Optimal laser control for ultrafast selection of closely lying excited states whose energy separation is smaller than the laser bandwidth is reported on the two-photon transition of atomic cesium; Cs (6S→7D
j , J=5/2 and 3/2). Selective excitation was carried out by pulse shaping of ultrashort laser pulses which were adaptively modulated in a closed-loop learning system handling eight parameters representing the electric field. Two-color fluorescence from the respective excited states was monitored to measure the selectivity. The fitness used in the learning algorithm was evaluated from the ratio of the fluorescence yields. After fifty generations, a pair of nearly transform-limited pulses were obtained as an optimal pulse shape, proving the effectiveness of the "Ramsey fringes" mechanism. Spherical silicon solar cell is solar cell that uses silicon spheres of 1 mm in diameter. Purpose of my research is establishment of a manufacturing technique of the spherical silicon. The spherical silicon is produced by the dropping solidification method: a capillary jet of liquid metal is split into droplets and the droplets are solidified during free fall. Minagawa et al. found that mono-dispersed droplets could be generated by applying an intermittent electromagnetic pinch force to a liquid metal jet. However, the droplets frequently coalesced during free fall before completing its solidification, therefor mono-dispersed spherical silicon could not be obtained. In order to solve this problem, an electrification method was investigated in this study. The droplets are electrified by applying a high voltage. It is expected that a repulsive force will act upon the charged droplets and prevent them from coalescence. Experiments on droplets electrification were carried out with capillary jet of water and liquid gallium. Droplet charges were measured by an electrometer. It was found that the droplet charge depends on some parameters such as a jet diameter, shape of an electrode and a value of applied voltage. The experimental results were compared with a theoretical analysis, which has proposed by Brandenberger et al., and the agreement between the two was quite well. It was concluded that a droplet charge can be estimated by the theoretical model.

Developments are discussed which can be used to improve the accuracy of microseismic monitoring for seismic event location, and case studies are presented for two North American hard rock mines. In the first, a comparison was made between the use of a conventional single velocity model and a variable 3D velocity model, which accounts for voids and different geological domains, on location accuracy at a mine employing the room and pillar mining method. The results showed that the modal average for event location accuracy improved from 14 m to 5 m using the 3D velocity models, while the single velocity model failed to detect 26% of the seismic events located by the 3D velocity model. In the second case study, the distribution of seismic events was further enhanced by using a collapsing method, an algorithm which relocates individual events within their error ellipsoids. The method was applied to two large event sequences which occurred in close proximity at a mine using the stoping method, and two different seismic event distributions were identified. Tomato mottle mosaic virus (ToMMV) is an emerging seed-transmissible tobamovirus that infects tomato and pepper. Since the first report in 2013 in Mexico, ToMMV has spread worldwide, posing a serious threat to the production of both crops. To prevent the spread of this virus, early and accurate detection of infection is required. In this study, we developed a detection method for ToMMV based on reverse-transcription loop-mediated isothermal amplification (RT-LAMP). A LAMP primer set was designed to target the genomic region spanning the movement protein and coat protein genes, which is a highly conserved sequence unique to ToMMV. This RT-LAMP detection method achieved 10-fold higher sensitivity than conventional RT-polymerase chain reaction methods and obtained high specificity without false positives for closely related tobamoviruses or healthy tomato plants. This method can detect ToMMV within 30 min of direct sampling of an infected tomato leaf using a toothpick and therefore does not require RNA purification. Given its high sensitivity, specificity, simplicity, and rapidity, the RT-LAMP method developed in this study is expected to be valuable for point-of-care testing in field surveys and for large-scale testing.

A velocity model is presented which calculates microseismic event locations and can account for multiple complex shaped geological units. The method allows the addition of multiple voids, which could be air filled, brine filled, or cement paste backfilled. Source mechanism results are displayed in three different ways: beach balls, strain tensor, and discrete fracture network. The model was applied to two case studies in North America. In the first, the use of a 3D velocity model with six horizontal layers and the 3D shape of a brine filled cavern is allowing microseismic events to be accurately resolved, especially those occurring near to the boundaries of the cavern. Objective: The regulation of food intake is a major research area in the study of obesity, which plays a key role in the development of metabolic syndrome. Gene targeting studies have clarified the roles of hypothalamic neurons in feeding behavior, but the deletion of a gene has a long-term effect on neurophysiology. Our understanding of short-term changes such as appetite under physiological conditions is therefore still limited. Methods: Targeted recombination in active populations (TRAP) is a newly developed method for labeling active neurons by using tamoxifen-inducible Cre recombination controlled by the promoter of activity-regulated cytoskeleton-associated protein (Arc/Arg3.1), a member of immediate early genes. Transgenic mice for TRAP were fasted overnight, re-fed with normal diet, and injected with 4-hydroxytamoxifen 1 h after the refeeding to label the active neurons. The role of labeled neurons was examined by expressing excitatory or inhibitory designer receptors exclusively activated by designer drugs (DREADDs). The labeled neurons were extracted and RNA sequencing was performed to identify genes that are specifically expressed in these neurons. Results: Fasting-refeeding activated and labeled neurons in the compact part of the dorsomedial hypothalamus (DMH) that project to the paraventricular hypothalamic nucleus. Chemogenetic activation of the labeled DMH neurons decreased food intake and developed place preference, an indicator of positive valence. Chemogenetic activation or inhibition of these neurons had no influence on the whole-body glucose metabolism. The labeled DMH neurons expressed prodynorphin (pdyn), gastrin-releasing peptide (GRP), cholecystokinin (CCK), and thyrotropin-releasing hormone receptor (Trhr) genes. Conclusions: We identified a novel cell type of DMH neurons that can inhibit food intake and promote feeding-induced positive valence. Our study provides insight into the role of DMH and its molecular mechanism in the regulation of appetite and emotion.
