameter of 36 mm was designed, manufactured and tested. Especially, in order to investigate the influence of difference in blade cord length and blade thickness on performance characteristics, the performance characteristics obtained by the designed axial fans with difference blade shapes were examined. Also, by using CFD, the same flow field as the experiment was visualized. It was found that the lift of the blade was increased and the performance was improved in high flow rate region by thinning of the blade thickness and by extending the blade chord length.

In the operation of a centrifugal compressor of turbocharger, instability phenomena such as rotating stall and surge are induced at a lower flow rate close to the maximum pressure ratio. In this study, the compressed air at the exit of centrifugal compressor was re-circulated and injected to the impeller inlet by using two injection nozzles in order to suppress the surge phenomenon. The most effective circumferential position was examined to reduce the flow rate at the surge inception. Moreover, the influences of the injection on the fluctuating property of the flow field before and after the surge inception were investigated by examining the frequency of static pressure fluctuation on the wall surface and visualizing the compressor wall surface by oil-film visualization technique.

The initial oxidation behavior of a clean Ni 3 Al (210) surface was studied at 300 K using a supersonic O 2 molecular beam (O 2 SSMB) having an O 2 translational energy of 2.3 eV, and real-time photoemission spectroscopy performed with high-brilliance synchrotron radiation. The evolution behaviors of the O 1s, Ni 2p, Al 2p, and Ni 3p spectra were examined during irradiation with the O 2 SSMB. The spectral analysis revealed that both the Al atoms and the Ni atoms on the surface were oxidized; however, the oxidation of Al progressed much faster than that of Ni. The oxidation of Al began to occur and AlO x was formed at an oxygen coverage of 0.26 monolayer (ML) (1 ML was defined as the atomic density of the Ni 3 Al (210) surface) and saturated at an oxygen coverage of 2.5 ML. In contrast, the oxidation of Ni commenced a little late at an oxygen coverage of 1.6 ML and slowly progressed to saturation, which occurred at an oxygen coverage of 4.89 ML.

Activation of the EGFR pathway is one of the mechanisms inducing acquired resistance to anaplastic lymphoma kinase (ALK) tyrosine kinase inhibitors (TKI) such as crizotinib and alectinib. Ceritinib is a highly selective ALK inhibitor and shows promising efficacy in non–small cell lung cancers (NSCLC) harboring the ALK gene rearrangement. However, the precise mechanism underlying acquired resistance to ceritinib is not well-defined. This study set out to clarify the mechanism in ALK-translocated lung cancer and to find the preclinical rationale overcoming EGFR pathway–induced acquired resistance to ALK-TKIs. To this end, ceritinib-resistant cells (H3122-CER) were established from the H3122 NSCLC cell line harboring the ALK gene rearrangement via long-term exposure to ceritinib. H3122-CER cells acquired resistance to ceritinib through EGFR bypass pathway activation. Furthermore, H3122 cells that became resistant to ceritinib or alectinib through EGFR pathway activation showed cross-resistance to other ALK-TKIs. Ceritinib and afatinib combination treatment partially restored the sensitivity to ceritinib. Implications: This study proposes a preclinical rationale to use ALK-TKIs and afatinib combination therapy for ALK-translocated lung cancers that have acquired resistance to ALK-TKIs through EGFR pathway activation. Background: Endobronchial ultrasound-guided transbronchial needle aspiration (EBUS-TBNA) is a widely available diagnostic tool for suspected stage I/II sarcoidosis. Combination of EBUS-TBNA and transbronchial lung biopsy (TBLB) has been proposed as diagnostic procedure in clinical settings. Objectives: The aim of this study was to assess the diagnostic yield of combined EBUS-TBNA and TBLB and identify the markers correlated with a high diagnostic rate. Methods: We retrospectively analyzed the data of 37 patients with suspected stage I/II sarcoidosis with enlarged hilar or mediastinal lymph nodes on computed tomography (CT) images. These patients had been scheduled to undergo EBUS-TBNA and TBLB. Serum levels of sarcoidosis markers (angiotensin-converting enzyme [ACE], soluble interleukin-2 receptor [sIL-2R], and lysozyme), CT findings, and examination techniques were evaluated as predictive markers for diagnosis. Results: Of the 37 patients, 32 had undergone both EBUS-TBNA and TBLB, while the remaining 5 patients had only undergone EBUS-TBNA. The diagnosis was confirmed by TBLB in 16 of the 32 patients (50.0%), EBUS-TBNA in 31 of the 37 patients (83.8%), and combined TBLB and EBUS-TBNA in all patients (100.0%). The serum level of sIL-2R, but not that of ACE or lysozyme, was correlated with successful diagnosis by EBUS-TBNA. Conclusion: In patients with stage I/II sarcoidosis, the serum level of sIL-2R is a promising and useful marker for predicting the diagnosis by EBUS-TBNA and reducing the burden of additional TBLB and its possible complications. We had developed the third-generation density functional (3G DF) method, which performed electronic state calculation with high efficiency in parallel computer by performing only matrix operation during the SCF calculation. Recently, hole mobilities in polyethylene (PE) oligomers has been investigated by means of Marcus theory. Although it is well known that Marcus theory is successfully applied to various organic semiconductors, strictly speaking, charge transfer in organic molecules requires quantum mechanical treatment to account for quantum nuclear tunneling between vibrational levels. We evaluate hole transfer rates between PE oligomers by Fermi's golden rule (FGR) rate expression and by classical Marcus rate expression at various temperature and energetic disorder. The results indicate that Marcus theory can be a good approximation for describing hole transfer between long polyethylene chain with small reorganization energy or in highly energetically disordered system. However, hole hopping rate between C10H22 molecules computed by Marcus theory was roughly 100 times smaller than that computed through FGR rate expression. High-temperature approximation must be applied with caution especially for quantitative evaluation of hole transfer rates in PE.

Hole mobility in crystalline polyethylene (PE) is evaluated from an atomistic point of view, with the combination of quantum chemical calculation and kinetic Monte Carlo simulations. Hole hopping rate between PE chains are computed by Fermi’s golden rule and Marcus rate expression. It turns out that hole transfer in PE occurs in a hopping regime rather than in a band regime even for crystalline structure without any inherent structural disorders. The results indicate that in crystalline PE, hole hops between localized states that are created by polaronic effect. The hole mobility is strongly dependent on the chain length, and increases with increasing chain-length. When the chain length was comparable to the thickness of lamellar crystals, i.e., the characteristic length scale of crystal in the direction of c axis, hole mobility and activation energy for hole transfer were in reasonable agreement with experimental values. Simulated results support the experimental prediction that fast and slow mobility are due to charge transfer in the crystalline region and amorphous region, respectively. Our findings show that high hole mobility observed in crystalline PE, regardless of the small inter-molecular electronic couplings, is due to the small reorganization energy, which is realized through the intra-molecular hole delocalization, and due to quantum tunneling effects owing to the small energetic disorder and high-frequency intra-molecular phonon modes.

A pretreatment procedure consisting of water vapor oxidation followed by H2 reduction has been developed for enhancing the catalytic activity of as-rolled Ni3Al foils for steam reforming of methane. The catalytic activities of the as-rolled Ni3Al foils in the temperature range of 873–1023 K at a steam to carbon ratio of 1.0 were found to be significantly improved by the pretreatment procedure consisting of a water vapor oxidation at 873–923 K for 1 h followed by a H2 reduction at 873 K for 1 h. Surface characterization of the foils before and after the reactions revealed that many fine NiO particles were formed on the surface by the water vapor oxidation at 873–923 K, whereas much less NiO particles were formed by the water vapor oxidation at low temperatures ( 973 K). During the subsequent H2 reduction treatment at 873 K, These NiO particles were reduced to metallic Ni. It is supposed that the enhancement in catalytic activity is attributed to the fine metallic Ni particles formed on the foil surface by the pretreatment procedure.

In recent years, excess electron transfer in organic liquids has attracted increasing interest owing to the emerging class of liquid organic semiconductors. In this study, to achieve a comprehensive understanding of electron conduction in liquids, we investigate hopping electron conduction in liquids from an atomistic viewpoint. High-pressure liquid benzene is chosen as a simple model system. Hopping electron mobility is computed using a combination of molecular dynamics simulations, quantum chemical calculations, and kinetic Monte Carlo methods. The computed electron mobility is in good agreement with the experimental values. Because the amplitude of the intermolecular vibration observed in liquids is larger compared to that in solids, the effect of dynamic disorder on electron mobility is investigated. Epidermal growth factor receptor (EGFR) mutation-positive lung cancer accounts for a significant subgroup of non-small cell lung cancers (NSCLC). Approximately 4-10% of EGFR mutations in NSCLC are EGFR exon 20 insertion mutations, which are reportedly associated with resistance to EGFR tyrosine kinase inhibitor (EGFR-TKI) treatment. NSCLC patients carrying these mutations are rarely treated with EGFR-TKIs. The purpose of this study was to evaluate the efficacy of afatinib or osimertinib plus cetuximab combination therapy in experimental NSCLC models with EGFR exon 20 insertion mutations.The EGFR mutations examined in this study were A763_Y764insFQEA, Y764_V765insHH, A767_V769dupASV, and D770_N771insNPG. Ba/F3 cells constitutively expressing wild type or mutated EGFR were used to determine the efficacy of afatinib or osimertinib plus cetuximab combination therapy in vitro. To determine the efficacy of the combination therapy in vivo, female BALB/c-nu mice were injected subcutaneously with 1 million Ba/F3 cells carrying EGFR A767_V769dupASV or Y764_V765insHH.We observed a mild but significant (P 0.05) additive effect of the combination therapy against several EGFR exon 20 insertion mutations in vitro. Regarding EGFR A767_V769dupASV and EGFR Y764_V765insHH, cetuximab and afatinib single treatment did not induce significant inhibition of tumor formation; however, afatinib plus cetuximab combination treatment induced significant (P 0.05) tumor growth inhibition without significant body weight loss or skin rash.The combination therapy induced a more potent inhibitory effect against several EGFR exon 20 insertion mutations than either therapy alone. Cetuximab can potentially increase the efficacy of afatinib or osimertinib in NSCLC with EGFR exon 20 insertion mutations.

The catalytic properties of single-phase Ni3Sn powder in the production of hydrogen via the decomposition of methanol were investigated in isothermal tests at 713, 793, and 873 K. The catalytic activity of Ni3Sn significantly increased with time at 793 and 873 K, but not at 713 K, suggesting that Ni3Sn is spontaneously activated at temperatures above 793 K. At these temperatures, Ni3Sn showed high selectivity for H2 and CO production and low selectivity for CH4, CO2, and H2O production, indicating that methanol decomposition was the main reaction, and that side reactions such as methanation and water–gas shift reaction were suppressed. Surface analysis revealed that fine Ni3Sn particles were formed during the reaction, accompanied by a small amount of deposited carbon. The formation of these particles was suggested to be the cause for the spontaneous activation of Ni3Sn.
