Located along the beam collision axis and shielded from the interaction point by at least 100 m of concrete and rock, the FPF will house experiments that will detect particles outside the acceptance of the existing large LHC experiments and will observe rare and exotic processes in an extremely low-background environment. In this work, we summarize the current status of plans for the FPF, including recent progress in civil engineering in identifying promising sites for the FPF and the experiments currently envisioned to realize the FPF's physics potential. Thermal stress is expected to compromise the persistence of tropical corals throughout their biogeographic ranges, making many reefs inhospitable to corals by the end of the century. We integrated models of local predictions of thermal stress throughout the coming century, coral larval dispersal, and the persistence of a coral’s metapopulation(s) in the Caribbean to investigate broad trends in metapopulation fragmentation and decline. As coral reef patches become inhospitable throughout the next century, the metapopulation of Orbicella annularis is predicted to fragment, with sub-networks centered around highly connected patches and thermal refuges. Some of these are predicted to include the reefs of Colombia, Panama, Honduras, Guatemala, Belize, Southern and Northern Cuba, Haiti, and the Bahamas. Unknown coral population demographic parameters, such as lifetime egg production and stock-recruitment rates, limit the model’s predictions; however, a sensitivity analysis demonstrates that broadscale patterns of fragmentation and metapopulation collapse before the end of the century are consistent across a range of potential parameterizations. Despite dire predictions, the model highlights the potential value in protecting and restoring coral populations at strategic locations that are highly connected and/or influential to persistence. Coordinated conservation activities that support local resilience at low coral cover have the potential to stave off metapopulation collapse for decades, buying valuable time. Thermal refuges are linchpins of metapopulation persistence during moderate thermal stress, and targeted conservation or restoration that supports connectivity between these refuges by enhancing local population growth or sexual propagation may be critically important to species conservation on coral reefs.

Climate change is radically altering coral reef ecosystems, mainly through increasingly frequent and severe bleaching events. Yet, some reefs have exhibited higher thermal tolerance after bleaching severely the first time. To understand changes in thermal tolerance in the eastern tropical Pacific (ETP), we compiled four decades of temperature, coral cover, coral bleaching, and mortality data, including three mass bleaching events during the 1982 to 1983, 1997 to 1998 and 2015 to 2016 El Niño heatwaves. Higher heat resistance in later bleaching events was detected in the dominant framework-building genus, Pocillopora, while other coral taxa exhibited similar susceptibility across events. Genetic analyses of Pocillopora spp . colonies and their algal symbionts (2014 to 2016) revealed that one of two Pocillopora lineages present in the region ( Pocillopora “ type 1”) increased its association with thermotolerant algal symbionts ( Durusdinium glynnii ) during the 2015 to 2016 heat stress event. This lineage experienced lower bleaching and mortality compared with Pocillopora “type 3”, which did not acquire D. glynnii . Under projected thermal stress, ETP reefs may be able to preserve high coral cover through the 2060s or later, mainly composed of Pocillopora colonies that associate with D. glynnii . However, although the low-diversity, high-cover reefs of the ETP could illustrate a potential functional state for some future reefs, this state may only be temporary unless global greenhouse gas emissions and resultant global warming are curtailed.

Limited dispersal of individuals between generations results in isolation by distance, in which individuals further apart in space tend to be less related. Classic models of isolation by distance assume that dispersal distances are drawn from a thin-tailed distribution and predict that the proportion of the genome that is identical by descent between a pair of individuals should decrease exponentially with the spatial separation between them. However, in many natural populations, individuals occasionally disperse over very long distances. In this work, we use mathematical analysis and coalescent simulations to study the effect of long-range (power-law) dispersal on patterns of isolation by distance. We find that it leads to power-law decay of identity-by-descent at large distances with the same exponent as dispersal. We also find that broad power-law dispersal produces another, shallow power-law decay of identity-by-descent at short distances. These results suggest that the distribution of long-range dispersal events could be estimated from sequencing large population samples taken from a wide range of spatial scales.

Caribbean coral reefs are experiencing a shift to algal dominance at the expense of stony corals. Determining the factors leading to algal phase shifts is crucial for assuring the survival of Caribbean coral reefs. In this study, factors controlling the growth of the abundant brown macroalgae Dictyota spp. were investigated by varying herbivory pressure (caging) and nutrients (fertilizer addition) on coral reefs near St. Thomas (US Virgin Islands). Experiment 1 measured Dictyota heights and percent cover at 3 sites (11-20 m depth) and showed no growth response to nutrient addition and a weak negative response to herbivory. To confirm results of Experiment 1, a caging and nutrient manipulation (Experiment 2) was conducted at one site (14 m depth) using the dependent variable Dictyota biomass. A strong negative response of growth to nutrient addition was shown, presumably because of nutrient inhibition, and an equally negative response to herbivory (loss of ~50% biomass over 21 d). The inhibitory effect of fertilization on growth was confirmed in a third experiment that showed increasing biomass loss over 4 treatment levels of increasing fertilizer addition (0 [ambient], 5, 10, 20 g). Overall, Dictyota was not nutrient limited at any sites, and was weakly controlled by herbivore populations. Factors responsible for Dictyota abundance on Caribbean reefs may reflect decreased herbivory caused by overfishing and reductions in coral cover and do not appear to be affected by recent changes in nitrogen or phosphorus load. This study reinforces the need for conservation and management of herbivores in coral reef ecosystems, to mitigate the effects from anthropogenic stressors.

Orbicellid corals are threatened primary reef-building corals throughout the Caribbean in shallow and mesophotic coral ecosystems (MCEs), yet a poor understanding of where they occur limits population monitoring and management. The goals of this study were to predict suitable habitat for orbicellid coral species and to identify how abiotic environmental factors constrain that habitat on the eastern Puerto Rico Shelf. The probability of occurrence for Orbicella annularis and O. faveolata/O. franksi (combined) from shallow to mesophotic depths on the eastern Puerto Rico Shelf was predicted using maximum entropy models. Contributions of abiotic predictors, including bathymetry, seafloor topography, temperature, wave exposure, and bottom velocity, were assessed. Model performance was assessed using area under the receiver operating characteristic curve, standard error of the replicate model runs, and mean absolute error. Both O. annularis and O. faveolata/O. franksi distributions were best predicted by rugosity, temperature, and wave exposure. O. faveolata/O. franksi occurred at shallow and mesophotic depths, and acted as a proxy for identifying the spatial extent of MCEs, contrary to O. annularis, which was predicted at shallow depths. Results for O. faveolata/O. franksi in mesophotic depths indicated potential for large areas of unexplored and unmonitored MCEs along the southeast shelf of St. Thomas, US Virgin Islands and within the Virgin Passage. These spatial predictions of potential mesophotic reef habitats will provide direction for future MCE exploration efforts.

Stony coral tissue loss disease (SCTLD) is a widespread and deadly disease that affects nearly half of Caribbean coral species. To understand the microbial community response to this disease, we performed a disease transmission experiment on US Virgin Island (USVI) corals, exposing six species of coral with varying susceptibility to SCTLD. The microbial community of the surface mucus and tissue layers were examined separately using a small subunit ribosomal RNA gene-based sequencing approach, and data were analyzed to identify microbial community shifts following disease acquisition, potential causative pathogens, as well as compare microbiota composition to field-based corals from the USVI and Florida outbreaks. While all species displayed similar microbiome composition with disease acquisition, microbiome similarity patterns differed by both species and mucus or tissue microhabitat. Further, disease exposed but not lesioned corals harbored a mucus microbial community similar to those showing disease signs, suggesting that mucus may serve as an early warning detection for the onset of SCTLD. Like other SCTLD studies in Florida, Rhodobacteraceae, Arcobacteraceae, Desulfovibrionaceae, Peptostreptococcaceae, Fusibacter, Marinifilaceae, and Vibrionaceae dominated diseased corals. This study demonstrates the differential response of the mucus and tissue microorganisms to SCTLD and suggests that mucus microorganisms may be diagnostic for early disease exposure.

Many studies have evaluated the impacts of hurricanes on coral communities, but far less is known about impacts, recovery, and resilience of sponge communities to these extreme events. In September 2017, St. Thomas, U.S. Virgin Islands, was impacted by two Category 5 hurricanes within 2 weeks: Hurricanes Irma and Maria. Such extreme events occurring in such rapid succession are virtually unprecedented. Pre-hurricane (2015, 2016) surveys of permanent transects at six sites around St. Thomas were compared with those at 10 weeks post-hurricanes (December 2017) to evaluate storm impacts on sponges and on benthic coral reef constituents. These surveys also established a baseline for evaluating future recovery. Percent cover of sponges declined by 24.9% post-hurricanes. In contrast, sponge density increased by 43.9% from 2015 to 2016 and declined slightly after the hurricanes. Overall sponge volume did not vary over time, and whereas sponge diversity was similar in 2015 and 2016, it increased post-hurricanes. Sponge morphologies were differentially affected by the hurricanes; the proportion of upright sponges declined by 36.9%, while there was a 24.4% increase in encrusting sponges. Coral and macroalgal cover did not change significantly over the sampling period, while percent cover of epilithic algae increased and non-living substrata decreased from 2015 to 2016 but did not change further post-hurricanes. At all sites, recruitment and/or regrowth of sponges was observed within 10 weeks post-hurricanes, indicating potential resilience in Caribbean sponge communities. Whether these sponge communities return to pre-hurricane conditions and how long that will take remains to be seen.

Quantum geometry is a key quantity that distinguishes electrons in a crystal from those in the vacuum. Its study continues to provide insights into quantum materials, uncovering new design principles for their discovery. However, unlike the Berry curvature, an intuitive understanding of the quantum metric is lacking. Here, we show that the quantum metric of Bloch electrons leads to a momentum-space gravity. In particular, by extending the semiclassical formulation of electron dynamics to second order, we find that the resulting velocity is modified by a geodesic term and becomes the momentum-space dual of the Lorentz force in curved space. We calculate this geodesic response for magic-angle twisted bilayer graphene and show that moir\'e systems with flat bands are ideal candidates to observe this effect. Extending this analogy with gravity further, we find that the momentum-space dual of the Einstein field equations remains sourceless for pure states while for mixed states it acquires a source term that depends on the von Neumann entropy, for small entropies. We compare this stress-energy equation with the weak-field limit of general relativity and conclude that the von Neumann entropy is the momentum-space dual of the gravitational potential. Consequently, the momentum-space geodesic equation for mixed states is modified by a term resembling an entropic force. Our results highlight connections between quantum geometry, momentum-space gravity and quantum information, prompting further exploration of this dual gravity in quantum materials., Comment: 21 pages, 4 figures. Ciguatera poisoning (CP) poses a significant threat to ecosystem services and fishery resources in coastal communities. The CP-causative ciguatoxins (CTXs) are produced by benthic dinoflagellates including Gambierdiscus and Fukuyoa spp., and enter reef food webs via grazing on macroalgal substrates. In this study, we report on a 3-year monthly time series in St. Thomas, US Virgin Islands where Gambierdiscus spp. abundance and Caribbean-CTX toxicity in benthic samples were compared to key environmental factors, including temperature, salinity, nutrients, benthic cover, and physical data. We found that peak Gambierdiscus abundance occurred in summer while CTX-specific toxicity peaked in cooler months (February–May) when the mean water temperatures were approximately 26–28 °C. These trends were most evident at deeper offshore sites where macroalgal cover was highest year-round. Other environmental parameters were not correlated with the CTX variability observed over time. The asynchrony between Gambierdiscus spp. abundance and toxicity reflects potential differences in toxin cell quotas among Gambierdiscus species with concomitant variability in their abundances throughout the year. These results have significant implications for monitoring and management of benthic harmful algal blooms and highlights potential seasonal and highly-localized pulses in reef toxin loads that may be transferred to higher trophic levels.

White plague disease has caused widespread coral mortality and affects over 30 Caribbean coral species, yet how different coral species respond to disease exposure has not been tested experimentally. This study quantified white plague transmission from Orbicella franksi to three susceptible and abundant coral species in the U.S. Virgin Islands: Orbicella annularis, Siderastrea siderea, and Porites astreoides. White plague was observed in 83% of tested O. annularis corals within 5.8 ± 1.1 (mean ± SE) d, 42% of S. siderea corals within 9.1 ± 1.4 d, and 17% of P. astreoides corals within 12 ± 0.87 d. Results from this study indicate that: (1) O. annularis is significantly more susceptible to white plague than S. siderea and P. astreoides, and (2) white plague infection occurs more rapidly in O. annularis than in P. astreoides. These results are important for understanding how multi-species diseases may impact coral species assemblages.

Stony coral tissue loss disease (SCTLD) was initially documented in Florida in 2014 and outbreaks with similar characteristics have since appeared in disparate areas throughout the northern Caribbean, causing significant declines in coral communities. SCTLD is characterized by focal or multifocal lesions of denuded skeleton caused by rapid tissue loss and affects at least 22 reef-building species of Caribbean corals. A tissue-loss disease consistent with the case definition of SCTLD was first observed in the U.S. Virgin Islands (USVI) in January of 2019 off the south shore of St. Thomas at Flat Cay. The objective of the present study was to characterize species susceptibility to the disease present in St. Thomas in a controlled laboratory transmission experiment. Fragments of six species of corals (Colpophyllia natans, Montastraea cavernosa, Orbicella annularis, Porites astreoides, Pseudodiploria strigosa, and Siderastrea siderea) were simultaneously incubated with (but did not physically contact) SCTLD-affected colonies of Diploria labyrinthiformis and monitored for lesion appearance over an 8 day experimental period. Paired fragments from each corresponding coral genotype were equivalently exposed to apparently healthy colonies of D. labyrinthiformis to serve as controls; none of these fragments developed lesions throughout the experiment. When tissue-loss lesions appeared and progressed in a disease treatment, the affected coral fragment, and its corresponding control genet, were removed and preserved for future analysis. Based on measures including disease prevalence and incidence, relative risk of lesion development, and lesion progression rates, O. annularis, C. natans, and S. siderea showed the greatest susceptibility to SCTLD in the USVI. These species exhibited earlier average development of lesions, higher relative risk of lesion development, greater lesion prevalence, and faster lesion progression rates compared with the other species, some of which are considered to be more susceptible based on field observations (e.g., P. strigosa). The average transmission rate in the present study was comparable to tank studies in Florida, even though disease donor species differed. Our findings suggest that the tissue loss disease affecting reefs of the USVI has a similar epizootiology to that observed in other regions, particularly Florida.

Background Understanding trends in surgical volumes can help Ambulatory Surgery Centers (ASCs) prevent clinician burnout and provide adequate staffing while maintaining the quality of patient care throughout the year. Health insurance deductibles reset in January each year and may contribute to an annual rhythm where the levee of year-end deductibles is breached in the last few months of every year, resulting in a flood of cases and several accompanying challenges. This study aims to identify and analyze monthly and yearly surgical volume patterns in ASCs and explore a relationship with the deductible reset. Methods De-identified, aggregate visit data for 2016–2019 were obtained retrospectively from 14 ambulatory surgery centers within the same benchmarking consortium in the Southeast. The ASCs subspecialty types consisted of orthopedics, urology, otolaryngology, and multispecialty. Kaiser Family Foundation survey data from 2016 to 2019 was used to inform deductible trends. Augmented Dickey-Fuller tests, linear regressions, and two-sample T-tests were conducted to explore and establish patterns in surgical volume between 2016 and 2019. Results Overall, average orthopedic surgical volume increased 38.04% from January to December in 2016–2019 with an average difference of 64 cases (95% CI: 47–80), while that of all ASCs combined increased 19.24% within the same timeframe with an average difference of 37 cases (95% CI: 21–52). Average health insurance deductibles rose 12% from $1476 to $1655 within the same timeframe. Regression analysis showed a stronger association between year and volume for orthopedic ASCs (R (Claxton et al., 2019) [2] = 0.796) than for all ASCs combined (R (Claxton et al., 2019) [2] = 0.645). Regression analysis also showed a stronger association between month and volume for orthopedic ASCs (R (Claxton et al., 2019) [2] = 0.488–0.805) than for all ASCs combined (R (Claxton et al., 2019) [2] = 0.115–0.493). Conclusion This study is first to identify regular and predictable yearly and monthly increases in orthopedic ASCs surgical volume. The study also identifies yearly increases in surgical volume for all ASCs. The combination of increasing yearly demand for orthopedic surgery and growing association between month and volume leads to an unnecessary year-end rush. Caribbean coral cover has decreased substantially in recent decades, with much of the live coral being replaced by macroalgae. Encrusting red algae in the genus Ramicrusta have become abundant throughout the region and have demonstrated widespread harm to corals by overgrowing living tissue, causing colony mortality, and impairing coral recruitment. In this research, Ramicrusta textilis was identified by morpho-anatomy and DNA sequencing from nine sites around St. Thomas, US Virgin Islands, and 3D photogrammetry was used to measure the rate of algal growth on stony corals. 3D models of individual coral colonies (five species plus controls, N = 72) competing with R. textilis revealed differential competitive abilities among taxa, with Siderastrea siderea being the only species capable of inhibiting overgrowth by the alga (mean linear algal growth − 1.1 mm yr−1). Important reef building coral species such as Orbicella annularis and Orbicella faveolata were poor competitors (mean linear algal growth + 15 mm yr−1 and + 7.7 mm yr−1, respectively), indicating that the emergence of the alga could have significant impacts on Caribbean coral reef species diversity, community composition, and structural complexity.

Stony coral tissue loss disease (SCTLD) was first observed in the United States Virgin Islands in January 2019 on a reef at Flat Cay off the island of St. Thomas. A year after its emergence, the disease had spread to several reefs around St. Thomas causing significant declines in overall coral cover. Rates of tissue loss are an important metric in the study of coral disease ecology, as they can inform many aspects of etiology such as disease susceptibility and resistance among species, and provide critical parameters for modeling the effects of disease among heterogenous reef communities. The present study quantified tissue loss rates attributed to SCTLD among six abundant reef building species (Colpophyllia natans, Montastraea cavernosa, Diploria labyrinthiformis, Pseudodiploria strigosa, Orbicella annularis, and Porites astreoides). Field-based 3D models of diseased corals, taken approximately weekly, indicated that the absolute rates of tissue loss from SCTLD slowed through time, corresponding with the accumulation of thermal stress that led to mass bleaching. Absolute tissue loss rates were comparable among species prior to the bleaching event but diverged during and remained different after the bleaching event. Proportional tissue loss rates did not vary among species or through time, but there was considerable variability among M. cavernosa colonies. SCTLD poses a significant threat to reefs across the Caribbean due to its persistence through time, wide range of susceptible coral species, and unprecedented tissue loss rates. Intervention and management efforts should be increased during and immediately following thermal stress events in order maximize resource distribution when disease prevalence is decreased.

Ecosystem degradation due to anthropogenic activities is the primary issue of our times. Theoretical analyses as well as efforts to restore and manage ecosystems depend on comprehensive metrics of ecosystem function. In the case of complex ecosystems such as tropical coral reefs—especially where monitoring, management, and restoration are important—multiple metrics reflecting key functional groups are required to accurately reflect ecosystem function and when necessary, diagnose degree and kind of ecosystem degradation. We propose inclusion of the generalist ectoparasite functional group as a measure of ecosystem function of coral reefs. This functional group is adaptable to loss of other community members and may experience an increase in abundance as ecosystem function declines. Fish‐parasitic gnathiid isopods are a member of this group, resident though inconspicuous in coral‐reef communities. On Caribbean coral reefs, based on 938 light‐trap samples, we observed a negative correlation between abundance of smaller‐sized gnathiids and abundance of live coral, a natural predator of gnathiids. Plots grouped by coral cover—a measure of success of the ecosystem engineer—and ectoparasite abundance varied significantly in community composition including abundance of macroalgae, turf algae, and farming Stegastes spp. damselfish reflecting shifts in community structure. Changes in gnathiid abundance with respect to the abundance of organisms participating in each of the core functional processes driving coral‐reef ecosystems reflect broad connectivity of gnathiid parasites across the ecosystem. We conclude that the hyperabundance of a small, cryptic, generalist parasite, when used in combination with a metric of abundance of the primary ecosystem engineer, can provide one nuanced measure of the ecosystem vulnerability to collapse.

Purpose: To assess the efficacy and safety of amniotic fluid therapy injections in patients with mild to moderate trigger finger. Methods: All participants received 1 mL of amniotic fluid injected into the tendon sheath of the affected tendon. Pretreatment and posttreatment data were collected for triggering frequency, Disabilities of the Arm, Shoulder, and Hand (DASH) questionnaire scores, and numerical pain rating scale scores. Results: Of 111 digits from 96 patients, 51% experienced clinically notable improvement and did not receive an alternative treatment. Average length of follow-up was 11 months. From baseline to end of follow-up, average pain score (0–10) decreased from 5.19 to 1.19 (P < .001), median triggering per day decreased from 5 to 0 (P < .001), and median DASH score (1–100) decreased from 20 to 6.03 (P < .001). There was a 50% success rate in patients with diabetes and a 52.6% success rate in digits diagnosed with concomitant Dupuytren contracture in the same hand. Conclusions: Amniotic fluid therapy injections may offer a biologic alternative for conservative treatment of trigger finger, particularly for patients with diabetes. Decreased pain, decreased triggering, and improved DASH scores offer preliminary evidence supporting the use of amniotic injections for stenosing tenosynovitis. Ciguatera fish poisoning is a seafood-toxin illness resulting from consumption of fish contaminated with ciguatoxins. Managing ciguatera fish poisoning is complex. It is made easier, however, by local fishers from endemic areas reporting regional predictability for local fish species' ciguatera fish poisoning risk, which the present study then tested. We investigated the prevalence of ciguatoxins in 4 commonly marketed and consumed species (Balistes vetula, Haemulon plumierii, Ocyurus chrysurus, and Epinephelus guttatus) across an oceanic gradient (north, south, east, and west) from the US Virgin Islands. Fish muscle extracts were analyzed for Caribbean ciguatoxins using an in vitro mouse neuroblastoma (N2a) cytotoxicity assay and confirmed by liquid chromatography-tandem mass spectrometry (LC-MS/MS). Fish collected from the north location had 0 fish with detectable ciguatoxins; this site also had the greatest wave energy. Caribbean ciguatoxins in fish ranged from 0.01 to 0.11, 0.004 to 0.10, and 0.005 to 0.18 ng Caribbean ciguatoxin-1 eq/g, from the west, east, and south respectively. Ciguatoxin-like activity was detectable by the N2a assay in 40, 41, 50, and 70% of H. plumierii, O. chrysurus, B. vetula, and E. guttatus, respectively. Of the fish collected, 4% had Caribbean ciguatoxin levels exceeding the US Food and Drug Administration guidance of 0.1 ng Caribbean ciguatoxin-1 eq/g fish. These findings concurred with spatial ciguatera fish poisoning prevalence information provided by local fishers in the US Virgin Islands and demonstrate how partnerships between researchers and fishers can aid the improvement of science-based ciguatera fish poisoning management. Mesophotic coral ecosystems (MCEs) are deep (> 30 m), light-dependent communities that are abundant in many parts of the global ocean. MCEs are potentially connected to shallow reefs via larval exchange and may act as refuges for reef organisms. However, MCE community level recovery after disturbance, and thus, community resilience, are poorly understood components of their capacity as refuges. To assess the potential for disturbance and growth to drive community structure on MCEs with differential biophysical conditions and coral communities, we collected colonies of Orbicella franksi and Porites astreoides and used computerized tomography to quantify calcification. The divergence of coral growth rates in MCEs with different environmental conditions may be species specific; habitat-forming O. franksi have slow and consistent growth rates of ~ 0.2 cm yr−1 below 30 m, regardless of mesophotic habitat, compared to ~ 1.0 cm yr−1 in shallow-water habitats. Slow skeletal growth rates in MCEs suggest that rates of recovery from disturbance will likely also be slow. Localized buffering of MCEs from the stressors affecting shallow reefs is therefore crucial to the long-term capacity of these sites to serve as refugia, given that skeletal extension and recovery from disturbance in MCEs will be significantly slower than on shallow reefs.

The Gambierdiscus genus is a group of benthic dinoflagellates commonly associated with ciguatera fish poisoning (CFP), which is generally found in tropical or sub-tropical regions around the world. Morphologically similar species within the genus can vary in toxicity; however, species identifications are difficult or sometimes impossible using light microscopy. DNA sequencing of ribosomal RNA genes (rDNA) is thus often used to identify and describe Gambierdiscus species and ribotypes, but the expense and time can be prohibitive for routine culture screening and/or large-scale monitoring programs. This study describes a restriction fragment length polymorphism (RFLP) typing method based on analysis of the large subunit rDNA that can successfully identify at least nine of the described Gambierdiscus species and two Fukuyoa species. The software programs DNAMAN 6.0 and Restriction Enzyme Picker were used to identify a set of restriction enzymes (SpeI, HpyCH4IV, and TaqαI) capable of distinguishing most of the known Gambierdiscus species for which DNA sequences were available. This assay was tested using in silico analysis and cultured isolates, and species identifications of isolates assigned by RFLP typing were confirmed by DNA sequencing. To verify the assay and assess intra-specific heterogeneity in RFLP patterns, identifications of 63 Gambierdiscus isolates comprising ten Gambierdiscus species, one ribotype, and two Fukuyoa species were confirmed using RFLP typing, and this method was subsequently employed in the routine identification of isolates collected from the Caribbean Sea. The RFLP assay presented here reduces the time and cost associated with morphological identification via scanning electron microscopy and/or DNA sequencing, and provides a phylogenetically sensitive method for routine Gambierdiscus species assignment.

The genus Gambierdiscus is a recognized group of marine epiphytic-benthic dinoflagellates that produce the toxins that cause ciguatera fish poisoning (CFP). To date, thirteen species and six ribotypes of Gambierdiscus have been identified, and multiple species commonly co-occur within a single site or epiphyte community. Toxicity can vary by species, thus it is important to be able to differentiate among species for research and monitoring purposes. Gambierdiscus species have very similar morphological characteristics and are difficult or impossible to distinguish using light microscopy. DNA sequencing has been an important tool in the definition of Gambierdiscus species, but it can be time-consuming and relatively expensive. To provide an alternative approach, a PCR-RFLP protocol was developed for efficient, rapid, and cost-effective identification of Gambierdiscus strains isolated from the Gulf of Mexico and Caribbean Sea, where CFP cases and Gambierdiscus spp. have been reported. The assay targets the D1-D2 hypervariable regions of the large subunit ribosomal RNA gene and uses a single restriction enzyme, BsrI. This method produces distinct RFLP banding patterns for the six species of Gambierdiscus reported from the Gulf of Mexico and Caribbean Sea, and also distinguishes them from four Pacific endemic species. This method was successfully used to type 465 clonal isolates of Gambierdiscus from the U.S. Virgin Islands and Akumal Beach - Mexico This BsrI PCR-RFLP method expands the tools available to researchers and managers engaged in monitoring activities and ecological studies.

Purpose: Trapeziectomy with suture button suspensionplasty (SBS) to treat thumb carpometacarpal (CMC) arthritis has been proposed as an alternative to ligament reconstruction tendon interposition. There have been limited large-scale or long-term reports regarding SBS outcomes. Single-surgeon intermediate follow-up is reported. Methods: We conducted a retrospective review of patients undergoing SBS procedures by a single surgeon. Implant manufacturer and postoperative immobilization protocol were recorded. Surgical outcomes, complications, and revision procedures were identified. Postoperative Disabilities of the Arm, Shoulder, and Hand scores were collected. Results: A total of 242 SBS surgeries were included, involving 215 patients, average age 64.82 years (range, 42–86 years). Average follow-up was 35 ± 25 months. In all, 183 Arthrex and 59 Stryker systems were used, 42 of which were immobilized for 6 weeks after surgery and 200 of which were mobilized at 2 weeks afterward. Postoperative Disabilities of the Arm, Shoulder, and Hand surveys were completed by 122 patients (57%), with an average score of 12. No scaphometacarpal abutment was reported. Thirteen complications were reported (5%), 7 of which were implant-associated (3%) and 6 of which were not (2%). Implant-associated complications consisted of 3 suture button pull-outs, 2 thumb–index metacarpal abutments, one suture tail irritation, and one index metacarpal fracture. Operative revision was required in 4 of 7 implant-associated cases and 5 of 6 non–implant associated cases. No suture button pull-outs required revision surgery. Conclusions: Results for a large series of SBS for CMC arthroplasty with intermediate follow-up revealed excellent clinical outcomes and low complication rates. Clinical relevance: Suture button suspensionplasty as an alternative to ligament reconstruction tendon interposition may be a viable option for treating thumb CMC arthritis. In addition, a technique to manage thumb–index metacarpal abutment is described. Disturbances are a natural part of the ecology of reef ecosystems including mesophotic coral ecosystems (MCEs). Storms, thermal stress, and volcanism are all documented as direct or indirect impacts on MCEs and have been shaping these systems for millennia. In general, anthropogenic disturbances are increasingly challenging community resistance and resilience and, in some cases, altering community composition. Potential anthropogenic disturbances to MCEs include the effects of climate change (warming waters, extreme temperature fluctuations, sea level rise, and increased intensity and frequency of storms), ocean acidification, physical impacts (marine debris, anchoring, benthic infrastructure, and other mechanical disturbances), harvesting for fisheries and the aquarium trade, impacts from coastal development (turbidity and sedimentation), pollution, invasive species introduction, and increases in disease outbreaks. Many of these disturbances are shown to impact MCEs, with subsequent degradation occurring just as these systems are coming into increasing scientific and management focus. Thermal stress and ocean acidification are suggested to pose the greatest existential threat to MCEs, while many local disturbances are amenable to local management strategies. Increasing knowledge of the distribution and structure of MCEs is a critical first step in management.

Accounting of the goods and services provided by ecosystems to human communities provides a basis for informed sustainable development, policy, and conservation decision-making. Coral reefs provide a myriad of such goods and services to coastal communities through direct provisioning (e.g., calories and natural products), environmental supporting and regulating services (e.g., nutrient or trophic cycling and stock support), and cultural products (e.g., tourism and culturally important ecosystems). Mid-depth coral communities (30–150 m), or mesophotic coral ecosystems (MCEs), are not generally addressed in ecosystem services accounting for coral reefs; however, they may share many of the services associated with shallow coral reefs, as well as provide unique ecosystem services of their own. The growing understanding that MCEs occupy large areas of previously uncharacterized insular and continental shelves suggests coral reef valuations need to account for these ecosystems. As shallower resources continue to decline due to anthropogenic pressures, it is crucial that we understand how MCEs support coastal ecosystems and human communities. Here, we explore the development of an ecosystem services framework for MCEs in the context of those provided by shallow coral reefs and present a baseline for further development as new data and information about MCEs become available. We recommend future research properly account for and valuate MCE ecosystem services, both individually and as they relate to ecosystem services for shallow-water reefs.

The rapid deterioration of coral reefs worldwide has led to a growing interest in identifying areas that can offer protection against adverse conditions including coral reef communities at intermediate (~15 to 30 m) and mesophotic (≥30 m) depths. However, various concepts regarding the protective potential of deeper coral reef communities, and subsequent roles in overall reef resilience and persistence, remain poorly defined. Herein, we organize these ideas into an initial conceptual framework and review for scleractinian corals how these ideas may be supported by the limited empirical data that is currently available. We distinguish between the concepts of “depth refuges,” “depth refugia,” and “depth resilience areas,” based on the nature (i.e., avoidance versus resilience) and temporal scope of protection. Although past examples have confirmed the role of mesophotic coral ecosystems as short-term, ecological “refuges,” there is thus far little support that they comprise long-term “refugia.” In contrast, the concept of “deep resilience areas,” reef communities that persist long-term through disturbance by resistance and recovery, remains largely unexplored. In terms of the functional roles of such protected areas in the overall coral reef ecosystem, we distinguish between the concepts of “reseeding” and “local persistence.” The potential to actively reseed shallow reefs may be ecologically important, but only for a small proportion of shared biodiversity, whereas the potential to promote persistence of local biodiversity may apply across a broad range of coral reef species. Although empirical evidence remains very limited, we hope that the incipient conceptual delineations presented here provide a constructive reference for further discussion and research into the ecological importance of deep reef communities.

Anchored mesh screens have been suggested as a standardized approach to monitor the cell abundances of epiphytic dinoflagellates in benthic habitats, including toxigenic members of the Gambierdiscus genus responsible for ciguatera poisoning (CP). Here we deployed screens for 24h at eight sites in the Florida Keys and St. Thomas (US Virgin Islands) to evaluate their performance relative to the traditional method of assessing Gambierdiscus abundance in which cell counts are normalized to wet weight of host algae. The 30-month study (April 2013 – August 2015) involved monthly sampling at sites where screens were suspended at near-bottom locations for a 24h period and retrieved, with concurrent collections of macrophytes; including Halimeda, Laurencia, and Thalassia in the Florida Keys, and Dictyota in both regions. Gambierdiscus cells were identified and enumerated in the screen and macrophyte samples, and several regression techniques were evaluated (linear regression using untransformed and log-transformed data; negative binomial distribution (NBD) regression) to determine how well the screen-derived data could estimate algal cell concentrations on the host algae. In all cases, the NBD models performed the best based on Akaike Information Criteria values, although 38% of the regressions were not statistically-significant, including all of the St. Thomas sites. The r(2) values were all < 0.75 and averaged 0.36, indicating relatively poor fit of the screen data. False negative results (regression models underestimating actual cell abundances) were common occurrences, ranging from 5 to 74% of the scenarios tested. In summary, these results indicate that 24h screen deployments do not appear to be consistent in all situations. Caution is therefore needed when considering 24h screens as a standardized monitoring approach for quantifying Gambierdiscus population dynamics across geography and ecosystems. Furthermore, neutral (artificial) substrates may not adequately capture either the host preference or palatability that likely influence the initial vector of toxin incorporation in the food web via herbivory on these macrophytes.

Ciguatera fish poisoning (CFP) is a syndrome caused by the bioaccumulation of lipophilic ciguatoxins in coral reef fish and their subsequent consumption by humans. These phycotoxins are produced by Gambierdiscus spp., tropical epiphytic dinoflagellates that live on a variety of macrophytes, as well as on dead corals and sand. Recent taxonomic studies have identified novel diversity within the Gambierdiscus genus, with at least 18 species and several sub-groups now identified, many of which co-occur and differ significantly in toxicity. The ability to accurately and quickly distinguish Gambierdiscus species in field samples and determine community composition and abundance is central to assessing CFP risk, yet most Gambierdiscus species are indistinguishable using light microscopy and other enumeration methods are semi-quantitative. In order to investigate the spatial and temporal dynamics of Gambierdiscus species and community toxicity, new tools for species identification and enumeration in field samples are needed. Here, fluorescence in situ hybridization (FISH) probes were designed for seven species commonly found in the Caribbean Sea and Pacific Ocean, permitting their enumeration in field samples using epifluorescence microscopy. This technique enables the assessment of community composition and accurate determination of cell abundances of individual species. Molecular probes detecting G. australes, G. belizeanus, G. caribaeus, G. carolinianus, G. carpenteri, and the G. silvae/G. polynesiensis clade were designed using alignments of large subunit ribosomal RNA (rRNA) sequences. These probes were tested for specificity and cross-reactivity using Gambierdiscus cultures, and a series of experiments were performed in which field samples spiked with known concentrations of Gambierdiscus cultures were analyzed to confirm that Gambierdiscus can be successfully detected and enumerated by FISH in the presence of detritus and other organisms. These probes were then used to characterize Gambierdiscus community structure of field samples collected from the Florida Keys and Hawai’i, USA. The probes revealed the co-occurrence of multiple species at each location. Time-series FISH analyses of samples collected from the Florida Keys quantified seasonal shifts in community composition as well as fluctuations in overall Gambierdiscus cell abundance. Application of species-specific FISH probes provides a powerful new tool to those seeking to target individual Gambierdiscus species, including significant toxin-producers, in field populations. Moving forward, analysis of Gambierdiscus community composition across multiple environments and over time will also allow species dynamics to be linked to environmental parameters, improving our ability to understand and manage the current and changing risks of CFP worldwide.

Coral reef ecosystems are declining due to multiple interacting stressors. A bioassessment framework focused on stressor-response associations was developed to help organize and communicate complex ecological information to support coral reef conservation. This study applied the Biological Condition Gradient (BCG), initially developed for freshwater ecosystems, to fish assemblages of U.S. Caribbean coral reef ecosystems. The reef fish BCG describes how biological conditions changed incrementally along a gradient of increasing anthropogenic stress. Coupled with physical and chemical water quality data, the BGC forms a scientifically defensible basis to prioritize, protect and restore water bodies containing coral reefs. Through an iterative process, scientists from across the U.S. Caribbean used fishery-independent survey data and expert knowledge to develop quantitative decision rules to describe six levels of coral reef ecosystem condition. The resultant reef fish BCG provides an effective tool for identifying healthy and degraded coral reef ecosystems and has potential for global application.

Population genetic studies provide insights into intraspecific diversity and dispersal patterns of microorganisms such as protists, which help understanding invasions, harmful algal bloom development and occurrence of seafood poisoning. Genetic differentiation across geography has been reported in many microbial species indicating significant dispersal barriers among different habitats. Temporal differentiation has been less studied and its frequency, drivers and magnitude are poorly understood due to a lack of integral studies. The toxic dinoflagellate species Gambierdiscus caribaeus was sampled during two years in the Florida Keys, and repeatedly from 2006 to 2016 at St. Thomas, US Virgin Islands (USVI), including a three-year period with monthly sampling, to compare spatial and temporal genetic differentiation. Samples from the USVI site showed high temporal variability in local population structure, which correlated with changes in salinity and benthic habitat cover. In some cases, temporal variability exceeded spatial differentiation, despite apparent lack of connectivity and dispersal across the Greater Caribbean Region based on the spatial genetic data. Thus, local processes such as selection might have a stronger influence on population structure in microorganisms than geographic distance. The observed high temporal genetic diversity challenges the prediction of harmful algal blooms and toxin concentrations, but illustrates also the evolutionary potential of microalgae to respond to environmental change.
