Background: Animal diseases that are endemic like tsetse transmitted trypanosomosis cause the continuous expenditure of financial resources of livestock farmers and loss of productivity of livestock. Estimating the cost of controlling animal trypanosomosis can provide evidence for priority setting and targeting cost-effective control strategies., Methodology: A cross-sectional survey to estimate the economic cost of bovine trypanosomosis was conducted in cattle-keeping communities living around Murchision falls National Park, in Buliisa district Uganda. Data was collected on herd structure, the cost of treatment and control, prevalence of morbidity and mortality rates due to trypanosomosis, and salvage sales losses in cattle herds in the last year., Results: In this study, 55.4% (n = 87) of the households reported their cattle had been affected by trypanosomosis during the previous last year. There was a high economic cost of trypanosomosis (USD 653) per household in cattle-keeping communities in Buliisa district of which 83% and 9% were due to mortality and milk loss respectively/ High mortality loss was due to low investment in treatment. The study showed that prophylactic treatment 3 times a year of the whole herd of cattle using Samorin ® (Isometamidium chloride) at a cost of USD 110 could drastically reduce cattle mortality loss due to trypanosomosis due to trypanosomosis with a return on investment of USD 540 annually per herd. This could be coupled with strategic restricted insecticide spraying of cattle with deltamethrin products., Conclusion: The results show a high economic cost of trypanosomosis in cattle-keeping communities in Buliisa district, with cattle mortality contributing the largest proportion of the economic cost. A cross-sectional study aimed to elucidate the prevalence of bovine trypanosomosis and its potential risk factors was conducted in tsetse suppression and non-suppression areas of South Omo Zone, Southern Ethiopia from November 2018- May 2019. A total of 1284 blood samples from local zebu cattle (642 each in dry and wet season) were examined by using buffy coat technique and thin blood smear method. The overall prevalence was 11.05 % with 14.33 % in dry and 7.78 % in wet season. According to multiple logistic regression analysis of tsetse suppression areas, higher prevalence in female than male (OR = 0.48, 95 % CI: 0.27, 0.83), in poor (OR = 3.25, 95 % CI: 1.26, 11.09) and medium (OR = 2.07, 95 % CI: 0.74, 7.37) than good body conditioned animals was recorded. Moreover, tethered animals (OR = 2.07, 95 % CI: 1.06, 3.92) were more likely to be infected than communal grazers and also higher prevalence in dry season than wet season (OR = 0.52, 95 % CI: 0.30, 0.87). Similarly, in tsetse non-suppression areas, higher prevalence in female than male (OR = 0.48, 95 % CI: 0.27, 0.85) and in wet season (OR = 0.41, 95 % CI: 0.23, 0.7) than dry season was recorded. Trypanosoma congolense and Trypanosoma vivax were found in cattle with the former more prevalent in both areas. Overall pooled mean packed cell volume (PCV) of parasitaemic animals (23.57 ± 3.13) was significantly lower than aparasitaemic animals (27.80 ± 4.95). Similarly, parasitaemic animals from tsetse suppression areas and tsetse non-suppression areas had significantly lower mean PCV than their aparasitaemic counterparts. Mean PCV of T. congolense (23.59 ± 3.22) infected animals was not different (P > 0.05) from T. vivax infected animals (23.26 ± 3.31). It was also indicated that the probability of anaemic animals to be parasitaemic was significantly higher (P < 0.05) than non-anaemic animals in both areas. In conclusion, the prevalence of trypanosomosis revealed its endemicity which bottlenecked the livestock production and productivity in the study area despite of tsetse suppression activities. A mixed method survey was conducted among pastoral and agro pastoral communities surrounding Murchison Falls National Park, Uganda to assess knowledge, attitudes and practices about control of bovine trypanosomosis. A total of 96.8% (n = 152) of the participants had seen tsetse flies, and close to 91.7% (n = 116) of the participants had heard about bovine trypanosomosis. Bovine trypanosomosis was reported as a major disease in their area by about 73.9% (n = 116). There was a significant difference (P < 0.05) in the level of awareness and perception about tsetse and bovine trypanosomosis across the study sub counties. The majority of the farmers (60.5%) stated that grazing near national parks was the main cause of bovine trypanosomosis. A small proportion of farmers associated sharing grazing land and watering points with wildlife (19.1%) and grazing cattle in tsetse fly-infested areas (8.3%) as the causes of trypanosomosis. The communities in the study sub counties were aware of at least one or two clinical signs of bovine trypanosomosis. Spraying cattle with insecticide and avoiding grazing animals in tsetse-infested areas were the control practices. Curative trypanocides were mainly used to treat their cattle against trypanosomosis. Bush clearing, targets and traps as tsetse fly control measures were less practiced by the farmers. Treatment of cattle was based on observation of clinical signs due to absence of blood diagnostic facilities. Implementing regular tsetse fly population monitoring surveys and promotion of disease rapid diagnostic tools at farm level as long-term strategies are key for effective control of the disease.

African animal trypanosomosis (AAT) is transmitted cyclically by tsetse flies and mechanically by biting flies (tabanids and stomoxyines) in West Africa. AAT caused by Trypanosoma congolense, T. vivax and T. brucei brucei is a major threat to the cattle industry. A mathematical model involving three vertebrate hosts (cattle, small ruminants and wildlife) and three vector flies (Tsetse flies, tabanids and stomoxyines) was described to identify elimination strategies. The basic reproduction number (R0) was obtained with respect to the growth rate of infected wildlife (reservoir hosts) present around the susceptible population using a next generation matrix technique. With the aid of suitable Lyapunov functions, stability analyses of disease-free and endemic equilibria were established. Simulation of the predictive model was presented by solving the system of ordinary differential equations to explore the behaviour of the model. An operational area in southwest Nigeria was simulated using generated pertinent data. The R0 < 1 in the formulated model indicates the elimination of AAT. The comprehensive use of insecticide treated targets and insecticide treated cattle (ITT/ITC) affected the feeding tsetse and other biting flies resulting in R0 < 1. The insecticide type, application timing and method, expertise and environmental conditions could affect the model stability. In areas with abundant biting flies and no tsetse flies, T. vivax showed R0 > 1 when infected wildlife hosts were present. High tsetse populations revealed R0 <1 for T. vivax when ITT and ITC were administered, either individually or together. Elimination of the transmitting vectors of AAT could cost a total of US$ 1,056,990 in southwest Nigeria. Hence, AAT in West Africa can only be controlled by strategically applying insecticides targeting all transmitting vectors, appropriate use of trypanocides, and institutionalising an appropriate barrier between the domestic and sylvatic areas., Competing Interests: The authors have declared that no competing interests exist.

African animal trypanosomiasis (AAT), a disease complex caused by tsetse fly-transmitted Trypanosoma brucei brucei, T. congolense savannah ITS, and T. vivax, continues to inflict heavy losses to the animal industry in terms of decreased livestock production and productivity. Live bait technology and chemotherapy have been used as a control strategy in northern Uganda since 2006 with minimal success. Here, we report the results of a cross-sectional study carried out in Lango subregion, Uganda, to assess the species prevalence of bovine trypanosome in cattle using the internal transcribed spacer (ITS) of trypanosome ribosomal DNA (rDNA). Blood samples were collected from 1090 cattle by ear vein puncture and screened using a single pair of primers designed to amplify ITS ribosomal DNA (rDNA). Our results indicate an overall prevalence of 40.18% (438/1090, 95% CI 30.82-54.51). T. vivax constituted 32.66% (356/1090), T. congolense 2.39% (26/1090), T. brucei 1.28% (14/1090), T. godfreyi 0.09%(1/1090), T. brucei and T. congolense 0.36% (4/1090), T. brucei and T. vivax 1.47% (16/1090), T. vivax and T. congolense 1.65% (18/1090), T. vivax and T. simiae 0.18% (2/1090), and T. vivax and T. godfreyi 0.09% (1/1090) of infections. Over 91.7% of infections involved single species, while 9.5% were mixed infections. Over 90.2% (37/41) of the mixed infections involved T. vivax as one of the species, while 53.7% (22/41) involved T. congolense. The high prevalence of AAT and the continued presence of T. brucei raise public health concerns because of the zoonotic implications. An integrated approach that involves mass treatment of cattle, vector, and animal movement control should be adopted to reduce the risk of both AAT and HAT.

In this study, we evaluated the therapeutic efficacy of diminazene diaceturate at a dose of 7 mg/kg (DA), imidocarb dipropionate at 4.8 mg/kg (IMD), isometamidium chloride at 0.5 and 1.0 mg/kg (ISM 0.5 and ISM 1.0) and combinations applied through different methods to treat Trypanosoma vivax in experimentally infected calves. Thirty male Girolando calves were kept indoors and infected intravenously with T. vivax trypomastigotes (approximately 1 × 10 6 ). On D-1, the calves were randomized based on the quantity of infecting parasites per animal, yielding six groups of five animals each: G1: positive control group without treatment; G2 animals treated with DA on Day 0 intramuscularly (IM); G3 animals treated with IMD on Day 0 and D + 14 subcutaneously; G4 animals treated with ISM 0.5 on Day 0 IM; G5 animals treated with ISM 1.0 on Day 0 IM; G6 animals received DA on Day 0 and ISM 1.0 on D + 14, both IM. Throughout 180 days, blood samples were collected for the evaluation of T. vivax using the Woo, Brener and PCR methods. The results indicated that the treatment protocols with DA and/or ISM 0.5 and ISM 1.0 had high efficacy (100 %) against T. vivax. Interestingly, cattle that received ISM remained free of parasites until D + 180. In contrast, animals treated with IMD had relapsed T. vivax detected on the 10 th and 14 th days post-treatment (DPT). Cattle that received ISM 1.0 did not exhibit relapsed T. vivax in the blood, even after reinfection performed on the 50 th DPT. However, treatment with DA on Day 0 failed to prevent a new infection of T. vivax on the 50 th DPT. The animals that received ISM 1.0 had a transient decrease in packed cell volume similar to that found in the control group. The reappearance of T. vivax in herds in Brazil treated with DA likely occurred due to the short half-life of the drug and not necessarily due to T. vivax resistance to DA., (Copyright © 2020 Elsevier B.V. Significant increases in human and livestock populations coupled with agricultural practices have changed the socioeconomic perspectives of livestock diseases. Evaluating the socioeconomic impact of bovine trypanosomosis and its vectors (Glossina, Tabanus and Stomoxys) from the perspective of the livestock owners is of great significance. Participatory rural appraisal was conducted among 209 livestock owners (focus groups) to determine the behavioural practices of animal husbandary to bovine trypanosomosis. In Nigeria, common Trypanosoma species found in cattle are Trypanosoma vivax, Trypanosoma congolense and Trypanosoma brucei. Trypanosomosis peaks were reported by owners to be in the months of March-August. A total of 70.8% (95%CI 64.32-76.56%) cattle owners perceived trypanosomosis as a major disease in their herd, 13.4% (95%CI 9.43-18.68%) practiced transhumance in the wet season and 93.9% (95%CI 88.58-96.92%) make use of trypanocides, and approximately US$ 8.4 million is spent annually on trypanocides in southwest Nigeria livestock industry. About 60.5% (95%CI 51.84-68.48) make use of insecticides against transmitting vectors, and only 1.9% (95%CI 0.75-4.82%) have ever heard of any form of government intervention scheme. Estimated losses ≥ US$ 426 (80-100% loss) can be incurred on a single animal depending on the size and market value. There is significant increase (16.2%, 95%CI 11.15-23.00%, P < 0.05) in the mortality rate of bovine trypanosomosis when compared to other livestock diseases. It will therefore be useful to involve the livestock owners with devising new and integrated measures for reducing the impact of this trypanosomosis.

The study assessed an integrated trypanosomosis control strategy in drug-resistant hotspot villages of northern Togo. This strategy comprised (i) rational trypanocidal drug use in symptomatic cattle, (ii) vectors and ticks control by targeted bi-monthly insecticidal spraying of the lower body parts of cattle and (iii) strategic deworming with Albendazole in the beginning and the end of the rainy season. The program was implemented between June 2014 and October 2015 in four villages in northern Togo, which had been previously identified as drug resistant hotspots for diminazene diaceturate (DA) and isometamidium chloride (ISM). The integrated control strategy was implemented in eight cattle herds at risk of the disease from two villages. Twelve herds from two other villages served as controls where trypanosomosis management and deworming remained under control of the farmers. Trypanocidal drug use during the study period was recorded by the intervention team based on the farmers' reports and own observations. Cattle herds were followed-up for trypanosomosis symptoms which were recorded at 3 to 4-month intervals, while extensive trypanosome diagnostics and recording of the packed cell volume were done before and after the intervention. Intervention herds had a significantly lower risk of trypanosome infection with a risk ratio of 0.18 (95% CI: 0.04, 0.91; p = 0.03), but no significant effect on mean packed cell volume was observed. However, trypanocidal treatments per animal per year were lower in intervention herds compared to control herds (0.3 vs 5 for DA and 0.8 vs 2 for ISM). The CRISPR-Cas system, which was originally identified as a prokaryotic defense mechanism, is increasingly being used for the functional study of genes. This technology, which is simple, inexpensive and efficient, has aroused a lot of enthusiasm in the scientific community since its discovery, and every month many publications emanate from very different communities reporting on the use of CRISPR-Cas9. Currently, there are no vaccines to control neglected tropical diseases (NTDs) caused by Trypanosomatidae, particularly Human African Trypanosomiasis (HAT) and Animal African Trypanosomoses (AAT), and treatments are cumbersome and sometimes not effective enough. CRISPR-Cas9 has the potential to functionally analyze new target molecules that could be used for therapeutic and vaccine purposes. In this review, after briefly describing CRIPSR-Cas9 history and how it works, different applications on diseases, especially on parasitic diseases, are reviewed. We then focus the review on the use of CRISPR-Cas9 editing on Trypanosomatidae parasites, the causative agents of NTDs, which are still a terrible burden for human populations in tropical regions, and their vectors., (Copyright © 2018 Elsevier B.V. Tsetse flies (Diptera, Glossinidae) transmit human and animal trypanosomoses in Africa, respectively a neglected human disease (sleeping sickness) and the most important constraint to cattle production in infested countries (nagana). We recently developed a methodology to map landscape friction (i.e. resistance to movement) for tsetse in West Africa. The goal was to identify natural barriers to tsetse dispersal, and potentially isolated tsetse populations for targeting elimination programmes. Most species distribution models neglect landscape functional connectivity whereas environmental factors affecting suitability or abundance are not necessarily the same as those influencing gene flows. Geographic distributions of a given species can be seen as the intersection between biotic (B), abiotic (A) and movement (M) factors (BAM diagram). Here we show that the suitable habitat for Glossina palpalis gambiensis as modelled by Maxent can be corrected by landscape functional connectivity (M) extracted from our friction analysis. This procedure did not degrade the specificity of the distribution model (P=0.751) whereas the predicted distribution area was reduced. The added value of this approach is that it reveals unconnected habitat patches. The approach we developed on tsetse to inform landscape connectivity (M) is reproducible and does not rely on expert knowledge. Despite extensive attempts over many decades to control African Animal Trypanosomiasis (AAT) across the tsetse fly belt of Nigeria, AAT persists as major animal health problem causing severe morbidity and mortality in livestock. The large agricultural losses in turn have severe adverse impacts on sustainable agricultural development. Despite this, in the past 50 years there have been no significant national control programs against AAT. Background: Animal African trypanosomiasis (AAT) and its tsetse vector are responsible for annual losses estimated in billions of US dollars ($). Recent years have seen the implementation of a series of multinational interventions. However, actors of AAT control face complex resource allocation decisions due to the geographical range of AAT, diversity of ecological and livestock systems, and range of control methods available., Methods: The study presented here integrates an existing tsetse abundance model with a bio-economic herd model that captures local production characteristics as well as heterogeneities in AAT incidence and breed. These models were used to predict the impact of tsetse elimination on the net value of cattle production in the districts of Mambwe, in Zambia, and Faro et Déo in Cameroon. The net value of cattle production under the current situation was used as a baseline, and compared with alternative publicly funded control programmes. In Zambia, the current baseline is AAT control implemented privately by cattle owners (Scenario Z0). In Cameroon, the baseline (Scenario C0) is a small-scale publicly funded tsetse control programme and privately funded control at farm level. The model was run for 10 years, using a discount rate of 5%., Results: Compared to Scenario C0, benefit-cost ratios (BCR) of 4.5 (4.4-4.7) for Scenario C1 (tsetse suppression using insecticide treatment of cattle (ITC) and traps + maintenance with ITC barrier), and 3.8 (3.6-4.0) for Scenario C2 (tsetse suppression using ITC and traps + maintenance with barrier of targets), were estimated in Cameroon. For Zambia, the benefit-cost ratio calculated for Scenarios Z1 (targets, ITC barrier), Z2 (targets, barrier traps), Z3 (aerial spraying, ITC barrier), and Z4 (aerial spraying, barrier traps) were 2.3 (1.8 - 2.7), 2.0 (1.6-2.4), 2.8 (2.3-3.3) and 2.5 (2.0-2.9), respectively. Sensitivity analysis showed that the profitability of the projects is relatively resistant to variations in the costs of the interventions and their technical efficiency., Conclusions: It is envisioned that the methodologies presented here will be useful for the evaluation and design of existing and future control programmes, ensuring they have tangible benefits in the communities they are targeting.

Background: Tsetse-borne African animal trypanosomosis (AAT) greatly influences livestock distribution and significantly slows livestock productivity in sub-Saharan Africa. While a number of control methods targeting the vector tsetse are in field application, treatment with the few available trypanocides continues to be the most widely applied control method. Unfortunately, improper and frequent use of these few available drugs, accelerated by poor veterinary service delivery, promotes trypanosome drug resistance, the magnitude of which has not been delineated. In the present study, current practices on trypanocide application for the control of bovine trypanosomosis in the field in Tanzania were studied with a view to providing policy advice on the safe and sustainable use of trypanocides., Methods: A cross-sectional study was conducted using a semi-structured questionnaire administered to a total of 200 randomly selected livestock keepers in selected pastoral and agropastoral areas within three districts in eastern and north eastern Tanzania., Results: In total, 50% of respondents in all three study districts had primary level education; over 40% had informal education and only 5% with university education (all from one district, Pangani). Age-wise, most respondents were aged 30-59 years with exception of Korogwe where 35% were aged 20-29 years. Over 95% of respondents had knowledge on tsetse as a vector of trypanosomosis and correctly identified tsetse in provided pictures. Furthermore, 98.7% of the respondents applied pyrethroids for vector control. Regarding parasite control practices, this study revealed a high degree of variation in trypanocides usage and the intervals of their application. Whereas only 20% of respondents use chemoprophylaxis for trypanosomosis control, the majority (69-95%) wrongly use diminazene aceturate thinking it is prophylactic, while only 5-30% used the prophylactic drug isometamidium chloride. Most of the respondents (95% in Korogwe, 60% in Pangani and 93.1% in Mvomero) administered the drugs on their own. Improper trypanocides administration was high as respondents in Korogwe (75%) and Mvomero (72%) administered the drugs intravenously with a view to achieving faster drug effect contrary to manufacturers' recommendations, while 40% of respondents from Pangani used both intravenous and intramuscular routes. Additionally, respondents did not observe the recommended withdrawal periods for the drugs., Conclusions: This study revealed a high level of trypanocides misuse which poses a high risk of trypanosome drug resistance development as well as risks to human health from drug residues in consumed animal products. This calls for improvements in veterinary service delivery in pastoral and agropastoral areas of Tanzania to counteract the misuse of chemotherapeutics.

African Animal Trypanosomosis (AAT) is a major disease of cattle in Togo and its control is essentially based on chemotherapy. However, because of excessive use of trypanocides during the past decades, chemo-resistance in the parasites has developed. In order to assess the current situation of AAT and resistance to trypanocidal drugs in Northern Togo, a study was conducted on cattle from December 2012 to August 2013 in the regions of Kara and Savanes. An initial cross-sectional survey was carried out in 40 villages using the Haematocrit Centrifugation Technique (HCT). Out of these, 5 villages with a trypanosome prevalence of >10% were selected for a block treatment study (BT) with diminazene diaceturate (DA: 3.5mg/kg for a 14-day follow-up) and isometamidium chloride (ISM: 0.5mg/kg for a 28-day follow-up). Positive blood samples collected during the parasitological surveys and an equivalent number of negatives were further analyzed by PCR-RFLP for trypanosome species confirmation and molecular diagnosis of resistance to DA in Trypanosoma congolense. The results from 1883 bovine blood samples confirmed a high overall trypanosome prevalence of 10.8% in Northern Togo. PCR-RFLP revealed that T. congolense is the dominant pathogenic trypanosome species (50.5%) followed by T. vivax (27.3%), and T. brucei (16.2%). The BT showed varying levels of treatment failures ranging from 0 to 30% and from 0 to 50% for DA and for ISM respectively, suggesting the existence of resistant trypanosome populations in the study area. Our results show that AAT still represents a major obstacle to the development of cattle husbandry in Northern Togo. In areas of high AAT risk, a community-based integrated strategy combining vector control, rational use of trypanocidal drugs and improving the general condition of the animals is recommended to decision makers., (Copyright © 2017 Elsevier B.V. Background: African animal trypanosomosis (AAT), transmitted by tsetse flies and tick-borne diseases are the main constraints to livestock production in sub-Saharan Africa. Vector control methods such as pour-on offer individual protection against ticks but not against tsetse so far, for which protection has always been communal, through a reduction of their density. The latter requires the treatment of a large part of the herd in a given landscape and is not instantaneous., Methodology/principal Findings: Two prospective surveys were conducted to evaluate the efficacy and persistence of a pour-on formulation composed of cypermetrhin, chlorpyrifos, piperonyl butoxid and citronella (Vectoclor, CEVA Santé Animal). In experimental conditions, tsetse flies were exposed to treated and control cattle. Flies knockdown and engorgement rates were determined and the product persistence was assessed as the time for these parameters to drop below 50% (T50). T50 was 37 days (95%CI: [33-41] days) and 46 days (95%CI: [39-56] days) for the knockdown and engorgement rates respectively. In field conditions, two cattle herds were monitored following a case-control experimental design, in the Adamaoua region of Cameroon. One herd was treated once with Vectoclor pour-on (treated group) and the second used as a control group (not treated). Ticks infestation rate, trypanosomosis prevalence and packed-cell volume were measured over the two months following treatment. The treatment was highly effective against ticks with a complete elimination three days after application in the treated group. Trypanosomosis prevalence was also significantly reduced during the study (by 4, P<0.001) and PCV of the treated group increased significantly in the same time (P<0.001), contrary to the control group., Conclusions/significance: The protection of this new pour-on against tsetse bites and trypanosomosis is demonstrated here for the first time. Moreover, this insecticide and repellent mixture offer a longer persistence of the efficacy against both tsetse and ticks than similar products currently on the market. It offers a great new opportunity for an integrated AAT control strategy including the treatment of residual cases with trypanocides. African animal trypanosomosis is a great obstacle to livestock production where tsetse flies play a major role. Metekel zone is among the tsetse-infested areas. Community-based tsetse fly and trypanosomosis control using targets was conducted from June 2011 to May 2012 in Metekel zone, Ethiopia, to decrease trypanosomosis and tsetse fly. Cloth screen targets were developed, impregnated with 0.1 % deltamethrin, and deployed alongside rivers by the research team together with the community animal health workers. Monthly parasitological and entomological data were collected, processed, and compared with similar data collected before control. Overall average tsetse fly (Glossina tachinoides) density decreased from 1.13 to 0.18 fly/trap/day after control. The density was decreased in all sites with no significant difference among the sites. However, higher decrements were observed in the dry and late dry seasons by more than 12 and 6 times, respectively. The reduction in overall apparent prevalence of trypanosomosis caused by Trypanosoma congolense, Trypanosoma brucei, and Trypanosoma vivax from 12.14 % before to 3.61 % after control coincides with the tsetse fly reduction. In all the study sites, significant reduction was observed before and after control. The highest decrement was observed in the late dry season when the apparent prevalence was reduced from 7.89 to 1.17 % before and after control, respectively. As this approach is simple, cost-effective, and appropriate for riverine tsetse species, we recommend to be scaled up to other similar places.

This study builds upon earlier work mapping the potential benefits from bovine trypanosomosis control and analysing the costs of different approaches. Updated costs were derived for five intervention techniques: trypanocides, targets, insecticide-treated cattle, aerial spraying and the release of sterile males. Two strategies were considered: continuous control and elimination. For mapping the costs, cattle densities, environmental constraints, and the presence of savannah or riverine tsetse species were taken into account. These were combined with maps of potential benefits to produce maps of benefit-cost ratios. The results illustrate a diverse picture, and they clearly indicate that no single technique or strategy is universally profitable. For control using trypanocide prophylaxis, returns are modest, even without accounting for the risk of drug resistance but, in areas of low cattle densities, this is the only approach that yields a positive return. Where cattle densities are sufficient to support it, the use of insecticide-treated cattle stands out as the most consistently profitable technique, widely achieving benefit-cost ratios above 5. In parts of the high-potential areas such as the mixed farming, high-oxen-use zones of western Ethiopia, the fertile crescent north of Lake Victoria and the dairy production areas in western and central Kenya, all tsetse control strategies achieve benefit-cost ratios from 2 to over 15, and for elimination strategies, ratios from 5 to over 20. By contrast, in some areas, notably where cattle densities are below 20per km(2), the costs of interventions against tsetse match or even outweigh the benefits, especially for control scenarios using aerial spraying or the deployment of targets where both savannah and riverine flies are present. If the burden of human African trypanosomosis were factored in, the benefit-cost ratios of some of the low-return areas would be considerably increased. Comparatively, elimination strategies give rise to higher benefit-cost ratios than do those for continuous control. However, the costs calculated for elimination assume problem-free, large scale operations, and they rest on the outputs of entomological models that are difficult to validate in the field. Experience indicates that the conditions underlying successful and sustained elimination campaigns are seldom met. By choosing the most appropriate thresholds for benefit-cost ratios, decision-makers and planners can use the maps to define strategies, assist in prioritising areas for intervention, and help choose among intervention techniques and approaches. Trypanosomosis is one of the most significant infectious threats to cattle in sub-Saharan Africa, and one form has also spread to Asia and South America. The disease is caused by a complex of trypanosome species, and the species and strain of parasite can have a profound influence upon the epidemiology of the host-parasite-vector relationships, the severity and course of infection, and, consequently, the implementation and development of control methods. This review will summarise our current knowledge of the relationship between trypanosome species/genotype and the phenotype of disease in cattle, and the implications that this has for ongoing efforts to develop diagnostics, drugs and vaccines for the control of cattle trypanosomosis.

In Senegal, a project has been undertaken to eradicate a population of tsetse flies (Glossina palpalis gambiensis) from a prime area for intensifying livestock production--the coastal region of Niayes. The project is intended to remove the constraint of trypanosomosis and allow the ecological intensification of cattle production. A cross-sectional analysis of ten case studies was the inductive phase of an assessment to gauge the impact of removing trypanosomosis on livestock production strategies. The methodology used was comprehensive analysis, with participatory epidemiology tools to understand farmers' rationales. The authors analysed the strategies of three main types of livestock producer (agro-pastoralists, mixed crop/livestock farmers and intensive dairy farmers). The strategies were in line with the farmers' goals and their ability to mobilise the socio-technical network. The risk management of trypanosomosis has been incorporated into livestock management practices through the use of trypanotolerant breeds, medical prophylaxis or placing livestock in low-risk areas. Removing the risk of disease would therefore have a major impact on decisions about the composition and strategic direction of herds. This change in the animal health environment would steer livestock production along different routes of intensification in a highly competitive environment. The indicators of innovation capacity revealed by this study will be used to quantitatively monitor various change scenarios, taking livestock producers' reasoning into account, in order to assess the socio-economic impact of eradicating the tsetse fly population in this area. The methodology presented in the study can be used to understand the impact of controlling other vector-borne infections on the innovation dynamics of livestock producers.

Endemic animal diseases such as tsetse-transmitted trypanosomosis are a constant drain on the financial resources of African livestock keepers and on the productivity of their livestock. Knowing where the potential benefits of removing animal trypanosomosis are distributed geographically would provide crucial evidence for prioritising and targeting cost-effective interventions as well as a powerful tool for advocacy. To this end, a study was conducted on six tsetse-infested countries in Eastern Africa: Ethiopia, Kenya, Somalia, South Sudan, Sudan and Uganda. First, a map of cattle production systems was generated, with particular attention to the presence of draught and dairy animals. Second, herd models for each production system were developed for two scenarios: with or without trypanosomosis. The herd models were based on publications and reports on cattle productivity (fertility, mortality, yields, sales), from which the income from, and growth of cattle populations were estimated over a twenty-year period. Third, a step-wise spatial expansion model was used to estimate how cattle populations might migrate to new areas when maximum stocking rates are exceeded. Last, differences in income between the two scenarios were mapped, thus providing a measure of the maximum benefits that could be obtained from intervening against tsetse and trypanosomosis. For this information to be readily mappable, benefits were calculated per bovine and converted to US$ per square kilometre. Results indicate that the potential benefits from dealing with trypanosomosis in Eastern Africa are both very high and geographically highly variable. The estimated total maximum benefit to livestock keepers for the whole of the study area amounts to nearly US$ 2.5 billion, discounted at 10% over twenty years--an average of approximately US$ 3300 per square kilometre of tsetse-infested area--but with great regional variation from less than US$ 500 per square kilometre to well over US$ 10,000. The greatest potential benefits accrue to Ethiopia, because of its very high livestock densities and the importance of animal traction, but also to parts of Kenya and Uganda. In general, the highest benefit levels occur on the fringes of the tsetse infestations. Background: Trypanosomosis, via causing anaemia, emaciation, production loss and death, is arguably the most important constraint to livestock development in Sub-Saharan countries, including Ethiopia and its impact in Baro-Akobo and Gojeb river basins (endemic areas for tsetse flies) is unknown. This study was carried out from November 2011 to April 2012 to assess farmers' perception on the presence, impact, management and the need of intervention programs of bovine trypanosomosis and tsetse fly in selected districts located in Baro-Akobo and Gojeb river basins, Southwestern Ethiopia. A standardized questionnaire survey was employed to collect the relevant information from the farmers., Results: The result of this study showed that 94.1% of the respondents considered bovine trypanosomosis as an economically important cattle disease which accounted for 64.6% of the total annual deaths in the year 2011/2012. Estimated mean annual financial loss via mortality due to trypanosomosis was reported to be 3501 Ethiopian Birr (US$200)/household. The reported trypanosomosis suggestive signs were consistent with published reports and farmers strongly associated the occurrence of the disease with biting flies (particularly, tsetse fly). Respondents also explained that the seasonality of the disease and its vectors, i.e. May and June are peak risk months of the year. Chemotherapy was reported the major method to combating the problem, mean frequency of treatment being 5.7 times per animal per year. Because of the economic burden of the disease, farmers expressed their strong interest and support for the establishment of intervention program in their area., Conclusion: The study revealed that livestock keepers are familiar with bovine trypanosomosis and its vectors as well as its impacts. Thus, trypanosomosis and tsetse control strategies should be integrated with the local communities' participation to minimize the impacts of the disease and its vectors in the area.

Background: In Chad, several species of tsetse flies (Genus: Glossina) transmit African animal trypanosomoses (AAT), which represents a major obstacle to cattle rearing, and sleeping sickness, which impacts public health. After the failure of past interventions to eradicate tsetse, the government of Chad is now looking for other approaches that integrate cost-effective intervention techniques, which can be applied by the stake holders to control tsetse-transmitted trypanosomoses in a sustainable manner. The present study thus attempted to assess the efficacy of restricted application of insecticides to cattle leg extremities using footbaths for controlling Glossina m. submorsitans, G. tachinoides and G. f. fuscipes in southern Chad., Methodology/principal Findings: Two sites were included, one close to the historical human African trypanosomiasis (HAT) focus of Moundou and the other to the active foci of Bodo and Moissala. At both sites, a treated and an untreated herd were compared. In the treatment sites, cattle were treated on a regular basis using a formulation of deltamethrin 0.005% (67 to 98 cattle were treated in one of the sites and 88 to 102 in the other one). For each herd, tsetse densities were monthly monitored using 7 biconical traps set along the river and beside the cattle pen from February to December 2009. The impact of footbath treatment on tsetse populations was strong (p < 10(-3)) with a reduction of 80% in total tsetse catches by the end of the 6-month footbath treatment., Conclusions/significance: The impact of footbath treatment as a vector control tool within an integrated strategy to manage AAT and HAT is discussed in the framework of the "One Health" concept. Like other techniques based on the treatment of cattle, this technology should be used under controlled conditions, in order to avoid the development of insecticide and acaricide resistance in tsetse and tick populations, respectively.

Trypanosomosis is an important disease affecting humans as well as animals. It remains a big constraint to livestock productions in tropical areas. The objective of this study was to assess the importance of trypanosomosis among cattle diseases in Burkina Faso, mainly in tsetse-challenged areas, and to capture information on how farmers apply available methods for controlling the disease. A survey has been carried out in 29 villages of Burkina Faso in three regions (north, southwest, and west regions). One hundred and thirty-four cattle breeders were interviewed individually with a questionnaire. The results indicate that among the 16 diseases mentioned by cattle breeders, trypanosomosis is the most important one in tsetse-challenged areas. Overall, 76.12 % of the breeders mentioned it as the most important disease, while 54.55 % of the farmers in the southwest region and 70.91 % of the farmers in the west region ranked it as a priority disease. Chemoprophylaxis/chemotherapy is widely used as a control method. Isometamidium chloride and diminazene aceturate were used by 53.49 and 46.52 % of the responders, respectively. Among farmers, 85.55 % ranked diminazene aceturate as the less efficient while 14.45 % ranked isometamidium chloride as the most efficient trypanocid. Trypanocidal drug quality and drug resistance were raised as a major concern by 30.77 and 50 % of the respondents, respectively. According to them, zebu cattle are more susceptible to trypanosomosis than taurine Baoule cattle and their crosses with zebu, emphasizing that crossing susceptible breeds with trypanotolerant ones, could be used as part of an integrated control strategy.

This study was conducted to assess the impact of tsetse and trypanosomosis control measures on cattle herd size and composition, herd dynamics, and milk yield in Wolaita and Gamogofa Zones, southern Ethiopia. The study showed that the average number of cattle herd size in tsetse challenged areas was significantly higher than those in tsetse-controlled areas. The number of non-pregnant dry cows, bulls and oxen were significantly higher in tsetse challenged areas than the other two study areas. The rate of cattle addition to and disposal from the herd were significantly higher in tsetse challenged areas. Cows in Southern Tsetse Eradication Project (STEP) and community tsetse controlled areas were able to give 26-27 %, 25-29 % and 17-21 % more daily milk yield at the beginning, middle and end of lactation, respectively, than those in tsetse-challenged areas. In addition, cows in STEP and community tsetse controlled areas had lactation length longer by 1.20 to 1.35 months; age at first calving was shorter by 5.30 to 5.10 months; and calving interval was shorter by 4.20 to 3.20 months than cows in tsetse-challenged area, respectively. Hence, tsetse and trypanosomosis control both by the community and project would play key role in the improvement of cattle productivity.

The effect of tsetse/trypanosomiasis control on cattle herd composition and growth and mortality of calves in tsetse controlled (by Southern Tsetse Eradication Project (STEP)) and uncontrolled blocks in southern Ethiopia was assessed. Structured questionnaire was used to interview 182 households to estimate cattle herd composition and calf mortality. Calves were bled to examine the presence of trypanosomes by the buffy coat technique. Forty NGU traps were deployed and fly catches determined. A case-control study was performed on 40 calves for 6 months to estimate calve growth parameters. Accordingly, the mean cattle herd size was lower in tsetse-controlled block than in the uncontrolled block, whereas the relative number of calves in a herd tend to be higher in the tsetse-controlled block (P = 0.06). While there was no report of cattle mortality in tsetse-controlled block, 16.48 % of the respondents have lost calves in tsetse-uncontrolled block in 1 year time. The prevalence of trypanosome positive calves was 2.95 % for uncontrolled block but no positive case in tsetse-controlled block. The apparent densities of flies/trap/day in tsetse-uncontrolled block were 30-fold higher than in tsetse-controlled block (P < 0.01). The case-control study revealed that the mean body weight gain of calves in tsetse-controlled block (40.23 ± 0.7 kg) was significantly higher than that of the uncontrolled block (34.74 ± 0.68 kg). The above findings strongly suggest that the intervention by the STEP project has significantly reduced tsetse population and trypanosomiasis consequently contributing to improved calf growth and survival.

Tsetse-transmitted livestock trypanosomosis affects livestock in large parts of sub-Saharan Africa. In southern Africa two epidemiological situations can be distinguished. The disease can have an endemic nature with high morbidity and low mortality in the livestock population. Endemic livestock trypanosomosis is found mainly in areas where cattle constitute the main host of tsetse and reservoirs of trypanosomes. Epidemic trypanosomosis, with high morbidity and high mortality is found in areas where wildlife persist as main reservoir and where livestock come into contact with tsetse flies transmitting trypanosomes from the sylvatic reservoir. Based on the differences in impact of the disease on livestock health in these two epidemiological settings, the appropriateness of the available trypanosomosis control tools differs. In trypanosomosis endemic areas, trypanocidal drug use could be the most suitable approach. Possible problems associated with the development of resistance in trypanosomes to the drugs need to be investigated further. Studies were made of the efficacy of using smoke and housing to protect cattle from tsetse (Diptera: Glossinidae) in Zimbabwe. The efficacy of smoke was assessed by its effect on catches in Epsilon traps baited with a blend of acetone, 1-octen-3-ol, 4-methylphenol and 3-n-propylphenol. The efficacies of different types of kraal (enclosure) were gauged according to the catches of electrocuting targets (E-targets), baited with natural ox odour, placed within various designs of kraal. Smoke from burning wood (Colophospermum mopane) or dried cow dung reduced the catch of traps by approximately 50-90%. Kraals with a continuous wooden or netting wall, 1.5 m high, reduced catches of E-targets by approximately 75%. Arrangements of electric nets were used to assess the numbers of tsetse attacking live cattle within kraals and/or near sources of smoke. The results confirmed findings with traps and E-targets: kraals reduced the numbers of tsetse that fed by approximately 80% and smoke reduced the numbers attracted by approximately 70%; the use of both reduced overall attack rates by approximately 90%. The inclusion of 4-methylguaiacol, a known repellent for tsetse and a natural component of wood smoke, halved the catches of traps and E-targets and the numbers of tsetse attacking cattle. The practical benefits and difficulties of using repellents and/or housing to manage trypanosomiases are discussed.

We conducted a field trial among Maasai cattle-keepers in Nkuruman and Nkineji areas of Kenya to evaluate the effectiveness of a synthetic tsetse-repellent technology developed for the control of trypanosomosis in cattle. The technology was a repellent (2-methoxy 4-methylphenol) emitted from dispensers attached to collars worn by cattle. Treatment was allocated at the herd level to ensure adequate protection of all the animals in a herd, with measurements of effectiveness conducted at the individual-animal level. The trial began in April 2005 and ran for 16 months including a baseline phase of 4 months. We recruited 12 herds in each area using a restricted random-sampling technique and distributed them equally into intervention (repellent) and control groups. Sample size was determined using a formal power calculation. Effectiveness or minimal worthwhile difference was defined as a 50% reduction in the incidence of trypanosome infection in the treated versus control group (effectiveness below which the technology was considered by experts as not viable compared to existing control techniques). All the animals in the recruited herds were screened monthly (buffy-coat technique) for trypanosome infections. The analysis followed the principle of intention-to-treat by which subjects are analysed according to their initial treatment assignment, regardless of the mechanical performance of the device. Crude and adjusted effects of the technology were 23% (p<0.001) and 18% (p=0.08) reduction in the infection incidence in the treatment compared to the control groups, respectively. The impact of the technology estimated in this study did not achieve the threshold of 50% reduction in the trypanosome infection incidence set a priori to indicate effectiveness (p<0.001). We therefore concluded that the prototype repellent technology package was not sufficiently effective in reducing trypanosome infection incidence under natural tsetse challenge to merit commercial development., (Copyright © 2010 Elsevier B.V. The present study was conducted to determine the prevalence and incidence of trypanosomosis and to investigate some factors influencing them in an agro-pastoral area of southwestern Burkina Faso. A total of 363 crossbred cattle (Baoule-zebu peul), which were bred under natural trypanosomosis challenge, were monitored monthly for parasitaemia, packed cell volume (PCV) and serological analyses over 2years. The parasitological prevalence estimated at the beginning of the survey using the buffy coat technique (BCT) was 7.54%. As much as 66.7% of all trypanosome infections were due to Trypanosoma vivax, 23.8% due to Trypanosoma congolense and 9.5% due to T. vivax/T. congolense mixed infections. The monthly serological incidence varied from 0.29% to 19.29%. The season was the most important factor influencing the serological prevalence and incidence and the animal PCV. The dry hot season is associated with increasing seroprevalences and incidences and consequently a decreasing average of PCV. International research and development efforts in Africa have brought ecological and social change, but analyzing the consequences of this change and developing policy to manage it for sustainable development has been difficult. This has been largely due to a lack of conceptual and analytical models to access the interacting dynamics of the different components of ecosocial systems. Here, we examine the ecological and social changes resulting from an ongoing suppression of trypanosomiasis disease in cattle in an agropastoral community in southwest Ethiopia to illustrate how such problems may be addressed. The analysis combines physiologically based demographic models of pasture, cattle, and pastoralists and a bioeconomic model that includes the demographic models as dynamic constraints in the economic objective function that maximizes the utility of individual consumption under different level of disease risk in cattle. Field data and model analysis show that suppression of trypanosomiasis leads to increased cattle and human populations and to increased agricultural development. However, in the absence of sound management, these changes will lead to a decline in pasture quality and increase the risk from tick-borne diseases in cattle and malaria in humans that would threaten system sustainability and resilience. The analysis of these conflicting outcomes of trypanosomiasis suppression is used to illustrate the need for and utility of conceptual bioeconomic models to serve as a basis for developing policy for sustainable agropastoral resource management in sub-Saharan Africa.

African animal trypanosomoses are the main parasitological constraints to livestock production in many sub-Saharan African countries infested with tsetse flies. A prospective survey was implemented in Dafinso (Burkina Faso) to assess the effect of deltamethrin 0.005% (Vectocid(ND), CEVA Santé Animale) impregnation of cattle on trypanosomes transmission in cattle. Two herds were involved in the survey. They were watered at two different waterpoints located on the same river harboring a Guinean riparian forest infested with two different species of tsetse flies (Diptera: Glossinidae), Glossina palpalis gambiensis Vanderplank and G. tachinoides Westwood. Animals belonging to one of the herds were impregnated with deltamethrin applied with a footbath whereas the other herd was used as control. The overall incidence of cattle trypanosomoses was reduced (p=0.01) from 0.76 (control group) to 0.11 (footbath-treated group). A positive effect of the footbath treatment on packed-cell volume was observed (p<0.001). The conditions requested to use a footbath to prevent cattle trypanosomoses are discussed.

The best technical package for the future comprises trypanocidal drugs for temporary relief and the use of insecticide-treated cattle, artificial baits and aerial spraying to attack the vector, to so give more lasting security. Whether this can speed the previously slow progress will depend on overcoming past hindrances to tsetse control: sporadic support, disputes over its desirability, difficulties of sustaining international operations, and poor planning in some instances. The Pan-African Tsetse and Trypanosomiasis Campaign intends to speed the progress but will fail unless it improves its image by breaking its association with the sterile insect technique and quickly executing some cheap and effective operations in large areas. Even then, there could be severe brakes due to Africa's political and financial instability. Overall, the pace of control is likely to increase, but perhaps only a little.

Research in control of tick-borne diseases and trypanosomosis, and their vectors, namely, ticks and tsetse flies respectively, has been on going for decades. However, very little attention has been paid to the socio-economic factors that are likely to influence the outcome of the interventions in the control of these diseases. Thus, this study was designed to investigate these factors, mainly the intra-household factors influencing decision-making in the control of Vector-borne diseases in the pastoralist areas of Uganda. These factors included: indigenous technical knowledge, household economic factors, and gender. Both qualitative and quantitative methods were used in the collection and analysis of data. The tools used for data collection included among others, participatory learning and action (PLA), and Case studies. The findings included the following: In pastoralist households, a big proportion of the household budget was allocated to vector-borne diseases control. In the male-headed households, men dominated decision-making on vector-borne diseases control, although the goals and priorities of men and women in these households were not the same. Also, vector-borne disease control was predominantly by use of modern veterinary drugs, and pastoralists treated sick cattle by themselves even in situations where there were veterinary personnel.

We tested the efficacy of two formulations of isometamidium in a tsetse-infested farm in North Cameroon from 20 August 2000 to 5 January 2001. A total of 90 adult cattle were used in three groups of 30 each corresponding to two treated and one untreated control. Drug efficacies were evaluated in terms of reduction of parasite incidence in the host's blood, maintenance of packed-cell volume (PCV) and weight gains. Both drugs reduced the incidence of parasites even though re-infections 2 weeks after treatment were common. PCV values were similar in both treated groups but higher than in the untreated control. Body weight changes followed a similar trend with the control losing weight from a mean of 427+/-119kg at the beginning to 398+/-93kg in 4 months. Weights increased from 375+/-76 and 396+/-110 to 396+/-69 and 418+/-112kg in the Veridium and Trypamidium groups, respectively. Efficacy was similar between the two formulations of isometamidium in the prophylaxis of bovine trypanosomosis. However, the presence of parasites in some animals barely 2 weeks after treatment suggested that either infections were not cleared or residual drug effects were not sufficient to prevent re-infections.

In large parts of Africa the control of livestock trypanosomiasis relies on the use of trypanocidal drugs. Resistance against the available compounds is developing rapidly in the trypanosome population. The effect of the development of drug resistance on the fitness of the trypanosome is not well known. To determine the effect of the development of resistance to isometamidium chloride on the trypanosome's transmissibility, transmission experiments were conducted. Use was made of three isogenic clones of Trypanosoma congolense with different susceptibility to the drug. The infection rate in Glossina morsitans morsitans differed significantly between clones and was significantly higher in tsetse flies infected with the T. congolense clone with the highest level of drug resistance.

The incidence of trypanosome infection was monitored in dairy cattle during a 6-month trial in Busia and Teso districts, western Kenya, to assess the efficacy of insecticide-treated netting for protection against tsetse flies. Frequently, the fragile netting did not last longer than 2 months because of destruction by strong wind or animal movements. Also, many farmers let their cattle graze freely outside the units during the day, despite technical advice, resulting in exposure of the free-ranging animals to habitats suitable for tsetse and thereby an increased risk of trypanosome infections. The trial groups thus comprised 34 animals from 11 dairy units that were continuously protected, and 153 animals from 46 dairy units that were partially protected. The control group consisted of 162 animals in 42 unprotected units. The phase-contrast buffy-coat technique was used for parasitological monitoring. The mean hazard rate for trypanosomes was significantly lower in protected cows, with a value of 0.007 as opposed to 0.02 for the control animals. Mean packed cell volumes (PCV) were significantly higher in protected cattle (29.7%) than in unprotected ones (27.6%). Farmers with protected animals also reported fewer nuisance flies and mosquitoes in their compounds.

Isometamidium chloride has remained a very important prophylactic and therapeutic drug against trypanosomosis in cattle since its introduction into the market in the 1950s with, unfortunately, a concomitant development of resistance in trypanosomosis endemic areas. Amplified Fragment Length Polymorphism (AFLP) was used to compare two isogenic clones of Trypanosoma congolense. The parent clone, sensitive to isometamidium, has a CD50 (the curative dose that gives complete cure in 50% of the animals) in the mouse of 0.018 mg/kg and its derivative exposed to increasing doses of isometamidium, has a CD50 that is 94-fold higher. Sixty-four combinations of eight Eco RI and eight Mse I primers were used in comparative AFLP analysis to detect subtle genetic differences between the two clones. Thirty-five polymorphic fragments of DNA that were observed only in the resistant clone were purified and then sequenced. The nucleotide sequences were used in searching the GeneDB T. congolense database to find surrounding sequences upstream of an open reading frame and downstream to a stop codon. The sequences of the open reading frames were subsequently compared to the sequences in the genomic databases. A predicted gene coding for an 854 amino acids protein was thus identified. The protein contains a putative ATP binding site, Walker B and LSGG motifs and eight predicted trans-membrane domains. The gene in the resistant strain of T. congolense has a triplet insertion coding for an extra lysine. Using polymerase chain reaction-restriction fragment length polymorphism, the insertion was sought in the genomes of 35 T. congolense strains isolated from different geographic origins and whose response to isometamidium chloride had been determined through single dose mouse tests. The presence of the insertion, specifying an extra codon was found to always be present in the genomes of T. congolense clones that were resistant to isometamidium chloride.

A pilot survey using a structured questionnaire was conducted in Tororo and Busia districts of Uganda on the knowledge and attitudes of cattle owners regarding tsetse fly and trypanosomosis control, in order to understand factors that hindered their full participation. A total of 81 cattle owners was randomly selected and interviewed, of which 92.5% were aware of tsetse flies and trypanosomosis and 87.6% recognised animal trypanosomosis as a problem in the area. Most cattle owners were aware of tsetse fly trapping (76.5%), isometamidium chloride use (55.5%), diminazene aceturate use (48%) and pour-on applications (18.5%). However, knowledge did not coincide with the application of control measures. Despite the widespread awareness, tsetse fly trapping and pour-on applications were used by only a small percentage of cattle owners (7.5% applied tsetse fly trapping while 76.5% were aware of it; 1.2% applied pour-on insecticides while 18.5% were aware of them). Differences between awareness and application were highly significant for tsetse fly trapping (chi2 = 67.8, d.f. = 1, P < 0.001) and pour-on applications (chi2 = 10.8, d.f. = 1, P < 0.05), but not for isometamidium chloride use (chi2 = 0.08, d.f. = 1, P = 0.77) and diminazene aceturate use (chi2 = 0.00, d.f. = 1, P = 1.00). Most cattle owners (97.5%) were willing to participate in future control programmes, but preferred participating on a group basis (85.2%) rather than individually (14.8%). The 4 most favoured control options in order of importance were: fly traps supplied by the government and maintained by cattle owners; contribution of labour by cattle owners for trap deployment; self-financing of trypanocidal drugs and self-financing of pour-on insecticide. The control options that should be selected in order to elicit full participation by cattle owners are discussed.

The objective of this study was to apply transition models to distinguish between factors associated with both incident and persistent trypanosome infections. Data collected from 1561 cattle were analyzed from a long-term study involving 8 herds in which both trypanosome infections (a total of 56,931 cattle sampling-months) and tsetse (Glossina spp.) challenge were monitored monthly from March 1986 to March 1998. Both pour-on and insecticide-target tsetse control programs and mass treatment with diminazene aceturate before tsetse control were associated with significant decreases in both incidence and persistence of trypanosome infection relative to noncontrol periods, as were seasonal and sex effects. The magnitudes of the effects were, however, often different for new and persistent infections. For persistence of infection, there were 2 trends. In general, the duration of infection increased during the study, despite the regular treatment with diminazene aceturate. The transition model had 2 major benefits. The first was to identify an increasing duration of infections with time, taking into account other factors associated with increasing infection risk. The second was to highlight different patterns in the effects of certain factors on new and persistent trypanosome infections.

The protective efficacy of isometamidium chloride (ISMM) and diminazene aceturate (DIM) against Trypanosoma brucei, Trypanosoma congolense and Trypanosoma vivax infections in cattle under a suppressed tsetse population was assessed in southeast Uganda. A total of 66 and 57 trypanosome-infected cattle were treated with ISMM and DIM, respectively together with 177 trypanosome-free animals not treated were followed for 12 months, checked every 4 weeks. There was no statistical difference in the mean time to infection with any trypanosome species in animals treated with ISMM or DIM. However, the mean time to trypanosome infection was significantly longer for treated animals than controls. The mean time to infection with each of the three trypanosome species differed significantly, with the average time to T. vivax infection the lowest, followed by T. congolense and then T. brucei. The protective efficacy of DIM was as good as that of ISMM; implying curative treatments against trypanosomosis are sufficient for combination with tsetse control. Isometamidium chloride or DIM had the highest impact on T. brucei and T. congolense infections in cattle.

A trial to evaluate the efficacy of a 1% cyfluthrin pour-on formulation (Cylence, Bayer) in reducing the incidence of bovine trypanosomosis was conducted in an area of ca. 2000 km2 of the Eastern Province of Zambia. The trial area was cultivated and carried a cattle population of approximately 11 animals/km2. Cattle were the main host of tsetse. Following the free of charge treatment of the adult cattle at intervals of 7 weeks and at a dosage of 15 ml/100 kg body weight, there was an increase in the average packed cell volume in the herd although the decline in the incidence of trypanosomal infections was more prolonged. The monthly incidence of trypanosomal infections started to decline substantially 8 months after the treatments were initiated. No trypanosomal infections were detected from 10 months after the start of the trial.

The distributions of insecticide-treated cattle from sites in Tanzania and Zimbabwe were assessed from interviews with livestock owners, analysis of secondary livestock data and mapping technologies. The time-course of tsetse control operations at these sites were then simulated using a mathematical model that assumed diffusive movement and logistic growth in fly populations. A simulation of a tsetse control operation in Mudzi district, north-east Zimbabwe, was in accord with observations that the use of insecticide-treated cattle was unable to prevent substantial re-invasion of tsetse from Mozambique, consequent on the patchy distribution of cattle. The simulation was also consistent with the observed efficacy of a 10-km wide barrier of insecticide-treated targets deployed evenly at 4 km/(-2). Simulation of a control operation on Mkwaja Ranch in Tanzania was in accord with the observation that the use of insecticide-treated cattle reduced the tsetse population on the ranch by c. 90%. Insecticide-treated cattle were used to better effect in the Kagera Region of Tanzania. Simulation of this operation predicts that the deployment of 35,000 treated cattle in the area would result in > 99% control of the tsetse population, consistent with the observed decline, by 1-2 orders of magnitude, in cases of trypanosomiasis in the region. The greater success of the Kagera operation was due to the size and shape of the treated area and, particularly, to the restriction of re-invasion to 20% of the perimeter, compared with > 80% on Mkwaja. Simulation was used to assess how tsetse control could have been improved at Mkwaja. The results suggest that splitting herds into smaller, more numerous, units could have achieved some improvement but, in general, the disease problem would not have been solved by the use of insecticide-treated cattle alone. Only by deploying odour-baited targets in ungrazed areas, or in a 1-3-km barrier around the ranch, could substantially better control (99-99.9%) have been achieved. Sensitivity analyses of the Mkwaja simulation showed that the general conclusions were robust to assumptions regarding cattle distribution and the rates of fly movement and growth. Properly managed and appropriately applied insecticide-treated baits are powerful weapons for tsetse control but should not be used without regard to potential levels of re-invasion, consequent largely on considerations of the size and shape of the treatment area and the density and distribution of the baits.

A study using a structured questionnaire was conducted in Busia District, Western Kenya and Kwale District, Coastal Kenya to obtain qualitative and quantitative information from 256 cattle owners about their production systems, their perceptions of the diseases encountered in their cattle, the drugs used, and other measures adopted to control trypanosomiasis in cattle. The predominant production system was mixed crop-livestock with farmers owning 2-11 local cattle on holdings between 2 and 5 ha. Approximately 15% of disease episodes in cattle were perceived to be trypanosomiasis, although the farmers' ability to make diagnoses was limited in that over half of the diagnoses were inconsistent with the clinical signs described. Drugs were generally obtained from agro-veterinary shops, and the farmers themselves administered slightly more than half of these. One third of drug treatments given to sick cattle were trypanocides, but over half of these trypanocidal treatments were given to cattle believed to have diseases other than trypanosomiasis.

A survey to monitor the use of trypanocidal drugs by cattle breeders was conducted in Zambia. Use was made of a questionnaire and of the isometamidium-ELISA technique. One hundred and twenty-two farmers and 50 veterinary assistants were interviewed. The isometamidium-ELISA was used to monitor the isometamidium serum concentration in 72 cattle, 1 week after unsupervised treatment by 56 farmers and 16 veterinary assistants. Although there was no clear indication of underestimation of the weight of the animals and although farmers had adequate knowledge of the correct usage of isometamidium, the results suggest frequent underdosing when considering isometamidium serum concentrations 1 week after treatment. In 76% of the cases, the expected protection period was equal or shorter than 28 days and equal or shorter than 33 days in 90% of the treated cattle., (Copyright 2002 Elsevier Science B.V.)

Cattle from 50 farms in Mukono County, Uganda, were monitored for trypanosomes every second month over an 18-month period (1995-1996) by mini-anion exchange chromatography and haematocrit centrifugation techniques. Eighteen trypanosome isolates collected from cattle during this period were characterised in cattle, goats and mice for their sensitivity to homidium, isometamidium and diminazene; 10 of the isolates were selected randomly, 8 were from animals that had the highest serum isometamidium concentrations at the time the isolates were collected. All the isolates contained only Trypanosoma brucei and/or T. vivax. In nai;ve Boran (Bos indicus) cattle the isolates exhibited low pathogenicity and were sensitive to diminazene aceturate at 3.5 mg/kg body weight (bw) and isometamidium chloride at 0.5 mg/kg bw. In goats, 5 of 8 isolates were highly pathogenic, producing clinical signs indicative of central nervous system involvement within 60 days of infection; all such isolates contained T. brucei. However, all 8 populations were sensitive in goats to diminazene aceturate at 3.5 mg/kg bw. In contrast, 4 populations were refractory to treatment with isometamidium chloride at 0.5 mg/kg bw in at least 1 out of 3 goats each. Furthermore, 5 populations were refractory to treatment with homidium chloride at 1.0 mg/kg bw in a minimum of 2 out of 3 goats each. In mice, the 50% curative dose values for 11 Mukono isolates that contained T. brucei ranged from 0.30 to 1.89 mg/kg bw for diminazene aceturate, from 0.02 to 0.17 mg/kg bw for isometamidium chloride and from 0.90 to 4.57 mg/kg bw for homidium chloride. Thus, by comparison to reference drug-sensitive populations, all the stabilates were highly sensitive to diminazene and isometamidium, while some expressed low levels of resistance to homidium.

This study was conducted in Masaba and Masafu Sub-counties, Busia District, Uganda to assess the effect on the tsetse fly population of first treating all cattle with 1% w/v deltamethrin pour-on for a few months, followed by treating 10% of the cattle population. Treatment of all cattle for 6 months resulted in a significant reduction in the density of tsetse flies from 6.3 to 0.1 flies/trap/day (FTD), a 98.4% reduction. During the same period, the point prevalence of bovine trypanosomosis dropped from 37.7% to 2.9% (a 92.3% reduction). Treatment was resumed six months later, but this time only 10% of the cattle population received the pour-on treatment at three week intervals for a period of one year. This treatment maintained the tsetse fly density between 0 and 0.5 FTD and the prevalence of bovine trypanosomosis generally remained below 10%. In conclusion, under the local prevailing conditions, treatment of all communally grazed cattle with deltamethrin pour-on effectively suppressed the Glossina fuscipes fuscipes population. However, subsequent treatment of 10% of the cattle probably failed to control the tsetse fly population at a level sufficient to reduce trypanosomosis to acceptable levels.

Participatory research on bovine trypanosomiasis was conducted with Orma pastoralists in Tana River District, Kenya. The use of participatory methods to understand local perceptions of disease signs, disease causes, disease incidence by cattle age group, seasonal patterns of disease and preferences for indigenous and modern control methods are described. Results indicated that local characterization of diseases called gandi and buku by Orma pastoralists was similar to modern veterinary knowledge on chronic trypanosomiasis and haemorrhagic trypanosomiasis (due to Trypanosoma vivax), respectively. The mean incidence of gandi varied from 10.2% in calves to 28.6% in adult cattle. The mean incidence of buku varied from 3.1% in calves to 9.6% in adults. Pearson correlation coefficients for disease incidence by age group were 0.498 (P < 0.01) and 0.396 (P < 0.05) for gandi and buku, respectively. Informants observed cases of trypanosomiasis in 24.1% of cattle (all age groups); these cases accounted for 41.8% of all sick cattle during the preceding 12-month period. Eight indigenous and three modern trypanosomiasis control methods were identified. Results indicated that an integrated approach to trypanosomiasis control based on private, individual action was well established in the assessment area. When presented with four different trypanosomiasis control methods, community representatives selected 'better use of trypanocides' as the most preferred intervention and 'community-based tsetse control' as the least preferred intervention. This finding prompted researchers to modify the original project activities. Constraints facing the sustainability of community-based tsetse control are discussed.
