Novel evidence is presented allowing further clarification of the mechanism of the slow-binding thymidylate synthase (TS) inhibition by N 4 -hydroxy-dCMP (N 4 -OH-dCMP). Spectrophotometric monitoring documented time- and temperature-, and N 4 -OH-dCMP-dependent TS-catalyzed dihydrofolate production, accompanying the mouse enzyme incubation with N 4 -OH-dCMP and N 5,10 -methylenetetrahydrofolate, known to inactivate the enzyme by the covalent binding of the inhibitor, suggesting the demonstrated reaction to be uncoupled from the pyrimidine C(5) methylation. The latter was in accord with the hypothesis based on the previously presented structure of mouse TS (cf. PDB ID: 4EZ8), and with conclusions based on the present structure of the parasitic nematode Trichinella spiralis , both co-crystallized with N 4 -OH-dCMP and N 5,10 -methylenetetrahdrofolate. The crystal structure of the mouse TS-N 4 -OH-dCMP complex soaked with N 5,10 -methylenetetrahydrofolate revealed the reaction to run via a unique imidazolidine ring opening, leaving the one-carbon group bound to the N(10) atom, thus too distant from the pyrimidine C(5) atom to enable the electrophilic attack and methylene group transfer.

Despite the substantial amount of genomic and transcriptomic data available for a wide range of eukaryotic organisms, most genomes are still in a draft state and can have inaccurate gene predictions. To gain a sound understanding of the biology of an organism, it is crucial that inferred protein sequences are accurately identified and annotated. However, this can be challenging to achieve, particularly for organisms such as parasitic worms (helminths), as most gene prediction approaches do not account for substantial phylogenetic divergence from model organisms, such as Caenorhabditis elegans and Drosophila melanogaster, whose genomes are well-curated. In this paper, we describe a bioinformatic strategy for the curation of gene families and subsequent annotation of encoded proteins. This strategy relies on pairwise gene curation between at least two closely related species using genomic and transcriptomic data sets, and is built on recent work on kinase complements of parasitic worms. Here, we discuss salient technical aspects of this strategy and its implications for the curation of protein families more generally.

Parasitic worms of the genus Trichinella (phylum Nematoda; class Enoplea) represent a complex of at least twelve taxa that infect a range of different host animals, including humans, around the world. They are foodborne, intracellular nematodes, and their life cycles differ substantially from those of other nematodes. The recent characterization of the genomes and transcriptomes of all twelve recognized taxa of Trichinella now allows, for the first time, detailed studies of their molecular biology. In the present study, we defined, curated, and compared the protein kinase complements (kinomes) of Trichinella spiralis and T. pseudospiralis using an integrated bioinformatic workflow employing transcriptomic and genomic data sets. We examined how variation in the kinome might link to unique aspects of Trichinella morphology, biology, and evolution. Furthermore, we utilized in silico structural modeling to discover and characterize a novel, MOS-like kinase with an unusual, previously undescribed N-terminal domain. Taken together, the present findings provide a basis for comparative investigations of nematode kinomes, and might facilitate the identification of Enoplea-specific intervention and diagnostic targets. Importantly, the in silico modeling approach assessed here provides an exciting prospect of being able to identify and classify currently unknown (orphan) kinases, as a foundation for their subsequent structural and functional investigation., (Copyright © 2016 Stroehlein et al.)

Trichinellosis is a globally distributed helminthic infection. There is a considerable interest in developing new anti-helminthic drugs affecting all the developmental stages of Trichinella. Acetazolamide (carbonic anhydrase (CA) inhibitor) involves a novel mechanism of action by inhibiting such an essential enzyme for parasite metabolism. This work aimed to study the effect of acetazolamide against different stages of T. spiralis in experimental animals. Mice were divided into three groups: group I: infected and treated with acetazolamide on day 2 post infection (P.I.), group II: infected and treated with acetazolamide on day 12 P.I., and group III: infected non-treated. From each group, small intestine and muscles were removed for histopathological and immunohistochemical studies. Also, total adult and muscle larval count were estimated. We found that acetazolamide was effective in reduction of both adult and muscle larval counts. When given early, the effect was more pronounced on the adults (62.7 %). However, the efficacy of the drug against muscle larvae was increased when given late (63 %). Improvement of the intestinal histopathological changes was observed in all the treated groups. Degeneration of encysted larvae with minimal pathologic changes of infected skeletal muscle was observed in the treated groups. Expression of matrix metalloproteinase-9 showed a statistically significant decrease in the intestinal and muscle tissues in all treated groups as compared to the control group. In conclusion, the present study revealed that acetazolamide, carbonic anhydrase inhibitor, could be a promising drug against both adults and larvae of T. spiralis.

Background: Thymidylate synthase is a housekeeping gene, designated ancient due to its role in DNA synthesis and ubiquitous phyletic distribution. The genomic sequences were characterized coding for thymidylate synthase in two species of the genus Trichinella, an encapsulating T. spiralis and a non-encapsulating T. pseudospiralis., Methods: Based on the sequence of parasitic nematode Trichinella spiralis thymidylate synthase cDNA, PCR techniques were employed., Results: Each of the respective gene structures encompassed 6 exons and 5 introns located in conserved sites. Comparison with the corresponding gene structures of other eukaryotic species revealed lack of common introns that would be shared among selected fungi, nematodes, mammals and plants. The two deduced amino acid sequences were 96% identical. In addition to the thymidylate synthase gene, the intron-less retrocopy, i.e. a processed pseudogene, with sequence identical to the T. spiralis gene coding region, was found to be present within the T. pseudospiralis genome. This pseudogene, instead of the gene, was confirmed by RT-PCR to be expressed in the parasite muscle larvae., Conclusions: Intron load, as well as distribution of exon and intron phases in thymidylate synthase genes from various sources, point against the theory of gene assembly by the primordial exon shuffling and support the theory of evolutionary late intron insertion into spliceosomal genes. Thymidylate synthase pseudogene expressed in T. pseudospiralis muscle larvae is designated a retrogene.

Nematode parasites of the genus Trichinella are intracellular and distinct life cycle stages invade intestinal epithelial and skeletal muscle cells. Within the genus, Trichinella spiralis and Trichinella pseudospiralis exhibit species-specific differences with respect to host-parasite complex formation and host immune modulation. Parasite excretory-secretory (ES) proteins play important roles at the host-parasite interface and are thought to underpin these differences in biology. Serine proteases are among the most abundant group of T. spiralis ES proteins and multiple isoforms of the muscle larvae-specific TspSP-1 serine protease have been identified. Recently, a similar protein (TppSP-1) in T. pseudospiralis muscle larvae was identified. Here we report the cloning and characterisation of the full-length transcript of TppSP-1 and present comparative data between TspSP-1 and TppSP-1.

Trichinella species are parasitic nematodes of great economical and health importance. In this study, approximately 400 bp of the mitochondrial cytochrome c-oxidase (COI) gene from six species of Trichinella parasites from China and two reference species (T3 and T5) was sequenced and compared. The results showed that parasites from Yunnan, Hubei, and Dongbei (north-eastern) of China were very similar to the reference T1 isolated from Dongbei. The T2 parasite was, however, more closely related to the T5 (T. murrelli); and a parasite from Henan was very different from the rest of the parasites. These results suggested that genetic differences among most of the geographic strains in China existed. The Henan isolate may represent a new strain or species that requires further investigation.

The complete coding sequence of manganese superoxide dismutase (Mn-SOD) of Trichinella pseudospiralis (Tp) was obtained and characterized for the first time by degenerative reverse transcription polymerase chain reaction (RT-PCR) and rapid amplification of complementary DNA ends (RACE) reactions. The open reading frame of Tp Mn-SOD contained 663 nucleotides, encoding 220 amino acid residues. This included the conserved histidine and aspartate residues for metal binding, cysteine residues for disulfide bond formation, and arginine residue for directing the superoxide ion to the protein. The presence of mitochondrial transit peptides and maturation cleavage site suggest that the cloned Tp Mn-SOD gene is a mitochondrial enzyme. It is a single copy gene containing three introns. Northern blotting suggested that the expression level of Mn-SOD is lower than Cu/Zn SOD in infective stage larvae. Semi-quantitative RT-PCR demonstrated that a single dominant transcript of Tp Mn-SOD was highly expressed in the infective-stage larvae but not in adult worms. The information provides a better understanding of the highly compartmentalized superoxide dismutases of adenophorean nematodes.

Copper/zinc (Cu/Zn) superoxide dismutase (SOD) activity was identified for the first time in both crude somatic extracts (CE) and excretory/secretory (E/S) products of Trichinella pseudospiralis. It was the dominant SOD in infective-stage larvae. Native polyacrylamide gel electrophoresis of CE and E/S products yielded a prominent band, which was cyanide-sensitive and was partly inhibited by hydrogen peroxide in SOD assay. Cytosolic Cu/Zn SOD was cloned. The 471-bp full-length cDNA sequence contained an open reading frame of 157 amino acids. The gene contained three introns. Quantitative reverse transcription-polymerase chain reaction indicated that the expression of cytosolic Cu/Zn SOD was substantially higher in infective-stage larvae than in adult worms. Cluster analysis showed that the sequence of the Cu/Zn SOD of T. pseudospiralis, an adenophorean nematode, is related to those of Brugia pahangi, Acanthocheilonema viteae, Onchocerca volvulus, and Haemonchus contortus (all belonging to the sercenentean group).

In December 2001, the routine inspection of a wild boar intended for human consumption revealed the presence of Trichinella ssp. larvae. Biological, morphological and genetic analyses demonstrated the parasite to be Trichinella pseudospiralis. This is the second report of T. pseudospiralis in the United States and the first report of the parasite in a food animal species in the U.S.

Crude extract specific activities of thymidylate synthase, dUTPase, thymidine kinase and dihydrofolate reductase were high during the development of Caenorhabditis elegans, the dauer larva activities being similar to those previously determined in Trichinella spiralis and T. pseudospiralis muscle larvae (with the exception of thymidine kinase, not detected in Trichinella). High thymidylate synthase expression in developmentally arrested larvae, demonstrated also at the mRNA and protein levels, is in agreement with a global cell cycle arrest of dauer larvae and indicates this unusual cell cycle regulation pattern can be shared by developmentally arrested larvae of C. elegans and the two Trichnella species. Hence, the phenomenon may be characteristic for developmentally arrested larvae of different nematodes, rather than specific for the parasitic Trichinella muscle larvae. Endogenous C. elegans thymidylate synthase was purified and its molecular properties compared with those of the recombinant protein, expression of the latter in E. coli cells confirming the NCBI database sequence identity.

Infection of Trichinella spp. is widespread among wildlife in Slovakia and the red fox (Vulpes vulpes) is the main reservoir of Trichinella britovi. Trichinella spiralis has been rarely documented in sylvatic and domestic animals of this country. During routine examination of domestic pigs at the slaughter, Trichinella larvae were detected by artificial digestion in a domestic pig of a large-scale breeding farm in Eastern Slovakia. The parasite has been identified by molecular (PCR) and biochemical (allozymes) analyses and by the morphology of the nurse cell as the non-encapsulated species Trichinella pseudospiralis infecting both mammals and birds. The epidemiological investigation carried out at the farm level revealed the presence of the same parasite species in other three pigs of 192 examined (2.1%), in 3 of 14 (21.4%) examined synanthropic rats (Rattus norvegicus) and in a domestic cat. The farm was characterized by inadequate sanitary conditions, insufficient nutrition, cannibalism and the presence of rat population. A different profile has been observed at the phosphoglucomutase locus in T. pseudospiralis isolates from Slovakia in comparison with the T. pseudospiralis reference isolate from the Palearctic region. This is the first documented focus of T. pseudospiralis from Central Europe. The detection in domestic pigs of a non-encapsulated parasite infecting both mammals and birds stresses the need to avoid the use of trichinelloscopy to detect this infection at the slaughterhouse.

Nitric oxide has been extensively studied as an effector molecule of the host immune response against both protozoa and helminths, but parasites can also produce this molecule, through the action of nitric oxide (NO) synthases or NO synthases-like enzymes. The aim of this study was to verify the possible production of NO by Trichinella britovi L(1) larvae and the enzymes involved in this process. The NO synthase immunoreactivity and putative nitric oxide synthase-activity was analysed using antibodies to mammalian NO synthase III and to nitrotyrosine with immunohistochemistry, gold immunocytochemistry and immunoblot analysis and NADPH-diaphorase histochemistry. Our results show that T. britovi L(1) larvae possess an enzymatic activity capable of producing NO. The localisation of this activity, according to the NADPH-diaphorase histochemistry, is both at the cuticular and the internal level. This localisation is confirmed by nitrotyrosine immunohistochemistry both under optical and electron microscopy. Using the NO synthase III antibody, a similar pattern of labelling was found: in particular, electron microscopy showed a localisation of this immunoreactivity in the cuticle and in the stichocytes, where only the alpha2 granules contained gold particles, mainly concentrated at their periphery. Four polypeptides reacting to the NO synthase III antibody are revealed by Western blotting. Their molecular weight ranged from 38 to 50 kDa. A significant reaction of the anti-nitrotyrosine antibody to polypeptides 95, 60, 48 and 39 kDa from the same sample suggested the presence of different nitrosylated proteins.

The presence of cholinesterases (ChE) is reported in T. pseudospiralis excretion-secretion products (ESP) by spectrophotometric method, using acetylthiocholine (ATCI) and butyrilthiocholine (BTCI) as substrates. By inhibition assays, we found that T. pseudospiralis release both acetyl- and butiryl-cholinesterases (AchE and BchE, respectively). The sedimentation coefficientes of these enzymes were determined by sucrose density gradient. We studied the in vivo ChE secretion by immunoblot assays using AchE from Electrophorus (electric eel) and sera from normal or infected mice with T. pseudospiralis or T. spiralis. The presence of anti-AchE antibodies was only demonstrated in the sera from T. pseudospiralis infected mice. Moreover the in vivo secretion was corroborated by the high difference determinate between the ChE activity of the immuno complexes from T. pseudospiralis infected sera and the immunocomplexes from T. spiralis infected sera as well as normal sera. Finally, we analyzed the effect of the organophosphate Neguvón (metrifonate) on the ChE activity from the T. pseudospiralis ESP. The drug inhibits in part this activity. Moreover Neguvón (metrifonate) showed a high activity against the T. pseudospiralis viability.

Isoenzyme-based approach was applied to compare Trichinella spiralis, T. britovi and T. pseudospiralis species. Among 13 enzyme systems examined, esterase (EST), malic enzyme (ME) and phosphoglucomutase (PGM) have been found as fully diagnostic, with no common allele in species studied. Adenosine deaminase (ADA), adenylate kinase (AK), hexokinase (HK), peptidase leucyl-alanine (PEP-C) and fructose-bis-phosphatase (FBP) have been capable of distinguishing the two species from resulting profiles. In addition, ADA, AK and PGM displayed the enzyme expression in the lowest amounts of muscle larvae in systems tested (100 larvae/100 microliters of extracts). Based on allozyme data, T. pseudospiralis has been found as the most distinct species within the group of taxa. Only a subtle genetic variability was recorded for T. pseudospiralis in which solely phosphoglucomutase exhibited variant patterns. In addition to the study of reference isolates, T. spiralis from lowland fox in Eastern Slovakia has been evidenced by use of genetic markers. This finding has proved that T. britovi is not the exclusive species parasitizing in the sylvatic ecosystem of the Slovak region.

Delineation of the genus Trichinella into a more complex group of parasites has substantially motivated investigators to better identify and characterize the species and genotypes that form the basis of their investigations. Because of the cosmopolitan geographical distribution and broad host range that typify this genus, assigning unique biological, immunological and biochemical characters to each species and genotype has been essential for researchers to further advance this field. Numerous groups have developed simple methods to differentiate the genotypes, and by so doing, have generated diagnostic keys that accurately reflect the distinct differences among parasites of this group. Throughout the years, many methods have been used to accomplish this task, beginning with isoenzyme analyses and the use of repetitive DNA probes, to employing the polymerase chain reaction (PCR) and more state-of-the-art technologies. This review article summarizes the development of these methods with emphasis on molecular techniques and the ultimate goal of providing a simple, rapid and reproducible test to differentiate Trichinella parasites at the highest level of sensitivity, i.e. single parasite.

Thymidylate synthase, dihydrofolate reductase and dUTPase specific activities were found to remain at a high and constant level in crude extracts from adult worms of Trichinella spiralis, as well as from muscle larvae of both Trichinella spiralis (isolated 1-24 months after infection) and Trichinella pseudospiralis (isolated 5.5-13 months after infection). The results obtained with Trichinella pseudospiralis muscle larvae isolated with the use of pepsin did not differ from those obtained when pepsin was not used. No thymidine kinase activity could be detected in muscle larvae of either species and thymidine phosphorylase could be found only in T. pseudospiralis larvae isolated without the use of pepsin. Muscle larvae of both species contained orotidylate phosphoribosyl transferase activity, pointing to a possibility of 5-fluorouracil activation. Uridine phosphorylase, another enzyme involved in 5-fluorouracil anabolism, was also present in T. pseudospiralis muscle larvae. Results of comparative studies on inhibition of purified T. spiralis and rat thymidylate synthases by substrate (4-thio-5-fluoro-dUMP, 2-thio-5-fluoro-dCMP and N4-hydroxy-dCMP) and cofactor (ZD 9331) analogues indicated only dUMP analogues to show feeble selectivity towards the parasite enzyme. A hypothesis is discussed, assuming high expression of thymidylate synthase in muscle larvae to be connected with their cells being arrested in the cell cycle.

A novel acidic extracellular single-stranded endonuclease was demonstrated for the first time in the excretory-secretory (E-S) products of 2 species of Trichinella. Unlike the double-stranded endonuclease reported earlier, the single-stranded molecule is divalent cation independent and is detected in both T. spiralis and T. pseudospiralis E-S products. It hydrolysed single-stranded DNA and RNA at comparable rates. The single-stranded endonuclease was sensitive to inhibition by Zn2+ and to high concentrations of NaCl. Zymographic analysis indicated that it was encoded by at least 3 peptides of Mr approximately 50-60 kDa. The rate of hydrolysis of single-stranded targets by the E-S products was substantially higher than that of the double-stranded molecule. Due to the differences in peptide profile, divalent cation dependence, and species-specific expression, the single and double-stranded endonucleases are likely to be encoded by different proteins and may have different functions.

Eight taxa have recently been proposed as being encompassed by the genus Trichinella on the basis of allozyme and biological data. In this paper we show that an analogous 8 taxon structure for this genus results from the random amplified polymorphic DNAs (RAPDs). Five 10-mer or 20-mer primers were used under different polymerase chain reaction (PCR) conditions to produce multiband RAPD fingerprints from muscle larvae of 40 isolates of Trichinella spp. The resulting RAPD data were analysed following the numerical taxonomic approach, and the resulting classification was compared to that derived from allozyme data. The agreement found between allozymes and RAPDs, while supporting the polyspecific structure of the genus Trichinella, confirms the potential of RAPDs as a tool for the detection of cryptic species. The selected primers were tested on individual muscle larvae in an attempt to standardize a RAPD assay for the routine identification of the 8 taxa of Trichinella. Only 1 of the 5 primers yielded reproducible fingerprints from the single larvae. Using this primer, the 5 species and the 3 other taxa of the genus Trichinella can be identified in a single assay without the need for massive in vivo parasite production.

A new outbreak of trichinellosis occurred in France in December 1993 and involved around 550 patients. The authors report here how recent knowledge on Trichinella have been helpful to investigate this outbreak.

In four enzymes among eight ones examined the differences were revealed in the intensity of staining of certain zones (G-6-P, ES) as well as in electrophoretic mobility of some loci (PGD, RGM) between the capsule forms of trichinellae and Trichinella pseudospiralis. As a result, the validity of isolation of T. pseudospiralis Garkavi, 1972 is confirmed.

The aim of this study was to compare the in vitro and in vivo activities of superoxide dismutase, an enzyme acting against free-radicals, in Trichinella spiralis and T. pseudospiralis isolates. In vitro, the activity of the superoxide dismutase in T. pseudospiralis was determined to be about two times higher than in T. spiralis. Only one band representing superoxide dismutase was found by electrophoretic analysis. In vivo, in muscles infected with T. spiralis, a strong histochemical reaction in the capsule and the layer directly adhering to it was found. In muscles infected with T. pseudospiralis, the activity was most marked in the vicinity of the larva and becomes less marked with the distance. The character of the reaction indicated that the enzyme is excreted by the larvae.

Biological, biochemical, and genetic relationships of Trichinella isolates were assessed and compared from 3 species of Illinois fur-bearing mammals. Tongue muscle collected from 1987 through 1989 from 323 raccoons (Procyon lotor), 9 red fox (Vulpes fulva), and 1 coyote (Canis latrans) were digested and Trichinella muscle larval prevalences and mean intensities (larvae/g) determined. The prevalence and mean intensity of tongue muscle-stage larvae were 2.8% and 44.4% and 326 and 2 larvae/g for raccoon and red fox, respectively. The single coyote examined for muscle larvae was negative. Seven of 13 Trichinella isolates (5 raccoon, 2 red fox) were maintained and amplified in mice. Comparative analyses of DNA from larvae of these isolates and from other Trichinella isolates were performed by dot-blot hybridization using specific repetitive DNA probes. DNA from the 7 Illinois sylvatic isolates reacted with the Trichinella T5 isolate DNA probe pUPB-3.7 and did not cross-react with the Trichinella spiralis probe pBP-2. The sylvatic isolates of Trichinella were also analyzed using 27 allozymes. Their allozymic patterns were similar to Trichinella reference strain T5 from Pennsylvania, except for mannose phosphate isomerase. These results suggest that the Illinois wildlife isolates belong to the Trichinella T5 genotype according to the classification system established by the International Trichinella Reference Centre and as defined here by positive hybridization to the pUPB-3.7 probe. Results from Southern blot analyses using rRNA as a probe and allozyme patterns revealed some heterogeneity among geographical isolates of the T5 genotype.

The taxonomic status of variants within the genus Trichinella is problematical. Some authors recognise no fewer than four species (Trichinella spiralis, T. pseudospiralis, T. nativa and T. nelsoni), others regard T. nativa and T. nelsoni as strains of T. spiralis (T. spiralis var nativa or sylvatica), while others consider the genus to be monospecific, with a variety of more or less well defined isolates. Much of the current evidence adduced to support these various positions is similar to that used pre-1983. It derives from studies of the incidence of Trichinella infections in wild and in domestic animals, comparisons of infectivity of different isolates in laboratory animals and studies of immunity. However, it has become clear that infectivity and epidemiological studies are unreliable tools for discriminating between isolates of Trichinella and it has been shown that differences in the elicitation of immune responses are as much a function of the host as of the parasite. The introduction of monoclonal antibody technology has, however, permitted the identification of specific antigens in different isolates. The information is as yet scant, and one antigen does not a species make. Isozyme analysis provides some support for separating the various isolates of Trichinella into distinct groups, but cannot of itself shed light on the species problem until certain conditions are met. These conditions are difficult to achieve even in organisms abundantly available and without the baggage of the parasitic habit. Isozyme analysis is probably best used to support the newer studies of genomic DNA. Recent analyses of DNA by restriction endonucleases and dot-blot hybridisation techniques show ample promise of insights into speciation, and a new technique for amplifying the DNA from a single larva by the polymerase chain reaction offers exciting prospects. However, the position yet remains as stated in the first section of this abstract.

To evaluate biological and biochemical variability in nonencapsulated Trichinella isolates, biological and allozymic studies were conducted on isolates of Trichinella collected from a raptoral bird (Aquila rapax) and a fox (Vulpes corsac) in Kazakhstan and from a dasyurid marsupial (Dasyurus maculatus) on the island of Tasmania, Australia. Allozyme profiles of bird and marsupial isolates showed close similarity with the type isolate of Trichinella pseudospiralis. The avian and fox isolates successfully interbred with the type T. pseudospiralis isolate, but they failed to interbreed with 3 encapsulating species, Trichinella spiralis, Trichinella nativa, and Trichinella britovi. The reproductive index assessed in 4 inbred and 1 outbred strains of mice was lower for the avian isolate than for the marsupial and the type T. pseudospiralis isolates (P < 0.001).

The analysis of genetic, biochemical, and biological data on about 300 Trichinella isolates, reported in the literature, allows a taxonomic revision of this genus. We propose the recognition of 5 sibling species, Trichinella spiralis (Owen, 1835) sensu stricto; Trichinella nativa Britov and Boev, 1972; Trichinella pseudospiralis Garkavi, 1972; Trichinella nelsoni Britov and Boev, 1972 sensu stricto; and Trichinella britovi n. sp., on the basis of biochemical and biological characteristics. Trichinella britovi n. sp. is characterized by distribution in the Palaearctic Region; newborn larvae (NBL) production in vitro of 35-55 NBL/72 hr; nurse cell development time (NC d.t.) between 24 and 42 days postinfection (d.p.i.); low reproductive capacity index (RCI) in mice, rats, and pigs; low resistance to freezing; 1 unique marker allozyme; and moderate pathogenicity for humans. The new species is most similar to Trichinella nativa but differs from it in 4 allozymes, in having less resistance to freezing, in having a different pattern of major ribosomal DNA fragments after endonuclease digestion, and in distribution area. Trichinella nativa is characterized by a holarctic distribution; hosts that are sylvatic mammals; NBL production in vitro 28-54/72 hr; NC d.t. between 20 and 30 d.p.i.; low RCI in mice, rats, and pigs; high resistance to freezing; 2 unique marker allozymes; and moderate to severe pathogenicity for humans. Trichinella spiralis sensu stricto is characterized by a cosmopolitan distribution in domestic pigs, associated wildlife, and humans; high NBL production in vitro (greater than 90 NBL/72 hr); NC d.t. between 16 and 37 d.p.i.; high RCI in mice, rats, and pigs; no resistance to freezing; 6 unique marker allozymes; and high pathogenicity for humans.(ABSTRACT TRUNCATED AT 250 WORDS)

Allozyme analysis was carried out on 152 Trichinella isolates from synanthropic and wild animals and from humans; the isolates were collected from 5 continents. The analysis, involving 27 enzymes, revealed the presence of 8 distinct gene pools, termed T1-T8. Four of the genetic groups represent the 4 previously proposed species: Trichinella spiralis sensu stricto (T1), Trichinella nativa (T2), Trichinella nelsoni (T7), and Trichinella pseudospiralis (T4). The other 4, T3, T5, T6, and T8 are distinct from previously described species. The absence of allozymic hybrid patterns among even sympatric groups indicates a lack of gene flow among the groups. Principal component analysis and the unweighted pair group method of analysis were used to assemble allozyme patterns of the 152 isolates into discrete groups and to show their relative relationships. Both analyses indicated the presence of 8 primary clusters that correlated with the gene pools revealed by direct allozyme profile analysis. The absence of evidence of gene flow among the gene pools and the high level of allozymic differentiation between the cluster groups support the concept that the genus Trichinella is composed of several sibling species.

An isolate of Trichinella obtained from a wild boar in Yugoslavia did not form cysts in the musculature of its natural host. Subsequent inoculation into experimental hosts demonstrated that some larvae became encysted only after extended time periods, whereas others remained unencapsulated. Histological staining of larvae in the musculature demonstrated no deposition of collagen typically seen for Trichinella spiralis spiralis, Trichinella spiralis nativa, or Trichinella spiralis nelsoni. The Yugoslavian isolate, given the name of Zagreb isolate after the University where it was first studied, had low infectivity for pigs and mice. Isozyme analysis demonstrated greater homology with T. s. nelsoni than with other subspecies of Trichinella. Restriction fragment length polymorphisms and dot blot analyses further demonstrated the distinctive nature of this isolate. These results suggest that lack of cyst formation might be characteristic of isolates other than those designated Trichinella pseudospiralis and that this character might be important in the classification of Trichinella.

Isoenzyme analysis was used to characterize 47 isolates of Trichinella nelsoni from Europe (EUR) and two from Kenya and Tanzania (AFR). In all, 27 isoenzymes showed polymorphism within the species. The EUR parasites, isolated from domestic, synanthropic, sylvatic animals and man, showed isoenzymatic profiles different from those exhibited by AFR parasites isolated from sylvatic animals. The EUR parasites showed polymorphism due to three isoenzymes, i.e. ME, MPI and PGM. Euclidean distance and a dendrogram were used to evaluate the relationships among isolates. The Euclidean values showed that EUR and AFR genotypes are distant from each other and from the reference strains T. spiralis s.str. and T. pseudospiralis. The genetic data indicate the absence of gene flow between AFR and EUR isolates. The biochemical results suggest the presence of genetic heterogeneity in T. nelsoni and support the existence of a zoogeographical segregation of the two gene pools. The authors propose that as operational labels, code T3 be assigned to EUR isolates and code T7, to AFR isolates.

An isolate of Trichinella of polar bear origin was studied by isoenzymatic typing. It was found referable to Trichinella nativa. While the Wistar rats proved nearly refractory to this isolate, the Swiss albino mice were highly susceptible. Ninety-one per cent of the cystic lesions in the diaphragm of the polar bear contained viable larvae after over 20 years of acquisition of the infection by the host which is a case of extreme adaptability of the parasite to its host. The anatomo-pathological aspects of these lesions are studied and the zoonotic significance of this isolate examined.

Epidemiological investigations of an outbreak of trichinellosis were carried out in a domestic swine herd and it was established that the parasite also occurred in rats, and in skunks, opossums, and raccoons. Because considerable uncertainty exists regarding the role of sylvatic trichinellosis as a reservoir for the synanthropic cycle, studies were conducted to determine the genetic nature of the various isolates from this ecosystem. Pig infectivity trials, isoenzyme analyses, and repetitive DNA sequence analyses were performed. The results showed that all isolates from the farm environs were genetically similar and that they are related to Trichinella spiralis isolated from domestic pigs. The implication of these findings, in contrast to studies on isolates from wildlife elsewhere, is that this parasite is transmitted from domestic swine to sylvatic hosts and that any control or eradication efforts must take into account the potential for reinfection of hogs from wild animals.

Soluble protein profiles and isozyme patterns of eight enzymes were compared for extracts of muscle stage larvae of the seven Trichinella isolates, using isoelectric focusing in polyacrylamide gel. Soluble protein profiles and isozyme patterns of four enzymes: malic enzyme, glucosephosphate isomerase, phosphoglucomutase, superoxide dismutase of them were clearly divided into four types. T. pseudospiralis from a racoon and the Polar strain from a polar bear formed type 1 and type 2. The Iwasaki strain from a Japanese black bear and the Yamagata strain from a racoon dog, both from Japan, were type 3. Type 4 consisted of three remaining strains, the Polish strain from a wild pig, the USA strain from a pig and the Thai strain from a human case, which have similar infectivities to pigs. The Thai strain varied a bit electrophoretically from other members of type 4. Zymograms of adenylate kinase and malate dehydrogenase were similar in types 2 and 3. The 6-phosphogluconate dehydrogenase zymogram of type 3, similar to that of type 4, was different from that of type 2. It is assumed from the data that type 3 (Japanese strain) was genetically intermediate to types 2 and 4. T. pseudospiralis and the Polar strain had a common main isozyme of 6-phosphogluconate dehydrogenase. The zymogram of lactate dehydrogenase was common except for T. pseudospiralis.

In an attempt to find possible targets for benzimidazole action in muscle-stage larvae of Trichinella spiralis, the effects of mebendazole and thiabendazole were tested in vivo by oral treatment of infested mice and in vitro by including these anthelmintics in an adequate maintenance medium containing decapsulated larvae. The effects of the anthelmintics on succinate dehydrogenase and fumarate reductase activities, measured in the mitochondrial fraction obtained from the in vivo- or in vitro-treated larvae showed that only thiabendazole causes significant inhibition of fumarate reductase activity. On the other hand, measurements of free glucose, glycogen reserves and soluble protein in the treated larvae indicate that in vivo, mebendazole and thiabendazole clearly diminish free glucose levels, although in vitro only mebendazole produces the same diminution. Both the glycogen and protein contents of the larvae remained unchanged after treatment in vivo or in vitro. The importance of these findings with regard to a possible site of action for mebendazole and thiabendazole is discussed.

Twenty-three Trichinella isolates have been examined by the isoenzyme typing of ten enzyme systems (LDH, ME, 6PGDH, G6PDH, GOT, AK, PGM, ACON, MPI, GPI). Four different zymodemes were obtained. All the examined isolates have shown an electrophoretic behaviour like one or other of four reference strains. The isolates from Italy and Yugoslavia have an electrophoretic mobility like T. nelsoni reference strain. The isolates from France, Holland, Poland and U.S.A. have an electrophoretic mobility like T. spiralis reference strain. For T. nativa and T. pseudospiralis we have only the reference strains. These results support the validity of the taxonomy of Trichinella genus in four good species.

The exoglycosidases beta-N-acetyl-D-glucosaminidase, beta-N-acetyl-D-galactosaminidase, alpha-1-fucosidase, alpha-D-glucosidase and alpha-D-mannosidase, and a non-specific acid phosphohydrolase are present at high levels in extracts of adult and muscle-stage (L1) Trichinella spiralis and at lower (5-30-fold) levels in extracts of the newborn larvae. The enzyme activities from the L1 extract were characterized. All displayed maximum activity at acid pH. beta-N-acetyl-D-glucosaminidase and beta-N-acetyl-D-galactosaminidase had identical molecular weights (110 000), pH optima (5.0), and isoelectric points (5.7) indicating that both of these substrate specificities reside in the same protein molecule. alpha-1-Fucosidase had a molecular weight of 125 000 and exhibited two pH optima (5.0 and 6.0) and four isoelectric points (5.9, 6.4, 6.7 and 7.1) indicating its presence in multiple molecular forms. alpha-D-Glucosidase had a molecular weight of 85 000, a pH optimum of 6.0 and an isoelectric point of 5.2; alpha-D-mannosidase had a molecular weight of 192 000, a pH optimum of 6.0 and an isoelectric point of 4.5; and acid phosphatase had a molecular weight of 81 000, a pH optimum of 6.0 and two isoelectric points (4.8 and 5.9) indicating its existence in two molecular forms. The same glycosidases and acid phosphatase were detected also in culture fluids collected after 15-20-h incubation of both L1 and adults. As in the worm extracts, beta-N-acetyl-D-glucosaminidase was present in these culture fluids at the highest activity with acid phosphatase present at the next highest activity.

LDH isoenzymes of Trichinella larvae isolated from experimentally infected guinea pigs were investigated between days 30 and 400 inclusive after infection. It was found that on the 30 th day after infection the larvae have two isoenzymes while from the 40th to the 400 th day--three isoenzymes. The first of the latter is of fast responce and is located in the anod area near the indicator while the two others are of slow response and are located between the isoenzymes LDH-3 and LDH-5 of the muscular tissue of guinea pigs. This indicates that energetic metabolism of Trichinella larvae proceeds in muscular tissues by aerobic and anaerobic way. Since electrophoretic specificity of LDH-isoenzymes of Trichinella larvae differ from that of LDH-isoenzymes of muscular tissue the former are apparently characteristic of Trichinella larvae.

Type 4 consisted of three remaining strains, viz. the Polish strain from a wild pig, the USA strain from a pig, and the Thai strain from a human case, all of which have similar infectivity to pigs. The isozyme patterns of esterase 1, beta-N-acetylglucosaminidase, and peptidase were similar in types 2 and 3. Those of esterase D were common to types 2-4 but not to type 1. In the zymogram of mannosephosphate isomerase, types 2-4 but not type 1 had one common band, whereas in the other bands type 2 was markedly distinguished from types 3 and 4. In the present study, the molecular phylogenic tree was constructed for the first time on the basis of our present and previous electrophoretic data by the use of cluster analysis, and the evolutionary process was considered as follows: T. pseudospiralis (type 1) and T. spiralis (the common ancestor of types 2-4) were initially separated. Next, the common ancestor of the strains from wild carnivores (types 2 and 3) and type 4 were separated. Finally, the Polar strain (type 2) and the Japanese strain (type 3) were separated.

The isoenzyme profiles of five isolates of the supposed 'species' of Trichinella, T. nativa, T. spiralis and T. nelsoni were compared. Four enzymes (AK, PGM, MPI and GPI) gave good resolution and clearly differentiated T. Spiralis from the other two species. T. nativa and T. nelsoni had similar isoenzyme patterns; the two separate isolates of T. nativa and T. spiralis used gave similar results, thus indicating the validity and the reproducibility of the technique. The value of enzyme electrophoresis for specific and subspecific classification of Trichinella is discussed and compared with the more traditional methods of taxonomy which have failed to resolve the controversy surrounding speciation.

In order to understand why different stages of Trichinella spiralis vary in their susceptibility to killing by leukocytes, the effects of artificially generated oxidants on different stages of this parasite were compared. More than 90% newborn larvae were killed after incubation in acetaldehyde-xanthine oxidase or glucose-glucose oxidase. On the other hand, fewer than 10% of adult worms or muscle larvae were killed when incubated under identical conditions. Thus, only the stages which are resistant to killing by leukocytes are resistant to killing by oxidants. The larvicidal effect of acetaldehyde-xanthine oxidase was blocked by the addition of either superoxide dismutase or catalase and was partially inhibited by radical scavengers and singlet oxygen quenchers. The oxidant resistant adults and muscle larvae contained 3-5 times more superoxide dismutase and at least five times more glutathione peroxidase than the oxidant sensitive newborn larvae. In contrast, all 3 stages lacked detectable amounts of catalase and contained roughly equivalent amounts of reduced glutathione. Accordingly, adults and muscle larvae may be more resistant to killing by leukocytes than newborn larvae because they contain better oxidant defenses.

Succinate dehydrogenase and fumarate reductase activities from a particulate fraction of 4 isolates of the supposed 'species' of Trichinella, T. spiralis, T. pseudospiralis, T. nativa and T. nelsoni have been determined using spectrophotometric methods. The 'in vitro' effect of the anthelmintics thiabendazole, mebendazole and ivermectin on succinate dehydrogenase and fumarate reductase activities from the above isolates is also described. The significance of the results is discussed in the context of the controversy that surrounds speciation of the genus Trichinella.

The epidemiological picture of trichinellosis in Thailand is reported. In the last 25 years, about 3000 people have been affected with Trichinella and 85 patients died. The hill-tribe pig has been the main source of human infections. Parasites isolated from a patient in 1986 were shown, through isoenzymatic typing of 10 gene-enzyme systems, to be T. spiralis s.str. The epidemiological and zoogeographical implications are discussed.

Seven isolates of the genus Trichinella were evaluated for their genetic identity and variability using starch gel electrophoresis. The various strains exhibited polymorphism in 8 of 11 enzyme systems tested and the strains could be segregated into at least 3 basic patterns for geographical isolates of Trichinella: pigs, wild carnivores and Trichinella spiralis var. pseudospiralis. There was also evidence for further differences between isolates from wild carnivores. The isozyme technique appears to have potential for zoogeographic studies in the genus Trichinella.
