Abstract The PRISM project, funded by the EU’s Innovative Medicines Initiative, has identified a transdiagnostic, pathophysiological relationship between the integrity of the default mode network (DMN) and social dysfunction. To explore the causal link between DMN integrity and social behaviour, we employed a preclinical back-translation approach, using focal demyelination of the forceps minor to disrupt DMN connectivity in mice. By applying advanced techniques such as functional ultrasound imaging and automated analysis of social behaviour, we demonstrated that reduced DMN connectivity leads to impaired social interactions and increased anxiety in mice. Notably, these effects were reversible, indicating that the forceps minor, a critical fibre tract connecting key DMN regions in the prefrontal cortex, plays a crucial role in social function. This study provides direct evidence for a causal relationship between DMN integrity and social dysfunction, with potential implications for developing targeted treatments in precision psychiatry.

Abstract The Claustrum/dorsal endopiriform cortex complex (CLA) is an enigmatic brain region with extensive glutamatergic projections to multiple cortical areas. The transcription factor Nurr1 is highly expressed in the CLA, but its role in this region is not understood. By using conditional gene-targeted mice, we show that Nurr1 is a crucial regulator of CLA neuron identity. Although CLA neurons remain intact in the absence of Nurr1, the distinctive gene expression pattern in the CLA is abolished. CLA has been hypothesized to control hallucinations, but little is known of how the CLA responds to hallucinogens. After the deletion of Nurr1 in the CLA, both hallucinogen receptor expression and signaling are lost. Furthermore, functional ultrasound and Neuropixel electrophysiological recordings revealed that the hallucinogenic-receptor agonists’ effects on functional connectivity between prefrontal and sensorimotor cortices are altered in Nurr1-ablated mice. Our findings suggest that Nurr1-targeted strategies provide additional avenues for functional studies of the CLA.

Background. In certain cases of lateral edentation, some treatment options are limited due to the patient’s desire or maybe due to the fact that biology could not permit otherwise. Hence, in these scenarios the options to use the implant therapy are limited. Case report. 50-year-old female patient was referred to clinic for a full mandibula rehabilitation. In this case the patient was diagnosed with Kennedy Class I. In this situation, the treatment option was fixed metal-ceramic crowns with precision attachments (keyway attachments) for a removable prothesis. Aim. To emphasize the fact that, from a socio-economic point of view, the patient tends to choose the more accessible treatment option, but also the time component has a major impact on the choice they make, the development time of a treatment plan being very important to the patient. Material and methods. 10 cases of partial edentation were analyzed for the possibility of realizing two treatment plans, namely the version with implants or with removable prosthesis. The clinical implications of various aspects of this area were monitored and displayed as results. Results. Patients were much more receptive to the removable prosthesis with precision attachments treatment compared to the insertion of implants. The former has a similar result to the latter, but in a much shorter time span and with lower costs. Conclusions. From the analyzed it was confirmed that edentulous patients can be successfully treated with fixed prosthodontics associated with a removable partial prosthesis with precision attachments with the same benefits that the implant works might have brought them, the patient being satisfied with the quality of life gained as a result of benefitting from removable prosthesis in a relatively convenient period of time.

Background: Abfraction lesions are manifested as damage to hard tissues in the cervical area of dental crowns. The study aimed to assess the direct restoration of abfraction lesions according to the modified United States Public Health Service (USPHS) criteria for 24 months. The restorations were accomplished with Fuji Bulk–GC, Omnichroma Flow-Tokuyama, and Beautifil® II–Shofu dental materials, and the therapy was or was not associated with wearing thermoformed mouthguards. Methods: From the 53 selected and analyzed patients (n = 53), 28 patients (with restorations of abfraction lesions) belonged to the 1st group and 25 patients (with 105 restorations, who also wore mouthguards) belonged to the 2nd group. Blind determination assessments were effectuated at baseline and after 2, 6, 12, 18, and 24 months. Results showed that, regardless of the rating score, there are no significant statistical differences in the evaluation criteria between the two groups of patients Conclusions: For each material, the scores of USPHS criterion presented good clinical performances after 24 months, with no significant statistical differences between the fillings and the applied therapy in the two groups of patients.

The application of the Coandă effect to the directional control of a jet or thrust is presented. Deviation of the thrust force by direct flow can be achieved by using the Coandă effect to change the angle of the primary jet engine exhaust nozzle. Major interest in the study of this phenomenon is caused by the possibility of using this effect for aircrafts with short take-off and landing, for thrust vectoring. The numerical investigations are performed using a RANS solver with an adequate turbulence model, showing a change of the jet direction. Thus, the conditions and the limits within which one can benefit from the advantages of Coandă-type flows are determined.
