The sheep bot fly, Oestrus ovis, is a very common myiasis of nasal and sinus cavities of sheep and goats causing severe welfare and production implications. As the viability of O. ovis adult flies strictly depends on larval abilities to assimilate and to stock nutrients from the host, it was necessary to investigate proteolytic activities in larval excretory/secretory products (ESP). ESP of O. ovis larvae degrade mucosal and plasmatic components such as mucin, albumin or immunoglobulin G. A preliminary biochemical characterization, using substrate gel analysis and inhibitor sensitivity, demonstrated the presence of at least six major serine proteases (molecular weights from 20 to 100 kDa), mainly trypsin-like, secreted in the digestive tube of larvae. Their involvement in larval trophic activity and evasion from the host immune response is further discussed as O. ovis excretory/secretory serine proteases could represent potential vaccinal targets.

The aims of this study were to characterise trichostrongyle egg excretion in dairy goat flocks under various epidemiological conditions, and to assess the relative importance of animals in their first lactation and goats with high levels of milk production on the total egg output of the flock. The repeatability of egg excretion was estimated on 4 different dates in 14 dairy goat farms in France. Egg outputs were positively skewed on most farms suggesting an aggregated distribution of parasites. Within-year repeatability coefficients ranged from 0.29 to 0.82 suggesting that, on most farms, animals with high egg excretion on one occasion are likely to show high rates of excretion on other occasions. Comparisons of egg outputs from the selected subgroups showed that the high producing goats contributed prominently to egg excretion, particularly at times of peak parasitism. Results from goats in their first lactation were more equivocal. These results provide a rationale for anthelmintic strategies in dairy goats based on phenotypic characters.

Concurrent infections of sheep with Oestrus ovis and trichostrongyles of the digestive tract are common in the field. In order to examine the possible occurrence of interactions between these two parasites and the consequences on parasite biology and the associated pathophysiological changes, an experimental study was conducted on four groups of naïve sheep. Group O was infected repeatedly with O. ovis larvae from D0 to D42 of experiment; group T received a single infection of Trichostrongylus colubriformis on D49; sheep from group OT received both infections, and animals from group C remained uninfected as controls. Parasitological and pathophysiological parameters were measured regularly. At necropsy (D94), the mucosal cellular responses in the nasal cavities and digestive tract from all animals were analysed from histological sections. Infection of the digestive tract with nematodes did not modify the biology of Oestrus populations. In contrast, the presence of O. ovis was related to significant reductions in nematode egg excretion, worm fecundity and worm burdens. These changes were associated with significant modifications in tissular populations of mast cells, globule leucocytes and eosinophils in the respiratory and digestive tracts. These results indicate that parasitic infection in one particular anatomical site induces "at distance" inflammatory reactions of the whole mucosal system. The relationships between the cellular response and the changes in worm biology are discussed.

Background and Purpose For decades, inhibitors of the cystic fibrosis transmembrane conductance regulator (CFTR) chloride channel have been used as tools to investigate the role and function of CFTR conductance in cystic fibrosis research. In the early 2000s, two new and potent inhibitors of CFTR, CFTRinh-172 and GlyH-101, were described and are now widely used to inhibit specifically CFTR. However, despite some evidence, the effects of both drugs on other types of Cl−-conductance have been overlooked. In this context, we explore the specificity and the cellular toxicity of both inhibitors in CFTR-expressing and non–CFTR-expressing cells. Experimental Approach Using patch-clamp technique, we tested the effects of CFTRinh-172 and GlyH-101 inhibitors on three distinct types of Cl− currents: the CFTR-like conductance, the volume-sensitive outwardly rectifying Cl− conductance (VSORC) and finally the Ca2+-dependent Cl− conductance (CaCC). We also explored the effect of both inhibitors on cell viability using live/dead and cell proliferation assays in two different cell lines. Key Results We confirmed that these two compounds were potent inhibitors of the CFTR-mediated Cl− conductance. However,GlyH-101 also inhibited the VSORC conductance and the CaCC at concentrations used to inhibit CFTR. The CFTRinh-172 did not affect the CaCC but did inhibit the VSORC, at concentrations higher than 5 µM. Neither inhibitor (20 µM; 24 h exposure) affected cell viability, but both were cytotoxic at higher concentrations. Conclusions and Implications Both inhibitors affected Cl− conductances apart from CFTR. Our results provided insights into their use in mouse models.

Previous studies using the patch-clamp technique demonstrated the presence of a small conductance Cl−channel in the apical membrane of respiratory gill cells in primary culture originating from sea bass Dicentrarchus labrax. We used the same technique here to characterize potassium channels in this model. A K+channel of 123 ± 3 pS was identified in the cell-attached configuration with 140 mM KCl in the bath and in the pipette. The activity of the channel declined rapidly with time and could be restored by the application of a negative pressure to the pipette (suction) or by substitution of the bath solution with a hypotonic solution (cell swelling). In the excised patch inside-out configuration, ionic substitution demonstrated a high selectivity of this channel for K+over Na+and Ca2+. The mechanosensitivity of this channel to membrane stretching via suction was also observed in this configuration. Pharmacological studies demonstrated that this channel was inhibited by barium (5 mM), quinidine (500 μM), and gadolinium (500 μM). Channel activity decreased when cytoplasmic pH was decreased from 7.7 to 6.8. The effect of membrane distension by suction and exposure to hypotonic solutions on K+channel activity is consistent with the hypothesis that stretch-activated K+channels could mediate an increase in K+conductance during cell swelling.

L’étude avait pour objectif de compléter les données sur l’épidémiologie des strongyloses gastrointestinales dans l’espèce caprine et de préciser l’influence relative de divers facteurs sur ce parasitisme et ses conséquences sur la production laitière. Plusieurs paramètres parasitologiques et physiopathologiques ont été mesurés dans un troupeau de 120 chèvres laitières au pâturage pendant deux ans. Les pics de parasitisme ont été observés en début d’automne et les genres dominants identifiés étaient Teladorsagia et Trichostrongylus. La conduite du troupeau selon un système de pâturage tournant ou continu n’a eu que peu d’influence sur les infestations par les nématodes. En revanche plusieurs facteurs de réceptivité de l’hôte ont été identifiés. Au cours des deux années, une excrétion d’œufs de parasites plus forte, entraînant une contamination accrue du pâturage, a été relevée dans deux groupes d’animaux au sein du troupeau : d’une part, chez les chèvres en première saison de pâturage et notamment les primipares ; d’autre part, chez les chèvres présentant le meilleur niveau de production de lait. Chez ces dernières, des répercussions physiopathologiques plus marquées du parasitisme ont aussi été notées, en particulier lors des pics d’excrétion d’œufs de strongles. The infection was not influenced by the mode of grazing (rotational vs continuous). In contrast, host factors were identified which modulate the of parasitism. In the first grazing season, and particularly those in first lactation, excreted more eggs and hence contributed largely to pasture contamination. In addition, the goats within the flock with the highest level of milk produced were more severely infected. They were also more to the impact of parasitism on milk yield. The relevancy of the identification of these animals a nock as a major epidemiological risk for infection is discussed in relation with a possible targeted application of anthelmintic treatments.

Patch-clamp experiments were undertaken on primary cultures of respiratory cells originating from sea bass (Dicentrarchus labrax) gills. A small-conductance Cl- channel of 8 pS was characterized in cell-attached configuration with 140 mM N-methyl-D-glucamine-Cl in the pipette and bath solutions. No activity was recorded below a membrane holding potential of +20 mV (-Vp, referenced to the pipette solution), and the channel showed an inward rectification. In the inside-out configuration the Cl- channel was active at all membrane holding potentials. Its open probability strongly increased with membrane depolarization. The channel activity could be increased by the application of protein kinase A+ATP. This channel was inhibited by 5-nitro-2-(3-phenylpropylamino)-benzoic acid, diphenylamino-2-carboxylic acid, and I- and was insensitive to 4,4'-diisothiocyanostilbene-2,2'-disulfonic acid. The presence of Cl- channels in the apical membrane of respiratory cells provides additional evidence for an important role of this cell type in the control of ion homeostasis of seawater fish.

Many monoclonal antibodies reactive with bovine leukocyte differentiation antigens are now available. Immunohistochemical staining on frozen sections using these monoclonal antibodies permits study of the functional morphology of bovine lymph nodes. Our study confirms usually accepted notions (B and T dependent-zones) and supplies complementary data about the repartition of CD4 cells (particularly intrafollicular positive cells), gamma delta T cells, MHC II expression and dendritic leukocytes.

In France a survey of the use of antimicrobials in veterinary medicine was set up in connection with the Committee for the Marketing Authorization of Veterinary Medicinal Products. This survey has emphasized the importance of antimicrobials in veterinary medicine and the reality of antibiotic resistance in both man and animals. Certain epidemiological data tend to demonstrate that the use of antibiotics in animals increases the risk of antibiotic resistance in man, through selection in animals of resistant pathogenic bacteria or of a non-pathogenic resistant bacteria carrying resistance plasmids. Four proposals are put forward for the control of these different risks: reserve certain antibiotics (life-saving antibiotics) for human medicine, conform to a code of good practices for antibiotic use in animals, strengthen and connect up existing networks for the epidemiosurveillance of antibiotic resistance, provide for studies on the ecological impact of antibiotics in the registration dossiers for veterinary medicinal products. These evolutive measures, based on scientific progress in microbiology, epidemiology and clinical studies should respond satisfactorily to the expectations of all concerned.

International audience; In conventional mice, murine strains of prions show highly specific phenotypes, as defined by several parameters including incubation period of disease, nature of clinical signs, western blot profile of abnormal prion protein (PrPSc), distribution of vacuoles and PrPSc in the brain. More surprisingly, this remains independent of where prions have had access to the brain, supporting the view that, through a determinism which remains obscure, the multiplication of the infectious agent and the PrP conversion process is favoured in brain specific areas. We sought to examine if the inoculation of natural scrapie strains to transgenic mice expressing ovine PrP would lead to similar findings. We have therefore 1) analysed the phenotype of the cloned ovine prion strain 127S after intraperitoneal or intracerebral infection of tg338 mice overexpressing the VRQ allele of sheep prion protein; 2) compared the cerebral PrPSc distribution of this strain with another cloned and phenotypically distinct one, LAN19K, after intravitreal injection. Our results show that 1) the apparent phenotype of the 127S strain is clearly influenced by the route of infection since intraperitoneally versus intracerebrally tg338 mice show different clinical signs, levels of PrPSc accumulation and distribution pattern of PrPSc and vacuoles in brain; 2) 127S and LAN19K show a similar propagation into the brain until the terminal stage of the disease, which was mainly restricted to the ocular projections. This common PrPSc distribution pattern strikingly differed from that obtained after intracerebral or intraperitoneal inoculation of these strains. Our data therefore suggest that, depending on the model, the capacity of prion strains to replicate in brain specific areas may be influenced by the route of infection.

Although the use of tanniferous plants or condensed tannins as an alternative to anthelmintics to control gastrointestinal nematodes has been largely documented in sheep, studies remain scarce in goats. The objective of this study was therefore to assess the possible impact of condensed tannins in goats infected with adult Haemonchus contortus. Two groups of cull goats were experimentally infected with 10.000 L3 of H. contortus. After 4 weeks, quebracho extracts, representing 5% of the diet DM, were administered for 8 days to one of the two groups. Goats of the second group remained as controls. One week after the end of quebracho administration, the goats were euthanised. Individual egg excretion and pathophysiological parameters were measured weekly during the study. At the end of the study, worm counts were assessed and histological samples from the abomasa were taken to count the numbers of mucosal mast cells, globule leukocytes and eosinophils. The administration of tannins was associated with a significant decrease in egg excretion, which persisted until the end of experiment. This reduction was not associated with any difference in worm number but with a significant decrease in female fecundity. No significant changes in the mucosal density of the three inflammatory cell types were detected between the two groups. These results indicate that the major consequence of tannin consumption in goats is a reduction in worm fecundity and egg output, which does not seem related to significant changes in the local mucosal response.

International audience; The ubiquitously expressed cyclin T1 gene encodes for a protein involved in human immunodeficiency virus type 1 (HIV-1) transcription activation. The goat gene was recently shown to share an expression pattern similar to that of its endogenous counterpart when incorporated into mice using a BAC insert. To assess if its promoter could target ubiquitous expression of the bovine Prnp in transgenic mice, two constructs carrying either 1 or 30 kb of cyclin T1 5'-flanking sequences were built and microinjected. Both constructs resulted in the unexpected high male germ cell-specific expression of the prion protein. These data re-question the suspected location of the cyclin T1 gene regulatory elements.

Infection by Oestrus ovis is common in Lacaune dairy ewes of Roquefort cheese area (Aveyron, France). It is believed by local breeders that there is a close relationship between nasal myiasis and the incidence of enzootic nasal tumour. In order to check these anecdotal reports, a serological survey was done on 658 breeding ewes before turn-out and 897 breeding and primiparous (hoggets) ewes at the end of the grazing season. By the time of sampling, it was clear whether the sheep were infected at the end of the winter or had been re-infected over summer. In April and September, 40.7 and 26.3%, respectively, were free of O. ovis infection, indicating that the autumn treatment was not completely effective and that O. ovis adult flies were circulating during the summer in many flocks. There were no differences in the incidence of adenocarcinoma between the groups indicating that there is no relationship between O. ovis infection and the presence of the cancer. Differences in milk production between the three groups were not statistically significant (Anova test P>0.05). In flocks where 1-5% of the ewes were infected or in non-infected flocks, ewes produced 3.6 and 8.56%, respectively, more milk than ewes from flocks where more than 5% of animals were infected. For primiparous ewes, the differences were of 8.5 and 12.24%.

Infections of the gastrointestinal tract with parasitic nematodes remain one of the main limiting factors in grazing dairy goats. The usual mode of control of these parasitic diseases has up to now been based on the repeated use of anthelmintics. However, the prevalence rates of anthelmintic resistances, in particular to benzimidazoles, are now particularly high in the French dairy goat production. This situation makes it mandatory to reconsider the usual mode of control of these nematodes and to look for short term, alternative solutions which combine the control of gastrointestinal infections and management of anthelmintic resistances. One of the possible options is to leave a part of the flock without treatment during the grazing season in order to maintain alleles of susceptibility to anthelmintics within the worm populations. Previous epidemiological observations identifying the categories of host populations at risk are presented which provide the rationale for targeted applications of treatments. The results of assays on experimental flocks and from farm surveys examining the advantages and drawbacks of selective treatments are presented. The value of these results in combination with other alternative solutions of control are discussed in order to use minimum treatments with maximum benefits.

The mean (sd) concentration of plasma 20beta-dihydrocortisol in 126 scrapie-affected sheep was 5-5 (7.0) ng/ml compared with 1.1 (0.7) ng/ml in 52 healthy sheep. The mean (sd) concentration of creatinine in the urine of 93 scrapie-affected sheep was 2.43 (1.56) microg/ml compared with 0.94 (0.86) pg/ml in 49 healthy sheep and 1.10 (0-95) pg/ml in 25 sheep with other diseases. These discriminant analyses carried out on healthy and scrapie-affected sheep showed that plasma 20beta-dihydrocortisol and urinary creatinine were the best predictors of the disease, and classified correctly 98 per cent of healthy sheep and 82 per cent of scrapie-affected sheep.

The targeted application of anthelmintic treatments represents one of the current available solutions to slow down the development of anthelmintic resistance within worm populations. Within an experimental flock of dairy goats, control of gastrointestinal parasitism by such selective treatments, targeting the most receptive animals within a flock, was previously found to be effective and to have no detrimental consequences on milk production. The objectives of the current study were to verify the validity of this method in farm conditions. Eleven dairy goat farms from three main areas of production in France were surveyed for 2 years. In six farms, the survey was prolonged for a third year. During year 1, systematic treatments were applied during the grazing season whereas in year 2 and or year 3, treatments were given exclusively to the goats in first lactation and to the multiparous ones with the highest potential of milk production. The level of nematode infection was measured four times per year through individual coproscopical examinations and the mean annual production of milk was recorded. No significant changes in egg excretion nor in milk production were noticed in any farm during years 2 or 3 after switching from the systematic to the selective mode of treatments. These results confirmed that targeted application of anthelmintics might represent a way to combine the control of trichostrongyles and the prevention of anthelmintic resistance.

The effects of copper supplementation on the copper status of 40 late-pregnant Aubrac beef cows grazing a copper-deficient pasture and later fed a marginally deficient diet were studied for five months. They were divided into four equal groups; the control group received no copper supplement, groups 1 and 2 received copper as copper sulphate at 10 and 30 mg/kg of diet dry matter (DM), respectively, for five months, and group 3 received 120 mg/kg of diet dry matter for 10 days. Plasma copper concentration and the activity of erythrocyte superoxide dismutase (esoD) were measured at the beginning of the experiment, in the cows and calves during weeks 1 and 3 after calving, and in the calves before they were turned out to pasture at a mean (sd) age of 51 (26) days. In spite of the low dietary copper content (4.2 mg/kg of DM), the plasma copper concentration of the control cows increased during the winter. All the copper supplements resulted in normal and similar plasma copper concentrations in the cows after calving, but the concentration decreased slightly between weeks 1 and 3 after calving in the group supplemented for 10 days. The treatments did not affect the eSOD of the cows. The calves born to the four groups showed the same patterns of plasma copper and esoD. Compared with the cows, the calves had low plasma copper concentrations at week 1 and values in the normal range at week 3; their esoD was high at weeks 1 and 3 but decreased after week 3.

From December 1998 to March 2000, caecum and ascendant colon of 42 horses naturally infected with cyathostomes were collected during routine necropsy or from a local slaughterhouse. Changes in the numbers of mucosal and submucosal mast cells (MMC and SMMC), intraepithelial, mucosal and submucosal eosinophils (IE, ME and SME) in the large intestine were investigated by histochemical techniques in relation to the worm burdens. The effect of age was examined in three subgroups: 6-24-month-old horses (group 1), 2-10-year-old horses (group 2) and horses more than 10 years of age (group 3). No globule leucocytes were detected in any sections. No significant variations with breed or sex were observed in cell counts. The main variations were higher eosinophil counts in groups 2 and 3 and a marked increase of the MMC counts in the oldest horses (group 3). For each cell type, the infiltration was homogeneous and generalised along the large intestine. In the whole horse sample, the IE numbers were the only parameters that correlated with the MMC and SMMC counts. Very few significant relationships were found between mast cells and eosinophils in groups 1 and 3, whereas numerous positive correlations were recorded in group 2. In the whole horse sample, several correlations were found between different cell counts and cyathostome burdens. The numbers of larvae, adult worms, and the total worm burdens were related to some of the tissular eosinophil counts while the percentage of early third stage larvae (EL3) was linked to mast cell densities. These relations between cells and worm populations showed variations with age. In group 1, most of the significant associations were found between eosinophil counts (IE and SME) and the total numbers of larvae and worms; in group 2, they were noticed between the three eosinophil types and the total cyathostome burdens. In group 3, a MMC hyperplasia was observed and correlations were mostly recorded between these MMC and the total numbers of adult worms or the percentage of EL3. Several associations were also detected between eosinophils (mainly ME and/or IE) and different cyathostome burdens. These variations in the relationship between inflammatory cells and cyathostomes seemed to be consistent with the cellular changes observed among the three age groups. These results suggest that eosinophil and mast cell infiltrations quantified in the large intestine wall might be associated with cyathostome infection.

Resistance to anthelmintics in nematode parasite of the digestive tract is a major concern in small ruminants and particularly in goats. One possible solution to limit the development and spread of resistance is to give treatments on a selective basis, i.e. by targeting the most susceptible animals within a flock rather than treating all the animals. In dairy goats, epidemiological studies have shown that, within a flock, goats in first lactation and the multiparous ones with the highest level of milk production were highly receptive to parasite infection. The objective of the study was to assess whether selective treatments could achieve a level of control of nematodes similar to systematic drenching. A similar experimental design was applied for two successive years. An experimental flock of 120 dairy goats was divided into two groups. All the goats from group SYS (systematic treatment) were drenched at mid-grazing season (July). In contrast, anthelmintics in group SEL (selective treatment) were restricted to the goats in first lactation plus the high producers. Overall, these treated goats represented 48% of the flock in year 1 and 66% in year 2. After treatments, both groups grazed on separate pastures. Parasitological and pathophysiological measurements were performed monthly. The results indicate a similar level of egg excretion in the two groups. The pathophysiological parameters (pepsinogen and phosphate concentrations) were also similar in the two groups, as well as the milk production for the 2 years. These results suggest that a targeted use of anthelmintics may allow efficient control of gastrointestinal nematodes whilst resulting in a predicted reduction in the selection pressure for the development of anthelmintic resistance.

Forty-two horses from Normandy (France) were examined post-mortem for small strongyle infections from October to March. In the positive horses, total worm numbers ranged from 234 to 90,247 (mean 11,297). Encysted larvae represented the major part of the total cyathostome burdens with a high percentage (83%) being early third stage larvae. They were mostly recovered from the caecum (48%) and ventral colon (40%) and were less present in the dorsal colon (12%). Adult cyathostomes were mainly located in the ventral colon (64%) and less frequently in the dorsal colon (27%) and caecum (9%). Twenty species of Cyathostominae were identified. The 10 most prevalent species (in sequence of prevalence) were Cyathostomum coronatum, Cylicocyclus nassatus, Cylicocyclus insigne, Cyathostomum catinatum, Cylicostephanus goldi, Poteriostomum imparidentatum, Cyathostomum labiatum, Cylicocyclus ultrajectinus, Cylicostephanus calicatus and Cylicostephanus minutus which comprised 84% of the total adult population. Twelve species showed a site preference in the ventral colon, five in the dorsal colon and only one in the caecum while two species were collected in nearly equal numbers from the ventral and dorsal colon. The number of species per horse ranged from 1 to 12 with a median of 5. Infections with singletons occurred in 12.5% of the positive horses while multiple infections were encountered in 87.5%. A positive correlation was found between the intensity of cyathostome infection and its diversity, measured either by the number of occurring species or Shanon indexes.

(Diptera: Oestridae), the nasal bot fly, has a relatively short free-living life cycle outside of the host, and therefore it is necessary to know when the parasitic period occurs in order to prevent the clinical signs and economic losses caused by this parasite. The length of this parasitic portion of the life cycle is quite variable: a few weeks to several months depending on the season and climatic conditions. Surveys of Oestrus ovis larval populations in sheep show different results on the number of generations according to the local climate. Mean monthly larval profiles of L1 and L3 burdens of sheep from West African Sahelian countries, Mediterranean countries (Morocco, Tunisia and Sicily) and Southwest France were compared. Valuable information on the suspected extension of the fly season is obtained showing the period of infection in each area. Les profils des populations mensuelles moyennes de larves 1 et 3 relevés dans des pays sahéliens, dans des régions méditerranéennes (Maroc, Tunisie et Sicile) ainsi que dans le sud de la France, ont été comparés. Des informations intéressantes sur la durée de la période d'activité des mouches ont ainsi été obtenues. Ces éléments pourraient être une aide efficace pour déterminer les périodes de traitement et le médicament le mieux adapté au risque parasitaire présent.

Parasite infection of the gastrointestinal tract with helminths or protozoa induces detrimental effects on host tissues and host physiology, which have been extensively studied and reviewed. However, parasitism of the digestive system is also associated with adaptive, compensatory phenomena based on changes in host physiology or structures and which tend to counterbalance the negative consequences. The objective of this review is to describe these adaptive processes and their possible underlying mechanisms. Different processes which tend to attenuate the effect of either the loss of appetite, the intestinal malabsorption or the increased tissue losses have been assessed. These processes have been reported both for helminth and protozoan infections, where they present similar characteristics. The mechanisms involved in the adaptation to parasitism remain largely unidentified. The role of feedback mechanisms based on host regulation, possibly through gastrointestinal hormones, has been raised. On the other hand, some data support the proposal that parasites themselves may initiate some of the adaptive processes and consequently favour their own survival. These adaptive phenomena appear to be an essential component in the dynamic balance between host and parasites. Also, parasite infections represent unique models to study the adaptation of the gastrointestinal tract to aggressors.

National audience; Un suivi épidémiologique, fondé sur des prélèvements mensuels, a été réalisé dans quatre exploitations caprines du sud-est de la France (Drôme et Ardèche) afin d'obtenir des informations sur la diversité et l'intensité du parasitisme digestif des chèvres adultes et examiner son impact sur l'évolution des courbes de lactation. Le but était aussi de vérifier si l'analyse de ces courbes peut servir d'outil d'aide à la décision pour l'administration de traitements antihelminthiques.
