Summary: We recently demonstrated that vaccines comprising antigenic peptides conjugated to a glycolipid agonist, termed glycolipid-peptide (GLP) vaccines, efficiently generate substantial numbers of long-lived CD8+ liver-resident memory T (Trm) cells that are crucial for protection against malaria liver-stage infection. To understand the underlying mechanism, we examined the prerequisites for priming, differentiation, and secondary boosting of liver Trm cells using these GLP vaccines. Our study revealed that generation of long-lived liver Trm cells relies on CD8+ T cell priming by type 1 conventional dendritic (cDC1) cells, followed by post-priming exposure to a combination of vaccine-derived inflammatory and antigenic signals. Boosting of liver Trm cells is feasible using the same GLP vaccine, but a substantial delay is required for optimal responses due to natural killer T (NKT) cell anergy. Overall, our study unveils key requirements for the development of long-lived liver Trm cells, offering valuable insights for future vaccine design.

Summary: The cGAS-STING pathway responds to cytosolic DNA to elicit host immunity to infection. The activation of stimulator of interferon genes (STING) can trigger a number of critical cellular responses including inflammation, noncanonical autophagy, lipid metabolism, senescence, and cell death. STING-mediated immunity through the production of type I interferons (IFNs) and nuclear factor kappa B (NF-κB)-driven proinflammatory cytokines is primarily driven via the effector protein TBK1. We have previously found that IκBα kinase epsilon (IKKε), a homolog of TBK1, can also facilitate STING-NF-κB responses. Therefore, a thorough understanding of how IKKε participates in STING signaling is essential. Here, we used a combination of genetic and biochemical approaches to provide mechanistic details into how IKKε confers non-IFN (e.g., NF-κB and MAPK) STING responses in macrophages, including in the absence of TBK1. We demonstrate a conserved mechanism of STING binding between TBK1 and IKKε. These findings strengthen our understanding of cGAS-STING signaling and the preservation of host immunity in cases of TBK1-deficiency.

6. Conceptualising social media addiction: a longitudinal network analysis of social media addiction symptoms and their relationships with psychological distress in a community sample of adults

Abstract Background Problematic social media use has been identified as negatively impacting psychological and everyday functioning and has been identified as a possible behavioural addiction (social media addiction; SMA). Whether SMA can be classified as a distinct behavioural addiction has been debated within the literature, with some regarding SMA as a premature pathologisation of ordinary social media use behaviour and suggesting there is little evidence for its use as a category of clinical concern. This study aimed to understand the relationship between proposed symptoms of SMA and psychological distress and examine these over time in a longitudinal network analysis, in order better understand whether SMA warrants classification as a unique pathology unique from general distress. Method N = 462 adults (M age = 30.8, SD age = 9.23, 69.3% males, 29% females, 1.9% other sex or gender) completed measures of social media addiction (Bergen Social Media Addiction Scale), and psychological distress (DASS-21) at two time points, twelve months apart. Data were analysed using network analysis (NA) to explore SMA symptoms and psychological distress. Specifically, NA allows to assess the ‘influence’ and pathways of influence of each symptom in the network both cross-sectionally at each time point, as well as over time. Results SMA symptoms were found to be stable cross-sectionally over time, and were associated with, yet distinct, from, depression, anxiety and stress. The most central symptoms within the network were tolerance and mood-modification in terms of expected influence and closeness respectively. Depression symptoms appeared to have less of a formative effect on SMA symptoms than anxiety and stress. Conclusions Our findings support the conceptualisation of SMA as a distinct construct occurring based on an underpinning network cluster of behaviours and a distinct association between SMA symptoms and distress. Further replications of these findings, however, are needed to strengthen the evidence for SMA as a unique behavioural addiction.

Dendritic cells (DCs) are crucial for the efficacy of cancer vaccines, but current vaccines do not harness the key cDC1 subtype required for effective CD8+ T-cell-mediated tumor immune responses. Vaccine immunogenicity could be enhanced by specific delivery of immunogenic tumor antigens to CD141+ DCs, the human cDC1 equivalent. CD141+ DCs exclusively express the C-type-lectin-like receptor CLEC9A, which is important for the regulation of CD8+ T cell responses. This study developed a new vaccine that harnesses a human anti-CLEC9A antibody to specifically deliver the immunogenic tumor antigen, NY-ESO-1 (New York esophageal squamous cell carcinoma 1), to human CD141+ DCs. The ability of the CLEC9A-NY-ESO-1 antibody to activate NY-ESO-1-specific naïve and memory CD8+ T cells was examined and compared with a vaccine comprised of a human DEC-205-NY-ESO-1 antibody that targets all human DCs. Human anti-CLEC9A, anti-DEC-205 and isotype control IgG4 antibodies were genetically fused to NY-ESO-1 polypeptide. Cross-presentation to NY-ESO-1-epitope-specific CD8+ T cells and reactivity of T cell responses in patients with melanoma were assessed by interferon γ (IFNγ) production following incubation of CD141+ DCs and patient peripheral blood mononuclear cells with targeting antibodies. Humanized mice containing human DC subsets and a repertoire of naïve NY-ESO-1-specific CD8+ T cells were used to investigate naïve T cell priming. T cell effector function was measured by expression of IFNγ, MIP-1β, tumor necrosis factor and CD107a and by lysis of target tumor cells. CLEC9A-NY-ESO-1 antibodies (Abs) were effective at mediating delivery and cross-presentation of multiple NY-ESO-1 epitopes by CD141+ DCs for activation of NY-ESO-1-specific CD8+ T cells. When benchmarked to NY-ESO-1 conjugated to an untargeted control antibody or to anti-human DEC-205, CLEC9A-NY-ESO-1 was superior at ex vivo reactivation of NY-ESO-1-specific T cell responses in patients with melanoma. Moreover, CLEC9A-NY-ESO-1 induced priming of naïve NY-ESO-1-specific CD8+ T cells with polyclonal effector function and potent tumor killing capacity in vitro. These data advocate human CLEC9A-NY-ESO-1 Ab as an attractive strategy for specific targeting of CD141+ DCs to enhance tumor immunogenicity in NY-ESO-1-expressing malignancies.

What if researchers interested in ‘the past’ used their noses? This open access book makes the case for a more imaginatively interdisciplinary approach to sensory heritage and history, arguing that we can and should engage our noses as a research tool for articulating the past. Assessing how both we and our ancestors approach, understand and conceptualise smell, Tullett shows how archives can be ‘re-odorized’ to uncover narratives that are only implicit in or obscured by the historical record. From perfume libraries to organic compounds emitted by historical objects, this book acts as a guide for employing our olfactory senses when researching and studying history in order to understand and communicate the past more fully. Employing ‘olfactory figures’ examples, Smell and the Past shows how historical narratives and arguments can be found through a structured olfactory experience, and demonstrates how our understanding of the past and its relationship with the present is enriched by opening our minds and using our noses. - Abstract
Research assessing personality traits and religiosity across cultures has typically neglected variation across religious affiliations and has been limited to a small number of personality traits. This study examines the relationship between the Big Five personality traits and their facets, two theoretically distinct measures of religiosity, and twelve other personality traits across seven religious affiliations and 61 countries/regions. The proportion of participants following a religion varied substantially across countries (e.g., Indonesia = 99%; Estonia = 7%). Both measures of religiosity were related to agreeableness, conscientiousness, happiness, and fairness; however; relations with religiosity as a social axiom were stronger and less variable across religious affiliations. Additionally, personality-religiosity links were more robust in low-development, high-conflict, and collectivist nations.

The term ‘Rock Art’ is loosely used in this article to refer to prehistoric carvings and paintings. Rock art research has changed profoundly in the last two decades. Partly, this is due to the introduction of more ‘scientific’ methodologies such as digital recording, to overcome the subjective nature of analogue documentation methods. Digital recording offers not only ‘pretty pictures’ but more immediate and quantifiable datasets and methods of analysis. As a result, new research implementing complex, multi-scalar and inter-relational analyses, which do not focus solely on the motifs or the landscape location, but encompass many variables of the rock art assemblages, have been successful in bringing rock art to wider narratives of prehistory. This article reflects on the interaction between rock art and digital archaeology, considering how the application of digital resources has changed the way we think, record and conduct research in this field. It will be illustrated by two main case studies from Iberia: Schematic Art in its painted form, and Atlantic Rock Art, a carving tradition.

24. A Simple and Rapid Protocol for the Isolation of Murine Bone Marrow Suitable for the Differentiation of Dendritic Cells

The generation of bone-marrow-derived dendritic cells is a widely used approach in immunological research to study antigen processing and presentation, as well as T-cell activation responses. However, the initial step of isolating the bone marrow can be time-consuming, especially when larger numbers of precursor cells are required. Here, we assessed whether an accelerated bone marrow isolation method using centrifugation is suitable for the differentiation of FMS-like tyrosine kinase 3 ligand-driven dendritic cells. Compared to the conventional flushing method, the centrifugation-based isolation method resulted in a similar bone marrow cell yield on Day 0, increased cell numbers by Day 8, similar proportions of dendritic cell subsets, and consequently a higher number of type 1 conventional dendritic cells (cDC1) from the culture. Although the primary purpose of this method of optimization was to improve experimental efficiency and increase the output of cDC1s, the protocol is also compatible with the differentiation of other dendritic cell subsets such as cDC2 and plasmacytoid dendritic cells, with an improved output cell count and a consistent phenotype.

Abstract The study used regularized partial correlation network analysis (EBICglasso) to examine the structure of DSM-5 internet gaming disorder (IGD) symptoms (network 1); and the associations of the IGD symptoms in the network with different types of motivation as defined in the self-determination theory i.e., intrinsic motivation (engaging in an activity for something unrelated to the activity), identified regulation (engaging in the activity because it aligns with one’s values and/or goals), external regulation (engagement in activity being driven by external rewards and/or approval), and amotivation (engaging in an activity without often understanding why) (network 2). Participants were 968 adults from the general community. They completed self-rating questionnaires covering IGD symptoms and different types of motivation. The findings for network 1 showed mostly positive connections between the symptoms within the IGD network. The most central symptom was loss of control, followed by continuation, withdrawal symptoms, and tolerance. In general, these symptoms were more strongly connected with each other than with the rest of the IGD symptoms. The findings for network 2 showed that the different types of motivation were connected differently with the different IGD symptoms. For instance, the likeliest motivation for the preoccupation and escape symptoms is intrinsic motivation, and for negative consequences, it is low identified regulation. Overall, the findings showed a novel understanding of the structure of the IGD symptoms, and the motivations underlying them. The clinical implications of the findings for assessment and treatment of IGD are discussed.

32. Design and connectivity : the case of Atlantic rock art

Circles, cup-marks and wavy lines are some of the most emblematic motifs associated with Atlantic Rock Art. The term 'Atlantic' was only introduced in the 1940s and is used throughout this thesis as it reflects the widespread distribution of the prehistoric assemblage of rock art, but also the geographic scope of this investigation. This particular iconography is known from Portugal, through to Spain, Ireland, England and up to Scotland, sharing a number of characteristics. Prior to the use of this expression, Atlantic Art was known by a variety of designations that demonstrate the fragmented character of its historiography and the regional nature of investigations. In 1997 Bradley's study introduced a turning point in investigations, with an inter-regional approach and the premise of Landscape Archaeology. This contrasted with traditional studies, more focused on the motifs and creation of typologies, failing to view Atlantic Art holistically, as a socially meaningful practice. In this thesis I set out to investigate differences and similarities of Atlantic Art. I define what its quintessential characteristics are beyond the motif typologies, and identify regional variations. Contextualizing these similarities and deviations, I assess the social and cultural implications of its creation and use. In each one of my five study areas (one in each country), I subjected empirical data to a three-scale investigation: i) Graphic - to study the motifs, ii) Sensorial - to study the rock medium and iii) Environmental - to study the landscape placement. These were developed under principles of Relational Ontology and Assemblage Theory, combining a multi-scalar methodology with a dynamic perspective of the data, explored through a detailed categorical scheme and its analysis with a Presence/Absence Matrix (PAM), spatial analysis carried out with GIS and Social Network Analysis (SNA) to relate and explore the differences and similarities, relationships and connectivity between the study areas. Concepts of developmental psychology and cultural transmission were used to posit that the tradition spread through methods of teaching. Contextualizing the tradition chronologically, it became clear that it formed another transformative processes that characterised the Neolithic.

They find that high expression of LRRN4CL in melanoma cells increases lung metastases in mice and correlates with poorer survival in melanoma patients, suggesting it could be a promising drug target.

- Abstract
Current coronavirus disease 2019 vaccines face limitations including waning immunity, immune escape by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) variants, limited cellular response, and poor mucosal immunity. We engineered a Clec9A-receptor binding domain (RBD) antibody construct that delivers the SARS-CoV-2 RBD to conventional type 1 dendritic cells. Compared with non-targeting approaches, single dose immunization in mice with Clec9A-RBD induced far higher RBD-specific antibody titers that were sustained for up to 21 months after vaccination. Uniquely, increasing neutralizing and antibody-dependent cytotoxicity activities across the sarbecovirus family was observed, suggesting antibody affinity maturation over time. Consistently and remarkably, RBD-specific follicular T helper cells and germinal center B cells persisted up to 12 months after immunization. Furthermore, Clec9A-RBD immunization induced a durable mono- and poly-functional T-helper 1-biased cellular response that was strongly cross-reactive against SARS-CoV-2 variants of concern, including Omicron subvariants, and with a robust CD8+ T cell signature. Uniquely, Clec9A-RBD single-shot systemic immunization effectively primed RBD-specific cellular and humoral immunity in lung and resulted in significant protection against homologous SARS-CoV-2 challenge as evidenced by limited body weight loss and approximately 2 log10 decrease in lung viral loads compared with non-immunized controls. Therefore, Clec9A-RBD immunization has the potential to trigger robust and sustained, systemic and mucosal protective immunity against rapidly evolving SARS-CoV2 variants.

- Abstract
People in economically advantaged nations tend to evaluate their life as more positive overall and report greater well-being than people in less advantaged nations. But how does positivity manifest in the daily life experiences of individuals around the world? The present study asked 15,244 college students from 62 nations, in 42 languages, to describe a situation they experienced the previous day using the Riverside Situational Q-sort (RSQ). Using expert ratings, the overall positivity of each situation was calculated for both nations and individuals. The positivity of the average situation in each nation was strongly related to the economic development of the nation as measured by the Human Development Index (HDI). For individuals’ daily experiences, the economic status of their nation also predicted the positivity of their experience, even more than their family socioeconomic status. Further analyses revealed the specific characteristics of the average situations for higher HDI nations that make their experiences more positive. Higher HDI was associated with situational experiences involving humor, socializing with others, and the potential to express emotions and fantasies. Lower HDI was associated with an increase in the presence of threats, blame, and hostility, as well as situational experiences consisting of family, religion, and money. Despite the increase in a few negative situational characteristics in lower HDI countries, the overall average experience still ranged from neutral to slightly positive, rather than negative, suggesting that greater HDI may not necessarily increase positive experiences but rather decrease negative experiences. The results illustrate how national economic status influences the lives of individuals even within a single instance of daily life, with large and powerful consequences when accumulated across individuals within each nation.

49. Social transformations from the Middle Bronze Age to the Middle Iron Age in Central Southern Britain

The Later Prehistory of Wiltshire is included within social models built on the neighbouring counties of Dorset (Sharples 1991a) and Hampshire (Cunliffe 1984a) or general accounts of southern Britain (Barrett 1980b; Brück 1999a; Hill 1995a; Rowlands 1980). These focus on hillforts, bronze or independent farmsteads. Utilizing the wealth of new data accumulated by developer funded work, this study re-examines the evidence using a landscape scale approach influenced by community studies. It reveals that current approaches fail to explain the evidence from the study area. Towards the end of the Bronze Age, there is a trend towards transhumance with seasonal, pastoral camps and linear earthworks. Coinciding with the fall from grace of bronze as social mediator, animals became one way through which relations were negotiated. The trend continues into the Earliest Iron Age when large midden sites around Pewsey indicate the exploitation of iron deposits. The middens become centres of craft production for these transhumant communities and facilitate the growth of a broad affinity across the region. The supremacy of the middens lasts as little as 200 years before new sources of iron, continental imports and recycling cause many to be abandoned. However, the contacts made between communities at these sites facilitate the mobilization of the labour required for hillfort construction. The rational for their construction varies according to the prevailing social and economic needs but most of these goals are achieved by the Early Iron Age when most are abandoned. The developed hillforts of the later Middle Iron Age are different to their forebears, sit within an otherwise unsettled landscape and indicate a rise in the level of conflict along the western margin of the area.
