Giriş: İnsülin ve kolon, akciğer ve endometrium kanserleri gibi bazı kanser türleri arasında pozitif bir korelasyon olduğu bilinmektedir. Prostat kanseri de bunlardan birisidir. Bu çalışmadaki amacımız insülin geni 3" UTR bölgesinde lokalize olan INS+1127 PstI polimosfizminin prostat kanseri etiyolojinde rolünü araştırmaktır. Materyal ve Metot: Çalışmaya 71 prostat kanseri tanısı almış hasta ve 119 normal kontrol bireyi dahil edilmiştir. INS+1127 PstI polimorfizm araştırması için PCR ve RFLP tekniği kullanılmıştır. Bulgular: Çalışmanın sonucunda INS geni PstI polimorfizmi genotipleri prostat kanserli hastalarda (n:71) %67,6 CC, %28,2 CT, %4,2 TT; kontrol grubunda (n: 119) %78,2 CC, %21 CT, %0,8 TT olarak belirlenmiştir. Sonuç: Hasta ve kontroller arasındaki genotip dağılımında istatistiksel olarak anlamlı bir farklılık bulunamamıştır (P=0,078).

Purpose: There is positive correlation between insulin and various cancers such as colon, lung and endometrium. Protate cancer is the one of them. The aim of this study, to determine the incidance of INS+1127 PstI polymorphism, located in the 3' UTR of insulin gene, between prostate cancer cases and controls and to clarify if this polymorphism have any role in the etiology of prostate cancer., Methods: We investigated 71 prostate cancer cases and 119 normal controls unrelated to patients. INS+1127 PstI polymorphism was investigated with PCR and RFLP method., Results: At the end of study; INS+1127 PstI polymorphism genotypes out to be 67.6 % CC, 28.2 % CT, 4.2 % TT among prostatic cancer individuals and 78.2% CC, 21% CT, 0.8% TT among control group individuals., Conclusion: As a result, there was no significant difference between patients and control group, in the genotype distribution.

Meme kanseri, tüm dünyada, kadınlar arasında en sık görülen kanser türüdür. Meme kanserinde kemoterapi genellikle adjuvan ve neoadjuvan olarak uygulanmaktadır. Adriamisin ve taksoter, meme kanseri kemoterapisinde sıkça kullanılan ajanlardır.Kemoterapi, yüksek maliyetli bir tedavi türüdür. Kemoterapide kullanılan ajanların toksik yan etkileri kullanımlarını kısıtlayan bir unsurdur. Uygulanan rejimlere bireysel cevap farklılıkları diğer bir problemdir. Bu nedenle, kemoterapiye yanıt hakkında bilgi verebilecek öngörüsel markırların belirlenmesi amacıyla çalışmalar yapılmaktadır.TWIST transkripsiyon faktörü, gelişimde mezoderm oluşumu ve farklılaşmasında görevli bir bazik heliks lup heliks proteinidir. İnsanlarda Twist proteinini kodlayan TWIST1 genindeki mutasyonlar Seathre-Chotzen Sendromu'na yol açmaktadır. Twistin anti-apoptotik, anjiyojenik ve metastatik süreçlerde rol oynadığı belirlenmiştir. Son dönemlerde, Twist transkripsiyon faktörünün, kemoterapötik ajanlara dirençte de rolü olduğu bildirilmektedir.Çalışmamızda, daha önce kemoterapi almamış hastalardan alınan meme tümörlerinde Twist expresyonu ile adiramisin ve taksoter hassasiyeti arasında bir ilişki olup olmadığını araştırmak amaçlanmıştır. Tümörlerin bu iki ajana hassasiyeti ATP-TCA ile belirlenmiştir.Çalışma sonunda, meme tümörlerinde Twist ekspresyon artışı ile adriamisine hassasiyetin azalması arasında istatistiksel olarak anlamlı bir ilişki olduğu görülmüştür. Tümörlerin taxotere hassasiyeti ile twist ekspresyon farklılıkları arasında ilişki görülmemiştir. Sonuç olarak, Twist, meme tümörlerinde adriamisin hassasiyetinin belirlenmesinde öngörüsel bir markır adayı olarak görünmektedir.Anahtar kelimeler: Twist, ATP-TCA, meme, tümör Breast cancer is the most prevalent cancer among women worldwide. Chemotherapy is being applied in adjuvant and neoadjuvant settings in breast cancer therapy. Adriamycin and taxotere are chemotherapeutics that are commonly used in breast cancer chemotherapy.Chemotherapy is an expensive therapy option. Besides, toxicity of the chemotherapeutics is one of the most important problem affecting their use. Another problem about chemotherapy is the different response rates of the individual patients to the chemotherapeutics used. So, there are many studies aiming to find predictive markers which give information about response to chemotherapeutics.TWIST transcription factor is a basic-helix-loop-helix protein regulating the mesoderm formation and differantiation during development. Mutations of the TWIST1 gene, coding for the twist protein is the cause of Sheatre-Chotzen Syndrome in humans. It has been determined that twist is a part of anti-apoptotic, angiogenic and metastatic processes. Twist transcription factor has also been reported have role in resistance to chemotherapeutic agents.We aimed to investigate the association between twist expression and response to adriamycin and taxotere in breast tumors of the patients, who did not recieve any chemotherapy before. ATP-TCA has been used to determine the responiveness of the tumors to these chemotherapeutics.According to the results of the study, there was a statistically significant correlation between increased levels of twist gene expression and decreased response rates to adriamycine in breast tumors There was no correlation between the twist expression levels and response to taxotere in the tumors. In conclusion, it seems that Twist is a candidate predictive marker for determining the responsiveness of breast tumors to adriamycin chemotherapy.Keywords: Twist, ATP-TCA, breast, tumor 112
